本实验应用离体大鼠Langendorff灌流模型,研究模拟缺血预处理(IP)对心脏的保护作用。发现IP可引起心室颤动阈(VFT)升高、心律失常发生率和严重程度降低、心室有效不应期短时延长。IP对心脏的保护作用在预缺血后30min已基本消失。提示IP对离体大鼠心肌有保护作用,且时间较为短暂。 相似文献

本工作分别在大鼠的多种器官上观察到缺血预处理(PC)对缺血/再灌注损伤的保护作用具有器官普遍性,其对血管床的保护是其器官保护的机制之一;在体外培养的乳鼠(兔)心肌细胞及大鼠(兔)胸主动脉血管平滑肌细胞等多种细胞等多种细胞上观察到蛋白激酶C(PKC)激活及PKC介导的蛋白磷酸化增强是PC细胞保护的共有环节。碱性成纤维细胞生长因子可以模拟PC的细胞保护作用,提示药物预处理对于防治缺血有关的疾病可能上人 相似文献

目的:研究肢体缺血预处理对大鼠肝缺血/再灌注损伤是否具有保护作用。方法:雄性SD大鼠32只,随机分为对照组(S组);缺血/再灌注组(I/R组);经典缺血预处理组(IPC组);肢体缺血预处理组(远端缺血预处理组,RPC组)。S组仅行开腹,不作其他处理;IPC组以肝缺血5min作预处理;RPC组以双后肢缺血5min,反复3次作预处理,2个预处理组及I/R组均行肝缺血1h再灌注3h。取血用于血清谷丙转氨酶(ALT)与血清谷草转氨酶(AST)检测。切取肝组织用于测定湿干比(W/D)、中性粒细胞(PMN)计数及观察显微、超微结构的变化。结果:与I/R组比较,IPC组,RPC组ALT,AST,W/D值,及PMN计数均明显降低(P〈0.01),肝脏的显微及超微结构损伤减轻。结论:肢体缺血预处理对大鼠肝脏I/R损伤有明显的保护作用,强度与经典缺血预处理相当,其机制可能与抑制肝脏炎症反应、减轻肝脏水肿、改善肝组织微循环有关。 相似文献

目的:探讨肾缺血预处理对家兔心脏缺血/再灌注(I/R)损伤的影响及意义。方法:32只大耳白家兔随机分为假手术(SO)、心脏I/R、经典缺血预处理(CIPC)及肾缺血预处理(RIPC)4组。观察各组心肌梗塞面积、左室舒缩功能、心脏超微结构及心律失常发生率的变化。结果:CIPC、RIPC组,心肌梗塞面积、再灌性心律失常发生率较I/R组明显降低,左室舒缩功能明显恢复(P<0.01),心脏超微结构损伤明显减轻。结论:RIPC可诱导出与CIPC类似的心脏保护效应。 相似文献

目的:探讨连续多次短暂缺血预处理对缺血/再灌注损伤心肌的保护作用及机制。方法:采用大鼠离体心脏Lan-gendorff灌流模型,观察缺血预处理对心肌缺血/再灌注后不同时间点冠脉流出液中AST、CPK、UDH及冠脉流量,心肌组织中SOD、LPO以及再灌注性心律失常的影响。结果:缺血预处理可以减少缺血/再灌注损伤的心肌冠脉流出液中AST、CPK、LDH的含量,提高心肌SOD活性,降低LPO水平,并且抑制再灌注性心律失常的发生,提高再灌注期间的冠脉流量。结论:缺血预处理对心肌缺血/再灌注损伤具有一定保护作用。 相似文献

目的:探讨缺血预处理对大鼠缺血再灌注心肌细胞凋亡及相关基因Bcl-2和Bax蛋白表达的影响。方法:制备缺血预处理(IP)和缺血再灌注损伤(I/R)模型,采用末端标记技术(TUNEL)检测心细胞凋亡,应用免疫组织化学方法检测Bcl-2和Bax的蛋白表达,结果:缺血再灌注组心肌细胞凋亡率明显比正常对照组高(P<0.05),而缺血预处理组心肌细胞凋亡率明显比缺血再灌注组低(P<0.05),缺血再灌注组Bcl-2表达阳性细胞率明显比正常组低(P<0.05),而缺血预处理组Bcl-2表达阳性细胞率明显较缺血再灌注组高(P<0.05)。缺血再灌注组Bax表达阳性细胞率明显较正常组高,而缺血预处理组Bax表达阳性细胞率明显较缺血再灌注组低(P<0.05)。结论:缺血再灌注可诱导心肌细胞凋亡,缺血预处理可减少心肌细胞凋亡,Bcl-2和Bax的蛋白表达在心肌凋亡发生中起重要作用,缺血预处理可上调Bcl-2蛋白表达和下调Bax蛋白表达。 相似文献

目的 :观察缺血预处理 (IPC)对大鼠肺缺血 /再灌注 (I/R)损伤的保护作用 ,并初步探讨其作用机制。方法 :建立离体大鼠肺灌流模型 ,36只wistar大鼠随机分为对照组、I/R组和IPC组 ,处理完毕后分别测定平均肺动脉压(MPAP)、肺组织湿 /干重比、支气管肺泡灌洗液中肺表面活性物质磷脂及表面张力改变 ,肺组织标本送电镜检查。结果 :①电镜下观察IPC组肺损伤明显减轻。②肺组织湿 /干重比值IPC组为 4.41± 0 .2 4,显著低于I/R组 ,但仍高于缺血前 (P <0 .0 1) ;③IPC组大鼠缺血 1h后MPAP为 ( 1.88± 0 .2 9)kPa ,明显低于I/R组 (P <0 .0 1) ;④IPC组支气管肺泡灌洗液中总磷脂为 ( 2 33 .42± 14.0 5 ) μg/kg ,大聚体为 ( 10 5 .39± 6 .17) μg/kg ,与I/R组相比显著增高 ,但低于对照组 (P <0 .0 1) ,三组之间小聚体含量没有显著差异 ;⑤IPC组表面张力为 ( 36 .88± 3.49)mN/m ,显著低于I/R组 ,与对照组相比则无显著性差异 (P >0 .0 5 )。结论 :缺血预处理对大鼠肺I/R损伤有保护作用 ,保护机制可能与促进肺表面活性物质 (PS)磷脂分泌、改善PS组成 ,从而提高PS功能有关。 相似文献

Oxidative stress can cause extensive damage to cardiac tissue under reperfusion conditions. However, preconditioning the myocardium may diminish these negative effects and alleviate reperfusion injury. There are a variety of preconditioning therapies, such as ischemic preconditioning (IPC) and hypoxic preconditioning (HPC), each targeting specific channels, receptors, and/or intracellular molecules. Ischemic preconditioning involves brief periods of ischemia followed by brief periods of reperfusion, thus strengthening the cardiac resistance for a longer period of ischemia. IPC involves complex mechanisms, some of which are still not completely understood today. Nevertheless, many studies have already established models of IPC. In addition, similar to IPC, HPC has also been recognized as preventing reperfusion injury. Reactive oxygen species (ROS) are known mediators of IPC and HPC. Particularly, mitochondria-generated ROS initiate activity of several beneficial preconditioning pathways. The role of ROS is paradoxical; low levels of ROS are key factors in signaling IPC/HPC, but high levels of ROS can contribute to increased oxidative stress on cardiomyocytes. Therefore, it is important to determine the molecular mechanism of IPC and HPC to avoid excessive accumulation of ROS to prevent cardiac injury. In this review, we will outline IPC and HPC, explaining the putative role of ROS in both pathways. We will also discuss preconditioning efficacy in certain conditions such as exercise and how the aging myocardium responds to preconditioning therapies. 相似文献

Effects of hypoxic preconditioning on adenosine (ADO) and its A(1) receptor were studied in Kunming mice. The ADO content and its metabolites in the brain were measured by a specific enzymatic method; a radioligand binding method was used to study the ADO A(1) receptor. The ADO content of the hippocampus in group C (exposure to 4 runs of hypoxia) was markedly higher than that in group A (control, without exposure to hypoxia and B (exposure to 1 run of hypoxia), showing that the ADO content could be cumulatively increased in the hippocampus, which was more sensitive to ischemia and hypoxia, during acute and repeated exposure to hypoxia. A(1) receptor density in group C was significantly lower than in group A and no difference was seen between groups B and C; A(1) receptor affinity in the hippocampus, pons and medula oblongata in group C was significantly higher than in group A, implying that during hypoxic preconditioning there might be some mechanisms preventing A(1) receptor density from decreasing further and making A(1) receptor affinity increase in some brain regions. These results indicate that cumulatively increased ADO in the hippocampus via A(1) receptor may play a neuroprotective role in the CNS as an inhibitory neuromodulator and thus contribute to the formation and development of acute hypoxic adaptation or tolerance. 相似文献

Ischemic preconditioning (IPC, i.e. increase in the organ resistance to a prolonged ischemia which occurs after a brief ischemic challenge) seems to be one of the most powerful endogenous cardioprotective mechanisms known to date. Current data regarding molecular mechanisms of early (classic) IPC as well as the second window of protection are reviewed in the context of the concept of sequential three-staged development of protective effect. we are aware refute it. We generalize the ABH in light of these data and observations. The population of zooxanthellae within
an organism is dynamic, the diversity of zooxanthellae is both surprising and difficult to ascertain, and field experiments
demonstrate both turn-over in zooxanthella types and habitat-holobiont correlations. Dynamic change in symbiont communities,
and the idea of an equilibrium or optimal community that matches the environment at a particular place and time, are concepts
that underlie or emerge from much of the recent literature. The mechanism we proposed to explain responses to acute bleaching
appears to operate continuously, thereby enabling the host-symbiont holobiont to track even subtle environmental changes and
respond promptly to them. These findings enhance the potential importance of the ABH in the outcomes of acute bleaching, which
can (1) accelerate this process of holobiont change, and (2) change the set of possible trajectories for how symbiont communities

The kudzu bug or bean plataspid, Megacopta cribraria (Fabricius), is native to Asia where it appears to be widely distributed (although the taxonomy is not entirely clear), but is infrequently a pest of legumes. This bug appeared in 2009 in the southeastern United States, where it is closely associated with kudzu, Pueraria montana Lour. [Merr.] variety lobata [Willd.] Maesen & S. Almeida. However, the insect has become a consistent economic pest of soybeans, Glycine max (L.) Merr., and some other leguminous crops in areas where large numbers can build in kudzu, in addition to being a considerable nuisance in urban landscapes where kudzu occurs. The insect has remarkable capacity for movement and has spread rapidly from nine Georgia counties in 2009 to seven states in 2012. Despite being a nuisance in urban areas and a crop pest, high populations of the bug also reduce the biomass of kudzu, which is itself a seriously problematic invasive weed, complicating the status of M. cribraria in its expanded range. Extant predators and a pathogen in the US have been observed attacking kudzu bugs in the laboratory and field, but no parasitism of eggs or nymphs has been observed to date. A single record of parasitism of an adult kudzu bug by a tachinid fly is known from the US, but no other adult parasitism has been observed in the US or elsewhere. Extant enemies may eventually significantly reduce the bug’s populations, but at present native enemies appear to be insufficient for the task, and exotic enemies from the kudzu bug’s native range may offer the best possibility for effective biological control in the US. Based on the available literature, the best option for an importation biological control program appears to be the platygastrid egg parasitoid Paratelenomus saccharalis (Dodd) because of its apparent host specificity, intimate biological linkages with M. cribraria, and wide geographic distribution in the Eastern Hemisphere. Other natural enemies may eventually emerge as good candidates for importation, but at present P. saccharalis appears to be the most promising. 相似文献

Ostracism refers to the general process of rejection and exclusion, observed in human groups and in many other species. It occurs as “shunning” in small homogenous groups like the Amish or as rejection among children. Ostracism in various forms is also deeply embedded in our own legal tradition, and is used in the formal and informal legal procedures of other cultures, used to maintain order, to punish deviance, and to increase social cohesion. Hence, it is plausible to hypothesize that human ostracism may have physiological substrates or biological functions in addition to cultural, moral, and legal dimensions. Biological research shows that human emotions (anger, fear, reassurance, self-confidence) involve responses of the limbic system as well as ideas or thoughts in the cerebral cortex and that human behavior continually integrates biological and cultural factors. The legal system expresses and channels human behavior. For this reason laws should be more effective if their functions complement (rather than ignore) the function of the behavior being regulated. To look at law as it affects human behavior in the light of the life sciences does not imply the intention of finding a universally valid “natural law,” akin to theological or ideological doctrine. On the contrary, we find that humans can form radically different social systems due to the plasticity of their behavior. However, an interdisciplinary analysis of ostracism as a common area of behavior, combining biology, law, and the social sciences can produce insights that none of these fields alone can provide. Such an approach should increase our understanding of human nature and the functions of law. 相似文献

Bryostatins are a family of protein kinase C modulators that have potential applications in biomedicine. Found in miniscule quantities in a small marine invertebrate, lack of supply has hampered their development. In recent years, bryostatins have been shown to have potent bioactivity in the central nervous system, an uncultivated marine bacterial symbiont has been shown to be the likely natural source of the bryostatins, the bryostatin biosynthetic genes have been identified and characterized, and bryostatin analogues with promising biological activity have been developed and tested. Challenges in the development of bryostatins for biomedical and biotechnological application include the cultivation of the bacterial symbiont and heterologous expression of bryostatin biosynthesis genes. Continued exploration of the biology as well as the symbiotic origin of the bryostatins presents promising opportunities for discovery of additional bryostatins, and new functions for bryostatins. 相似文献
