Degree of unsaturation of sn-2 located fatty acyl side-chainsin typical membrane phospholipids has a marked effect on surfaceten si on-associated parameters of monolayers of these lipidsover aqueous sub-phases. For a fixed area monolayer in a completelyexpanded state, an increase in the number of cis double bondswas found to cause a concomitant increase in surface tension.For the same fatty acids incorporated into phosphatidylcholinemolecules, surface pressure/molecular area isotherms show thatsn-2 linoleoyl monolayers manifest markedly higher surface pressuresthan sn-2 oleoyl ones in the fully compressed state. Furthermore,the isotherms and monolayer collapse points indicate a greaterrigidity of the oleoyl species monolayer. The strong correlationof these effects with molecular radius, rotational diffusionconstant and total molecular peripheral properties of the monolayermay be determined by rotational micro viscosity experiencedby the molecules. This in turn is determined by molecular radiusthrough the total molecular peripheral length. It is suggestedthat a contributing factor to membrane bioregulation may besuch changes in surface tension-associated parameters arisingfrom the structure of the unsaturated fatty acyl phospholipidside-chains. In a biological membrane the surface tension would,therefore, be an average of headgroup effects and of degreeof fatty acyl side-chain unsaturation. To investigate the genetic mechanisms regulating the transitionfrom vegetative to reproductive phase in Arabidopsis, doublemutants between two embryonic flower (emf) and 12 differentlate-flowering mutants were constructed and analyzed. Doublemutants in all combinations displayed the emf phenotypes withoutforming rosettes during early development; however, clear variationsbetween different double mutants were observed during late development,fwa significantly enhanced the vegetative property of both emfmutants by producing a high number of sessile leaves withoutany further reproductive growth in emf1 fwa double mutants.It also produced numerous leaf-like flower structures similarto those in leafy ap1 double mutant in emf1 fwa double mutants.Nine late-flowering mutants, ft, fca, ld, fd, fpa, fe, fy, fha,and fve, caused different degrees of increase in the numberof sessile leaves, the size of inflorescence, and the numberof flowers only in weak emf1 and emf2 mutant alleles background.Two late-flowering mutants, co and gi, however, had no effecton either emf1 and emf2 mutant alleles in double mutants. Ourresults suggest that FWA function in distinct pathways fromboth EMF genes to regulate flower competence by activating geneswhich specify floral meristem identity. CO and GI negativelyregulate both EMF genes, whereas the other nine late-floweringgenes may interact with EMF genes directly or indirectly toregulate shoot maturation in Arabidopsis. 1 To whom correspondence should be addressed.  相似文献

Permeability coefficients for the cell membrane of Nitella jlexilishave been determined for some organic substances by measuringtheir transcellular transport with the double chamber method.Benzyl alcohol is rapidly taken up in the vacuole of the cell.In the double chamber method benzyl alcohol taken up in thevacuole at one end of the cell is not as rapidly transportedto the other end as expected from the rate of protoplasmic streamingin the vacuole. This can be explained by assuming the presenceof a "non-streaming part" in the vacuole. The uptake of ureainto the streaming part of the cell is stimulated by directlyilluminating the part where the cell is immersed in urea solution. (Received March 10, 1971; )  相似文献

Near-isogenic lines have been produced in pea to examine theeffects of genes at the r and rb loci on the growth and developmentof the seed. The rb alleles were found to have an effect onthe growth of the testa as well as on that of the embryo incontrast to those at the r locus which only affect the latter.The relationship between the fresh and dry weights of the embryosof rb isoline and the double mutant differed from that of theround-seeded wild-type line. The use of the 0.22 µm Millipore cellulose membrane forthe measurement of the in vivo absorption of aquatic particlesby the light-transmission method has been explored relativeto the performance of the currently used 0.7 µm WhatmanGF/F glass-fiber filter. The standard solvent extraction processfor pigment removal has been replaced by a newly developed processbased on NaClO bleaching. The tests carried out indicate thatthe two filters exhibit comparable performances. It has beenshown that a double measurement, performed first in the particlesretained in the GF/F filter and then on the sample obtainedpassing the GF/F filtrate through a Millipore membrane, allowsfor the selective determination of the absorption of the particlefraction with diameter in the 0.22–0.7 µm range.  相似文献

The effects of increasing concentrations of soil copper on anumber of leaf structural parameters in oregano plants werestudied to determine the effect of copper toxicity. Copper-stressedleaves were small and chlorotic and underwent a thickening oftheir lamina, due principally to an increase in the number andvolume of mesophyll cells. The number of stomata and glandularand non-glandular hairs increased significantly. Chloroplastsof mesophyll cells declined dramatically in number and volume.Grana and stroma thylakoids of chloroplasts did not undergoany noticeable alterations, but starch grains disappeared, plastoglobulibecame larger and the double membrane limiting the chloroplastbecame dilated. The Permeability of the Guard Cell Plasma Membrane and Tonoplast   总被引:4,自引:0,他引:4  
Uptake experiments and efflux compartmental analysis of planthormones, osmotica and toxins using ‘isolated’ guardcells of Valerianella locusta and guard cell protoplasts (GCP)of Vicia faba were performed in order to study the permeabilityproperties of guard cell plasma membrane and tonoplast. Theplasma membrane of guard cells exhibits a higher permeabilitythan plasma membranes of mesophyll cells for most solutes investigated.The permeability coefficients (Ps calculated for the guard cellplasma membranes are also significantly higher than the Ps valuesfor the guard cell tonoplast. This applies also for protonatedABA. We suppose that the high permeability for ABAH could bepart of the target cell properties. A Collander analysis demonstratesa linear correlation between Ps, values and the ratio Kr/Mr1,5for both plasma membrane (r = 0.87) and for the tonoplast (r=0.93). Because of deviations from the observed correlations,the permeation of some solutes (ABA, GA, IAA through the tonoplast;methylamine through the plasma membrane) seems to be facilitatedby an additional transport mechanism. The Collander analysisof the plasma membrane of GCP shows very similar results tothe analysis of the plasma membrane of ‘isolated’guard cells, indicating that isolation of protoplasts does notalter the permeability of the guard cell plasma membrane. The supramolecular organization of the vacuole membrane (orof the membranes of mucilage sacs) in 27 species of red algaeis studied in replicas of rapidly frozen and fractured cells.Intramembranous particle complexes composed of four particles('tetrads' with average diameters between 8·5 and 14·5have been observed in the protoplasmic fracture (PF) face butmost clearly and more frequently in the exoplasmic fracture(EF) face of the vacuole membrane of all red algae investigated.The tetrads lie individually within the vacuole membrane orform clusters in several species and are randomly distributed.In the species Ceramium diaphanum var. strictum and Laurenciaobtusa the intramembranous particle complexes ('tetrads') havebeen observed both in the EF and PF faces of the vacuole membrane;the 'membrane tetrads' at least as regards these two speciesseem to span both the outer and inner leaflets of the vacuolemembrane ('transmembrane particles'). The occurrence of particletetrads in the plasma membrane is probably due to exocytosiseither of the Golgi vesicles or of the mucilage sacs. Tetradfrequency in the EF face of the vacuole membranes of the investigatedred algae varies between 2 and 87 µm-2, while that ofsingle particles varies between 102 and 695 µm-2. ThePF face of the vacuole membrane is characterized by a higherparticle density than the EF face. The particle densities ofthe PF and EF faces of the plasma membrane for a given speciesare higher than those of the corresponding fracture faces ofthe vacuole membrane. Some members of Bangiophycidae bear smallerprotein particles (diameter between 8·5 and 10·5nm) in comparison with those of Florideophycidae (diameter between10·5 and 14·5 nm). It is suggested, based uponthe particle tetrads lying in depressions of the vacuole membraneand the origin of vacuoles (mucilage sacs) from ER, that theparticle tetrads originate from the ER or the Golgi complex.Since vacuoles (mucilage sacs) in red algae, along with theGolgi complex, are involved in the synthesis and export of cellsurface polysaccharides, it could be assumed that the 'membrane-tetrads'within the vacuole membrane represent a membrane-bound multienzymecomplex, participating in the synthesis of amorphous extracellularmatrix polysaccharides.Copyright 1993, 1999 Academic Press Red algae, freeze-fracture, vacuole membrane, mucilage sacs, membrane tetrads, supramolecular organization  相似文献

The ultradian rhythmic movement of the lateral leaflets of Desmodiummotorium is accompanied by rhythmic changes of the extra- andintracellular electrical potentials in the pulvinus, which aremeasured in situ in the pulvinus against the bathing solutionof the petiole. Extra- and intracellular potentials oscillatewith 180'b0 phase difference to each other, as shown by simultaneousmeasurements of both types of potentials in the abaxial partof the pulvinus. Light-induced changes of these potentials movein opposite directions. The in situ membrane potential of themotor cells of the pulvinus was calculated from the differencebetween the extra- and intracellular potentials. It was foundto oscillate between –136 and –36 mV, in phase withthe intracellular and inverse to the extracellular potential.The phase relationship between the leaflet movement rhythm andthe in situ membrane potential rhythm was as follows: downwardmovement is preceded and accompanied by a strong depolarization,upward movement by hyperpolarization. Our results suggest that membrane depolarization in pulvinarmotor cells of Desmodium motorium drives and controls potassiumefflux and hyperpolarization potassium influx via potassiumchannels. A field experiment was conducted with a water-stressed treatmentand well-watered control using eight maize (Zea mays L.) cultivars.Effects of water deficits on cell membrane stability (CMS) measuredby the polyethylene glycol (PEG) test, leaf surface wax content,and relative growth rate were investigated. Cytoplasmic lipidcontent was also analysed. Cell membrane stability and leaf surface wax content increasedwith the degrees of stress. Tolerance to drought evaluated asincrease in CMS under water deficit conditions was well differentiatedbetween cultivars and was well correlated with a reduction inrelative growth rate under stress. A negative correlation wasfound between percentage injury in the PEG test and leaf surfacewax content. High phospholipid contents were observed in tissuesof drought tolerant cultivars under water deficit conditions. The greatest activity of protopectinase obtained from the growthof Rhizoctonia solani and Fusarium solani on autoclaved potatoplugs occurred at pH 6.5, and greatest activity of the ‘lossof viscosity’ enzyme was found at 6–5 for Rhizoctonia,and between 6.5 and 8.3 for Fusarium. Protopectinase enzymeobtained from double infections of the Fusarium spp. with Rhizoctonia,or by mixing the enzymes of individual Fusarium spp. with Rhizoctoniaenzyme, were more active than the enzymes from single inoculations.Cylindrocarpon radicicola enzyme was more active when obtainedfrom a pure culture than from double infection. Similarly, mixingthis enzyme with the enzyme of Rhizoctonia reduced its activity.The evidence indicated that the protopectinase of Rhizoctoniawas similar to that of Cylindrocarpon and differed from thatof the Fusarium spp. Using paper partition chromatography, two bands from Rhizoctoniacrude enzyme had a stimulatory effect on Fusarium enzyme, whileonly one band from Fusarium enzyme stimulated Rhizoctonia enzyme. The purified enzyme of Rhizoctonia degraded pectin to galacturonicacid. Fusarium pure enzyme degraded pectin to an intermediatestage. A mixture of the two enzymes degraded pectin to galacturonicacid, without the intermediate stage formed by Fusarium alonebeing detected. The role played by pectic enzymes upon the synergistic relationof Rhizoctonia solani and Fusarium solani on rotting potatotubers is discussed.  相似文献

In a series of dawn-to-dusk studies, we examined the natureand accessibility of nectar rewards for pollinating insectsby monitoring insect visits and the secretion rate and standingcrop of nectar in the British native plant species Salvia pratensis,Stachys palustris, S. officinalis, Lythrum salicaria, Linariavulgaris, the non-native Calendula officinalis, Petunia x hybrida,Salvia splendens, and the possibly introduced Saponaria officinalis.We also compared single with double variants ofLotus corniculatus, Saponaria officinalis, Petunia x hybrida andCalendula officinalis. All the British species studied are nectar-rich and are recommendedfor pollinator-friendly gardens. They showed maximal secretionrates of about 10–90 µg sugar per flower h-1, andmost had mean standing crops of about 5–60 µg sugarper flower. In all British species studied, the corolla wasdeep enough for the relatively long-tongued bumblebee Bombuspascuorum, but the shallower flowers of Lythrum salicaria werealso much visited by shorter-tongued bees and hoverflies, aswell as by butterflies. The exotic Salvia splendens, presumablycoevolved with hummingbirds in the Neotropics, has such deepflowers that British bees cannot reach the nectar except bycrawling down the corolla. With a secretion rate approaching300 µg sugar per flower h-1and little depletion by insects,S. splendens accumulated high standing crops of nectar. S. splendens,and single and double flowers of the two probably moth-pollinatedspecies Petunia x hybrida and Saponaria officinalis, receivedfew daytime visits despite abundant nectar but Calendula waswell visited by hoverflies and bees. We compared single anddouble variants of Lotus corniculatus,Petunia x hybrida andCalendula officinalis, and also Saponaria officinalis, the lastbeing probably introduced in Britain (Stace, 1997 New floraof the British Isles. 2nd edn. Cambridge: Cambridge UniversityPress). In Petunia, Saponaria and Lotus, double flowers secretedlittle or no nectar. In Calendula, where doubling involved achange in the proportion of disc and ray florets rather thanmodification of individual flower structure, double and singlecapitula had similar standing crops of nectar. Except inCalendula, exotic or double flowers were little exploited by insect visitors.In the exotics, this was probably due to the absence or scarcityof coevolved pollinators, coupled, in double flowers, with theabsence of nectar. Impedance of renal vascular smooth muscle cells (VSMCs) cultured on microelectrodes was measured by electric cell-substrate impedance sensing. Changes in measured impedance as a function of frequency were compared with the calculated values obtained from an extended cell-electrode model to estimate the junctional resistance, distance between the ventral cell surface and the substratum, and apical and basolateral membrane capacitances of renal VSMCs. This cell-electrode model was derived to accommodate the slender and rectangular shape of VSMCs. The calculated changes in impedance (Zcal) based on the model agreed well with the experimental measurement (Zexp), and the percentage error defined as |(ZcalZexp)/Zexp| was 1.0%. To test the sensitivity of the new model for capturing changes in cell-cell and cell-substrate interactions induced by changes in cellular environment, we then applied this model to analyze timpedance changes induced by an integrin binding peptide in renal VSMCs. Our result demonstrates that integrin binding peptide decreases junctional resistance between cells, increases the distance between the basolateral cell surface and substratum, and increases the apical membrane capacitance, whereas the basolateral membrane capacitance stays relatively stable. Quantitative study of the cytochrome c acting in the photosyntheticsystem of the blue-green alga Anabaena variabilis (M-2) wasdone with membrane fragments and intact cells. Membrane fragments highly active in the NADP+-Hill reaction(above 200 µmoles/mg chl.a;-hr) retained photoresponsivecytochrome c equal only one-tenth that of P700, while the plastocyanincontent was almost equal to that of P700. The cytochrome contentin intact cells was a little larger than that in membrane fragmentson the chlorophyll a basis. However, the values relative toP700 (1/9) and plastocyanin (1/10) were identical with thosein membrane fragments. The content was also far smaller thanthat of reaction center II's (1/6). If the cytochrome mediatesall electrons from reaction center II, the cytochrome oxidation-reductionshould have a rate constant of 2.4?102 sec–1 which isone order above of the rate constant of the cytochrome reduction(2.3 to 3.5?101sec–1). These quantitative relationshipsindicate that in Anabaena variabilis (M-2), c-type cytochrome,either cytochrome f or algal cytochrome c, cannot function inthe main electron flow between two reaction centers. The chloroplast NAD(P)H dehydrogenase (NDH) complex, which reducesplastoquinones in thylakoid membranes, is involved in PSI cyclicelectron flow and chlororespiration. In addition to land plants,the NDH complex is conserved in cyanobacteria. In this study,we identified a novel NDH-related gene of Arabidopsis, NDH-dependentcyclic electron flow 5 (NDF5, At1g55370). Post-illuminationincreases in chlorophyll fluorescence were absent in ndf5 mutantplants, which indicated that NDF5 is essential for NDH activity.Sequence analysis did not reveal any known functional motifsin NDF5, but there was some homology in amino acid sequencebetween NDF5 and NDF2, a known NDH subunit. NDF5 and NDF2 homologswere present in higher plants, but not cyanobacteria. A singlehomolog, which had similarity to both NDF5 and NDF2, was identifiedin the moss Physcomitrella patens. Immunoblot analysis showedthat NDF5 localizes to membrane fractions of chloroplasts. Thestability of NdhH, a subunit of the NDH complex, as well asNDF5 and NDF2, was decreased in ndf5, ndf2 and double ndf2/ndf5mutants, resulting in a loss of NDH activity in these mutants.These results indicated that both NDF5 and NDF2 have essentialfunctions in the stabilization of the NDH complex. We proposethat NDF5 and NDF2 were acquired by land plants during evolution,and that in higher plants both NDF5 and NDF2 are critical toregulate NDH activity and each other's protein stability, aswell as the stability of additional NDH subunits.  相似文献

The hyperpolarizing response of the Chara internodal cell wasstudied by applying the voltage clamp and constant current techniques.By assuming the membrane as an electromotive force (emf) inseries with a resistance r (which is the sum of a series resistancers and the membrane resistance rm), it was shown that the hyperpolarizingresponse was brought about not only by the increase in membraneresistance but by the increase in membrane emf. The time dependentcurrent-voltage (IVm) curve obtained under the voltageclamp during the hyperpolarizing response showed a negativeresistance. The relationship between a photochemically active chromoprotein(ACP) (cf. ref. 1) and photosystem II was investigated withmembrane fragments of Anabaena cylindrica, A. variabilis andP. boryaman. ACP was solubilized from membrane fragments of A. cylindricabut not from those of A. variabilis or P. boryanum, when themembrane fragments had been incubated in a dilute buffer andhad lost their Hill or photosystem II activity. In A. cylindrica,ACP-solubilization always occurred, independent of photosystemII inactivation, on incubation of the membrane fragments inmedia without PEG. However, the amount of ACP solubilizationaccompanying photosystem II inactivation was twice that withoutphotosystem II inactivation. The increase in ACP solubilizationaccompanying photosystem II inactivation. The kinetics resembledthose for the decrease in 695 nm fluorescence emitted by membranefragments at — 196?C (cf. 2). The ACP solubilized independent of photosystem II inactivationwas assumed to have been released during disruption of intactcells in the preparation of membrane fragments. The slow ACPsolubilization upon the inactivation of photosystem II was attributedto the pigment being bound to membranes. We assume that thephoto-reactive component of ACP, P690 (cf. 3, 4), is releasedfrom the membranes during photosystem II inactivation, and thatP690 is a component of photosystem II which emits the 695 nmfluorescence at — 196?C. The maturing egg of Histiopteris incisa has many features incommon with that of Pteridium, with which the species may berelated. The nuclear evaginations are however conspicuous inrespect of the extensive development of internal membrane systems,involving dilatation of the perinuclear space. In some in stancesthe inner membrane of the nuclear envelope, the proliferationof which gives rise to the dilatations, was strikingly tenuous,possibly indicating an extensive interchange between the perinuclearspace and that part of the nucleoplasm sequestered in the evaginations.The evaginations frequently contain an aggregate of electron-opaquematerial resembling structurally and in staining propertiessimilar aggregates in Pteridium, but in contrast to Pteridiumthere was no evidence that these aggregates were organized inor near the nucleolus. Plasma membranes were isolated using the aqueous polymer two-phasepartition method from the algae Chara corallina and Chara longifolia,algae which differ in their ability to grow in saline environments.Enrichment of plasma membrane and depletion of tonoplast relativeto the microsomal fraction was monitored using phosphohydrolaseassays and crossreactions to antibodies raised against higherplant transporters. Antibodies to the vacuolar ATPase and pyrophosphatasecross-reacted with epitopes in the microsomal fraction, butshowed little affinity for the plasma membrane fraction. Pyrophosphataseactivity also declined in the plasma membrane fraction relativeto the microsomal fraction. The V-type H+ -ATPase activity,sensitive to nitrate or bafilomycin, was low in both fractions,though the cross-reaction to the antibody was reduced in theplasma membrane fraction. By contrast, the antibody recognitionof a P-type H+-ATPase amino acid sequence from Arabidopsis didnot occur strongly in the anticipated 90–100 kDa range.While there was enhanced recognition of a polypeptide at around140 kDa in the plasma membrane fraction, salt treatment of Charalongifolia resulted in plasma membrane fractions with reducedamounts of this epitope, but no change in vanadate-sensitiveATPase activity, suggesting that it does not represent the onlyP-type ATPase. Using sperm ultrastructure the systematic placement and affinitiesof the caenogastropod family Plesiotrochidae are re-examined.The simultaneous hermaphrodite, Plesiotrochus crinitus Thiele,1930, produces both euspermatozoa (uniflagellate, fertile sperm)and paraspermatozoa (bi- or triflagellate, infertile sperm).Features of each type of sperm clearly indicate that the Plesiotrochidaeare closely related to the Campanilidae (Campaniloidea) andare not, as previously believed, referable to the superfamilyCerithioidea. Significant sperm synapomorphies of Plesiotrochus(Plesiotrochidae) and Campanile (Campanilidae) include the morphologyof the eusperm midpiece (seven to nine straight mitochondriasurrounded by a segmented, accessory sheath of membrane-boundvesicles) and morphology of the anucleate parasperm head (axialcore of mitochondria surrounded by a bilaterally symmetricalarrangement of axonemes and dense vesicles). The characteristicsubstructure of the cerithioidean eusperm midpiece (four straightmitochondria each containing parallel, cristal plates) is notobserved in Plesiotrochus or Campanile. Euspermatozoa of Plesiotrochusdiffer from Campanile principally in details of the acrosomalcomplex (Plesiotrochus with apical bleb, probable absence ofan accessory membrane; Campanile without apical bleb, accessorymembrane well developed), the transverse profile of all midpiecemitochondria (thin in Plesiotrochus; thick in Campanile), andmorphology of the annulus (double ring in Plesiotrochus; singlering in Campanile). In addition, all observed paraspermatozoaof Plesiotrochus are anucleate, whereas in Campanile anucleateand nucleate paraspermatozoa are present. On the basis of spermsynapomorphies of Plesiotrochus and Campanile, the Plesiotrochidaeare transferred from the Cerithioidea to the Campaniloidea.
