The process of dissociation into variants that differ in colony morphology occurring in batch cloned cultures of two Bacillus thuringiensis strains belonging to different subspecies was studied at optimal and elevated temperatures. An increase in the cultivation temperature to 40°C resulted in an increase in the fraction of R variants to 100% after 72 h of cultivation of either of the strains. This increase was not due to the selection of forms with greater resistance to elevated temperature. The level of resistance to elevated temperature was determined by the strain genotype and did not correlate with morphological characteristics of the colonies.  相似文献

The resistance to heat, as measured by D values and phantom thermal death time curves, was observed to increase for one of three strains of Clostridium perfringens type A subsequent to animal passage. Animal passage was accomplished by the force-feeding of germ-free mice with bacterial suspensions of the organism, followed by the force-feeding of additional gnotobiotic mice with the contaminated feces. For the one strain in which an increase in heat resistance was noted, the result could not be attributed to mouse feces per se, since the presence of sterile germ-free mouse feces in a suspending medium did not protect C. perfringens spores from elevated temperature destruction.  相似文献

We have analyzed two variants of Drosophila melanogaster (RS and RE) which lead to the dual phenotype of elevated DDC activity and increased resistance to dietary alpha-methyldopa relative to Oregon-R controls. Both phenotypes show tight genetic linkage to the dopa decarboxylase, Ddc, and l(2)amd genes (i.e., less than 0.05 cM distant). We find that low (Oregon-R), medium (RS) and high (RE and Canton-S) levels of DDC activity seen at both pupariation and eclosion in these strains are completely accounted for by differences in accumulation of DDC protein as measured by immunoprecipitation. Genetic reconstruction experiments in which Ddc+ and amd+ gene doses are varied show that increasing DDC activity does not lead to a measurable increase in resistance to dietary alpha-methyldopa. This suggests that the increased resistance to dietary alpha-methyldopa is not the result of increased DDC activity but, rather, results from increased l(2)amd+ activity. Both cytogenetic and molecular analyses indicate that these overproduction variants are not the result of small duplications of the Ddc and amd genes, nor are they associated with small (greater than or equal to 100 bp) insertions or deletions. Measurements of DDC activity in wild-type strains of Drosophila reveal a unimodal distribution of activity levels with the Canton-S and RE strains at the high end of the scale, the Oregon-R control at the low end and RS near the modal value. We conclude that accumulated changes in a genetic element (or elements) in close proximity to the Ddc+ and amd+ genes lead to the coordinated changes in the expression of the Ddc and amd genes in these strains.  相似文献

Facultatively and obligately thermophilic variants were isolated from 3 out of 12 tested mesophilic Bacillus megaterium strains. The variants occurred at a frequency of 10-8–10-9. The ability to grow at elevated temperatures was cured by means of treatment with acridine orange. Stable revertants were isolated from facultatively and obligately thermophilic variants. An unknown type of megacin was produced by the facultative thermophiles. This megacin attacked mesophilic and obligately thermophilic strains. The thermophiles displayed a few divergent taxonomic characteristics but a close relationship between the strains was indicated by the megacin spectrum and sensitivity to phage. Arrhenius plots revealed that the strains could be considered as temperature range variants and that the temperature characteristic increased with growth at a higher temperature range. Two strains of Escherichia coli were subjected to heat and cold-storage treatments to determine the stability of the fecal E. coli characteristics of gas production from lactose and indole production at elevated incubation temperatures. No variants were detected with repeated sublethal heat treatment. A high incidence of variants was observed with extended cold storage of the organisms in liquid and semisolid media, especially with poor nutrient composition, and in the absence of cryoprotective agents. The indole characteristic at elevated temperature was more stable than the production of gas from lactose. The critical temperature at which both gas production from lactose and the indole characteristic were lost was 44.5 degrees C. It appeared that the variants resulted from increased temperature sensitivity of the formic hydrogen lyase and tryptophanase enzymes, respectively.  相似文献

The activity of enzymes, inactivating levomycetin and penicillin in the cells of plague and pseudotuberculosis microbes bearing extrachromosomal determinants resistant to a number of antibiotics was studied as dependent on some cultivation parameters: population age, aeration rate and temperature. It was shown that the highest capacity for levomycetin acetylation was characteristic of the cells in the late logarithmic and early stationary growth phages. Accumulation of levomycetin O-acetothers in the incubation medium markedly increased, when the cells were grown under the conditions of intensive aeration. An increase in the cultivation temperature up to 37 degrees C was accompanied by a reliable decrease in the activity of levomycetin acetylase in the transconjugant plague and pseudotuberculosis microbes though no correlation with the resistance levels in the same strains to the above antibiotics was observed. Optimal conditions for penicillinase production were determined. The maximum levels of penicillinase were found in the cells of Y. pestis 556/106 Rn with the episotic resistance type in the early exponential developmental phase under the aeration conditions and the temperature of 28 degrees C.  相似文献

The dynamics and structure of the epidemic process of Salmonella infections among the population of Perm in 1983-1988 was studied and the results of evaluation of antibiotic resistance of the dominating Salmonella species analyzed. The study revealed that a decrease in salmonellosis morbidity caused by S. typhimurium was associated with a limited circulation of anthroponotic (antibiotic-resistant) variants of Salmonellae and a relative increase in the proportion of zoonotic (antibiotic-sensitive) strains. At the period of elevated morbidity this Salmonella infection affected mainly young children in cold months, whereas in recent years seasonal morbidity rises shifted to spring-summer and summer-autumn months, affecting older age groups of the population. The study also revealed that a rise in salmonellosis morbidity caused by S. enteritidis was due to increased circulation of zoonotic variants of Salmonellae. Changes in the epidemiological situation necessitate correction of the system of epidemiological surveillance on Salmonella infections with the emphasis on sanitation measures in stock-breeding farms with unfavorable epidemiological situation.  相似文献

The routinely used antibiotics and antiseptics were compared with the same staphylococcal isolates by MIC ranges, the mean MIC for the strains, proportion of the variants resistant to the drugs, distribution of the strains by the resistance spectra and the number increase rate for the resistant variants in natural populations within 5-10-year periods. It was concluded that when used locally the antiseptics had some advantages over the antibiotics especially with respect to hospital strains. The authors believe that in developing new drugs, ++re-estimation of the routinely used ones, choosing the optimal drug for a particular case it is more important to consider the heterogeneity of microbial populations, the frequency of resistant variants and activity of the mechanisms of their selection under hospital conditions.  相似文献

The ability of temperature variants of influenza A2 virus to produce plaques in chick embryo kidney tissue culture was studied at different temperatures. Definite differences in efficiency of plaque formation by cryophilic and thermophilic strains were observed at low and high temperatures. Differentiation of the temperature variants appears to reside in a number of genetic markers, designated rct-40, rct-28, and plaque size (S). Virulence of influenza A2 virus, enhanced after prolonged cultivation at high temperature, is probably related to its greater efficiency of plating at 40 C (rct-40(+)), formation of larger plaques at optimal (36 C) and high (40 C) temperatures, and to loss of ability to form plaques at 28 C (rct-28(-)).  相似文献

Resistance of 669 clinical strains of Proteus mirabilis to 18 chemotherapeutic drugs was studied in dynamics within 1970-1985. An increase in the number of cultures resistant to ampicillin and carbenicillin was noted while the number of cultures resistant to cephalosporines did not change. Within the period from 1970 to 1975 there was observed a marked increase in the number of Proteus strains resistant to aminoglycoside antibiotics. After that period their number gradually lowered and in 1985 reached the level of 1970. Beginning from 1973 there were observed a decrease in the number of Proteus chloramphenicol resistant strains and simultaneous occurrence of cultures sensitive to this antibiotic. The predominating number of the tested strains preserved during the whole observation period their resistance to tetracycline, doxycycline, rifampicin, novobiocin, furazolidone and furagin. No increase in the number of Proteus strains with multiple drug resistance including those resistant to 5-7 drugs was noted in the observation periods of 1970-1975, 1980 and 1985. The most frequent were Proteus strains resistant to 2-4 drugs. Among them cultures resistant to chloramphenicol and aminoglycoside antibiotics of the first generation predominated. Grouping of the strains by the same resistance spectra provided dividing the rested cultures of Proteus mirabilis into 69 variants.  相似文献

Rough and smooth variants of Bacillus stearothermophilus strains ATCC 12976 and 12980 were isolated. These variants showed morphologically different colonies. In both strains the rough variants were less heat-resistant than the smooth ones, and their activation occurred in a shorter time; on the other hand, their dipicolinic acid (DPA) content was higher. These results indicate that a relationship between higher DPA content and higher thermal resistance does not exist. Therefore, this evidence supports the hypothesis that DPA is not the determining factor in the degree of spore heat resistance but that it could, instead, have a role in maintaining thermal resistance obtained by other means.  相似文献

Mammalian cells of different origin (11 strains) were cultivated at the temperature of 25--41 degrees to measure the temperature limits of mitosis. Different strains of the cells reacted to the increase or decrease in the cultivation temperature in a dissimilar way. The difference between the upper temperature limit and the optimum one was not over 5 degrees. Cell division did not end with the temperature fall by over 10 degrees. Various cell strains responded to the temperature decrease in a different way. Most cellular population had three cell types. The majority of the cells were capable of dividing at the threshold temperature; some cells could enter mitosis without completing it and stop at the metaphase. The temperature limits of mitosis were not related to the species and tissue origin of the cells.  相似文献

A technique was developed which facilitates the application of elevated temperatures to strains of unknown heat sensitivity during attempts to eliminate antibiotic resistance. Cultures were subjected to a temperature gradient and the lowest temperatures which inhibited growth were found to be highly efficient in the elimination of gentamicin resistance in several strains of Staphylococcus aureus and S. epidermidis.  相似文献

The authors studied the behaviour of 11 mumps virus strains or variants including the thermolabile standard Jeryl Lynn strain under thermal charge (50 degrees C/30 min). Varants were obtained from the Soviet vaccinal strains Leningrad-3 by cultivation under various conditions. Incubation temperature and cellular substrate played an important role therein. Variants with various behaviour in the marker T50 resulted. It was found that passages at 32 degrees C at limited dilutions as well as those on chick embryos or in cultures of chicken fibroblasts increased their thermolability. Imipenem is often used to treat intensive care unit patients infected by Enterobacter aerogenes, but it is leading to an increasing number of antibiotic resistant strains. Clinical isolates and imipenem resistant variants presented a high level of resistance to beta-lactam antibiotic group and to chemically unrelated drugs. We report here that imipenem selects strains which contain active efflux pumps ejecting various unrelated antibiotics including quinolones, tetracycline, and chloramphenicol. An increase of AcrA, an efflux pump component, was observed in the imipenem resistant variants. The overexpression of marA, involved in the genetic control of membrane permeability via porin and efflux pump expression, indicated the activation of the resistance genetic cascade in imipenem resistant variants.  相似文献

The ability of purple photosynthetic bacteria to form phase variants (dissociants) was discovered. The R and M phase variants were isolated from the Rhodobacter sphaeroides population, and their affiliation with the parent strain was confirmed by the PCR of their 16S rRNA gene fragments. The R and M variants were different both in colony morphology and in resistance to some physical and chemical factors. The R variant of Rhb. sphaeroides had advantages when grown in the light, at elevated temperature, under aeration, and UV irradiation, whereas the M variant had advantages when grown aerobically in the dark or at increased NaCl concentrations.  相似文献

Beet necrotic yellow vein virus (BNYVV), vectored by Polymyxa betae, causes rhizomania in sugar beet. For disease control, the cultivation of hybrids carrying Rz1 resistance is crucial, but is compromised by resistance-breaking (RB) strains with specific mutations in the P25 protein at amino acids 67–70 (tetrad). To obtain evidence for P25 variability from soil-borne populations, where the virus persists for decades, populations with wild-type (WT) and RB properties were analysed by P25 deep sequencing. The level of P25 variation in the populations analysed did not correlate with RB properties. Remarkably, one WT population contained P25 with RB mutations at a frequency of 11%. To demonstrate selection by Rz1 and the influence of RB mutations on relative fitness, competition experiments between strains were performed. Following a mixture of strains with four RNAs, a shift in tetrad variants was observed, suggesting that strains did not mix or transreplicate. The plant genotype exerted a clear influence on the frequency of RB tetrads. In Rz1 plants, the RB variants outcompeted the WT variants, and mostly vice versa in susceptible plants, demonstrating a relative fitness penalty of RB mutations. The strong genotype effect supports the hypothesized Rz1 RB strain selection with four RNAs, suggesting that a certain tetrad needs to become dominant in a population to influence its properties. Tetrad selection was not observed when an RB strain, with an additional P26 protein encoded by a fifth RNA, competed with a WT strain, supporting its role as a second BNYVV pathogenicity factor and suggesting the reassortment of both types.  相似文献

Comparative characteristics of sporogenous and asporogenous Bacillus thuringiensis strains is carried out. Asporogenous strains are found to differ from wild type strains in a number of criteria, including colony morphology, character of growth on rich and poor media and UV-sensitivity. Sporogenous strains form R colonies, they are more stable and more rare produce variants forming S colonies. S colonies are typical for asporogenous mutants, and under the cultivation in unfavourable conditions (elevated temperature, a shift of pH, a change of an incubation regime) asporogenous strains dissociate with a high frequency into R form. Initial strains, which are multiple auxotrophs, under certain conditions can form "prototrophic" revertants which are unstable when incubated on rich media. Suppressor mutation is supposed to be a possible mechanism of the origination of "prototrophs".  相似文献

A comparative study was conducted on the adaptive mechanisms of the strains Arthrobacter oxydans K14 and Acinetobacter lwoffii EK30A isolated from permafrost subsoil sediments and of those of the analogous collection strains (Ac-1114 Type and BSW-27, respectively). In each pair of the strains compared, the strains differed in terms of (i) growth-related, physiological, and biochemical properties; (ii) resistance to stress factors; (iii) capacity for generation of dormant forms (DFs) under growth arrest conditions, and (iv) intrapopulation production of phase variants. The strains isolated from permafrost displayed a lower growth rate but were more resistant to repeated freezing-thawing treatment than the collection strains. Under the same growth conditions, the permafrost strains formed larger numbers of cystlike anabiotic DFs, extraordinarily small cells, and forms that became nonculturable during long-term storage. Resuscitation of the nonculturable forms resulted in a 2- to-7-fold increase in the percentage of FISH-detectable metabolically active cells. The permafrost strains were also distinguished by increased genome lability. This facilitated their dissociation into intrapopulation variants with phenotypically distinct colonial and morphological properties and different antibiotic resistance. The phenotypic variability was more prominent in Arthrobacter (for which it was not reported previously) than in Acinetobacter. In the populations produced by plating the dormant bacterial forms, the qualitative and quantitative characteristics of the phase variant spectra varied depending on the formation conditions and the composition of the solid media used for the plating. Thus, the permafrost isolates of A. oxydans and Ac. lwoffii were distinguished from their collection analogs by a more manifest adaptive potential including stress resistance, the intensity of DF generation under growth arrest conditions, and increased intrapopulation variability.  相似文献
