The distribution of purines and pyrimidines in desoxypentose nucleic acids prepared from a variety of animal and plant sources has been studied. 1. The nucleic acids were prepared from calf thymus, calf kidney, sheep spleen, horse spleen, chicken erythrocyte, turtle erythrocyte, trout sperm, shad testes, sea urchin sperm, wheat germ, and Pneumococcus Type III. 2. Separate hydrolyses were carried out for the determination of purines and pyrimidines. These procedures permitted nearly quantitative recovery of nucleic acid phosphorus in many of the preparations examined. 3. In the case of those preparations where a quantitative recovery was obtained it can be concluded that no bases other than adenine, guanine, thymine, and cytosine were present in appreciable amounts. 4. The distribution of purines and pyrimidines in all the nucleic acids studied renders the tetranucleotide hypothesis untenable. 5. The results of the analyses have indicated no great differences in the composition of these nucleic acids with respect to purines and pyrimidines.  相似文献

Due to its sensitivity to chirality, Raman optical activity (ROA), which may be measured as a small difference in vibrational Raman scattering from chiral molecules in right- and left-circularly polarized incident light, is a powerful probe of biomolecular structure in solution. Protein ROA spectra provide information on the secondary and tertiary structures of the polypeptide backbone, hydration, side-chain conformation, and structural elements present in denatured states. Nucleic acid ROA spectra yield information on the sugar ring conformation, the base stacking arrangement, and the mutual orientation of the sugar and base rings around the C-N glycosidic linkage. ROA is able to simultaneously probe the structures of both the protein and the nucleic acid components of intact viruses. This article gives a brief account of the theory and measurement of ROA and presents the ROA spectra of a selection of proteins, nucleic acids, and viruses which illustrate the applications of ROA spectroscopy in biomolecular research.  相似文献

The isoelectric characteristics of some nucleic acid preparations from rat liver have been examined. 10S and 4S RNA species and SV-DNA were found to have isoelectric points of 5.2, 6.0-6.7, and 4.35 respectively. The molecular charge ratios (net negative charge/nucleotide) were calculated. Using SV-DNA as a standard, these isoelectric characteristics and charge ratios have been interpreted as indicating that the 10S and 4S RNAs have 35 and 56% of the molecules involved in secondary structure.  相似文献

1. Nucleic acids of whole Drosophila adults were prepared in good yield and substantially free from impurities by new modifications of the phenol method. 2. The average molar base compositions of the DNA (41% of guanine+cytosine) and transfer RNA (60% of guanine+cytosine) resemble those of mammalian nucleic acids; the ribosomal RNA has a DNA-like molar base composition (43% of guanine+cytosine), and it is considered that this is reflected in the lower stability of its secondary structure compared with mammalian ribosomal RNA. 3. The two main ribosomal forms were separated and average base compositions and sedimentation values determined.  相似文献
