Calf brain tubulin purified by means of ammonium sulfate fractionation, ion-exchange chromatography, and MgCl2 precipitation contains a low level of Mg2+ -dependent GTPase activity, the protein preparation being essentially homogeneous according to conventional procedures. Tubulin was freed from this possibly contaminant enzyme activity by Sephacryl S300 gel chromatography. Soluble tubulin itself showed a ligand-induced Mg2+-dependent GTPase activity in the presence of colchicine, but not of tropolone methyl ether, podophyllotoxin, or vinblastine. Tubulin also hydrolyzed GTP when assembling into microtubules. This reaction proceeded in a nonlinear fashion and was suppressed together with microtubule assembly by lowering the protein concentration under the critical concentration, adequately modifying assembly buffer conditions, or using Ca2+, tropolone methyl ether, podophyllotoxin, or vinblastine. The number of molecules of GTP hydrolyzed per molecule of tubulin polymerized was estimated to vary between 0.9 and 2.1, depending on whether morpholineethanesulfonate or phosphate assembly buffers were employed.  相似文献

Free ribulose bisphosphate (RuBP4?) rather than its magnesium complex (RuBP-Mg2?) was the apparent substrate for spinach ribulose bisphosphate carboxylase/oxygenase. The apparent Km for total RuBP (pH 8.0 at 30° C) increased with increasing Mg2+ concentrations from 11.6 μM at 13.33 mM Mg2+ to 32.6 μM at 40.33 mM Mg2+. Similarly the apparent Km for RuBP-Mg2? complex increased with increasing Mg2+ from 9.4 μM at 13.33 mM Mg2+ to 29.7 μM at 40.33 mM Mg2+. However, the Km values for uncomplexed RuBP4? were independent of the (saturating) concentration of Mg2+ (Km=2.2 μM). The Vmax did not vary with the changing concentrations of Mg2+. In contrast, the Km for total RuBP remained constant with varying Mg2+ concentrations (Km=59.5 μM) for the enzyme from R. rubrum. The apparent Km for the RuBP-Mg2? complex decreased with increasing Mg2+ concentrations from 16.0 μM at 7.5 mM Mg2+ to 5.9 μM at 27.5 mM Mg2+. The initial velocity for the C. vinosum enzyme was also found to be independent of the (saturating) concentration of Mg2+ when total RuBP was varied in the assay. Thus the response to total RuBP by these two bacterial enzymes, which markedly differ in structure, was closely similar.  相似文献

Complexation in the H+-Si(OH)4-tropolone (HL) system was studied in 0.6 M (Na)Cl medium at 25° C. Speciation and formation constants were determined from potentiometric (glass electrode) and 29Si-NMR data. Experimental data cover the ranges 1.5 ? - log[H+] ? 8.4, 0.002 ? B ? 0.012 M, and 0 ? C ? 0.060 M (B and C stand for the total concentration of Si and tropolone, respectively). In acid solutions (-log[H+] ? 3) a hexacoordinated cationic complex, SiL3+, is formed with log K(Si(OH)4 + 3HL + H+ XXX SiL3+ + 4H2O) = 7.08 ± 0.03. Furthermore, the formation of a disilicic acid was established from 29Si-NMR data. The dimerization constant of Si(OH)4 was found to be 10 exp (1.2 ± 0.1). In model calculations the solubility of quartz and amorphous SiO2 in the presence of tropolone is demonstrated. Data were analyzed using the least-squares computer program LETAGROPVRID.  相似文献

The (Na++K+)-activated, Mg2+-dependent ATPase from rabbit kidney outer medulla was prepared in a partially inactivated, soluble from depleted of endogenous phospholipids, using deoxycholate. This preparation was reactivated 10 to 50-fold by sonicated liposomes of phosphatidylserine, but not by non-sonicated phosphatidylserine liposomes or sonicated phosphatidylcholine liposomes. The reconstituted enzyme resembled native membrane preparations of (Na++K+)-ATPase in its pH optimum being around 7.0 showing optimal activity at Mg2+: ATP mol ratios of approximately 1 and a Km value for ATP of 0.4 mM.Arrhenius plots of this reactivated activity at a constant pH of 7.0 and an Mg2+: ATP mol ratio of 1:1 showed a discontinuity (sharp change of slope) at 17 °C, With activation energy (Ea) values of 13–15 kcal/mol above this temperature and 30–35 kcal below it. A further discontinuity was also found at 8.0 °C and the Ea below this was very high (> 100 kcal/mol).Incresed Mg2+ concentrations at Mg2+: ATP ratios in excess of 1:1 inhibited the (Na++K+)-ATPase activity and also abolished the discontinuities in the Arrhenius plots.The addition of cholesterol to phosphatidylserine at a 1:1 mol ratio partially inhibited (Na++K+)-ATPase reactivation. Arrhenius plots under these conditions showed a single discontinuity at 20°C and Ea values of 22 and 68kcal/mol above and below this temperature respectively. The ouabain-insensitive Mg2+-ATPase normally showed a linear Arrhenius plot with an Ea of 8 kcal/mol. The cholesterol-phosphatidylserine mixed liposomes stimulated the Mg2+-ATPase activity, which now also showed a discontinuity at 20 °C with, however, an increased value of 14 kcal/mol above this temperature and 6 kcal/mol below. Kinetic studies showed that cholesterol had no significant effect on the Km for ATP.Since both of cholesterol and Mg2+ are know to alter the effects of temperature on the fluidity of phospholipids the above result are discussed in this context.  相似文献

(1) Contrary to what has usually been assumed, (Na+ + K+)-ATPase slowly hydrolyses AdoPP[NH]P in the presence of Na+ + Mg2+ to ADP-NH2 and Pi. The activity is ouabain-sensitive and is not detected in the absence of either Mg2+ or Na2+. The specific activity of the Na+ + Mg2+ dependent AdoPP[NH]P hydrolysis at 37°C and pH 7.0 is 4% of that for ATP under identical conditions and only 0.07% of that for ATP in the presence of K+. The activity is not stimulated by K+, nor can K+ replace Na+ in its stimulatory action. This suggests that phosphorylation is rate-limiting. Stimulation by Na+ is positively cooperative with a Hill coefficient of 2.4; half-maximal stimulation occurs at 5–9 mM. The Km value for AdoPP[NH]P is 17 μM. At 0°C and 21°C the specific activity is 2 and 14%, respectively, of that at 37°C. AMP, ADP and AdoPP[CH2]P are not detectably hydrolysed by (Na+ + K+)-ATPase in the presence of Na+ + Mg2+. (2) In addition, AdoPP[NH]P undergoes spontaneous, non-enzymatic hydrolysis at pH 7.0 with rate constants at 0, 21 and 37°C of 0.0006, 0.006 and 0.07 h?1, respectively. This effect is small compared to the effect of enzymatic hydrolysis under comparable conditions. Mg2+ present in excess of AdoPP[NH]P reduces the rate constant of the spontaneous hydrolysis to 0.005 h?1 at 37°C, indicating that the MgAdoPP[NH]P complex is virtually stable to spontaneous hydrolysis, as is also the case for its enzymatic hydrolysis. (3) A practical consequence of these findings is that AdoPP[NH]P binding studies in the presence of Na+ + Mg2+ with enzyme concentrations in the mg/ml range are not possible at temperatures above 0°C. On the other hand, determination of affinity in the (Na+ + K+)-ATPase reaction by competition with ATP at low protein concentrations (μg/ml range) remains possible without significant hydrolysis of AdoPP[NH]P even at 37°C.  相似文献

Cytosolic free magnesium (Mgf) is considered relatively constant. To test this concept, Mgf was estimated during hyperkalemic ventricular akinesis, normal and maximum adrenergic stimulation, and sulfate loading of the normoxic perfused guinea-pig heart. The Mgf estimates utilized a new sliding scale derived from the Mg2+-dependence of glyceraldehyde-3-phosphate dehydrogenase/phosphoglycerate kinase (GAPDH/PGK). The pseudo constant KGAPDH′·KPGK′ was measured as ([creatine phosphate][3-phosphoglycerate][lactate]KLDH/([creatine][Pi[glyceraldehyde 3-phosphate][pyruvate]KCK), which varied with magnesium due to KCK (CK, LDH = creatine kinase, lactate dehydrogenase). However, the correct magnesium dependencies of the true constants KGAPDH·KPGK and KCK were taken from the literature. The [Mg2+] at which pseudo KGAPDH′·KPGK′ equalled true KGAPDH·KPGK was the best estimate of Mgf. Mgf fell to ≈0.13 mM in hyperkalemic arrest from a control of ≈0.6 mM, rising to ≈0.85 mM only during maximum adrenergic stress. Mgf increased further to ≈1.3 mM during sulfate loading which induced ATP catabolism. Mgf and ATP were reciprocally related. Thus; (1) myocardial free [Mg2+] judged from GAPDH/PGK mass-action relations changed appreciably only under extreme physiological states; (2) ATP was a major chelator of Mg2+ in perfused myocardium, i.e., acute ATP pool size reduction may be associated with increments in Mgf.  相似文献

(1) The effects of calmodulin binding on the rates of Ca2+-dependent phosphorylation and dephosphorylation of the red-cell Ca2+ pump, have been tested in membranes stripped of endogenous calmodulin or recombined with purified calmodulin. (2) In Mg2+-containing media, phosphorylation and dephosphorylation rates are accelerated by a large factor (at 0°C), but the steady-state level of phosphoenzyme is unaffected by calmodulin binding (at 0°C and 37°C). In Mg2+-free media, slower rates of phosphoenzyme formation and hydrolysis are observed, but both rates and the steady-state phosphoenzyme level are raised following calmodulin binding. (3) At 37°C and 0°C, the rate of (Ca2+ + Mg2+)-ATPase activity is stimulated maximally by 6–7-fold, following calmodulin binding. At 37°C the apparent Ca2+ affinity for sustaining ATP hydrolysis is raised at least 20-fold, Km(Ca) ? 10 μM (—calmodulin) and Km(Ca) < 0.5 μM (+ calmodulin), but at 0°C the apparent Ca2+ affinity is very high in calmodulin-stripped membranes and little or no effect of calmodulin is observed (Km(Ca) ? 3–4 · 10-8 M). (Ca2+ + Mg2+)-ATPase activity in calmodulin activated membranes and at saturating ATP levels, is sharply inhibited by addition of calcium in the range 50–2000 μM. (4) A systematic study of the effects of the nucleotide species MgATP, CaATP and free ATP on (Ca2+ + Mg2+)-ATPase activity in calmodulin-activated membranes reveals: (a) In the 1–10 μmolar concentration range MgATP, CaATP and free ATP appear to sustain (Ca2+ + Mg2+)-ATPase activity equally effectively. (b) In the range 100–2000 μM, MgATP accelerates ATP hydrolysis (Km(MgATP) ? 360 μM), and CaATP is an inhibitor (Ki(CaATP) ? 165 μM), probably competing with MgATP fo the regulatory site. (5) The results suggest that calmodulin binding alters the conformational state of the Ca2+- pump active site, producing a high (Ca2+ + Mg2+)-ATPase activity, high Ca2+ affinity and regulation of activity by MgATP.  相似文献

The aim of this study was to determine from macroscopic current analysis how intracellular magnesium ions, Mg i 2+ , interfere with sodium channels of mammalian neurones. It is reported here that permeation across the sodium channel is voltage- and concentration-dependently reduced by Mg i 2+ . This results in a general reduction of sodium membrane conductance and an outward sodium peak current at large positive potentials. 30 mM Mg i 2+ leads to a negative shift of voltage dependence of sodium channel gating parameters, probably due to the surface potential change of the membrane. This shift alone is, however, insufficient to explain the reduction of outward sodium currents. The blockage by Mg i 2+ is decreased upon increasing intracellular or extracellular Na+ concentration, which suggests that Mg?' interferes with sodium permeation by competitively occupying sodium channels. Using a kinetic model to describe the sodium permeation, the dissociation constant (at zero membrane potential) of Mg i 2+ for the sodium channel has been calculated to be 8.65 ± 1.51 mM, with its binding site located at 0.26 ± 0.05 electrical distance from the inner membrane. This dissociation constant is smaller than that of Na i +, which is 83.76 ± 7.60 mM with its binding site located at 0.75 ± 0.23. The low dissociation constant of Mg i 2+ reflects its high affinity for the sodium channel.  相似文献

Although different routes for the S-nitrosation of cysteinyl residues have been proposed, the main in vivo pathway is unknown. We recently demonstrated that direct (as opposed to autoxidation-mediated) aerobic nitrosation of glutathione is surprisingly efficient, especially in the presence of Mg2+. In the present study we investigated this reaction in greater detail. From the rates of NO decay and the yields of nitrosoglutathione (GSNO) we estimated values for the apparent rate constants of 8.9±0.4 and 0.55±0.06 M−1 s−1 in the presence and absence of Mg2+. The maximum yield of GSNO was close to 100% in the presence of Mg2+ but only about half as high in its absence. From this observation we conclude that, in the absence of Mg2+, nitrosation starts by formation of a complex between NO and O2, which then reacts with the thiol. Omission of superoxide dismutase (SOD) reduced by half the GSNO yield in the absence of Mg2+, demonstrating O2 formation. The reaction in the presence of Mg2+ seems to involve formation of a Mg2+•glutathione (GSH) complex. SOD did not affect Mg2+-stimulated nitrosation, suggesting that no O2 is formed in that reaction. Replacing GSH with other thiols revealed that reaction rates increased with the pKa of the thiol, suggesting that the nucleophilicity of the thiol is crucial for the reaction, but that the thiol need not be deprotonated. We propose that in cells Mg2+-stimulated NO/O2-induced nitrosothiol formation may be a physiologically relevant reaction.  相似文献

Myosin reacted with N-ethylmaleimide in the presence of ADP lost its ability to be activated by actin. Subfragment 1 behaved similarly. About 2 moles of N-ethylmaleimide per mole of Subfragment 1 were required to eliminate actin activation of the Mg2+-ATPase activity. At the point at which actin activation was lost the K+-EDTA-ATPase activity was also lost, but the Ca2+-activated ATPase activity was increased. Kinetic measurements indicated that the labelling with N-ethylmaleimide in the presence of ADP reduced V (the ATPase activity at infinite actin concentration) but did not effect Kapp (which is related to the dissociation constant of the actin-Subfragment 1 complex). The Mg2+-activated activity of the reacted myosin alone remained unaltered and the ability to bind actin was retained. We propose that the N-ethylmaleimide labelling blocked the actin activation by preventing the accelerated release of hydrolysis products from the myosin.  相似文献

The observed equilibrium constants (Kobs) for the reactions of d-2-phosphoglycerate phosphatase, d-2-Phosphoglycerate3? + H2O → d-glycerate? + HPO42?; d-glycerate dehydrogenase (EC 1.1.1.29), d-Glycerate? + NAD+ → NADH + hydroxypyruvate? + H+; and l-serine:pyruvate aminotransferase (EC 2.6.1.51), Hydroxypyruvate? + l-H · alanine± → pyruvate? + l-H · serine±; have been determined, directly and indirectly, at 38 °C and under conditions of physiological ionic strength (0.25 m) and physiological ranges of pH and magnesium concentrations. From these observed constants and the acid dissociation and metal-binding constants of the substrates, an ionic equilibrium constant (K) also has been calculated for each reaction. The value of K for the d-2-phosphoglycerate phosphatase reaction is 4.00 × 103m [ΔG0 = ?21.4 kJ/mol (?5.12 kcal/mol)]([H20] = 1). Values of Kobs for this reaction at 38 °C, [K+] = 0.2 m, I = 0.25 M, and pH 7.0 include 3.39 × 103m (free [Mg2+] = 0), 3.23 × 103m (free [Mg2+] = 10?3m), and 2.32 × 103m (free [Mg2+] = 10?2m). The value of K for the d-glycerate dehydrogenase reaction has been determined to be 4.36 ± 0.13 × 10?13m (38 °C, I = 0.25 M) [ΔG0 = 73.6 kJ/mol (17.6 kcal/mol)]. This constant is relatively insensitive to free magnesium concentrations but is affected by changes in temperature [ΔH0 = 46.9 kJ/mol (11.2 kcal/mol)]. The value of K for the serine:pyruvate aminotransferase reaction is 5.41 ± 0.11 [ΔG0 = ?4.37 kJ/mol (?1.04 kcal/mol)] at 38 °C (I = 0.25 M) and shows a small temperature effect [ΔH0 = 16.3 kJ/ mol (3.9 kcal/mol)]. The constant showed no significant effect of ionic strength (0.06–1.0 m) and a response to the hydrogen ion concentration only above pH 8.5. The value of Kobs is 5.50 ± 0.11 at pH 7.0 (38 °C, [K+] = 0.2 m, [Mg2+] = 0, I = 0.25 M). The results have also allowed the value of K for the d-glycerate kinase reaction (EC 2.7.1.31), d-Glycerate? + ATP4? → d-2-phosphoglycerate3? + ADP3? + H+, to be calculated to be 32.5 m (38 °C, I = 0.25 M). Values for Kobs for this reaction under these conditions and at pH 7.0 include 236 (free [Mg2+] = 0) and 50.8 (free [Mg2+] = 10?3m).  相似文献

We aimed to study kinetics of modulation by intracellular Mg2+ of cardiac gap junction (Mg2+ gate). Paired myocytes of guinea-pig ventricle were superfused with solutions containing various concentrations of Mg2+. In order to rapidly apply Mg2+ to one aspect of the gap junction, the non-junctional membrane of one of the pair was perforated at nearly the connecting site by pulses of nitrogen laser beam. The gap junction conductance (Gj) was measured by clamping the membrane potential of the other cell using two-electrode voltage clamp method. The laser perforation immediately increased Gj, followed by slow Gj change with time constant of 3.5 s at 10 mM Mg2+. Mg2+ more than 1.0 mM attenuated dose-dependently the gap junction conductance and lower Mg2+ (0.6 mM) increased Gj with a Hill coefficient of 3.4 and a half-maximum effective concentration of 0.6 mM. The time course of Gj changes was fitted by single exponential function, and the relationship between the reciprocal of time constant and Mg2+ concentration was almost linear. Based on the experimental data, a mathematical model of Mg2+ gate with one open state and three closed states well reproduced experimental results. One-dimensional cable model of thirty ventricular myocytes connected to the Mg2+ gate model suggested a pivotal role of the Mg2+ gate of gap junction under pathological conditions.  相似文献

The inhibition of the polymerization of tubulin from cultured cells of rose (Rosa. sp. cv. Paul's scarlet) by colchicine and the binding of colchicine to tubulin were examined in vitro and compared with data obtained in parallel experiments with bovine brain tubulin. Turbidimetric measurements of taxol-induced polymerization of rose microtubules were found to be sensitive and semiquantitative at low tubulin concentrations, and to conform to some of the characteristics of a nucleation and condensation-polymerization mechanism for assembly of filamentous helical polymers. Colchicine inhibited the rapid phase of polymerization at 24°C with an apparent inhibition constant (K i) of 1.4·10-4 M for rose tubulin and an apparent K i=8.8·10-7 M for brain tubulin. The binding of [3H]colchicine to rose tubulin to form tubulin-colchicine complex was mildly temperature-dependent and slow, taking 2–3 h to reach equilibrium at 24°C, and was not affected by vinblastine sulfate. The binding of [3H]colchicine to rose tubulin was saturable and Scatchard analysis indicated a single class of low-affinity binding sites having an apparent affinity constant (K) of 9.7·102 M-1 and an estimated molar binding stoichiometry (r) of 0.47 at 24°C. The values for brain tubulin were K=2.46·106 M-1 and r=0.45 at 37°C. The binding of [3H]colchicine to rose tubulin was inhibited by excess unlabeled colchicine, but not by podophyllotoxin or tropolone. The data demonstrate divergence of the colchicine-binding sites on plant and animal tubulins and indicate that the relative resistance of plant microtubule polymerization to colchicine results from a low-affinity interaction of colchicine and tubulin.Abbreviations MT microtubule - TC tubulin-colchicine complex  相似文献

The effect of the hydrolysis product Pi and the artificial substrate p-nitrophenyl phosphate (p-nitrophenyl-P) on ouabain binding to (Na+ + K+)-activated ATPase was investigated.The hypothesis that (Mg2+ + p-nitrophenyl-P)-supported ouabain binding might be due to Pi release and thus (Mg2+ + Pi)-supported could not be confirmed.The enzyme · ouabain complexes obtained with different substrates were characterized according to their dissociation rates after removal of the ligands facilitating binding. The character of the enzyme · ouabain complex is determined primarily by the monovalent ion present during ouabain binding, but, qualitatively at least, it is immaterial whether binding was obtained with p-nitrophenyl phosphate or Pi.The presence or absence of Na+ during binding has a special influence upon the character of the enzyme · ouabain complex. Without Na+ and in the presence of Tris ions the complex obtained with (Mg2+ + Pi) and that obtained with (Mg2+ + p-nitrophenyl-P) behaved in a nearly identical manner, both exhibiting a slow decay. High Na+ concentration diminished the level of Pi-supported ouabain binding, having almost no effect on p-nitrophenyl phosphate-supported binding. Both enzyme · ouabain complexes, however, now resembled the form obtained with (Na+ + ATP), as judged from their dissociation rates and the K+ sensitivity of their decay. The complexes obtained at a high Na+ concentration underwent a very fast decay which could be slowed considerably after adding a low concentration of K+ to the resuspension medium. The most stable enzyme · ouabain complex was obtained in the presence of Tris ions only, irrespective of whether p-nitrophenyl phosphate or Pi facilitated complex formation. The presence of K+ gave rise to a complex whose dissociation rate was intermediate between those of the complexes obtained in the presence of Tris and a high Na+ concentration.It is proposed that the different ouabain dissociation rates reflect different reactive state of the enzyme. The resemblance between the observations obtained in phosphorylation and ouabain binding experiments is pointed out.  相似文献

Regulation of ADP-Glucose Pyrophosphorylase from Chlorella vulgaris   总被引:1,自引:1,他引:0  
ADP-glucose pyrophosphorylase was partially purified from Chlorella vulgaris 11h. 3-Phosphoglycerate activated the enzyme by lowering the Michaelis constant for glucose-1-phosphate (from 0.97 to 0.36 millimolar in the presence of 2 millimolar phosphoglycerate) and ATP (from 0.23 to 0.10 millimolar), as well as increasing the Vmax. Saturation curves for 3-phosphoglycerate were hyperbolic and the activator concentration at half Vmax value for 3-phosphoglycerate was 0.41 millimolar either in the presence or absence of phosphate. Phosphate inhibited the enzyme in a competitive manner with respect to glucose-1-phosphate, but did not affect the Michaelis constant value for ATP. 3-Phosphoglycerate changed neither the inhibitor concentration at half Vmax value of 1.0 millimolar for phosphate nor the hyperbolic inhibition kinetics for phosphate. The enzyme required divalent cations for its activity. The activation curves for Mn2+ and Mg2+ were highly sigmoidal. The activator concentration at half Vmax values for Mn2+ and Mg2+ were 2.8 and 3.7 millimolar, respectively. With optimal cations, the Michaelis constant values for ATP-Mn and ATP-Mg were 0.1 and 0.4 millimolar, respectively.  相似文献

The observed equilibrium constant Kobs for the hydrolysis of ATP to ADP and inorganic phosphate has been calculated as a function of pH and metal ion concentration pM (- log [M]) at 25 °C and μ = 0.2 with the use of literature values of the acid dissociation and complex dissociation constants for the phosphates.The resulting standard free energy changes ΔG °′ are presented by means of contour diagrams for the range pH 4–10 and pM 1–7. These maps summarize the results of some 1900 calculations per diagram, and clearly simulate a differential effect of the metal ions of interest, including Mg2+, Ca2+, Sr2+, Mn2+, Li+, Na+ and K+, on the equilibrium hydrolysis of ATP.  相似文献

A procedure was developed to isolate a membrane fraction of rat skeletal muscle which contains a highly active Mg2+-ATPase (5–25 μmol Pi/mg min). The rate of ATP hydrolysis by the Mg2+-ATPase was nonlinear but decayed exponentially (first-order rate constant ≥0.2 s?1 at 37°C). The rapid decline in the ATPase activity depended on the presence of ATP or its nonhydrolyzable analog 5′-adenylyl imidodiphosphate (AdoPP[NH]P). Once inactivated, removal of ATP from the medium did not immediately restore the original activity. ATP- or AdoPP[NH]P-dependent inactivation could be blocked by concanavalin A, wheat germ agglutinin or rabbit antiserum against the membrane. Additions of these proteins after ATP addition prevented further inactivation but did not restore the original activity. Low concentrations of ionic and nonionic detergents increased the rate of ATP-dependent inactivation. Higher concentrations of detergents, which solubilize the membrane completely, inactivated the Mg2+-ATPase. Cross-linking the membrane components with glutaraldehyde prevented ATP-dependent inactivation and decreased the sensitivity of the Mg2+-ATPase to detergents. It is proposed that the regulation of the Mg2+-ATPase by ATP requires the mobility of proteins within the membrane. Cross-linking the membrane proteins with lectins, antiserum or glutaraldehyde prevents inactivation; increasing the mobility with detergents accelerates ATP-dependent inactivation.  相似文献

31P NMR chemical shifts of salts of adenosine 5′-triphosphate and diphosphate: ATPH2?22(Me4N+) · H2O, ATPH2?22 Na+ · 3.5 H2O, ATPH2?2Mg2+ · 4 H2O, ATPH2?2Ca2+ · 2 H2O, ADPH2?2(Me4N+) · H2O and ADPH2?Mg2+ · 4 H2O have been measured in 0.02 M 2H2O solutions at 145.7 MHz (22° C) at constant p2H values (8.20 and 6.20). The results are compared with those obtained from salts of adenosine 5′-monophosphate and other simpler phosphomonoesters, e.g. AMP2?2(Me4N+), AMP2?Mg2+, AMPH?Me4N+ and (AMPH?)2Mg2+. It is concluded that the effects exerted by Mg2+ and Ca2+ on the 31P NMR shifts of dipoly- and tripolyphosphates relative to monovalent cations are due mainly to changes in conformation of the polyphosphate chain rather than to purely electronic factors associated with the binding of divalent cations to the phospho-oxyanions. The data are consistent with the existence of the following complexes at p2H 8.20: (MgPαPβ)ADP? and (MgPαPγ)ATP2?af (MgPαPβ)ATP2?af (MgPβPγ)ATP2? with the latter equilibrium relatively fast in the NMR time scale. Monoprotonation of the terminal phosphate appears to weaken the Mg2+-polyphosphate binding, particularly at Pβ of MgADPH and at Pβ and Pγ of MgATPH?. The Mg2+-polyphosphate binding weakens further at p2H 3.70, i.e. in MgATPH2. Possible implications of the results in the mechanism of actomyosin Mg2+-ATPase in muscle contraction are discussed.  相似文献

The kinetics of association and dissociation of Escherichia coli 30 S and 50 S ribosomal subunits appear to fit the simple scheme
over a wide range of Mg2+ and ribosome concentrations, for the preparations studied (which have a sharp [Mg2+]-dependence on the equilibrium degree of association, e.g. 10% to 90% for 1.5 mm to 3.5 mm). Both rate constants depend strongly upon magnesium ion concentration (k2 goes from 0.04 × 106 to 21 × 106m−1 s−1 as [Mg2+] goes from 1.5 mm to 8 mm; k1 goes from 150 to 2 s−1 in the interval 1.0 to 3.0 mm), but k1 may level off above 3.0 mm and k2 increases slowly at high [Mg2+]. (The highest rate may not be far from the diffusion-controlled limit.) The primary effect of Mg2+, as calculated from the rather large changes in binding as a function of [Mg2+], is to decrease the contribution of electrostatic repulsion to the free energy of activation; specific, or class-specific, interactions of di- and multivalent cations with unknown ribosomal substructures may modulate this effect.  相似文献
