The sensory arborizations of the femoral chordotonal organ (FECO), trochanteral campaniform sensilla (CS) and the trochanteral hairplate (THP) are restricted to the ipsilateral hemiganglion and possess large dendritic fields in the dorsal and ventral neuropiles. The efferent projections of the fast (Df) and the slow (Ds) depressor motoneurons project into the same areas. Three groups of trichoid sensilla located on the coxal and trochanteral cuticular walls have central contralateral projections and in one case intersegmental projections. All sensory projections terminate either just lateral of or in the Ventral Association Centre (VAC). Comparisons with the mechanoreceptors of other insects reveal similar projection patterns for some proprioceptors and non-homologous central configurations for others.  相似文献

Three groups of giant fibers are found in the cockroach ventral nerve cord. A latero-dorsal group (dorsal GIs), a latero-ventral group (ventral GIs) and a medio-ventral group. The morphology of all three groups of fibers within the thoracic ganglia is described. The morphology of the dorsal and ventral GI pathways in the abdominal and suboesophageal ganglia is also described. The projection patterns of the neurons in each ganglion are remarkably similar which suggests a common function. When motorneurons 5rl (depressor) and 6Br4 (levator) are stained simultaneously with the dorsal and ventral GI groups, some branches from both motor and giant neurons converge. The branching of the remaining medio-ventral group of fibers and their proximity to areas receiving motorneuronal input suggests that these are the small diameter axons described by Dagan and Parnas (1970).  相似文献

Summary The ocelli of Periplaneta americana were studied by light and electron microscopy. The view that the ocellus of the cockroach represents a degenerated structure can no longer be supported. All organelles necessary for function are present.The club-shaped retinular cells lie homogeneously distributed in the cupule of the ocellus. Rhabdoms are seen as sickle-, y-, x-or star-shaped structures with up to six cells in formation. Cells were found which had formed two rhabdomeres.The mass of cell organelles lies in the cytoplasm between the cell nucleus and the rhabdom. Smooth endoplasmic reticulum is wound into a spindle formation of considerable size at the origin of the axon in some cells. A cylindrical body in which 10–40 microtubules are packed, as yet unknown in insect retinular cells, is described.The receptory area of the ocellus terminates in a tapetum which contains granules, soluble in alcohol. The axon bundles of the retinular cells run through the tapetum and immediately thereafter make synaptic contact with dendrites of the ocellar nerve cells, while still within the ocellus.The authors are indebted to Mrs. Margaret Weber-Wood for her linguistic assistance  相似文献

Poly(ADP-ribosylation) is a post-translational modification of nuclear proteins typical of most eukaryotic cells. This process participates in DNA replication and repair and is mainly regulated by two enzymes, poly(ADP-ribose) polymerase, which is responsible for the synthesis of polymers of ADP-ribose, and poly(ADP-ribose) glycohydrolase, which performs polymer degradation. The aim of this work was to investigate in the cockroach Periplaneta americana L. (Blattaria: Blattidae) the behaviour of poly(ADP-ribosylation). In particular, we addressed: (i) the possible modulation of poly(ADP-ribosylation) during the embryonic development; (ii) the expression of poly(ADP-ribose) polymerase and glycohydrolase in different tissues; and (iii) the role of poly(ADP-ribosylation) during spermatogenesis. In this work we demonstrated that: (i) as revealed by specific biochemical assays, active poly(ADP-ribose) polymerase and glycohydrolase are present exclusively in P. americana embryos at early stages of development; (ii) an activity carrying out poly(ADP-ribose) synthesis was found in extracts from testes; and (iii) the synthesis of poly(ADP-ribose) occurs preferentially in differentiating spermatids/spermatozoa. Collectively, our results indicate that the poly(ADP-ribosylation) process in P. americana, which is a hemimetabolous insect, displays catalytical and structural features similar to those described in the holometabolous insects and in mammalian cells. Furthermore, this process appears to be modulated during embryonic development and spermatogenesis.  相似文献

A preparation of citrinin from an isolate of Penicillium citrinum disrupted the spiracle control mechanism of the cockroach, resulting in excessive evaporation of water. The insect rapidly lost body weight and died due to dehydration within 48 h of treatment with citrinin. Presumptive evidence is given here for the neurotoxic nature of the mycotoxin.  相似文献

A phospholipase has been identified in the fat body of the American cockroach, Periplaneta americana, which removes fatty acid from the sn-2 acyl position of an artificial substrate. The enzyme has been characterized using a crude preparation obtained by low-speed centrifugation of the homogenized tissue. With 1-hexadecanoyl-2-(1-pyrenedecanoyl)-sn-glycero-3-phosphocholine as the substrate, the K(m) has been estimated to be 1.17 microM and the v(max) 113.5 pmol/min/mg protein. The phospholipase has a pH optimum close to 7 and shows maximal activity at 50 degrees C. Activity of the phospholipase has been determined in cytosolic and plasma membrane fractions. The specific activity of the latter fraction is approximately twice that of the cytosol. The enzyme in both fractions is Ca(2+)-independent. Arch.  相似文献

We established a classical conditioning procedure for the cockroach, Periplaneta americana, by which odors were associated with reward or punishment. Cockroaches underwent differential conditioning trials in which peppermint odor was associated with sucrose solution and vanilla odor was associated with saline solution. Odor preference of cockroaches was tested by allowing them to choose between peppermint and vanilla sources. Cockroaches that had undergone one set of differential conditioning trials exhibited a significantly greater preference for peppermint odor than did untrained cockroaches. Memory formed by three sets of differential conditioning trials, with an inter-trial interval of 5 min, was retained at least 4 days after conditioning. This conditioning procedure was effective even for cockroaches that had been harnessed in plastic tubes. This study shows, for the first time in hemimetaborous insects, that both freely moving and harnessed insects are capable of forming olfactory memory by classical conditioning procedure. This procedure may be useful for future electrophysiological and pharmacological studies aimed at elucidation of neural mechanisms underlying olfactory learning and memory.  相似文献

This study examined the biochemical characteristics of α‐amylase and hormonal (adipokinetic hormone: AKH) stimulation of α‐amylase activity in the cockroach (Periplaneta americana) midgut. We applied two AKHs in vivo and in vitro, then measured resultant amylase activity and gene expression, as well as the expression of AKH receptor (AKHR). The results revealed that optimal amylase activity is characterized by the following: pH: 5.7, temperature: 38.4 °C, Km (Michaelis–Menten constant): 2.54 mg starch/mL, and Vmax (maximum reaction velocity): 0.185 μmol maltose/mL/min. In vivo application of AKHs resulted in significant increase of amylase activity: by two‐fold in the gastric caeca and 4–7 fold in the rest of the midgut. In vitro experiments supported results seen in vivo: a 24‐h incubation with the hormones resulted in the increase of amylase activity by 1.4 times in the caeca and 4–9 times in the midgut. Further, gene expression analyses reveal that AKHR is expressed in both the caeca and the rest of the midgut, although expression levels in the former were 23 times higher than levels in the latter. A similar pattern was found for the amylase (AMY) gene. Hormonal treatment did not affect the expression of either gene. This study is the first to provide evidence indicating direct AKH stimulation of digestive enzyme activity in the insect midgut, supported by specific AKHR gene expression in this organ.  相似文献

Summary Innervation of the antennal heart, an independent accessory circulatory motor in the head of insects, was investigated in the cockroach Periplaneta americana by use of axonal cobalt filling and transmission electron microscopy. The muscles associated with this organ are innervated by neurones located in a part of the suboesophageal ganglion, generally considered to be formed by the mandibular neuromere. Dorsal unpaired median (DUM) and paired contralateral neurones were stained. The axons of all these neurones run along the circumoesophageal connectives and through the paired nervus corporis cardiaci III into the corpora cardiaca. They pass through these organs forming fine arborizations there and exit anteriorly as a small pair of nerves which terminate at the antennal heart-dilator muscles. Numerous branches of these nerves extend beyond the lateral borders of the large transverse dilator muscle and terminate in the ampullar walls of the antennal heart. These neurosecretory fibres form neurohaemal areas which obviously release their products into the haemolymph, which is pumped into the antennae. The possible functions of the neurones associated with the antennal heart are discussed with respect to both, their role as a modulatory input for the circulatory motor and as a neurohormonal release site.  相似文献

The ultrastructural organization and the junctional complexes of peripheral nerves have been investigated in the cockroach Periplaneta americana. Nerve 5 is surrounded by a layer of connective tissue, the neural lamella, beneath which is a layer of perineurial glial cells wrapping the axons. Adjacent perineurial cells are joined to one another by septate, gap and tight junctions. Septate and gap junctions were observed in freeze-fracture replicas of main trunk nerve 5. Septate junctions were found as rows of PF particles mainly in perineurial cell membranes. Gap junctions exhibited EF macular aggregates in perineurial and subperineurial glial cells. During incubations in vivo with extracellularly applied ionic lanthanum, the lanthanum did not penetrate beyond the perineurium. Where nerve 5 branches and contacts the muscle, lanthanum penetrated freely between the muscle fibres and the nerve branches. In small peripheral branches where the axons are surrounded by single a glial layer, lanthanum is unable to penetrate to the axolemma.  相似文献
