Sulphur requirements ofCurvularia pallescens, Alternaria citri, andA. tenuis have been studied. The fungi exhibited selectivity towards different sulphur compounds. Magnesium sulphate gave best growth and sporulation of the three fungi and it could not be replaced by any other sulphur compound.  相似文献

The effect of different sulphur and phosphorus compounds on the growth and sporulation ofCurvularia pallescens Boed. has been studied. Nine different sulphur sources were tried but among them only magnesium sulphate yielded the best dry weight of the fungus. Zinc sulphate, sodium sulphate, sodium thiosulphate, potassium sulphate and calcium sulphate supported good growth. Poor growth was recorded on sodium bisulphite, ammonium sulphate, sodium sulphide and control. Sporulation was excellent on magnesium sulphate. It was good on zinc sulphate, sodium sulphate and potassium sulphate. On sodium thiosulphate, calcium sulphate, sodium bisulphite and control it was fair. Sodium sulphide and ammonium sulphate had inhibitory effect as sporulation was poor and nil on these two compounds respectively.Six phosphorus compounds were studied. Tripotassium phosphate gave best growth and excellent sporulation. Good growth and excellent sporulation was recorded on monobasic potassium phosphate and magnesium phosphate. Growth and sporulation were good on dibasic potassium phosphate and sodium dihydrogen phosphate. Ammonium phosphate was poorly utilized.  相似文献

The sulphur nutrition of three isolates ofAlternaria tenuis Auct., isolated from the diseased leaves ofMangifera indica L.,Musa paradisiaca L. andPsidium guajava L., was studied. They were grown on the medium devoid of sulphur as well as on media containing various sources of sulphur viz., ammonium sulphate, sodium hyposulphite, sodium thiosulphate, magnesium sulphate, potassium sulphate, potassium metabisulphite, zinc sulphate and thiourea. Sodium hyposulphite, sodium thiosulphate, magnesium sulphate, potassium sulphate and zinc sulphate were generally found to be satisfactory sources for the growth of all the isolates under study. Poor growth of the different isolates was observed on the medium devoid of sulphur.  相似文献

Twenty-oneRhizobium andBradyrhizobium strains were testedin vitro against the mycelial growth of three pathogenic fungi on solid and liquid media. All tested rhizobia and bradyrhizobia significantly suppressed the growth of the three soil-borne root-infecting fungi (Fusarium solani, Macrophominia phasolina andRhizoctonia solani) either in the absence or presence of iron. This indicates that the siderophore played a minor role in the biocontrol potential ofRhizobium andBradyrhizobium against pathogenic fungi. Pot experiments revealed that the numbers of propagules causing disease after 4 weeks of planting varied with species and host plant. The three most activeRhizobium andBradyrhizobium strains (R. leguminosarum bv.phaseoli TAL 182,B. japonicum TAL 377 andBradyrhizobium sp. (lupin) WPBS 3211 D) tested under greenhouse conditions for their ability to protect one leguminous (soybean) and two non-leguminous (sunflower and okra) seedlings from root rot caused byFusarium solani, Macrophominia phaseolina andRhizoctonia solani provided significant suppression of disease severity compared with nonbacterized control in both leguminous and non-leguminous seedlings.Bradyrhizobium sp. Summary Nitrogen and sulphur requirements ofColletotrichum inamdarii Lal isolated from the leaves ofCarissa carandas L. have been studied. DL-serine, L-asparagine and L-phenylalanine have been found to be of good nitrogen source followed by potassium nitrate, calcium nitrate, magnesium nitrate, DL-alanine, ammonium nitrate, glutamic acid, ammonium sulphate, DL-valine, aspartic acid, ammonium chloride, ammonium hydrogencarbonate, L-histidine and potassium nitrite. There was no growth in the absence of nitrogen.Sporulation was excellent on calcium nitrate and sodium nitrate, Very good on DL-serine, potassium nitrate, and magnesium nitrate. Good on L-asparagine, L-phenylalanine and ammonium oxalate. Fair on DL-alanine, DL-leucine, ammonium sulphate, DL-valine, ammonium chloride and L-histidine whereas poor on glutamic acid, aspartic acid, ammonium tartarate and ammonium nitrate. Few spores were observed on ammonium hydrogencarbonate but potassium nitrite did not show any sporulation.Amongst the sulphur compounds sodium bisulphate gave the best growth and good sporulation, followed by sodium thiosulphate, magnesium sulphate, ammonium sulphate and potassium sulphate. Summary Interactions of 46 isolates of fungi with an isolate ofM. phaseoli from cotton, was studied in agar culture. They were grouped into 4 different types of reactions.T. viride was found to inhibit the growth ofM. phaseoli and grow over its colony. The hyphae ofT. viride were observed to coil around the hyphae ofM. phaseoli. A majority of the isolates tested had no effect on the growth ofM. phaseoli. M. phaseoli, however, was found to overgrow the test fungi.  相似文献

Summary Field plots of Tifton loamy sand were treated with various soil pesticides and left exposed or covered with biodegradable paper or black polyethylene film mulch. Cucumber, squash, muskmelon, sweetcorn, and polebean were planted in one or more experiments. Trickle irrigation under the film was used in several tests. Isolations were made from roots of all crops except sweetcorn, and the fungi most commonly isolated wereFusarium oxysporum, Pythium spp.,F. solani, F. roseum, andRhizoctonia solani from cucurbits andF. solani andF. oxysporum from polebean. Significantly fewer fungi were isolated from plants grown in soil treated with DD-MENCS (20% methyl isothiocyanate +80% chlorinated C3 hydrocarbons) or methyl bromide-chloropicrin (2:1) (MBC) than from controls, and populations ofPythium spp.,F. solani, andF. oxysporum, were reduced in soil. Three isolates ofA. tenuis isolated from the diseased leaves ofMangifera indica l. Musa paradisiaca l. andPsidium guajava l. were investigated. They were grown on different sources of nitrogen viz., potassium nitrate, sodium nitrate, calcium nitrate, ammonium nitrate, sodium nitrite, ammonium sulphate, ammonium chloride, glycine, DL-valine, L-glutamic acid, urea, thiourea, L-asparagine and peptone. They were also grown on the medium lacking nitrogen. A wide variation was observed in the growth and reproduction of the different isolates. The growth of all of them was good on potassium nitrate, calcium nitrate, glycine, DL-valine, L-glutamic acid, L-asparagine and peptone but the sporulation was satisfactory on calcium nitrate only. Sodium nitrite supported moderate growth of banana leaf isolate whereas there was no growth of the other two isolates. None of the organisms could grow on the medium lacking nitrogen as well as on thiourea. The results obtained with the isolates under study have been compared with those of earlier investigators and it has been clearly established that the different isolates ofA. tenuis could show marked differences in their nitrogen requirements.  相似文献

The effect ofStachybotrys atra on fungi, actinomycetes and bacteria was studied in agar culture. Of the 27 genera and 52 species of fungi, 6 species of actinomycetes and 5 genera and 10 species of bacteria tested,S. atra was found to inhibit 95.7 % fungi, all the actinomycetes and 83.3 % of bacteria.Aspergillus ustus, 2 unidentified species ofFusarium, Rhizobium trifolii andPseudomonas spp. were however not inhibited. In experiments in soil,S. atra inhibited the growth ofTrichoderma viride. S atra was ineffective whereT. viride andS. atra were inoculated simultaneously or whereT. viride was added 7 days earlier. Besides,S. atra was not found to controlMacrophomina phaseoli infection on cotton. Soil culture ofS. atra showed phytotoxic effect on cotton seedlings.S. atra produced a dark brown to black thermostable toxic metabolite in the medium. The culture filtrate retarded the growth ofM. phaseoli in agar culture. Despite the importance ofS. atra as a fungistatic agent with wide antimicrobial spectrum, the use ofS. atra as a protectant against plant pathogens seems to be limited because of its phytotoxicity on cotton.  相似文献

Summary Growth response ofRhizoctonia solani Kühn, the damping-off fungus, to metabolites of selected antagonistic rhizospheric bacteria and fungi of some Egyptian cotton varieties, namely, two strains ofBacillus subtilis Cohn,Aspergillus terreus Thom, andAspergillus flavus Link produced in culture media containing nitrate- or ammonium-nitrogen sources, proved the potency ofB. subtilis metabolites in inhibitingR. solani mycelial growth whether from nitrate- or ammonium-nitrogen culture media. Metabolite filtrates ofB. subtilis (II) are more potent than those ofB. subtilis (I). Increasing concentration of bacterial metabolite filtrates resulted in a decreased mycelial dry weight ofR. solani. The bacterial inhibitory factor forR. solani mycelial growth is partially affected by heat. Metabolite filtrates ofA. terreus from nitrate-nitrogen are slightly more potent than from ammonium-nitrogen culture media while an opposite relation is evident withA. flavus metabolites. Growth responses ofR. solani to different experimental dilutions of metabolite filtrates ofA. terreus andA. flavus proved the intervention of the nutritive factor in witholding growth of the damping-off fungus.  相似文献

Effect of fourteen different nitrogen sources on five species ofSphaeropsis i. e.S. cactinae, S. cycadis, S. jodhpurensis, S. muehlenbeckiae andS. punicae, obtained respectively from the infected parts ofCitrus medica, Cycas circinalis, Cordia gharaf, Muehlenbeckia platycladoes andPunica granatum was studied, under cultural conditions on Asthana and Hawker's Medium A by substituting its nitrogen with equal quantity of an other nitrogen source. All the present species reproduced well on potassium nitrate, sodium nitrate, ammonium sulphate and ammonium nitrate. None of the present species could grow on sodium nitrite. Out of all amino acids, no one was found to be excellent for the growth and sporulation of the present fungi. Most of the amino acids supported poor growth and sporulation of three of the five species ofSphaeropsis, S. cactinae, S. cycadis andS. punicae on L-asparagine, Glycine, L-cystine and L-alanine. Maximum and minimum pH for growth and sporulation ranged from 6.5 to 7.8 and 5.3 to 4.2 respectively. Decrease in adjusted pH was also observed for all the species on L-asparagine and DL-valine.  相似文献

The effect of different carbon and nitrogen compounds on growth and sporulation ofC. microcephala (Wallr.)Tul. causing ergot disease of Bajra has been studied. Nine different sources of carbon were used but cane sugar was found to be the best source for both, growth and sportulation of the fungus. Glucose, sucrose and maltose gave good growth but fair sporulation. Lactose and sorbitaol proved to be the poor sources. However, fungus failed to utilize starch, dextrin and mannitol.Nineteen nitrogen compounds were tried for the growth and sporulation of the fungus. Best growth and sporulation were supported by peptone and glycine. L-asparagine, DL-valine, Urea, magnesium nitrate and L-proline supported good growth and fair sporulation except DL-valine where it was excellent. Poor growth was obtained on L-isoleucin, ammonium sulphate, potassium nitrate,-alanine, ammonium chloride, DL-aspartic acid and DL-methionine. Fungus failed to utilize thio-urea.  相似文献

Summary AlthoughRhizopus stolonifer, Aspergillus niger andRhizoctonia solani could be grown on gum arabic solutions of 1–4% concentrations, yet their growth was poor presumably due to lack of sufficient nitrogen. Gum arabic was found to be a good carbon source when it replaced sucrose in Czapek-Dox medium. The sporulation ofRhizopus was not affected by the presence of gum in the medium, while that ofAspergillus was appreciably affected. The number of sporulating heads was reduced, irregularity in the shape of the head due to variation in the length of spore chains was also noticed. A new medium of pH 5.6 was found satisfactory for fungal growth and could be recommended for soil isolations. Its composition is: 4% gum arabic, 1% peptone, 0.025% sodium chloride and 0.001% ferrous sulphate.  相似文献

Summary The physiological conditions governing growth and sporulation ofSaksenaea vasiformis Saksena, a fungus with outstanding morphological features quite peculiar for Mucorales, were determined. Earlier studies made byTiwari (1955) on a strain of the same species had shown that this fungus is incapable of sporulating on any synthetic medium normally employed for growing fungi.The fungus had been found to have a high tolerance for very low and high pH values. It showed maximum growth at two pH values, one near neutral point, at pH 6, and another at high alkaline pH value, i.e., pH 11. Reason for this behaviour of this fungus has already been discussed. The most suitable temperature for the growth of the fungus was found to be between 30–35° C.Nearly all the carbon, nitrogen and sulphur sources which generally favour growth of fungi were found to support vegetative growth of this fungus as well. However, sporulation in this fungus had peculiar nutritional needs. Only some of the carbon sources, viz., arabinose, rhamnose, sorbose, galactose, lactose and citric acid which supported poor growth, were found to support good or excellent sporulation. But it may be stated that not all carbon sources supporting poor growth could induce sporulation of the organism. Also none of the nitrogen or sulphur sources could induce the fungus to sporulate in presence of glucose as carbon source.  相似文献

Summary Major sclerotial polypeptides from the psychrophiles,Myriosclerotinia borealis (W 51),Coprinus psychromorbidus (LRS 131),Typhula idahoensis (W 21), andTyphula incarnata (W 21) were purified by using polyacrylamide gel electrophoresis and electroelution. Polyclonal antibodies were raised against these major sclerotial polypeptides. Immunofluorescence microscopy showed that the major sclerotial polypeptides from all four psychrophilic species were sequestered in discrete protein bodies of cultured and field-grown sclerotia. Western blot analysis indicated that all antisera reacted positively with their respective antigens, the major sclerotial polypeptides. Reciprocal immunological cross-reactions were observed between the major sclerotial polypeptides ofM. borealis (W 51) andT. idahoensis (W 21). Antiserum to the major sclerotial polypeptides of bothM. borealis andT. idahoensis also recognized the major sclerotial polypeptides ofC. psychromorbidus (LRS 131). A cDNA library was constructed using poly(A)+RNA fromPisum sativum which had been treated for 8 h with the fungusFusarium solani f. sp.phaseoli. Two thousand four hundred recombinant colonies were screened by differential colony hybridization using32P-labelled cDNAs prepared from RNA extracted from either noninoculated or inoculated pea tissue. cDNA clones were then selected, which showed greater hybridization with cDNA prepared from pea RNA 8 h post-inoculation than with a cDNA probe from 0 h. Seven distinct hybridization classes were chosen for further study. Northern blot analyses of total cellular RNAs inoculated for 16 h with eitherF. solani phaseoli or water demonstrated that each cDNA clone selected represents an mRNA species which increases substantially in abundance during infection. Results of3H-uridine pulse-labelling experiments suggested that enhanced synthesis is at least partially responsible for the accumulation of the fungus-inducible mRNAs which hybridized with the clones.  相似文献

Growth of the fungusMicrosporum gypseum and utilization of cystine during this growth was studied in a glucose-arginine medium containing either sodium sulphate, andL-cystine orDL-cystine. Replacement of sulphate withL-cystine brought about no significant changes in the growth of the microorganism. Utilization ofL-cystine as a source of carbon and nitrogen was rapid and complete and excess sulphur was excreted into the medium in the form of sulphate. Similarly excreted were also minute amounts of sulphite which immediately reacted with the remaining cystine to formS. sulphocysteine. Growth ofM. gypseum in a medium withDL-cystine was slow. Although this substance was not utilized as readily asL-cystine, its utilization was still complete and excess sulphur was similarly excreted in the form of sulphate and sulphite. The initial step in the utilization of theD-isomer is probably its extracellular deamination.  相似文献

The content of total, bound and osmotically free magnesium was estimated in various fungi and in the yeastSaccharomyces cerevisiae. Total magnesium increases at lower growth rates ofEndomyces magnussi andPenicillium chrysogenum 140A as well as during the logarithmic stage of growth ofPenicillium chrysogenum Q-176. The binding of magnesium requires orthophosphate, decreasing during lack of external phosphate when the intracellular concentration of free magnesium rises. The fungi were found to contain a novel form of bound magnesium, a polymeric magnesium orthophosphate (POMg), which appears to take part in the control of free magnesium level inPenicillium chrysogenum Q-176. The level of free magnesium is proportional to the growth rate ofEndomyces magnusii andPenicillium chrysogenum Q-176 and 140A. Total, as well as free, magnesium changes less than three-fold as external Mg concentration is changed 13,000-fold. The magnesium up against concentration gradients of 1∶25 to 1∶1300, the metal being distributed non-uniformation the cells ofSaccharomyces cerevisiae.  相似文献

Severity of stem-rot disease of peanut caused byRhizoctonia solani was reduced by 54.9 and 68% in plants of two cultivars treated in the greenhouse with antagonistic strains ofPseudomonas fluorescens. These strains were selected based on theirin vitro toxicity to mycelial growth and sclerotial germination ofR. solani. In field experiments, bacterization of peanuts withP. fluorescens resulted in taller plants (by 25.7%) and increased yields (by 59.0%).  相似文献

An experiment was carried out to study the colonization of soil microorganisms on cotton roots buried in the soil.Fusarium culmorum, F. solani, F. oxysporum, Macrophomina phaseoli andBacillus sp. were found to be the predominant colonizers followed byBotryodiplodia theobromae, Fusarium acuminatum, Aspergillus niger, A. flavus, Stachybotrys atra, Tilachlidium humicola andRhizopus sp. The practice of leaving cotton roots in the soil would therefore add to the building up of the inoculum potential of the soil inhabiting fungi several of which are serious root pathogens.  相似文献
