Impairment of cognitive functions, particularly long-term (episodic) and working memory, is one of the earliest prognostic symptoms of Alzheimer's disease, both cognitive impairment and neurodegeneration being mediated by amyloid-beta neurotoxicity. Effects of intracerebroventricular administration of amyloid-beta peptide (25-35) [A beta(25-35)] to rats on the retention of previously learned task in an 8-armed radial maze was studied. A beta(25-35) was injected bilaterally, at doses of 15 or 30 nmol/rat, 7 days after the preliminary learning. The performance in the maze was tested 60 days after the surgery. A beta(25-35) impaired the short-term memory, with no significant effect on the long-term memory. No dose dependence could be demonstrated. 相似文献

Summary A similar sequence of behavioral effects was observed for either cooling or heating; most effects occurred on changing temperature of entire fish or of only the cerebellum. On moderate heating or cooling, fish are hyperexcitable, spontaneously hyperactive; on further heating or cooling swimming is uncoordinated; when the subcerebellar structures are heated or cooled, equilibrium is disturbed; on further heating or cooling coma and respiratory failure ensue. Critical temperatures are modifiable by acclimation. The behavioral effects of cerebellectomy are additive with temperature effects on motor centers.Electrical activity of Purkinje neurons changes in the same thermal ranges as behavior. Inhibition via cerebellar interneurons is most sensitive and can be modified by acclimation. Ongoing activity increases with warming up to a blocking temperature; interspike interval histograms show pattern changes during warming. Activation via mossy fibers-granule cells is more sensitive than that via climbing fibers, and antidromic impulses are most resistant.A neuronal model based on inhibitory actions of Purkinje neurons on motor centers and parallel feedback excitatory pathways can explain both behavioral and electrical observations. 相似文献

L-精氨酸和氨基胍对大鼠内毒素性肺损伤治疗作用的实验研究 总被引:8,自引:0,他引:8
目的:观察L-精氨酸(L-arginine)和一氧化氮合酶抑制剂氨基胍(AG)对内毒素性肺损伤的治疗作用。方法:采用静脉注射脂多糖(LPS)制备内毒素性肺损伤大鼠模型。将48只SD大鼠随机分为6组:空白对照组、LPS模型组、AG治疗组(50mg/kg)、L-精氨酸(500mg/kg)、(250mg/kg)和L-精氨酸(250mg/kg)+AG(50mg/kg)治疗组。经腹腔给药,实验过程中监测大鼠平均动脉压(MAP),定时取静脉血测定血浆中NO含量,于规定时间处死大鼠,迅速取出肺脏,观察LPS引起大鼠急性肺损伤后肺系数、肺水肿情况和肺组织中丙二醛(MDA)含量、一氧化氮合酶(NOS)、超氧化物歧化酶(SOD)活性的变化,以及L-精氨酸和氨基胍分别单独给药和二者联合给药对内毒素性肺损伤的治疗作用。结果:氨基胍可明显升高MAP,降低肺系数和肺含水量,减少血浆中NO含量,可显著降低肺组织中NOS活性,减少MDA含量,增强SOD活性,改善肺损伤;L-精氨酸可明显降低肺系数和肺含水量,减少MDA含量,增强SOD活性;L-精氨酸与氨基胍联合应用亦得到上述类似结果。结论:L-精氨酸和氨基胍分别单独给药以及二者联合给药对内毒索性肺损伤均具有治疗作用。 相似文献

Unilateral intracerebroventricular (ICV) injections of a high dose of CCK7 have been reported to elicit barrel rotations accompanied by contralateral postural asymmetry; there was no spontaneous locomotor activity other than barrel rolling. The aim of the present study was to investigate the effects of lower doses of CCK-peptides on circling behavior; it was reasoned that if ambulation was present following unilateral ICV administrations of lower doses of CCK, then the contralateral postural asymmetry previously reported might be expressed as contraversive circling. In the present study, spontaneous locomotor activity was observed following ICV injections of lower doses of CCK sulfated octapeptide (CCK8), desulfated CCK octapeptide (dCCK8) and CCK tetrapeptide (CCK4). As postulated, contraversive circling was induced by CCK8 (0.5–5000 ng, ICV); the two other CCK fragments, dCCK8 and CCK4, were inactive in this respect. In addition, the contraversive circling bias induced by CCK8 (5.0 ng, ICV) was attenuated by co-injections of the CCK antagonist proglumide (10 and 100 ng) and by intraperitoneal injections of the dopamine (DA) antagonist haloperidol (0.05 and 0.1 mg/kg, 45 min prior to ICV CCK8). These data suggest that the effect is mediated by CCK receptors and through a facilitatory influence on central DA function. 相似文献

Active immunisation of albino rats by the BSA-conjugated CCK-4 induced formation of antibodies to the CCK-4 and some long-term changes of the rat behaviour. These changes were contrary to anxiogenic effect of the CCK-4 and demonstrated an anxiolytic effect of the immunisation. The data obtained suggest a possibility of an immunocorrection of pathological anxiety and fear by an inverse immunoregulation. 相似文献

This study analyzed the effects of L-arginine and non-specific nitric oxide (NO) synthase blocker (L-NAME) on structural and metabolic changes in experimental ischemia/reperfusion injury in the rat. Histopathological evaluation of rat tissues after reperfusion was also performed. The animals were divided into four groups: [1] nonischemic control, [2] ischemia 4 hrs/repefusion 30, 60, 120 min, [3] ischemia/reperfusion after L-arginine administration, [4] ischemia/reperfusion, after L-arginine, and L-NAME. L-arginine (500 mg/kg) and L-NAME (75 micromol/rat/day) were administrated orally for 5 days before experiment. Concentrations of free radicals, CD-62P, CD-54 and malonyl dialdehyde (MDA) in tissues, and MDA and NO levels in sera were determined. Free radical levels significantly increased in reperfused skeletal muscle, small and large intestines. In large bowel, reperfusion increased MDA levels and evoked a rise of endotoxin level while NO levels decreased. Histological studies showed an increase in the number of lymphocytes in both intestines. Administration of L-arginine reduced leukocyte adherence associated with ischemia-repefusion injury, decreased the levels of free radicals and MDA in the examined tissues, and inhibited the release of endotoxins into blood. L-arginine-treated animals showed higher serum NO levels and reduced leukocyte bowel infiltration. Concomitant L-NAME administration reduced serum NO and tissue free radical [corrected] levels, but did not affect intestinal leukocyte infiltration. L-arginine could ameliorate intestinal ischemia/reperfusion injury and constitute a possible protective mechanism by decreasing neutrophil-endothelial interactions, stimulating free radical scavenging and reducing lipid peroxidation. 相似文献

The in vitro effects of pH on goldfish (Carassius auratus) ovulation were investigated. Final oocyte maturation and follicular detachment were induced in vivo in gravid goldfish by HCG injections and elevated holding temperatures. Females were biopsied to determine the time of germinal vesicle breakdown and when oocytes should be removed for in vitro incubations. Prior to ovulation, the ovaries were removed, dissected and mature intrafollicular oocytes were incubated in media of varying pH (7.3-8.9). There was a significant increase in ovulation as the pH of the incubation increased and this ovulation could be blocked by indomethacin. Indomethacin did not inhibit the actual mechanism of oocyte expulsion since exogenous PGF2 alpha induced ovulation in indomethacin blocked incubates. Increased pH did not increase the ovulatory response observed with exogenous PGF2 alpha. The combined results suggest that an increase in incubation pH stimulates prostaglandin synthesis that, in turn, stimulates ovulation. 相似文献

Summary Hydrostatic pressure has been shown to reverse the effects of anaesthetics in intact animals. To account for this, a range of thermodynamic hypotheses and models based on the physicochemical properties of various hydrophobic systems have been put forward. All these hypotheses and models predict that temperature should have some simple effect equivalent to that of pressure. The present work was undertaken to study the relationship between n-butanol concentration, temperature and the performance of a conditioned response in the goldfish, to see if it fitted any of the theories.It was found that the concentration of n-butanol required to just block the conditioned response in the goldfish was maximal (18mM) at about 18 °C and that both raising the temperature and lowering it reduced the required concentration. The sharp reduction in successful behaviour at the normal thermal limits were little changed by the presence of the alcohol.The absence of a simple relationship between temperature and concentration indicates the necessity of proposing a more complex model than the simple hydrophobic site, and a cellular mechanism is put forward linking both membrane and metabolic events. 相似文献

We investigated whether dietary supplementation with L-arginine, the endogenous precursor of nitric oxide, might affect serum lipid levels and activities of intestinal mucosa enzymes in animals, in which diabetes was induced by administration of streptozotocin. Control and diabetic rats were fed diets with or without 2% L-arginine supplementation for 4 weeks. Diabetic rats had significantly higher concentrations of serum triglycerides and LDL-cholesterol than control rats. These alterations were partially reduced by L-arginine supplementation. Experimental diabetes did not influence the lactase and leucine aminopeptidase activity in the intestine, but the activity of alkaline phosphatase was increased. Furthermore, activities of maltase and sucrase in the intestinal mucosa were elevated in streptozotocin-induced diabetic rats and were restored to control levels after dietary L-arginine supplementation. On the basis of the present experimental evidence, dietary L-arginine supplementation appears to affect the metabolism of lipoproteins and might alleviate some gastrointestinal dysfunctions, commonly seen in diabetes mellitus. 相似文献

Apoptosis is a form of cell death, characterized by morphological and biochemical changes. Apoptosis occurs in the normal testis and in response to different agents. In this study, we investigated the effect of gonadotropin-releasing hormone (GnRH) in testicular apoptosis in the goldfish. GnRH is a decapeptide that is expressed in different tissues, including gonads in mammalian and non-mammalian species. While GnRH is considered to be a paracrine mediator of ovarian follicular atresia, the role of GnRH in the testis is less clear. In the present study, treatments with native salmon GnRH and chicken GnRH-II increased DNA fragmentation (a hallmark of apoptosis) in the mature goldfish testis. On the other hand, gonadotropin hormone was found to act as survival factor, by decreasing spontaneous and GnRH-induced DNA fragmentation in the goldfish testis. The results demonstrate that GnRH plays an important paracrine role in the control of apoptosis in the goldfish testis. 相似文献

In mammals, NPY is a key factor in the regulation of feeding behavior. In the present study, the effects of refeeding for 1-3 h in 72-75-h food deprived (FD) goldfish on the levels of NPY mRNA in telencephalon-preoptic (TEL-POA), hypothalamus (HYP) and optic tectum-thalamus (OT-THAL) were examined, using Northern blot analysis. Goldfish FD for 72 h exhibited a significant increase in NPY mRNA levels in all brain regions. At 1 h after 72-h FD (73-h FD), NPY mRNA was significantly increased in TEL-POA and OT-THAL, but remained the same as 72-h FD fish in HYP. At 3 h after 72-h FD (75 h), all brain regions exhibited a significant increase in NPY mRNA levels. However, subsequent refeeding for 1-3 h rapidly and completely reversed the effects of FD in all brain regions, reaching fed levels within 1-3 h of refeeding. Serum GH levels were highest in 72-h FD fish, but decreased significantly over 1-3 h after 72-h FD; whereas, refeeding reversed the increase in serum GH levels only at 3 h after refeeding. Taken together, these results further support that NPY is a physiological brain transducer involved in the regulation of daily appetite and feeding in goldfish. 相似文献

The effects of intraperitoneal thyroxine administration (1 microgram/body weight) on postnatal days 1-3 and the subsequent alterations upon the development of six self-grooming components were measured postnatally in male Wistar rats between days 1-60. Observations on self-grooming components showed that thyroxine-treated rats did not show consistent significant differences in the duration of grooming movements of short displacement directed to the forepaws and head throughout the study, as compared to controls. By contrast, after weaning, the duration of grooming movements of long displacement directed to the fur, genital area, and body scratching were significantly increased. The findings suggest that early thyroxine treatment may primarily interfere with the neural circuitry that modulate long displacement grooming movements, rather than with the substrates controlling short displacement grooming activities. Thus, the hormone might be acting upon neural modulatory systems of self-grooming having different vulnerability in relation to the level of maturity of the brain circuits underlying each grooming component. 相似文献

The catabolism and excretion of [26-14C]cholesterol was studied in rats on semisynthetic and commercial diets low in fat or containing 15% butter or corn oil. Rats on the low fat commercial diet oxidized the labeled cholesterol to 14CO2 at more than twice the rate of those on the semisynthetic diet. Fecal excretion of labeled lipid was also somewhat higher with the commercial diet. The added fats had little effect on rate of oxidation of cholesterol but dietary corn oil stimulated fecal excretion of labeled lipid. The rate of loss of labeled cholesterol through oxidation and excretion showed a positive correlation with cholesterol biosynthesis, as measured previously by acetate incorporation into cholesterol in rats on the same kinds of diet. A simple method for efficient trapping and counting of 14CO2 was developed, which facilitated measurement of low levels of 14CO2 in expired air. Estimation of bile acid production from the rate of oxidation of [26-14C]cholesterol to expired 14CO2 and the specific activity of plasma cholesterol gave somewhat higher values than those obtained by other methods. To demonstrate whether nutritional lithium deficiency is associated with behavioral changes, male Sprague-Dawley rats were placed on a lithium-deficient diet (Li content <.01 ppm). A lithium-deprived group, receiving drinking water containing 31 μM NaCl, were compared to a control group receiving drinking water containing 31 μM LiCl. Growth and general appearance were the same in both groups. However, lithium-deficient animals demonstrated decreased aggression in social interactions with other rats and also in response to handling. The phase of wheel-running activity was delayed by 0.8 h and exhibited decreased amplitude (p<.05). Other behaviors, including acquisition and retention of a passive avoidance response, were unaffected by lithium deprivation. An erratum to this article is available at . 相似文献
