In host-range studies, bean common mosaic virus strains (BCMV-NL1, -NL3 and -NY 15) usually induced distinct systemic symptoms in susceptible bean cultivars and latent infection in several Vigna genotypes (except NY15 which gave mosaic symptoms in the latter), while blackeye cowpea mosaic virus (B1CMV-W) caused distinct systemic symptoms in several Vigna genotypes and only weak systemic symptoms in a few bean genotypes only. Biologically, B1CMV-W was closest to BCMV-NY15 and less close to -NL1. When using antisera to the three BCMV strains and five strains of B1CMV (including a strain originally considered cowpea aphid-borne mosaic virus CAMV-Mor) in SDS-immunodiffusion and ELISA, BCMV-NL1 and -NY15 were found to be closely related to each other and to BICMV-Fla, -NR and -W, and less closely to BICMV-Ind and -Mor. Serological relationships of BCMV-NL1 and -NY15 to BCMV- NL3 were more distant, which is in line with the biological distinction of NL3 in causing temperature-independent necrosis in bean cultivars with the necrosis gene I. PAGE analysis of coat proteins revealed that the three strains of BCMV and B1 CMV-W have similar but non-identical molecular masses. Although molecular hybridisation may further elucidate quantitative relationships between potyvir-uses, variation within and among the potyviruses may continue to pose problems in their classification and identification.  相似文献

The growth and lipid content of three Botryococcus braunii strains from China (CHN), United Kingdom (UK) and Japan (JAP) were compared at three temperatures (20, 25 and 30 C), three irradiances (60, 100 and 300 W m−2) and four salinities (0, 0.15, 0.25, and 0.5 M NaCl) for 30 days, respectively. In the temperature trial, the UK strain and JAP strain grew faster at 25 C than at other temperatures, while the CHN strain performed equally well at 20 and 25 C. The JAP strain grew slowest among the three strains at all temperatures, whereas the growth rate of the CHN and UK strains was similar at all temperatures except at 20 C. The UK strain contained the highest lipid content, but the CHN strain had the lowest lipid content at most temperatures. In the light trial, the highest growth rate was found in the UK strain and the lowest growth rate was observed in the JAP strain at most irradiances. The UK and JAP strains contained more lipids than the CHN strain at 60 and 100 W m−2, but the lipid content was not significantly different among the three strains at 300 W m−2. In the salinity trial, both the CHN and UK strains grew faster than the JAP strain at all salinities, but the growth rate between the CHN and UK strains was not different. However, the CHN strain had the lowest lipid content whereas the UK strain produced the highest lipids at most salinities. Our results indicate that the CHN strain and the UK strain grow faster than the JAP strain, but the UK and JAP strains produce more lipids than the CHN strain. The UK strain should be considered as a potential B. braunii strain for the exploitation of renewable energy.  相似文献

香菇是我国深受欢迎的食药用菌,产量居食用菌产业之首。为拓宽栽培香菇菌株的遗传背景,解决现有主栽香菇菌株种性退化的问题,本研究以香菇‘808’菌株与3个香菇野生资源、3个栽培菌株(辽抚4号、BY1和荷香)开展单-双杂交育种,开发适合当地栽培环境的主栽品种。研究结果显示:在270对单-双杂交组合中,通过锁状联合观察和与亲本的拮抗试验,鉴定出89个杂交子,进一步通过栽培试验筛选出10个性状相对优良的候选菌株;最终通过农艺性状对比试验,筛选出ZJXG 5和ZJXG 8两个优良杂交菌株,这两个菌株均以当地长白山野生资源S1和本地主栽菌株BY1为双核杂交亲本选育而来,表现出超亲显著、菇形圆整、颜色呈浅褐色的特性,前4潮生物学效率在86%-88%之间。本研究结果揭示,在香菇杂交育种中,当亲本菌株来源地域与杂交后代菌株筛选地域相近时,单双杂交亲和率、杂交菌株的出菇率和丰产性指标较高,这提示我们,利用当地野生香菇资源更有利于创制适合当地自然环境的地方品种。  相似文献

The common bean (Phaseolus vulgaris) is a high protein crop and the main legume in the cropping system of western Kenya. Despite its importance, common bean yields are low (<1.0 t/ha) and declining. Bean common mosaic virus (BCMV) and bean common mosaic necrosis virus (BCMNV) are the most common and most destructive viruses and can cause a yield loss as high as 100%. In Kenya, a limited number of cultivars and exotic genotypes with resistance to BCMV and BCMNV strains have been reported. This study sought to determine the distribution and screen popular cultivars for resistance to the viruses. In October 2016 and May 2017, two diagnostic surveys for bean common mosaic disease (BCMD) were conducted in seven counties of western Kenya namely Bungoma, Busia, Homa Bay, Nandi, Vihiga, Kakamega and Siaya. Leaf samples showing virus-like symptoms were collected and analysed by ELISA. Sixteen popularly grown bean cultivars together with cowpea (Vigna unguiculata), soybean (Glycine max), green grams (Vigna radiata) and groundnut (Arachis hypogaea) were planted in a greenhouse in a completely randomized block design with three replicates. The plants were inoculated with BCMNV isolate at 3-leaf stage. Data were taken weekly for 3 weeks on type of symptoms expressed and number of plants infected. In total, 270 bean farms were visited. Symptoms of mosaic, downward curling, local lesions, stunting or a combination of these were observed during both surveys. Mean virus incidence was higher in the short rain season (50.2%) than in the long rain season (35.6%). The mean BCMD severity on a scale of 0–3 was highest (2.3) in Kakamega County and lowest (0.5) in Siaya. On variety resistance tests to BCMNV isolate, 10 bean cultivars were susceptible, four tolerant and two resistant. BCMNV is widely distributed across counties probably because of use of uncertified seeds by farmers and inoculum pressure from seed and aphid vector. For improved yields of common bean, farmers should be advised to plant certified seeds for all legumes in the cropping system.  相似文献

Eight isolates of Cucumber mosaic virus (CMV) isolated from seven different host species were tested for their virulence on potato cv. Desirée. Three isolates caused a systemic infection, of which one isolate from Asiatic lily (CMV-P26) and one from cucumber (CMV-J) appeared to be highly virulent, in contrast to the third isolate (CMV-M) that originated from cucumber and caused mild symptoms only. These three isolates were transmitted to 26 additional potato cultivars by mechanical inoculation in a greenhouse. All cultivars were infected with at least one CMV isolate and developed local chlorotic symptoms, but only 17 cultivars (including Desiree) developed primary systemic symptoms including necrosis, mosaic and/or malformation of leaves. Furthermore, in only five cultivars (including Desiree) CMV was transmitted to tubers and was subsequently detected in plants of the first and second vegetative progeny, the secondary symptoms of these plants being severe. The observed phenotypic responses of potatoes to CMV were not associated with the maturity type (early or late) or resistance to other viruses. Results of this study indicate a high level of biological variability among CMV isolates and that infection in potato depends on CMV isolate and potato cultivar.  相似文献

Abnormal celery plants (Apium graveolens) with symptoms typical of virus infection were collected from 28 farms in Cambridgeshire, Lancashire, Lincolnshire, and Norfolk; six different diseases were found. The causal viruses from three of these were isolated and partially characterised. Cucumber mosaic virus was quite common, but usually symptomless; it had no effect on the growth of four celery cultivars in field trials. Celery virus 036 was serologically related to western celery mosaic virus, had a host range apparently confined to Umbelliferae, and was very common in the Fens, causing ringspot symptoms in some cultivars and a loss in yield of up to 24%. This disease was much less severe than that caused by an American isolate of western celery mosaic virus in glasshouse comparisons. Celery virus 065 (an unidentified isometric virus, 28–30 nm in diameter) was less common, had a very wide host range, and caused stunting, necrosis, and chlorotic flecking in all cultivars with a loss in yield of up to 45%. Of the remaining three diseases, celery yellow spot was common; the causal agent was transmitted by mechanical inoculation, but very inefficiently, and although yield losses were not determined accurately, they were thought to be insignificant. While plants with symptoms typical of celery yellow net and celery strap leaf diseases were often found, no viruses could be isolated from them, despite repeated attempts.  相似文献

The biological and serological properties of two Japanese barley mild mosaic virus (BaMMV) strains (BaMMV-Kal and BaMMV-Nal) and a German BaMMV strain (BaMMV-M) were compared. Mechanical inoculation experiments showed that these three strains differed from one another in their ability to infect specific barley cultivars. BaMMV-Kal and BaMMV-M caused similar symptoms, but BaMMV-Nal clearly differed from them in its symptoms on some barley cultivars. The three BaMMV strains efficiently infected barley plants at 15°C, whereas at 20°C BaMMV-Kal and BaMMV-M also infected many plants but BaMMV-Nal infected only a few. BaMMV-Kal and BaMMV-M were indistinguishable by ELISA, while BaMMV-Nal was distinguished from both. The biological and serological variability reported shows that BaMMV occurs as two distinct strains in Japan.  相似文献

This study investigated the infection process of Mycosphaerella graminicola and enzyme activities related to reactive oxygen species (ROS) or oxylipin biosynthesis in four French wheat cultivars with variable resistance to M.?graminicola infection. At field level, cultivars Caphorn, Maxyl and Gen11 were susceptible, whereas Capnor showed high levels of quantitative resistance. Moreover, Capnor and Gen11 were tolerant, i.e., their yield was less affected by infection compared to non-tolerant Maxyl and Caphorn. These four cultivars were inoculated under laboratory conditions with the M.?graminicola IPO323 reference strain. Cytological and biochemical responses were studied on collected first plantlet leaves and several features discriminated between cultivars. However, resistance and tolerance had no impact on the fungal infection process. Levels of lipoxygenase (LOX), peroxidase (PO) and glutathione-S-transferase (GST) activities were also compared with regard to cultivar resistance or tolerance to M.?graminicola. LOX, PO and GST activities did not discriminate resistance and tolerance profiles, although a low level of PO in inoculated and non-inoculated plants could be associated with tolerance. In addition, cell necrosis correlated positively with LOX in non-tolerant cultivars, while mycelia surrounding stomata were positively correlated with PO in the resistant cultivar. GST activity presented correlations between cytological and biochemical parameters only for susceptible cultivars. Stomatal and direct penetration were positively correlated with GST activity in the susceptible non-tolerant cultivars, while these correlations were negative in the tolerant cultivar. When combining cytological and biochemical observations with resistance and tolerance profiles, for each cultivar and at each time point, cultivars could be classified in tight accordance with their previous field characterisation. Moreover, tolerance allowed us to distinguish susceptible cultivars when both biochemical and cytological parameters were considered together.  相似文献

Diseases affecting strawberry (Fragaria × ananassa Duch.) have been of major concern in recent years because of their widespread occurrence and potential for yield loss. Anthracnose, caused by the fungus Colletotrichum acutatum, is one of the most serious diseases of strawberry worldwide. Tissue-culture induced (somaclonal) variation provides one strategy for generating disease-resistant genotypes. As part of a program to generate strawberry germplasm resistant to anthracnose, an in vitro screening system was used to evaluate several commercial cultivars, Chandler, Delmarvel, Honeoye, Latestar, Pelican and Sweet Charlie propagated in vitro, and shoots regenerated from leaf explants of these cultivars for resistance to C.␣acutatum isolate Goff (highly virulent). Regenerants with increased levels of resistance were identified from all of the cultivars. The greatest increases in disease resistance were observed for regenerants from leaf explants of cultivars Pelican and Chandler that exhibited 17.5- and 6.2-fold increases in resistance, respectively. The highest levels of anthracnose resistance (2 to 6% leaf necrosis) were exhibited by regenerants from explants of cultivars Pelican and Sweet Charlie. These studies suggest that generating somaclonal variation may be a viable approach to obtaining strawberry plants with increased levels of anthracnose resistance.  相似文献

采用人工接种方法研究了166份黄淮海北部地区近年来生产上种植品种及近期育成品种(系)对SMV的抗性,利用黄淮海地区SMV流行株系SC3、SC6、SC7、SC11以及混合4个株系进行了抗性鉴定,从客观上评价了上述品种(系)的抗性。结果显示:对4个株系均表现抗病(中抗、高抗和免疫)的共82份,占49.4%;其中对3个株系表现高抗或免疫的32份,占19.3%;对4个株系表现高抗或免疫的23份,占13.9%。对混合株系表现抗病的108份,占65.1%。其中表现免疫的45份,占27.1%,表现高抗的29份,占17.5%。对4个株系和混合株系均表现抗病的62份,占37.3%;表现免疫和高抗的14份,占8.4%。近年育成品种对SC3、SC7株系较早期育成品种的抗性显著增强;来自河北、北京、山西的品种抗性较好,病情指数整体较低。鉴定筛选出对接种的4个株系及混合株系均表现免疫的品种冀豆9号和石豆6号,可作为抗病育种的重要抗源。本研究还发现部分品种对接种的4个株系和混合株系表现出抗性差异,表明SMV株系间存在着明显的互作。  相似文献

Six blackberry or hybrid berry cultivars and 19 raspberry cultivars were assessed for their infectibility with, and sensitivity to, graft inoculation with 10 distinct viruses found infecting Rubus in the UK. Cultivars were grafted with each of, two isolates of the pollen borne raspberry bushy dwarf virus (RBDV), five aphid borne viruses: black raspberry necrosis, raspberry leaf mottle (RLMV), raspberry leaf spot (RLSV), rubus yellow net and raspberry vein chlorosis (RVCV); and isolates of the nematode transmitted nepoviruses, arabis mosaic, raspberry ringspot, strawberry latent ringspot and tomato black ring. All tested cultivars were infectible with a resistance breaking isolate of RBDV but only about half of that number with the Scottish type isolate of the virus. The raspberry cvs Autumn Bliss, and occasionally Glen Garry and Glen Prosen, developed leaf yellowing symptoms following infection with RBDV, but none of the other infected cultivars showed obvious leaf symptoms when kept in a heated glasshouse during the growing season. All tested cultivars were infectible with each of the four viruses transmitted in nature by the aphid, Amphorophora idaei. Most were infected symptomlessly, but seven cultivars developed severe leaf spotting symptoms due to infection with RLMV or RLSV. All but one of the raspberry cultivars were infectible with RVCV, which is transmitted in nature by the aphid Aphis idaei, and almost all infected plants developed leaf symptoms; only one of the hybrid berry or blackberry cultivars tested was infected with RVCV. In tests with the four nepoviruses, all tested cultivars, except Tummelberry, were infectible with at least one or more of these viruses. However, cultivars responded differently to challenge inoculation with different isolates of individual nepoviruses. Several cultivars developed chlorotic leaf mottling following infection with some nepovirus isolates. The implications of these results for virus control are discussed in the light of the changing pattern of virus and virus vector incidence in the UK.  相似文献

Eighty soybeans (Glyane max Merr) cultivars, includingx a resistant line Oxley 615 were each inoculated with seven streams of soybean mosaic virus SMV. Susceptible cultivers produced smaller plants with delaved maturity, and reduced seed yield relative to the non inoculared plants Someptible cultivars had.a higher level of morrled seeds and seat transmission ot SMV from the morrled seeds than the resistance line Oxley 615. The SMV strain cultivar interaction was significant for all traits, suggsesting that soybean cultivars should be tested against specific SMV strains to determine their response to this virus.  相似文献

Three strains of Verticillium albo-atrum causing severe wilt of tomato (T), progressive (Hp ) and fluctuating (HF) wilt of hop, were inoculated through the roots of four tomato cultivars at different inoculum concentrations. Symptoms were assessed visually 42 days after inoculation, and quantitatively on the change in total leaf area compared with controls. Distribution of mycelium and tyloses was determined by sections at 2 cm intervals of root, stem and petiole. Cultivars Loran Blood and Moscow showed resistance to disease expression at all levels of inoculum concentration with the T strain. Bonny Best and Potentate were both susceptible to this strain, but whereas in Potentate, disease severity increased from mild to severe with increase in inoculum concentration, Bonny Best was severely diseased at the lowest level of inoculum. All cultivars showed some susceptibility to the HP and HF strains; the ‘resistance’ of Loran Blood and Moscow was no longer apparent and Bonny Best was most severely affected. The relative susceptibilities to the strains were HF Bonney Best > Loran Blood > Potentate > Moscow, HP Bonny Best > Loran Blood, Moscow > Potentate, T Bonny Best > Potentate > Loran Blood, Moscow. In general, vascular colonization was less in the cultivars Loran Blood and Moscow with all three fungal strains at io5propagules/ml level of inoculum, but this was not always correlated with an increase in disease severity. With the exception of the host-pathogen combinations Bonny Best/T, Bonny Best/HF, Potentate/T and Moscow/T, increasing the inoculum concentration to 107propagules/ml increased disease severity but had little or no effect of increasing vascular colonization. In Bonny Best/T, Bonny Best/HF and Potentate/T vascular colonization was reduced with the higher level of inoculum. Moscow showed complete resistance to symptom expression and little vascular colonization with the T strain at 105prop./ml. At 107prop./ml resistance was maintained but there was very extensive growth of mycelium in the vessels. Tylosis resulted from an interaction of host, fungal strain and the level of inoculum and was not always correlated with the degree of vascular colonization. Contrary to previous reports the resistant varieties Loran Blood and Moscow developed acute disease symptoms after inoculation with HP and HF and these were associated with a high level of tylosis rather than mycelial growth. Tylosis and disease severity but not mycelial growth increased with higher levels of inoculum. The results suggested that susceptibility to Verticillium wilt was a complex response depending on host cultivar, fungal strain and the initial inoculum concentration. In some cultivar-pathogen combinations susceptibility was directly proportional to the amount of mycelium present in the vessels, while in others a physiological resistance mechanism independent of the degree of colonization appeared to operate. In a third category, increased disease development rather than resistance was associated with high levels of tylosis.  相似文献

Sugarcane plantlets were sectioned halfway between the base and the youngest ligule and then inoculated by soaking the wound in a suspension of Xanthomonas campestris pv. vasculorum. The infection caused rapid necrosis of the inoculated leaves, chlorosis of uninoculated leaves, or death of the inoculated plantlet. New tillers sometimes showed chlorosis or white streaks. The effects of the inoculum concentration, the cultivar, and the bacterial strain on symptom severity were determined. The ranking of cultivars depended on the inoculum concentration, and strains were found to differ with regard to aggressiveness. However, cultivars and strains were more effectively classified in greenhouse trials. The poor expression of leaf resistance appeared to limit the use of the in vitro test.  相似文献

Seven Azospirillum strains induced more deformation of root hairs of wheat than did strains of Rhizobium leguminosarum, Azotobacter chroococcum, or Escherichia coli. Azospirillum sp. strain Sp245 caused the most deformation. Strain Sp245 (isolated from surface sterile roots of wheat) and strain Sp7 (isolated from the rhizosphere of a forage grass) were compared with regard to their effects on root hair deformation, their attachment to roots, and their effects on the growth of four wheat cultivars. The amount of deformation caused by the two strains in the four cultivars increased in the following order: cv. Tobari, cv. Tonari, cv. BH1146, cv. Lagoa. Strain Sp245 attached to the roots of all cultivars in low numbers, and attachment did not increase with time (up to 48 h). Strain Sp7 attached in higher numbers, and attachment increased with time. Inoculation of the four cultivars of wheat had pronounced effects on root mass measured at maturity. The magnitude of the effects in the four cultivars increased in the following order: Tobari, Tonari, BH1146, Lagoa; these effects were progressively more positive for strain Sp245 and progressively more negative for strain Sp7. Concentrations of N in wheat did not vary substantially between cultivars or strains. Concentrations of K and P did not vary substantially between cultivars but did vary between strains, Sp245 effecting increases and Sp7 effecting decreases.  相似文献

Slow-growing bacteria similar to the bacterium causing lettuce corky root (CR) in California (strain CA1) were isolated from muck soils of Florida, New York, and Wisconsin, using lettuce seedlings as bait. All strains were tested for reaction with polyclonal antibodies produced against strain CA1 and for pathogenicity on CR-susceptible (Salinas) and CR-resistant (Green Lake) lettuce cultivars in a greenhouse. Five strains from Florida, three from New York, and three from Wisconsin induced severe CR symptoms on Salinas and mild symptoms on Green Lake. All strains were gram-negative, aerobic, oxidase positive, and catalase positive and reduced nitrate to ammonia. Whole-cell fatty acid compositions were similar for all strains and resembled that of Pseudomonas paucimobilis. Since this fatty acid pattern is unique, it is suggested that CR of lettuce is caused by strains of the same bacterium in Florida, New York, Wisconsin, and California.  相似文献

Strain-genotype interaction of tobacco mosaic virus in tomato   总被引:1,自引:0,他引:1  
The symptoms and virus content of isogenic tomato genotypes differing by three tobacco mosaic virus (TMV) resistance factors, Tm-I, Tm-2 and Tm-22, were studied in relation to various isolates of TMV and four strains were identified. The common strain induced no symptoms on plants with any of the factors for resistance, one strain caused symptoms on Tm-I plants, one on Tm-2 plants and one on both Tm-I and Tm-2 plants and also on Tm-I Tm-2 plants. No strain induced symptoms on Tm-22 plants. The gene, Tm-I, was found to be dominant or incompletely dominant for preventing symptom development but was recessive or intermediate for limiting virus multiplication of the common strain. Both Tm-2 and Tm-22 were dominant for a hypersensitive response to the common strain. Virus multiplication was temperature-dependent. The background or varietal genotype did not affect virus multiplication. A systemic necrosis of Tm-22 plants occurred only when heterozygous Tm-22 was not protected by other factors against specific strains of TMV. The complexity of the host genotype, pathogen genotype and environment interactions are outlined and the exploitation of the resistance factors in tomato breeding discussed.  相似文献

Summary Three field experiments were conducted on ten cultivars of winterwheat and four cultivars of springwheat to estimate the growth promoting effect ofAzospirillum brasilense under varying nitrogen doses. Independent of cultivar selection or nitrogen dose a highly significant yield increase could be observed in winterwheat: strains S631 and SpBr14 increased the average grain yield with 9.14% and 14.82% respectively. When the yield components were studied a coinciding increase in ear density could be demonstrated of resp. 10.57% and 13.55%. Less significant results were obtained with springwheat although in one experiment strain SpBr14 significantly increased grain yield. As with winterwheat tillering of the plant was markedly affected by inoculation with both strains. In a companion greenhouse experiment it was found that inoculation with Azospirillum can cause a decrease in the root mass of wheatplants except when strain SpBr14 is used. Therefore it is suggested that the presence of a higher tillering together with an undisturbed nutrient uptake capacity can result in yield increases after inoculation withAzospirillum brasilense.  相似文献

Plants of several potato clones with major gene resistance to potato virus Y (PVY) developed necrotic local lesions and systemic necrosis after manual inoculation with common (PVYo) or veinal necrosis (PVYN) strains of the virus. The clones reacted similarly, although their resistance genes are thought to be derived from four different wild species of Solarium. Mesophyll protoplasts from each clone became infected when inoculated with RNA of PVYo by the polyethylene glycol method. The proportion of protoplasts infected, assessed by staining with fluorescent antibody to virus particles, was similar to that of protoplasts of susceptible potato cultivars. In contrast, plants of potato cultivars Corine and Pirola, which possess gene Ry from S. stoloniferum, developed few or no symptoms when manually inoculated or grafted with PVYo. Moreover, only very few protoplasts of these cultivars produced virus particle antigen after inoculation with PVYo RNA. The extreme resistance to PVY of cvs Corine and Pirola was therefore expressed by inoculated protoplasts whereas the resistance of the necrotic-reacting potato clones was not.  相似文献
