Oyster populations periodically exposed to runoff from acid sulfate soils (ASS) are of depressed abundance and have fewer smaller individuals than unaffected populations, despite having similar recruitment levels to unaffected sites during dry periods. We examined how the timing and duration of exposure to ASS runoff influences the growth and survival of successfully settled oysters. We predicted that among 6-month-old oysters, growth and survival would be (1) lower among individuals continuously exposed to ASS-acidified waters than those that are episodically exposed, and (2) most negatively affected during rainfall events, which enhance transport of ASS runoff to estuaries. Six-month-old Sydney rock oysters, Saccostrea glomerata, were deployed at ASS-affected and unaffected sites within each of two south-east Australian estuaries. After 10 weeks, oysters were transplanted within and across sites in an estuary and maintained in situ for another 10 weeks. Oysters that remained for 20 weeks at ASS-affected sites grew at just over half the rate of oysters at reference sites. Oysters transplanted from acidified to reference sites grew more than oysters transplanted from reference to acidified sites or oysters that remained at reference sites. Unexpectedly, overall oyster mortality was low. Greater rainfall, and hence a lower pH, is likely to have accounted for the greater impact of acidification on growth during the second 10 weeks. Where oysters recruit to a 6-month age cohort, they may be able to tolerate subsequent, moderate, acidification events. Reduced growth during acidification periods may be offset by positive growth during intervening dry periods.  相似文献

Climate warming and low pH environment are known to negatively impact all levels of aquatic organism from cellular to organism and population levels. For ammonotelic freshwater species, any abiotic factor fluctuation will cause disturbance to the fish, specifically at the gills which act as a multifunctional organ to support all biological processes. Therefore, this study was designed to investigate the effect of temperature (28 vs. 32°C) and pH (7.0 vs. 5.0) stress on the gill plasticity of Hoven's carp after 20 days of continuous exposure. The results demonstrated that high temperature and low pH caused severe changes on the primary and secondary lamellae as well as the cells within lamellae. An increasing trend of the proportion available for gas exchange was noticed at high temperature in both pH exposures, which resulted from a reduction of the primary lamellae width with elongated and thinner secondary lamellae compared to fishes at ambient temperature. Following exposure to high temperature and acidic pH, Hoven's carp experienced gill modifications including aneurysm, oedema, hypertrophy, curling of secondary lamellae, epithelial lifting, hyperplasia and lamellae fusion. These modifications are indicators of the coping mechanism of Hoven's carp to the changing environment in order to survive.  相似文献

1. The euryhaline fish Fundulus heteroclitus has an incipient lethal pH between 3.75 and 4.0 in fresh water.2. Fish exposed to pH 3.5 in sea water or fresh water died in about 3 hr, and had greatly elevated or depressed body sodium concentrations, respectively. The direction and degree of change in body sodium level depended on the sodium diffusion gradient between the environment and the fish. This is the first time that the death of fish in sea water at low pH has been shown to be associated with hypernatremia.3. Yet, sodium fluxes during the first hour of exposure to pH 3.5 in water of 3.5 or 35 ppt salinity were not different from controls, and body and plasma sodium concentration did not change during 2hr exposure to pH 3.5. This initial insensitivity of gill sodium regulation to blockage by low pH is quite different from the response of previously studied freshwater fish.4. The degree of acid tolerance displayed by F. heteroclitus is surprising considering its estuarine habits. This paradoxical tolerance appears to be a secondary consequence of its ability to adjust sodium balance in relation to rapid changes in salinity.  相似文献

This study investigated the relationship between level of stress in middle and high school students aged 12–18 and risk of atopic dermatitis. Data from the Sixth Korea Youth Risk Behavior Web-based Survey (KYRBWS-VI), a cross-sectional study among 74,980 students in 800 middle schools and high schools with a response rate of 97.7%, were analyzed. Ordinal logistic regression analyses were conducted to determine the relationship between stress and atopic dermatitis with severity. A total of 5,550 boys and 6,964 girls reported having been diagnosed with atopic dermatitis. Younger students were more likely to have atopic dermatitis. Interestingly, the educational level of parents was found to be associated with having atopic dermatitis and having more severe condition. In particular, girls with mothers with at least college education had a 41% higher risk of having atopic dermatitis and severe atopic condition (odds ratio (OR)) = 1.41, 95% CI, 1.22–1.63; P<0.0001) compared with those with mothers who had attended middle school at most. Similar trend was shown among both boys and girls for their father''s education level. The stress level was found to be significantly associated with the risk of atopic dermatitis. Compared to boys with who reported “no stress”, boys with “very high” stress had 46% higher the risk of having more severe atopic dermatitis (OR = 1.46, 95% CI, 1.20–1.78; P<0.0001), 44% higher (OR = 1.44, 95% CI, 1.19–1.73; P<0.0001) with “high” stress, and 21% higher (OR = 1.21, 95% CI, 1.00–1.45; P = 0.05) with “moderate” stress. In contrast, we found no statistically significant relationship between stress and atopic dermatitis in girls. This study suggests that stress and parents'' education level were associated with atopic dermatitis. Specifically, degree of stress is positively correlated with likelihood of being diagnosed with this condition and increasing the severity.  相似文献

Three patients with elevated blood lactate values are described. The first, despite moderate hyperlactatemia of 5.3 mEq./1. and severe acidosis with an arterial blood pH of 6.98, had no “excess lactate”. In a second patient, moderate acidosis with a pH of 7.27 and blood lactate of 7.5 mEq./1., of which 33% was excess lactate, was found to be secondary to tissue hypoxia on an ischemic basis and preceded the onset of clinical shock by four hours. A third patient, diabetic and under treatment with phenformin hydrochloride, presented with many features suggestive of pulmonary embolism, including marked pulmonary hypertension. A diagnosis of idiopathic lactic acidosis was established when the arterial blood pH was found to be 6.77 and a blood lactate value of 14.2 mEq./1., 60% as excess lactate, was discovered in the absence of a demonstrable cause of tissue hypoxia. Exploration of the pulmonary vascular bed showed no sign of mechanical blockage. The diagnostic, therapeutic and prognostic value of measuring blood lactic acid, and of quantitating the proportion circulating as “excess lactate”, is emphasized.  相似文献

Ammonium-oxidizing bacteria were examined in two wetland environments, a freshwater marsh and an estuarine bay, during a 2-year period. Two predominant types were consistently isolated, one from each environment. Both isolates were identified as species ofNitrosomonas. Using a closed culture, high cell density assay, the effects of temperature, pH, salinity, Na+, K+, nitrite, nitrate, and ammonium concentrations on ammonium oxidation were determined. Maximum activity was observed for the freshwater isolate at 35°C, pH 8.5, salinities of 0.3 to 0.5% Na+ and K+, and ammonium concentrations greater than 0.5 g/l. For the estuarine isolate, maximum activity was observed at 40°C, pH 8.0, salinities of 0.5 to 1.0%, 1.0% Na+ and K+, and 0.2 g/l ammonium. The estuarine isolate had a Na+ requirement which could be partially substituted by the K+, suggesting that the organism is a true estuarine bacterium. Nitrite inhibited both isolates at concentrations greater than 5 mg/l, whereas nitrate had no significant effect on either isolate.  相似文献

Introduction

The aims of this present study were firstly to assess the outcome, including functional course, in anti-Jo1 positive patients with antisynthetase syndrome (ASS), and secondly to determine predictive parameters of poor outcome in these patients.

Methods

The medical records of 86 consecutive anti-Jo1 patients with ASS were reviewed in 4 academic centers.

Results

13 patients (15.1%) achieved remission of ASS, whereas 55 (63.9%) improved and 18 (20.9%) deteriorated in their clinical status. Both steroid and cytotoxic drugs could be discontinued in only 4.7% of patients. ASS was associated with decreased quality of life at long-term follow-up: only 69.2% of patients considered to be in remission experienced a return to previous normal activities; and 24.7% of other patients with non-remitting ASS still had a marked reduction of activities (as shown by the disability scale of the Health Assessment Questionnaire). Decreased quality of life was further due to calcinosis cutis (8.1%) and adverse effects of steroid therapy (36%). Factors associated with ASS deterioration were older age, pulmonary and esophageal involvement, calcinosis cutis and cancer. Higher anti-Jo1 levels were further associated with disease severity in ASS patients.

Conclusions

The present study shows high morbidity related to ASS. Furthermore, we suggest that patients with predictive factors of ASS deterioration may require more aggressive therapy. Our findings also suggest that in anti-Jo1 patients with severe esophageal manifestations, combined high dose steroids and intravenous immunoglobulins might be proposed as the first line therapy. Finally, as cancer occurred in 14% of anti-Jo1 patients, our findings underscore that the search for cancer should be performed in these patients.  相似文献

The acidophilic red alga Cyanidium sp. is one of the dominant mat-forming species in the highly acidic waters of Río Tinto, Spain. The culture of Cyanidium sp., isolated from a microbial mat sample collected at Río Tinto, was exposed to 9 different pH conditions in a gradient from 0.5 to 5 for 24?h and its physiological status evaluated by variable chlorophyll a fluorescence kinetics measurements. Maximum quantum yield was determined after 30?min, 1?h, 2?h, 4?h, 6?h and 24?h of exposure after 15?min dark adaptation. The effect of pH on photochemical activity of Cyanidium sp. was observable as early as 30?min after exposure and the pattern remained stable or with only minor modifications for 24?h. The optimum pH ranged from 1.5 to 2.5. A steep decrease of the photochemical activity was observed at pH below 1 even after 30?min of exposure. Although the alga had tolerated the exposure to pH?=?1 for at least 6?h, longer (24?h) exposure resulted in reduction of the photochemical activity. At pH above 2.5, the decline was more moderate and its negative effect on photochemistry was less severe. According to the fluorescence measurements, the red alga Cyanidium sp. is well-adapted to prevailing pH at its original locality at Río Tinto, i.e. pH of 1 to 3. Introduction

Obesity in pregnancy is associated with increased risk of complications and adverse outcomes in mother and child. Childhood adverse experiences are known to have numerous negative physical and emotional sequelae. We aimed to examine if exposure to abuse and/or neglect in childhood increased the likelihood of pre-pregnancy obesity.

Methods

Demographic and clinical data including weight, height, mental health as measured by the General Health Questionnaire and exposure to childhood trauma as measured by the childhood trauma questionnaire was collected from 239 women attending antenatal care at an Australian tertiary hospital.

Results

More than one quarter of women were obese prior to pregnancy and approximately 20% of women self reported experiencing moderate to severe physical, sexual or emotional abuse. Almost 60% of women scored in the clinical range on the GHQ. Pre-pregnancy obesity in women attending antenatal care was associated with a self-reported history of emotional or physical abuse with those exposed to moderate or severe emotional or physical abuse having increased odds of being obese prior to pregnancy (O.R. and 95% CI: 2.40; 1.19–4.84 and 2.38; 1.18–4.79 respectively). There was no significant association between other forms of childhood maltreatment, demographic or current mental health status and pre-pregnancy obesity.

Conclusions

The high rates of obesity, mental health problems and self reported childhood maltreatment in the Australian antenatal population are serious public health concerns due to the extra health risks conferred on mother and offspring. Exposure to physical or emotional abuse during childhood increases the likelihood of obesity in women attending antenatal care. Further research is required to determine reasons for this association.  相似文献

We examined the genetic and physiological characteristics of chitin degrading enzymes expressed by fosmids cloned from two strains of chitinolytic gammaproteobacteria isolated from alkaline, hypersaline Mono Lake, California; and from a metagenomic library derived from an estuarine bacterial community (Dean Creek, Sapelo Island, GA, USA). The Mono Lake chitinolytic enzymes presented unique adaptations in terms of halo- and alkalitolerance. The sequence from one of the Mono Lake isolates (strain 12A) was a conventional family 18 glycosyl hydrolase; however, the expressed protein had a novel secondary activity peak at pH 10. We obtained a novel family 20 glycosyl hydrolase sequence from Mono Lake strain AI21. The activity of the expressed protein had a pH optimum of 10, several pH units higher than any other enzyme currently assigned to this family, and the enzyme retained 80% of its activity at pH 11. The enzyme was also halotolerant, retaining activity in salt solutions of up to 225 g l(-1). Sequence analysis indicated a molecular weight of approximately 90 kDa for the protein, and that it contained two active sites. Culture supernatant contained two chitinolytic proteins, 45 and 31 kDa, suggesting possible post-expression modification of the gene product. In contrast, the sequence found in the estuarine metagenomic library and the functional characteristics of the protein expressed from it were those of a conventional family 18 glycosyl hydrolase.  相似文献

Contagious mucocutaneous dermatitis is a frequently encountered disease of mountain hares (Lepidus timidus) in Finland. We describe the histopathologic changes and propose an etiologic cause for this disorder. Fifty-three cases collected during 1982-2000 were examined histologically. Transmission electron microscopy was performed in one case. In fully developed lesions, keratinocytes in epidermis and follicular infundibula were swollen and contained large eosinophilic intracytoplasmic inclusion bodies with marked reticular and ballooning degeneration. In later stages, there was marked necrosis and ulceration with severe pyogranulomatous and suppurative inflammation. At this stage, no viral inclusions were detectable, but secondary Staphylococcus warnerii infection was present in most cases. In late lesions, there was dermal fibrosis with epidermal hyperplasia. No spiral-shaped bacteria suggesting treponematosis were detected at any stage. Ultrastructurally, swollen epidermal and follicle infundibular cells contained round intracytoplasmic inclusion bodies with a myriad of virions typical of poxvirus with a biconcave nucleocapsid core, two lateral bodies, and a clearly discernible outer lipoprotein capsule. The findings suggest that contagious mucocutaneous dermatitis in mountain hares is a viral disease caused by a poxvirus. The disease is often complicated by secondary bacterial infection, most commonly S. warneri.  相似文献

A 51-year-old woman with a 20-year history of severe hypertension and target organ damage had nondiuretic hypokalemia, kaluresis, suppressed plasma renin activity and elevated urinary excretion of aldosterone. Renal arteriography demonstrated unilateral renal artery stenosis secondary to fibromuscular hyperplasia. Blood pressure responded only minimally to almost all antihypertensive agents. The horse mussel Atrina zelandica (Gray) is a large, suspension feeding pinnid bivalve, common in coastal and estuarine areas of northern New Zealand. As a suspension feeder, Atrina is likely to be influenced by suspended sediment loads. We conducted a laboratory experiment to determine the effect of short-term elevations in turbidity levels, such as those commonly recorded during storms, on the physiological condition and clearance rates of Atrina. We also conducted a field survey and a 3-month transplant experiment at multiple sites along a gradient of increasing suspended sediment load in a New Zealand estuary. Laboratory clearance rates of Atrina declined above a threshold suspended sediment concentration, and Atrina physiological condition at the end of this experiment was lower in high cf. low turbidity treatments. Decreases in Atrina condition were detected after exposure to elevated levels for only 3 days. The field survey and transplant experiment provided empirical evidence of a strong, negative effect of increasing suspended sediment flux on the physiological condition of Atrina. We suggest that relationships between the physiological condition of suspension feeders and sediment settling flux could provide a link between sediment inputs, which commonly occur as a result of catchment runoff during rainfall events, and the ecological health of estuarine and shallow coastal areas. Our study also demonstrated that Atrina have a natural distribution limit controlled by suspended sediment load. Thus, there is potential for larger-scale functional and structural effects on benthic communities in estuarine and coastal areas with high rates of sedimentation.  相似文献

Synopsis The objective was to determine the effects of acid and aluminum in acidified hard and soft water on the histology and morphometry of rainbow trout gills, and to determine relevant toxicity indicators within the gill tissue. Acid and aluminum promoted measurable primary epithelial hyperplasia which proved to be a reliable biological indicator of acid and aluminum contamination and possibly of some predictive value. Low levels of aluminum and acid resulted in hypertrophied chloride cells, suggesting a role in adapting to the contaminants. High concentrations of aluminum (>10 molI-1) caused chloride cell necrosis and consequently a decline in cell numbers over time. Aluminum precipitates accumulating within the chloride cell cytoplasm probably lead to impaired function prior to cell degeneration. The morphological alterations resulted in a decrease in water space between secondary lamellae (up to 40% within 14 d) which may reduce the efficiency of gas exchange. Twice the aluminum was required in hard water to elicit a similar soft water tissue response. Pathological changes were more severe with aluminum at pH 5.2 than at pH 4.7; results of aluminum speciation suggest that both labile and non-labile fractions are responsible for the induction of gill lesions. Low levels of aluminum may protect fish from the effects of high hydrogen ion concentration.  相似文献

Reactive oxygen and nitrogen species (ROS and RNS) produced by the phagocytic cells are the most common arsenals used to kill the intracellular pathogens. However, Leishmania, an intracellular pathogen, has evolved mechanisms to survive by counterbalancing the toxic oxygen metabolites produced during infection. Polyamines, the major contributor in this anti-oxidant machinery, are largely dependent on the availability of L-arginine in the intracellular milieu. Argininosuccinate synthase (ASS) plays an important role as the rate-limiting step required for converting L-citrulline to argininosuccinate to provide arginine for an assortment of metabolic processes. Leishmania produce an active ASS enzyme, yet it has an incomplete urea cycle as it lacks an argininosuccinate lyase (ASL). There is no evidence for endogenous synthesis of L-arginine in Leishmania, which suggests that these parasites salvage L-arginine from extracellular milieu and makes the biological function of ASS and the production of argininosuccinate in Leishmania unclear. Our previous quantitative proteomic analysis of Leishmania promastigotes treated with sub-lethal doses of ROS, RNS, or a combination of both, led to the identification of several differentially expressed proteins which included ASS. To assess the involvement of ASS in stress management, a mutant cell line with greatly reduced ASS activity was created by a double-targeted gene replacement strategy in L. donovani promastigote. Interestingly, LdASS is encoded by three copies of allele, but Western blot analysis showed the third allele did not appear to express ASS. The free thiol levels in the mutant LdASS-/-/+ cell line were decreased. Furthermore, the cell viability in L-arginine depleted medium was greatly attenuated on exposure to different stress environments and was adversely impacted in its ability to infect mice. These findings suggest that ASS is important for Leishmania donovani to counterbalance the stressed environments encountered during infection and can be targeted for chemotherapeutic purpose to treat visceral leishmaniasis.  相似文献

Previous studies have shown that isolates of Chattonella marina from Australia and Japan exhibit differences in tolerance to high intensities of visible light. Here we show that the Australian strain of C. marina produces around five times more UV-absorbing mycosporine amino acids (MAAs) than the Japanese strain. This corresponds with 66% increased growth by the Australian strain under UVB exposure compared to no UV exposure. The MAA mycosporine-glycine, which reportedly acts as an antioxidant, was found in high quantity (110 fg cell-1) in the Australian but was absent in the Japanese strain. In contrast, changes in the concentration of violaxanthin and zeaxanthin per cell were 4.7-4.8 times greater in the Japanese relative to the Australian strain suggesting that the Japanese strain uses a xanthophyll cycle to moderate inhibition by high photosynthetically active radiation (PAR) irradiance. Increased MAA production under high irradiance was also observed in other Australian strains of Chattonella, but not noted in other Japanese strains suggesting ecophenotypic adaptation due to differing environmental conditions.  相似文献

Planktonic larvae of estuarine crabs are commonly exported to the continental shelf for development and then return to coastal and estuarine areas as postlarvae (megalopae). Megalopae returning to estuaries must be adapted to survive in brackish water whereas those of coastally distributed species should not need such adaptations. We investigated 1) whether megalopae of the estuarine crab Callinectes sapidus and the coastal crab Callinectes similis undergo changes in salinity tolerance upon entry into an estuary and 2) what factors induce those changes. Megalopae were collected at a coastal site and a nearby estuarine site and exposed to a range of salinities (5, 10, 15, 20 and 30) for 6 h. Percent survival was determined after 24 h reintroduction to the collection site water. We also investigated 1) whether increased salinity tolerance was induced by reduced salinity or estuarine chemical cues, 2) the time to acclimation and 3) the salinity necessary for acclimation. C. sapidus megalopae from the estuarine site were more likely to survive exposure to low salinities than those from the coastal site. C. sapidus megalopae from the coastal site exhibited increased survival after acclimation to salinities of 27 and 23 for 12 h. Estuarine chemical cues had no effect on salinity tolerance. C. similis megalopae were less likely to survive at low salinities and did not exhibit an acclimation response upon exposure to reduced salinities. These results suggest that megalopae of C. sapidus are physiologically adapted to recruit to estuaries whereas megalopae of C. similis are unable to acclimate to low salinity conditions.  相似文献

There is now considerable evidence that, as the climate continues to warm, bushfires are becoming more common and severe, particularly in regions such as south-eastern Australia. The extraordinary Australian bushfires over the summer of 2019/2020 resulted in the burning of habitats such as highland peat swamps and intertidal estuarine wetlands over unprecedented spatial scales. Across New South Wales, these bushfires affected 183 ha of saltmarshes and 23 ha of mangroves in 19 estuaries. The percentage of fire-affected saltmarsh ranged from 51% to 81% in the worst impacted estuaries, although typically ≤15% of mapped saltmarsh was damaged. Just over 50% of mangroves were burnt in Wonboyn Lake (although this constituted <0.2 ha), whereas in all other estuaries, ≤5% of mangroves were burnt. At the state-wide scale, the likelihood of saltmarshes being affected by fire was unrelated to adjacent terrestrial vegetation; however, mangroves adjacent to burnt wet sclerophyll forest were more likely to burn than not. Burnt mangroves were almost exclusively associated with extreme or high severity fires in adjacent terrestrial vegetation, yet saltmarshes were also impacted in some cases by moderate or low-intensity fires. Many species of saltmarsh plants had re-sprouted or germinated after 6–24 months, but the extent of any recovery or changes in species composition were not quantified. The majority of fire-affected mangrove trees appeared to be dead 24 months after the fires, despite observations of epicormic growth on some trees after six months. Bushfire impacts to estuarine wetlands are likely to become more frequent and results from our work can help target hazard reduction burning that might be considered for minimising damage to mangroves. More work is required to better understand potential longer term impacts and the capacity for natural recovery of estuarine wetlands from bushfires.  相似文献

Guinea pigs with genetically determined resistance or susceptibility to infection with the nematode parasite Trichostrongylus colubriformis were allowed to become infected with the sarcoptid mite Trixacarus caviae. Compared with nematode-susceptible guinea pigs, nematode-resistant animals had larger populations of mites and developed a more severe dermatitis, with greater mast cell hyperplasia and many more infiltrating eosinophils. The results suggest that animals bred for resistance to one parasite may have greater susceptibility to other parasites.  相似文献
