The neutral carotenoids of wild-type Neurospora crassa and of carotenoid mutants at four discrete genetic loci were isolated using gradient elution chromatography on deactivated alumina columns. Carotenoids were identified by absorption spectrophotometry and thin layer cochromatography with carotenoid standards. Phytoene, phytofluene, -carotene, -carotene, neurosporene, torulene, lycopene, and 3,4-dehydrolycopene were isolated from wild type. Phytoene, phytofluene, -carotene, -carotene, neurosporene, -carotene, lycopene, and one unknown carotenoid, tentatively identified as 15,15-cis--carotene, were isolated from a yellow mutant, ylo-1. ylo-1 also contained residual carotenoids having similar absorption spectra to, but very different chromatographic behavior from, phytofluene, -carotene, -carotene, and lycopene. Albino and colored al-1 mutants contained large amounts of phytoene and only traces of other neutral carotenoids. Albino al-2 and al-3 mutants contained only traces of neutral carotenoids.  相似文献

The pigment composition of the light-harvesting complexes of Photosystem II (LHC II) has been determined for lettuce (Lactuca sativa). In common with other members of the composite, the photosynthetic tissues of this species may contain large amounts of the carotenoid lactucaxanthin (, -carotene-3,3'-diol) in addition to their normal compliment of carotenoids. The occurrence and distribution of lactucaxanthin in LHC II has been examined using isoelectric focusing of BBY particles followed by reversed-phase HPLC analysis of the pigments. The major carotenoids detected in LHC IIb, LHC IIa (CP29) and LHC IIc (CP26) purified from dark-adapted lettuce were lutein, violaxanthin, neoxanthin and lactucaxanthin. Lactucaxanthin has been shown to be a major component of PS II, accounting for 26% of total xanthophyll in both LHC IIb (23% total xanthophyll) and in the minor complexes (12–16%). In this study, LHC IIb was clearly resolved into four bands and their carotenoid composition determined. These four bands proved to be very similar in their pigment content and composition, although the relative amounts of neoxanthin and lutein in particular were found to increase from bands 1 to 4 (i.e. with increasing electrophoretic mobility). The operation of the xanthophyll cycle has also been examined in the LHC of L. sativa following light treatment. The conversion efficiency for violaxanthinzeaxanthin was nearly identical for each light-harvesting complex examined at 58–61%. Summary A Thiococcus sp. nov. gen., the first Thiorhodaceae known to contain bacteriochlorophyll b, was isolated from river mud using filtered light above 900 m as the selecting agent in enrichments. The orange-brown colour of the immotile, Gram-negative, photolithotrophic cocci is due to two new carotenoids, 3,4,3,4-tetrahydrospirilloxanthin and 3,4,3,4-tetrahydrospirilloxanthinal. The photosynthetic pigments appear to be associated with a unique internal membrane system consisting of branched membrane tubes of even diameter (approx. 450 Å) continuous with the cell membrane. The negatively stained tubes examined in the electron microscope reveal a surface fine structure of raised bodies arranged in a regular hexagonal pattern.Dedicated to Professor Dr. C. Three light intensity-dependent Chl b-deficient mutants, two in wheat and one in barley, were analyzed for their xanthophyll cycle carotenoids and Chl fluorescence characteristics under two different growth PFDs (30 versus 600 mol photons·m–2 s–1 incident light). Mutants grown under low light possessed lower levels of total Chls and carotenoids per unit leaf area compared to wild type plants, but the relative proportions of the two did not vary markedly between strains. In contrast, mutants grown under high light had much lower levels of Chl, leading to markedly greater carotenoid to Chl ratios in the mutants when compared to wild type. Under low light conditions the carotenoids of the xanthophyll cycle comprised approximately 15% of the total carotenoids in all strains; under high light the xanthophyll cycle pool increased to over 30% of the total carotenoids in wild type plants and to over 50% of the total carotenoids in the three mutant strains. Whereas the xanthophyll cycle remained fairly epoxidized in all plants grown under low light, plants grown under high light exhibited a considerable degree of conversion of the xanthophyll cycle into antheraxanthin and zeaxanthin during the diurnal cycle, with almost complete conversion (over 90%) occurring only in the mutants. 50 to 95% of the xanthophyll cycle was retained as antheraxanthin and zeaxanthin overnight in these mutants which also exhibited sustained depressions in PS II photochemical efficiency (Fv/Fm), which may have resulted from a sustained high level of photoprotective energy dissipation activity. Two cultures, a yeast (Rhodorula rubra GED8) and a yogurt starter (Lactobacillus bulgaricus 2–11+Streptococcus thermophilus 15HA), were selected for associated growth in whey ultrafiltrate (WU) and active synthesis of carotenoids. In associated cultivation with the yogurt culture L bulgaricus 2–11+S. thermophilus 15HA under intensive aeration (1.3 l–1min–1 air-flow rate) in WU (45 g lactose l–1), initial pH 5.5, 30 °C, the lactose-negative strain R. rubra GED8 synthesized large amounts of carotenoids (13.09 mg l–1 culture fluid). The carotenoid yield was approximately two-fold higher in association with a mixed yogurt culture than in association with pure yogurt bacteria. The major carotenoid pigments comprising the total carotenoids were -carotene (50%), torulene (12.3%) and torularhodin (35.2%). Carotenoids with a high -carotene content were produced by the microbial association 36 h earlier than by Rhodotorula yeast species. No significant differences were notd in the ratio between the pigments synthesized by R. rubra GED8+L. bulgaricus 2–11, R. rubra GED8+S. thermophilus 15HA, and R.rubra GED8+yogurt culture, despite the fact that the total carotenoid concentrations were lower in the mixed cultures with pure yogurt bacteria.  相似文献

A new purple nonsulfur bacterium was isolated from enrichment cultures of a sulfide-containing marine lagoon. The bacterium is similar to Rhodopseudomonas capsulata and is described as a new species of the genus Rhodopseudomonas: Rhodopseudomonas adriatica. Cells are non-motile, 0.5–0.8 m by 1.3–1.8 m, and multiply by binary fission. Intracytoplasmic membranes are of the vesicular type. The photosynthetic pigments are bacteriochlorophyll a and carotenoids of the spheroidene group. Growth is possible anaerobically in the light and at low pO2 in the dark. Biotin and thiamine are required as growth factors. A wide variety of organic compounds, as well as sulfide and thiosulfate, are used as photosynthetic electron donors. Sulfide is oxidized to elemental sulfur, which is subsequently converted to sulfate, whereas thiosulfate oxidation occurs without measurable intermediate. Rhodopseudomonas adriatica is unable to assimilate sulfate, growth is only possible in the presence of a reduced sulfur compound.  相似文献

Summary The two main carotenoids of Stigmatella aurantiaca were identified as 1,2-dihydro-1-hydroxy-3,4-dehydro-torulene glucoside (myxobactin) and 1,2-dihydro-1-hydroxy-4-keto-torulene glucoside (myxobacton). Both pigments occur as monoesters of various fatty acids. The structural formulas were established by chemical and chromatographical analysis and by visible, infrared, and mass spectroscopy.  相似文献

Summary A method for quantitative evaluation of the photokinetic action in Cyanophyceae is described. The value of the photokinetic action, i.e. the quotient of distances covered in light and dark, was determined forPhormidium autumnale in white and monochromatic light. From 0.02 to 30000 lux the photokinetic reaction ofPh. autumnale is positive, above 30000 lux negative. The zero threshold of photokinesis is at 0.01–0.02 lux, the optimum at 2000 lux.By means of an interference filter monochromator system the action spectrum of photokinesis was determined for the range between 317 and 800 m under conditions of equal quantum flux related to an energy of 1000 erg/cm2·sec at 670 m. These results were compared with the pigment absorptionin vitro andin vivo.The main maximum at 680 m and the smaller peak at 620 m coincide with the absorption maxima of chlorophyll a in the living cell. The region 430–440 m was also active without a distinct maximum. Irradiation of the principal absorption region of the carotenoids and phycobilins is only slightly effective for the latter. The distinct maximum at 390 m and the strong action of UV and blueviolet light must be due to an unknown pigment.Thus visible irradiation is photokinetically effective according to its absorption by chlorophyll a, whereas no or little relation exists to carotenoids and phycobilins.

About 20 different carotenoids were found in a strictly aerobic photosynthetic bacterium, Erythrobacter longus. All the carotenoids except the highly polar ones were identified as C40-skeletal carotenoids, which could be devided into three groups: (1) bicyclic carotenoids: -carotene and its hydroxyl derivatives; -cryptoxanthin, zeaxanthin, caloxanthin and nostoxanthin, (2) monocyclic carotenoids: rubixanthin, bacteriorubixanthin and bacteriorubixanthinal, which was a unique cross-conjugated carotenal, and (3) acyclic carotenoids: anhydrorhodovibrin and spirilloxanthin. Bacteriorubixanthinal and zeaxanthin were the major components. (3R)-3-Hydroxy--ionone has rarely been found in carotenoids of purple photosynthetic bacteria, while the acyclic carotenoids have been found exclusively in photosynthetic bacteria. Summary Cell differentiation in Anabaena cylindrica is accompanied with characteristic changes in the pigment composition of heterocysts and spores. In both the absence of phycocyanin is consistent with the lack of CO2-fixing ability previously reported. The presence of chlorophyll and -carotene suggests a functional photosystem I in heterocysts. In the spores chlorophyll is largely replaced by pheophytin. The quantitative distribution of carotenoids is also affected. An increase in the proportion of -carotene is characteristic of heterocysts while spores show a larger proportion of xanthophylls compared with the intact filament.  相似文献

Summary The influence of allelic state of gene B on skin pigmentation in two cultivars of Cucurbita pepo L. has been studied. Total carotenoids were lower at early stages of fruit development in cultivar (cv.) Early Prolific (EP) BB YY fruit skin, than in EP B + B + YY fruit skin, but no differences were observed in total skin carotenoids twenty days after anthesis. Total carotenoids were lower in cv. Fordhook Zucchini (FZ) BB yy fruit skin, than in FZ B + B + yy fruit skin at all developmental stages from anthesis to maturity. Both green and yellow tissues contained typical foliar carotenoids. The carotenoids from yellow fruit skin of both EP genotypes and of FZ BB were characterized by a low carotene: xanthophyll ratio, with a high proportion of the xanthophylls esterified to fatty acids. The xanthophylls of the yellow tissues were esterified with 120, 140, 160 fatty acids. The carotenoids from the green fruit skin of FZ B + B + had a higher percentage of carotenes (primarily -carotene) and a lower percentage of esterified xanthophylls. Spectral shapes of carotenoid fractions from all yellow tissues were similar and distinguishable from those of green FZ B + B + tissue. Carotenoid accumulation in the cotyledons of the mustard seedling (Sinapis alba L.) is controlled by light. Besides the stimulatory function of phytochrome in carotenogenesis the experiments reveal the significance of chlorophyll accumulation for the accumulation of larger amounts of acrotenoids. A specific blue light effect was not found. 4-O-Glycosylation of 2-azidoethyl 2,3,6-tri-O-benzyl-4-O-(2,3-di-O-benzyl-6-O-benzoyl--D-galactopyranosyl)--D-glucopyranoside with a disaccharide donor, 4-trichloroacetamidophenyl 4,6-di-O-acetyl-2-deoxy-3-O-(2,3,4,6-tetra-O-acetyl--D-galactopyranosyl)-1-thio-2-trichloroacetamido--D-galactopyranoside, in dichloromethane in the presence of N-iodosuccinimide and trifluoromethanesulfonic acid resulted in a tetrasaccharide, 2-azidoethyl (2,3,4,6-tetra-O-acetyl--D-galactopyranosyl)-(1 3)-(4,6-di-O-acetyl-2-deoxy-2-trichloroacetamido--D-galactopyranosyl)-(1 4)-(2,3-di-O-benzyl-6-O-benzoyl--D-galactopyranosyl)-(1 4)-2,3,6-tri-O-benzyl--D-glucopyranoside, in 69% yield. The complete removal of O-protecting groups in the tetrasaccharide, the replacement of N-trichloroacetyl by N-acetyl group, and the reduction of the aglycone azide group to amine led to the target aminoethyl glycoside of -D-Gal-(1 3)--D-GalNAc-(1 4)--D-Gal-(1 4)--D-Glc-OCH2CH2NH2 containing the oligosaccharide chain of asialo-GM1 ganglioside in 72% overall yield. Selective 3-O-glycosylation of 2-azidoethyl 2,3,6-tri-O-benzyl-4-O-(2,6-di-O-benzyl--D-galactopyranosyl)--D-glucopyranoside with thioglycoside methyl (ethyl 5-acetamido-4,7,8,9-tetra-O-acetyl-3,5-dideoxy-2-thio-D-glycero--D-galacto-2-nonulopyranosyl)oate in acetonitrile in the presence of N-iodosuccinimide and trifluoromethanesulfonic acid afforded 2-azidoethyl [methyl (5-acetamido-4,7,8,9-tetra-O-acetyl-3,5-dideoxy-D-glycero--D-galacto-2-nonulopyranosyl)oate]-(2 3)-(2,6-di-O-benzyl--D-galactopyranosyl)-(1 4)-2,3,6-tri-O-benzyl--D-glucopyranoside, the selectively protected derivative of the oligosaccharide chain of GM3 ganglioside, in 79% yield. During most infections the plasma levels of trace elements change, but it is not clear if this reflects changes in the infected tissues. Coxsackievirus B3 (CB3) infection may result in viral replication, subsequent inflammation and changed trace element levels in the myocardium. In the present study, the trace element levels in the plasma and heart of adult male A/J mice were determined during the pre-inflammatory stage (day 4) of CB3 myocarditis for the following trace elements: aluminium (Al), arsenic (As), calcium (Ca), cobalt (Co), copper (Cu), iron (Fe), magnesium (Mg), manganese (Mn), selenium (Se), silver (Ag), vanadium (V) and zinc (Zn). The severity of the infection was assessed through clinical signs of disease and trace element levels were measured through inductively-coupled plasma mass-spectrometry (ICP-MS). In the heart, the levels decreased for V (59%; p<0.01), Co (38%; p<0.01), Al (81%; p< 0.01), As (66%; p<0.01) and Se (16%; p<0.01). Increased levels were detected for Mn (13%; p<0.05), Fe (48%; p<0.01), Cu (34%; p<0.01) and Ag (46%; p< 0.01). In the plasma, decreases were detected in the level of Zn (32%; p<0.05), whereas increases were seen in Mn (362%; p<0.05), Fe (272%; p<0.01), Co (71%; p<0.05), Cu (25%; n.s.) and Mg (43%; p<0.01) levels. A correlation was found between the levels in plasma and myocardium for Co (r s=–0.636; p<0.05), Fe (r s=0.764; p<0.05), Mn (r s=0.682; p<0.05) and Mg (r s=–0.791; p<0.05). Thus, determination of some of these trace elements in the plasma may be useful to indicate target tissue involvement in the early pre- inflammatory stage of an infectious disease. Some of these elements are important nutrients for the immune system, while others may be associated with the development of disease complications, such as cardiac arrhythmias.  相似文献

The level of an important carotenoid (-carotene) in the gut of Periplaneta americana depends on the content of the carotenoid in food: a carotenoid-fortified diet causes accumulation of -carotene up to 10 g/g wet weight, while on a carotenoid-deficient diet the level of this substance is low (0.7 g/g wet weight). In the eye, in contrast to the gut, a constant level of -carotene (1.3-1.4 g/g wet weight) is found regardless of the diet. This phenomenon remained unchanged over three years of feeding of the cockroaches with the carotenoid-deficient diet, suggesting that P. americana produces carotenoids by de novo biosynthesis. This suggestion was confirmed in experiments using intraperitoneal injection of the exogenous carotenoid biosynthesis precursor [14C]mevalonic acid pyrophosphate followed by extraction of carotenoid and chromatographic purification of the labeled product. Injection of 3.4 nmoles [14C]mevalonic acid pyrophosphate transiently increased the -carotene content in eyes on days 2 and 4 after injection of the label. Purification of radiolabeled carotenoids from eye and gut by the transfer of carotenoids into a less polar solvent, alkaline hydrolysis (saponification), and chromatography on alumina and cellulose columns decreased the specific radioactivity to a constant level that cannot be further decreased by repeated chromatography. The elution profile of these purified preparations of -carotene after chromatography is characterized by coincidence of symmetric peaks of count and absorption. We suggest that to create the optimal carotenoid concentration in the eye, P. americana uses two biochemical mechanism: 1) it accumulates carotenoids in reserve in the gut when abundant supplies of carotenoids are available in the diet; 2) it synthesizes carotenoids de novo when its food is deficient in these compounds.  相似文献

The underwater light field in the Bellingshausen andAdmundsen Seas was characterised using data collectedduring the R/V Polarstern cruise ANT XI/3, from12.1.94 to 27.3.94. The euphotic zone varied from 24to 100 m depth. Spectral diffuse vertical attenuationcoefficients (K d ())were determined for 12narrow wavebands as well as for photosyntheticallyavailable radiation (PAR, 400–700 nm): K d (490)ranged from 0.03 to 0.26 m1; K d (550) from0.04 to 0.17 m1; K d (683) from 0.04 to0.17 m1; and K d (PAR) varied from 0.02 to0.25 m1. K d () for wavelengths centred at412 nm, 443 nm, 465 nm, 490 nm, 510 nm, 520 nm and550 nm were significantly correlated with chlorophyllconcentration (ranging from 0.1 to 6 mg m3). Thevertical attenuation coefficients for 340 nm and380 nm ranged from 0.10 to 0.69 m1 and from 0.05to 0.34 m1, respectively, and were also highlycorrelated with chlorophyll concentrations. These K d values indicate that the 1% penetration depthmay reach maxima of 46 m and 92 m for 340 nm and380 nm, respectively. The spectral radiancereflectances (Rr()) for 443 nm, 510 nm and 550 nmwere less than 0.01 sr1. Rr() for 665 nm and683 nm increased with depth up to 0.2 sr1 because ofchlorophyll fluorescence. Using a model that predicts downwardirradiances by taking into account the attenuation bywater and absorption by chlorophyll, we show thatchlorophyll fluorescence has a significant influenceon the red downward irradiance (E d (633, 665, 683))in deeper layers. The ability of the phytoplanktonpopulation to influence the light environment byautofluorescence and absorption processes depends onthe light conditions and on the photoacclimation ofthe cells, represented by the in vivo crosssection absorption coefficient of chlorophyll (a*). Theobtained mean chlorophyll-specific light attenuationcoefficients of phytoplankton in situ (k d ) are higherthan the in vivo absorption coefficient of chlorophyll,more than to be excepted from the scattering. a*(), m2 mg chl1, decreased due topackaging effect with increasing chlorophyllconcentrations.  相似文献

The low-copy-number and broad-host-range pSM19035-derived plasmid pBT233 is stably inherited in Bacillus subtilis cells. Two distinct regions, segA and segB, enhance the segregational stability of the plasmid. Both regions function in a replicon-independent manner. The maximization of random plasmid segregation is accomplished by the recombination proficiency of the host or the presence of the pBT233 segA region. The segA region contains two open reading frames (or) [ and ]. Inactivation or deletion of or results in SegA plasmids. Better than random segregation requires an active segB region. The segB region contains two ors (or and or). Inactivation of either of the orfs does not lead to an increase in cell death, but or plasmids are randomly segregated. These results suggest that pBT233 stabilization relies on a complex system involving resolution of plasmid oligomers (segA) and on the function(s) encoded by the segB region.  相似文献

We report here studies which characterize further the interaction ofN,N-dicyclohexylcarbodiimide with cytochromec oxidase leading to inhibition of H+ translocation by the enzyme. Further evidence is presented to show that the inhibition results from a real interaction of DCCD with the enzyme and cannot be accounted for by uncoupling and, contrary to recent criticisms, this interaction occurs specifically with subunit III of the enzyme even at relatively high inhibitor-to-enzyme stoichiometries. Summary In merodiploid cells containing a double dose of structural genes of RNA polymerase subunits-rpoBand rpoC-the rate of and subunits synthesis is 2 times higher than in haploidcells. Missence mutation rpoC1 (tsX) alters polypeptide and inducesthe and subunits synthesis at increased rate, particularly at a nonpermissive temperature. When rpoBCoperon carrying mutation rpoC1 is duplicated no dosage effect is observed. In the rpoC +/rpoC1 heterodiploid the rpoC1 mutation does not significantly accelerate RNA polymerase subunits synthesis i.e. is recessive with respect to rpoC + Rifampicin causes 6-fold stimulation of RNA polymerase subunits synthesis in a sensitive wild-type strain. The rpoC1 mutation itself accelerates the synthesis of these subunits 3-fold. In the presence of rifampicin the mutant strain produces 13–22-fold faster as compared to wild-type strain without the drug. Thus, the effects of rifampicin and the mutation are multiplied suggesting that these factors act independently. Similar data have also been obtained with rifampicin-treated cells of rpoB22 (ts22) amber-mutant.After UV-irradiation of cells and synthesis is depressed much stronger than the total protein synthesis. Infection with a transducing phage rif d-47 which carries rpoB gene provokes a higher rate of synthesis. When pre-irradiated cells (500 erg/mm2) are infected with this phage, the rate of synthesis grows 20-fold compared to irradiated, non-infected cells and 6.5-fold compared to intact cells.The data are discussed in terms of the possible regulatory mechanisms of RNA polymerase subunit synthesis.  相似文献
