Summary The streptococcal plasmid ERL1 determining inducible resistance to erythromycin, lincomycin, and staphylomycin S was isolated by dye-buoyant density centrifugation and shown to have a molecular weight of about 17.5 Mdal, as revealed by sedimentation through neutral sucrose gradients. In SM60 cells entering the stationary phase its covalently closed circular form was present to the extent of 5 copies per chromosomal genome equivalent. ERL1 was subject to the DNA restriction and modification mechanism discovered in strain 56188. It did not apear to exercise restriction of phage DNA but mediated a partial release of the restricted growth of A25.  相似文献

Summary Using recombinant DNA techniques, we introduced a previously cloned streptokinase gene from Streptococcus equisimilis into the Challis strain of S. sanguis (group H). The gene was expressed in the new host under the control of its own promoter and the gene product had biological properties identical to authentic streptokinase. However, the molecular weight of cloned streptokinase (42 K) as expressed by S. sanguis was substantially lower than that of authentic streptokinase (47 K). Since the cloned streptokinase gene encoded a 47 K mature protein, the lowered molecular weight of S. sanguis streptokinase may reflect posttranslational proteolytic cleavage, which leaves the biological activity of the gene product and its serological reactivity unimpaired.  相似文献

利用免疫蛋白组学方法,鉴定出链球菌2型(Streptococcus suis type2,SS2)江苏分离株HA9801具有免疫反应性的蛋白HM3。应用同源性比对、信号肽预测、跨膜区预测及亚定位预测等生物信息学方法对该蛋白进行分析,结果显示:同源性最高的蛋白为屎肠球菌胞外溶解物结合蛋白(41%);蛋白序列中含有信号肽结构;7-24位氨基酸为该蛋白的跨膜区;预测蛋白定位为除胞质外的未定位置。PCR扩增出该蛋白的一段基因定向克隆到表达载体pET-32a( )中并转化入BL21(DE3)宿主菌。重组菌经IPTG诱导后的SDS-PAGE图谱在46kDa处出现融合蛋白的条带。Westernblot表明,此融合蛋白可被SPF微型猪抗SS2血清所识别,提示该蛋白可作为该菌的亚单位疫苗的候选物。  相似文献

The low-copy-number and broad-host-range pSM19035-derived plasmid pBT233 is stably inherited in Bacillus subtilis cells. Two distinct regions, segA and segB, enhance the segregational stability of the plasmid. Both regions function in a replicon-independent manner. The maximization of random plasmid segregation is accomplished by the recombination proficiency of the host or the presence of the pBT233 segA region. The segA region contains two open reading frames (or) [ and ]. Inactivation or deletion of or results in SegA plasmids. Better than random segregation requires an active segB region. The segB region contains two ors (or and or). Inactivation of either of the orfs does not lead to an increase in cell death, but or plasmids are randomly segregated. These results suggest that pBT233 stabilization relies on a complex system involving resolution of plasmid oligomers (segA) and on the function(s) encoded by the segB region.  相似文献

A gene (dexS) coding for a Streptococcus suis capsular type 2 dextranase (DexS) was detected in a recombinant gene library constructed in phage λZapII, and its nucleotide sequence was determined. Sequence comparison showed that the dexS gene product had significant similarities with enzymes which hydrolyze glucose polymers. Moreover, conserved amino acids that are suggested to be part of the active site of the glucosidases are also found in DexS. The dexS gene, adjacent to the gene encoding a S. suis IgG-binding protein, encoded a protein of approximately 62 kDa which exhibited DexS activity.  相似文献

[背景] 化脓隐秘杆菌作为一种条件致病菌,常与蓝耳病病毒、猪圆环病病毒、巴氏杆菌等混合感染猪,引起各种非特异性化脓性感染,部分临床症状与副猪嗜血杆菌相似。本试验从江西某猪场一起因呼吸道症状急性死亡病猪的肺脏分离到一株具有β溶血特性、疑似化脓隐秘杆菌的细菌病原。[目的] 鉴定该病原的菌属种类、生物学特性及基因组特征,为该菌疾病的基础研究与临床防控奠定理论基础。[方法] 采用透射电镜、扫描电镜结合PCR鉴定等方法对菌属种类进行鉴定;通过动物实验、药敏试验,分析菌株的生物学特性;借助全基因组测序、生物信息学等技术发掘该菌的基因组特征。[结果] 形态观察、16S rRNA基因鉴定及系统发育分析证实,该分离菌株是化脓隐秘杆菌,命名为JX18;药敏试验表明,该菌株对克林霉素和林可霉素不敏感,对其余所选药物均敏感;动物实验结果显示,对小鼠腹腔攻毒后,小鼠腹部出现明显脓肿病灶,最高剂量组的小鼠全部死亡;全基因组测序结果表明,该菌株携带plo、nanH、nanP、fimA、fimC、fimE等重要毒力基因,并且与其他化脓隐秘杆菌菌株毒力基因的相似性较高;根据同源基因数据库(Cluster of Orthologous Groups,COG)预测,菌株JX18中参与碳水化合物代谢的基因占比最高;通过毒力基因数据库(virulence factor database,VFDB)、UniProtKB/Swiss-Prot等数据库预测,JX18菌株基因组中携带有多个组氨酸激酶、反应调节因子等与细菌双组分调控系统相关基因,以及多种与粘附、侵入及分泌系统相关的毒力基因。[结论] 在江西省分离鉴定出一株猪源强致病性化脓隐秘杆菌,丰富了猪源化脓隐秘杆菌病原信息,为进一步开展猪源化脓隐秘杆菌相关研究与防控奠定了理论依据。  相似文献

A cDNA library was constructed in λgt11 using poly(A)+ mRNA from early larvae of Onchocerca gibsoni. Screening of the library using serum from a single onchocerciasis patient yielded several strongly immunoreactive clones, one of which (OGK2) was found to encode a novel myosin-related protein. cDNA clone OGK2 contained an insert of 2017 bp, consisting of continuous open reading frame in frame with the vector; hence this clone encodes 671 amino acid residues of a larger protein. A fragment (619 nt) of the OGK2 cDNA was subcloned into the expression vector pGEX-1N to generate a glutathione S-transferase fusion protein. Polyclonal antiserum raised to this fusion protein strongly recognised an O. gibsoni protein of approximately 220 kDa. Immunolocalization studies indicated that this protein was associated predominantly with the hypodermis and a number of other specific membrane layers in the adult parasite. Myosin-related proteins are frequently immunodominant parasite antigens and in a number of studies have been shown to confer a degree of protective immunity against the corresponding parasite. Evaluation of the protective potential of the OGK2 protein, therefore, appears to be warranted.  相似文献

【目的】猪链球菌是一种能感染人和猪的人畜共患病病原,并且还可零星感染多种哺乳动物。本试验旨在调查流浪猫携带猪链球菌的情况。【方法】在流浪猫身上分离猪链球菌,经血清凝集实验和PCR检测,鉴定其血清型;经多序列位点分型分析,鉴定其ST型;将所分离的细菌与Gen Bank上已公布的猪链球菌构建16S rRNA的系统发育树,分析该菌株与其他猪链球菌的亲缘关系;药敏纸片法分析其耐药性;小鼠攻毒试验分析其毒力。【结果】本试验在流浪猫身上分离到一株猪链球菌,命名为m70,其血清型为9型。多序列位点分型显示,m70株属于一个新的ST型。与Gen Bank上已公布的猪链球菌16S rRNA进行系统发育树分析,结果显示m70属于一个单独的分支。m70与临床菌株的耐药情况相似,对四环素耐药,对红霉素中介耐药,对氨苄西林敏感。小鼠攻毒试验显示,感染10~8 CFU剂量m70的小鼠,死亡率达到60%–80%(3/5–4/5),3次攻毒试验的平均LD(50)为5.1×107 CFU;而本实验室保存的猪链球菌强毒株HA9801感染小鼠的平均LD(50)为3.9×107 CFU,两者之间没有显著差异(P〈0.05)。【结论】从流浪猫身上分离得到的猪链球菌m70属于优势血清型,且毒力较强,提示一些流行血清型的猪链球菌强毒株具有从流浪猫传染人的潜在风险。  相似文献

【背景】副乳房链球菌(Streptococcus parauberis)是重要的水产病原菌,该病原菌已逐渐出现新的血清型及多重耐药性状,因此亟须开发出一种新的抗菌药物用于该病害的防治。研究发现,前噬菌体编码的裂解酶能够有效地杀死其宿主,具有良好的抗菌应用前景。【目的】以副乳房链球菌前噬菌体裂解酶为对象,研究其杀菌宿主谱并优化其裂解活性的条件。【方法】利用PHASTER工具对副乳房链球菌菌株KRS02083全基因组序列分析发现,其前噬菌体包含一种裂解酶的基因Sply828;通过基因克隆、表达和纯化等技术得到裂解酶Sply828蛋白;通过浊度递减实验探究裂解酶Sply828对不同细菌的杀菌活性及其最适的裂解条件。【结果】裂解酶Sply828对鱼源副乳房链球菌具有最佳的杀菌活性,并发现该酶对处于指数生长期的细菌杀菌效果最好;其最适裂解温度为28°C,最适pH为6.2;Ca2+和Mg2+对该酶的杀菌活性有促进作用,但是Zn2+、Cu2+、Fe2+、Ni2+明显抑制...  相似文献

The isolation, purification, and partial characterization of a novel iron-containing protein from the sulfate-reducing anaerobic bacterium, Desulfovibrio gigas, is described. The highly insoluble protein was isolated from the cell debris following osmotic shock of the bacteria. The insoluble fraction consistently contained about 90% of the cell-associated iron. Elemental analysis of a crude protein preparation gave 5.3% iron, 2.9% sulfur and 11.9% nitrogen. An independent colorimetric iron analysis showed 6.4% iron. The iron could be dissociated from the protein by treatment with 5% SDS. The iron-free protein was purified by a combination of organic extraction and DEAE-cellulose chromatography. The purified protein showed only one major band, Mr 14 000, by SDS-polyacrylamide gel electrophoresis. The protein could be reconstituted upon treatment with an appropriate mixture of FeS and β-mercaptoethanol. The reconstituted protein had the same physical and chemical properties as the native protein. The amino acid composition was not unusual except for the high isoleucine content.  相似文献

Streptococcus thermophilus is a thermophilic gram-positive bacterium belonging to the lactic acid group. We report the isolation and characterization of a new 9.6-kDa DNA-binding protein, HSth, belonging to the HU family of nucleoid-associated proteins. The hsth gene was isolated in a 2.5-kb genomic region, upstream of a gene with strong homology to Lactococcus lactis pyrD. It is transcribed from a single E. coli sigma(70)-like promoter. Based on its high level of sequence similarity to B. subtilis and E. coli HU, HSth appears to be an HU homologue. The HSth protein shows biochemical and functional properties typical of HU proteins from gram-positive bacteria, being heat-stable, acid-soluble, and homodimeric. When expressed in HU-deficient E. coli cells, HSth supported the growth of bacteriophage Mu as efficiently as E. coli HU homo- and heterodimeric proteins. It did not, however, display any IHF-specific functions. Finally, we show that HSth binds to linear DNA with no apparent specificity, forming protein-DNA complexes similar but not identical to those observed with E. coli HU proteins.  相似文献

猪链球菌(Streptococcus suis)是猪重要的细菌性病原,它可以导致猪的脑膜炎、败血症和关节炎等症状,给养猪业带来严重经济损失;同时该菌还可感染人,是一种人畜共患病原菌。应用分子流行病学方法,阐明猪链球菌病的流行病学特征,明确其毒力分型、时空分布、传播途径、传染源,确定传播的遗传决定因素等,将有助于猪链球菌病的防控。目前常用的分子流行病学方法主要有多位点序列分型、脉冲场凝胶电泳、全基因组测序和基于PCR的方法等。本文介绍了上述方法的原理以及在猪链球菌流行病学中的应用,并分析这几种方法的优缺点,从而为更好地揭示猪链球菌流行病学特征、制定猪链球菌病的防控策略提供参考。  相似文献

An anti-fungal protein GAFP-1 (Gastrodia anti-fungal protein, also called gastrodianin) was purified from Gastrodia elata B1. f. flavida S. Chow (Orchidaceae), a parasitic plant on the fungus Armillaria mellea. It can inhibit the hyphal growth of some phytopathogenic fungi such as Valsa ambiens, Rhizoctonia solani, Gibberella zeae, Ganoderma lucidum and Botrytis cinerea in vitro. GAFP-1 is a monomer with a molecular mass of 10 kDa and a pI of 8.45. The optimum pH for its inhibitory activity is 5.0 ∼ 6.0. GAFP-1 is insensitive to high temperatures. It can preserve 75% inhibitory activity after 30 min at 60°C. Amino acid composition analysis revealed that GAFP-1 is rich in Asp (22.1%), Gly (10.0 %) and Leu (9.4 %), and does not contain any Pro. The amino acid sequence of the N-terminal was determined and found to share high homology with those of other lectins from orchids such as Listera ovata and Epipactis helleborine. GAFP-1 could not agglutinate trypsin-treated rabbit erythrocytes. It could bind to chitin, immobilized mannose and N-acetylglucosamine in 50mM sodium acetate buffer (pH 5.0) with 2 M ammonium sulfate. These data suggest that GAFP-1 could be a lectin-like protein with strong inhibitory activity against certain fungal pathogens.  相似文献

【背景】马链球菌兽疫亚种(Streptococcus equi subsp. zooepidemicus,SeZ)是引起马腺疫的主要病原,还可引起猪链球菌病,加强该菌的地方株分子流行病学监测对有效防控相关疫病十分必要。【目的】对新疆地区2个马场SeZ分离株进行鉴定和药敏特性分析,并分析3株新疆分离株的分子流行与菌株的遗传进化特征。【方法】对分离纯化的3株病原菌(ZHZ113、ZHZ211和ZHZ523)进行染色观察、生化及药敏特性检测,对16S rRNA和SeM基因进行遗传进化分析,以链球菌7个管家基因arcC、nrdE、proS、spi、tdk、tpi和yqiL为目的基因对3株分离菌进行多位点序列分型(Multilocus Sequence Typing,MLST)研究。【结果】3株SeZ的药敏结果显示这3株分离菌对不同抗生素的耐药程度不同,但均对头孢西丁、庆大霉素、链霉素、红霉素、左氧氟沙星、环丙沙星、土霉素等11种药物敏感。16SrRNA基因序列分析显示这3株分离菌均属于Ⅱ群(兽疫链球菌)。3株菌的MLST分型结果分别为ST39、ST419、ST421型,其中ST419和ST42...  相似文献

Streptococcus pneumoniae type 2 pili are recently identified fimbrial structures extending from the bacterial surface and formed by polymers of the structural protein PitB. Intramolecular isopeptide bonds are a characteristic of the related pilus backbone protein Spy0128 of group A streptococci. Based on the identification of conserved residues in PitB, we predicted two intramolecular isopeptide bonds in PitB. Using a combination of tandem mass spectrometry and Edman sequencing, we show that these bonds were formed between Lys63-Asn214 and Lys243-Asn372 in PitB. Mutant proteins lacking the intramolecular isopeptide bonds retained the proteolytic stability observed with the wild type protein. However, absence of these bonds substantially decreased the melting temperature of the PitB-derivatives, indicating a stabilizing function of these bonds in PitB of the pneumococcal type 2 pilus.  相似文献

Mutacins are bactericidal substances of proteinaceous nature produced by Streptococcus mutans. Lantibiotics are antibacterial substances containing post-translationally modified amino acids such as lanthionine. Mutacin B-Ny266 was purified from the cell pellet of S. mutans strain Ny266 by ethanol extraction at pH 2.0 followed by reversed-phase chromatography (Sep-Pak® cartridge) and by HPLC on a C18 column. The mean purification factor was 3240±81 and the mean yield was 1.0±0.1%. Molecular mass of mutacin B-Ny266 as determined by mass spectroscopy is 2270.29±0.21 Da. The amino acid sequence of the purified active fraction was obtained by Edman degradation after treatment with alkaline ethanethiol. Twenty-one amino acids were detected in this analysis. Mutacin B-Ny266 belongs to the type A lantibiotics. The proposed sequence is: F–K–
