人血小板衍生生长因子BB亚型包涵体复性与纯化   总被引:1,自引:0,他引:1  
目的:优化人血小板衍生生长因子BB亚型(PGDF-BB)包涵体复性方法与纯化条件,获得具有较好生物活性的重组PGDF-BB蛋白。方法:对PGDF-BB包涵体以梯度尿素进行变性,选择最佳包涵体变性浓度;比较不同复性条件下的复性率,稳定PGDF-BB包涵体复性方法;参照该蛋白的理化性质,选择适合PGDF-BB重组蛋白的纯化方法。结果:原核系统内实现了PGDF-BB的高表达;通过优化包涵体复性方法,重组蛋白的包涵体复性率可达40%以上;经过多个纯化方法相结合,PGDF-BB的纯度达到95%。结论:通过实验条件的优化,提高了PGDF-BB包涵体复性率,获得高纯度、高生物活性的重组PGDF-BB蛋白。  相似文献

利用8 mol/L尿素溶液对表达在大肠杆菌包涵体中的GST-TRAF6融合蛋白进行变性,通过逐级稀释复性的方法对尿素溶解后的GST-TRAF6融合蛋白进行复性,将复性后的GST-TRAF6融合蛋白进一步利用谷胱甘肽琼脂糖树脂亲和层析的方法进行分离纯化,将分离纯化后的蛋白通过Western blot方法进行验证,最后利用体外泛素化反应检测经包涵体变性、复性和纯化后的GST-TRAF6融合蛋白的生物学活性。经过包涵体变性、梯度稀释复性和谷胱甘肽琼脂糖树脂亲和层析3个步骤后纯化得到纯度达90%以上、浓度为396 ng/μL的蛋白质溶液。利用GST蛋白作为对照,经Western blot验证表明,纯化得到的蛋白确为GSTTRAF6融合蛋白。进一步利用体外泛素化反应分析其泛素连接酶活性发现,17 ng/μL浓度的GST-TRAF6融合蛋白能够以泛素分子作为底物在5 min内快速催化自由泛素链的生成。结果表明,表达在大肠杆菌包涵体中的GST-TRAF6融合蛋白经尿素变性溶解后能够成功复性并分离纯化,在溶解性改变的同时恢复了其泛素连接酶活性。为从大肠杆菌包涵体中大规模分离纯化蛋白质提供了一种新的复性方法。  相似文献

基于蛋白质内含子的蛋白质纯化自我断裂标签已经被广泛使用超过15年之久.但这一系统体内表达过程的提前断裂一直是限制这一技术广泛应用的瓶颈,特别是在需要高温表达和长表达周期的真核表达系统中.本研究介绍了一种利用小肽控制的基于蛋白质内含子和非层析标签ELP(elastin-like polypeptide)的自我断裂系统.在这一系统中,蛋白质内含子的体内外活性严格受到其结构互补小肽控制.在体内表达不含有互补小肽时,蛋白质内含子不具有活性;而在体外添加结构互补小肽,蛋白质内含子结构恢复并发生C端断裂反应释放目的蛋白.由于非层析标签ELP的引入,因此整个纯化过程可以简单地通过几步机械沉淀完成.此外,这一系统反应pH、小肽与前体蛋白之间的摩尔比及断裂速率也一并进行了系统的研究.  相似文献

蛋白质内含子介导的断裂(切割)反应被用于蛋白质纯化、连接和环化等,但目前仍存在断裂效率低、断裂反应的不可控、产物复杂等问题。蛋白质内含子的定点突变可导致其N端或C端断裂。其末位氨基酸突变则剪接反应第3步天冬酰胺环化无法进行,发生N端断裂;其首位氨基酸发生突变则剪接反应第一步酰基重排及其后续步骤均无法进行,而天冬酰胺环化仍可进行,发生C端断裂。利用已获得的高剪接活性的S1和S11型断裂蛋白质内含子Ssp GyrB,分别将其参与剪接反应的首位半胱氨酸或末位天冬酰胺突变为丙氨酸,构建能够发生一端断裂的断裂蛋白质内含子。研究结果表明,突变后断裂蛋白质内含子的剪接反应几乎不发生,其断裂活性有不同程度的提高,获得了在大肠杆菌体内具有较高效断裂活性的断裂蛋白质内含子。这将为进一步研究其体外可控性剪接、构建高效的蛋白纯化系统和深入研究蛋白质内含子的剪接机制提供基础。  相似文献

带有BMP 7基因的大肠杆菌可以用来高量表达重组的人骨形态发生蛋白 7。升温诱导表达后 ,每升培养液大约可得到菌体湿重 3g ,其中目的蛋白约占菌体总蛋白量的 40 %。裂解离心 ,用低浓度变性剂洗涤初步纯化包涵体 ,上清中无目的蛋白损失 ,目的蛋白纯度提高到 60 %,将包涵体溶解于高浓度变性剂溶液中 ,然后在不同条件下用离子交换色谱法对变性状态下的蛋白质进行纯化 ,绝大部分杂蛋白被除去 ,目的蛋白纯度达 95 %以上 ,改变条件 ,可以减少rhBMP 7损失。并做Westernblot对目的蛋白进行特异性鉴定。  相似文献

以高效表达的基因工程菌为材料,对以包涵体形式存在的目的产物——心钠素(ANF)融合蛋白进行了酶切及分离纯化.先用TritonX-100和低浓度尿素、盐酸胍洗涤,提高包涵体的纯度.再用高浓度的尿素处理,使蛋白变性溶解,脱氧胆酸钠对蛋白有明显的促溶作用.解决变性蛋白质去除变性剂时的聚合问题.在非变性条件下进行SephadexG-75凝胶过滤,FPLC系统纯化,得到目的融合蛋白.用凝血酶对融合蛋白作特异性切割,释放出ANF小肽,经FPLC分离鉴定,与标准ANF有相同的TR值,说明其纯度已达均一.生物活性检测表明:有明显的放免活性和舒张血管、利尿的生理活性  相似文献

蛋白质内含子对于外显子的选择性限制了它的应用范围。目前,已经建立的卡那霉素定向进化系统适用于微小蛋白质内含子,但不适用于断裂蛋白质内含子。为了研究适用于断裂蛋白质内含子的定向进化的方法,我们引入了DNA展示系统。在该系统中,生物素化基因在人工细胞中与其表达的融合蛋白(内含子C端-外显子-生物素结合蛋白)形成DNA 蛋白质连接体。只有能与后续加入的N端蛋白质内含子前体(Flag-内含子N端)发生剪接反应的DNA 蛋白质连接体,才能被加上旗帜标签(Flag),从而被抗旗帜标签抗体纯化柱(Anti Flag antibody M2 agarose)筛选富集。为了验证该系统的可行性,实验构建了2个基因,即含有具剪接活性的蛋白质内含子的阳性基因IRC和含有不具剪接活性的蛋白质内含子的阴性基因IRCM。实验首先通过Western印迹和琼脂糖凝胶电泳证明,人工细胞中的体外转录翻译系统不仅可高表达500个氨基酸的蛋白质,而且表达的蛋白中内含子仍保持原有的剪接能力。生物素结合蛋白能结合95%的DNA,并且形成的DNA 蛋白质连接体可以被筛选富集,最终证明了该系统用于断裂蛋白质内含子定向进化筛选的可行性。随后,为了检测系统的富集效率,制备了人为的基因“突变库”,即将基因IRC和IRCM以摩尔比1:10的比例混合,经过2轮筛选后,阳性基因IRC可以被10倍富集,进一步证明了体外筛选方法的可行性。该方法为后续针对不同宿主进化出不同的断裂蛋白质内含子提供筛选方法支持,也为断裂蛋白质内含子的在生物技术和研究领域的应用奠定了基础。  相似文献

重组人粒细胞集落刺激因子(rhG-CSF)经大肠杆菌温度诱导表达后,其表达产物以包涵体形式存在,包涵体经过变性、复性和分离纯化等步骤处理后得到纯化的rhG-CSF.在一定的实验条件下用单甲氧基聚乙二醇活性酯(mPEG20k-NHS)对rhG-CSF进行化学修饰,所得修饰产物经分离纯化后获得PEG20K-rhG-CSF偶联物.与修饰前的rhG-CSF相比较,尽管修饰后的rhG-CSF体外生物学活性下降至修饰前的20%左右,但其在体内的作用时间能够得到显著的延长,药效有了明显提高.  相似文献

Hsp47 is regarded as a collagen-specific chaperone with several suggested roles in collagen biosynthesis under normal and disease conditions. We describe here a procedure for the expression and purification of Hsp47 in Escherichia coli using the IMPACT expression system (New England Biolabs) where the guest gene is fused to the adduct, intein, with a chitin-binding domain. Use of this system resulted in relatively high levels of soluble Hsp47 compared to other available protocols, especially when the bacterial cells were induced at 14 degrees C instead of 37 degrees C. The cell lysate was passed through a chitin-Sepharose affinity column and Hsp47 was cleaved from intein using beta-mercaptoethanol. Minor degradation products were subsequently removed using a hydroxylapatite column to yield milligram amounts of pure and active protein suitable for structural studies. Gel electrophoretic analysis of the purified protein indicated the presence of a small proportion of trimeric species when non-reducing conditions were used. The ability to form a trimer may be important for its role as a chaperone. The IMPACT system allows for radiolabelling of purified Hsp47 with (35)S for use in binding experiments. Illustrative data on collagen binding by (35)S-Hsp47 are shown.  相似文献

AIMS: To study the effect of 2450 MHz microwave radiation under vacuum (vacuum microwave or VM) on survival and injury of Escherichia coli and to search for possible nonthermal effects associated with VM. METHODS AND RESULTS: Destruction kinetics of E. coli in peptone water were determined in a continuous-flow vacuum system, heated by convection heating in a water bath or with microwaves (VMs). Vacuum was used to control the boiling point of water and to maintain temperature in the bacterial suspensions at specified levels (49-64 degrees C). CONCLUSIONS: z-Value in the water bath treatment was 9.1 degrees C while for VM at 510 and 711 W it was 6.2 and 5.9 degrees C, suggesting that E. coli is more sensitive to temperature changes under microwave heating. Arrhenius calculations of the activation energies of the destruction reactions suggest that the mechanism of destruction in VM may be different from that of conventional heat. The number of injured micro-organisms showed no significant differences among treatments. SIGNIFICANCE AND IMPACT OF THE STUDY: The impact of temperature on E. coli destruction was different when microwaves were the medium of heat transfer, suggesting the existence of factors other than heat contributing to the lethal effect of VM.  相似文献

Translational control directed by the eukaryotic translation initiation factor 2 alpha-subunit (eIF2alpha) kinase GCN2 is important for coordinating gene expression programs in response to nutritional deprivation. The GCN2 stress response, conserved from yeast to mammals, is critical for resistance to nutritional deficiencies and for the control of feeding behaviors in rodents. The mouse protein IMPACT has sequence similarities to the yeast YIH1 protein, an inhibitor of GCN2. YIH1 competes with GCN2 for binding to a positive regulator, GCN1. Here, we present evidence that IMPACT is the functional counterpart of YIH1. Overexpression of IMPACT in yeast lowered both basal and amino acid starvation-induced levels of phosphorylated eIF2alpha, as described for YIH1 (31). Overexpression of IMPACT in mouse embryonic fibroblasts inhibited phosphorylation of eIF2alpha by GCN2 under leucine starvation conditions, abolishing expression of its downstream target genes, ATF4 (CREB-2) and CHOP (GADD153). IMPACT bound to the minimal yeast GCN1 segment required for interaction with yeast GCN2 and YIH1 and to native mouse GCN1. At the protein level, IMPACT was detected mainly in the brain. IMPACT was found to be abundant in the majority of hypothalamic neurons. Scattered neurons expressing this protein at higher levels were detected in other regions such as the hippocampus and piriform cortex. The abundance of IMPACT correlated inversely with phosphorylated eIF2alpha levels in different brain areas. These results suggest that IMPACT ensures constant high levels of translation and low levels of ATF4 and CHOP in specific neuronal cells under amino acid starvation conditions.  相似文献

The purpose of this study was to investigate the effects of a floor heating and air conditioning system on thermal responses of the elderly. Eight elderly men and eight university students sat for 90 minutes in a chair under the following 3 conditions: air conditioning system (A), floor heating system (F) and no heating system (C). The air temperature of sitting head height for condition A was 25 degrees C, and the maximum difference in vertical air temperature was 4 degrees C. The air and floor temperature for condition F were 21 and 29 degrees C, respectively. The air temperature for condition C was 15 degrees C. There were no significant differences in rectal temperature and mean skin temperature between condition A and F. Systolic blood pressure of the elderly men in condition C significantly increased compared to those in condition A and F. No significant differences in systolic blood pressure between condition A and F were found. The percentage of subjects who felt comfortable under condition F was higher than that of those under condition A in both age groups, though the differences between condition F and A was not significant. Relationships between thermal comfort and peripheral (e.g., instep, calf, hand) skin temperature, and the relationship between thermal comfort and leg thermal sensation were significant for both age groups. However, the back and chest skin temperature and back thermal sensation for the elderly, in contrast to that for the young, was not significantly related to thermal comfort. These findings suggested that thermal responses and physiological strain using the floor heating system did not significantly differ from that using the air conditioning system, regardless of the subject age and despite the fact that the air temperature with the floor heating system was lower. An increase in BP for elderly was observed under the condition in which the air temperature was 15 degrees C, and it was suggested that it was necessary for the elderly people to heat the room somehow in winter. Moreover, it is particularly important for elderly people to avoid a decrease in peripheral skin temperature, and maintain awareness of the warmth of peripheral areas, such as the leg, in order to ensure thermal comfort.  相似文献

AIMS: Bacteria are important residents of water systems, including those of space stations which feature specific environmental conditions, such as lowered effects of gravity. The purpose of this study was to compare responses with modelled reduced gravity of space station, water system bacterial isolates with other isolates of the same species. METHODS AND RESULTS: Bacterial isolates, Stenotrophomonas paucimobilis and Acinetobacter radioresistens, originally recovered from the water supply aboard the International Space Station (ISS) were grown in nutrient broth under modelled reduced gravity. Their growth was compared with type strains S. paucimobilis ATCC 10829 and A. radioresistens ATCC 49000. Acinetobacter radioresistens ATCC 49000 and the two ISS isolates showed similar growth profiles under modelled reduced gravity compared with normal gravity, whereas S. paucimobilis ATCC 10829 was negatively affected by modelled reduced gravity. CONCLUSIONS: These results suggest that microgravity might have selected for bacteria that were able to thrive under this unusual condition. These responses, coupled with impacts of other features (such as radiation resistance and ability to persist under very oligotrophic conditions), may contribute to the success of these water system bacteria. SIGNIFICANCE AND IMPACT OF THE STUDY: Water quality is a significant factor in many environments including the ISS. Efforts to remove microbial contaminants are likely to be complicated by the features of these bacteria which allow them to persist under the extreme conditions of the systems.  相似文献

Detection of fish nocardiosis by loop-mediated isothermal amplification   总被引:4,自引:0,他引:4  
AIMS: Loop-mediated isothermal amplification (LAMP) is a novel method that amplifies DNA with high specificity and rapidity under isothermal conditions. In this study, using the LAMP method, a protocol for detecting Nocardia seriolae which is a causative agent of fish nocardiosis, was designed. METHODS AND RESULTS: A set of four primers, two inner and two outer, were designed based on the sequence of the 16S-23S ribosomal RNA internal transcribed spacer region of N. seriolae. Time and temperature conditions for detection of N. seriolae were optimized for 60 min at 65 degrees C. Other fish pathogen was not amplified by this LAMP system. The detection of N. seriola using LAMP was found to be more sensitive than that by polymerase chain reaction. CONCLUSIONS: LAMP is a highly sensitive and rapid diagnostic procedure for detection of N. seriolae. SIGNIFICANCE AND IMPACT OF THE STUDY: LAMP is a useful diagnostic method for fish nocardiosis.  相似文献

AIMS: The aim of this study was to investigate the influence of environmental and physiological factors on the susceptibility of Escherichia coli to the Curvularia haloperoxidase system. METHODS AND RESULTS: The Curvularia haloperoxidase system is a novel enzyme system that produces reactive oxygen species which have an antimicrobial effect. Escherichia coli MG1655 was exposed to the Curvularia haloperoxidase system under different temperatures and NaCl concentrations and after exposure to different stress factors. Temperature clearly affected enzymatic activity with increasing antibacterial effect at increasing temperature. The presence of NaCl interfered with the enzyme system and in the presence of 1% NaCl, no antibacterial effect could be observed at pH 7. Cells grown at pH 8.0 were in one experiment more resistant than cells grown at pH 6.5, whereas cells grown in the presence of 2% NaCl were more susceptible to the Curvularia haloperoxidase system. CONCLUSIONS: Environmental and physiological factors can affect the antibacterial activity of the Curvularia haloperoxidase system. SIGNIFICANCE AND IMPACT OF THE STUDY: The study demonstrates a systematic approach in assessing the effect of environmental and physiological factors on microbial susceptibility to biocides. Such information is crucial for prediction of application as well as potential side-effects.  相似文献

AIM: To develop a mathematical method to estimate non-isothermal microbial growth curves in foods from experiments performed under isothermal conditions and demonstrate the method's applicability with published growth data. METHODS AND RESULTS: Published isothermal growth curves of Pseudomonas spp. in refrigerated fish at 0-8 degrees C and Escherichia coli 1952 in a nutritional broth at 27.6-36 degrees C were fitted with two different three-parameter 'primary models' and the temperature dependence of their parameters was fitted by ad hoc empirical 'secondary models'. These were used to generate non-isothermal growth curves by solving, numerically, a differential equation derived on the premise that the momentary non-isothermal growth rate is the isothermal rate at the momentary temperature, at a time that corresponds to the momentary growth level of the population. The predicted non-isothermal growth curves were in agreement with the reported experimental ones and, as expected, the quality of the predictions did not depend on the 'primary model' chosen for the calculation. CONCLUSIONS: A common type of sigmoid growth curve can be adequately described by three-parameter 'primary models'. At least in the two systems examined, these could be used to predict growth patterns under a variety of continuous and discontinuous non-isothermal temperature profiles. SIGNIFICANCE AND IMPACT OF THE STUDY: The described mathematical method whenever validated experimentally will enable the simulation of the microbial quality of stored and transported foods under a large variety of existing or contemplated commercial temperature histories.  相似文献

Dissipative particle dynamics with energy conservation (eDPD) was used to investigate conduction heat transfer in two dimensions under steady-state condition. Various types of boundary condition were implemented to the conduction domain. Besides, 2D conduction with internal heat generation was studied and the heat generation term was used to measure the thermal conductivity and diffusivity of the eDPD system. The boundary conditions used include both the Neumann and Dirichlet boundary conditions. The Neumann boundary condition was applied via adiabatic surfaces and surfaces exposed to convection heat transfer. The DPD simulations were compared to analytical solutions and finite-difference techniques. It was found that DPD appropriately predicts the temperature distribution in the conduction regime. Details of boundary condition implementation and thermal diffusivity measurement are also described in this paper.  相似文献

AIMS: The inactivation of Bacillus subtilis 168 spores subjected to the combined stress of pH, temperature and sodium chloride in a buffer system was modelled. METHODS AND RESULTS: Bacillus subtilis 168 spore suspension in 50 mmol l-1 potassium phosphate buffer was heated in an open system using a block heater. A second order polynomial equation was used to describe the relationship between pH, temperature, sodium chloride concentration and the logarithm of the decimal reduction time (D-value) of the spores. Response surface graphs were constructed to predict the inactivation within the experimental domain. The data obtained were also compared with those reported for B. subtilis in different media and foods included in a large reference-based database of thermal inactivation (ThermoKill Database, TKDB R9100), which was constructed in the laboratory. CONCLUSIONS: All the variables studied seemed to have a significant effect on the inactivation of B. subtilis 168 spores in potassium phosphate buffer. The coefficient of determination, r2, and an analysis of the residuals from the model indicated the adequacy of the model to predict the inactivation of B. subtilis spores within the range of the experimental variables studied. SIGNIFICANCE AND IMPACT OF THE STUDY: The findings of this study will enable a better understanding of the inactivation of B. subtilis spores under the influence of the studied environmental variables. The model can be used by food industries to assess and monitor the shelf life of food products in the event of a chance contamination by B. subtilis spores.  相似文献
