Intrachromosomal asymmetry, one of the two aspects of karyotype asymmetry, increases because of shifts of the centromere position from the median/submedian to terminal or subterminal. Conventional measures of asymmetry use comparisons with the most symmetrical karyotype and thus are an absolute measure of variation in the centromeric index. Conversely, the CVCI index, the coefficient of variation for the centromeric index, only evaluates the relative variation in the centromeric index. We performed a detailed analysis of the capability of the CVCI index and conventional measures of karyotype asymmetry to accurately characterize differences in intrachromosomal asymmetry. Our results show that the CVCI index is unable to accurately evaluate intrachromosomal asymmetry. In fact, the CVCI index evaluates a different measure than other conventional measures of karyotype asymmetry. Therefore, to avoid confusion, we propose that the variation evaluated by the CVCI index should be named heterogeneity of the centromeric index.  相似文献

Despite extensive literature on the diversity of karyotypes in Allium is available, no attempt to analyse these data together, within a robust phylogenetic framework, has been carried out so far. Thus, we examined patterns and trends in chromosome evolution across the genus. Based on literature survey, karyo-morphometric features for 207 species belonging to 12 subgenera of Allium were obtained. Included in the data-set were basic chromosome number (x), somatic chromosome number (2n), total haploid (monoploid) chromosome length (THL) and three different measures defining karyotype structure: CVCI, measuring how heterogeneous are centromeres positions in a karyotype, CVCL and MCA, quantifying interchromosomal and intrachromosomal karyotype asymmetry, respectively. Trends in karyotype evolution were analysed by phylogenetic regressions and independent contrasts. Mean karyotypes highlighted differences and similarities in karyotype structure between the 12 subgenera. Further differences were noted when the two parameters for analysing karyotype asymmetry were assessed. In addition, by examining the effects of increasing karyotype dimensions (a proxy for genome size) on karyotype structure and asymmetry, it was shown that in Allium species, the DNA was added proportionally to their arm lengths. Overall, p = 8 and somehow intermediate karyotype asymmetry levels seem to represent plesiomorphic character-states in Allium.  相似文献

Karyotypes in 16 representative taxa of the Ophrys genus are compared, based on Feulgen-stained somatic metaphase chromosomes. The karyotypes of O. omegaifera subsp. israelitica, O. ulupinara, O. lycia, O. argolica subsp. lucis, O. argolica subsp. lesbis, O. climacis and O. reinholdii subsp. reinholdii are described for the first time. Karyological analyses indicate relationships among the species with respect to their asymmetry indices. All Ophrys taxa studied were diploid with 2n = 2x = 36 chromosomes. One B chromosome has been detected among the chromosomes of O. argolica subsp. lucis. All karyotypes are symmetrical, consisting of metacentric and submetacentric chromosomes. The longest chromosomes of all the investigated specimens contain a secondary constriction. It is determined that there is a correlation between the total number of chromosomes having secondary constrictions and the evolutionary development order of the taxa. Based on nuclear DNA content, analysis was carried out by flow cytometer using propodium iodide as fluorochrome, 2C nuclear DNA content of 16 Ophrys species varying between 20.80 pg (O. argolica subsp. lucis) and 23.11 pg (O. omegaifera subsp. israelitica). Karyotype asymmetry relationships are discussed according to the bidimensional scatter plots of A1–A2, CVCL–CVCI, CVCL–MCA and CVCI–MCA.  相似文献

Eight species of Ophrys sect. Pseudophrys (Orchidaceae) were cytogenetically studied. The analysed species possess the most symmetrical karyotypes of the genus (MCA ranged from 10.21 to 15.87 and CVCL from 19.61 to 23.93) with 2n = 2x = 36, being composed of mainly metacentric chromosomes. The karyotype formulae were: 36m for Ophrys archimedea, 32m + 4sm for O. flammeola, 32m + 4sm for O. funerea, 36m for O. laurensis, 36m for O. lojaconoi, 34m + 2sm for O. lucifera, 34m + 2sm for O. obaesa and 36m for O. pallida. Karyotype morphometric characters were evaluated by calculating MCA and CVCL for the assessment of karyotype asymmetry, and CVCI for the evaluation of heterogeneity in the position of the centromeres. The relationships between species were thus finally elucidated. The species characterised by wide distribution show greater karyomorphological distance than those with restricted distribution. The possible evolutionary role of chromosomal rearrangements as well as gene mutations in the speciation of Ophrys is discussed.  相似文献

The culture of green mussel (Perna viridis) in the Gulf of Thailand depends on natural spat which are believed to come from spawning grounds adjacent to major river mouths. In the present paper, genetic diversity of spatial and temporal populations of green mussel in the Gulf of Thailand was investigated using five microsatellite loci. The results showed moderate genetic variation of all 11 populations (averaged number of alleles per locus, A = 10.4–12.2; effective number of alleles per locus, A e = 5.36–6.59; mean allelic richness, A r = 10.23–12.06; observed heterozygosity, H o = 0.52–0.63, and expected heterozygosity, H e = 0.66–0.73) without significant differences among populations. No sign of bottleneck or genetic disequilibrium was observed. Genetic differentiation among spatial populations was low (F ST = 0.0046, CI0.95 = 0.0020–0.0083 for the samples collected in January, 2007, and F ST = 0.0088, CI0.95 = 0.0010–0.0162 for the samples collected in July, 2007) while temporal variation was significant as revealed by the analysis of molecular variance. Multidimensional scaling separated temporal population groups with minor exception. The assignment test revealed that most of the recruits were from other populations.  相似文献

The relationship between developmental stability and morphological asymmetry is derived under the standard view that structures on each side of an individual develop independently and are normally distributed. I use developmental variance of sizes of parts, VD, as the converse of developmental stability, and assume that VD follows a gamma distribution. Repeatability of asymmetry, a measure of how informative asymmetry is about VD, is quite insensitive to the variance in VD, for example only reaching 20% when the coefficient of variation of VD is 100%. The coefficient of variation of asymmetry, CVFA, also increases very slowly with increasing population variation in VD. CVFA values from empirical data are sometimes over 100%, implying that developmental stability is sometimes more variable than any previously studied type of trait. This result suggests that alternatives to this model may be needed.  相似文献

Characters which are closely linked to fitness often have low heritabilities (VA/VP). Low heritabilities could be because of low additive genetic variation (VA), that had been depleted by directional selection. Alternatively, low heritabilities may be caused by large residual variation (VR=VPVA) compounded at a disproportionately higher rate than VA across integrated characters. Both hypotheses assume that each component of quantitative variation has an independent effect on heritability. However, VA and VR may also covary, in which case differences in heritability cannot be fully explained by the independent effects of elimination‐selection or compounded residual variation. We compared the central tendency of published behavioural heritabilities (mean=0.31, median=0.23) with morphological and life history data collected by 26 ). Average behavioural heritability was not significantly different from average life history heritability, but both were smaller than average morphological heritability. We cross‐classified behavioural traits to test whether variation in heritability was related to selection (dominance, domestic/wild) or variance compounding (integration level). There was a significant three‐way interaction between indices of selection and variance compounding, related to the absence of either effect at the highest integration level. At lower integration levels, high dominance variance indicated effects of selection. It was also indicated by the low CVA of domestic species. At the same time CVR increased disproportionately faster than CVA across integration levels, demonstrating variance compounding. However, neither CVR nor CVA had a predominant effect on heritability. The partial regression coefficients of CVR and CVA on heritability were similar and a path analysis indicated that their (positive) correlation was also necessary to explain variation in heritability. These results suggest that relationships between additive genetic and residual components of quantitative genetic variation can constrain their independent direct effects on behavioural heritability.  相似文献

In order to refine the measure of karyotype asymmetry a new chromosomal parameter of dispersion index is proposed that has the potential to decipher even the minor karyotypic variations, thus permitting further evolutionary gradations to the karyotype asymmetry classes of Stebbins. The higher the dispersion index, the more specialized would be the karyotype. The dispersion index takes into account the variance lor gradual change in chromosome size within a complementvis-a-vis variance for the position of centromere in a karyotypic totality. The dispersion index is calculated as the proportionate measure of centromeric gradient to the coefficient of variation for chromosome length; wherin centromeric gradient = length of median short arm — length of median chromosome. Thus, the three most important karyotypic criteriaviz., differences in: absolute chromosome size, position of centromere and relative chromosome size, are all covered in the proposed parameter. The effectiveness of dispersion index has been tested on a plant taxa,Papaver L., where karyomorphological details, nuclear DNA content, and morphotaxonomic parameters have been amply elucidated from an evolutionary stand point. It is hoped that dispersion index would find immense utility in delimiting species interrelationships particularly in the closely related taxa, when applied in conjunction with other systematic parameters.  相似文献

This study focuses on karyological investigation of four taxa of Dianthus (Caryophyllaceae) including D. crossopetalus, D. orientalis subsp. stenocalyx, D. orientalis subsp. gorganicus, and Dianthus sp. distributed in Northeast of Iran. The karyotype asymmetry/symmetry was evaluated using two methods: 1) CVCL (coefficient of variation of chromosome length) and CVCI (heterogeneity of the centromeric index); and 2) MCA (mean centromeric asymmetry). The karyotype asymmetry was also used to investigate the relationships among the taxa. Results obtained from the current study revealed that there are two different ploidy levels (2n = 2x = 30 and 2n = 4x = 60) among the investigated taxa. The indices CVCL and MCA described karyotype asymmetry correctly based on variation in chromosome length. Intraspecific trait variation (ITV), based on available genetic diversity, is one of the major means plant populations can respond to environmental variability. The study of functional trait variation and diversity has become popular in ecological research, for example, as a proxy for plant performance influencing fitness. Up to now, it is unclear which aspects of intraspecific functional trait variation (iFDCV) can be attributed to the environment or genetics under natural conditions. Here, we examined 260 individuals from 13 locations of the rare (semi‐)dry calcareous grassland species Trifolium montanum L. in terms of iFDCV, within‐habitat heterogeneity, and genetic diversity. The iFDCV was assessed by measuring functional traits (releasing height, biomass, leaf area, specific leaf area, leaf dry matter content, Fv/Fm, performance index, stomatal pore surface, and stomatal pore area index). Abiotic within‐habitat heterogeneity was derived from altitude, slope exposure, slope, leaf area index, soil depth, and further soil factors. Based on microsatellites, we calculated expected heterozygosity (He) because it best‐explained, among other indices, iFDCV. We performed multiple linear regression models quantifying relationships among iFDCV, abiotic within‐habitat heterogeneity and genetic diversity, and also between separate functional traits and abiotic within‐habitat heterogeneity or genetic diversity. We found that abiotic within‐habitat heterogeneity influenced iFDCV twice as strong compared to genetic diversity. Both aspects together explained 77% of variation in iFDCV ( = .77, F2, 10 = 21.66, p < .001). The majority of functional traits (releasing height, biomass, specific leaf area, leaf dry matter content, Fv/Fm, and performance index) were related to abiotic habitat conditions indicating responses to environmental heterogeneity. In contrast, only morphology‐related functional traits (releasing height, biomass, and leaf area) were related to genetics. Our results suggest that both within‐habitat heterogeneity and genetic diversity affect iFDCV and are thus crucial to consider when aiming to understand or predict changes of plant species performance under changing environmental conditions.  相似文献

Response to selection depends on heritable genetic variation, which is affected by environmental conditions. The present study experimentally assessed whether the effect of light-related stress and the attenuating effect of shade as a facilitator of seedling germination, survival and growth affect the expression of heritable variation and the potential for a response to selection in the columnar cactus Pilosocereus leucocephalus. A reciprocal transplant experiment combined with the artificial manipulation of light/shade conditions within greenhouses was performed using seeds from controlled crosses of two natural populations (demes PN and SI). Additive genetic variance (VA), heritability (h2) and the coefficient of variation of additive variance (CVA) were estimated for per cent of germination, per cent of seedling survival and growth (biomass) under each treatment combination. Although all three recruitment traits showed evidence of different from zero heritability, this result was highly dependent upon the particular transplant site, deme and light treatment combination. The deme that is still not locally adapted (SI) showed significant heritability for all traits and much more potential for a response selection as indicated by a higher CVA than the locally adapted deme PN. The effect of light conditions on the expression of VA, h2 and CVA depended on whether the deme was grown in its native or an alien site, but this interaction was only detected for the less adapted deme of SI. Shade conditions promoted by facilitation reduced the evolutionary potential for germination of both demes through an attenuation of genetic differences among genotypes.  相似文献

Five species (14 ecotypes) belonging to three subgenera of ornamental-medicinal Iranian Fritillaria plant were chromosomally and karyotypically assessed, using squash technique and 1 % (w/v) aceto-orcein stain. All species were diploid (2n = 2x = 24) having mean chromosome length of 16.8 μm (14.2–18.6 μm). The satellites varied in number (1–4 pairs) and in size (1.27–3.01 μm): mostly locating on long arms. Four chromosome types (“m”, “sm”, “st”, “T”) formed 11 different karyotypic formulas: the latter is being reported for the first time in some ecotypes in either S1 or S4. Nine chromosomal parameters were calculated. ANOVA verified intra- and inter-specific chromosomal variation in examined Iranian Fritillaria species. Twelve different methods were used to assess the degree of karyotype asymmetry. Among those, one qualitative (Stebbins classification) and seven quantitative (TF %, CVTL, DI, AsK %, A2, Rec, CG %) parameters verified that S2 and S5 species were recognized as having the most asymmetrical and symmetrical karyotypes, respectively.  相似文献

Fluorescent chromosome banding and measurements of nuclear DNA content by image cytometry of Feulgen-stained cells were performed in one sample each of eight diploid (2n?=?24) species of Solanum: S.?endoadenium, S.?argentinum, S.?pseudocapsicum, S.?atropurpureum, S.?elaeagnifolium, S.?sisymbriifolium, S.?chenopodioides, and S.?palustre. The species studied could be distinguished by heterochromatin amount, banding patterns, and genome size. They exhibited only GC-rich heterochromatin and showed a comparatively low heterochromatin amount (expressed as percentage of haplotype karyotype length), ranging from 2.10 in S.?argentinum to 8.37 in S.?chenopodioides. Genome size displayed significant variation between species, with 1C-values ranging from 0.75?pg (735?Mbp) in S.?palustre to 1.79?pg (1,754?Mbp) in S.?sisymbriifolium. No significant correlation between genome size and heterochromatin amount was observed, but intrachromosomal asymmetry index (A 1) was negative and significantly correlated with heterochromatin amount. DNA content was positively and significantly correlated with karyotype length. DNA C-value distribution in the genus as well as karyotype affinities and relationships between species are discussed in relation to different infrageneric classifications of Solanum.  相似文献

In order to explore ontogenetic variation in leaf-level physiological traits of Betula pendula trees, we measured changes in mass- (A mass) and area-based (A area) net photosynthesis under light-saturated conditions, mass- (RSmass) and area-based (RSarea) leaf respiration, relative growth rate, leaf mass per area (LMA), total nonstructural carbohydrates (TNC), and macro- and micronutrient concentrations. Expanding leaves maintained high rates of A area, but due to high growth respiration rates, net CO2 fixation occurred only at irradiances >200 μmol photons m–2 s–1. We found that full structural leaf development is not a necessary prerequisite for maintaining positive CO2 balance in young birch leaves. Maximum rates of A area were realized in late June and early July, whereas the highest values of A mass occurred in May and steadily declined thereafter. The maintenance respiration rate averaged ≈8 nmol CO2 g–1 s–1, whereas growth respiration varied between 0 and 65 nmol CO2 g–1 s–1. After reaching its lowest point in mid-June, leaf respiration increased gradually until the end of the growing season. Mass and area-based dark respiration were significantly positively correlated with LMA at stages of leaf maturity, and senescence. Concentrations of P and K decreased during leaf development and stabilized or increased during maturity, and concentrations of immobile elements such as Ca, Mn and B increased throughout the growing season. Identification of interrelations between leaf development, CO2 exchange, TNC and leaf nutrients allowed us to define factors related to ontogenetic variation in leaf-level physiological traits and can be helpful in establishing periods appropriate for sampling birch leaves for diagnostic purposes such as assessment of plant and site productivity or effects of biotic or abiotic factors. 以欧亚温带草原东缘生态样带为平台,以样带上未放牧和重度放牧配对样地为研究对象,开展重度放牧对欧亚温带典型草原土壤氮矿化及其温度敏感性的影响研究。结果表明:(1)在室内培养条件下,土壤氮积累量和土壤净氮矿化速率呈现出干燥度指数较大的样点显著大于干燥度指数最低的样点(P0.05)。在相对湿润的样点,土壤氮素矿化周转速率较快;(2)重度放牧对不同样点土壤氮积累量和土壤氮矿化速率的影响是不同的。在干燥度指数较高样点,重度放牧样地土壤铵态氮减少量和速率较未放牧样地低(P0.05),重度放牧显著降低了土壤硝态氮积累量、无机氮积累量、硝化速率、净氮矿化速率(P0.05);在干燥度指数较低样点,重度放牧样地土壤铵态氮减少量和速率较未放牧样地高(P0.05),重度放牧对土壤硝态氮积累量、无机氮积累量、硝化速率、净氮矿化速率影响不大(P0.05);(3)土壤氮矿化作用温度敏感性(Q_(10))变化范围在1.61—2.06,重度放牧对Q_(10)无显著影响。随着纬度的升高,Q_(10)呈升高趋势。Q_(10)与基质质量指数以及表观活化能与基质质量指数均呈显著的负相关关系(P0.05);(4)土壤硝态氮积累量、无机氮积累量、硝化速率、净氮矿化速率对重度放牧的响应比与干燥度指数呈极显著负相关(P0.01),重度放牧对欧亚温带典型草原土壤氮矿化的影响受气候条件(温度和降水)的调控。  相似文献

Cytogenetic characterization by karyotyping and determination of DNA content by flow cytometry of five species of Chrysolaena (Vernonieae, Asteraceae) was performed. This is the first study of nuclear DNA content realized in the genus. The 2C-values were compared with the ploidy level and the total karyotype length (TKL) of each species. Mitotic analysis revealed a base chromosome number x = 10 for all entities and different ploidy levels, from diploid (2n = 2x = 20) to octoploid (2n = 8x = 80). All species showed bimodal karyotypes composed of metacentric and submetacentric chromosomes. The average chromosome size (ML) varied from 1.86 μm to 2.70 μm, while the TKL ranged from 18.65 μm to 80.55 μm. The intrachromosomal asymmetry index (A1) varied from 0.27 to 0.38, while the interchromosomal asymmetry index (A2) ranged from 0.19 to 0.25. A new cytotype is reported for the first time for C. propinqua. Accessory chromosomes found in C. verbascifolia, C. cognata, C. flexuosa, and C. propinqua are also reported as new.  相似文献

Assessing the genetic adaptive potential of populations and species is essential for better understanding evolutionary processes. However, the expression of genetic variation may depend on environmental conditions, which may speed up or slow down evolutionary responses. Thus, the same selection pressure may lead to different responses. Against this background, we here investigate the effects of thermal stress on genetic variation, mainly under controlled laboratory conditions. We estimated additive genetic variance (VA), narrow-sense heritability (h2) and the coefficient of genetic variation (CVA) under both benign control and stressful thermal conditions. We included six species spanning a diverse range of plant and animal taxa, and a total of 25 morphological and life-history traits. Our results show that (1) thermal stress reduced fitness components, (2) the majority of traits showed significant genetic variation and that (3) thermal stress affected the expression of genetic variation (VA, h2 or CVA) in only one-third of the cases (25 of 75 analyses, mostly in one clonal species). Moreover, the effects were highly species-specific, with genetic variation increasing in 11 and decreasing in 14 cases under stress. Our results hence indicate that thermal stress does not generally affect the expression of genetic variation under laboratory conditions but, nevertheless, increases or decreases genetic variation in specific cases. Consequently, predicting the rate of genetic adaptation might not be generally complicated by environmental variation, but requires a careful case-by-case consideration.Subject terms: Evolutionary genetics, Climate-change ecology, Biodiversity  相似文献

The present work provides new data on the banding pattern of diploid Cobitis taenia and its triploid hybrid females, which belong to the diploid–polyploid complex in the Vistula River tributary. C-banding, silver-staining (Ag), and fluorescent staining with chromomycin A3 techniques were used to describe the diploid and triploid karyotype. The karyotype of Cobitis taenia of 2n=48 was characterised by one pair of NOR-bearing subtelocentric chromosomes and at least four chromosomes with CMA3-positive sites. The C-positive heterochromatin was present in the centromeres of almost all chromosomes and the pericentromeric regions of several metacentric and submetacentric chromosomes. The triploid females of 3n=74 had two pairs of chromosomes with active NORs. The NORs-sites were located terminally on two biarmed and two uniarmed chromosomes. The CMA3-staining revealed at least six A3-positive sites. The C-banded and A3-stained triploid karyotype was composed of haploid set of Cobitis taenia and diploid set of unidentified species, so heterochromatin pattern confirmed the possibility of their hybrid origin. The characteristics of banded diploid and triploid karyotype, and the hypothetical karyotype of an unknown species of 2n=50 is discussed. Karyotypic characters, mitotic metaphase chromosomes, monoploid idiograms and karyograms of Minuartia anatolica (Boiss.) Woronow var. phrygia (Bornm.) McNeill, Minuartia anatolica (Boiss.) Woronow var. scleranthoides (Boiss. & Noe) McNeill, Minuartia corymbulosa (Boiss. & Balansa) McNeill var. gypsophilloides McNeill and Minuartia aksoyi M.Koç & Hamzao?lu were investigated for the first time. Analysis of somatic metaphases showed that the chromosome numbers and the formulas of these taxa were 2n = 24 = 14m + 6sm + 4st for Minuartia anatolica var. phrygia, 2n = 14 = 6m + 8sm for Minuartia anatolica var. scleranthoides, 2n = 14 = 6m + 4sm + 4st for Minuartia corymbulosa var. gypsophilloides and 2n = 30 = 14m + 10sm + 6st for Minuartia aksoyi. No satellites were observed in the karyotypes of these taxa. Karyotype asymmetry was estimated by many different methods, namely the Stebbins classification, the karyotype asymmetry index (As K %), the total form percent (TF %), the Rec and Syi indices, the intrachromosomal asymmetry index (A1) and interchromosomal asymmetry index (A2), the dispersion index (DI), the degree of asymmetry of karyotype (A index) and the asymmetry index (AI).  相似文献

To investigate annual variation in soil respiration (R S) and its components [autotrophic (R A) and heterotrophic (R H)] in relation to seasonal changes in soil temperature (ST) and soil water content (SWC) in an Abies holophylla stand (stand A) and a Quercus-dominated stand (stand Q), we set up trenched plots and measured R S, ST and SWC for 2 years. The mean annual rate of R S was 436 mg CO2 m−2 h−1, ranging from 76 to 1,170 mg CO2 m−2 h−1, in stand A and 376 mg CO2 m−2 h−1, ranging from 82 to 1,133 mg CO2 m−2 h−1, in stand Q. A significant relationship between R S and its components and ST was observed over the 2 years in both stands, whereas a significant correlation between R A and SWC was detected only in stand Q. On average over the 2 years, R A accounted for approximately 34% (range 17–67%) and 31% (15–82%) of the variation in R S in stands A and Q, respectively. Our results suggested that vegetation type did not significantly affect the annual mean contributions of R A or R H, but did affect the pattern of seasonal change in the contribution of R A to R S.  相似文献
