固态下酵母自溶可以有效促进菌体内多种活性物质的释放,进而提高酵母类产品的品质。通过优化自溶温度、自溶时间及自溶促进剂锌离子浓度以获得固态发酵下酵母自溶的最佳工艺,对固态发酵物料中游离氨基酸、可溶性蛋白、α-氨基氮含量和A260/A280等指标的分析来确定固态酵母自溶工艺条件,在此基础上以自溶温度40 ℃、50 ℃、55 ℃;作用时间12、18、24 h;锌离子添加浓度2、4、8 mg/kg设置L9(33)正交试验,进一步优化固态酵母自溶的工艺参数。结果表明酵母自溶的最佳工艺条件为:自溶温度55 ℃、作用时间18 h、锌离子浓度2 mg/kg,此时其可溶性蛋白含量可达9.31 mg/g、游离氨基酸14.36 mg/g、α-氨基氮10.16 μg/g、A260/A280为1.73。经工艺优化后,可显著提高酵母自溶产物可溶性蛋白、游离氨基酸和α-氨基氮的含量,从而明显提高了复合菌培养物的品质。  相似文献

为探究短时微流水处理对池塘养殖鳙鱼(Aristichthys nobilis)肌肉滋味品质的影响, 采用微流水装置对池塘养殖鳙鱼进行处理, 研究处理时间(0、2d、4d、6d、8d和10d)对池塘养殖鳙鱼肌肉中基本营养成分、游离氨基酸、脂肪酸、核苷酸及其降解产物、甜菜碱含量和感官评分的影响, 以期为提升鳙鱼滋味品质提供新方法。结果表明: 随着微流水处理时间的延长, 鳙鱼肌肉中总游离氨基酸含量、甜味氨基酸含量、腺苷酸(Adenosine monophosphate, AMP)含量和甜菜碱含量呈现先增加后降低的趋势, 均在处理6d时含量最高, 较未处理组分别增加了17.74%、39.45%、28.00%和50.16%; 灰分和鲜味氨基酸含量呈现增加的趋势; 粗脂肪、总脂肪酸含量、饱和脂肪酸含量、单不饱和脂肪酸含量、多不饱和脂肪酸含量、苦味氨基酸含量、酸味氨基酸含量、次黄嘌呤核苷(Hypoxanthine riboside, HxR)和次黄嘌呤(Hypoxanthine, Hx)含量则逐渐降低; 粗蛋白含量在处理过程中无显著变化。滋味分析仪(电子舌)和感官评分结果表明, 微流水处理显著提高了鳙鱼肌肉鲜味和甜味, 降低了其咸味、苦味和腥味, 且处理6d时鳙鱼肌肉的滋味、气味、质地和色泽感官评分最高。故短时微流水处理可显著提高鳙鱼的肌肉滋味品质, 不降低营养品质, 适宜的处理时间为6d。  相似文献

本论文探究了培养条件对德氏乳杆菌保加利亚亚种KLDS 1.9201和唾液链球菌嗜热亚种KLDS 3.021自溶的影响.结果表明,随着培养温度(30℃~55℃)、盐浓度(0%~5%)以及酸度值(pH5.0~7.5)的升高,KLDS 1.9201和KLDS 3.0201不仅自溶度相应增大,而且在磷酸缓冲液中释放的核酸和蛋白含量也呈递增趋势.高温55℃,pH 7.5和5%乳酸盐浓度的培养环境使KLDS 1.9201和KLDS 3.0201在培养45 h后的自溶度分别达到了55%和36.3%,显著高于低自溶培养环境下(30℃、pH 5.5和0%乳酸盐)KLDS 1.9201和KLDS 3.0201的自溶度9.96%和8.6%(P<0.05).透射电镜观察表明高自溶和低自溶培养条件对酸奶菌株菌体形态的影响是截然不同的,且酸奶菌株自溶时发生的细胞壁降解程度与自溶光密度值相对应.  相似文献

用200~300nm的波长范围对富铬酵母及普通酵母的溶液进行紫外扫描,发现在λ_(260nm)处有一特征紫外吸收峰,富铬酵母细胞的铬含量与其吸收峰的光密度呈线性关系;在不同的温度和pH值条件下,通过紫外吸收峰的测定,富铬酵母溶液在酸性条件下稳定,在碱性条件下不稳定。溶液的λ_(260nm)紫外吸收峰随着温度的上升而升高。从氨基酸含量分析结果看,富铬酵母的谷氨酸、甘氨酸、组氨酸、丙氨酸和赖氨酸含量高于普通酵母,但其他氨基酸含量比普通酵母低。  相似文献

以珍珠贝母为原料 ,通过细胞自溶破壁 ,使牛黄酸溶出 ,以离子交换树脂法提取纯化溶液中牛磺酸。实验结果表明 :细胞自溶破壁的最佳条件为温度 4 0℃ ,p H5.5,自溶时间 4 0 h;纯化可使牛磺酸量占氨基酸总量的 80 .6%,回收率为 87.5%。  相似文献

啤酒酿造过程中正常情况下的酵母不会发生自溶,但如果细胞衰老死亡或操作不当则会出现酵母自溶现象.虽然自溶的细胞在生产中占的比例较小,但自溶后的酵母将胞内物质释放到酒液中,产生酵母味,严重影响了啤酒的口感和外观质量.早期研究认为酵母细胞壁在酵母自溶过程中不发生水解,即自溶后的细胞仅剩下空的细胞外壳.现在研究发现酵母在自溶时其细胞壁也会发生水解作用,葡聚糖酶将构成细胞壁成分的葡聚糖进行分解,分解产物最终释放到酒液里和细胞质内物质共同影响啤酒的质量.  相似文献

以聚乙烯醇为唯一碳源从环境中筛选获得了高效降解聚乙烯醇的微生物菌株XT11,初步鉴定为假单胞菌属(Pseudomonas sp.).对菌株Pseudomonas XT11的生长过程及PVA降解过程进行了研究,发现该菌株在54 h内可将1 g/L的聚乙烯醇(PVA)降解.同时研究了温度、pH值及酵母膏浓度对该菌株降解PVA的影响,结果表明其最适温度、pH值和酵母膏浓度分别为30℃、7.0和0.5 g/L.研究了PVA浓度对PVA降解率的影响,发现随着PVA浓度的增大,PVA的降解率降低.  相似文献

细胞自噬(autophagy)是一种主要由溶酶体介导的降解通路,作为细胞维持内环境稳态的一种保护性机制,不仅通过将长寿命蛋白和衰老细胞器降解为小肽或氨基酸为细胞提供再生资源,而且也可作为防御机制抵抗病原微生物感染和寄生. 自噬缺失与许多疾病如癌症、心血管疾病等的发生关系密切,在机体生理、病理过程中发挥重要作用. 本文拟就细胞自噬与病毒感染、机体免疫的关系加以综述,以期为研究细胞自噬的发生、参与机体免疫、发挥抗病毒感染作用及其分子机制提供参考,也为进一步研究抗病毒治疗的靶标提供新思路.  相似文献

用常规方法测定了大菱鲆鱼皮的基本营养成分,用氨基酸自动分析仪分析其氨基酸构成,并对大菱鲆鱼皮中的氨基酸组成进行了营养学评价。结果表明,大菱鲆鱼皮蛋白质含量为14.8%,脂肪含量为1.8%,氨基酸含量为27.24%,其中含量较高的是甘氨酸、丙氨酸、脯氨酸和精氨酸,占总氨基酸的59.73%;必需氨基酸、甜味氨基酸、苦味氨基酸、鲜味氨基酸和涩味氨基酸的含量分别占总氨基酸的20.81%、60.61%、30.07%、10.43%和0.01%。甜味氨基酸、苦味氨基酸、鲜味氨基酸是大菱鲆鱼皮氨基酸的主要部分,构成了大菱鲆鱼皮的主要味道,大菱鲆鱼皮是一种口感柔滑、味道鲜美的理想补强食品。  相似文献

It has now been over twenty years since a novel herpesviral genome was identified in Kaposi's sarcoma biopsies. Since then, the cumulative research effort by molecular biologists, virologists, clinicians, and epidemiologists alike has led to the extensive characterization of this tumor virus, Kaposi's sarcoma-associated herpesvirus(KSHV; also known as human herpesvirus 8(HHV-8)), and its associated diseases. Here we review the current knowledge of KSHV biology and pathogenesis, with a particular emphasis on new and exciting advances in the field of epigenetics. We also discuss the development and practicality of various cell culture and animal model systems to study KSHV replication and pathogenesis.  相似文献

Comprises species occurring mostly in subtidal habitats in tropical, subtropical and warm-temperate areas of the world. An analysis of the type species, V. spiralis (Sonder) Lamouroux ex J. Agardh, a species from Australia, establishes basic characters for distinguishing species in the genus. These characters are (1) branching patterns of thalli, (2) flat blades that may be spiralled on their axis, (3) width of the blade, (4) primary or secondary derivation of sterile and fertile branchlets and (5) position of sterile and fertile branchlets on the thalli. Application of the latter two characters provides an important basic method for separation of species into three major groups. Osmundaria , a genus known only in southern Australia, was studied in relation to Vidalia , and its separation from the Vidalia assemblage is not accepted. Species of Vidalia therefore are transferred to the older genus name, Osmundaria. Two new species, Osmundaria papenfussii and Osmundaria oliveae are described from Natal. Confusion in the usage of the epithet, Vidalia fimbriala Brown ex Turner has been clarified, and Vidalia gregaria Falkenberg, described as an epiphyte on Osmundaria pro/ifera Lamouroux, is revealed to be young branches of the host, Osmundaria prolifera.  相似文献

Fifteen chromosome counts of six Artemisia taxa and one species of each of the genera Brachanthemum, Hippolytia, Kaschgaria, Lepidolopsis and Turaniphytum are reported from Kazakhstan. Three of them are new reports, two are not consistent with previous counts and the remainder are confirmations of very scarce (one to four) earlier records. All the populations studied have the same basic chromosome number, x = 9, with ploidy levels ranging from 2x to 6x. Some correlations between ploidy level, morphological characters and distribution are noted.  相似文献

采用原位分子杂交方法检测HCV RNA及HBV X基因;采用免疫组织化学方法研究HCV核心抗原,非结构区C33c抗原及HBxAg在肝细胞肝癌中的定位及分布.结果表明(1)HCV RNA、HBV X基因在肝细胞肝癌组织检出率分别为40%(55/136)和82%(112/136).HCV RNA定位于癌细胞的胞浆内,阳性细胞呈散在、灶状及弥漫分布三种形式;HBV X基因在肝癌细胞中的分布呈胞浆型、核型及核浆型,阳性细胞也呈上述三种分布形式;(2)HCV C33c抗原、核心抗原在肝细胞肝癌中的阳性率为81%(133/164)及86%(141/164).C33c抗原定位于癌细胞及肝细胞的胞浆内;核心抗原既定位于癌细胞核中,又可定位于胞浆中.C33c抗原阳性细胞以灶状分布为主;而核心抗原阳性细  相似文献

For a plant selection model with frequency-independent viabilities, fertilities and selfing rates, it is shown that apart from global fixation, for certain parameter combinations a protected polymorphism and facultative fixation (either allele may become fixed according to initial frequencies) may both occur. Facultative fixation requires different selling rates for the dominant and recessive type. Protection of the polymorphism requires resource allocation for male and female function. In this connection the problem of purely genetically caused population extinction is discussed.
For general frequency dependence and regular segregation, the chances for establishment of a completely recessive gene are compared to those of a completely dominant gene. It is proven that the process of establishment of the recessive gene, despite a fitness advantage, may be considerably endangered by drift effects if random mating prevails. The recessive gene may reach the same effectivity in establishment as a dominant gene, only if the recessive homozygote mates exclusively with its own type during the period of establishment.  相似文献
