Quercus ilex L. (holm oak) coppices, widespread around the Mediterranean basin, are probably the result of 5 000 years of prolonged human disturbance of the original Quercus pubescens Willd. (downy oak) forests. Since disturbance has almost ceased in recent years, a question arises as to the development of these coppices: will the Q. pubescens forests return, or will Q. ilex remain the dominant species? To investigate the phenomenon, we analyzed the first stages, i.e. germination of the two species in holm oak coppices. Our experiments show that both species germinated better in coppices than in clearings or clear-cuts. Moreover, Q. pubescens appears to be slightly favored above Q. ilex and it is suggested auto-allelopathy is involved, at least partially inhibiting the germination of Q. ilex.  相似文献

Chloroplast (cp) and mitochondrial (mt) DNA variation were studied in 97 populations of cork oak (Quercus suber) in Morocco; in 31 of these populations, holm oak (Quercus ilex), a clearly distinct species, also occurred and was compared with Q. suber. Three cpDNA and one mtDNA primer pairs were used in the survey, each in combination with one restriction enzyme. Six haplotypes belonging to two very divergent lineages were detected; one lineage predominates in each species, and is probably ancestral, as inferred from comparisons with other oak species. In the mixed-species populations, cytoplasmic genomes were frequently shared across species, as indicated by an introgression ratio of 0.63. This index is a new measure of the propensity of species to share locally genetic markers, varying from zero (complete differentiation) to one (no differentiation). By contrast, more closely related deciduous oak species (Q. robur, Q. petraea and Q. pubescens) have introgression ratios varying from 0.82 to 0.97. The introgression events appear to have been more frequent in the direction Q. ilex (female) x Q. suber (male), a finding which seems attributable to the flowering phenology of these two species. This asymmetry may have favoured immigration of Q. suber beyond its main range, in regions already colonized by Q. ilex. There, rare hybridization and further introgression through long distance pollen flow have established populations that are morphologically indistinguishable from Q. suber but that have cytoplasmic genomes originating from the local Q. ilex populations.  相似文献

Gene flow is particularly frequent in the genus Quercus (oaks), especially between closely related species. We focus here on Quercus ilex and the cork-producing Quercus suber , which occasionally hybridize although they are phylogenetically markedly separated. Morphological observations were combined with both allozymic and chloroplastic diagnostic markers to characterize hybridization and introgression and to infer their dynamics in two French regions (French Catalonia and Provence), which are separated by several hundred kilometres. Some hybrids were found in both regions, indicating recent hybridization events. As expected from previous studies, most hybrids resulted from ♀ Q. ilex  × ♂ Q. suber crosses, but our data showed that the reciprocal cross is also possible. Partial independence between nuclear and chloroplastic introgression was observed in the two species. Nuclear introgression was limited in both species and both regions, with no preferred direction. In Provence, chloroplastic introgression was very rare in both species. Conversely, all Q. suber individuals from French Catalonia were introgressed by Q. ilex chlorotypes. This might be explained by introgression in the Iberian Peninsula antedating the first occurrence of the two species in French Catalonia. We also observed a new chlorotype that was created locally, and was exchanged between the two species. However, the two species still remain genetically differentiated. The dynamics and complexity of exchanges and the factors determining them (including human management of Q. suber ) are discussed.  相似文献

Background and Aims

Quercus suber and Q. ilex are distantly related and their distributions partially overlap. They hybridize occasionally, but the complete replacement of Q. suber chloroplast DNA (cpDNA) by that of Q. ilex was identified in two specific geographical areas. The objective of this study was to determine whether the contrasting situation reflected current or recent geographical interspecies gene flow variation or was the result of ancient introgression.

Methods

cpDNA PCR-RFLPs (restriction fragment length polymorphisms) and variation at ten nuclear microsatellite loci were analysed in populations of each species, in 16 morphologically intermediate individuals and the progeny of several of them. Interspecies nuclear introgression was based on individual admixture rates using a Bayesian approach with no a priori species assignment, and on a maximum-likelihood (ML) method, using allele frequencies in the allopatric populations of each species as controls. Gene flow was compared specifically between populations located within and outside the specific areas.

Key Results

High interspecies nuclear genetic differentiation was observed, with twice the number of alleles in Q. ilex than in Q. suber. According to Bayesian assignment, approx. 1 % of individuals had a high probability of being F1 hybrids, and bidirectional nuclear introgression affected approx. 4 % of individuals in each species. Hybrid and introgressed individuals were identified predominantly in mixed stands and may have a recent origin. Higher proportions including allospecific genes recovered from past hybridization were obtained using the ML method. Similar rates of hybridization and of nuclear introgression, partially independent of cpDNA interspecies transfer suggestive of gene filtering, were obtained in the populations located within and outside the areas of complete cpDNA replacement.

Conclusions

The results did not provide evidence for geographical variation in interspecies gene flow. In Central-Western Spain, forests and woodlands composed of Quercus sp. support outstanding levels of biodiversity, but there is increasing concern about their long-term persistence due to a lack of regeneration. We hypothesize that this regenerative lack is operating on a large geographic scale; that there are differences in the abundance of regeneration between three oak species; that oak regeneration is governed mainly by forest management and structure; and that shrubs act as important physical protectors of seedlings and saplings. We analyzed whether densities of oak seedlings and saplings in several size classes were related to stand-structure, understory, and physiographic variables potentially affecting regeneration. Data collected at a regional level (1 km × 1 km grid) by the Spanish Forest Inventory were evaluated from 2,816 plots. Results revealed that regeneration failure was common for all size categories, from small seedlings to large saplings, and for the three oak species studied, especially the evergreens. Of the Quercus ilex, Q. suber, and Q. pyrenaica plots studied, 49%, 62%, and 20% were lacking any small seedlings, and 82%, 96%, and 56% did not have any large saplings, respectively. Regeneration was positively correlated with tree cover and density, especially of small and medium-sized trees, and negatively correlated with the presence of large trees, indicating that regeneration failure is mostly associated with more open, uniform, and/or aged woodlands. Regeneration densities of Q. ilex and Q. suber were positively correlated with all understory variables, suggesting that the presence of pioneer shrubs represent a major safe site for early tree recruitment, independent from specific shrub species.  相似文献

Abstract

Random Amplified Polymorphic DNA (RAPD) and Inter-Simple Sequence Repeats (ISSR) markers were employed to examine samples from Quercus cerris, Q. suber and Q. crenata in order to test the hypothesis of the hybrid origin of Q. crenata from Q. cerris and Q. suber in a part of its distribution area where the two putative parents do not overlap at present. Leaves from 21 Q. crenata and 37 Q. cerris individual trees were collected at natural sites in northern Italy, where Q. suber is currently lacking; 21 Q. suber and six Q. crenata plants from central Italy were also analysed. Results from Unweighted Pair Group Method with Arithmetic mean (UPGMA) analysis and principal component analysis (PCA) implied that exchange of neutral markers has been considerable between the three species, while differences in morphological characters have remained comparatively stable. The Mantel test indicated low correlation between RAPD- and ISSR-based similarity matrices, showing that the two screening techniques reveal unrelated estimates of genetic relationships. Hybrid indices computed for both markers displayed an intermediate position of Q. crenata individuals between the two putative parents shifted toward Q. cerris. Results from the present study corroborate the hypothesis of a hybrid origin for Q. crenata occurring in northern Italy, and suggest asymmetrical backcrossing with Q. cerris acting as the recurrent parent.  相似文献

This paper describes research on the application of tissue culture techniques to the micropropagation of cork oak (Quercus suber L.), a forest species of ecological and industrial importance in the Mediterranean area. Apical buds and nodal stem segments were employed as initial explants. Their origins were young seedlings, stump sprouts and sprouts formed on cuttings collected from old trees.The action of the mineral medium and growth regulators was studied in the multiplication stage. Media with low concentrations of ions, such as Sommer's or Heller's, are more suitable for growth and proliferation of explants than other media richer in salts. It was also observed that cytokinin (BA) must be present for the culture development. Adding low concentrations of auxin (NAA) to the medium improves the multiplication rate, especially in vegetative material of adult origin.The auxin type is the most important factor in the promotion of rhizogenesis. The method of application determines the quality of the root system. Treatment with low concentrations of IBA added to the rooting medium gives the best results.High sucrose concentration also improves rooting. Identifying patterns of fine-scale genetic structure in natural populations can advance understanding of critical ecological processes such as dispersal and gene flow across heterogeneous landscapes. Alpine ungulates generally exhibit high levels of genetic structure due to female philopatry and patchy configuration of mountain habitats. We assessed the spatial scale of genetic structure and the amount of gene flow in 301 Dall’s sheep (Ovis dalli dalli) at the landscape level using 15 nuclear microsatellites and 473 base pairs of the mitochondrial (mtDNA) control region. Dall’s sheep exhibited significant genetic structure within contiguous mountain ranges, but mtDNA structure occurred at a broader geographic scale than nuclear DNA within the study area, and mtDNA structure for other North American mountain sheep populations. No evidence of male-mediated gene flow or greater philopatry of females was observed; there was little difference between markers with different modes of inheritance (pairwise nuclear DNA F ST = 0.004–0.325; mtDNA F ST = 0.009–0.544), and males were no more likely than females to be recent immigrants. Historical patterns based on mtDNA indicate separate northern and southern lineages and a pattern of expansion following regional glacial retreat. Boundaries of genetic clusters aligned geographically with prominent mountain ranges, icefields, and major river valleys based on Bayesian and hierarchical modeling of microsatellite and mtDNA data. Our results suggest that fine-scale genetic structure in Dall’s sheep is influenced by limited dispersal, and structure may be weaker in populations occurring near ancestral levels of density and distribution in continuous habitats compared to other alpine ungulates that have experienced declines and marked habitat fragmentation.  相似文献

Seasonal changes in pigment composition of sun and shade leavesof cork oak (Quercus suber) were studied under field conditionsin Portugal. Expanding leaves showed a high concentration ofxanthophyll cycle components, violaxanthin, antheraxanthin andzeaxanthin. The pool of violaxanthin plus antheraxanthin pluszeaxanthin (V+A+Z) varied greatly between the seasons, beinghigher at the end of summer and in winter when photosynthesiswas limited by water stress and cold, respectively. The sizeof V+A+Z pool was associated to synthesis of zeaxanthin in responseto an excess of light. In sun leaves, midday A+Z relative contentwas positively correlated with the V+A+Z pool, whereas in shadeleaves A+Z decreased with leaf ageing. In both leaf types A+Zwas positively correlated with the non-photochemical quenching(NPQ) of chlorophyll a fluorescence. However, in winter NPQdid not change significantly throughout the day, whereas the(A+Z)/(V+A+Z) increased following the typical daily trend observedin other seasons. The potentially confounded effects of factors affecting breeding dispersal have rarely been simultaneously examined. The consequences of breeding dispersal are even less studied, presenting a paradox: breeding dispersal seldom seems to improve breeding success, despite its presumed adaptiveness. We studied the causes and consequences of breeding dispersal in female-philopatric eiders (Somateria mollissima) in relation to the spatiotemporal predictability of nest success. Previous nest fate, breeding experience, and breeding density simultaneously affected breeding dispersal. Dispersal distances were longer among inexperienced breeders and after failed breeding. Individual dispersal distances decreased with increasing nest-site-specific breeding density, whereas island-specific nesting success peaked at intermediate densities. The fate of neighbouring nests (‘public information’) did not influence dispersal. Breeding dispersal was unrelated to subsequent hatching success, controlling for individual quality (body condition, breeding experience, previous nest fate), while it delayed hatch date, which is likely to impair reproductive success. This delay may result from the loss of acquired information of local breeding conditions, prolonging nest prospecting and establishment, also helping explain why breeding dispersal did not increase at high breeding densities, despite a potential reduction in nesting success. In long-lived species, however, dispersal-induced reductions in reproductive output in one season could be offset by improved parental survival prospects. Careful nest prospecting may be profitable, because overall nest success had a strong island-specific component but showed weak temporal variation, and successive individual nest fates were predictable between years. Once a safe nest site is found, females may breed at the same place successfully for many years.  相似文献

Procedures have been developed to standardize the multiplicationstage during mature cork-oak (Quercus suber L.) micropropagation.Axillary and terminal buds were established on Gresshoff andDoy basal medium containing 1 mg I–1 of 6-benzylaminopurine(BAP). Initiation of cultures was possible all over the year.The effects of BAP, Z, IBA, 1AA and NAA and various nutrientformulae on shoot growth and proliferation was investigated.BAP was more suitable than zeatin. Shoot proliferation and elongationwere strongly improved by the combination BAP/IAA in the presenceof low salt media, like Gresshoff and Doy or Woody Plant medium.Both rates were significantly increased when a double-phaseculture system was used. Shoots have been multiplied for 1 yearat the rate of three to four-fold every 4 weeks without anydecline of vigour. Rooting was achieved by briefly dipping thebasal ends of in vitro regenerated shoots in an IBA concentratedsolution. The results here reported constitute a promising steptowards large scale in vitro propagation of a species in whichconventional vegetative propagation by cuttings is very difficult. The phenology of Quercus suber L., a dominant species of the montados in the Iberian Peninsula, was studied for 2 years in southwest Portugal. The seasonal progression of phenological events was analyzed in seven trees. Selected branches were examined monthly for shoot elongation, leaf number, branching, flowering, and fruiting. Radial stem growth and specific leaf weight were also studied. Active growth was observed from early spring to early summer. Reserves accumulated during winter and high photosynthetic activity in early spring apparently supported this strong development. The growth flush started with stem radial increment, which seemed to be impaired by spring rainfall. Male inflorescence production was the next phenological event. Old leaves were shed during new twig and leaf emergence. Shoot elongation and the number of new leaves produced were well correlated with the previous-year shoot's length, and were not clearly related to climatic factors. Radial growth resumed in autumn at a lower rate than in the previous spring, a possible consequence of a reserve depletion due to lower photosynthetic production in summer and investment on fruit maturation, which was complete by late autumn. Premature and excessive new leaf production were apparently subjected to self-pruning strategies related to the development of each tree's crown. Younger cork-oaks produced shorter and fewer shoots per module, and more sclerophyllous leaves than the older ones. A high intra-specific variability was observed in all the results.  相似文献

The regeneration of somatic seedlings from selected 100-year-old cork oak trees is reported. The induction of somatic embryogenesis from leaves of epicormic shoots was significantly affected by genotype, harvesting time and their interaction. Leaves from all five selected trees produced somatic embryos when the segments of branches used as sources of epicormic shoots were collected in May. Genotype, but not the level of photosynthetically active radiation, affected the proliferation of the embryogenic lines and the number of detachable embryos that could be obtained from them. Genotype also affected several steps leading to conversion of somatic embryos, from germination to complete acclimatisation of somatic seedlings. Almost 40% of the somatic embryos from all lines germinated, showing coordinated root and shoot growth. Although the mean percentage of recovery for the whole process was low, plants could be regenerated from four of the five trees tested.  相似文献

Abstract

Drought treatments in holm-oak (Quercus ilex) seedlings induce variations in total root length, number of root apices, shoot/root dry weight, and root electrolyte leakage. When drought treatments last for more than 50 days a considerable number of fine lateral roots die, irrespective of branching order or distribution within the root system. Scorching of drought-treated seedlings induces a transient stimulation of root growth. These results indicate that root turnover is deeply affected during treatments, with survival of seedlings being entrusted to the tolerance of a number of roots situated in the deeper region of the root system. Activity of the meristematic tissue present within the apices of these surviving roots supports regeneration of above-ground lost organs during recovery. Knowledge of the mechanisms ensuring the survival of Mediterranean tree seedlings following drought and fire is useful for developing models of vegetation dynamics.  相似文献

Very similar genome sizes, similar karyotypes and heterochromatin organisation, and identical number/position of ribosomal loci characterise the common oak (Q. robur) and the cork oak (Q. suber), two distantly related oak species. Representational Difference Analysis (RDA) was used to subtract the genome of Q. suber from the genome of Q. robur in order to search for genome differentiation. A library of 400 clones (bearing RDA fragments) representing genome differences between the two species was obtained. Seven Q. robur-specific DNA sequences were analysed with respect to their molecular and chromosome organisation. All belong to the dispersed repetitive component of the genome, as revealed by Southern hybridisation and in situ hybridisation. They are present in the Q. robur genome in between 100 and 700 copies, and are distributed along the length of almost all chromosomes. A search for homologies between RDA fragments and sequences in Genbank revealed similarities of all RDA fragments with known retrotransposons. The RDA fragments were also tested for their presence/absence in the genomes of six additional oak species belonging to different phylogenetic groups, in order to examine the evolutionary dynamics of these DNA sequences.  相似文献

Summary Osmoticum (high sucrose concentration) and chilling (4°C) treatments were assayed in order to promote germination (radicle elongation) and plant conversion of Quercus suber somatic embryos. Those treatments were applied before embryos were set to germinate on Murashige and Skoog (MS) liquid medium. Chilling for 2 mo. improved germination rate (best results close to 100%). On the other hand, that treatment increased plant conversion, especially when high sucrose concentration was used. The best results were obtained with 150 g l−1 sucrose (75% of cultured embryos converted to plants). Incubation conditions during germination were also studied. Although there were no significant differences on final results, warmer temperatures and darkness increased germination rate. The influence of the different treatments on shoot morphology was also studied. The highest percentages of shoots with normal leaves obtained were close to 25%.  相似文献

Here, we provide the first report on flavonoid content in holm oak (Quercus ilex L.) leaves, analyzed by HPLC–MS/MS. Flavanols and flavonols were the predominant groups, although proanthocyanidins and many soluble tannins had a relevant presence in all leaf samples. Seasonal variation of flavonoids was determined in extracts from Q. ilex leaves during resprouting after a forest fire in two Mediterranean forests. Similar seasonal trends were observed over 2 years during the two main stress seasons (winter and summer). The most abundant flavonoid was the flavanol epicatechin, which showed similar values during the two seasons. Hexosides of the flavonols, quercetin, kaempferol and rhamnetin showed considerably higher content in winter, especially at the lowest temperatures. These variations in both forests are discussed on the basis of the chlorophyll fluorescence results obtained. Anthocyanins were found practically absent in mature leaves. Nutrient or water availability differences between sites or seasons were not related to changes in leaf flavonol-hexoside content.  相似文献

We have analysed the distribution of epigenetic marks for histone modifications at lysine residues H3 and H4, and DNA methylation, in the nuclei of mature pollen cells of the Angiosperm tree Quercus suber; a monoecious wind pollinated species with a protandrous system, and a long post-pollination period. The ultrasonic treatment developed for the isolation of pollen nuclei proved to be a fast and reliable method, preventing the interference of cell wall autofluorescence in the in situ immunolabelling assays. In contrast with previous studies on herbaceous species with short progamic phases, our results are consistent with a high level of silent (5-mC and H3K9me2) epigenetic marks on chromatin of the generative nucleus, and the prevalence of active marks (H3K9me3 and H4Kac) in the vegetative nucleus. The findings are discussed in terms of the pollination/fertilization timing strategy adopted by this plant species.  相似文献
