In Rhodobacter sphaeroides, transfer of the first electron in quinol oxidation by the bc(1) complex shows kinetic features (a slow rate (approx. 1.5 x 10(3)/s), high activation energy (approx. 65 kJ/mol) and reorganization energy, lambda (2.5 V)) that are unexpected from Marcus theory and the distances shown by the structures. Reduction of the oxidized iron-sulfur protein occurs after formation of the enzyme-substrate complex, and involves a H-transfer in which the electron transfer occurs through the approx. 7 A of a bridging histidine forming a H-bond with quinol and a ligand to 2Fe-2S. The anomalous kinetic features can be explained by a mechanism in which the electron transfer is constrained by coupled transfer of the proton. We discuss this in the context of mutant strains with modified E(m,7) and pK for the iron-sulfur protein, and Marcus theory for proton-coupled electron transfer. We suggest that transfer of the second proton and electron involve movement of semiquinone in the Q(o) site, and rotation of the Glu of the conserved -PEWY- sequence. Mutational studies show a key role for the domain proximal to heme b(L). The effects of mutation at Tyr-302 (Tyr-279 in bovine sequence) point to a possible linkage between conformational changes in the proximal domain, and changes leading to closure of the iron-sulfur protein access channel at the distal domain.  相似文献

Ras proteins regulate cell growth and differentiation, and their mutation plays a major, direct role in causing human cancer. For years, their precise function has been a mystery. One of the pathways Ras controls has recently been identified. It consists of a cascade of kinases (Raf, MEK and MAP kinases) that transmits signals from the plasma membrane to the nucleus. The role of Ras is remarkably simple: it recruits Raf from the cytoplasm to the plasma membrane. Once in the membrane, Raf is activated and the kinase cascade is initiated. In 2014, Islamist jihadist groups overran a Lebanese border town and besieged it for four days, spreading terror across the town and the country as a whole. In response, the Lebanese army launched a violent counterattack on these groups with the aid of Hizbullah in what became known as the Battle of Arsal. Declaring the area a security zone, the army restricted mobility and placed Lebanese citizens and Syrian refugees under military confinement. While the battle ended with a government-declared victory that celebrated the ‘reclamation of state sovereignty’, these events disrupted social life in unprecedented ways. This article focuses on the phenomenological experience of sovereignty and the price paid for the desire for this state. I explore the ruptures people experienced in their existence (their right to life, subsistence, and movement under indiscriminate shelling) and existentiality (their reflections on their human existence in time, place, and relationship to others in a social world that is no longer recognizable). I argue for a concept of existential displacement that focuses on the experience of citizens as well as refugees for a broader understanding of the ruptures and dispossessions taking place in conflict.  相似文献

The telomeres are the nucleoproteic structures present at the ends of eukaryotic chromosomes. One can compare them to the protective ends of a shoelace; when the ends get eroded, the shoelace disintegrates and we dispose of it. The same thus applies to the chromosomes; when telomeres reach a critical threshold for function, the genome becomes unstable and the cell senesces. Increased fire frequency can significantly erode both soil properties and plant–pollinator interactions affecting plant reproductive success but they have seldom been assessed simultaneously. Here, we evaluate soil properties, pollinator assemblage and the reproductive success of two native Fabaceae herbs, Desmodium uncinatum and Rhynchosia edulis, growing in unburned, low and high fire frequency sites of Chaco Serrano across two consecutive years. Desmodium uncinatum is outcrossing with a high dependence on pollinators, whereas R. edulis is autogamous and completely independent of pollinators. We found that soil water content, nitrates and electrical conductivity significantly decreased in low and high fire frequency sites. Pollinator richness and composition visiting each plant species was similar across all fire frequency scenarios. However, fruit set of the exogamous D. uncinatum was strongly reduced in frequently burned sites, whereas fruit set of the autogamous R. edulis showed no significant changes. In both species, the probability of setting fruits was positively related to soil quality across fire frequency scenarios, implying that decreased reproduction was mainly driven by limitation of abiotic resources shaped by increased fire frequency. Because the pollinator-dependent D. uncinatum has a higher reproductive cost, reduced soil quality induced by fire frequency had stronger effects on its reproduction. Chronic reduction of sexual reproduction in frequently burned sites with depleted soils will limit population recruitment with negative consequences on long-term plant population persistence.  相似文献

Although ketamine is widely used as an analgesic agent and has an anti-allodynic effect on neuropathic pain, the underlying analgesic mechanisms are not fully explained by the modern 'neuronal-based' theories. As emerging studies have focused on the critical role of spinal astrocytes in the pathological pain states, we have hypothesized that there exist some 'astrocytes-related' mechanisms in the analgesic function of ketamine. In the present study, using the spinal nerve ligation (SNL) pain model, we investigated the anti-nociceptive effects of intraperitoneal or intrathecal ketamine on SNL-induced neuropathic pain response, meanwhile, we investigated the astrocytic activation after ketamine administration on SNL rats. Behavioral data showed that either intraperitoneal or intrathecal ketamine inhibited SNL-induced allodynia, however, immunohistochemistry showed that SNL induced astrocytic activation was suppressed by intrathecal but not intraperitoneal ketamine. Using quantitative Western blot analysis, our report showed that intrathecal ketamine down-regulated glial fibrillary acidic protein expression, suggesting inhibition of SNL-induced astrocytic activation, which wasn't influenced by intraperitoneal administration. We conclude that intraperitoneal ketamine could alleviate SNL-induced neuropathic pain via the classical 'neuronal-based' mechanisms, but in addition, 'astrocytes-related' mechanisms were also important underlying the anti-allodynic effect of intrathecal ketamine.  相似文献

The Brief Piping Signal of the Honey Bee: Begging Call or Stop Signal?   总被引:2,自引:0,他引:2  
For over 40 yr, investigators have recognized that the brief piping signal plays a role in the foraging operation of a honey bee colony. The function of this signal, however, remains uncertain. The main objective of this study was to determine whether, under normal foraging conditions, bees following waggle dancers produce brief piping signals to beg nectar samples from the dancers. We made observations on waggle dancers and their followers in an undisturbed colony whose foragers gathered nectar and pollen from flowers. We found that waggle dancers do often receive brief piping signals, that the bees producing these signals are generally dance followers, and that these signals increase a waggle dancer's tendency to stop dancing. We also found, however, that the brief piping signal is clearly not a begging call; 0 of 41 waggle dancers that received a piping signal from a dance follower gave a nectar sample to the bee that produced the signal. Our results support the hypothesis that the brief piping signal is a stop signal; it serves to shut off waggle dancing. But why some dance followers pipe the dancer they are following, thereby inhibiting her dancing, remains unclear and warrants further investigation.  相似文献

Isolated heart preparations of frog and rat were used to test the validity of the adrenoceptor interconversion hypothesis. This hypothesis claims that low temperature converts the inotropic beta-adrenoceptors in isolated frog and rat heart to alpha-adrenoceptors. The present results do not support the adrenoceptor interconversion hypothesis. In the isolated frog ventricle, lowering the temperature from 24 C to 14 C did not significantly alter the inotropic potency of the sympathomimetic drugs isoprenaline, epinephrine, and phenylephrine and did not reduce the potency of the beta-adrenoceptor blocking drug propranolol as an epinephrine antagonist. In the isolated rat left atrium, lowering the temperature from 31 C to 17-19 C did not significantly change the inotropic potency of isoprenaline, norepinephrine and phenylephrine, did not diminish the potency of propranolol, and did not increase the potency of the alpha-adrenoceptor blocking drug phentolamine.--Benfey, B. Actin is an abundant protein in most nonmuscle cells. It has often been observed in isolated nuclei, yet cytoplasmic contamination was of course initially regarded as the most plausible origin. Numerous studies on nuclear actin appeared in the 1970s and 1980s, but the picture remained rather muddy. The viewpoint at that time was that actin-shown to move freely between cytoplasm and nucleus-was a mere "thermodynamic wanderer," transiently occupying the nucleus. More recently, evidence has been mounting that actin's presence in the nucleus is not simply governed by the laws of diffusion. The same holds true for the finding of various actin-related proteins in the nucleus, and the case for nuclear myosin, specifically myosin I, is now quite convincing. Moreover, the first intimations of functional roles of nuclear actin are now emerging. Here we examine the overall subject from cell biological and chemical perspectives. The major issue is no longer the presence of actin in the nucleus but rather its supramolecular organization, intranuclear locations, and, of course, functions. These issues interface with recent findings that reveal a surprisingly diverse repertoire of actin conformations and oligomer and polymer forms beyond monomeric G-actin and polymeric F-actin. We present ideas for advancing the nuclear actin field and call for a renewed attack on this major problem in cell biology.  相似文献

Spike length (SL), spikelet number (SPN) per spike, kernel number per spike (KNPS), and thousand-kernel weight (TKW) have strong genetic associations with kernel weight per spike (KWPS) in wheat. To investigate their genetic relationships at the individual quantitative trait locus (QTL) level, both unconditional and conditional QTL mapping for KWPS with respect to SL, SPN, KNPS, and TKW were conducted. Two related F8:9 recombinant inbred line populations, comprising 485 and 229 lines, respectively, were used. The trait phenotypic performances of each population were evaluated in four different environments. Unconditional QTL mapping analysis identified 22 putative additive QTL for KWPS, five of which were stable QTL, and only QKwps-WJ-1B.2 showed significant additive-by-environment interaction effects. In comparison with unconditional QTL mapping analysis, conditional QTL mapping analysis indicated that, at the QTL level, KNPS and TKW contributed more to KWPS than did SL and SPN. Any unconditional QTL for KWPS detected in this study was associated with at least one of its four related traits. The present study will provide assistance in the understanding of the genetic relationships between KWPS and its related traits.  相似文献

After the Second World War, in Italy Q Fever or Coxiellosis has been shown a significant relevance, a recrudescence with an epidemic state for over ten years. Later, the infectious disease occurred as endemic since the 80s, the outbreaks were just isolated. Workflows analysis of some authors has demonstrated the spread out of the infection throughout Italian herds with a prevalence ranging from 1.2 per cent to 10 per cent. Our survey carried out throughout Campania area in cattle has shown a real positivity over 14 per cent performing the IFAT for the detection of IgG antibodies for Coxiella burnetii. Therefore, it has been so important to stress the influence of cattle farming management in stables as a real risk of Coxiellosis. In countries where adulthood is considered to be attained at age eighteen, 2011 can be the point at which the diphosphoinositol polyphosphates might formally be described as "coming of age", since these molecules were first fully defined in 1993 (Menniti et al., 1993; Stephens et al., 1993b). But from a biological perspective, these polyphosphates cannot quite be considered to have matured into the status of being independently-acting intracellular signals. This review has discussed several of the published proposals for mechanisms by which the diphosphoinositol polyphosphates might act. We have argued that all of these hypotheses need further development.We also still do not know a single molecular mechanism by which a change in the levels of a particular diphosphoinositol polyphosphate can be controlled. Yet, despite all these gaps in our understanding, there is an enduring anticipation that these molecules have great potential in the signaling field. Reflecting our expectations of all teenagers, it should be our earnest hope that in the near future the diphosphoinositol polyphosphates will finally grow up.  相似文献
