This study presents a combined method to analyze extracellular fungal laccases using a new anti-laccase antibody together with the identification of tryptic laccase peptides by mass spectrometry (nanoLC–ESI–MS/MS). The polyclonal anti-laccase antibody LccCbr2 was raised against peptides designed from the copper binding region II of fungal laccases using in silico data obtained from GenBank database. As a consequence, detection requires denaturation of the enzymes due to the stable conformation of the copper binding region II. The specificity of the antibody was shown with denatured laccase Lcc1 of Coprinopsis cinerea and laccase of Hypholoma fasciculare. LccCbr2 detected amounts as low as 5 ng of highly purified laccase, indicating a possible use of the antibody for quantification of laccase proteins. Denatured extracellular laccases from culture supernatants of the basidiomycetes C. cinerea, H. fasciculare, Lentinula edodes, Mycena sp., Piriformospora indica, Pleurotus cornucopiae, Pleurotus ostreatus, Pycnoporus cinnabarinus, Trametes versicolor and furthermore the ascomycete Verpa conica were detected with apparent molecular masses between 60 and 70 kDa by LccCbr2. The identity of extracellular laccases from C. cinerea, H. fasciculare, P. ostreatus, P. cinnabarinus and T. versicolor were verified by tryptic peptides using nanoLC–ESI–MS/MS.  相似文献

一株新分离的云芝栓孔菌Z-1应用于木质素降解及染料脱色   总被引:1,自引:0,他引:1  
木质素是一种非结晶性的复杂三维网状酚类高分子聚合物,被认为是木质纤维素生物质抗降解的天然屏障。探索并开发高效降解木质素的微生物资源已成为近些年来的研究焦点。本研究对新分离的一株具有潜在木质素降解能力的菌株Z-1开展了系列研究。首先通过形态学和系统发育分析将菌株Z-1鉴定为云芝栓孔菌Trametes versicolor。平板定性检测初步表明云芝栓孔菌T. versicolor Z-1具有较强的产过氧化物酶和漆酶能力。以木质素为唯一碳源时,T. versicolor Z-1对木质素的降解率和脱色率分别可达13.38%和26.43%。酶活检测分析表明该菌主要是通过分泌漆酶和锰过氧化物酶(manganese peroxidase,MnP)降解木质素。利用傅里叶变换红外光谱(fourier transform-infrared spectroscopy,FT-IR)、扫描电镜(scanning electron microscope,SEM)及气相色谱-质谱联用(gas chromatography-mass spectroscopy,GC-MS)对云芝栓孔菌T. versicolor Z-1降解后木质素残渣结构以及代谢物的鉴定分析结果证实了该菌对木质素的强降解能力,并表明该菌对木质素的降解途径包括酚醚键的断裂、芳香环侧链氧化裂解以及芳香环开环反应等。此外,该菌还对多种芳香类染料展现出了强的脱色能力,其中对刚果红、孔雀石绿和考马斯亮蓝R-250 3种染料的脱色率达到100%。本研究表明云芝栓孔菌T. versicolor Z-1具有应用于工业化木质素降解与芳香化合物类染料脱色的开发前景。  相似文献

Extracellular laccases produced by three different wood-rotting fungi, Cerrena unicolor, Heterobasidion annosum and Trametes versicolor, were immobilized via covalent bonds formation on DEAE-Granocel 500, CM-Granocel 500, and acrylic carriers. Out of the tested carriers, only the DEAE-Granocel 500, which was activated by divinyl sulphone appeared to be a suitable matrix for the expression of enzymic activity. Only one laccase of all the tested enzymes produced by C. unicolor showed the best binding to the carrier and a satisfactory enzymic activity. The immobilized laccase exhibited the highest enzymic activity at pH 5.2 and it was more resistant to thermal denaturation than the native enzyme. At 90 °C, it retained 75% activity compared to the free enzyme. It was also more stable during storage at 4 °C: after 4 months the immobilized laccase retained 98% of initial activity. Immobilized C. unicolor laccase was active in 10–60% concentration of methanol, acetone, isopropanol or acetonitrile. The best enzymic activity was observed in 20% solution of acetonitrile in buffer.  相似文献

The influence of alcohol, 4-acetylamino,2,2,6,6'-tetramethylpiperidinyloxy (4-acetylamino-TEMPO) and laccase (from Trametes versicolor, TvL) concentration in the aerobic oxidation of furfuryl alcohol was investigated. Studies show that the K m for 4-acetylamino-TEMPO is around 6.3 mM (V max=0.18 mM min-1) using 6.6 U mL-1 of laccase and a furfuryl alcohol concentration of 140 mM. Under these optimized conditions, the reaction rate is still dependent on the concentration of enzyme in solution. Laccase can be reused, with a residual activity of around 25%. An important conclusion is that laccase is not stable in the presence of oxoammonium salts, presumably due to degradation via oxidation of essential amino acid residues or the glycosyl moieties on the periphery of the enzyme.  相似文献

为了探明漆酶在斑玉蕈生长发育过程中的功能,对斑玉蕈转录测序预测的13个漆酶基因序列进行分析、鉴定和构建分子系统发育树;检测了不同生长发育时期漆酶的活性和漆酶基因表达水平。研究结果显示:13个基因片段中有10个是漆酶基因。不同的漆酶同工酶之间进化关系存在明显差异,大多数漆酶与木腐菌(金针菇Flammulina filiformis和侧耳属Pleurotus)进化关系较近。对斑玉蕈不同生长发育时期的酶活检测结果显示,从斑玉蕈的菌丝恢复期到钉头期,漆酶活性逐渐升高,而在子实体形成后期酶活逐渐降低。对培养40d、60d和80d的菌丝样品以及不同生长发育时期的样品进RT-qPCR检测,结果显示在菌丝营养生长时期,大多数漆酶基因在第40-60天表达量持续增加1-3倍,而在第60-80天时表达量出现降低的情况。而在生殖生长时期,大多数漆酶基因在转色期或者原基期相对表达量达到最大值,并在子实体期出现降低,这与漆酶活性的检测具有一致性。lcc3lcc7lcc8lcc9在斑玉蕈生殖生长过程中相对表达量出现了10-100倍的上调。这说明从菌丝培养到菌丝扭结形成子实体和子实体发育的过程中,不同的漆酶可能发挥着不同的作用,表达量较高的漆酶基因可能对基质降解和子实体形成起主要作用。  相似文献

Phenols removal in musts: Strategy for wine stabilization by laccase   总被引:1,自引:0,他引:1  
The potential of laccase from Trametes versicolor for phenolic removal in must for wine stabilization was evaluated through a combination of an analytical methodology (capillary zone electrophoresis) and kinetics of phenols removal as the total antioxidant potential variation. Total phenolic content, total antioxidant potential and polyphenols were monitored from 0 to 3 h of must treatment. The results indicated that the treatment of a red must with laccase affect mainly the phenolic compounds responsible for the must antioxidant properties. The treatment of white musts with laccase showed higher reduction in total phenol than in the total antioxidant potential. Phenol degradation by laccase was very fast for catechins, and slowly for stilbenes (cis- and trans-resveratrol) and derivatives of cinnamic (ferulic and caffeic) and benzoic (syringic, vanillic, and gallic) acids. It is possible to conclude in this case that the use of laccase in white wines is perfectly feasible. This would allow softer and ecologically correct treatments, which would diminish the cost of processing and avoid deterioration of wines for long storage times.  相似文献

The explosive 2,4,6-trinitrotoluene (TNT) is considered a toxic environmental pollutant that contaminates the soil and ground water. The white rot fungus Phanerochaete chrysosporium is well known for the degradation of TNT under ligninolytic condition. Very few, if any, studies have been done using other white rot fungi. In this study four fungal species, namely, P. chrysosporium, Kuehneromyces mutabilis, Hypholoma fasciculare, and Phlebia radiata, were used to investigate TNT degradation. All fungi were grown under ligninolytic (low-nitrogen) and nonligninolytic (high-nitrogen) conditions containing 25 parts per million (ppm) (0.11 mM) of TNT. Analysis by high-performance liquid chromatography (HPLC) showed biotransformation of TNT under both conditions. Complete degradation occurred under ligninolytic conditions (peroxidase enzymes were present) by P. chrysosporium and P. radiata. A nitrite release assay at 6 days indicated the denitrifying abilities of all the tested varieties of white rot fungi. For both ligninolytic and non-ligninolytic conditions, mass-balance studies showed biotransformation of 0.5 μ Ci 14C-labeled TNT with pregrown mycelial pellets of all fungal species, in which 5% to 15% of the TNT was converted to CO2. These studies show that TNT may be degraded by several other species of white rot fungi and provided additional information on the biodegradation of nitroaromatic compounds in the environment.  相似文献

The ability of several white-rot fungal strains to remove and detoxify acetone extractives (pitch or resin) in Scots pine sapwood was investigated in stationary laboratory batch assays. Fungal pretreatment provided up to 62% total pitch reduction and significant decreases in pitch toxicity. The best strains were Bjerkandera sp. strain BOS55, Stereum hirsutum and Trametes versicolor that eliminated over 93% of the problematic triglyceride fraction and 58–87% of other lipophilic extractive classes in only 2 weeks. Fungal removal of the wood extractives was accompanied by a 7.4–16.9-fold decrease in their inhibitory effect, as determined in the Microtox bioassay. Wood pretreatment by Bjerkandera sp. and T. versicolor caused limited losses of woody mass (less than 4% in 4 weeks); whereas S. hirsutum led to somewhat higher mass losses (7% in 4 weeks). These results indicate the potential of white rot fungi to control pitch deposition problems in pulping and to reduce the aquatic toxicity caused by naturally-occurring lipophilic extractives in forest industry effluents.  相似文献

Coprinellus congregatus secreted a laccase isozyme when the culture was transferred to an acidic liquid medium (pH 4.1). The laccase cDNA gene (clac2) was used as a probe for cloning of the genomic laccase gene (lac2) including the promoter (Plac2). The open reading frame (ORF) of lac2 had 526 deduced amino acids and four conserved copper binding domains as other fungal laccases. Recombinant plasmid (pRSlac2p-cDNA) of lac2 cDNA with its own promoter was transformed in Saccharomyces cerevisiae. Expression of the transformed lac2 gene was induced by oxidative stress (H2O2) in yeast and the survival rate of the transformed yeast strain was greatly increased when compared with that of the control strain transformed with pRS316 yeast vector.  相似文献

生存在不同基底颜色环境下的爬行动物种群通常表现出丰富的体色地理变异, 其体色变化的潜在机制具有多样性。变色沙蜥(Phrynocephalus versicolor)和草原沙蜥(P. frontalis)具有较近的遗传关系, 曾被认为与荒漠沙蜥(P. przewalskii)组成同一系统发育种组。本文应用光纤光谱仪(AvaSpec-2048), 通过记录沙蜥背部体表12个部位的皮肤光反射率, 定量比较在黑化环境下的深色变色沙蜥与非黑化环境下的浅色草原沙蜥自然体色变异, 研究其种群体色变异是否具有时间可逆性, 并探讨基底颜色对沙蜥体色的影响机制。研究结果表明, 黑化生境下的变色沙蜥体色显著深于非黑化枯黄色生境下的草原沙蜥。此外, 对黑化与非黑化样本开展的生境互换移植围栏实验, 即把枯黄色生境中非黑化的草原沙蜥移植于黑色的基底环境中饲养, 把黑化生境中黑化的变色沙蜥移植于枯黄色生境中饲养。结果表明, 饲养1周后黑化群体背部6个检测部位的光反射率显著变大, 其他部位均无显著变化; 而非黑化群体只有左后肢和背部右上方2个部位的皮肤光反射率发生显著变化, 其他部位反射率无显著变化。结果表明, 变色沙蜥体色变异能力比草原沙蜥强, 体色表型可能已经在两个近缘沙蜥物种中稳定遗传, 基底生境颜色的短期变化在统计学上能引起肉眼难以识别的轻微的体色变异, 个体发育相关的一些遗传因素可能对体色变异起控制 作用。  相似文献

丰林国家级自然保护区木腐真菌多样性与寄主倒木的关系   总被引:1,自引:0,他引:1  
木腐真菌是一类以木材为生长基质的大型真菌, 通过分泌各种水解酶全部或部分降解木材中的木质素、纤维素和半纤维素, 促进森林生态系统的物质循环, 具有重要的生态功能。本研究调查了丰林国家级自然保护区固定样地中木腐真菌的多样性和倒木特征, 并进行了木腐真菌的物种多样性和数量与倒木的种类、数量、腐朽程度、直径大小等的相关性分析。结果显示: 在样地内共采集木腐真菌标本295份, 经鉴定为93种, Shannon多样性指数为3.86, Simpson指数为0.96。相关性分析发现木腐真菌的数量和种类与直径为2-5 cm和5-10 cm的倒木、2级腐烂的倒木和红松倒木均显著相关。样地中优势倒木寄主分别为槭属(Acer)、榛属(Corylus)、云杉属(Picea)和松属(Pinus), 这4类倒木上生长的木腐真菌种类组成具有明显的差异, 槭属和榛属倒木上的共有优势种主要是三色拟迷孔菌(Daedaleopsis tricolor)、云芝(Trametes versicolor)和桦附毛孔菌(Trichaptum pargamenum), 而松属和云杉属的共有优势种主要有白囊耙齿菌(Irpex lacteus)、云芝、冷杉附毛孔菌(Trichaptum abietinum)和褐紫附毛孔菌(T. fuscoviolaceum)。倒木产生真菌子实体的概率研究表明, 同一类寄主倒木上发生木腐真菌子实体的概率在调查面积增加到0.36 ha后趋于一个定值, 松属倒木中仅有10.2%产生真菌子实体, 槭属和云杉属分别是12.9%和13.4%, 榛属最高, 达到53.7%。本研究结果对于预测森林生态系统中木腐真菌的发生具有重要理论意义。  相似文献

Bioremediation of munitions-contaminated soil requires effective transformation and detoxification of high concentrations of 2,4,6-trinitrotoluene (TNT). Pseudomonas aeruginosa strain MX, isolated from munitions-contaminated soil, aerobically transformed TNT (100 mg/L) in culture medium within 15 h, causing transient accumulation of hydroxylaminodinitrotoluenes (HADNTs). The predominance of 2-hydroxylamino-4,6-dinitrotoluene (2HADNT), as well as 2-amino-4,6-dinitrotoluene (2ADNT) and 4,4' ,6,6' -tetranitro-2,2' -azoxytoluene (2,2'AZT), indicated preferential reduction of the TNT ortho nitro group. While only 12% of the TNT was transformed to 2ADNT, up to 65% was transformed to tetranitroazoxytoluenes (AZTs), which accumulated as a precipitate. The precipitate was formed by microscopic particles adhering to bacterial cells, which subsequently formed clusters containing lysed cells. Toxicity toward bacteria was primarily attributed to 2ADNT, because pure AZTs preincubated with sterile medium had little effect on the strain. While the culture medium containing TNT exhibited toxicity toward corn (Zea mays L.) and witchgrass (Panicum capillare L.), little phytotoxicity was observed after incubating with P. aeruginosa strain MX for 4 d. Strong binding of HADNTs to soil and low AZT bioavailability may further promote the detoxification of TNT in soil.  相似文献

本研究利用愈创木酚和苯胺蓝固体培养基对菌株进行初筛,利用形态学和分子生物学对筛选出的菌株进行鉴定,以黄孢原毛平革菌Phanerochaete chrysosporium CGMCC 5.0776为对照,利用其对玉米秸秆进行预处理并测定木质素和纤维素的降解率,测定筛选菌株在预处理玉米秸秆过程中漆酶、锰过氧化物酶(manganese peroxidase,MnP)和木质素过氧化物酶(lignin peroxidase,LiP)活性。结果表明:利用愈创木酚和苯胺蓝固体培养基,从16株白腐真菌菌株中筛选出2株具有较高漆酶或MnP活性的菌株,鉴定其为桦栓孔菌Trametes betulina (L.) Pilát(ZT-153)和亚黑管孔菌Bjerkandera fumosa (Pers.) P. Karst.(ZT-307),测定T. betulina ZT-153和B. fumosa ZT-307对玉米秸秆酸不溶木质素降解效率分别为13.60%和21.87%,较对照P. chrysosporium CGMCC 5.0776高1.58%和9.85%,对纤维素的降解率较低,分别为4.10%和4.50%。2株菌株在预处理玉米秸秆过程中,T. betulina ZT-153表现出漆酶和MnP活性,B. fumosa ZT-307只表现出LiP活性。其中B. fumosa ZT-307对玉米秸秆酸不溶木质素的降解效率最高,在秸秆资源的综合利用方面具有较好的潜力和应用前景。  相似文献

真菌漆酶(laccase)是一种多酚氧化酶,在真菌生长发育中具有重要作用。本研究采用根癌农杆菌介导转化的方法,以香菇Lentinula edodes菌株W1为受体菌株,在Leactin基因启动子调控下过表达Lelcc1基因;对其中7个单拷贝插入的转化子进行qRT-PCR分析,7个Lelcc1基因表达量较出发菌株W1均上调了1.5-8倍。进一步分析了这7个转化子的遗传稳定性;挑取了3个稳定的转化子进行表型分析,主要包括不同培养基中的生长速度、代料栽培过程中菌棒转色程度、以及透射电镜观察菌丝细胞的超微结构。发现转化子和受体菌株W1的生长速度无显著差异,但代料栽培过程中转化子较W1转色更快,菌棒表面颜色更深;透射电镜观察菌丝细胞的超微结构发现在细胞壁厚度、细胞膜形态等方面,其中两个超量表达转化子与W1间存在显著差异。结果表明,香菇Lelcc1基因可能参与了转色过程中色素合成或积累,也可能与细胞壁形成有关。  相似文献

A thermostable laccase was isolated from an unidentified fungal isolate [Enz. Microb. Technol. 33 (2003) 212], and tentatively named UD4. This work indicates that the enzyme has unique properties other than its thermostability. Investigation into the kinetic parameters of the thermostable laccase yielded an unusually high affinity for ABTS as a substrate (low Km) when compared with available published data for other laccase isozymes. The specificity constant (kcat/Km) was found to be considerably higher than laccase from other sources and is comparable to “white” laccase from Pleurotus ostreatus (POXA1). However, POXA1 isozyme exhibits a large turnover number (kcat) that contributes to its high specificity constant whereas the high specificity constant for UD4 laccase is achieved by having a high substrate affinity. The UD4 thermostable laccase, like most other laccases, is able to utilize guaiacol as a substrate, whereas POXA1 is unable to oxidize guaiacol, indicating a broader substrate range for the thermostable laccase from UD4. The thermostable laccase is inhibited by sodium azide through non-competitive inhibition, and by thioglycolic acid and hydroxylamine through competitive inhibition. The high specificity constant, substrate affinity and broader substrate range of the thermostable laccase from UD4 indicates that it is a highly favourable candidate enzyme for industrial application.  相似文献

The degradation of 2,4,6-trinitrotoluene (TNT) by seven strains of white rot fungi was examined in two different media containing 50 mg L−1 of TNT. When TNT was added into a nutrient-rich YMG medium at the beginning of the incubation, four of the fungal strains completely removed TNT during several days of incubation and showed higher removal rates than those of Phanerochaete chrysosporium. TNT added into YMG medium after a 5-day preincubation period completely disappeared within 12 hours, and the removal rates were higher than those in N-limited minimal medium. Isomers of hydroxylamino-dinitrotoluene were identified as the first detectable metabolites of TNT. These were transformed to amino-dinitrotoluenes, which also disappeared during further incubation from cultures of Irpex lacteus. During the initial phase of TNT degradation by I. lacteus, dinitrotoluenes were also detected after the transient formation of a hydride-Meisenheimer complex, indicating that I. lacteus used two different pathways of TNT degradation simultaneously. We investigated the influence of potential laccase inducers with environmental relevance on extracellular laccase activity and removal of the xenoestrogen technical nonylphenol (tNP) by the aquatic hyphomycete Clavariopsis aquatica. Concomitantly, we identified two putative laccase gene fragments (Icc1 and Icc2) and have followed their expression during removal of tNP under different conditions. Our results indicate a significant effect of copper on extracellular laccase activity in supernatants of fungal cultures. Laccase activity was highest in the presence of copper when added together with vanillic acid, followed by copper when used alone. Only slight laccase activities were recorded in the presence of only vanillic acid, whereas in the absence of either compound laccase activities were negligible. Laccase activity was well correlated with the removal efficiency of tNP, indicating the involvement of laccase in tNP bioconversion. Overall, Icc2 was less expressed than Icc1. The expression of Icc1 and Icc2 correlated only partially with the measured laccase activity, suggesting the existence of cell-associated laccase fractions not detectable in fungal culture supernatants and/or the existence of additional laccase genes.  相似文献

The behaviour of three different laccase-based graphite biosensors was studied in view of their use in agricultural or industrial waters polluted by phenolic compounds. Catechol was used as a substrate model. Laccase from Trametes versicolor was immobilized on one biosensor (type A electrode) by adsorption while, on the other two biosensor types, laccase was covalently bound through the carboxylic groups created on the graphite by means of treatment with an electric potential difference (type B electrode) or with nitric acid (type C electrode). In the latter two cases, hexamethylenediamine and glutaraldehyde were used as the spacer and the coupling agent, respectively. The extension of linear response range and the sensitivity and time stability of each biosensor type were investigated. The type C biosensor gave the best results and its electrochemical properties proved comparable to those reported by other authors.  相似文献
