We have cloned and sequenced the genes atpB and atpE, coding for CF1 subunits and , respectively, of the chloroplast genome of the brown alga Dictyota dichotoma. Although the coding site of atpE cannot be demonstrated by heterologous Southern hybridizations, a 417 bp reading frame 3 to atpB was identified as the gene atpE by sequence similarities with atpE genes from other sources. A maximum sequence identity of 30% is found between the predicted amino acid sequence of the Dictyota subunit and the corresponding cyanobacterial subunits. Including conserved amino acid replacements, the Dictyota subunit exhibits about 70% sequence similarity with the cyanobacterial and land plant subunits. As in cyanobacteria, the atpE gene does not overlap the preceding gene atpB. The deduced amino acid sequence of atpB is 74–79% identical to the corresponding cyanobacterial and chloroplast subunits. Entirely conserved are regions referred to as the catalytic and/or regulatory sites of ATP formation, including interacting regions between subunits and . A phylogram predicted from F1/CF1- subunits of eleven different organisms suggests a common evolutionary origin of plastids from chlorophytes and brown algae.  相似文献

Temperature dependencies were established for the egg-to-mummy and mummy-to-adult phases, for mummy mortality, and for parasitism of Aphidius ervi Haliday, Aphidius rhopalosiphi De Stefani-Perez, and Praon volucre (Haliday) (Hymenoptera, Aphidiidae), three parasitoids of Sitobion avenae (Fabricius) (Homoptera, Aphididae), at 8°C, 12°C, 16°C, 20°C, and 25°C on winter wheat (cv. Haven). A physiological model described temperature-dependent development over the full temperature range, whereas a linear model was fitted for data above 8°C and used to estimate the lower temperature thresholds and day-degrees (° D) required for development. The thresholds for A. ervi were 2.2°C for egg-mummy development and 6.6°C for mummy-adult development, those for A. rhopalosiphi were 4.5°C and 7.2°C, and those for P. volucre were 3.8°C and 5.5°C. The time to develop into mummies and adults differed significantly between the three species: A. ervi development into mummies required an average of 159 ° D, while development into adults took an average of 73 ° D. The corresponding average times required for A. rhopalosiphi and P. volucre to develop mummies were 124° D and 126° D, while their development into adults required an average of 70° D and 150° D, respectively. Mummy mortality was 25–35% at 8°C and less at the higher temperatures tested, but began to increase again at 25°C, showing a quadratic relationship between mortality and temperature. Parasitization was very low or, in the case of P. volucre, absent up to 12°C and thereafter increased with increasing temperature. The relationship between parasitization, recorded as percent aphids mummified, and temperature was linear at the temperatures tested and depended on species. A. ervisuperparasitized 11.1% aphids at 20°C and 16.6% aphids at 25°C, whereas superparasitism was low in A. rhopalosiphi and absent in P. volucre. From 16°C to 25°C the P. volucre sex ratio increased. For A. ervi and A. rhopalosiphi there was no trend with temperature, but at 20°C and 25°C it was close to even. Field data for 1996 and 1997 allowed for a comparison of actual and expected emergence of overwintering mummies. In both years, parasitoids were predicted to have emerged from overwintering mummies well in advance of the onset of aphid infestation, and more than a month earlier than the first parasitized aphids were found in winter wheat. Observations from trap plants in other crops supported the predictions of the models. Other factors that can affect biological control by cereal aphid parasitoids are discussed.  相似文献

Summary The dorsal tegument of the mature cercaria of Notocotylus attenuatus is a syncytial, cytoplasmic layer, containing two types of secretory granule which are identifiable ultrastructurally. The type 1 secretory bodies are electron lucid, whereas most type 2 granules have a banded appearance. The ventral tegument contains granules which are secreted from the type 3 cells; the type 3 granules are membrane bound, electron dense, and consist of both an amorphous and a finely striated zone. The type 4 cells mainly contain cigar-shaped granules consisting of an amorphous core surrounded by concentric striations. The granules exhibit structural variability in shape and content. The type 4 cells undergo a cellular migration to the tegument during encystment. The structure of the posterior-lateral glands and mode of secretion of the granules are described. Possible functions of microtubules are discussed for each cell type. Details of some secretory processes involved in the formation of the hemispherical cyst wall are described. The layers of the cyst wall may be related to the granular contents of the various parenchymal cells of the cercaria. The tegument of the metacercaria originates primarily from the cytoplasm of the type 1, type 2, type 3 and type 4 cells.  相似文献

Complete nuclear-encoded small-subunit 18S rRNA (=SSU rRNA) gene sequences were determined for the prasinophyte green alga Mantoniella squamata; the charophycean green algae Chara foetida, Coleochaete scutata, Klebsormidium flaccidum, and Mougeotia scalaris; the bryophytes Marchantia polymorpha, Fossombronia pusilla, and Funaria hygrometrica; and the lycopod Selaginella galleottii to get a better insight into the sequential evolution from green algae to land plants. The sequences were aligned with several previously published SSU rRNA sequences from chlorophytic and charophytic algae as well as from land plants to infer the evolutionary relationships for major evolutionary lineages within the Chlorobionta by distance matrix, maximum parsimony, and maximum likelihood analyses. Phylogenetic trees created by the different methods consistently placed the Charophyceae on the branch leading to the land plants. The Charophyceae were shown to be polyphyletic with the Charales (charalean algae) diverging earlier than the Coleochaetales, Klebsormidiales, Chlorokybales, and Zygnematales (charophycean algae) which branch from a point closer to the land plants in most analyses. Maximum parsimony and maximum likelihood analyses imply a successive evolution from charophycean algae, particularly Coleochaetales, to bryophytes, lycopods, and seed plants. In contrast, distance matrix methods group the bryophytes together with the charophycean algae, suggesting a separate evolution of these organisms compared with the club moss and the seed plants. Summary Brassica napus cybrid plants which contain novel nucleus-mitochondria-chloroplast combinations have been constructed, via protoplast fusion. Such fusions resulted in mitochondrial DNA plasmids being lost (at a frequency of 12.5%) or, more surprisingly, being transferred from mitochondria of one protoplast population to mitochondria of the other population (at a frequency of 6.1%). Mitochondria containing their new DNA complement became the dominant organelle population in regenerated plants and were faithfully maternally inherited through successive sexnal generations. No concomitant alterations in mitochondrial chromosome organization or nuclear chromosome number occurred. Protoplast fusion can, therefore, cure plant mitochondria of extrachromosomal DNA and, more importantly, be used to transform plant mitochondria with naturally occurring mitochondrial plasmids. The potential for mitochondrial transformation with recombinant vectors is discussed.  相似文献

The ultrastructure of dikaryotic haustoria of sevenPucciniastrum species,Thekopsora galii, Naohidemyces vaccinii, andCalyptospora goeppertiana was investigated.Pucciniastrum actinidiae, P. agrimoniae, P. pyrolae, andCalyptospora goeppertiana revealed haustoria whose necks were wrapped by a fold of the extrahaustorial matrix. The matrix-fold ofCalyptospora goeppertiana was characteristically shaped.Pucciniastrum circaeae, P. epilobii, P. hikosanense, P. styracinum, Thekopsora galii, andNaohidemyces vaccinii showed typical haustorial necks which were not sheathed by a matrix-fold. Haustorial necks which were wrapped by a fold of the extrahaustorial matrix were designated velopedunculate, and those which were naked gymnopedunculate. The genes were cloned for the two apoprotein subunits, and ,of phycocyanin from the cyanobacterium Spirulina maxima = Arthrospiramaxima) strain F3. The - and -subunit gene-coding regionscontain 489 bp and 519 bp, respectively. The -subunit gene is upstreamfrom the -subunit gene, with a 111-bp segment separating them.Similarities between the -subunits of S. maxima and nine othercyanobacteria were between 58% and 99%, as were those between the -subunits. Atriplex gmelini plants were regenerated via organogensis from hypocotyl explants. Callus lines were induced from the hypocotyl explants on Linsmaier and Skoog (LS) medium supplemented with 1 M benzyladenine and 5 M -naphthaleneacetic acid in the dark. Shoots were regenerated from the callus lines on LS medium supplemented with 20 M thidiazuron and 0.1 M -naphthaleneacetic acid under a high-intensity light condition (450 mol m–2 s–1). The regenerated shoots were rooted on LS medium without growth regulators to obtain fully developed plants. We succeeded in transforming Atriplex gmelini from callus lines using Agrobacterium tumefaciens.  相似文献

-Oxidation enzymes were detected both in the mitochondria and microbodies of pea cotyledons. Intact mitochondria did not show -oxidation enzyme activity but in ruptured mitochondria this activity was high. It is apparent that the mitochondrial membrane barrier prevents rapid access of acyl-CoA substrates to matrix -oxidation sites. Removal of the membrane barrier permits rapid access of acyl-CoAs and these enzyme activities may then be measured.  相似文献

Summary The genetical control of basidiospore production by sporophores of the fungus Coprinus congregatus was studied. This species is characterized by a bipolar compatibility control, and homokaryons with complementary alleles A1 and A2 can be distinguished apart. We confirmed that the pale mushroom phenotype of the fungus is determined by a nuclear gene symbolized pal. This gene also controls a sporeless character and segregates independently of the mating-type locus. Dikaryons homoallelic for the pal allele produce typical pale and sporeless sporophores, while heteroallelic (pal +, pal ) and homoallelic (pal +, pal +) dikaryons produce normal or almost normal sporulating sporophores. In order to segregate homokaryons homoallelic for the pal gene (A1, pal ; A1, pal +, A2, pal ; A2, pal +), the following protocols were used: (a) the dikaryotization of stock homokaryons containing the pal + allele and of each mating type, A1 or A2, by dikaryotic mycelia homoallelic for the pal allele; (b) the culturing of homokaryotic mycelia issuing from the germination of basidiospores from sporophores produced by dikaryotic mycelia heterokaryotic for the pal gene; (c) the culturing of mycelia grown from protoplasts obtained from dikaryons homoallelic for the pal allele (D6 strain), and from homokaryons heteroallelic for the pal gene (H8), or homoallelic for pal #x002B;+ allele (H7). These techniques enabled us to segregate homokaryons of the four types defined above and were indispensable in the segregation of the pal homoallelic homokaryons as no basidiospores were produced by typical pale mushrooms.  相似文献

Fowl plague virus, strain Dutch, was metabolically labeled withd-[2-3H]mannose, or withd-[6-3H]glucosamine, and the small subunit (HA2; 0.8 mg in total) of the viral hemagglutinin was isolated by preparative sodium dodecylsulfate-polyacrylamide gel electrophoresis. After proteolytic digestion, the radioactive oligosaccharides were sequentially liberated from the glycopeptides by treatment with different endo--N-acetylglucosaminidases and with peptide:N-glycosidase or, finally, by hydrazinolysis. In this manner, four groups of glycans could be obtained by consecutive gel filtrations and were subfractionated by HPLC. The structures of the individual oligosaccharides were analyzed by micromethylation, by acetolysis or by digestion with exoglycosidases. Interspecific hybridizations were carried out between the two tetraploidsElymus caucasicus andE. longearistatus, and 23 tetraploids and hexaploids ofElymus containing SH, SY, SYH, and SYW genomes and representing various geographical regions. Meiotic pairing was studied in the two target species and their hybrids. It is concluded from this study that (i) interspecific hybridization is fairly easy to perform although strong reproductive barriers exist between the species; (ii)Elymus caucasicus andE. longearistatus are allotetraploids, and share the diverged SY genomes; (iii) the divergence of SY genomes is correlated with the geographic distance between theElymus spp. studied.  相似文献

Elicitins are 10 kDa holoproteins secreted by Phytophthora fungi, that elicit an incompatible hypersensitive reaction, leading to resistance against fungal and bacterial plant pathogens. Comparison of primary sequences of -elicitins and -elicitins indicated several potential necrotic activity-determining residues. All of the highly necrotic -elicitins have a hydrophilic residue (usually lysine) at position 13, whereas in the less necrotic -elicitins this residue is replaced by a valine. Here, we report the synthesis and expression of a gene encoding a highly necrotic elicitin, -cryptogein, and we show that the substitution of Lys-13 of this recombinant protein by a valine leads to a drastic alteration to the necrotic activity of the recombinant protein.  相似文献

At 5 and 10 g ml-1 concentration, zearalenone (F-2), a mycotoxin produced by a number of species of the genus Fusarium, causes an inhibition of the oxidative phosphorylation of isolated plant mitochondria, while at 20 and 40 g ml-1 it causes uncoupling. However, when the mitochondria are pre-incubated for 20 min with F-2, the uncoupling appears to be the prevailing effect. F-2 is also able to inhibit the mitochondrial ATPase activity (Mg2+-dependent). Conversely, F-2 (40 g ml-1) does not alter the ATP level of maize roots and only slightly affects the ATPase activity of pea stem and maize root microsomal fractions. In addition, F-2 (10–40 g ml-1) inhibits ATP synthesis catalyzed by rat liver mitochondria. InEchinops the flowers are surrounded by several scales and initiated in an acropetal and spiral succession on a cone-like inflorescence axis (Figs. 1–6). The floral organs originate in the following sequence: petals—stamens—carpels—pappus. The petals arise from a meristematic rim and therefore are already interconnected when they arise as primordia. This sympetalous zone remains rather inconspicuous for a long period, but eventually, the elongated corolla tube is formed through intercalary growth in a ring zone. Thereby, the stamens are moved upwards and form ledges on the corolla tube (Fig. 34). In the inferior ovary the usual zones of the typical angiospermous gynoecium can be distinguished, namely a synascidiate, symplicate and hemisymplicate zone. The ovule is borne on carpellary tissue.

The control regions of mitochondrial DNA of two insects, Schistocerca gregaria and Chorthippus parallelus, have been isolated and sequenced. Their sizes are 752 by and 1,512 bp, respectively, with the presence of a tandem repeat in C. parallelus. (The sequences of the two repeats are highly conserved, having a homology of 97.5%.) Comparison of their nucleotide sequences revealed the presence of several conserved sequence blocks dispersed through the whole control region, showing a different evolutionary pattern of this region in these insects as compared to that in Drosophila. A highly conserved secondary structure, located in the 3 region near the small rRNA gene, has been identified. Sequences immediately flanking this hairpin structure rather than the sequences of this structure themselves are conserved between S. gregaria/C. parallelus and Drosophila, having a sequence consensus of TATA at 5 and GAA(A)T at 3. The motif G(A)nT is also present in the 3 flanking sequences of mammalian, amphibian, and fish mitochondrial L-strand replication origins and a potential plant mitochondrial second-strand-replication origin, indicating its universal conservation and functional importance related to replication origins. The stem-and-loop structure in S. gregaria/C. parallelus appears to be closely related to that found in Drosophila despite occupying a different position, and may be potentially associated with a second-strand-replication origin. This in turn suggests that such a secondary structure might be widely conserved across invertebrates while their location in the control region may be variable. We have looked for such a conserved structure in the control regions of two other insects, G. firmus and A. mellifera, whose DNA sequences have been published, and their possible presence is discussed.Mitochondrial control regions characterized to date in five different insect taxa (Drosophila, G. firmus, A. mellifera, S. gregaria, and C. parallelus) may be classed into two distinct groups having different evolutionary patterns. It is observed that tandem repetition of regions containing a probable replication origin occurred in some species from disjunct lineages in both groups, which would be the result of convergent evolution. We also discuss the possibility of a mechanism of parahomologous recombination by unequal crossing-over in mitochondria, which can explain the generation of such tandemly repeated sequences (especially the first critical repetition) in the control region of mtDNA, and also their convergent evolution in disjunct biological lineages during evolution. The structure of a new nonasaccharide isolated from human milk has been investigated. Zusammenfassung Im Ovar von Chironomus sind in Phase 1 des 4. Larvenstadiums polygonal abgeflachte Innenzellen von kleineren Außenzellen umgeben, die Bakteroide und Phagosomen enthalten; zwischen den Innenzellen liegen unregelmäßige Zelltrümmer (keimbahnbegleitende Substanzen). Zu Beginn der Ovariolenbildung werden in Phase 3 durch Spalträume zwei Schichten der Außenzellen voneinander getrennt, von denen die innere (Follikel- und Eikanalepithel) regelmäßige Buchten bildet. In diese Buchten wandern von innen Zellpaare ein, die an synaptischen Komplexen bzw. multiplen Chromatinstrukturen als Ei- und Nährzellen kenntlich sind. Zwischen beiden Zellen sind Fusome häufig, die später in eigentümlicher Weise geschlossen werden. Zwischen den Eikanalzellen entsteht in Phase 5 durch Spaltbildung der Eikanal; in Phase 7 sind die Eikanalzellen auffallend glykogenreich. Kurz vor der Vitellogenese treten im Bereich der Oocyte Membransysteme und annulated lamellae auf; akzessorische Kerne werden als Ausstülpungen des Oocytenkernes gebildet und später abgeschnürt. In Phase 9 sind an der Peripherie der Eizelle Mikrovillisäume und Pinocytosebläschen sichtbar. Die distalen Zellen der Ovariole haben Eioder Nährzellcharakter, sind aber bei Ch. melanotus nicht von Follikelzellen umgeben und werden beim weiteren Ovariolenwachstum reduziert. Trotz extrem geringer Nährzellzahl der Follikel scheint das Chironomus-Ovar funktionell nicht von anderen polytroph meroistischen Insektenovarien unterschieden.

Summary In the ovary of Chironomus during phase 1 of the fourth larval instar, polygonally flattened inner cells are surrounded by smaller outer cells which contain bacteroids and phagosomes. Irregular cell remnants (germ line accompanying substances) lie among the inner cells. At the beginning of ovariole formation in phase 3, two layers of outer cells are separated by the formation of fissures. The inner layer of these cells (follicle- and egg-passage epithelium) forms regular invaginations. Cell pairs, identified as oocytes and nurse cells by synaptic complexes or multiple chromatin structures, wander from inside into the invaginations. Frequently between the two cells are fusomes, which later close in a characteristic manner. During phase 5, an egg passage is formed as a fissure among the egg-passage cells. During phase 7, the egg passage cells are conspicuously full of glycogen. Shortly before vitellogenesis membrane systems and annulated lamellae appear in the region of the oocyte. Accessory nuclei are formed by a tieing-off of projections of the the oocyte nucleus. During phase 9, microvilli and pinocytotic vesicles can be seen at the periphery of the oocyte. The distal cells of the ovariole are of oocyte or nurse cell nature, but in Ch. melanotus they are not surrounded by follicle cells and are reduced during further ovariole growth. In spite of the extremely small number of nurse cells in the follicle, the Chironomus ovary apparently does not differ functionally from other polytrophic meroistic insect ovaries.
