The present study was conducted to investigate the effects of chromium histidinate (CrHis) against experimentally induced type II diabetes and on chromium (Cr), zinc (Zn), selenium (Se), manganese (Mn), iron (Fe), and copper (Cu) in serum, liver, and kidney of diabetic rats. The male Wistar rats (n = 60, 8 weeks old) were divided into four groups. Group I received a standard diet (12% of calories as fat); group II were fed standard diet and received CrHis (110 mcg CrHis/kg body weight per day); group III received a high-fat diet (HFD; 40% of calories as fat) for 2 weeks and then were injected with streptozotocin (STZ) on day 14 (STZ, 40 mg/kg i.p.; HFD/STZ); group IV were treated as group III (HFD/STZ) but supplemented with 110 mcg CrHis/kg body weight per day. The mineral concentrations in the serum and tissue were determined by atomic absorption spectrometry. Compared to the HFD/STZ group, CrHis significantly increased body weight and reduced blood glucose in diabetic rats (p < 0.001). Concentrations of Cr, Zn, Se, and Mn in serum, liver, and kidney of the diabetic rats were significantly lower than in the control rats (p < 0.0001). In contrast, higher Fe and Cu levels were found in serum and tissues from diabetic versus the non-diabetic rats (p < 0.001). Chromium histidinate supplementation increased serum, liver, and kidney concentrations of Cr and Zn both in diabetic and non-diabetic rats (p < 0.001). Chromium supplementation increased Mn and Se levels in diabetic rats (p < 0.001); however, it decreased Cu levels in STZ-treated group (p < 0.001). Chromium histidinate supplementation did not affect Fe levels in both groups (p > 0.05). The results of the present study conclude that supplementing Cr to the diet of diabetic rats influences serum and tissue Cr, Zn, Se, Mn, and Cu concentrations.  相似文献

We investigated the effects of supplemental chromium (Cr) as Cr (III) picolinate on pigs fed high-fat diets (HFD) in a 56-day experiment. Thirty-two crossbred pigs (9.6 kg) were allotted to four treatments with four blocks and two pigs/pen. Treatments included: (1) low-fat diet (fat < 3.5%; LFD) with no Cr, (2) HFD (fat > 30%) with no Cr, (3) HFD with 1,000 ppb Cr, and (4) HFD with 2,000 ppb Cr. Pigs fed HFD gained weight faster, consumed less, and had lower feed:gain (p < 0.05). Pigs fed HFD had higher respiration rates than pigs fed LFD on d 41 (p < 0.05). Plasma insulin on d 14 linearly decreased with Cr (p = 0.05). Plasma cholesterol concentrations were higher in the pigs fed HFD than those fed LFD, but were largely unaffected by supplemental Cr. Consumption of HFD resulted in greater carcass weight, perirenal fat, and backfat measures (p < 0.01) compared with the LFD group. Cr resulted in linear reductions of hot carcass weight (p = 0.08) and average backfat (p < 0.05). The effects of Cr on carcass fat measures were more pronounced in castrated males than in females. These results indicate that Cr attenuates some effects of a HFD, mainly body fat accretion of pigs, and especially in castrated pigs.  相似文献

Tissue-specific accumulation of phenylpropanoids was studied in mycorrhizas of the conifers, silver fir (Abies alba Mill.), Norway spruce [Picea abies (L.) Karst.], white pine (Pinus strobus L.), Scots pine (Pinus silvestris L.), and Douglas fir [Pseudotsuga menziesii (Mirbel) Franco], using high-performance liquid chromatography and histochemical methods. The compounds identified were soluble flavanols (catechin and epicatechin), proanthocyanidins (mainly dimeric catechins and/or epicatechins), stilbene glucosides (astringin and isorhapontin), one dihydroflavonol glucoside (taxifolin 3′-O-glucopyranoside), and a hydroxycinnamate derivative (unknown ferulate conjugate). In addition, a cell wall-bound hydroxycinnamate (ferulate) and a hydroxybenzaldehyde (vanillin) were analysed. Colonisation of the root by the fungal symbiont correlated with the distribution pattern of the above phenylpropanoids in mycorrhizas suggesting that these compounds play an essential role in restricting fungal growth. The levels of flavanols and cell wall-bound ferulate within the cortex were high in the apical part and decreased to the proximal side of the mycorrhizas. In both Douglas fir and silver fir, which allowed separation of inner and outer parts of the cortical tissues, a characteristic transversal distribution of these compounds was found: high levels in the inner non-colonised part of the cortex and low levels in the outer part where the Hartig net is formed. Restriction of fungal growth to the outer cortex may also be achieved by characteristic cell wall thickening of the inner cortex which exhibited flavanolic wall infusions in Douglas fir mycorrhizas. Long and short roots of conifers from natural stands showed similar distribution patterns of phenylpropanoids and cell wall thickening compared to the respective mycorrhizas. These results are discussed with respect to co-evolutionary adaptation of both symbiotic partners regarding root structure (anatomy) and root chemistry. Acute exposure to hexavalent chromium [Cr(VI)] compounds can cause hepatotoxicity. Reactive intermediates and free radicals generated during reduction process may be responsible for Cr(VI) toxicity. In this study, the effects of pretreatment or posttreatment of taurine on Cr(VI)-induced oxidative stress and chromium accumulation in liver tissue of Swiss Albino mice were investigated. Single intraperitoneal (ip) potassium dichromate treatment (20 mgCr/kg), as Cr(VI) compound, significantly elevated the level of lipid peroxidation as compared with control group (p < 0.05). This was accompanied by significant decreases in nonprotein sulfhydryls (NPSHs) level, superoxide dismutase (SOD), and catalase (CAT) enzyme activities as well as a significant chromium accumulation in the tissue (p < 0.05). Taurine administration (1 g/kg, ip) before or after Cr(VI) exposure resulted in reduction of lipid peroxidation (p < 0.05) showed rebalancing effect on tissue NPSH levels either in pretreatment or in posttreatment (p < 0.05). Enzyme activities of SOD and CAT were restored by taurine pretreatment (p < 0.05), whereas posttreatment had less pronounced effects on these parameters. On the other hand, taurine treatment, before or after exposure, could exert only slight decreases in tissue Cr levels (p > 0.05). In view of the results, taurine seems to exert some beneficial effects against Cr(VI)-induced oxidative stress in liver tissue.  相似文献

Present study investigates relationships between total and bioaccessibility of trace elements (Cd, Co, Cr, Cu, Mn, NI, Pb, V, and Zn) concentrations in sediment and their bioaccumulation in species in Shadegan wetland in southwest of Iran. Bioavailability factor (BAF) and translocation factor (TF) were calculated in plants and trophic transfer factor (TTF) was determined in bird species. For this purpose, sampling of sediments, aquatic plants including Phragmites australis, Typha australis, Scripus maritimus and two bird species encircling Porphyrio porphyrio and globally threatened Marmaronetta angustirostris were carried out during winter 2009. Result of chemical analysis show that bioaccessibility concentrations of Mn (8.31 mg/kg), V (1.33 mg/kg), and Pb (1.03 mg/kg) are higher than other metals. The uptake trend of trace elements in plant decreases as root > stem > leaf. Accumulation levels of trace elements in different tissues of P. porphyrio and M. angustirostris are almost identical and considerable. Accumulation and toxicity of Cd in birds is more than plants. In addition, BAF of V, Pb, and Cr indicates high accumulation by plants and great pollution rate in the area of study. In S. maritimus TF for Mn, Cu, Pb, and V are high whereas in T. australis, Cu and Pb posses the highest TF. Also Cr, Co, Mn, Ni, and Zn have higher TF from stem to leaf than root to stem in P. australis. Finally, TTFs were compared in various bird species.  相似文献

This study was conducted to determine the effects of dietary supplementation with Cr nicotinate and Cr chloride and their optimum inclusion rate on performance, carcass traits, meat oxidative stability, serum metabolites, hematological parameters, and liver chromium concentration in heat-stressed broilers. A total number of 420, 1-day-old male broiler chicks were randomly assigned to seven treatments with four replicates of 15 chicks. The dietary treatments consisted of the basal diet supplemented with 0 (control), 500, 1,000, and 1,500 μg/kg Cr in the form of Cr nicotinate and Cr chloride. Chicks were raised for 6 weeks in heat stress condition (33 ± 2°C). Supplements of organic and inorganic Cr particularly at 1,500 μg/kg incorporation increased feed consumption (P < 0.05) and body mass gain of broilers (P < 0.01). Cr supplementation increased carcass yield and decreased abdominal fat (P < 0.01). Supplementation of 1,500 μg/kg Cr nicotinate (P < 0.05) enhanced liver Cr concentration. Storage time increased lipid oxidation of meat (P < 0.01). Cr decreased lipid oxidation of breast and thigh muscles over 2 (P < 0.01) or 6 (P < 0.05) days of storage time. Birds fed 1,500 μg/kg Cr nicotinate, had lower concentration of serum glucose and triglyceride at 21 days (P < 0.05). Hematological parameters tested at 21 and 42 days, were not influenced. The results suggested that dietary Cr supplementation regardless of its source have a positive effect on productive, and carcass traits, also enhances oxidative stability of refrigerated meat in broilers reared under heat stress conditions.  相似文献

Bifidobacterium longum grew at 65 L pilot scale of the membrane bioreactor (MBR), externally fitted with ceramic membrane (0.7 m2). Cell mass at the MBR reached 22.18 g L−1 as dry cell weight in 12 h, which is 8.44 times higher than cell mass attained at the vial culture. The growth rate in the vial culture was μ = 0.385 h and at the batch culture was μ = 1.13 h in the exponential period and μ = 0.31 h−1 in the stationary period. In the fed-batch mode was μ = 1.102 h−1 for 6 h with inoculation and declined to μ = 0.456 h−1 with feeding of feed medium. The growth rate at the MBR was μ = 0.134 h−1. The number of viable cells was 6.01 × 1012 cfu L−1 at the batch culture, but increased to 1.15 × 1014 cfu L−1 at the MBR culture. The specific growth rate of viable cell number (colony-forming units per liter, per hour) improved by 6.01 times from the batch to the MBR culture. The wall shear stress mainly generated by the pump, and the membrane incorporated into the MBR was controlled during the cultivation at the MBR. The viability of B. longum declined to under 10% in the first 2 weeks of the 4-week stability test (40°C) as B. longum was exposed to over wall shear stress 713 Pa, but the viability improved to 30–40% in wall shear stress of 260 Pa or STR culture. The loss in the cell viability can be saved by managing with wall shear stress during the cultivation at the MBR.  相似文献

The effects of supplemental chromium (Cr) as chromic chloride hexahydrate in incremental dose levels (0, 0.5, 1.0, and 1.5 mg/day for 240 days) on metabolism of nutrients and trace elements were determined in dwarf Bengal goats (Capra hircus, castrated males, average age 3 months, n = 24, initial mean body weight 6.4 ± 0.22 kg). Live weight increased linearly (p < 0.05) with the level of supplemental Cr. Organic matter and crude protein digestibility, intake of total digestible nutrients, and retention of N (g/g N intake) increased (p < 0.05) in a dose-dependent linear manner. Serum cholesterol and tryacylglycerol concentrations changed inversely with the dose of supplemental Cr (p < 0.01). Supplemental Cr positively influenced retention of copper and iron (p < 0.05) causing linear increase (p < 0.01) in their serum concentrations. It was concluded that Cr supplementation may improve utilization of nutrients including the trace elements and may also elicit a hypolidemic effect in goats. However, further study with regards to optimization of dose is warranted.  相似文献

Trace element content in hair is affected by the age of the donor. Hair samples of subjects from four counties in China where people are known to have long lifespan (“longevity counties”) were collected and the trace element content determined. Samples were subdivided into three age groups based on the age of the donors from whom these were taken: children (0–15 years); elderly (80–99 years); and centenarians (≥100 years). We compared the trace element content in hair of different age groups of subjects. Support vector machine classification results showed that a non-linear polynomial kernel function could be used to classify the three age groups of people. Age did not have a significant effect on the content of Ca and Cd in human hair. The content of Li, Mg, Mn, Zn, Cr, Cu, and Ni in human hair changed significantly with age. The magnitude of the age effect on trace element content in hair was in the order Cu > Zn > Ni > Mg > Mn > Cr > Li. Cu content in hair decreased significantly with increasing age. The hair of centenarians had higher levels of Li and Mn, and lower levels of Cr, Cu, and Ni comparing with that of the children and elderly subjects. This could be a beneficial factor of their long lifespan.  相似文献

Cellular events which occur prior to cell collapse were examined in the root cortex of rice (Oryza sativa L.) during aerenchyma formation. Cell collapse started at a specific position in the mid cortex. These cells were distinct in shape from those located towards the periphery. Furthermore, cell collapse was preceded by acidification and the loss of plasma-membrane integrity in cells of the mid cortex. Subsequent death of neighboring cells followed a radial path. Microinjection of molecules of different sizes conjugated with fluorescein isothiocyanate (FITC) showed a molecular exclusion limit of between 9.3 and 19.6 kDa in the root cortex. Furthermore, large molecules, i.e. those around 9.3 kDa, were predominantly transferred in a radial direction, which coincided with the path of sequential cell death. The mRNA and protein for NADH-dependent glutamate synthase (NADH-GOGAT; EC 1.4.1.14) in root tips of rice (Oryza sativa L. cv. Sasanishiki) plants increases dramatically within 12 h of supplying a␣low concentration (>0.05 mM) of ammonium ions (T.␣Yamaya et al., 1995, Plant Cell Physiol 36: 1197–1204). To identify the specific cells which are responsible for this rapid increase, the cellular localization of NADH-GOGAT protein was investigated immunocytologically with an affinity-purified anti-NADH-GOGAT immunoglobulin G. When root tips (>1 mm) of rice seedlings which had been grown for 26 d in water were immuno-stained, signals for the NADH-GOGAT protein were detected in the central cylinder, in the apical meristem, and in the primordia of the secondary roots. Signals for ferredoxin-dependent GOGAT (Fd-GOGAT; EC 1.4.7.1) protein were also seen in the same three areas. When the roots were supplied with 1 mM ammonium ions for 24 h, there were strong signals for the NADH-GOGAT protein in two cell layers of the root surface, i.e. epidermis and exodermis, in addition to the cells giving signals in the absence of ammonium ions. The supply of ammonium ions was less effective on the profile of signals for Fd-GOGAT. Although the supply of ammonium ions had less effect on the expression of cytosolic glutamine synthetase (GS; EC 6.3.1.2), this enzyme was also found to be located in the epidermis and exodermis, as well as in the central cylinder and cortex. The results indicate that NADH-GOGAT, coupled to the cytosolic GS reaction, is probably important for the assimilation of ammonium ions in the two cell layers of the root surface. The present study was carried out to assess the impact of wastewater on parsley (Petroselinum crispum). The parameters determined for soil were pH, electrical conductivity (EC), soil organic matter (SOM), nutrient elements (Ca, Mg, Na, K, Mn, Cu, Zn, and Fe), and heavy metals (Cd, Cr, Ni, and Pb), while the parameters determined for the plant included pigment content, dry matter, nutrient element, and heavy metals. SOM, EC, and clay contents were higher, and pH was slightly acidic in soil treated with wastewater compared to control soil. The enrichment factors (EF) of the nutrient elements in contaminated soil are in the sequence of Na (2) > Ca (1.32) > Mn = Mg (1.17) > Cu (1.11) > Zn (1.08) > Fe (1.07) > K (0.93), while EF in parsley are Na (6.63) > Ca (1.60) > Mg (1.34) > Zn (1.15) > Fe (0.95) > Cu = K (0.90) > Mn (0.85). Application of wastewater significantly decreased dry matter, while photosynthetic pigment content increased in parsley. The enrichment of the heavy metals is in the sequence: Cd (1.142) > Pb (1.131) > Ni (1.112) > Cr (1.095). P. crispum shows a high transfer factor (TF > 1) for Cd signifying a high mobility of Cd from soil to plant. Thus, although the wastewater irrigation in parsley production aims to produce socioeconomic benefits, study results indicated that municipal wastewater is not suitable for irrigation of parsley because it has negative effects on plant and causes heavy metal accumulation.  相似文献

In 10- to 16-year-old children, inhabitants of industrial towns of Ukraine, an X-ray fluorescence analysis of the content of some chemical elements in hair samples allowed us to find a deficiency of Zn and Cu against the background of excesses of Ca, Cr, Ni, Mo, and Cd. Comparison of the parameters of EEG frequency components and levels of Ca, Zn, Cu, Fe, Mn, Co, Cr, Mo, Ni, Sr, Pb, and Cd showed that there are significant correlations between the normalized values of spectral powers of many frequency components of the ongoing EEG recorded in different functional states (eyes closed/open, solving an arithmetic task, and phono-/photostimulation) and concentrations of ten of the above-mentioned trace elements. Comparative estimation of the neurotropicity of the elements showed the following sequence of numbers of significant correlations (shown in parentheses) from the total set of possible comparisons: Cd (35) > Ni (31) > Cr (19) > Sr (17) > Pb (16) > > Ca (10) > Cu (7) > Mo (3) > Zn (2) > Fe (1). The intensity of correlations (values of the correlation coefficients) varied from 0.26 to 0.42 at 0.05 < P < 0.001; more frequently, such correlations were observed under conditions of EEG recording with the eyes closed (39) and upon solving an arithmetic task (33).  相似文献

We present an analysis of X-ray-induced damage in ΦX174 plasmid DNA, applying doses between D = 250 and 1,500 Gy. To analyse this damage in detail, the distribution of plasmid fragments after irradiation have been determined by scanning force microscopy. The results show that even for the lowest dose of D = 250 Gy, a significant amount of double-strand breaks are observed. For increasing dose, the percentage of small fragments increases and is accompanied by a shortening of the average fragment length from < L> = 1,400 nm for a dose of D = 250 Gy to < L> = 1,080 nm after irradiation with D = 1,500 Gy. The most crucial parameter, the average number of double-strand breaks per broken plasmid (<DSBb> ) has been determined for the first time for the applied doses. The results show that the average number of DSBs per broken plasmid <DSBb> increases almost linearly from a value of <DSBb> = 1.3 after irradiation with D = 250 Gy to <DSBb> = 1.7 after exposure to D = 1,500 Gy. The presented results show that the amount of DSBs induced by X-ray radiation in plasmid DNA can be calculated with high accuracy by means of scanning force microscopy, providing relevant information regarding the interaction of X-rays with DNA molecules.

The single and joint toxicity effects of Cu, Cr(III), and Cr(VI) on the root elongation of pakchoi in solution and soil were investigated. The median effective concentration (EC50) was determined to examine the toxic thresholds of the test elements. The results showed that individual contamination by Cu, Cr(III), or Cr(VI) can inhibit the root elongation of pakchoi. The EC50 values of the test elements were 2.02 mg/L and 195.8 mg/kg, 62.2 mg/L and 1,773 mg/kg, and 6.88 mg/L and 8.08 mg/kg in solution and soil, respectively. Toxic unit (TU) was introduced to determine the outcome in combined tests, and different behaviors were observed in both solution and soil. The coexistence of Cu and Cr(III) in solution exhibited an antagonistic effect (EC50mix = 1.76 TUmix), whereas a synergistic effect was observed in soil (EC50mix = 0.76 TUmix). In contrast, combined Cu–Cr(VI) showed a less than additive toxicity both in solution and soil, with EC50mix values of 3.31 and 1.24 TUmix. In conclusion, the coexistence of toxicity in Cu–Cr(III) and Cu–Cr(VI) differs from the toxicity exhibited individually by Cu, Cr(III), and Cr(VI). Heavy metal interaction also changes depending on the medium.  相似文献

An attempt has been made to analyze some trace elements and electrolytes like Zn, Cu, Mn, Fe, Co, Na, K, Ca, and Li present in the Swertia chirayita roots and leaves. The concentration of Ca in all the samples was more than 1,346.0 mg/kg and the concentration of other elements was found in the order K > Ca > Fe > Na > Mn > Zn > Co > Cu > Li in different samples of S. chirayita.  相似文献

To identify the importance of arbuscular mycorrhizal fungi (AMF) colonizing wetland seedlings following flooding, we assessed the effects of AMF on seedling establishment of two pioneer species, Bidens frondosa and Eclipta prostrata grown under three levels of water availability and ask: (1) Do inoculated seedlings differ in growth and development from non-inoculated plants? (2) Are the effects of inoculation and degree of colonization dependent on water availability? (3) Do plant responses to inoculation differ between two closely related species? Inoculation had no detectable effects on shoot height, or plant biomass but did affect biomass partitioning and root morphology in a species-specific manner. Shoot/root ratios were significantly lower in non-inoculated E. prostrata plants compared with inoculated plants (0.381 ± 0.066 vs. 0.683 ± 0.132). Root length and surface area were greater in non-inoculated E. prostrata (259.55 ± 33.78 cm vs. 194.64 ± 27.45 cm and 54.91 ± 7.628 cm2 vs. 46.26 ± 6.8 cm2, respectively). Inoculation had no detectable effect on B. frondosa root length, volume, or surface area. AMF associations formed at all levels of water availability. Hyphal, arbuscular, and vesicular colonization levels were greater in dry compared with intermediate and flooded treatments. Measures of mycorrhizal responsiveness were significantly depressed in E. prostrata compared with B. frondosa for total fresh weight (−0.3 ± 0.18 g vs. 0.06 ± 0.06 g), root length (−0.78 ± 0.28 cm vs.−0.11 ± 0.07 cm), root volume (−0.49 ± 0.22 cm3 vs. 0.06 ± 0.07 cm3), and surface area (−0.59 ± 0.23 cm2 vs.−0.03 ± 0.08 cm2). Given the disparity in species response to AMF inoculation, events that alter AMF prevalence in wetlands could significantly alter plant community structure by directly affecting seedling growth and development.  相似文献

This experiment was carried out to investigate the effects of different levels of organic and inorganic chromium on the performance, immune function and some serum mineral concentrations of broilers under heat stress condition (23.9–37°C cycling). A total of 150 one-day-old broiler chicks according to a completely randomized design were assigned into five treatment groups. Each treatment consisted of three replicates and each replicate contained ten chicks. Chicks were fed on corn–soybean meal basal diets with added different concentrations of chromium (0, 600 and 1,200 μg kg−1 chromium chloride or 600 and 1,200 μg/kg chromium l-methionine) from 1 to 49 days of age. Humoral immunity was assessed by intravenous injection of 7% sheep red blood cell (SRBC) followed by evaluation of serum for antibody titers in primary and secondary responses. Cell-mediated immunity was assessed by the cutaneous basophil hypersensitivity (CBH) test to phytohemagglutinin (PHA)-P at day 32 and PHA-M at day 48. Heterophil/lymphocyte (H/L) ratio was also measured as a reliable indicator of stress. The body mass, feed intake and conversion ratio were not influenced by dietary chromium (P > 0.05). Dietary supplementation of both organic and inorganic chromium significantly increased primary and secondary antibody responses (P < 0.01), and also improved H/L ratio (P < 0.05), CBH response (P < 0.01) as well as relative weights of thymus (P < 0.05) and spleen (P < 0.01). Both dietary organic and inorganic chromium caused an increase in serum concentrations of Cr and Zn (P < 0.01), but decreased the serum concentration of Cu (P < 0.01). These results suggest that supplemental chromium especially in organic form offers a good management practice to reduce heat stress-related depression in immunocompetence of broiler chicks.  相似文献

A fluorimetric ratio technique was elaborated to measure apoplastic pH in the outer root cortex of maize (Zea mays L.) grown hydroponically. A newly synthesized fluorescent probe, fluorescein boronic acid (pKa = 5.48), which covalently binds to the cell wall of the outer cell layers, was used. Under conditions of saturating ion concentrations the apoplastic pH was determined along the root axis ranging from 1 to 30 mm behind the root tip. Apoplastic pH was recorded for root segment areas (1 mm2), and pH values of high statistical significance were obtained. With an external solution of pH 5, the apoplastic pH was about pH 5.1 in the division zone, between pH 4.8 and 4.9 in the elongation region and about pH 4.9 in the root hair zone. At an external pH of 8.6, the difference between the external pH and the apoplastic pH was considerably more, with a pH of 5.2–5.3 in all root zones. Addition of 1 mM NH4 + caused a small apoplastic pH decrease (0.05 of a pH unit) in all root zones. Apoplastic alkalization upon application of 6 mM NO3 was highest (0.3 of a pH unit) in the zone where root hairs emerge; in the division and early elongation zones, apoplastic pH increased only transiently. In the presence of 10 mM HCO3 , NO3 elicited a higher and persistent alkalization (0.06–0.25 of a pH unit) in all root zones. Application of fusicoccin reduced apoplastic pH from 4.85 to 4.75 in the elongation zone, while inhibition of the H+-ATPase with vanadate alkalized the apoplast in the root hair zone from pH 5.4 to 5.6. The observed pH differences along the root axis upon differential N supply and application of HCO3 provide evidence that this new pH technique is a useful tool with which to measure apoplastic pH, and in future may permit measurements at microsites at the cell level by use of microscope imaging. The shoot apex of Dennstaedtia cicutaria consists of three zones—a zone of surface initials, a zone of subsurface initials, and a cup-shaped zone that is subdivided into a peripheral region and central region. A diffuse primary thickening meristem, which is continuous with the peripheral region of the cup-shaped zone, gives rise to a broad cortex. The roots occurring on the rhizomes are initiated very near the shoot apex in the outer derivatives of the primary thickening meristem. The roots that occur on the leaf bases also differentiate from cortical cells. Eventually, those cortical cells situated between the newly formed root apical cell and the rhizome procambium (or leaf trace) differentiate into the procambium of the root trace, thus establishing procambial continuity with that of the rhizome or leaf trace. Parenchymatous root gaps are formed in the rhizome stele and leaf traces when a few of their procambial cells located directly above the juncture of the root trace procambium differentiate into parenchyma. As the rhizome procambium or leaf trace continues to elongate, the parenchyma cells of the gap randomly divide and enlarge, thus extending the gap.  相似文献
