Ejector loop reactors (ELR) are successfully used in industrial chemical processes for gas/liquid reactions. They achieve higher mass transfer rates compared to the stirred‐tank reactor (STR) at comparable specific power input. Insufficient oxygen transport and shear stress induced growth inhibition are limiting parameters during microbial fermentation. Due to its better mass transfer characteristics, the ELR was expected to have beneficial effects on biomass and recombinant protein production. One concern, however, was whether the ELR's shear stress characteristics would have a negative effect. This study evaluated the suitability of using the Buss‐Loop® Reactor (BLR), one of the most advanced ELR technologies, as a bioreactor. The well‐studied STR was used as a reference. A lab scale BLR was adapted for microbial fermentation. Mass transfer rates and specific power inputs were within the same order of magnitude in the ELR and the reference STR. Maximum values of 207 and 205 h?1 at power inputs of 6.9 and 9.7 W/L were measured in the ELR and STR, respectively. During batch fermentation of Escherichia coli K12 MG1655, maximum cell densities were higher in the ELR (OD600 of 22) than in the STR (OD600 of 18). Green fluorescence protein (GFP) production with pGS1 was comparable; however, more GFP was released into the media in the ELR. This indicates higher cell disruption compared to the STR. Despite this drawback of the first prototype, our work clearly demonstrates the potential of the ELR as a system for microbial fermentations.  相似文献

The glucose oxidase system was adapted for estimation of the overall oxygen transfer rate in a periodic pressure oscillating, solid-state bioreactor. Enzyme concentration of 40 ml enzyme preparation L−1 was found adequate to give linear gluconic acid production and attain maximal oxygen absorption rates. At 4 atm and 30°C, the oxygen transfer rate reached 892 mmol kg−1 initial dry matter h−1 in this system, while only 121 mmol kg−1 initial dry matter h−1 was obtained in a conventional static tray bioreactor.  相似文献

Miniature parallel bioreactors are becoming increasingly important as tools to facilitate rapid bioprocess design. Once the most promising strain and culture conditions have been identified a suitable scale-up basis needs to be established in order that the cell growth rates and product yields achieved in small scale optimization studies are maintained at larger scales. Recently we have reported on the design of a miniature stirred bioreactor system capable of parallel operation [Gill et al. (2008); Biochem Eng J 39:164-176]. In order to enable the predictive scale-up of miniature bioreactor results the current study describes a more detailed investigation of the bioreactor mixing and oxygen mass transfer characteristics and the creation of predictive engineering correlations useful for scale-up studies. A Power number of 3.5 for the miniature turbine impeller was first established based on experimental ungassed power consumption measurements. The variation of the measured gassed to ungassed power ratio, P(g)/P(ug), was then shown to be adequately predicted by existing correlations proposed by Cui et al. [Cui et al. (1996); Chem Eng Sci 51:2631-2636] and Mockel et al. [Mockel et al. (1990); Acta Biotechnol 10:215-224]. A correlation relating the measured oxygen mass transfer coefficient, k(L)a, to the gassed power per unit volume and superficial gas velocity was also established for the miniature bioreactor. Based on these correlations a series of scale-up studies at matched k(L)a (0.06-0.11 s(-1)) and P(g)/V (657-2,960 W m(-3)) were performed for the batch growth of Escherichia coli TOP10 pQR239 using glycerol as a carbon source. Constant k(L)a was shown to be the most reliable basis for predictive scale-up of miniature bioreactor results to conventional laboratory scale. This gave good agreement in both cell growth and oxygen utilization kinetics over the range of k(L)a values investigated. The work described here thus gives further insight into the performance of the miniature bioreactor design and will aid its use as a tool for rapid fermentation process development.  相似文献

In this study, we evaluated the feasibility of using mass cultivation of the adventitious roots of Echinacea purpurea in balloon type bubble (air-lift) bioreactors to produce caffeic acid derivatives, which have pharmaceutical and therapeutic values. An approximately 10 fold increase in biomass and secondary compounds was observed after 4 weeks of culture in balloon type bubble bioreactors (5 L capacity containing 4 L of half strength MS medium). In addition, a linear relationship was observed between the concentration of biomass and the sucrose and ion consumption rate. Furthermore, the concentration of biomass in the bioreactor culture was found to increase as the conductivity decreased. An inoculum density of 7 g/L FW and an aeration rate of 0.1 vvm were found to be suitable for inducing the accumulation of biomass and secondary metabolites. Of the three caffeic acid derivatives evaluated (caftaric acid, chlorogenic acid, and cichoric acid), the concentration of cichoric acid was the highest (26.64 mg/g DW).  相似文献

A method for the continuous culture of microorganisms is described which employs growth-dependent pH changes to control the rate of addition of fresh medium to a culture vessel. The apparatus (the phauxostat) supports, at constant pH, longterm continuous culture at rates near or at the maximum of which the organisms are capable. The buffering capacity of the inflowing medium determines the steady-state population density of the culture, but the rate of growth is independent of the buffering capacity. The fundamental theory of operation is tested and some basic parameters of growth are estimated using Escherichia coli B growing continuously in media containing glucose, glycerol or DL-lactate.Joseph C. Soluble microbial products (SMPs) tend to accumulate in the membrane bioreactor (MBR) systems as a consequence of high membrane rejection and apparently low biodegradability within the wastewater treatment system. The extension of the activated sludge models (ASMs) with SMPs, therefore, has received crucial importance in recent days, particularly considering their potential use as indicators of the membrane fouling propensity. This paper presents a critical review of the formation and degradation kinetics of SMP subdivisions that have so far been used for the mathematical modelling of MBR. The paper identified a simplified approach to incorporate the kinetics of the SMP formation and degradation in the general mathematical models of MBR. It suggested that the inclusion of only four additional linear differential equations in the ASM1-SMP integrated mathematical model could simulate well the effluent quality and membrane fouling prediction. The model would also serve as a useful tool in optimizing operation conditions for better treatability and fouling control.  相似文献
