Pinus sylvestris seedlings, colonised by ectomycorrhizal (EM) fungi from either of two different soils (untreated forest soil and a limed soil from a clear cut area), were grown with or without biotite as a source of K. The biotite was naturally enriched in 87Sr and the ratio of 87Sr/ 86Sr in the plant biomass was estimated and used as a marker for biotite weathering and compared to estimates of weathering based on foliar content of K. Different nutrient regimes were used to expose the seedlings to deficiencies of K with and without an application of nitrogen (NH4NO3) in excess of seedling demand. The seedlings were grown for 220 days and the elemental composition of the shoots were analysed at harvest. The EM colonisation was followed by analysing the concentration of ergosterol in the roots and the soils. Bacterial activity of the soil was estimated by the thymidine incorporation technique. The concentration of organic acids in the soil solution was measured in the soil in which seedlings colonised by EM fungi from the untreated forest soil were grown. It was found that seedlings colonised by EM fungi from untreated forest soil had taken up more K in treatments with biotite addition compared to seedlings colonised by EM fungi from the limed forest soil (p<0.05). Seedlings from untreated forest soil had larger shoots and contained more K when grown with biotite compared to KCl as K source, indicating that biotite had a stimulatory effect on the growth of these seedlings which was not related to K uptake. Seedlings from the limed soil, on the other hand, had similar foliar K content when grown with either biotite or KCl as K source. The larger uptake of K in seedlings from untreated forest soil was not an effect of a more developed EM colonisation of the roots since seedlings from the limed soil had a higher ergosterol concentration both in the soil and in the roots. Nutrient regimes had no significant influence on the total uptake of K but the 87Sr/ 86Sr isotope ratio in the plant biomass indicated that seedlings grown with excess nitrogen supply had taken up proportionally less Sr from the biotite (1.8% of total Sr content) compared to seedlings grown with a moderate nitrogen supply (5.0%). Furthermore, seedlings grown with excess nitrogen supply had a reduced fungal colonisation of roots and soil and bacterial activity was lower in these soils. The 87Sr/ 86Sr ratio in the plant biomass was positively correlated with fungal colonisation of the roots (r 2=0.98), which may indicate that the fungus was involved in releasing Sr from the biotite. Uptake of K from biotite was not related to the amount of organic acids in the soil solution. Oxalic acid was positively related to the amount of ergosterol in the root, suggesting that oxalic acid in the soil solution originates from the EM symbionts. The accuracy of the estimations of biotite weathering based on K uptake by the seedlings in comparison with the 87Sr/86Sr isotope ratio measured in the shoots is discussed. The aim of this study was to test the potential of four isolates of ectomycorrhizal (EM) fungi to utilize organic nitrogen (N) at two different substrate pHs. The organic N source (15N labelled lyophilised fungal mycelium) was mixed with either untreated peat/sand mixture (pH 4.9) or peat/sand mixture limed to a pH of 5.9 and put in cylindrical containers added to each pot. The content of the containers was separated from the roots of Pinus sylvestris seedlings by a nylon mesh and a 2 mm air gap to reduce diffusion of labelled N to the roots. The mycorrhizal plants (except those colonized by Suillus variegatus 2) took up significantly more 15N from the labelled mycelium than uncolonized seedlings. Liming significantly reduced the uptake of 15N by one of the EM fungi (unidentified) but not the other tested species (Paxillus involutus and two isolates of S. variegatus). The EM fungal isolates differed in their influence on the bacterial activity of the soil. This was reduced with P. involutus at both pH levels and increased with one of the two S. variegatus isolates at the high pH and with the other S. variegatus isolate at the low pH level. Liming the soil generally increased bacterial activity. The influence of liming on the proportion of organic N uptake in relation to inorganic N uptake by ectomycorrhizal trees is discussed.  相似文献

Pinus sylvestris seedlings infected with either the ectomycorrhizal (ECM) fungus Paxillus involutus or Suillus variegatus were exposed to a range of Cd or Zn concentrations. This was done to investigate the relationship between the sensitivity of ECM fungi and their host plants over a wide range of concentrations. P. involutus ameliorated the toxicity of Cd and Zn to P. sylvestris with respect to root length, despite significant inhibition of ECM infection levels by Cd (Cd EC50 [effective concentration which inhibits ECM infection by 50%] values were: P. involutus 3.7 μg g-1 Cd; S. variegatus 2.3 μg g-1 Cd). ECM infection by P. involutus also decreased Cd and Zn transport to the plant shoots at potentially toxic concentrations and also influenced the proportion of Zn transported to the roots and shoots, with a higher proportion retained in the roots of the seedlings. ECM infection did increase host biomass production, but this was not affected by the presence of Cd or Zn. Root and shoot biomass production by P. sylvestris, in both the presence and absence of ECM fungi, was unaffected by Cd and Zn at all concentrations tested.  The abilities of three ectomycorrhizal fungi, Paxillus involutus, Suillus variegatus and Lactarius rufus, to utilize organic and inorganic nitrogen sources were determined by measuring the growth and endogenous free polyamines (putrescine, spermidine and spermine) of pure culture mycelium. Differences were found in the utilization of the nitrogen sources and in the polyamine concentrations between the fungal species and between isolates of L. rufus. All the fungi grew well on ammonium and on several amino acids. Endogenous polyamine levels varied with the nitrogen source. Spermidine was commonly the most abundant polyamine; however, more putrescine than spermidine was found in P. involutus growing on inorganic nitrogen or arginine. Low amounts of spermine were found in S. variegatus and some samples of L. rufus. None or only a trace of spermine was found in P. involutus mycelium. In all fungi, putrescine concentrations were higher with ammonium than with the nitrate treatment. The total nitrogen content of peat did not determine the ability of L. rufus strains isolated from peatland forest sites to utilize organic nitrogen. Accepted: 27 November 1998  相似文献

 Nitrogen deposition and intentional forest fertilisation with nitrogen are known to affect the species composition of ectomycorrhizal fungal communities. To learn more about the mechanisms responsible for these effects, the relations between fungal growth, nitrogen uptake and nitrogen availability were studied in ectomycorrhizal fungi in axenic cultures and in symbiosis with pine seedlings. Effects of different levels of inorganic nitrogen (NH4) on the mycelial growth of four isolates of Paxillus involutus and two isolates of Suillus bovinus were assessed. With pine seedlings, fungal uptake of 15N-labelled NH4 was studied in short-term incubation experiments (72 h) in microcosms and in long-term incubation experiments (3 months) in pot cultures. For P. involutus growing in symbiosis with pine seedlings, isolates with higher NH4 uptake were affected more negatively at high levels of nitrogen availability than isolates with lower uptake. More NH4 was allocated to shoots of seedlings colonised by a high-uptake isolate, indicating transfer of a larger fraction of assimilated NH4 to the host than with isolates showing lower NH4 uptake rates. Thus low rates of N uptake and N transfer to the host may enable EM fungi avoid stress induced by elevated levels of nitrogen. Seedlings colonised by S. bovinus transferred a larger fraction of the 15N label to the shoots than seedlings colonised by P. involutus. Seedling shoot growth probably constituted a greater carbon sink in pot cultures than in microcosms, since the mycelial growth of P. involutus was more sensitive to high NH4 in pots. There was no homology in mycelial growth rate between pure culture and growth in symbiosis, but N uptake in pure culture corresponded to that during growth in symbiosis. No relationship was found between deposition of antropogenic nitrogen at the sites of origin of the P. involutus isolates and their mycelial growth or uptake of inorganic nitrogen. Kinetics of net phosphate (Pi) uptake was measured on intact ectomycorrhizal and non‐mycorrhizal Pinus sylvestris seedlings using a semihydroponic cultivation method. The depletion of Pi in a nutrient solution was assessed over a 160–0.2 μM Pi gradient. Growth of the pine seedlings was P limited and measurements were performed 7 and 9 weeks after inoculation. Three ectomycorrhizal fungi were studied: Paxillus involutus, Suillus bovinus and Thelephoraterrestris. Pi uptake was extremely fast in plants colonised by P. involutus. The Pi concentration dropped below 0.2 μM within 4–5 h. In plants colonised with S. bovinus this occurred in 5–6 h and in plants associated with T. terrestris 8 h were needed to run through the whole concentration range. Non‐mycorrhizal plants of similar size and nutrient status decreased Pi to a concentration between 1 and 2 μM in 18 h. Data were curve fitted to a two‐phase Michaelis‐Menten equation. The apparent kinetic constants, Km and Vmax, for the high affinity Pi uptake system of the pine roots could be estimated accurately. Vmax of this system was up to 7 times higher in pines associated with P. involutus than in non‐mycorrhizal seedlings. The intact extraradical mycelium greatly increased the absorption surface area of the roots (Vmax). Non‐mycorrhizal plants had a Km between 7.8 and 16.4 μM Pi. Plants mycorrhizal with P. involutus had Km values between 2.4 and 7.2, plants colonised with S. bovinus had a Km between 5.1 and 12.3, and seedlings associated with T. terrestris had a Km from 4.6 to 10.1 μM Pi. All 3 ectomycorrhizal fungi had a strong impact on the Pi absorption capacity of the pine seedlings. The results also demonstrated that there is substantial heterogeneity in kinetic parameters among the different mycorrhizal root systems.  相似文献

 Embryogenic cell masses of three Scots pine (Pinus sylvestris) cell lines K779, K884 and K1009 were cultivated with the ectomycorrhizal (ECM) fungi Laccaria bicolor, L. proxima, Pisolithus tinctorius, Paxillus involutus and two strains of Suillus variegatus. The average growth ratio of the slowly proliferating cell line K1009 was improved by L. proxima and S. variegatus strain H, while of the rapidly proliferating lines K779 and K884 the non-mycorrhizal controls grew best. The fungi caused two distinct reactions in embryogenic cultures. In the positive reaction, the shape and light yellow colour of the cultures resembled the controls, while in the negative reaction the embryogenic cells became brown and necrotic and the fungi grew aggressively over them. These reactions to the fungi did not correlate completely with effects on the growth ratio. All the cell lines enhanced the radial growth of S. variegatus H and of P. tinctorius, while the Laccaria species and S. variegatus strain 1 thrived better alone. This study shows that early-stage embryogenic cells of Scots pine and ECM fungi are able to interact. As some fungi produced a positive reaction or even increased proliferation, they could be used to enhance somatic embryogenesis of Scots pine. Specific fungi might be used to induce the growth of slowly proliferating cell lines, and knowledge of positive cell line-fungus interactions could be useful in work with later stages of somatic embryogenesis, such as rooting. The effects of slow (apatite, biotite) and fast-release nutrients (P, K, Mg) on C and N mineralization in acid forest soil were studied. These nutrients were applied alone or together with urea or urea and limestone. The production of CO2 in the soil samples taken one and three growing seasons after the application was lower in the soils treated with the fast-release nutrients than in the untreated soils. Similar reduction of microbial activity was not seen after the apatite and apatite+biotite treatments. In the first growing season, urea and urea+limestone enhanced CO2 production, but after three growing seasons, the opposite was true. Apatite and apatite+biotite added together with urea did not compensate for the decreasing effect of urea on the CO2 production. The addition of fast-release salts increased somewhat the concentration of NH inf4 sup+ in the soil and more NH4 + accumulated during laboratory incubation in the soil samples taken one growing season after the application. The urea addition immediately increased the concentrations of NH4 + and of NO3 in the soil, but, three growing seasons after application, urea had only a slight increasing effect on mineral N content of the soil. Slow-release nutrients seem to have a more favourable effect than fast-release salts on nutrient turnover in acid forest soil.  相似文献

The higher levels of nitrogen in ammonium form stimulated the growth of mycelia and increased the accessible as well as the total acid phosphatase activity of Paxillus involutus (Batsch) Fr. isolates grown in pure culture. Rates of mycelia growth and acid phosphatase activities varied widely from one isolate to another. Scots pine (Pinus sylvestris L.) seedlings were inoculated with different P. involutus isolates in axenic conditions. Shoots of pine seedlings with mycorrhizae contained more phosphorus than shoots of non-mycorrhizal seedlings. The relations between growth and phosphatase activity of P. involutus isolates and their efficiency in supplying the host plant with phosphate are discussed.  相似文献

A greenhouse experiment was performed to evaluate the effect of Norway spruce (Picea abies (L.) Karst.) seedlings on net nutrient availability in five different growing media containing F- or H-layer and mineral soil originating from a haplic podzol in northern Sweden. The initial total amounts of eight nutrient elements (N, K, P, Ca, Mg, Mn, Fe, Zn) and exchangeable amounts of the same elements were analyzed in pots with or without spruce seedlings. In the planted pots seedling nutrient uptake was also estimated. After 26 weeks, higher net nutrient availability with seedlings was found in 25 out of the 40 (62%) growing media and nutrient element combinations. A positive seedling effect on net nutrient availability might be explained by rhizodeposition stimulating the soil microorganism activity and accelerating the weathering of minerals or by seedling roots promoting the nutrient providing processes through changes in soil chemical and physical properties. Nitrogen availability was primarily affected by what part of the forest floor the growing medium contained although the positive response to seedling presence was apparent. The positive net availability response of P, Ca, Mg, Mn, Fe and Zn to seedling presence was on the other hand relatively strong. In the case of P, K, and Zn the growing medium composition (if the F- and H-layer was pure or mixed with mineral soil) was also an important factor for the estimated net availability. Pure F-and H-layer provided greater P- and K-availability while the availability of Zn increased when mineral soil was added. The influence of growing plants ought to be considered when soil samples are used for assessing the nutrient availability.  相似文献

The effect of three different nitrogen sources on the growth of external ectomycorrhizal mycelium was studied in Perspex micorocosms. Nonsterile peat was used as substrate. Five different fungal isolates growing in symbiosis with pine seedlings were investigated: two isolates of Paxillus involutus, one of Suillus bovinus and two unidentified ectomycorrhizal fungi isolated from ectomycorrhizal root tips. Three different nitrogen sources were used: ammonium as (NH4)2SO4, nitrate as NaNO3 and a complete nutrient solution (Ingestad 1979), and three different nitrogen concentrations, 1, 2 or 4 mg N/g dry wt. of peat. The mycelial growth of all fungi was found to be negatively affected by the nitrogen amendments, although the sensitivity to nitrogen varied between the isolates. One of the unidentified isolates was extremely sensitive and growth was completely inhibited by all nitrogen treatments. In contrast, the growth of one of the P. involutus isolates was only slightly reduced by the nitrogen amendments. The different nitrogen sources all reduced growth, and since no significant difference was found between the nitrogen sources or between the different nitrogen concentrations the results were pooled to give one value that summarized the effect of nitrogen on mycelial growth. Thus, the mycelial growth of one of the two P. involutus isolates was reduced to approximately 80% of the growth in the control, the other P. involutus and one of the unidentified fungi, vgk 2 89.10, were reduced to 40–50% of the control growth, S. bovinus to 30% of the control and the most sensitive fungus, the unidentified isolate vg 1 87.10, was reduced to 3% of the growth in the control treatment. In all experiments, the shoot to root ratio generally increased, mainly as a result of increased shoot growth.  相似文献

Effects of manipulation of litter and humus layers (removal, doubling and control treatments) on the colonization potential of ectomycorrhizal fungi were studied in two secondary stands of Pinus sylvestris (5 and 18 years old) in The Netherlands. Five-mont-hold, sterile-grown Scots pine seedlings, inoculated with Laccaria bicolor, Paxillus involutus or Rhizopogon luteolus and noninoculated seedlings were used as baits. The seedlings were harvested after one growing season. For comparison, sporocarps of ectomycorrhizal fungi were also investigated. Genus composition on the seedlings was independent of initial inoculum, but determined by both treatment and age of the stands. In both stands, removal of litter and humus layers increased, and addition of organic material decreased the number of ectomycorrhizal types on the seedlings. Not all indigenous genera were observed by either outplanting seedlings or sporocarp surveys.  相似文献

The aim of this study was to investigate the potential of different isolates of ectomycorrhizal (EM) fungi to enhance the growth of Pinus sylvestris seedlings in five natural peat substrates with different nitrogen concentrations, and the effect of the Scots pine seedlings and fungal inoculum on the formation of dissolved inorganic and organic nitrogen in peat. Utilization of different organic nitrogen compounds by microbial community in the peat was also investigated using Biolog MT MicroPlates. Inoculation of the seedlings with EM fungi enhanced seedling growth. Piloderma croceum increased root growth especially, whereas Lactarius rufus increased needle growth and Suillus variegatus I, II and III improved both root and needle growth. All the EM fungi also significantly affected stem growth. Nitrogen concentration of the peat did not affect seedling growth as much as the EM fungi. At the lowest peat N concentration (1.17%) NH 4 + mineralisation was lower and DON (dissolved organic nitrogen) accumulation higher than at higher peat N concentrations. The EM fungal isolates had different effects on NH 4 + and DON accumulation/degradation in peat. The EM fungal isolates significantly increased NH 4 + formation in the peat, whereas L. rufus and P. croceum had an opposite effect on DON accumulation. S. variegatus I significantly decreased the DON concentrations during peat incubation. The N concentration of the peat slightly affected the utilization of amino acids and polyamines by the microbial community, whereas inoculation with S. variegatus I, II or III had no effect.  相似文献

Guayule seedlings were inoculated with two Glomus species in pasteurised soil and grown in the glasshouse without added fertiliser for 8 wk prior to transplanting to the field. The survival rate of transplanted guayule seedlings was increased by inoculation with vesicular arbuscular mycorrhizal fungi compared with uninoculated controls. Inoculated guayule had greater concentrations of Ca, Fe, Mg and Mn at six months of age, and greater concentrations of Ca, Mg, and Zn at 12 months of age than did uninoculated plants. The latex content of both roots and shoots of guayule was greater in inoculated than in uninoculated guayule plants at 12 and 18 months of age. The resin content remained unchanged between treatments irrespective of sampling date.  相似文献

Tap and primary lateral roots of seedlings of the putatively non-mycorrhizal Banksia ericifolia became marginally colonised when grown in an established mycelium of an arbuscular mycorrhizal (AM) fungus in the laboratory. A similar degree of colonisation was found in seedlings from an open woodland. All colonies lacked arbuscules. Two factors influencing colonisation and associated growth of host plants were examined experimentally: concentration of P in the soil and organic energy associated with the fungus. While some inoculated seedlings were slightly smaller when colonised by AM fungi, the results were inconsistent and never statistically significant. Seedlings take up insignificant quantities of soil P during early growth, even in the presence of abundant added P. Though colonisation was minor in all cases, an existing mycelium, whether or not connected to a companion plant, slightly increased the amount of root of B. ericifolia colonised by an AM fungus. All seedlings grew slowly. Shoots were significantly larger than roots, until the initiation of proteoid roots which commenced at about 40 days after germination, with both relatively high and low P supply.  相似文献

Although it is well established that ectomycorrhizas improve the mineral nutrition of forest trees, there has been little evidence that they mediate uptake of divalent cations such as Mg. We grew nonmycorrhizal seedlings and seedlings mycorrhizal with Paxillus involutus Batsch in a sand culture system with two compartments separated by a 45-μm Nylon mesh. Hyphae, but not roots, can penetrate this net. Labeling the compartment only accessible to hyphae with 25Mg showed that hyphae of the ectomycorrhizal fungus Paxillus involutus transported Mg to their host plant. No label was found in nonmycorrhizal control plants. Our data support the idea that ectomycorrhizas are important for the Mg nutrition of forest trees. Summary Seedlings of Pinus resinosa Ait. in test tubes were inoculated with the ectomycorrhizal fungus Paxillus involutus Fr. or with discs of sterile modified Melin-Norkrans (MMN) medium. Paxillus involutus was also inoculated to control tubes in the absence of Pinus resinosa seedlings. In vivo labelling of proteins in Pinus resinosa roots and in Paxillus involutus mycelium was carried out using 35S l-methionine 1, 2, 3, 4, 5 and 7 days after inoculation. Sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDSPAGE) of the protein extracts from the four treatments and autoradiography demonstrated that the presence of root exudates altered protein synthesis in Paxillus involutus as three major bands disappeared when Paxillus involutus was exposed to root exudates. Protein synthesis in Pinus resinosa was also altered when Paxillus involutus was introduced into the tubes, since at least two bands were more intense when seedlings were inoculated with Paxillus involutus, as compared to control roots. No difference was observed in the growth and the label incorporation of Paxillus involutus growing with or without root exudates. Ectomycorrhizal roots were not formed during this experiment. Gene regulation in this ectomycorrhizal association occurs, therefore, prior to the formation of ectomycorrhizal roots.  相似文献

The effects of slow and fast-release fertilizers (P, K, Mg) on the movement and availability of nutrients in acid forest soil were studied. Fast-release superphosphate, potassium chloride and magnesium sulphate and slow-release apatite (P) and biotite (K, Mg) were applied alone or together with urea or urea+limestone. The nutrient content in the organic horizon was determined one growing season and three growing seasons after the application, and in the mineral layer after one growing season. The movement of nutrient ions in the organic horizon was studied by an ion exchange resin bag method during a 5-month period following application. The fast-release salts immediately increased the soluble P and exchangeable K and Mg contents in the organic and mineral soils and in the resin bags. After three growing seasons the effect of K application in the organic layer was non-detectable and that of P had clearly diminished. Apatite gradually increased soluble P content in the organic layer, but biotite had only a minor effect on the K and Mg contents. The nutrients from the fast-release fertilizers had clearly become available and mobile in the year of application and were thus susceptible to leaching. The rate of nutrient release from apatite and biotite is slower and the added nutrients are retained in the organic horizon. Slow-release compounds, like apatite and biotite, might be potential fertilizers for counteracting acidic deposition and subsequent nutrient losses.  相似文献

Phytoextraction has been proposed in recent years as an environmentally and cost-efficient treatment technique for the remediation of heavy-metal contaminated sites. In particular, plants that are fast growing, metal accumulating, and economically interesting, such as sunflowers or trees, recently became more important in research on phytoextraction. Heavy metal uptake of trees can be strongly influenced by ectomycorrhizal fungi. We investigated the possibility of enhancing phytoextraction of Cd by willows (Salix viminalis) and poplars (Populus canadensis) in association with three well known ectomycorrhizal fungi (Hebeloma crustuliniforme, Paxillus involutus and Pisolithus tinctorius). A pot experiment was conducted using Cd polluted soil from a contaminated site. Four replicates of each combination of fungus and tree species, and controls without fungal inoculum, were set up. After a growth period of 11 weeks, yields and Cd concentrations in roots, stems, and leaves were measured. In addition, the total Cd uptake, the transfer to roots, and the translocation to stems and leaves were calculated. The association of P. canadensis with P. involutus led to a highly significant increase of Cd concentrations, in particular in the leaves, which contained 2.74 ± 0.34 mg Cd per kg dry matter. Compared to the control this is an enhancement of nearly 100%. The fungi also significantly enhanced the translocation from the roots to the leaves, leading to a concentration ratio (leaves/roots) of 0.32 ± 0.06 compared to 0.20 ± 0.02 of the control plants. Additionally, P. involutus significantly enhanced the total Cd extraction by P. canadensis. Similar effects were not observed by other fungi or in association with S. viminalis.  相似文献

Production of free and conjugated polyamines by one strain of Laccaria proxima (Boud.) Maire, three strains (H, O, K) of Paxillus involutus (Batsch) Fr., and one strain of Pisolithus tinctorius was studied in vitro. Spermidine (Spd) was the main polyamine in the 4-week-old mycelium of all the fungi. It was mainly present in the free form, but it also occurred in conjugated forms. Paxillus involutus strain H released large amounts of free putrescine (Put), and the Pisolithus tinctorius released a compound probably related to cadaverine (Cad). On the other hand, these two fungi contained less conjugated polyamines than the other fungi. In addition to the amounts, the forms (perchloric acid soluble and insoluble) of conjugated polyamines in the mycelium varied between species and strains. L. proxima contained nearly as much insoluble conjugated Spd as free Spd, whereas Paxillus involutus strains O and K contained relatively large amounts of soluble conjugated Spd. The results suggest that ectomycorrhizal fungal species and strains differ in their ability and need to produce conjugated polyamines. The small amounts of soluble conjugated polyamines found in the culture filtrates indicate that some specific conjugated polyamines may be involved in polyamine translocation across the plasma membrane.  相似文献
