The fungal species from rhizosphere and rhizoplane of perennial grasses of the Western Ghats of India were studied for their pathogenicity, antagonism in vitro, substrate and root colonization abilities, rhizosphere competence, growth in different soil pH and inoculum shelf-life. Out of 138 non-pathogenic fungal isolates tested, 85 were antagonistic in vitro to chilli anthracnose pathogen Colletotrichum capsici. Fifteen isolates with >60% inhibition zone to pathogen culture had saprophytic and root and rhizosphere colonization abilities. The sorghum grain inocula of test antagonistic fungi- Fusarium oxysporum, Chaetomium globosum and Trichoderma harzianum had the shelf-life of 90 days at 20?±?2?°C and required optimum soil pH of 6.5. The above fungal isolates when tested for biocontrol of anthracnose disease in greenhouse and field caused reduction in seedling mortality and decreased disease incidence and severity at various plant growth stages and significant reduction in chilli fruit and seed infection. The test antagonistic fungi promoted seedling and mature plant growth and increased fruit and seed yield. Populations of these antagonistic fungi were fairly high in chilli rhizosphere at harvest. The present study indicated that antagonistic fungi from grass rhizosphere and rhizoplane could be used to control anthracnose and promote plant growth, and increase yield of chilli in field.  相似文献

The role of organic acids in the mobilization of plant nutrients from the rhizosphere was assessed in seven contrasting soil types. The results indicated that malate was poor at mobilizing micronutrients from all the test soils, whilst citrate was capable of mobilizing significant quantities. Citrate was also capable of mobilizing P from one soil which possessed a large Ca-P fraction. This mobilization of P was due to both the complexing action of the citrate anion and due to the dissolution properties of the protons released from citric acid upon equilibrium with the soil solution. The reaction of citrate with cations was found to be near instantaneous with significant absorption to the solid phase in some soils at low concentrations. Soil decomposition studies indicated that citrate was rapidly broken down in organic soils but was more resistant to degradation in subsoil horizons. It was concluded that organic acids can be expected to be of little consequence in nutrient mobilization from high pH soils, whilst in acid soils they may be involved both in a more general mechanism for micronutrient uptake or as a potential Al detoxification mechanism.Abbreviations C6H8O7 Citric acid and H-citrate indicates - C6H5O7Na3 whilst Na-citrate indicates  相似文献

植物根际是指植物根系与土壤的交界面,是根系自身生命活动和代谢对土壤影响最直接、最强烈的区域,其物理、化学和生物性质不同于土体土壤。在这个区域里,与植物发生相互作用的大量微生物,被称为根际微生物。根际微生物在植物的生长发育和植物病虫害的生物防治等方面都具有十分重要的意义。本文总结了根际微生物群落构建的研究现状,介绍了根际微生物的经典和最新的研究方法,包括根箱法、同位素技术以及高通量测序、菌群定量分析、高通量分离培养等方法在根际微生物研究中的应用,讨论了植物根系分泌物(碳水化物、氨基酸、黄酮类、酚类、激素及其信号物质)和土壤物理化学性质对根际微生物群落的影响,概述了根际微生物-植物的互作机制,以及根际微生物群落对植物的促生作用、提高植物抗逆性和抑制作用,并对根际微生物群落研究中存在的问题和未来发展方向进行了展望。  相似文献

The role of proteinaceous amino acids in rhizosphere nutrient mobilization was assessed both experimentally and theoretically. The degree of adsorption onto the soil's solid phase was dependent on both the amino acid species and on soil properties. On addition of amino acids to both soil and freshly precipitated Fe(OH)3, no detectable mobilization of nutrients (K, Na, Ca, Mg, Cu, Mn, Zn, Fe, S, P, Si and Al) was observed, indicating a very low complexation ability of the acidic, neutral and basic amino acids. This was supported by results from a solution equilibria computer model which also predicted low levels of amino acid complexation with solutes present in the soil solution. On comparison with the Fe(OH)3 and equilibria data obtained for the organic acid, citrate, it was concluded that amino acids released into the rhizosphere have a limited role in the direct acquisition of nutrients by plants. The effectiveness of root exudates such as amino acids, phytosiderophores and organic acids in nutrient mobilization from the rhizosphere is discussed with reference to rhizosphere diffusion distances, microbial degradation, rate of complexation and the root's capacity to recapture exudate-metal complexes from the soil.  相似文献

Penicillium expansum, Botrytis cinerea and Colletotrichum acutatum are the most common postharvest pathogens of apples, peaches and nectarines. In this study, 96 bacteria were isolated from halophytes rhizosphere and assayed for biocontrol activity under in vitro conditions. Among the 96 isolates tested, isolate S16 effectively inhibited the growth of P. expansum, B. cinerea and C. acutatum. The isolate S16 has reduced 78.33±1.53 to 82.98±2.13% of disease severity in apples, peaches and nectarines. Matrix-assisted laser desorption ionisation-time of flight mass spectrometry of the antifungal compounds revealed three lipopeptide complexes, namely surfactins, iturins and fengycins. Lipopeptides and hydrolytic enzymes produced by the isolate S16 play an important role in the antifungal activity. Polymerase chain reaction analysis using ituD, srfAD, fenD and fenE gene-specific primers showed that the isolate S16 carry sequences similar to ituD, srfAD, fenD and fenE genes. Based on the 16S rDNA sequencing, the effective isolate S16 was identified as Bacillus subtilis.  相似文献

采用抗菌谱测定以及BOXAIR-PCR、生理生化特征和16S rDNA序列分析等方法对分离自10种作物根际的55株拮抗细菌的多样性及主要拮抗菌类群进行了分析.结果表明: 根际拮抗细菌的拮抗作用具有丰富的多样性;BOXAIR-PCR分析中供试菌在721%相似性水平上可以聚为7个群,850%相似性水平上聚为25个群;所有供试根际拮抗菌分别属于芽孢杆菌属(Bacillus)芽孢杆菌属(Paenibacillus)、短芽孢杆菌属(Brevibacillus)、假单胞菌属(Pseudomonas)和产碱菌属(Alcaligenes).作物根际拮抗菌具有丰富的遗传多样性和拮抗性能多样性.  相似文献

Abstract

Three antagonists: Pseudomonas fluorescens (Pf1), Bacillus subtilis and Trichoderma viride, were tested alone and in combination for suppression of onion leaf blight (Alternaria palandui) disease under glasshouse and field conditions. The average mean of disease reduction was 24.81% for single strains and 42.44% for mixtures. In addition to disease suppression, treatment with a mixture of antagonists promoted plant growth in terms of increased plant height and ultimately bulb yield. Though seed treatment of either single strain or strain mixtures alone could reduce the disease, subsequent application to root, leaves or soil further reduced the disease and enhanced the plant growth. The mixture consisting of Pseudomonas fluorescens Pf1 plus Bacillus subtilis plus Trichoderma viride was the most effective in reducing the disease and in promoting plant growth and bulb yield in greenhouse and field tests.  相似文献

Summary Biological control of Verticillium wilt disease with antagonistic micro-organisms was studied. Antagonism of some fungi, isolated from tomato rhizosphere, toVerticillium albo-atrum R & B. was observedin vitro. A clearly defined zone, in which the growth of the pathogen was inhibited, was observed withPenicillium spp. (includingPenicillium chrysogenum Thom) andFusarium culmorum (S.G. Sm) Sacc., whileTrichoderma viride pers. ex Fries,Gliocladium spp. andPenicillium vermiculatum Dangeard, suppressed the growth ofV. albo-atrum by penetrating, and overgrowing it. OnlyT. viride andP. vermiculatum culture filtrate added to the Dox's agar, reduced the radial growth ofV. alboatrum. Root-dip application of culture filtrates ofT. viride andP. chrysogenum was found to be most effective in controlling the disease, followed by other species ofPenicillium andGliocladium spp. WhileFusarium culmorum provided no control. Improvement of plant height and vigour with a better yield due to culture filtrate treatment occurred. Root-dip application of antagonistic fungal propagules (T. viride, P. chrysogenum) to tomato seedlings was also very effective in controlling wilt in tomato plants grown inV. albo-atrum infested soil. The mineral phosphorus supply produced two outbreaks in the bacterial population of the barley rhizosphere and rhizoplane but inhibited the growth of fungal mycelium. The inhibition of mycelial growth might be due to the exudation of specific inhibitors by barley roots, since the most pronounced inhibition was observed at high doses of supplementary phosphorus.  相似文献

The enemy release hypothesis predicts that alien populations are more vigorous and grow faster in introduced areas than within their natural range due to lack of natural enemies. An extension of this theory would be that the lack of natural enemies on an herbivore species will indirectly increase the number of host plant species that it can infest. The eucalyptus gall wasp Ophelimus maskelli is a recent introduction to the Mediterranean basin. Its arrival was followed after about two years by its parasitoid, Closterocerus chamaeleon. We evaluated the range of hosts used by the gall wasp by surveying 50 Eucalyptus species, 37 Eucalyptus hybrids and 18 E. globulus half-sibling families, before and after the establishment of the parasitoid. We found a consistent reduction in the observed host range of the gall wasp after the establishment of the parasitoid with a reduction of 83% on Eucalyptus species, 84% on hybrids, and 94% on E. globulus half-siblings. Host range narrowed down from 18 to three closely-related host species, all in the section Exsertaria, series Exsertae, E. camaldulensis, E. tereticornis and E. rudis, which are probably the standard hosts of O. maskelli. In the remaining affected species the intensity of attack decreased significantly in all studied regions. These findings show that, besides the direct gains from classical biological control, there may be indirect benefits through a natural-enemy-induced reduction in the range of host plants used by herbivorous insect pests.  相似文献

The aim of the study was to investigate the ability of Zea mays L. roots to regulate the amount of free amino acids present in the rhizosphere. The active uptake of amino acids was shown to conform to Michaelis-Menten kinetics. Comparison of amino acid-N and NO3-N kinetic parameters and soil solution concentrations showed that root uptake of free amino acids from soil may contribute significantly to a plant's N budget. The influx of amino acids also helps to minimize net C/N losses to the soil, and is therefore important in regulating the size of the rhizosphere microbial population. Experimental data and a computer simulation model of amino acid influx/efflux in a sterile solution culture, showed that roots were capable of re-sorping over 90% of the amino acids previously lost into solution as a result of passive diffusion.  相似文献

根系构型是最重要的植物形态特征之一,具有可塑性,既由遗传因素控制,又受到许多环境因子的调控。近年的大量研究表明,根际微生物能够调控植物的根系构型,进而影响植物的一系列生理与生态过程。综述丛枝菌根真菌(AMF)、根瘤菌、植物根际促生菌(PGPR)等重要根际微生物类群对植物根系构型的调控模式以及相应的调控机理,并对进一步的研究进行了展望,旨在为今后的相关研究和实际应用提供参考。  相似文献

Pollen and charcoal analysis of a 5.9-m-long sediment profile from a swamp on an alluvial terrace on the edge of La Selva biological station, Costa Rica (10°26'23" N, 84°00'24" W, 36 m asl), documents three millennia of human and natural disturbance within a lowland tropical rain forest. The record indicates that the highly diverse rain forest that presently surrounds the Cantarrana swamp regrew following forest clearing and maize agriculture that ended only a few centuries ago. The first maize pollen appears in sediments deposited ∼1070 calendar-year BP. Older sediments below the 'maize zone' contain macroscopic charcoal, abundant microscopic charcoal, and possible pollen indicators of forest clearance, perhaps signaling local cultivation of root crops that left no pollen in the sediments. Interpretation of local archaeology indicates that La Selva reserve may have been most heavily populated during the El Bosque and La Selva regional archaeological phases from 2250–950 yr BP. However, the distribution of maize pollen in the sediments is clear evidence that the reserve was also occupied during the later La Cabaña phase (950–400 yr BP), from which few artifacts have yet surfaced. Natural forest disturbance from treefalls and stream dynamics, and hydrological shifts associated with late-Holocene climate variability, form a backdrop to the human land-use history preserved in the Cantarrana sediment profile.  相似文献

祁连山高山植物根际土放线菌生物多样性   总被引:2,自引:0,他引:2  
从祁连山老虎沟不同海拔位点的15种植物根际土中培养得到78株特异表型放线菌,并结合菌体形态、生理代谢特征、抗菌活性及16S rDNA序列对其生理及系统发育多样性进行了研究。结果表明,分离菌株分属于链霉菌属(Streptomyces spp.)(73株)、诺卡氏菌属(Nocardia spp.)(4株),另有1株与GenBank中同源性最高的菌株Micromonospora saelicesensis相似性达92%,为1潜在新种。链霉菌属为主要类群,占分离菌株的93.6%,该属菌株在5个海拔位点的15种植物根际土中均有分布,但存在海拔位点、植物种类的差异性和特异性;诺卡氏菌属的菌株仅见于海拔2200 m的猪毛菜、海拔2800 m的钉柱萎陵菜和3800 m处的甘肃蚤缀根际土中;1潜在新种分离自海拔2200 m处的沙生针茅根际土。次级代谢物产生和拮抗性筛选研究结果表明:H2O2酶、脂酶2(Tween-40)、脲酶、蛋白酶、脂酶3(Tween-80)、淀粉酶、H2S、脂酶1(Tween-20)、可溶性色素及有机酸这10类次级代谢物产生菌分别占供试菌株的89.7%、82.1%、70.5%、62.8%、53.8%、52.6%、48.7%、44.9%、32.1%和17.9%,其中,淀粉酶、脂酶1、色素和有机酸仅由链霉菌产生;有29株放线菌对参试人类病原菌具有抑制作用,占供试菌株的37.2%,分布于5个海拔位点的12种植物根际土,其中,从药用植物甘肃黄芪和四裂红景天根际土中分离到的抗性菌株占拮抗性放线菌总数的60%。研究表明,高山地区植物根际土放线菌资源丰富,菌株生理功能多样,是新放线菌种和生物活性物质的重要资源库。  相似文献

Infective and effective Frankia were shown to occur in five diverse tropical forest soils of Costa Rica. Results of a plant infection assay indicated that Frankia is a common component of the soil biota in low and high elevation, primary and secondary forest soils. This is the first report of Frankia in lowland tropical rainforests of the Americas. These results suggest either a nonsymbiotic population of soil Frankia, the presence of unknown actinorhizal host species, or an ability of Frankia to be dispersed over long distances.  相似文献

Four antagonistic bacterial isolates, Bacillus subtilis, Bacillus sp., Pseudomonas corrugata 1 and P.-corrugata 2, isolated from the rhizosphere of tea plants growing in different geographical locations in India, were tested as microbial inoculants for hardening of tissue-cultured tea plants raised in the laboratory prior to the transfer to open land. Bacterial inoculations resulted in enhanced survival (up to 100, 96, and 88%), as against 50, 52, and 36% survival observed in the corresponding control plants, in rainy, winter and summer seasons, respectively. Rhizoplane and rhizosphere soil analyses showed that the major biotic factor responsible for mortality following the transfer of tissue culture raised plants to soil was fungal attack (Fusarium oxysporum). Bacterial inoculations also resulted in plant growth promotion of tissue culture as well as seed raised plants of tea.  相似文献
