【目的】旨在探究硫酸亚铁(FeSO4)对缺铁性贫血(iron deficiency anemia, IDA)小鼠肠道健康的影响。【方法】选取45只24日龄体重(16.0±1.2) g的雄性昆明小鼠,随机分成3组,每组15只,即对照组(正常饲料,饮蒸馏水)、IDA组(低铁饲料造模2周,饮去离子水)、IDA-Fe2+组(造模结束后灌胃FeSO4 3周,饮去离子水),实验结束时采样。【结果】试验结束时,与IDA组相比,IDA-Fe2+组小鼠的红细胞、血红蛋白、红细胞压积、平均红细胞体积、平均红细胞血红蛋白量、平均红细胞血红蛋白浓度等贫血指标均恢复至正常水平(P>0.05),表明FeSO4具有改善IDA的功能;与对照组相比,IDA和IDA-Fe2+组小鼠的氧化应激指标和肿瘤坏死因子α均显著升高(P<0.01)、紧密连接蛋白Occludin显著降低(P<0.001),IDA-Fe2+组与IDA组除结肠总抗氧化能力(TAC)外均无差异显著性(P>0.05)。结肠组织...  相似文献

OBJECTIVE: To determine the effectiveness of physician probability estimates calculated on the basis of findings from history-taking and physical examination in the diagnosis of iron deficiency anemia in elderly patients. DESIGN: Prospective study. SETTING: Two community hospitals offering secondary and tertiary care. PATIENTS: A total of 259 patients over 65 years of age found to have previously undiagnosed anemia. MEASURES: Physician estimates of the likelihood of iron deficiency before (pretest probability) and after (post-test probability) the laboratory test results were available. The hemogram was available to the physicians when they made their pretest probability estimates. Because the serum ferritin level proved to be the most powerful of the laboratory test results studied, the likelihood ratios associated with the post-test estimates were compared with the ratios associated with the serum ferritin level. MAIN RESULTS: The post-test probability estimates were influenced by the serum ferritin level and the pretest estimates. The post-test estimates derived from the findings obtained through history-taking and physical examination and the laboratory test results (including the serum ferritin level) were slightly less accurate in predicting iron deficiency than the serum ferritin level alone. Nevertheless, a model in which the pretest estimates were used in addition to the serum ferritin level to predict iron deficiency proved to be more powerful than the serum ferritin level alone (p = 0.006). This indicated that the limitations of the post-test estimates were due to a misinterpretation of the serum ferritin level and that the findings from history-taking and physical examination added important diagnostic information. CONCLUSIONS: Physicians must be aware of test properties to provide optimal care to their patients. If test results are properly interpreted, pretest probabilities derived from findings obtained through history-taking and physical examination can add useful information that will lead to more accurate diagnoses.  相似文献

采用雌性Sprague-Dwley(SD)大鼠构建缺铁性贫血模型。将24只雌性SD大鼠随机均分为两个大组,实验组饲以低铁饲料和去离子水,对照组饲以基础饲料和自来水。第30天处死所有SD大鼠,取血测定血红蛋白含量、红细胞压积;每只取2g肝组织匀浆离心,测定肝脏中铁的含量。结果表明,本研究成功构建SD大鼠缺铁性贫血模型,为营养学、内分泌学和毒理学研究提供新的动物模型。  相似文献

The effects of iron deficiency and iron reconstitution on the ultrastructure of the unicellular cyanobacterium Anacystis nidulans R2 were studied by electron microscopy. Low-iron cells, grown with different amounts of aeration, were analyzed at 6, 12, and 24 h after the addition of iron. Low-iron cells had a decrease in the quantities of membranes, phycobilisomes, and carboxysomes and a large increase in glycogen storage granules. In cells aerated with gentle shaking, the addition of iron caused the number of carboxysomes to increase rapidly within 6 h. This was paralleled by a decrease in the quantity of glycogen storage granules. Carboxysomes were associated with the nucleoplasmic face of the inner photosynthetic membrane in normal, but not low-iron, cells; they once more contacted the membrane by 6 h after iron addition. Phycobilisome assembly was apparent by 6 h, and the number of phycobilisomes increased throughout reconstitution. Membrane restoration was accomplished in two stages: (i) components were added to preexisting membranes until about 12 h, and (ii) new membranes were synthesized beginning at 12 to 18 h. Low-iron cells grown by bubbling with air had only one to two concentric layers of membrane per cell. The addition of iron led to a pattern of reconstitution that was similar to that described above with two important exceptions. Under these conditions, the number of carboxysomes remained low and the carboxysomes rarely contacted the photosynthetic membranes. New membranes were not synthesized until the culture had reached the late-logarithmic growth phase and after all other morphological features had returned to normal.  相似文献

The reversibility of the alterations in norepinephrine (NE) content and turnover in interscapular brown adipose tissue and heart of iron-deficient rats has not been demonstrated. We therefore examined NE metabolism in age-matched male Sprague-Dawley rats depleted of iron by dietary means and after repletion with iron dextran. Heart NE content was 58, 61, and 85% of controls at 0, 3, and 7 days after repletion, whereas interscapular brown adipose tissue-NE content was 87, 103, and 104% of controls. Fractional heart NE turnover was 225% greater in iron-deficient anemics than controls but normalized within 3 days. Interscapular brown adipose tissue NE turnover was 58%, 46%, and 20% above controls in iron-deficient rats after 0, 3, or 7 days of iron repletion. Hematocrit returned to 80% of normal in 7 days. Liver triiodothyronine production also increased to 80% of control in this period. A second experiment used isovolemic exchange transfusion to examine the influence of anemia per se on these alterations in organ NE turnover. Acute correction of anemia in iron deficiency did not alter brown fat NE turnover. Heart NE turnover was significantly lower in anemic animals regardless of iron status. Defects in heart and brown fat NE metabolism are reversible within 7 days of iron treatment as are alterations in triiodothyronine production. Anemia per se has little effect on brown fat NE metabolism but does dramatically decrease heart NE content.  相似文献

Heme synthesis from [2-14C]glycine was studied in liver and red blood cells. In normal rats liver contained two early [14C] heme peaks maximal at 1 and 4.5 h, followed by a long plateau of heme labeling. These phases were present in both microsomes and mitochondria. Cycloheximide suppressed formation of the first but not the second heme component. All phases of hepatic heme labeling were reduced in iron-deficient rats, with better preservation ofthe microsomal fraction. In iron-deficient rats responding to iron therapy, the first peak merged with an enlarged and premature second component; the increase was most marked in mitochondria. Thus, labeled heme metabolism was less perturbed in microsomes than mitochondria in both of these conditions. Peripheral blood also contained a [14C] heme peak at 1 h in all experimental groups. This was highest with the increased eythroid response observed in iron-treated rats. The first heme peak, present in both hepatic and erythroid cells, may represent a pool of free or unassigned heme. The later heme component may reflect formation of hemoproteins, which could be related directly or in directly to the initial, rapid turnover heme component.  相似文献

Diagnostic information on adolescents may be elicited from both youths and their parents, especially for depressive and suicidal symptomatology. The objective of this study was to examine the degree of agreement between parent and adolescent reports of major psychiatric disorders, at the diagnostic and at the symptom level, in a severely affected inpatient clinical population. 64 parent–adolescent pairs were interviewed separately with the semi-structured diagnostic interview Kiddie-SADS-PL. Symptomatology was also assessed with 11 self-report and parent-report scales, all translated, adapted and in most cases validated in Iceland. A total of 25 subscales were included to assess emotional dimensions such as depression or anxiety and cognitive dimensions such as attention deficit or self-concept. Good agreement was found for social phobia and fair agreement for generalized anxiety disorder. Although parent–youth agreement was poor in most cases at the symptoms level, significant correlations indicated consistency for most severity scores, except those related to depressive symptomatology, attention deficit, separation anxiety or conduct disorder. The low agreement between reports of suicidal ideation is in line with results from previous studies and suggests that parents might under- or over-estimate this symptomatology. The combination of data obtained with diagnostic interviews and rating-scales confirmed results from prior empirical work, giving greater weight to parents’ reports of observable behavior and to adolescents´ reports of subjective experiences, especially depressive symptomatology. Our findings suggest that both parent and child informants are necessary to obtain adequate assessments in adolescents. Further research should explore the correspondence between discrepant diagnoses and external criteria such as parental psychopathology or parent–child relationships and attachment. Psychoanalysis could benefit from cognitive neuroscience and use cognitive assessments as interesting tools. Thus, cognitive assessments can show discrepant results according to parents’ or adolescents’ reports and can therefore shed light on the parent–child interaction and relational dynamics. Inversely, cognitive neuroscience could benefit from psychoanalysis by taking into account, when interpretating the scores, the relational dynamics and the personal history of the rater.  相似文献

Iron chlorosis is very common on alkaline soils such as calcareous ones, since iron availability is limited by high pH. Under these conditions of iron deficiency, graminaceous plant species induce special mechanisms for iron acquisition, involving enhanced release of iron chelators called phytosiderophores. On the other hand, it is known that most of salt soils have alkaline pH. So, plants growing on this kind of soils are often subjected simultaneously to salinity and iron deficiency. This work aimed at (i) studying the physiological responses of barley (Hordeum vulgare L.) to iron deficiency, and (ii) evaluating the effect of salt on the iron nutrition and the phytosiderophore release. For this purpose, seedlings of Hordeum vulgare L. were cultivated under controlled conditions, either in a complete nutrient solution with or without NaCl, or in an iron free nutrient solution containing or not NaCl. The plant morphological aspect, chlorophyll content of young leaves, iron status, biomass production, and phytosiderophore release by roots were assessed. Plants subjected to Fe deficiency exhibited a severe chlorosis, accompanied by a significant biomass reduction. These plants developed more lateral roots than the control with a highly stimulated phytosiderophore release. However, the latter was greatly diminished when iron deficiency was associated to salinity. A depressive effect of salt on iron acquisition in plants subjected only to salt stress which was also observed and further confirmed by the important decrease of efficiency in iron acquisition. These results suggest that salinity may reduce capacity of plants to acquire iron from alkaline soils by inhibiting phytosiderophore release.  相似文献

Phosphate (Pi) is an essential element for plant development and metabolism. Due to its low availability and mobility in soils, it is often a limiting nutrient for their growth. This phenomenon is reinforced by the formation of insoluble complexes in the environment with many cations, affecting the solubility of both phosphate and associated ions. This interaction is investigated here for iron, a strong phosphate chelator. Depleting the medium in phosphate clearly resulted in an increase of iron content in Arabidopsis. These modifications triggered molecular responses linked with iron status (transport, homeostasis and accumulation). Interestingly, physiological modifications affecting iron storage were also observed. The accumulation of phosphate/iron complexes in the vacuoles of plants grown in Pi-rich medium disappeared in Pi-depleted medium in favor of accumulation of iron inside the chloroplasts, likely associated with ferritin.  相似文献

Adult rats raised on 90% iron deficient diet for 6 weeks showed a decrease in the levels of brain phosphatidylinositol (60%), phosphatidylinositol 4-phosphate (25%) and phosphatidylinositol 4,5-bisphosphate (30%), the precursors for the second messengers, diacylglycerol and inositol phosphates. Rehabilitation to normal diet for 24 days was found to restore the levels to control values. The data suggests that iron deficiency could lead to alterations in the functions related to phosphoinositide-linked receptor system(s).  相似文献

The survival probabilities of newly-formed colonies of organisms arising from the branching process formulation of individual reproduction are examined. Six types of 2-parameter discrete offspring distributions common to mathematical ecology are compared with respect to survival of newly formed colonies of offspring. It is found that the survival value of the distribution can be rank ordered in the following descending order: modified Poisson (highest) Neyman A, geometric Poisson, Pólya-Aeppli, negative binomial and the modified geometric. Causal factors for these differences and practical implications of these results are discussed.  相似文献

This study was designed to measure the effect of iron supplementation on antioxidant status in iron-deficient anemia, including the time for hemoglobin normalization and at the time of filling of iron body stores. The extent of plasma lipid peroxidation was evaluated by measuring the levels of malondialdehyde and glutathione peroxidase (GSH-Px), and the activities of superoxide dismutase (SOD) and catalase in 63 patients with iron-deficiency anemia before and after 6 wk of iron supplementation and at the time when body iron stores are saturated. After 6 wk of iron supplementation, a significant decrease of oxidative stress was observed in the treated subjects relative to controls (p<0.05). No significant differences existed between treated patients at 6 wk and at the end of the study. The erythrocyte levels of catalase, SOD, and GSH-Px were significantly lower in treated patients relative to controls (p<0.05). These levels increased after 6 wk of supplementation (p<0.05) and showed no significant differences with those at the end of the study.  相似文献

Effect of dietary iron deficiency on mineral levels in tissues of rats   总被引:3,自引:0,他引:3  
To clarify the influence of iron deficiency on mineral status, the following two synthetic diets were fed to male Wistar rats: a control diet containing 128 micrograms iron/g, and an iron-deficient diet containing 5.9 micrograms iron/g. The rats fed the iron-deficient diet showed pale red conjunctiva and less reactiveness than the rats fed the control diet. The hemoglobin concentration and hematocrit of the rats fed the iron-deficient diet were markedly less than the rats fed the control diet. The changes of mineral concentrations observed in tissues of the rats fed the iron-deficient diet, as compared with the rats fed the control diet, are summarized as follows: . Iron concentrations in blood, brain, lung, heart, liver, spleen, kidney, testis, femoral muscle, and tibia decreased; . Calcium concentrations in blood and liver increased; calcium concentration in lung decreased; . Magnesium concentration in blood increased; . Copper concentrations in blood, liver, spleen and tibia increased; copper concentration in femoral muscle decreased; . Zinc concentration in blood decreased; . Manganese concentrations in brain, heart, kidney, testis, femoral muscle and tibia increased. These results suggest that iron deficiency affects mineral status (iron, calcium, magnesium, copper, zinc, and manganese) in rats.  相似文献

Cultures of the nitrogen-fixing cyanobacterium Anabaena variabilis PCC 6309 were grown under several iron concentrations, and changes in growth rates and chlorophyll and phycocyanin concentrations were determined. The total amount of ferredoxin present in the cells was found to be dependent on the concentration of iron in the media, as was the pattern distribution of the two different forms of the iron-sulfur protein described in this organism. Flavodoxin was not found to be present in these cells even when they were grown in the absence of added iron, indicating that this flavoprotein does not replace ferredoxin in this particular strain.  相似文献
