This is the first report on population variability of nonacosan-10-ol and n-alkanes in needle epicuticular waxes of Macedonian pine (Pinus peuce GRISEB.) Hexane extracts of needle samples, originating from two natural populations in Montenegro (Zeletin and Sjekirica) and from one population in Serbia (Mokra Gora) were analyzed by gas chromatography (GC) and gas chromatography/mass spectrometry (GC/MS). The amount of nonacosan-10-ol varied individually from 41.3 to 72.31% (average 55.9%), with the Sjekirica population being statistically divergent (64.4% on average). The results showed n-alkanes in epicuticular waxes ranging from C?? to C??. The most abundant alkanes were C??, C??, C??, and C?? (15.5, 11.1, 10.6, and 10.5% on average, resp.). The carbon preference index of Pinus peuce ranged from 1.0 to 4.3 (1.9 on average). Average chain length ranged from 18.4 to 27.7 (average 25.7). A high level of inidividual quantitative variation in all of these hydrocarbon parameters was also detected. These results were compared with published data on other species from the Pinus genus.  相似文献

An n-hexane extract of fresh, mature leaves of Ludwigia adscendens, containing a thin layer of epicuticular waxes, has been analysed for the first time by TLC, IR and GC using standard hydrocarbons. The leaves contained 22 identified long chain (C15-C36) n-alkanes, accounting for 74.27% of the hydrocarbons present, and an unknown number of unidentified branched chain alkanes. The predominant n-alkane was C25 (11.02%), whilst C18 (7.62%), C20 (6.14%), C29 (5.36%) and C27 (5.29%) n-alkanes were moderately abundant: the C35 homologue was present only in minor amounts (0.22%).  相似文献

The seasonal development of adaxial Prunus laurocerasus leaf surfaces was studied using newly developed methods for the mechanical removal of epicuticular waxes. During epidermal cell expansion, more than 50 microg leaf(-1) of alkyl acetates accumulated within 10 d, forming an epicuticular wax film approximately 30 nm thick. Then, alcohols dominated for 18 d of leaf development, before alkanes accumulated in an epicuticular wax film with steadily increasing thickness (approximately 60 nm after 60 d), accompanied by small amounts of fatty acids, aldehydes, and alkyl esters. In contrast, the intracuticular waxes stayed fairly constant during development, being dominated by triterpenoids that could not be detected in the epicuticular waxes. The accumulation rates of all cuticular components are indicative for spontaneous segregation of intra- and epicuticular fractions during diffusional transport within the cuticle. This is the first report quantifying the loss of individual compound classes (acetates and alcohols) from the epicuticular wax mixture. Experiments with isolated epicuticular films showed that neither chemical conversion within the epicuticular film nor erosion/evaporation of wax constituents could account for this effect. Instead, transport of epicuticular compounds back into the tissue seems likely. Possible ecological and physiological functions of the coordinate changes in the composition of the plant surface layers are discussed.  相似文献

For the first time, the n‐alkane distribution and variability of the epicuticular waxes within 22 Sedum taxa was reported with focus on the chemotaxonomy of native Sedum representatives from the central Balkan Peninsula, compared to their relations with four other species of the Crassulaceae family. By GC/MS and GC‐FID identification and quantification, it was established that n‐alkanes C27, C29, C31, C33, and C35 were the dominant constituents of the examined epicuticular wax samples. Applying multivariate statistical analyses including agglomerative hierarchical clustering (AHC) and principal component analysis (PCA), the relation according to the n‐alkane composition between the examined samples was established. It was shown that the n‐alkane variability of the central Balkan Sedum species was considerable and that n‐alkanes might not be very reliable taxonomic markers for these species.  相似文献

The influence of leaf epicuticular waxes of nine woody species from cerrado (a savanna ecosystem of Brazil) on the foraging behavior of the leaf-cutting antAtta sexdens rubropilosa Forel, 1908 (Formicidae, Attini) was investigated. Some waxes showed neither inhibitory nor attractive effect over a series of tests, while a second group caused a delayed rejection. Waxes of two species were shown to be strongly deterrent to the ants. Since different waxes caused different responses, it is concluded that their chemical composition, rather than hydrophobicity or mechanical constraints, accounts for the observed effects. Plant species with leaves rejected by the ants did not necessarily have repellent waxes. On the other hand, no species with acceptable leaves yielded waxes with deterrent effects.  相似文献

Cerrado is a savanna ecosystem of central and southeastern Brazil. Many woody species of cerrado have thick layers of cork. The present work aimed to characterize, by GC/MS analysis, the constituents of n-hexane extracts from the cork of common species from cerrado. Cork samples from 31 individuals, corresponding to 14 species and six families, were analyzed. Similarities and differences were noticed between cork and cuticular waxes regarding profiles of lipophilic constituents. The distribution of cork constituents was analyzed using the UPGMA clustering method and DICE coefficient. All clusters in the dendrogram obtained comprise individuals from a same species, suggesting that the distribution of lipophilic cork constituents is useful for species characterization and possibly also for species identification, resembling results commonly obtained with molecular markers. Seven samples of Bignoniaceae, corresponding to two genera and seven species, emerged in a common cluster, in an arrangement in accordance with the recent segregation of Tabebuia species to a new genus Handroanthus. The markers analyzed were not efficient regarding characterization of other families.  相似文献

The leaf cuticle is covered by epicuticular wax consisting mainly of straight-chain aliphatic hydrocarbons with a variety of substituted groups. Studies have been concentrated on n-alkanes in epicuticular wax of Winged bean [Psophocarpus tetragonolobus (Stickm.) DC.]. Hydrocarbon constituents especially n-alkane analyses of seven cultivars of Winged bean [Psophocarpus tetragonolobus (Stickm.) DC.] have been undertaken. All the n-alkanes in between C14–C18 and C20–C38 are present in each of the species. Among the species, amount of n-alkanes is maximum in IC112417 and relatively low in EC38825. Scanning electron microscopic views were also taken for epicuticular layers and their hydrocarbons of the leaves of all the genotype species of the plant. Qualitative and quantitative characterization of n-alkanes present in the epicuticular wax extracted from the mature leaves can be used as an effective tool in chemo taxonomical work and also for the study of genotypic variation of the different cultivars.  相似文献

Foliar epicuticular waxes of specimens of 13 Croton species native in Brazil were extracted. The fractions containing alkanes and primary alcohols were isolated by preparative thin layer chromatography. Derivatized n-primary alcohols were identified by gas chromatography (GC) coupled with mass spectrometry and n-alkanes by GC and comparison with known standards. Relative abundances were estimated by GC coupled with flame ionization detector. The distribution of constituents of both classes was analyzed by cluster analysis, using the UPGMA method and Euclidean distances. The chemical affinities among species were compared with published data of molecular phylogenetic relationships. The distribution of n-alkanes and primary alcohols were shown to be useful markers of Croton species. Primary alcohols were more consistent than n-alkanes for species fingerprinting.  相似文献

本试验选用6个抗病性不同的甘蓝型油莱品种,研究其叶表皮蜡质组成及结构与菌核病抗性的关系。结果表明,抗病品种在去除叶表皮蜡质后病情指数显著增加;感病品种无显著变化。不同抗性品种(系)间除酯类组分含量无显著差异外,其余蜡质组分含量差异显著。相关分析表明,蜡质组分中酯类含量与病情指数呈显著负相关关系,醇类、酮类含量与病情指数呈显著正相关,其余组分和蜡质总量与病情指数无显著相关关系。抗性品种叶表皮蜡质中烷类及酯类所占比重较高,而易感品种酮类比重较高。扫描电镜结果显示,抗病品种(系)的蜡质晶体主要为颗粒状、杆状、丝状;而感病品种(系)的蜡质晶体中不规则片状晶体所占比例较大。这些结果说明油菜叶表皮蜡质的组分及结构可能是抗病品种抵抗和延迟病原菌侵入的机制之一。  相似文献

Whether the rumen microbes are able to synthesize and/or degrade long-chain alkanes in anaerobic conditions remains a question to be answered before these hydrocarbons can be confidently used as duodenal flow or rumen transit markers. In this context, an experiment in vitro was carried out to establish whether within a rumen liquor fermentation system, n-alkanes can be derived from de-waxed structures of the plant or from non-alkane wax components (long-chain fatty alcohols, long-chain fatty acids and esters), or may be metabolized by bacteria to other components or to shorter-chain hydrocarbons. Ryegrass was labelled with 14C in growth chambers under controlled conditions in order to use it as a substrate. The labelled material obtained was separated in three fractions: labelled alkanes, labelled de-waxed plant and labelled wax components without the alkanes. These fractions were used for three different incubations in vitro, which objectives were as follows: 1. To check whether rumen bacteria can synthesize alkanes from carbon structures other than waxes (e.g. sugars). 2. To verify whether rumen bacteria can metabolize the n-alkanes to other compounds. 3. To check whether rumen bacteria can synthesize n-alkanes from other carbon compounds from waxes. The results showed that there was neither bacterial synthesis nor metabolism of the n-alkanes in in vitro conditions.  相似文献

Aim  To determine if the distributions of lizard species from Seasonally Dry Tropical Forest (SDTF) enclaves within the Cerrado biome in central Brazil are associated with the Tropical Seasonal Forests Region, a recently proposed phytogeographic unit of South America, corroborating the existence of a Pleistocenic Arc of SDTFs.
Location  SDTF remnants in the Paranã River valley, municipality of São Domingos, Goiás, Brazil.
Methods  Lizards were extensively sampled using haphazard sampling, funnel traps, and pitfall traps with drift fences during four expeditions. The composition of the SDTF lizard assemblage was compared with those from other South American phytogeographic regions (Caatinga, Cerrado, Chaco, Llanos, and the dry forests of Colombia and Bolivia), based on the literature and our own unpublished data.
Results  The SDTF lizard assemblage contained 20 species, including 11 species with extensive distributions among the regions considered, seven species shared exclusively with Cerrado localities, a single species shared exclusively with other SDTFs, and one endemic species. The presence of Lygodactylus klugei (Smith, Martin & Swain, 1977), presumably endemic to the Pleistocenic Arc formed by the Tropical Seasonal Forests Region, considerably extends the known distribution of this species, suggesting historical connections between Caatinga and Cerrado SDTF enclaves.
Main conclusions  The composition of the lizard assemblage in Cerrado SDTF enclaves seems to corroborate the recent proposal that the SDTF should be recognized as a phytogeographic unit (or dominium). The presence of disjunct populations and endemic species highlights the urgency of considering the uniqueness of the Paranã River valley SDTFs and the importance of its conservation.  相似文献

Cuticular waxes play a pivotal role in limiting transpirational water loss across the plant surface. The correlation between the chemical composition of the cuticular waxes and their function as a transpiration barrier is still unclear. In the present study, intact tomato fruits (Lycopersicon esculentum) are used, due to their astomatous surface, as a novel integrative approach to investigate this composition- function relationship: wax amounts and compositions of tomato were manipulated before measuring unbiased cuticular transpiration. First, successive mechanical and extractive wax-removal steps allowed the selective modification of epi- and intracuticular wax layers. The epicuticular film consisted exclusively of very-long-chain aliphatics, while the intracuticular compartment contained large quantities of pentacyclic triterpenoids as well. Second, applying reverse genetic techniques, a loss-of-function mutation with a transposon insertion in a very-long-chain fatty acid elongase beta-ketoacyl-CoA synthase was isolated and characterized. Mutant leaf and fruit waxes were deficient in n-alkanes and aldehydes with chain lengths beyond C30, while shorter chains and branched hydrocarbons were not affected. The mutant fruit wax also showed a significant increase in intracuticular triterpenoids. Removal of the epicuticular wax layer, accounting for one-third of the total wax coverage on wild-type fruits, had only moderate effects on transpiration. By contrast, reduction of the intracuticular aliphatics in the mutant to approximately 50% caused a 4-fold increase in permeability. Hence, the main portion of the transpiration barrier is located in the intracuticular wax layer, largely determined by the aliphatic constituents, but modified by the presence of triterpenoids, whereas epicuticular aliphatics play a minor role.  相似文献

Needles of Taxus baccata L. were covered with tubular epicuticular wax crystals varying in diameters (100 and 250 nm) and lengths (300-500 and 500-1000 nm) on the abaxial and adaxial surfaces, respectively. Various sampling protocols were employed to study the chemical composition of the needle waxes on three different levels of spatial resolution. First, a dipping extraction of whole needles yielded the total cuticular wax mixture consisting of very long chain fatty acids (21%), alkanediols (19%), phenyl esters (15%), and secondary alcohols (9%) together with small amounts of aldehydes, primary alcohols, alkanes, alkyl esters, and tocopherols. Second, waxes from both sides of the needle were sampled separately by brushing with CHCl3-soaked fabric glass. Both sides showed very similar qualitative composition, but differed drastically in quantitative aspects, with nonacosan-10-ol (18%) and alkanediols (33%) dominating the abaxial and adaxial waxes, respectively. Third, the epi- and intracuticular wax layers were selectively sampled by a combination of mechanical wax removal and brushing extraction. This provided direct evidence that the tubular wax crystals contained high percentages of nonacosane-4,10-diol and nonacosane-5,10-diol on the abaxial surface, and nonacosan-10-ol on the adaxial surface of the needles. Together with these compounds, relatively large amounts of fatty acids and smaller percentages of aldehydes, primary alcohols, alkyl esters, and alkanes co-crystallized in the epicuticular layer. In comparison, the intracuticular wax consisted of higher portions of cyclic constituents and aliphatics with relatively high polarity. The formation of the tubular crystals is discussed as a spontaneous physico-chemical process, involving the establishment of gradients between the epi- and intracuticular wax layers and local phase separation.  相似文献

以野生型拟南芥与蜡质突变体cer1、cer4为试验材料,通过研究核盘菌胁迫对拟南芥茎表皮蜡质结构及组分含量的影响,揭示核盘菌侵染与表皮蜡质的关系。扫描电镜结果显示,野生型拟南芥蜡质晶体以垂直于表面的杆状、块状结构为主;突变体cer1晶体类型以水平的松针状、块状结构为主;突变体cer4蜡质晶体以垂直片层结构为主。核盘菌胁迫下,拟南芥蜡质晶体结构及分布形态发生变化。蜡质层结构在核盘菌胁迫下表现为:杆状、松针状蜡质晶体减少—蜡质晶体熔融—表皮"囊状凸起"—表皮膜层破裂。这些结构变化有利于病菌突破角质层屏障而侵入到植株体内。色质谱分析结果显示:与野生型相比,cer1突变体烷、次级醇、酮类显著减少;cer4突变体表现为一级醇含量减少。接种核盘菌后,野生型拟南芥与蜡质突变体一级醇类显著增加(cer1增加不显著);烷类、次级醇类、酮类含量与蜡质总量均显著减少,表明蜡质前体物质在受到核盘菌胁迫后更多地通过酰基还原途径生成一级醇,从而减少了由脱羰基途径所生成的蜡质组分。核盘菌通过改变表皮蜡质晶体结构与化学组分分泌量来促进侵染。  相似文献

Two main hypotheses have been proposed to explain the diversification of the Caatinga biota. The riverine barrier hypothesis (RBH) claims that the São Francisco River (SFR) is a major biogeographic barrier to gene flow. The Pleistocene climatic fluctuation hypothesis (PCH) states that gene flow, geographic genetic structure and demographic signatures on endemic Caatinga taxa were influenced by Quaternary climate fluctuation cycles. Herein, we analyse genetic diversity and structure, phylogeographic history, and diversification of a widespread Caatinga lizard (Cnemidophorus ocellifer) based on large geographical sampling for multiple loci to test the predictions derived from the RBH and PCH. We inferred two well‐delimited lineages (Northeast and Southwest) that have diverged along the Cerrado–Caatinga border during the Mid‐Late Miocene (6–14 Ma) despite the presence of gene flow. We reject both major hypotheses proposed to explain diversification in the Caatinga. Surprisingly, our results revealed a striking complex diversification pattern where the Northeast lineage originated as a founder effect from a few individuals located along the edge of the Southwest lineage that eventually expanded throughout the Caatinga. The Southwest lineage is more diverse, older and associated with the Cerrado–Caatinga boundaries. Finally, we suggest that C. ocellifer from the Caatinga is composed of two distinct species. Our data support speciation in the presence of gene flow and highlight the role of environmental gradients in the diversification process.  相似文献

The tree species composition of seasonally dry tropical forests (SDTF) in north-eastern and central Brazil is analyzed to address the following hypotheses: (1) variations in species composition are related to both environment (climate and substrate) and spatial proximity; (2) SDTF floristic units may be recognized based on peculiar composition and environment; and (3) the Arboreal Caatinga, a deciduous forest occurring along the hinterland borders of the Caatinga Domain, is one of these units and its flora is more strongly related to the caatinga vegetation than to outlying forests. The study region is framed by the Brazilian coastline, 50th meridian west and 21st parallel south, including the Caatinga Domain and extensions into the Atlantic Forest and Cerrado Domains. Multivariate and geostatistic analyses were performed on a database containing 16,226 occurrence records of 1332 tree species in 187 georeferenced SDTF areas and respective environmental variables. Tree species composition varied significantly with both environmental variables and spatial proximity. Eight SDTF floristic units were recognized in the region, including the Arboreal Caatinga. In terms of species composition, its tree flora showed a stronger link with that of the Cerrado Dry Forest Enclaves. On the other hand, in terms of species frequency across sample areas, the links were stronger with two other units: Rock Outcrops Caatinga and Agreste and Brejo Dry Forests. There is a role for niche-based control of tree species composition across the SDTFs of the region determined primarily by the availability of ground water across time and secondarily by the amount of soil mineral nutrients. Spatial proximity also contributes significantly to the floristic cohesion of SDTF units suggesting a highly dispersal-limited tree flora. These units should be given the status of eco-regions to help driving the conservation policy regarding the protection of their biodiversity.  相似文献

This is the first report of population variability of the contents of n‐alkanes and nonacosan‐10‐ol in the needle epicuticular waxes of Serbian spruce (Picea omorika). The hexane extracts of needle samples originated from three natural populations in Serbia (Vranjak, Zmajeva?ki potok, and Mile?evka Canyon) were investigated by GC and GC/MS analyses. The amount of nonacosan‐10‐ol varied individually from 50.05 to 74.42% (65.74% in average), but the differences between the three investigated populations were not statistically confirmed. The results exhibited variability of the composition of n‐alkanes in the epicuticular waxes with their size ranging from C18 to C35. The most abundant n‐alkanes were C29, C31, and C27 (35.22, 13.77, and 12.28% in average, resp.). The carbon preference index of all the n‐alkanes (CPItotal) of the P. omorika populations (average of populations IIII) ranged from 3.3 to 11.5 (mean of 5.9), while the average chain length (ACL) ranged from 26.6 to 29.2. The principal component and cluster analyses of the contents of nine n‐alkanes showed the greatest difference for the population growing in the Mile?evka Canyon. The obtained results were compared with previous literature data given for other Picea species, and this comparison was briefly discussed.  相似文献

The mosses Andreaea rupestris, Pogonatum aloides and P. urnigerum contain surface waxes in amounts of 0.05–0.12% dry wt. The waxes consisted of esters (C38-C54), primary alcohols (C20-C32), free fatty acids (C16-C30), and alkanes (C21-C31). Additionally, aldehydes (C22-C30) were major constituents in the wax of P. urnigerum. The classes and their chain length distributions in the surface waxes of these mosses are comparable to those of epicuticular waxes of higher plants.  相似文献
