Evolution of linkage disequilibrium of the founders in exponentially growing populations was studied using a time-inhomogeneous It? process model. The model is an extension of the diffusion approximation of the Wright-Fisher model. As a measure of linkage disequilibrium, the squared standard linkage deviation, which is defined by a ratio of the moments, was considered. A system of ordinary differential equations that these moments obey was obtained. This system can be solved numerically. By simulations, it was shown that the squared standard linkage deviation gives a good approximation of the expectation of the squared correlation coefficient of gamete frequencies. In addition, a perturbative solution was obtained when the growth rate is not large. By using the perturbation, an asymptotic formula for the squared standard linkage deviation after a large number of generations was obtained. According to the formula, the squared standard linkage deviation tends to be 1/(4Nc), where N is the current size of the population and c is the recombination fraction between two loci. It is dependent on neither the initial effective size, the growth rate, nor the mutation rate. In exponentially growing populations, linkage disequilibrium will be asymptotically the same as that in a constant size population, the effective size of which is the current effective size.  相似文献

Epidemiological studies related to radiofrequency (RF) electromagnetic fields (EMF) have mainly used crude proxies for exposure, such as job titles, distance to, or use of different equipment emitting RF EMF. The Royal Norwegian Navy (RNoN) has measured RF field emitted from high‐frequency antennas and radars on several spots where the crew would most likely be located aboard fast patrol boats (FPB). These boats are small, with short distance between the crew and the equipment emitting RF field. We have described the measured RF exposure aboard FPB and suggested different methods for calculations of total exposure and annual dose. Linear and spatial average in addition to percentage of ICNIRP and squared deviation of ICNIRP has been used. The methods will form the basis of a job exposure matrix where relative differences in exposure between groups of crew members can be used in further epidemiological studies of reproductive health. We introduce a generalization of the well-known ARCH process, widely used for generating uncorrelated stochastic time series with long-term non-Gaussian distributions and long-lasting correlations in the (instantaneous) standard deviation exhibiting a clustering profile. Specifically, inspired by the fact that in a variety of systems impacting events are hardly forgot, we split the process into two different regimes: a first one for regular periods where the average volatility of the fluctuations within a certain period of time is below a certain threshold, , and another one when the local standard deviation outnumbers . In the former situation we use standard rules for heteroscedastic processes whereas in the latter case the system starts recalling past values that surpassed the threshold. Our results show that for appropriate parameter values the model is able to provide fat tailed probability density functions and strong persistence of the instantaneous variance characterized by large values of the Hurst exponent (H>0.8), which are ubiquitous features in complex systems.  相似文献

A beam formed radiofrequency (RF) exposure-incubator employing a horn antenna, a dielectric lens, and a culture case in an anechoic chamber is developed for large scale in vitro studies. The combination of an open type RF exposure source and a culture case through which RF is transmitted realizes a uniform electric field (+/-1.5 dB) in a 300 x 300 mm area that accommodates 49 35 mm diameter culture dishes. This large culture dish area enables simultaneous RF exposure of a large number of cells or various cell lines. The RF exposure source operates at 2142.5 MHz corresponding to the middle frequency of the downlink band of the International Mobile Telecommunication 2000 (IMT-2000) cellular system. The dielectric lens, which has a gain of 7 dB, focuses RF energy in the direction of the culture case and provides a uniform electric field. The culture case is sealed and connected to the main unit for environmental control, located outside the anechoic chamber, via ducts. The temperature at the center of the tray, which contains the culture dishes in the culture room, is maintained at 37.0 +/- 0.2 degrees C by air circulation. In addition, the appropriate CO2 density and humidity supplied to the culture case realizes stable long-term culture conditions. Specific absorption rate (SAR) dosimetry is performed using an electric field measurement technique and the Finite Difference Time Domain (FDTD) calculation method. The results indicate that the mean SAR of the culture fluid at the bottom of the 49 (7 x 7 array) culture dishes used in the in vitro experiments is 0.175 W/kg for an antenna input power of 1 W and the standard deviation of the SAR distribution is 59%. When only 25 culture dishes (5 x 5 array) are evaluated, the mean SAR is 0.139 W/kg for the same antenna input power and the standard deviation of the SAR distribution is 47%. The proliferation of the H4 cell line in 72 h in a pair of RF exposure-incubators reveals that the culture conditions are equivalent to those of a common CO2 incubator.  相似文献

ABSTRACT: BACKGROUND: The symptom of tongue deviation is observed in a stroke or transient ischemic attack. Nevertheless, there is much room for the interpretation of the tongue deviation test. The crucial factor is the lack of an effective quantification method of tongue deviation. If we can quantify the features of the tongue deviation and scientifically verify the relationship between the deviation angle and a stroke, the information provided by the tongue will be helpful in recognizing a warning of a stroke. METHODS: In this study, a quantification method of the tongue deviation angle was proposed for the first time to characterize stroke patients. We captured the tongue images of stroke patients (15 males and 10 females, ranging between 55 and 82 years of age); transient ischemic attack (TIA) patients (16 males and 9 females, ranging between 53 and 79 years of age); and normal subjects (14 males and 11 females, ranging between 52 and 80 years of age) to analyze whether the method is effective. In addition, we used the receiver operating characteristic curve (ROC) for the sensitivity analysis, and determined the threshold value of the tongue deviation angle for the warning sign of a stroke. RESULTS: The means and standard deviations of the tongue deviation angles of the stroke, TIA, and normal groups were: 6.9 [PLUS-MINUS SIGN] 3.1, 4.9 [PLUS-MINUS SIGN] 2.1 and 1.4 [PLUS-MINUS SIGN] 0.8 degrees, respectively. Analyzed by the unpaired Student's t-test, the p-value between the stroke group and the TIA group was 0.015 (>0.01), indicating no significant difference in the tongue deviation angle. The p-values between the stroke group and the normal group, as well as between the TIA group and the normal group were both less than 0.01. These results show the significant differences in the tongue deviation angle between the patient groups (stroke and TIA patients) and the normal group. These results also imply that the tongue deviation angle can effectively identify the patient group (stroke and TIA patients) and the normal group. With respect to the visual examination, 40% and 32% of stroke patients, 24% and 16% of TIA patients, and 4% and 0% of normal subjects were found to have tongue deviations when physicians "A" and "B" examined them. The variation showed the essentiality of the quantification method in a clinical setting. In the receiver operating characteristic curve (ROC), the Area Under Curve (AUC, = 0.96) indicates good discrimination. The tongue deviation angle more than the optimum threshold value (= 3.2[DEGREE SIGN]) predicts a risk of stroke. CONCLUSIONS: In summary, we developed an effective quantification method to characterize the tongue deviation angle, and we confirmed the feasibility of recognizing the tongue deviation angle as an early warning sign of an impending stroke.  相似文献

The quantitative analysis of amino acids (AAs) in single dry blood spot (DBS) samples is an important issue for metabolic diseases as a second-tier test in newborn screening. An analytical method for quantifying underivatized AAs in DBS was developed by using liquid chromatography coupled with tandem mass spectrometry (LC–MS/MS). The sample preparation in this method is simple and ion-pairing agent is not used in the mobile phase that could avoid ion suppression, which happens in mass spectrometry and avoids damage to the column. Through chromatographic separation, some isomeric compounds could be identified and quantified, which cannot be solved through only appropriate multiple reactions monitoring transitions by MS/MS. The concentrations of the different AAs were determined using non-deuterated internal standard. All calibration curves showed excellent linearity within test ranges. For most of the amino acids the accuracy of extraction recovery was between 85.3 and 115 %, and the precision of relative standard deviation was <7.0 %. The 35 AAs could be identified in DBS specimens by the developed LC–MS/MS method in 17–19 min, and eventually 24 AAs in DBS were quantified. The results of the present study prove that this method as a second-tier test in newborn screening for metabolic diseases could be performed by the quantification of free AAs in DBS using the LC–MS/MS method. The assay has advantages of high sensitive, specific, and inexpensive merits because non-deuterated internal standard and acetic acid instead of ion-pairing agent in mobile phase are used in this protocol.  相似文献

A high-performance liquid chromatographic method for the determination of bufaralol, a benzofuran analogue, in plasma is described.The unchanged drug, the major metabolites and an internal standard are extracted from plasma, purified by back-extraction steps and thereafter separated using a reversed-phase liquid chromatographic system. The detection is carried out by means of a fluorescence detector and an UV detector connected in series. The sensitivity of the assay for the unchanged drug and the major metabolite is about 1 ng/ml plasma using a 0.5 ml specimen per analysis and the relative standard deviation of the whole assay lies in the range ± 4–5%.The procedure was successfully used to determine plasma levels in volunteers following a single oral dose of 40 mg of bufaralol. The results obtained using the new high-performance liquid chromatographic method were compared with those determined by another method which combines gas chromatography with mass fragmentography, and it was found that these two sets of results coincided quite well.  相似文献

A number of functions for the autofocusing of microscopes and other optical instruments are to be found in the literature. In this article we compare 11 of them to determine, in an objective manner, which functions are most suitable for implementation with real-time video acquisition systems. Three different images, each representing a typical class of image, are used in the comparison. Among the best focus functions found in our study are the squared magnitude gradient, the squared Laplacian, and the normalized image standard deviation.  相似文献

Because commercial lots of methylene green are known to be heterogeneous, we report a thin layer chromatographic method for checking purity. We also describe a simple and effective flash chromatographic purification procedure for subsequent purification. The coefficient of variation has been used in many evolutionary studies. However, a strong negative correlation between this index and size may artificially inflate the apparent variability of small traits. This is most pronounced when variables whose size differs by more than an order of magnitude are compared or when the index is applied to variables whose size is within an order of magnitude of their measurement error. When this is likely to affect conclusions, other indexes of variability should be considered. One alternative is to use the standard deviation of log-transformed data; however, this index is sometimes still correlated with variable size so care should be exercised in its use. Another alternative is to regress the standard deviation onto mean variable size; however, this method may also be misleading if variables are not randomly distributed about the regression line. As an example of the effect of bias in the coefficient of variation, previous studies of mammalian dental variability profiles were re-evaluated. It was found that variation among teeth is relatively homogenous, both within and among species. The exception is that the canines of some mammalian species have variability that is considerably higher than would be expected from tooth size. Previous explanations of variability patterns that invoked developmental fields are incompatible with the new data.  相似文献

Ablative techniques have been sought in many circumstances as alternatives to surgical resection/incision. Besides being minimally invasive, potential benefits of ablation include greater speed and improved access to target tissue compared with other surgical techniques. There is a wide variety of ablation technologies currently in use for medical treatment. These include but are not limited to tissue heating by radiofrequency (RF) current, microwaves, laser, and high intensity ultrasound. RF is among the most heavily used because of its relatively low complexity and cost. Ablative techniques have proven to be viable alternatives to surgical resection/incision of tissue. Although there are other means of tissue heating besides RF, RF is the most commonly used technique in operating rooms because of the reliability of transmural lesions and the low complexity of the system. Optimal systems account for the heterogeneous nature of tissue and variations in tissue property through the ablation cycle. It is important to monitor and assure adequate energy delivery by selecting the appropriate configuration of devices. Energy delivery varies between the various generators and systems, some more responsive than others with relative to changes in tissue impedance that will affect the end results of the operation.  相似文献

Molecular dynamics simulations of simple bicrystal systems have been much used as a tool to explore how the migration of grain boundaries varies with their structure and with experimental conditions. In order to permit the exploration of a large parameter space, many studies are forced to rely on a small number of simulations (often a single simulation) for each configuration. The motion of a grain boundary is inherently statistical and any variability in the measured grain boundary velocity should be taken into account in subsequent analysis of trends in grain boundary mobility. Here we present the results of large numbers of simulations of equivalent boundaries, which show that this variability can be large, particularly when small systems are simulated. We show how a bootstrap resampling approach can be used to characterise the statistical uncertainty in boundary velocity using the information present in a single simulation. We show that the approach is robust across a variety of system sizes, temperatures and driving force strengths and types, and provides a good order-of-magnitude measure of the population standard deviation across multiple equivalent simulations.  相似文献

A simplified method for titrations of biochemical systems is described as well as extensive error propagation through the data analysis. This work uses a Tronac Model 450 isoperibol titration calorimeter. Sample volumes of 2 ml are used and total heats of less than 5 mcal can be routinely measured. The binding of 3'-CMP to bovine pancreatic ribonuclease A is used to illustrate the methods. The binding enthalpy can be determined with a standard deviation of 1.5% and the free energy with a standard deviation of 2 to 3%.  相似文献

目的 观察是否可以通过对伊马替尼(Imatinib,IM)进行血药浓度监测提高疗效,减少药物不良反应。方法 选取2013~2018年就诊于我院的慢性粒细胞白血病(chronic myelogenous leukemia, CML)患者,分为试验组(药物监测组),对照组(常规经验治疗组)。对服药3个月、6个月、12个月、18个月,进行疗效及不良反应评估及比较。结果 共有51人入选此次临床试验。其中试验组35人,对照组16人。结果 服用伊马替尼400mg/d时,血药浓度568.00~3 989.66ng/ml,均数(标准差):1 716.46ng/ml(788.96);服用伊马替尼300mg/d时,血药浓度720.89~1 497.11ng/ml,均数(标准差):971.67ng/ml(204.02)。达到主要分子学反应(major molecular response, MMR)的伊马替尼血药浓度高于未达到稳态时的伊马替尼血药浓度。两组不良反应评级有统计学差异。试验组III级及以上不良反应发生率明显小于对照组。结论 伊马替尼的稳态血浆药物谷浓度存在较大个体差异,这种个体差异与疗效和不良反应存在相关性。通过治疗药物监测(therapeutic drug monitoring, TDM)可以在确保疗效的同时,减少伊马替尼在治疗慢性粒细胞白血病中的不良反应。结果尚需大样本临床试验进一步验证。伊马替尼药物代谢个体差异的原因需要大样本遗传药理学研究进一步探讨。  相似文献

Considerable interest has focused on idiotypic cross-reactivity among antibody molecules. Cross-reactive idiotypes (Id) on monoclonal and polyclonal rheumatoid factors (RF) have been found frequently. Sufficient attention has not been directed, however, to the proportion of RF exhibiting the cross-reactivity, leaving the impression of extensive RF cross-reactivity when, in fact, this might represent a small minority of total RF molecules in a given individual. We have examined the polyclonal RF from patients with rheumatoid arthritis (RA) for cross-reactive Id in three different assays and with different Id-anti-Id systems. First, a sensitive liquid-phase radioimmunoassay was used in which panels of sera were tested for inhibition of different, idiotypically unrelated, Id-anti-Id interactions. When compared with normal sera, some of the sera from patients with RA caused minimal inhibition of Id-anti-Id interactions. None, however, caused marked inhibition of any Id-anti-Id system. Secondly, the panels of sera were also tested in a direct binding ELISA to detect partially cross-reactive Id that may not have been identified in the inhibition radioimmunoassay without differing results. Finally, results similar to the autologous Id-anti-Id inhibition assay were also found when the panels of RA sera were tested in two nonautologous Id-anti-Id systems, in which the anti-Id reacted with other than their own Id. These studies indicate that although cross-reactivity with some RF of an individual's total RF population may be seen frequently, an individual's repertoire of RF is itself private, quite diverse, and unique to that individual.  相似文献

A stochastic model for hospital infection incorporating both direct transmission and indirect transmission via free-living bacteria in the environment is investigated. We examine the long term behavior of the model by calculating a stationary distribution and normal approximation of the distribution. The quasi-stationary distribution of the model is studied to investigate the models’ behavior before extinction and the time to extinction. Numerical results show agreement between the calculated distributions and results of event-driven simulations. Hand hygiene of volunteers is more effective in terms of reducing the mean (or standard deviation) of the stationary distribution of colonized patients and the expected time to extinction compared to hand hygiene of health care workers (HCWs), on the basis of our parameter values. However, the indirect (or direct) transmission rate can lead to either increase or decrease in the standard deviation of the stationary distribution, but the impact of the indirect transmission is much greater than that of the direct transmission. The findings suggest that isolation of new admitted colonized patients is most effective in reducing both the mean and standard deviation of the stationary distribution and measures related to indirect transmission are secondary in their effects compared to other interventions.  相似文献

A method is presented to estimate the biometric parameters of a quantitative trait locus linked to a genetic marker when both loci are segregating in the F-2 generation of a cross between two inbred lines. The method, which assumes underlying normal distributions, is a combination of maximum likelihood and moments methods and uses the statistics of the genetic marker genotype samples for the quantitative trait to estimate the recombination frequency between the two loci and the means and variances of the genotypes of the quantitative trait locus. With this method, the genetic parameters of a locus affecting plant height linked to an electrophoretic marker for esterase were accurately estimated from a sample of 1596 F-2 progeny of a cross between two species of Lycopersicon (tomato). Linkage distance between the two loci was 38 map units and the effect of the quantitative trait locus was 1.6 phenotypic standard deviation units. Accurate estimates of the genetic parameters and linkage distance for populations of 2000 individuals simulated with a segregating codominant locus with an effect of 1.63 standard deviations linked to a genetic marker with .2 recombination were also derived by this method. The method is not effective in distinguishing between complete and partial linkage in samples of only 500 individuals or for quantitative loci with effects less than a phenotypic standard deviation. The method is more effective for codominant than for dominant loci.  相似文献

The linear gradient mode of chromatography is the most widely employed mode of operation in ion-exchange chromatographic separations. However, in recent years, the displacement mode has received considerable attention because of its promise of high throughput and high resolution. To enable a comparison of these two modes of chromatography, it is essential to identify the optimum operating conditions for each. We employed an iterative algorithm to carry out the necessary optimization. The Steric Mass Action model of ion-exchange chromatography is used in concert with the solid-film linear-driving force model to describe the chromatographic behavior of the solutes in these systems. The performances of displacement and gradient modes of chromatography are compared for different types of separation problems. It turns out that for "easy" separations, both the modes are equally effective. However, for challenging separations, the displacement mode is superior to the gradient mode. Our results shed significant light on the performance of gradient and displacement modes in protein ion-exchange systems.  相似文献
