During mouse gastrulation, the primitive streak is formed on the posterior side of the embryo. Cells migrate out of the primitive streak to form the future mesoderm and endoderm. Fate mapping studies revealed a group of cell migrate through the proximal end of the primitive streak and give rise to the extraembryonic mesoderm tissues such as the yolk sac blood islands and allantois. However, it is not clear whether the formation of a morphological primitive streak is required for the development of these extraembryonic mesodermal tissues. Loss of the Cripto gene in mice dramatically reduces, but does not completely abolish, Nodal activity leading to the absence of a morphological primitive streak. However, embryonic erythrocytes are still formed and assembled into the blood islands. In addition, Cripto mutant embryos form allantoic buds. However, Drap1 mutant embryos have excessive Nodal activity in the epiblast cells before gastrulation and form an expanded primitive streak, but no yolk sac blood islands or allantoic bud formation. Lefty2 embryos also have elevated levels of Nodal activity in the primitive streak during gastrulation, and undergo normal blood island and allantois formation. We therefore speculate that low level of Nodal activity disrupts the formation of morphological primitive streak on the posterior side, but still allows the formation of primitive streak cells on the proximal side, which give rise to the extraembryonic mesodermal tissues formation. Excessive Nodal activity in the epiblast at pre‐gastrulation stage, but not in the primitive streak cells during gastrulation, disrupts extraembryonic mesoderm development.  相似文献

The well known and most important function of nucleoli is ribosome biogenesis. However, the nucleolus showed delayed development and malfunction in somatic cell nuclear transfer (NT) embryos. Previous studies indicated that nearly half rRNA genes (rDNA) in somatic cells were inactive and not transcribed. We compared the rDNA methylation level, active nucleolar organizer region (NORs) numbers, nucleolar proteins (upstream binding factor (UBF), nucleophosmin (B23)) distribution, and nucleolar-related gene expression in three different donor cells and NT embryos. The results showed embryonic stem cells (ESCs) had the most active NORs and lowest rDNA methylation level (7.66 and 6.76%), whereas mouse embryonic fibroblasts (MEFs) were the opposite (4.70 and 22.57%). After the donor cells were injected into enucleated MII oocytes, cumulus cells and MEFs nuclei lost B23 and UBF signals in 20 min, whereas in ESC-NT embryos, B23 and UBF signals could still be detected at 60 min post-NT. The embryos derived from ESCs, cumulus cells, and MEFs showed the same trend in active NORs numbers (7.19 versus 6.68 versus 5.77, p < 0.05) and rDNA methylation levels (6.36 versus 9.67% versus 15.52%) at the 4-cell stage as that in donor cells. However, the MEF-NT embryos displayed low rRNA synthesis/processing potential at morula stage and had an obvious decrease in blastocyst developmental rate. The results presented clear evidences that the rDNA reprogramming efficiency in NT embryos was determined by the rDNA activity in donor cells from which they derived.  相似文献

The nucleolus constitutes a cytologically visible phenotype for ribosomal DNA (rDNA). Nucleolar size, as determined by silver staining, is a good indicator of cell proliferation rate and biosynthetic activity. Nevertheless, the relationship between rDNA content and sexual dimorphism for nucleolar size is not well documented. In the present study, the impact of sex and ploidy level on nucleolar size is investigated in three haplo/diploid and three diplo/diploid species of insect. Nucleolar sizes are found to be proportional to ploidy level in the haplo/diploid hymenopterans Trypoxylon albitarse and Nasonia vitripennis. Conversely, in the ant Messor barbarus, nucleolar sizes are larger in haploid males (winged) than diploid females (apterous). Among the diplo/diploid species, evidence for gene dosage compensation on nucleolar activity is suggested by the absence of sex differences in Drosophila simulans, a species in which rDNA is limited to the X chromosome. By contrast, in the grasshopper Stenobothrus festivus, another species with rRNA genes restricted to the X chromosome, the size of the nucleolus is significantly larger in females than in males. Additionally, in the grasshopper Chorthippus parallelus, where rDNA is distributed evenly on several autosomes of males and females, the females also show larger nucleoli than males. In both grasshopper species, the magnitude of the female/male ratio for nucleolus area is very similar to the body size ratio, suggesting that body size, as well as sex, ploidy, gene dosage and physiological activity, may be an important determinant of nucleolus area.  相似文献

Summary: Mouse embryos homozygous for the allele eedl7Rn5‐3354SB of the Polycomb Group gene embryonic ectoderm development (eed) display a gastrulation defect in which epiblast cells move through the streak and form extraembryonic mesoderm derivatives at the expense of development of the embryo proper. Here we demonstrate that homozygous mutant ES cells have the capacity to differentiate embryonic cell types both in vitro as embryoid bodies and in vivo as chimeric embryos. In chimeric embryos, eed mutant cells can respond to wild‐type signals and participate in normal gastrulation movements. These results indicate a non–cell‐autonomous function for eed. Evidence of mutant cell exclusion from the forebrain and segregation within somites, however, suggests that eed has cell‐autonomous roles in aspects of organogenesis. In the naturally synchronous mitosis of the syncytial plasmodium of Physarum polycephalum , the nucleolus disintegrates in prophase, releasing a large amount of ribosomal RNA. Using biotinylated rDNA probes, we studied by high-resolution in situ hybridization the behavior of this nucleolar RNA throughout mitosis. Our results demonstrate that this rRNA is stable and maintained within the mitotic nucleus mainly, but not exclusively, associated with fibrillar nucleolar remnants. The distribution of these rRNA molecules on both sides of the cleavage plane in telophase is indicative of a precise mechanism of mitotic partition of the nucleolar components, supporting our recent findings concerning the rDNA minichromosomes (Puvion-Dutilleul and Pierron, 1992, Exp. Cell Res. 203, 354-364). Taking advantage of the stability of this RNA component in mitosis, we unambiguously demonstrate that the nucleolar remnants are the precursors of the prenucleolar bodies appearing in the newly divided nuclei which, by fusion, reconstitute a single nucleolus. Our data exemplify the persistence of the nucleolar rRNA in mitosis and demonstrate that in Physarum, following its disintegration, the nucleolus is segregated and inherited.  相似文献

SYNOPSIS. Studies on the nudeohis in various species of Amphibia,including the Mexican axolotl, have contributed to an understandingof the function of the nucleolar organizing region. In the firstpart of this paper we have reviewed some of the research onnucleolar variants in the axolotl. These variants were recognizedin different strains of laboratory reared animals. The phenotypesinclude diminished nucleolar size and possible alterations inthe levels of rDNA. The expression of several of these variantnucleolar phenotypes does not, however, reflect a simple deletionof rRNA genes. Evidence from analyses of several mutant andapparently normal strains indicate that some animals have relativelyhigh levels of rRNA redundancy, while others, including someanimals which display variant nucleolar sizes, display "normal"levels. Data on the inheritance of rRNA genome sizes in the axolotlis reviewed. The observation that intermediate levels of redundancywere detected in some crosses is discussed in terms of inheritancedata from other organisms. Arabidopsis thaliana 45S ribosomal genes (rDNA) are located in tandem arrays called nucleolus organizing regions on the termini of chromosomes 2 and 4 (NOR2 and NOR4) and encode rRNA, a crucial structural element of the ribosome. The current model of rDNA organization suggests that inactive rRNA genes accumulate in the condensed chromocenters in the nucleus and at the nucleolar periphery, while the nucleolus delineates active genes. We challenge the perspective that all intranucleolar rDNA is active by showing that a subset of nucleolar rDNA assembles into condensed foci marked by H3.1 and H3.3 histones that also contain the repressive H3K9me2 histone mark. By using plant lines containing a low number of rDNA copies, we further found that the condensed foci relate to the folding of rDNA, which appears to be a common mechanism of rDNA regulation inside the nucleolus. The H3K9me2 histone mark found in condensed foci represents a typical modification of bulk inactive rDNA, as we show by genome-wide approaches, similar to the H2A.W histone variant. The euchromatin histone marks H3K27me3 and H3K4me3, in contrast, do not colocalize with nucleolar foci and their overall levels in the nucleolus are very low. We further demonstrate that the rDNA promoter is an important regulatory region of the rDNA, where the distribution of histone variants and histone modifications are modulated in response to rDNA activity.  相似文献

Mixl1 is the only member of the Mix/Bix homeobox gene family identified in mammals. During mouse embryogenesis, Mixl1 is first expressed at embryonic day (E)5.5 in cells of the visceral endoderm (VE). At the time of gastrulation, Mixl1 expression is detected in the vicinity of the primitive streak. Mixl1 is expressed in cells located within the primitive streak, in nascent mesoderm cells exiting the primitive streak, and in posterior VE overlying the primitive streak. Genetic ablation of Mixl1 in mice has revealed its crucial role in mesoderm and endoderm cell specification and tissue morphogenesis during early embryonic development. However, the early lethality of the constitutive Mixl1?/? mutant precludes the study of its role at later stages of embryogenesis and in adult mice. To circumvent this limitation, we have generated a conditional Mixl1 allele (Mixl1cKO) that permits temporal as well as spatial control of gene ablation. Animals homozygous for the Mixl1cKO conditional allele were viable and fertile. Mixl1KO/KO embryos generated by crossing of Mixl1cKO/cKO mice with Sox2‐Cre or EIIa‐Cre transgenic mice were embryonic lethal at early somite stages. By contrast to wild‐type embryos, Mixl1KO/KO embryos contained no detectable Mixl1, validating the Mixl1cKO as a protein null after Cre‐mediated excision. Mixl1KO/KO embryos resembled the previously reported Mixl1?/? mutant phenotype. Uniparental silencing of 35S rRNA genes (rDNA), known as nucleolar dominance (ND), is common in interspecific hybrids. Allotetraploid Tragopogon mirus composed of Tragopogon dubius (d) and Tragopogon porrifolius (p) genomes shows highly variable ND. To examine the molecular basis of such variation, we studied the genetic and epigenetic features of rDNA homeologs in several lines derived from recently and independently formed natural populations. Inbred lines derived from T. mirus with a dominant d‐rDNA homeolog transmitted this expression pattern over generations, which may explain why it is prevalent among natural populations. In contrast, lines derived from the p‐rDNA dominant progenitor were meiotically unstable, frequently switching to co‐dominance. Interpopulation crosses between progenitors displaying reciprocal ND resulted in d‐rDNA dominance, indicating immediate suppression of p‐homeologs in F1 hybrids. Original p‐rDNA dominance was not restored in later generations, even in those segregants that inherited the corresponding parental rDNA genotype, thus indicating the generation of additional p‐rDNA and d‐rDNA epigenetic variants. Despite preserved intergenic spacer (IGS) structure, they showed altered cytosine methylation and chromatin condensation patterns, and a correlation between expression, hypomethylation of RNA Pol I promoters and chromatin decondensation was apparent. Reversion of such epigenetic variants occurred rarely, resulting in co‐dominance maintained in individuals with distinct genotypes. Generally, interpopulation crosses may generate epialleles that are not present in natural populations, underlying epigenetic dynamics in young allopolyploids. We hypothesize that highly expressed variants with distinct IGS features may induce heritable epigenetic reprogramming of the partner rDNA arrays, harmonizing the expression of thousands of genes in allopolyploids.  相似文献

The rDNA transcribed region (TR) was tested for its accessibility for RsaI recognizing 15 TR sites, DNase I, and photoinducible arylazide (N-(4-azido-2-hydroxybenzoyl)-N,N"-diaminoheptane acetate) in isolated nuclei, and for the arylazide in intact cells. Arylazide readily entered the cells and did not appreciably affect the chromatin structure. Its photolysis products efficiently modified DNA in accessible sites. Single-strand breaks made by DNase I were not transformed into two-strand ones in rDNA TR, suggesting the necessity of denaturing electrophoresis for such an analysis. About 70% of all rDNA copies proved poorly accessible for endonucleases and arylazide, the accessibility being higher in their 18S and 5.8S rRNA gene regions than in the regions of the external transcribed spacers (ETSs) and the 28S rRNA gene. Proteinase K disrupted this structure, and the corresponding copies were extracted from nuclei. This explained whyin situ hybridization occasionally fails to reveal rDNA in the nucleolar fibrillar center (FC) on electron microscopic preparations. In other rDNA copies, TR (excluding 5"-ETS) was accessible for nucleases and arylazide. These copies were not extracted from nuclei treated with proteinase K. Some of their RsaI sites were protected by tightly bound proteins. Seven such regions were identified in TR. Possible association of the molecular structure, nucleolar location, and functional state of rDNA is discussed.  相似文献

Summary In this work we have used a method that allows a rapid and precise quantification of rRNA genes. With the purpose of examining small numbers of rRNA genes, we used Drosophila melanogaster embryos which are inviable because their sex chromosomes carry extensive rDNA deletions. Two of the mutants, B s Ybb 1 and Ybb l , appear to be completely devoid of rDNA. Summary Bruchidius embryos are shown to be well suited for biochemical studies during early embryogenesis. Mass cultivation is easy, and highly synchronized embryos can be obtained in large numbers (104–105 eggs). A method for in vivo incubation is described which allows the labelling of newly synthesized RNA. The kinetics of3H-ruidine uptake, phosphorylation and incorporation into RNA are presented. By autoradiography, the distribution of newly synthesized RNA is shown. Thereby, stage-specific differences were found in the labelling pattern of vitellophage nuclei, of blastoderm nuclei and of the nuclei of pole cells. The labelling of the cytoplasm remains weak until cellular blastoderm is formed. During late blastoderm and at gastrulation this label increases markedly. Gel electrophoresis of isolated RNA shows that at cellular blastoderm formation most of the label occurs in a region between 18 S and 7 S. Later on, at the onset of gastrulation, the3H-uridine incorporation found in isolated RNA is raised about 10 fold and rRNA synthesis becomes prominent. In a chase experiment, the processing of precursor RNA molecules into shorter RNA species, especially into mature rRNA and 5S RNA, is shown. The advantages of theBruchidius embryo for the biochemical analysis of early RNA synthesis and the regulation of rRNA synthesis in insect embryos are discussed.Dedicated to Professor Dr. Dr. h. c.
