A study was made on the mass cultivation of Nannochloropsis sp. in newly designed annular reactors operated under natural, artificial or combined illumination. The annular reactor consists of two 2-m-high Plexiglas cylinders of different diameter placed vertically one inside the other so as to form an annular culture chamber. Artificial illumination is supplied by lamps placed inside the inner cylinder. Two annular reactors of different diameter (50 and 91 cm), light path (4.5 and 3.0 cm) and illuminated surface area (5.3 and 9.3 m2) were experimented with. The effect of two different artificial light sources (fluorescent tubes and metal halide lamps) on culture productivity was investigated in both systems. The highest productivity on a per reactor basis (about 34 g (d. wt) reactor–1 24 h–1) was achieved with the larger reactor illuminated by a 400-W metal halide lamp. From February to May a 91-cm reactor illuminated only with natural light was operated in parallel with a 91-cm reactor subjected to combined illumination. Under natural illumination productivity increased from 16.6 g (d. wt) reactor–1 d–1 in February to 34.1 g (d. wt) reactor–1 d–1 in May. Under combined illumination productivity was 41.3 g (d. wt) reactor–1 d–1 in February and increased up to 48.3 g (d. wt) reactor–1 d–1 in May. Although the culture exposed to combined illumination always attained higher yields, the productivity gap between the two cultures decreased gradually along the season as solar radiation and minimum night temperatures increased. A 1200-L plant made of ten 50-cm annular reactors was set up and operated for two years with combined illumination yielding an average of 270 g of dry Nannochloropsis sp. biomass per day. More than 2000 L of concentrate suspension (50 g (d. wt) L–1) of Nannochloropsis sp. were produced and successfully used by fish hatcheries as live feed for rotifers and for rearing seabream larvae with the green-water technique. This study indicates that the annular reactor can be profitably used for long-term cultivation of Nannochloropsis in temperate climates. Besides reliability and ease of operation, the main advantage of the system is that it can be used under natural illumination, yet artificial light can be also supplied to maintain high productivity levels in winter or on cloudy days. 相似文献

The cell growth and eicosapentaenoic acid (EPA) yields of Nannochloropsis sp. were enhanced in the fed-batch cultures. With feeding glucose solution, the biomass reached 1.1 g dry wt l(-1) after 10 days' culture, which was 40% higher than that obtained in the batch culture (0.8 g dry wt l(-1)). With supplement of nitrate solution, the biomass reached 1 g dry wt l(-1), and reached the stationary phase 2 days earlier than the others. The maximum of biomass (1.2 g dry wt l(-1)) was obtained with the supplement of the mixture of glucose and nitrate solution. The EPA yields of Nannochloropsis sp. after 10 days' growth in the fed-batch cultures were 52 mg l(-1), 43 mg l(-1) and 56 mg l(-1) with, respectively, addition of nitrate, glucose and both together. Nannochloropsis sp. was grown under mixotrophic conditions with 30 mM glucose as carbon source, reaching 507 mg dry wt l(-1) after 8 d. This was 1.4-fold of that obtained under photoautotrophic conditions. Under mixotrophic and photoautotrophic cultivations, the net photosynthetic rate of Nannochloropsis sp. did not change but the respiratory rate increased in the mixotrophic cultivation. The yield of eicosapentaenoic acid was 22 mg l(-1) in the mixotrophic cultivation and 20 mg l(-1) under the photoautotrophic cultivation. 相似文献

Photoacclimation in the marine eustigmatophyte Nannochlropsis sp., used extensively as a food chaincomponent in aquaculture, was studied both in thelaboratory and outdoors. Cell-chlorophyll andcarotenoids were used as markers to assessphotoacclimation to strong light, as well as todecreasing growth irradiance due to cellproliferation. Focusing on practical aspects involvedin mass cultivation, three different approaches wereused as follows: (a) cultures initially exposed to lowlight (150 mol photon m-2 s-1) thentransferred to strong light (1000 to 3000 molphoton m-2 s-1); (b) initially low celldensity cultures grown in reactors of differentlight-paths, exposed to strong PFD, in the laboratoryand outdoors; (c) initially low or high cell densitycultures exposed to strong light. As has already beenestablished in many reports, cell-chlorophyllrepresented a sensitive parameter in assessing cellresponse to changes in the intensity of the lightsource as well as to modifications in the light regimeto which the cells were exposed. Cell-chlorophyllconcentration sharply decreased initially upontransferring the culture from low PFD cell-1 tohigh PFD cell-1 due to either culture dilution(i.e. decrease in cell density and mutual shading) orto an increase in PFD. After some 7 days ofphotoacclimating to 2000 and 3000 mol photonm-2 s-1, chlorophyll a content began to riseat a much faster rate than cell number, which alsoincreased in response to the higher irradiance.Cell-chlorophyll in the culture exposed to 2000mol photon m-2 s-1 increased afteracclimation earlier and at a faster rate than in theculture exposed to 3000 mol photon m-2s-1, indicating the later irradiance affected astronger stress. The length of the reactor's lightpath exerted a decisive effect on cell response tostrong light through its influence on the light regimein the culture. Upon a sharp increase in PFD,carotenoids in the 1-cm reactor increased in muchhigher rate than chlorophyll, compared with the 3-cmlight path reactors. This marked difference in cellresponse to a shift-up in light was attributed to thevast variations in the light regime associated withdifferences in the length of the light path and areal density. Growth oflow cell density cultures ceased temporarily upontransfer to strong light, in contrast with high celldensity cultures transferred to strong light, whichcontinued growth without a lag. 相似文献

To increase eicosapentaenoic acid (EPA, 20:5, n-3) content in the marine alga Nannochloropsis sp., the effect of CO2 concentration during cultivation has been investigated. In a batch culture under normal atmospheric conditions (0.037% CO2), the EPA content per cell increased during the first 1.5 days and then decreased immediately even though the cells were in an exponential growth phase. Increasing the CO2 concentration to 0.3% and 2% over day 1.5 retained the EPA content at the higher concentration for another 1 and 2 days, respectively, suggesting that the EPA accumulation is enhanced by elevated concentrations of CO2. EPA accumulation in response to elevated CO2 concentrations was also observed during a later growth phase when CO2 was introduced after the decrease of EPA content. The addition of CO2 caused a slight decrease in the pH of the medium though this was not the cause of the observed EPA accumulation as addition of acidic buffer did not affect the EPA content. The maximum EPA production was obtained when 2% CO2 was supplied 12 h prior to the end of the exponential growth. The total EPA production during 4-day cultivation was about twice that obtained with ambient air. These results suggest that the available CO2 concentration affects the EPA content in Nannochloropsis sp. 相似文献

Optimization of growth and fatty acid composition of a unicellular marine picoplankton,Nannochloropsis sp., with enriched carbon sources 总被引:5,自引:0,他引:5
A unicellular marine picoplankton, Nannochloropsis sp., was grown under CO2-enriched photoautotrophic or/and acetate-added mixotrophic conditions. Photoautotrophic conditions with enriched CO2 of 2800 l CO2 l–1 and aeration gave the highest biomass yield (634 mg dry wt l–1), the highest total lipid content (9% of dry wt), total fatty acids (64 mg g–1 dry wt), polyunsaturated fatty acids (35% total fatty acids) and eicosapentaenoic acid (EPA, 20:53) (16 mg g–1 dry wt or 25% of total fatty acids). Mixotrophic cultures gave a greater protein content but less carbohydrates. Adding sodium acetate (2 mM) decreased the amounts of the total fatty acids and EPA. Elevation of CO2 in photoautotrophic culture thus enhances growth and raises the production of EPA in Nannochloropsis sp. 相似文献

Nannochloropsis sp. was grown with different levels of nitrate, phosphate, salinity and temperature with CO2 at 2,800 μl l−1. Increased levels of NaNO3 and KH2PO4 raised protein and polyunsaturated fatty acids (PUFAs) contents but decreased carbohydrate, total lipid and total fatty acids (TFA) contents. Nannochloropsis sp. grew well at salinities from 22 to 49 g l−1, and lowering salinity enhanced TFA and PUFAs contents. TFA contents increased with the increasing temperature but PUFAs contents decreased. The highest eicosapentaenoic acid (EPA, 20:5ω3) content based on the dry mass was above 3% under low N (150 μM NaNO3) or high N (3000 μM NaNO3) condition. Excessive nitrate, low salinity and temperature are thus favorable factors for improving EPA yields in Nannochloropsis sp. 相似文献

Nannochloropsis sp. was grown semicontinuously with a rate of daily renewal of the culture media of 40% of the volume of the culture under different irradiances (40, 60, 80, 220 and 480 mol quanta m–2 s–1). Under the conditions tested, light saturation was achieved at 220 mol quanta m–2 s–1 with no significant increase in steady-state cell density or of dry weight productivity with higher irradiance, reaching values of 115 × 106 cells ml–1 and 375 mg l–1 day–1 respectively. C/N ratios clearly indicated the point of light saturation, decreasing with increasing irradiance for light-limited conditions and increasing for light-saturated conditions. Under light-limited conditions, an increase in the irradiance produced an increase in the protein percentage of the organic fraction to the detriment of lipids and carbohydrates, while small changes were recorded under light-saturated conditions. The degree of unsaturation of fatty acids was lower with increasing irradiance, with a three-fold decrease of the percentage of total n–3 fatty acids, from 29 to 8% of total fatty acids, caused mainly by a decrease of eicosapentaenoic acid (EPA) (20:5n–3). The microalga reached its maximal value of dry weight productivity (375 mg l–1 day–1), EPA productivity (3.2 mg l–1 day–1) and maximal protein content (36% of the organic content) at the point at which light saturation was achieved. A strain of Nannochloropsis isolated originally from the East China Sea and obtained from Institute of Hydrobiology, Chinese Academy of Sciences was shown to utilize glucose or ethanol for mixotrophic and heterotrophic growth. The highest cell density, 550 mg L− 1 dry weight after culture for 8 days, was obtained during mixotrophic culture with 30 mM glucose. The organic carbon sources had no effect on the net photosynthetic rate, but enhanced the respiratory rate. The addition of an organic carbon source led to an increase in the cell lipid content and a decrease in their eicosapentaenoic acid (EPA) content. The EPA yield was 21.9 mg L− 1 using photoautotrophic culture, and 23.4 mg L− 1 and 23.0 mg L− 1, respectively, in mixotrophic cultivation with glucose or ethanol as the carbon source. 相似文献

In order to study the effects of different nitrogen source and concentrationon the growth rate and fatty acid composition, a marine microalga Ellipsoidion sp. with a high content of eicosapentaenoic acid (EPA) wascultured in media with different nitrogen sources and concentrations.During the pre-logarithmic phase, the alga grew faster with ammoniumas N source than with nitrate, but the reverse applied during thepost-logarithmic phase. The alga grew poorly in N-free mediumor medium with urea as the sole N source. In the same growth phase,ammonium medium resulted in higher yield of total lipid, but the EPA yielddid not differ significantly different from that using nitrate medium. Themaximum growth rate occurred in medium containing 1.28 mmolL-1 sodium nitrate, while maximum EPA and total lipid contents werereached at 1.92 mmol L-1, when EPA accounted for 27.9% totalfatty acids. The growth rate kept stable when NH4Cl ranged from0.64 to 2.56 mmol L-1, and the maximum content of total lipidand EPA occurred in the medium with 2.56 mmol L-1NH4Cl. The EPA content was higher in the pre- thanpost-logarithmic phase, though the total lipid content was lower. Pigment composition and its variation with culture agewere analyzed in six strains of Nannochloropsis(Eustigmatophyceae). The capacity for accumulationof the ketocarotenoids astaxanthin and canthaxanthinwas higher in N. salina and N. gaditanathan in the other strains studied here. Theinfluence of salinity (15 to 100 practical units) onpigment production was studied in N. gaditana,where a defined pattern of variation could not befound apart from a notable increase in zeaxanthin at100. In cultures grown in a photobioreactor and athigh cell densities of about 109 cells mL-1,pigment production reached: 350 mg L-1 forchlorophyll a, 50 mg L-1 for violaxanthin,5 mg L-1 for canthaxanthin, 3 mg L-1 forastaxanthin. The highest contents of canthaxanthin andastaxanthin obtained in experiments with N.gaditana were 19.4 and 14.6 ng pigment (106cells)-1, respectively, which accounts for 0.7%dry weight. By means of xanthophyll cycle inductionthrough exposure of cells to high irradiance and at40 °C, conversion of violaxanthin intozeaxanthin may attain up to 70% of the violaxanthincontent, which corresponds to 0.6% dry weight. Theresults indicate that interest in Nannochloropsis as a source of valuable pigments isnot related to its capacity for single pigmentaccumulation, but the availability of a range ofpigments such as chlorophyll a, zeaxanthin,canthaxanthin and astaxanthin, each with highproduction levels. 相似文献

To extract the microalgal lipid in situ, biocompatible solvents were screened for lipid milking of Nannochloropsis sp. in an aqueous–organic system. The effects of organic solvents on the microalgal growth, the lipid extractability, the dehydrogenases activity and the cell membrane integrity were investigated by UV–visible spectrophotometer, FT-IR spectroscopy, 2,3,5-triphenyltetrazolium chloride (TTC) and Evans Blue stain method, respectively. The results showed that alkane solvents with log P > 5.5 were biocompatible while the hydrophilic solvents with log P < 5.5 were toxic to Nannochloropsis sp. due to the deactivated dehydrogenase and increased cell membrane permeability. As 10% (v/v) hexadecane was used to establish biphasic system, the total lipid production of Nannochloropsis sp. was increased by 28.9% compared to the control. The screened biocompatible solvent hexadecane enhanced not only the algal growth but also the lipid accumulation, showing an effective way to facilitate the process for in situ lipid milking from Nannochloropsis sp. 相似文献

5-Aminolevulinic acid (ALA) was produced aerobically in the dark during growth on glucose by a newly isolated Chlorella sp. When levulinic acid (20 mM), a competitive inhibitor of ALA dehydratase, was added repeatedly to the medium, about 1.5 mM of ALA was produced extracellularly. Glutamate (30 mM) added with levulinic acid (20 mM, given repeatedly) enhanced ALA production up to 1.9 mM, indicating that ALA might be synthesized via the C-5 pathway.K. Many industrial wastes contain Cr(VI), a carcinogen and mutagen, the toxicity of which can be ameliorated by reduction to Cr(III). Microbacterium sp. NCIMB 13776 andDesulfovibrio vulgaris NCIMB 8303 reduced Cr(VI) to Cr(III) anoxically using 25 mM sodium citrate buffer (pH 7), with 25 mM sodium acetate and 25 mM sodium formate as electron donors at 30 °C, under which conditions the rates of reduction of 500 M sodium chromate were 77 and 6 nmol h–1 mg dry cell wt for D. vulgaris and Microbacterium sp., respectively, these being increased to 127 and 17 nmol h–1 mg dry cell wt in the presence of 20 mM MOPS/NaOH buffer. 相似文献

A fermentation process for the microbial production of a new lipid surface-active compound, 7,10-dihydroxy-8 (E)-octadecenoic acid (OCD), has been established using a vegetable oil as carbon source in a coordinated carbon/nitrogen feed strategy. The taxonomic positions of soil isolates known as Streptomyces groups A, B and C were clarified. Comparative 16S rDNA sequence studies indicated that representatives of all three taxa
formed distinct phyletic lines within the Streptomyces tree though the group A strains were shown to be related to Streptomyces griseus and associated validly described species. The taxonomic integrity of all three groups was highlighted by DNA:DNA relatedness
and ribotype data though the group A strains encompassed a higher degree of genetic variation than the group B and C strains.
In light of these and earlier phenotypic data it is proposed that Streptomyces groups A, B and C be given species status as Streptomyces sanglieri sp. nov., Streptomyces aureus sp. nov. and Streptomyces laceyi sp. nov., respectively.
