桦褐孔菌三萜化合物提取工艺的优选   总被引:2,自引:0,他引:2  
为了提高桦褐孔菌三萜化合物的产量,对其三萜化合物的提取工艺进行了初步优选。试验以桦褐孔菌发酵菌丝体为材料,齐墩果酸为标准品,用香草醛比色法测桦褐孔菌三萜化合物含量。通过单因素实验考察了不同提取剂、提取剂用量、提取温度、提取时间对桦褐孔菌三萜化合物提取的影响。结果表明,提取的最佳工艺为:以异丙醇为提取溶剂,用量为7倍,最佳提取温度为80℃,提取时间2h,在此条件下三萜化合物占菌丝体干质量的9.03%。该工艺的三萜桦褐物的提取率高,可用于桦褐孔菌中三萜化合物的提取。  相似文献

通过单因素实验和正交实验对影响桦褐孔菌三萜提取的异丙醇体积浓度、硫酸铵质量浓度、料液比和超声时间等因素进行探讨,旨在建立桦褐孔菌Inonotus obliquus菌丝体三萜类化合物的提取工艺。结果表明,提取桦褐孔菌三萜的最佳条件为:异丙醇体积浓度40%,硫酸铵质量浓度0.125g/mL,料液比(W/V)0.9:1,超声时间为35min,三萜提取率为6.18%,提取效率明显高于常规超声波提取法(5.38%)。利用该方法获得的三萜具有较强清除自由基的能力。通过正交实验获得的桦褐孔菌三萜的提取工艺是一种经济、有效的方法。  相似文献

The submerged cultivating conditions for triterpenoids production from Antrodia cinnamomea were optimized using uniform design method and the one-factor-at-a-time method was adopted to investigate the effect of plants oils and glucose supply on triterpenoids production and mycelia growth. Corn starch and culturing time were identified as more significant variables for triterpenoids production. The optimal conditions for triterpenoids production was 20.0 g/L corn starch, 20.0 g/L wheat bran, 1.85 g/L MgSO4, initial pH 3 and 16 days of cultivation. In addition, investigation of plant oils and glucose supply showed that 0.3 % (v/v) olive oil supply at the beginning of fermentation stimulated mycelia growth and significantly increased triterpenoids production; 0.2 % (w/v) glucose supplement at 10th day enhanced production of triterpenoids with slight effect on biomass, which is reported for the first time. The triterpenoids production experimentally obtained under the optimal conditions was 7.23 % (w/w). The uniform design method may be used to optimize many environmental and genetic factors such as temperature and agitation that can also affect the triterpenoids production from A. cinnamomea.  相似文献

The isolation of pentacyclic triterpenoids from seven species of fresh mangrove leaves using a simple and rapid method is described. The leaves were homogenized using chloroform—methanol and the extract was diluted with water to precipitate out triterpenoids which were separated into neutral and acidic fractions. These were analysed by gas-liquid chromatography as acetyl and trimethylsilyl ether derivatives on a 3% OV-17 column. Sterols were isolated from the chloroform layer by preparative thin layer chromatography and were analysed by gas-liquid chromatography as their trimethylsilyl ether derivatives on a 3% OV-17 column. The triterpenoids found were α-amyrin, β-amyrin, lupeol, oleanolic acid and ursolic acid in most of the samples. Sterols found in all the samples were cholesterol, campesterol, stigmasterol, sitosterol and stigmast-7-en-3β-ol. Retention indices of the triterpenoids and sterols have been determined.  相似文献

本文报道了基于香草醛-高氯酸显色反应的分光光度法定量测定灵芝三萜的修正方法,并对该方法应用进行了探讨和优化。采用此方法检测了灵芝子实体中含量较高的几种三萜酸,结果表明若采用齐墩果酸为标准品检测灵芝三萜,检测结果远低于真实值。在光谱分析上,研究表明对紫外-可见光扫描吸收峰进行面积积分,获得的标准曲线的线性关系更优。  相似文献

本研究考察乙醇浓度对灵芝子实体中三萜成分提取的影响及提取物抗前列腺癌细胞LNCaP的增殖活性和迁移能力,为提取灵芝三萜时乙醇浓度的选择和灵芝抑制前列腺相关疾病产品开发提供依据。研究采用高效液相(HPLC)色谱法测定了灵芝醇提物中目前具有代表性的9种三萜的含量;并用抑制前列腺癌细胞LNCaP的细胞活性和细胞迁移能力的实验研究灵芝提取物的抗前列腺癌的作用。HPLC测定结果显示,当乙醇浓度为50%-80%时,9种灵芝三萜的含量随着乙醇浓度的升高而增大;当乙醇浓度继续升高至95%时,提取的三萜含量反而有所降低。细胞实验结果显示,4个浓度乙醇的灵芝提取物都具有良好的抑制LNCaP肿瘤细胞增殖作用和阻滞LNCaP细胞迁移能力。因此在使用乙醇提取灵芝三萜时,乙醇浓度升高有利于三萜的提取;在生产灵芝抗前列腺相关产品时,提取灵芝子实体中抗前列腺增生的活性成分时选用80%的乙醇较为适宜。  相似文献

Two unusual triterpenoids, (24S)-24-methyl-25,32-cyclo-5alpha-lanosta-9(11)-en-3beta-ol and (24S)-24-methyl-25,32-cyclo-cycloartane-3beta-ol, were isolated from leaves of Pandanus boninensis along with known triterpenoids and lignans. Agrostophyllinol and agrostophyllinone, two new triterpenoids, were isolated from the orchid Agrostophyllum brevipes. Agrostophyllinone was also isolated from another orchid Agrostophyllum callosum. The structures of agrostophyllinol and agrostophyllinone were established as 24-methylene-lanosta-9(11)-en-3beta-ol (5a) and 24-methylene-lanosta-9(11)-en-3-one (5c), respectively, from spectral and chemical evidence. 为了更好地发挥紫芝的药用功效,本文优选了紫芝子实体三萜类化合物的提取条件。采用L9(34)正交实验设计,以提取溶媒、提取方法、提取时间和提取次数四因素三水平的提取工艺优选。各因素重要性依次为提取溶媒>提取次数>提取方法>提取时间。本文确定了以氯仿或丙酮为提取溶剂提取紫芝中三萜类物质的标准化方法。在提取过程中如果采用甲醇、乙醇或水作为提取溶媒,虽然提取效率高,但含糖、氨基酸等其它非三萜成分多,三萜类物质提取不全。  相似文献

Four iridal-type triterpenoids, two of which were new compounds, have been isolated from rhizomes of Iris tectorum Maxim. Their structures were determined by 1D and 2D NMR spectroscopy and ESI-MS spectrometry. The compounds were identified as the iritectols A and B, and the known iridobelamal A and isoiridogermanal. The presence of epoxide and tetrahydrofuran functions are not common in previously isolated iridal-type triterpenoids.  相似文献

三萜化合物具有可观的药用价值和经济价值,但是目前的生产过程复杂、产量低,利用微生物异源合成三萜化合物已成为当前研究趋势,大肠杆菌作为常用萜类合成底盘细胞具有异源合成三萜化合物及其前体的天然优势和研究前景。对三萜化合物微生物异源合成研究进展进行了综述,从三萜化合物合成代谢途径、关键酶的特点及大肠杆菌三萜表达模块和底盘细胞适配三个方面对该途径进行了阐述和分析,针对实现大肠杆菌高效合成三萜类化合物所需要解决的基础问题进行讨论,为扩展大肠杆菌作为三萜化合物合成底盘细胞提供建议和前景分析。  相似文献

以不同灵芝品种、不同栽培基质、不同管理方式、不同生长时期获得的灵芝子实体为原料,用95%乙醇超声提取,对提取物进行了含量测定、三萜组成分析和体外抗肿瘤实验。结果表明,不同子实体醇提物中三萜和甾醇类物质的含量在4.60-6.20mg/g之间;高效液相分析发现10种三萜化合物的种类和含量在样品间存在明显差异。所有灵芝子实体醇提物对肿瘤细胞L1210的增殖均有一定的抑制作用,开伞期的灵芝子实体醇提物抑制肿瘤增殖的活性优于其他生长时期。  相似文献

Nineteen triterpenoids including two new tirucallane ones, fisuphanins A and B, were isolated from the whole plant of Euphorbia fischeriana Steud. A new 12-membered macrolide (1) along with two known C-31 and four known C-32 cycloartane triterpenoids (27) were isolated from the tubers of Bletilla striata. Their structures were determined by spectroscopic analysis, including 1D NMR, 2D NMR, and HRESIMS. The chemotaxonomic significance of these isolates, especially C-31 and C-32 cycloartane triterpenoids, are discussed. To our knowledge, this is the first report on the co-occurrence of the unusual 24-methyl, 24-ethyl, and 24,24-dimethyl cycloartane triterpenoids in the family Orchidaceae.  相似文献

为准确测定灵芝孢子粉中三萜的含量,运用高效液相建立适合孢子粉的分析测定方法。通过对前处理条件的优化,确定40%乙醇为孢子粉中等极性三萜酸类的最佳提取溶剂,浓缩倍数是子实体提取条件的50倍。通过色谱柱和洗脱条件的优化,建立了包括灵芝酸I、灵芝烯酸C、灵芝酸C2等13种标准品测定方法,方法学考察显示该分析方法精密度、重复性、稳定性的RSD值均小于5%,可以用于灵芝孢子粉中三萜类成分的定量检测。通过5组样品的分析发现,灵芝酸C6、灵芝酸G、灵芝酸A、灵芝酸D、灵芝酸F是灵芝孢子粉中的主要三萜类成分,其中灵芝酸A含量最高,平均占样品三萜总量的比例达19.71%;三萜类成分的溶出量与是否破壁没有相关性。三萜类成分在灵芝孢子粉和灵芝孢子油产品中的含量非常低,孢子粉的三萜含量为14.24-99.70μg/g,仅为子实体的1/100,灵芝孢子油中三萜含量也均低于50μg/g,因此三萜类成分不适合作为灵芝孢子粉及其相关产品的定量检测指标。  相似文献
