The electrophysiological responses of water, salt and sugar receptors in the labellar chemosenory hair of Drosophila were investigated. In contrast to the responses of large flies such as blowfly and fleshfly, spike height changed in parallel with the spike frequencies in all the three kind of receptors, and at the same time, the spike also changed in shape: when the receptor potential was small, the spike was small and biphasic, but when the receptor potential was large, the spike was large and monophasic. These phenomena are consistently explained by assuming that antidromic conduction of spikes in the distal process of the receptor cells is blocked due to inactivation of Na channels by the depolarizing receptor potential.  相似文献

An experimentally recorded time series formed by the exact times of occurrence of the neuronal spikes (spike train) is likely to be affected by observational noise that provokes events mistakenly confused with neuronal discharges, as well as missed detection of genuine neuronal discharges. The points of the spike train may also suffer a slight jitter in time due to stochastic processes in synaptic transmission and to delays in the detecting devices. This study presents a procedure aimed at filtering the embedded noise (denoising the spike trains) the spike trains based on the hypothesis that recurrent temporal patterns of spikes are likely to represent the robust expression of a dynamic process associated with the information carried by the spike train. The rationale of this approach is tested on simulated spike trains generated by several nonlinear deterministic dynamical systems with embedded observational noise. The application of the pattern grouping algorithm (PGA) to the noisy time series allows us to extract a set of points that form the reconstructed time series. Three new indices are defined for assessment of the performance of the denoising procedure. The results show that this procedure may indeed retrieve the most relevant temporal features of the original dynamics. Moreover, we observe that additional spurious events affect the performance to a larger extent than the missing of original points. Thus, a strict criterion for the detection of spikes under experimental conditions, thus reducing the number of spurious spikes, may raise the possibility to apply PGA to detect endogenous deterministic dynamics in the spike train otherwise masked by the observational noise.  相似文献

Patterns of carbon integration in aclonal species are poorly understood in spite of their potential to influence individual fitness. To provide more information about these patterns, we performed a defoliation experiment with P. aristata. We examined, at the metameric level, the reproductive responses to the removal of the major carbon sources within metamers. Bracts on marked reproductive spikes and leaves subtending these spikes were removed at three stages of reproductive maturity: spike elongation, flowering, and fruiting. Spike dry weight and length, capsule number, seeds per capsule, and seed weight were measured. We tested the hypothesis that seed weight would respond least to defoliation. We also performed a complementary 14C translocation experiment to measure the amount of radioactive carbon moving into the marked spikes from outside the metamer. Defoliation depressed all components of reproduction within marked spikes, and little 14C was translocated from outside the metamer into the reproductive spikes, even those that were defoliated. Both results support the view that reproductive metamers in this species are largely autonomous with respect to their carbon budget. Defoliation during spike elongation most depressed reproduction, and bract removal depressed reproduction more than did leaf removal. The data suggest that bracts compensate for leaf removal by increasing their photosynthetic rate; however, the ability to compensate differs among plant populations. Of all the reproductive components, seed weight was least affected by defoliation. The data show, however, that the time of defoliation relative to reproductive development influences which reproductive components are affected.  相似文献

Current neurophysiological research has the aim to develop methodologies to investigate the signal route from neuron to neuron, namely in the transitions from spikes to Local Field Potentials (LFPs) and from LFPs to spikes.LFPs have a complex dependence on spike activity and their relation is still poorly understood1. The elucidation of these signal relations would be helpful both for clinical diagnostics (e.g. stimulation paradigms for Deep Brain Stimulation) and for a deeper comprehension of neural coding strategies in normal and pathological conditions (e.g. epilepsy, Parkinson disease, chronic pain). To this aim, one has to solve technical issues related to stimulation devices, stimulation paradigms and computational analyses. Therefore, a custom-made stimulation device was developed in order to deliver stimuli well regulated in space and time that does not incur in mechanical resonance. Subsequently, as an exemplification, a set of reliable LFP-spike relationships was extracted.The performance of the device was investigated by extracellular recordings, jointly spikes and LFP responses to the applied stimuli, from the rat Primary Somatosensory cortex. Then, by means of a multi-objective optimization strategy, a predictive model for spike occurrence based on LFPs was estimated.The application of this paradigm shows that the device is adequately suited to deliver high frequency tactile stimulation, outperforming common piezoelectric actuators. As a proof of the efficacy of the device, the following results were presented: 1) the timing and reliability of LFP responses well match the spike responses, 2) LFPs are sensitive to the stimulation history and capture not only the average response but also the trial-to-trial fluctuations in the spike activity and, finally, 3) by using the LFP signal it is possible to estimate a range of predictive models that capture different aspects of the spike activity.  相似文献

Carex rainbowii (Cariceae, Cyperaceae), a new species from the Drakensberg mountains (KwaZulu-Natal province), is described and illustrated. It was found in the shady understory of the Afromontane forest in the Cathedral Peak area. An additional, nearby population was also identified based on previously collected herbarium material. Morphological and molecular (cpDNA 5′trnK intron and nrDNA ITS and ETS sequences) data were used to evaluate the taxonomic status of these populations and shed light on their systematic placement. Our data strongly support their taxonomic identity and inclusion in Carex sect. Sylvaticae. The new species can be readily distinguished from other related taxa mainly by the frequently androgynecandrous terminal spike, dense female spikes, hyaline glumes, as well as by some quantitative features. This finding implies a considerable biogeographic disjunction from the mainly Eurasian-North African range of the remaining species of sect. Sylvaticae, a pattern also found in the related sections Ceratocystis, Rhynchocystis and Spirostachyae. Comments are provided on previous misidentifications of C. rainbowii as the closely related Carex sylvatica. Data pertinent to the conservation status of the species are provided.  相似文献

The barley mutant, poly-row-and-branched spike (prbs) showed altered inflorescence morphology: complete conversion of the rudimentary lateral spikelets in two-rowed barley into fully developed fertile spikelets similar to the six-rowed phenotype, and additional spikelets in the middle of spike. Moreover, branched spikes emerged in progeny from a cross between the mutant and a six-rowed barley cultivar. Morphological observation of the development of immature spikes of the mutant and descendants with branched spikes showed that the Prbs gene is involved in spikelet development in the triple-mound stage. In mutant prbs, new meristems initiated at the flanks of lateral spikelets and middle spikelet meristems were converted to branch meristems, developing branched spikes. These observations suggested that the Prbs gene plays a crucial role in spikelet initiation and identity maintenance. The Prbs gene may be an important modifier in inflorescence differentiation from a panicle into a spike. The branched spikes emerging in hybrids from a cross between the mutant and six-rowed barley cultivar were not conferred by the gene vrs1 or Int-c, which decide spike morphology in six-rowed barley. These results imply that although six-row genes vrs1 and Int-c and prbs have similar effects on lateral spikelet development, they have different functions in branched spikes. The Prbs gene was mapped to chromosome 3H between SSR marker Bmag0023 and marker Cbic60 at a genetic distance of 3.3 and 5.4 centimorgans (cM), respectively.  相似文献

The influence of neural morphology and passive electrical parameters on the width and amplitude of extracellular spikes is investigated by combined analytical and numerical investigations of idealized and anatomically reconstructed pyramidal and stellate neuron models. The main results are: 1), All models yield a low-pass filtering effect, that is, a spike-width increase with increasing distance from soma. 2), A neuron's extracellular spike amplitude is seen to be approximately proportional to the sum of the dendritic cross-sectional areas of all dendritic branches connected to the soma. Thus, neurons with many, thick dendrites connected to soma will produce large amplitude spikes, and therefore have the largest radius of visibility. 3), The spike shape and amplitude are found to be dependent on the membrane capacitance and axial resistivity, but not on the membrane resistivity. 4), The spike-amplitude decay with distance r is found to depend on dendritic morphology, and is decaying as 1/rn with 1 ≤ n ≤ 2 close to soma and n ≥ 2 far away.  相似文献

A study of the negative phase of the spikes recorded extra cellularly from insect mechanoreceptor has been performed in order to characterize some electrical properties of the dendrite which contains the transducing part of the sensory neuron. These properties have been investigated in mechanoreceptors of the metathoracic leg of the locust Schistocerca gregaria by firing antidromic action potentials both at rest and during mechanical or electrical stimulation. The amplitude of the negative phase of the spike appears to be correlated with the polarization of the dendritic membrane, although when bursts of action potentials are applied, the relation is more complex, including a depressive influence of a given spike on the following spike. The receptor potential and the antidromic dendritic spikes both originate in the same region of the dendrite but they involve different ionic processes. Our results indicate that the dendrite is electrically excitable. The spike which originates in the dendrite has an initial negative phase with a small superimposed positive component. A spike of this shape is never observed under natural stimulation. It is proposed that the negative phase of the antidromic impulse provides a suitable means for studying the variations in electrical polarization of the dendrite which cannot be recorded directly.  相似文献

Sensory neurons code information about stimuli in their sequence of action potentials (spikes). Intuitively, the spikes should represent stimuli with high fidelity. However, generating and propagating spikes is a metabolically expensive process. It is therefore likely that neural codes have been selected to balance energy expenditure against encoding error. Our recently proposed optimal, energy-constrained neural coder (Jones et al. Frontiers in Computational Neuroscience, 9, 61 2015) postulates that neurons time spikes to minimize the trade-off between stimulus reconstruction error and expended energy by adjusting the spike threshold using a simple dynamic threshold. Here, we show that this proposed coding scheme is related to existing coding schemes, such as rate and temporal codes. We derive an instantaneous rate coder and show that the spike-rate depends on the signal and its derivative. In the limit of high spike rates the spike train maximizes fidelity given an energy constraint (average spike-rate), and the predicted interspike intervals are identical to those generated by our existing optimal coding neuron. The instantaneous rate coder is shown to closely match the spike-rates recorded from P-type primary afferents in weakly electric fish. In particular, the coder is a predictor of the peristimulus time histogram (PSTH). When tested against in vitro cortical pyramidal neuron recordings, the instantaneous spike-rate approximates DC step inputs, matching both the average spike-rate and the time-to-first-spike (a simple temporal code). Overall, the instantaneous rate coder relates optimal, energy-constrained encoding to the concepts of rate-coding and temporal-coding, suggesting a possible unifying principle of neural encoding of sensory signals.  相似文献

Human astroviruses (HAstVs) are a major cause of gastroenteritis. HAstV assembles from the structural protein VP90 and undergoes a cascade of proteolytic cleavages. Cleavage to VP70 is required for release of immature particles from cells, and subsequent cleavage by trypsin confers infectivity. We used electron cryomicroscopy and icosahedral image analysis to determine the first experimentally derived, three-dimensional structures of an immature VP70 virion and a fully proteolyzed, infectious virion. Both particles display T = 3 icosahedral symmetry and nearly identical solid capsid shells with diameters of ~ 350 Å. Globular spikes emanate from the capsid surface, yielding an overall diameter of ~ 440 Å. While the immature particles display 90 dimeric spikes, the mature capsid only displays 30 spikes, located on the icosahedral 2-fold axes. Loss of the 60 peripentonal spikes likely plays an important role in viral infectivity. In addition, immature HAstV bears a striking resemblance to the structure of hepatitis E virus (HEV)-like particles, as previously predicted from structural similarity of the crystal structure of the astrovirus spike domain with the HEV P-domain [Dong, J., Dong, L., Méndez, E. &; Tao, Y. (2011). Crystal structure of the human astrovirus capsid spike. Proc. Natl. Acad. Sci. USA 108, 12681–12686]. Similarities between their capsid shells and dimeric spikes and between the sequences of their capsid proteins suggest that these viral families are phylogenetically related and may share common assembly and activation mechanisms.  相似文献

Neurons encode information in sequences of spikes, which are triggered when their membrane potential crosses a threshold. In vivo, the spiking threshold displays large variability suggesting that threshold dynamics have a profound influence on how the combined input of a neuron is encoded in the spiking. Threshold variability could be explained by adaptation to the membrane potential. However, it could also be the case that most threshold variability reflects noise and processes other than threshold adaptation. Here, we investigated threshold variation in auditory neurons responses recorded in vivo in barn owls. We found that spike threshold is quantitatively predicted by a model in which the threshold adapts, tracking the membrane potential at a short timescale. As a result, in these neurons, slow voltage fluctuations do not contribute to spiking because they are filtered by threshold adaptation. More importantly, these neurons can only respond to input spikes arriving together on a millisecond timescale. These results demonstrate that fast adaptation to the membrane potential captures spike threshold variability in vivo.  相似文献

Space-clamped squid axons treated with low calcium and computed Hodgkin-Huxley (HH) axons were stimulated by steps of superthreshold current from 101 to 400% of the rheobasic value over a temperature range of 5–27°C. The natural frequency of sustained repetitive firing of real and computed axons depended weakly upon stimulus intensity and strongly upon temperature, with a Q10 of 2.7 (experimental) and 2.6 (computed). For real axons, but not the computed axon, the intervals between the first two spikes were shorter than between subsequent spikes. Constant spike frequencies from 75 Hz at low intensities and temperatures to 330 Hz at high intensities and temperatures were soon achieved. Subthreshold and superthreshold responses were sometimes intermixed in a train of responses from a real axon responding to a constant step of current, but not predicted by HH. The time interval following a spike was always longer than that following a subthreshold oscillation in slightly decalcified real axons, as Huxley and FitzHugh also found for computed axons. There was a bias toward spikes at the beginning of the train and toward subthreshold responses later on. Some repeated patterns were found, every second, third, or fourth response being a spike. Neither the HH equations nor the computed or experimental threshold behaviors show a critical temperature to support a membrane phase transition.  相似文献

The development and morphology of the wheat spike is important because the spike is where reproduction occurs and it holds the grains until harvest. Therefore, genes that influence spike morphology are of interest from both theoretical and practical stand points. When substituted for the native chromosome 2A in the tetraploid Langdon (LDN) durum wheat background, the Triticum turgidum ssp. dicoccoides chromosome 2A from accession IsraelA confers a short, compact spike with fewer spikelets per spike compared to LDN. Molecular mapping and quantitative trait loci (QTL) analysis of these traits in a homozygous recombinant population derived from LDN × the chromosome 2A substitution line (LDNIsA-2A) indicated that the number of spikelets per spike and spike length were controlled by linked, but different, loci on the long arm of 2A. A QTL explaining most of the variation for spike compactness coincided with the QTL for spike length. Comparative mapping indicated that the QTL for number of spikelets per spike overlapped with a previously mapped QTL for Fusarium head blight susceptibility. The genes governing spike length and compactness were not orthologous to either sog or C, genes known to confer compact spikes in diploid and hexaploid wheat, respectively. Mapping and sequence analysis indicated that the gene governing spike length and compactness derived from wild emmer could be an ortholog of the barley Cly1/Zeo gene, which research indicates is an AP2-like gene pleiotropically affecting cleistogamy, flowering time, and rachis internode length. This work provides researchers with knowledge of new genetic loci and associated markers that may be useful for manipulating spike morphology in durum wheat.  相似文献

Responses of the antennal thermosensitive neuron of the ground beetle Platynus assimilis to warming from 20 to 50 °C were measured and analysed. During warming, neurons switched from regular spiking to bursting. ISI analysis showed that the number of spikes in the burst and spike frequency within the burst were temperature dependent and may precisely encode unfavourably or dangerously high temperatures in a graded manner. In contrast, regular spikes of the neuron encode moderate temperatures at 20-30 °C. The threshold temperature of spike bursting varied in different neurons from 25 to 47 °C. As a result, the number of bursting neurons increased with temperature increase. Therefore, in addition to the burst characteristics, the total number of bursting neurons may also contain useful information on external temperature. A relationship between the spike bursts and locomotor activity of the beetles was found which may have importance in behavioural thermoregulation of the species. At 44.4 ± 0.6 °C, first indications of partial paralysis (of the hind legs) were observed. We emphasize, that in contrast to various sensory systems studied, the thermoreceptor neuron of P. assimilis has a stable and continuous burst train, no temporal information is encoded in the timing of the bursts.  相似文献

Wheat grain yield consists of three components: spikes per plant, grains per spike (i.e. head or ear), and grain weight; and the grains per spike can be dissected into two subcomponents: spikelets per spike and grains per spikelet. An increase in any of these components will directly contribute to grain yield. Wheat morphology biology tells that a wheat plant has no lateral meristem that forms any branching shoot or spike. In this study, we report two novel shoot and spike traits that were produced from lateral meristems in bread wheat. One is supernumerary shoot that was developed from an axillary bud at the axil of leaves on the elongated internodes of the main stem. The other is supernumerary spike that was generated from a spikelet meristem on a spike. In addition, supernumerary spikelets were generated on the same rachis node of the spike in the plant that had supernumerary shoot and spikes. All of these supernumerary shoots/spikes/spikelets found in the super wheat plants produced normal fertility and seeds, displaying huge yield potential in bread wheat.  相似文献

BackgroundRecent findings have shown that imaging voluntarily activated motor units (MUs) by decomposing ultrasound-based displacement images provides estimates of unfused tetanic signals evoked by spinal motoneurons’ neural discharges (spikes). Two methods have been suggested to estimate its spike trains: band-pass filter (BPM) and Haar wavelet transform (HWM). However, the methods’ optimal parameters and which method performs the best are unknown. This study will answer these questions.MethodHWM and BPM were optimized using simulations. Their performance was evaluated based on simulations and 21 experimental datasets, considering their rate of agreement, spike offset, and spike offset variability to the simulated or experimental spikes.ResultsA range of parameter sets that resulted in the highest possible agreement with simulated spikes was provided. Both methods highly agreed with simulated and experimental spikes, but HWM was a better spike estimation method than BPM because it had a higher agreement, less bias, and less variation (p < 0.001).ConclusionsThe optimized HWM will be an important contributor to further developing the identification and analysis of MUs using imaging, providing indirect access to the neural drive of the spinal cord to the muscle by the unfused tetanic signals.  相似文献

The ionic mechanism underlying optimal stimulus shapes that induce a neuron to fire an action potential, or spike, is relevant to understanding optimal information transmission and therapeutic stimulation in the nervous system. Here we analyze for the first time the ionic basis for stimulus optimality in the Hodgkin and Huxley model and for eliciting a spike in squid giant axons, the preparation for which the model was devised. The experimentally determined stimulus is a smoothly varying biphasic current waveform having a relatively long and shallow hyperpolarizing phase followed by a depolarizing phase of briefer duration. The hyperpolarizing phase removes a small degree of the resting level of Na + channel inactivation. This result together with the subsequent depolarizing phase provides a signal that is energetically more efficient for eliciting spikes than rectangular current pulses. Sodium channel inactivation is the only variable that is changed during the stimulus waveform, other than the membrane potential, V. The activation variables for Na + and K + channels are unchanged throughout the stimulus. This result demonstrates how an optimal stimulus waveform relates to ionic dynamics and may have implications for energy efficiency of neural excitation in many systems including the mammalian brain.  相似文献
