The influence of iron, aluminium and of the combined application of both metals on microbial biomass and production of siderophores by three fungi (Aspergillus nidulans, Neurospora crassa and Hymenoscyphus ericae) were investigated. All three species showed a strong iron regulation and Al-sensitivity of siderophore biosynthesis although several differences remained species dependent. Inhibitory effects of Fe and Al on siderophore-production were additive and the higher binding capacity of siderophores towards iron could be compensated by a higher Al-availability. Although pH itself is also important for regulation of siderophore biosynthesis, an indirect effect of Al on siderophore production via an Al-induced pH decrease could be outlined. The toxic effects of Al resulting in a reduced biomass production were compensated by high Fe-availability, whereas the addition of DFAM, a bacterial siderophore, enhanced Al-toxicity.  相似文献

The photo-induced toxicity of anthracene was investigated as the mortality in Antarctic shallow water amphipod, Gondogeneia antarctica, at different concentrations of anthracene and different periods of exposure to natural sunlight and artificial UVA and UVB radiations. When exposed to natural sunlight, animals contaminated in the dark and placed in clean water or in anthracene solutions showed different degrees of mortality, dose–time dependent. Effects were even more evident when these animals were exposed to artificial UVA or UVB radiations. Depuration seemed to be a slow process. The effects of UV radiation and anthracene alone and the effects of the interactions of these two stressors implied that solar radiation is an important parameter that deserves consideration in the environmental assessment of polycyclic aromatic hydrocarbons in Antarctic coastal waters. G. antarctica proved to be a good bioindicator for the phototoxicity of anthracene in Antarctic shallow waters.  相似文献

A distinct inhibition of the growth of a typical soil-born bacterium, Arthrobacter sp. PI/1-95, by aluminium was established. The content of intracellular water and (consequently) the cell size of the bacterium significantly increased when cells came into contact with aluminium. Both parameters were also increased when applying HCl, but Al caused the greater change, so that a distinct discrimination between the effects of aluminium on the one hand and a sole pH-effect on the other hand was possible. The presented results point to an osmo-regulative disorder connected with a malfunction of cell membrane and cell wall, which seems to be a probable mechanism for an Al-toxicity in connection with Arthrobacter sp. A solution culture experiment was carried out to study the effects of interactions between aluminium (Al) and phosphorus (P) on Al-toxicity under conditions of suboptimal P supply. The experiment was conducted in a growth chamber with seedlings of the Al-sensitive sorghum genotype TAM428 (Sorghum bicolor (L.) Moench). Phosphorus deficiency differed from Al toxicity in its effect on shoot/root ratio and root morphological charateristics. Results indicated that there were positive effects of Al on the uptake and assimilation of P. Therefore, it was unlikely that an Al-induced P deficiency could account for the observed reduction in plant biomass. Plants suffered more from Al toxicity at very low P supply. Moreover, decreasing P supply resulted in increased root H-ion efflux density. In the soil, where a rhizosphere can be formed, this would make the plant even more susceptible to Al. Dry matter yield of the plants was affected more severely by Al at the first harvest (14 days) than at the second (35 days), but the opposite was true for P. Aluminium-inhibited root development and reduced uptake of N, K and Mg (but not Ca) may be partly responsible for the growth depression. Increasing the P supply exerted certain roles in eliminating Al phytotoxicity, possibly through improved root development and nutrient uptake. The detrimental influence of Al on biomass could be overcome by doubling the P supply.  相似文献

Fitness‐related traits are often affected by temperature. Heat‐resistant genotypes could influence the dependence of fitness traits on temperature, which should be important in adaptation to directional changes in temperature including global warming. Here, we tested temperature‐dependent variation in longevity and fecundity between Drosophila melanogaster Meigen (Diptera: Drosophilidae) genotypes that differ in heat‐resistance QTL. Longevity and fecundity were affected by heat‐resistance genotypes at constant moderate and high temperature. However, these differences between heat‐resistant and heat‐sensitive genotypes disappeared in a cyclic thermal regime. Analysis with the logistic mortality function indicated that mortality patterns are dependent on temperature and genotype. The results suggest that genotype*temperature interactions are substantial for senescence‐related traits. In particular, fluctuating temperatures can drastically reduce any differences in life‐history traits between heat‐resistance genotypes, even if such genotypes differentially affect the traits at constant temperatures.  相似文献

Abstract. The present experiments are part of a larger study designed to investigate the influence of husbandry parameters on the life history of the ramshorn snail, Marisa cornuarietis, in order to identify suitable husbandry conditions for maintaining multi‐generation populations in the laboratory for use in ecotoxicological testing. In this paper we focus on the effects of a combination of food types and feeding frequencies (i.e., the frequency with which the snails were offered food) on juvenile growth and survival at different temperatures. Offspring produced in the laboratory by wild specimens of M. cornuarietis, from Puerto Rico, were used to test the effects of three types of food (lettuce, alginate with fish food, alginate with snail mix) fed at three frequencies (given ad libitum on 4/4, 2/4, or 1/4 d) on juvenile survival and growth. The 4‐d feeding regimens were repeated four times, giving a total of 16 d for the experiments. The experiments were conducted at two temperatures (22° and 25°C) under a 12 h light:12 h dark photoperiod. Juvenile growth rates increased with increasing feeding frequency for all food types. The most rapid growth rates occurred in the high‐frequency lettuce treatments and the slowest growth rates in the low‐frequency lettuce and alginate with snail mix treatments. Juvenile snails grew faster at 25° than at 22°C, and mortality was about twice as high at the lower temperature. Growth rates were used to provide a rough estimate of time to maturity, which was determined to take about twice as long at 22° than at 25°C. The results showed that lettuce is the best food if supplied in abundance, but effects on growth are very dependent on feeding frequency and temperature. We conclude that 25°C is a more appropriate temperature for maintaining populations than 22°C, that lettuce provides a suitable food source, and that food should be supplied continuously for husbandry and toxicity testing of populations of M. cornuarietis.  相似文献

The adverse effects of high temperatures on the early life stages of anadromous whitefish Coregonus lavaretus were experimentally examined by assessing fertilization success, the percentage of developmental abnormalities, cumulative mortality and the rate of embryogenesis across a range of temperatures. Temperatures ≥ 7° C increased the proportion of unfertilized and abnormally dividing eggs, deformed embryos and consequent mortality. The higher the temperature, the more severe were the effects. When eggs were fertilized and constantly incubated at various temperatures, the effective level for 50% of the eggs and embryos (EL50) of temperature was 7·6° C at the developmental stage when eye pigmentation was visible. Fewer developmental abnormalities and a lower cumulative mortality rate were observed when embryos were exposed to high temperatures from the later, gastrula stage, than from fertilization or the four‐cell stage. Irrespective of retarded development in terms of day‐degrees (i.e. the sum of daily mean temperatures), a high incubation temperature reduced the development time of C. lavaretus, leading to earlier hatching, and hatched fry were shorter than at the reference temperature of 4–5° C. Global warming will particularly pose risks for stenothermic species such as C. lavaretus, with early life stages being especially susceptible. Thus, relatively small increases and fluctuations in river water temperatures during the spawning season of this anadromous species may have substantial negative impacts on its recruitment and population persistence.  相似文献

The effects of highly and moderately acid soils on total biomass, biomass partitioning, fine root characteristics and nutritional status of beech seedlings (Fagus sylvatica L.) were studied in a growth chamber experiment. In Haplic Arenosols seedlings grew slowly but equally well without damage symptoms in a highly acid and a moderately acid soil horizon. The moderately acid Ah+Bw-horizon of a Eutric Cambisol was favourable to seedling growth. The fine root development was reduced in the highly acid A+Bw-horizon of a Dystric Cambisol and in the Ah+E-horizon of a Haplic Podzol, the latter of which also caused increased mortality. Seedling growth in the B2-horizon of the Haplic Podzol was vigorous, in spite of a higher level of extractable Al and lower base saturation as compared with the Ah+E-horizon. These results are interpreted in relation to soil acidity, soil Al and nutritional status of the seedlings. We conclude that neither Al-toxicity nor nutrient deficiency cause the damage symptoms observed in the Ah+E-horizon of a Haplic Podzol and the fine root reduction in the A+Bw-horizon of a Dystric Cambisol. The damage symptoms of the PZhA treatment seems to be more the result of H-toxicity or H-related factors other than nutrient shortage or Al-toxicity. Climate change is the most significant stressor that is anticipated increasingly to affect human and global ecosystems. Arthropods, including insects, are particularly vulnerable to global warming and this group is often used for various ecotoxicological tests. In addition, temperature is one of the most important toxicity‐modifying factors in ecotoxicology. Therefore, temperature dependent toxicological research is required to obtain ecologically relevant conclusions during the current era of rapid climate change. This study shows that two midge species (Chironomus riparius Meigen and C. yoshimatsui Martin et Sublette) exhibit different developmental characteristics and responses to cadmium and lead heavy metals with temperature. The former species is an internationally standardized test species in ecotoxicological studies, whereas the latter species is native to Korea. Hence, even though these two species belong to the same genus, Chironomus, their development differs with temperature, which leads to different responses to heavy metals. There was a decline in developmental time (from egg and larva to pupa) for both species with temperature; however, there was a species difference in the rate of decline. In the acute toxicity test, the 48‐hr LC50 values for cadmium and lead decreased with temperature for both species. In the chronic toxicity test, emergence rates tended to decrease with temperature, except for when C. yoshimatsui was exposed to cadmium.  相似文献

Aluminum is an abundant metal in the earth’s crust that turns out to be toxic in acidic environments. Many plants are affected by the presence of aluminum at the whole plant level, at the organ level, and at the cellular level. Tobacco as a cash crop (Nicotiana tabacum L.) is a widely cultivated plant worldwide and is also a good model organism for research. Although there are many articles on Al-phytotoxicity in the literature, reviews on a single species that are economically and scientifically important are limited. In this article, we not only provide the biology associated with tobacco Al-toxicity, but also some essential information regarding the effects of this metal on other plant species (even animals). This review provides information on aluminum localization and uptake process by different staining techniques, as well as the effects of its toxicity at different compartment levels and the physiological consequences derived from them. In addition, molecular studies in recent years have reported specific responses to Al toxicity, such as overexpression of various protective proteins. Besides, this review discusses data on various organelle-based responses, cell death, and other mechanisms, data on tobacco plants and other kingdoms relevant to these studies.  相似文献

The toxicity and anticholinesterase activity of tremorgenic fungal metabolites, territrems, to the corn earworm, Helioverpa zea (Boddie) (Lepidoptera, Noctuidae) were examined. In oral toxicity studies, territrem A significantly inhibited growth by 40% at 25 ppm and by 89% at 250 ppm. Territrem B and an epoxy-derivative significantly inhibited growth by ca. 45% at 250 ppm. Piperonyl butoxide administered orally synergized the toxicity of the territrems tested. In topical toxicity studies, the epoxy-derivative caused 26% mortality and 25% growth retardation at 10 mg/gm insects. Territrem A and B were not significantly lethal, but did reduce growth by 15–20% at 10 mg/gm insect. Paraoxon tested in the same way caused 100% mortality at 25 ppm orally and 10 mg/gm topically. However, all territrems tested in vitro as inhibitors of H. zea head acetylcholinesterase were at least comparable to or were more active than paraoxon. Topically administered epoxy-territrem B also inhibited H. zea head acetylcholinesterase. The potential for development of new insecticides from a territrem lead structure is discussed.  相似文献

Natural reproduction of pallid sturgeon Scaphirhynchus albus has been limited for decades and a recruitment bottleneck is hypothesized to occur during the larval stage of development. In this study, we evaluated the effects of water velocity and temperature on the swimming activity, energy use, settling behaviour and mortality of endogenously feeding larvae. The swimming activity of drifting sturgeon larvae (i.e., fish exhibiting negative rheotaxis) increased at low water velocity. In subsequent experiments, we observed greater energy depletion and resultant mortality of larvae in no-flow environments (0 cm s−1) compared to tanks with water velocity ranging from 3.5 to 8.3 cm s−1. The growth rate of drifting larvae was positively related to water temperature (18.7–23.3°C), but reduced growth rate at low water temperature (18.7°C) resulted in protracted development that extended average drift duration by ~4 days compared to larvae reared at 23.3°C. This study provides evidence that cooler summer water temperatures, characteristic of present-day conditions in the upper Missouri River, can reduce larval development and extend both the drift duration and distance requirements of S. albus. Moreover, if dispersed into low velocity environments, such as in reservoir headwaters, larvae may experience increased mortality owing to a mismatch between early life stage drift requirements and habitat conditions in the river. Manipulation of water releases to increase seasonal water temperature below dams may aid survival of S. albus larvae by shortening the time and distance spent drifting.  相似文献

The aim of this study was to determine the suitability of water quality in the Roanoke River of North Carolina for supporting shortnose sturgeon Acipenser brevirostrum, an endangered species in the United States. Fathead minnows Pimephales promelas were also evaluated alongside the sturgeon as a comparative species to measure potential differences in fish survival, growth, contaminant accumulation, and histopathology in a 28‐day in situ toxicity test. Captively propagated juvenile shortnose sturgeon (total length 49 ± 8 mm, mean ± SD) and fathead minnows (total length 39 ± 3 mm, mean ± SD) were used in the test and their outcomes were compared to simultaneous measurements of water quality (temperature, dissolved oxygen, pH, conductivity, total ammonia nitrogen, hardness, alkalinity, turbidity) and contaminant chemistry (metals, polycyclic aromatic hydrocarbons, organochlorine pesticides, current use pesticides, polychlorinated biphenyls) in river water and sediment. In the in situ test, there were three non‐riverine control sites and eight riverine test sites with three replicate cages (25 × 15‐cm (OD) clear plexiglass with 200‐μm tear‐resistant Nitex® screen over each end) of 20 shortnose sturgeon per cage at each site. There was a single cage of fathead minnows also deployed at each site alongside the sturgeon cages. Survival of caged shortnose sturgeon among the riverine sites averaged 9% (range 1.7–25%) on day 22 of the 28‐day study, whereas sturgeon survival at the non‐riverine control sites averaged 64% (range 33–98%). In contrast to sturgeon, only one riverine deployed fathead minnow died (average 99.4% survival) over the 28‐day test period and none of the control fathead minnows died. Although chemical analyses revealed the presence of retene (7‐isopropyl‐1‐methylphenanthrene), a pulp and paper mill derived compound with known dioxin‐like toxicity to early life stages of fish, in significant quantities in the water (251–603 ng L?1) and sediment (up to 5000 ng g?1 dry weight) at several river sites, no correlation was detected of adverse water quality conditions or measured contaminant concentrations to the poor survival of sturgeon among riverine test sites. Histopathology analysis determined that the mortality of the river deployed shortnose sturgeon was likely due to liver and kidney lesions from an unknown agent(s). Given the poor survival of shortnose sturgeon (9%) and high survival of fathead minnows (99.4%) at the riverine test sites, our study indicates that conditions in the Roanoke River are incongruous with the needs of juvenile shortnose sturgeon and that fathead minnows, commonly used standard toxicity test organisms, do not adequately predict the sensitivity of shortnose sturgeon. Therefore, additional research is needed to help identify specific limiting factors and management actions for the enhancement and recovery of this imperiled fish species.  相似文献

There has been no consensus on whether Prorocentrum minimum is “toxic,” despite sporadic reports suggesting possible shellfish toxicity and laboratory studies showing harmful effects of this dinoflagellate on molluscan shellfish. Shellfish toxicity outbreaks associated with natural blooms of P. minimum have been confounded by co-occurrence of other toxic phytoplankton. Laboratory studies have demonstrated unequivocally that some P. minimum isolates can produce toxins that kill mice on injection, but the bioactive compound or compounds remain unidentified, and accumulation of toxin in grazing mollusks has not been demonstrated. Laboratory experiments testing the responses of grazing mollusks to P. minimum cultures have yielded variable results, ranging from mortality in scallops and oysters to normal growth of oysters. Effects observed in the laboratory include rejection as pseudofeces by clams, poor larval development in oysters, tissue pathologies (sometimes transient) in oysters and scallops, and systemic immune responses in oysters and scallops. Several recent studies have provided evidence that variation in toxicity of P. minimum is dependent on environmental conditions and their effects on the physiology of this dinoflagellate. Accordingly, seemingly conflicting observations from field and laboratory studies may be explained by transient toxin expression in P. minimum.  相似文献

Osteochondral defects can degenerate into osteoarthritis and currently there are no good treatment alternatives available to most Orthopaedic surgeons. Osteochondral allografting can restore damaged joint surfaces but its clinical use is limited by poor access to high quality tissue. Vitrification of osteochondral tissue would allow the banking of this tissue but requires high concentrations of cryoprotective agents. This study was designed to ascertain dimethyl sulfoxide (DMSO) toxicity kinetics to chondrocytes in situ after exposure to DMSO at different temperatures recorded as a function of time. Porcine osteochondral dowels were exposed to 1, 3, 5, and 6M DMSO at 4, 22, and 37°C for 0.5 min to 120 min. Chondrocyte recovery was determined by membrane integrity (Syto 13 and ethidium bromide) and mitochondrial (WST-1) assays. Results demonstrated that cell recovery was concentration, temperature and time dependent. At higher concentrations and temperatures, significant cell loss occurred within minutes. A rate constant calculated for chondrocyte death was dependent on temperature. 1 M DMSO appeared relatively non-toxic. This experiment established a method to examine systematically toxicity parameters for chondrocytes in situ and this data can be used to tailor vitrification protocols by limiting exposure temperature and time or lowering DMSO concentrations below toxic levels recorded.  相似文献

A 40-day culture experiment of water hyacinth was made in 4 different water temperatures, 15, 20, 25 and 30°C, which were combined with 4 levels of concentration of culture solution, 1/3, 1, 3 and 9-fold of the standard solution containing 28 ppm of totalN and 7.7 ppm of totalP. The optimum condition for obtaining the maximum plant growth shifted from 30°C: 3-fold condition in the early stage to 20–25°C: 3-fold condition in the later stages of the experiment. The relation between the fresh weight biomass per 100-l tank,w, and the concentration of culture solution,f, was expressed successfully by a reciprocal equation,1/w=A F/f+A F f/(1-f/C F)+B F, in whichA f,A f′, andB f are time dependent coefficients andC f is the upper limit of the concentration to permit plant growth which can change with time. 1. The glacial relict, Mysis relicta, which occurs mainly in unproductive cold water habitats, shows increased mortality rates in temperatures above 16–18 °C. Using a 12‐year data set the growth, mortality and fat stores of Mysis in Lough Neagh, an increasingly hypertrophic lake which lacks a thermal refuge and which is subject to warming, were investigated. 2. Mysis showed a significant reduction in fat stores on reaching sexual maturity, contrary to studies elsewhere. 3. The contributions of temperature and eutrophication (total phosphorus, chlorophyll‐a concentrations) to growth, mortality and the timing and duration of developmental events were investigated by regression. 4. Water temperatures rose between 1994 and 2005 and the time when temperatures exceeded 16 °C doubled over this period. Juvenile and adult growth rates declined and the duration of the juvenile stage increased in warmer years. Eutrophication also affected these variables but, in general, to a lesser extent than temperature. 5. Year class strength (YCS) declined by a factor of 10 over the study period. YCS was independent of temperature up to 2000 °C degree days (>4 °C) but declined steeply above that point. Temperature was more important than eutrophication in predicting mortality and YCS. YCS was positively correlated with the length of time water temperature was below 7 °C, the upper temperature for Mysis breeding. 6. An established bioenergetic model predicted that water temperature and the timing of embryo release would have appreciable effects on Mysis viability. It also indicated that Mysis would lose mass at summer temperatures, consistent with the observed fat decline.  相似文献

In vivo historical control data, including mortality, body weight, food and water consumption, and clinical signs in F344/DuCrj rats were obtained from 11 long-term toxicity and carcinogenicity studies conducted at the Biosafety Research Center, Foods, Drugs and Pesticides, (An-Pyo Center) during the last five years. Survival at 109 weeks of age was 80.2% (min: 74%, max: 90%) in males and 80.5% (min: 72%, max: 92%) in females. The maximum mean body weights of males and females were 443.3 +/- 15.8 g (mean +/- S. D.) and 295.7 +/- 13.3 g respectively. Male rats attained their maximum body weight at 82.6 +/- 5.3 weeks of age, the females at 103.5 +/- 2.5 weeks of age. Clinical symptoms increased with age, particularly after 84 weeks of age, and included: wasting, piloerection, palpable subcutaneous and abdominal masses, and decreased spontaneous movement. Lowered body temperature and auricular pallor occurred commonly in moribund animals. The nature and grade of toxicity in the treated animals were generally disclosed by comparing with the behavior and signs in the control animals. The use of in-house, historical control data can be useful in subsequent evaluations of chronic toxicity and carcinogenicity studies.  相似文献

Selective mortality within a population, based on the phenotype of individuals, is the foundation of the theory of natural selection. We examined temperature-induced shifts in the relationships among early life history traits and survivorship over the embryonic and larval stages of a tropical damselfish, Pomacentrus amboinensis. Our experiments show that temperature determines the intensity of selective mortality, and that this changes with ontogeny. The size of energy stores determined survival through to hatching, after which egg size became a good indicator of fitness as predicted by theoretical models. Yet, the benefits associated with egg size were not uniform among test temperatures. Initial egg size positively influenced larval survival at control temperature (29 °C). However, this embryonic trait had no effect on post-hatching longevity of individuals reared at the higher (31 °C) and lower (25 °C) end of the temperature range. Overall, our findings indicate that the outcome of selective mortality is strongly dependent on the interaction between environment conditions and intrinsic developmental schedules.  相似文献

Environmental temperature is an important determinant of physiological processes and life histories in ectotherms. Over latitudinal scales, variation in temperature has been linked to changes in life-history traits and demographic rates, with growth and mortality rates generally being greatest at low latitudes, and longevity and maximum length being greater at higher latitudes. Using the two-spined angelfish, Centropyge bispinosa, as our focal species, we compared growth patterns, growth rates, longevity, mortality, asymptotic length and maximum length across 22 reefs that span 13° of latitude within the Great Barrier Reef Marine Park (GBRMP) and the Coral Sea Marine Park (CSMP), Australia. We found no predictable latitudinal variation in mortality rates, growth patterns, growth rates, asymptotic or maximum length of C. bispinosa at regional to biogeographic scales. However, C. bispinosa consistently exhibited reduced longevity at lower, warmer latitudes within the CSMP. The greatest differences in mean maximum length of C. bispinosa were between continental (GBRMP) and oceanic (central CSMP) reefs of similar latitude, with individuals being larger on average on continental versus oceanic reefs. The lack of predictable life-history and demographic variation in C. bispinosa across a 13° latitudinal gradient within the CSMP, coupled with differences in mean maximum length between continental and oceanic reefs at similar latitudes, suggest that local environmental conditions have a greater influence than environmental temperature on the demographic rates and life-history traits of C. bispinosa.  相似文献
