Large quantities of cassava common mosaic virus (CCMV) were purified from systemically infected Nicotiana benthamiana plants. A polyclonal antiserum, with a titre of 1/128 in the tube precipitin test, was produced by immunising rabbits with purified virus. Viral antigens were detected in cassava, using both the double-antibody sandwich or plate-trapped antigen forms of enzyme-linked immunosorbent assay (ELISA). The virus reacted with antisera to the potexviruses potato virus X and tulip virus X in F(ab')2 ELISA. As determined by ELISA, isolates of CCMV from cassava and chaya are closely serologically related to each other. Leaf extracts from infected N. benthamiana plants were infective to a dilution of 10--4 but not 10--5; after heating for 10 min at 65 °C but not 70 °C; and after storage at room temperature for 14 days. The virus has a sedimentation coefficient of 126 S20,w, a single coat protein molecule of c . mol. wt 21 000, and a single-stranded RNA genome of c . mol. wt 2.0 ± 106. Several dsRNA species, including the putative viral replicative form of c . mol. wt 4.1 ± 106, were isolated from virus-infected cassava and N. benthamiana .  相似文献

Aflatoxin content and number of fungi in poultry feedstuffs from Indonesia   总被引:1,自引:1,他引:0  
The content of aflatoxin and associated fungi was determined in 56 samples, including 34 of corn, 10 of soybean meal, nine of rice bran and three of broken rice, collected from different poultry farms and poultry feedmills situated around Jakarta-Bogor, Indonesia.
Ninety-one per cent of the corn samples contained aflatoxins and the total concentration ranged from 22 to 6171 μ g/kg. With rice bran, 100% of the samples were positive for aflatoxin B1, ranging from 36 to 71 μ g/kg. No aflatoxin was detected in samples of soybean meal or broken rice. All the samples were contaminated by several fungi (8 times 103–5 times 106cfu/g) and further identification was limited to Aspergillus flavus and A. parasiticus. The dominant species was A. flavus (2 times 103–4 times 106cfu/g in corn samples, 1·0 times 103–1·0 times 105cfu/g in soybean meal, 2 times 104–4·4 times 105cfu/g in rice bran and 2 times 104–6 times 104cfu/g in broken rice). A virus obtained from naturally infected lucerne ( Medicago sativa ) in New Zealand reacted with antiserum to an Australian isolate of lucerne transient streak virus (LTSV). Some plants infected with New Zealand isolates showed yellow flecks along lateral veins of leaves; symptoms were transient in some lucerne plants but persistent in others. A New Zealand isolate (LTSV-NZ) infected 14 of 39 plant species tested by mechanical inoculation, but was not transmitted by five aphid species. In sap of Nicotiana clevelandii , LTSV-NZ was infective after storage for 4 wk at 20 oC, diluting to 10-5, or heating for 10 min at 70 oC. Purified virus preparations contained a single electrophoretic component and a single sedimenting component (s20w= 112 S ) which formed a single buoyant density component in CsCl (1.37 g cm-3) but two density components in Cs2SO4 (1.26 and 1.32 g cm-3). LTSV-NZ particles were stable in 10 ITIM EDTA at pH 5, but not at pH 8, being degraded into two sedimenting components of 105 S and 92 S. Particles contained c. 18% RNA in the form of one single-stranded RNA molecule of mol. wt 1–4 times 106, and a polypeptide of mol. wt c. 32 400. LTSV-NZ was serologically unrelated to 24 other isometric plant viruses. However, its properties are similar to those of southern bean mosaic virus and allied viruses. The number of aerobic bacteria in a blanket peat decreased with depth from 26 times 106/g dry peat in the surface layers to 0.5 times 106/g dry peat at 30–40 cm down the profile, thereafter remaining roughly constant. Obligate psychrophiles comprised <2.5% of this population. Anaerobes were most numerous, 9 times 106/g dry peat at 6–10 cm depth, decreasing to 0.5 times 106/g at 20–30 cm. Calculations indicated that these counts, 103–104-fold lower than the direct counts, substantially underestimated the active microbial population. Gram negative rods, the predominant aerobes in the surface layers, were replaced by unidentified Bacillus strains at 10–20 cm depth but became increasingly more numerous further down the profile. The Gram negative rods were the most numerous organisms/m2 but the Bacillus strains, one third of which were present as spores, made the largest contribution to the biomass/m2. Gram positive cocci, Arthrobacter and, infrequently, Nocardia were also isolated. Actinomyces -like forms were the predominant obligate anaerobes and were approximately three times more numerous than clostridia and a curved Gram negative rod.  相似文献

Data from analyses of the fermentation of cassava slurry were used to derive kinetic constants for the rate of hydrolysis of bound cyanide, formation of free cyanide, pH change and fermentation. The pseudo-kinetic constant for the decay of bound cyanide was 1·6 times 10-2/h and 2·5/h for free cyanide. Under the experimental conditions, the concentration of bound cyanide at any time during fermentation was 22·65e-0·016tμg/g, where t is time in h. The fermentation rate constant was determined as 2·2871 times 1019. Based on the empirical expressions of the rate data, it is believed that rate constants of the intrinsic parameters of fermentation of systems similar to cassava slurry could be determined.  相似文献

Dormant tuber tissue of Jerusalem artichoke ( Helianthus tuberosus L.) can be stimulated by wounding to initiate RNA and protein synthesis. No DNA synthesis or cell divisions occur unless an auxin is provided. Changes in polysomal profiles and levels of Poly(A)+-RNA in response to wounding and auxin treatment were studied. Polysomes were isolated at various times after excision and incubation of tissue in the presence or absence of 10−5 M 2,4-dichlorophenoxyacetic acid. Polysomal profiles were studied by sucrose density gradient centrifugation. Dormant tissue contained ribosomes mainly in monosome form. Within 4 h of excision, a significant increase in the polysomal fraction was observed both in control and auxin-treated tissue. Increases in polysomes continued during the next 20 h. Poly(A)+-RNA was isolated from total polysomal RNA by oligo(dT)-cellulose column chromatography. There was a large increase in the amount of poly(A)+-RNA within 4 h of excision. During the first 43 h of incubation, levels of total polysomal RNA as well as poly(A)+-RNA in tissue treated with 2,4-dichlorophenoxyacetic acid were significantly higher than those in controls.  相似文献

An isolate of apple mosaic virus, designated ApMV-J, was isolated from rose cv. Sweet Surrender in New York State, USA. Mechanical transmission of ApMV-J from rose to cucumber was facilitated by preparing inocula with 0.03 M sodium phosphate buffer containing 0.02 M 2-mercap-toethanol and 1 % polyvinylpyrolidone. Catharanthus roseus cv. Little Linda was a good virus maintenance host and Cucumis sativus cv. Lemon a good propagation host. Contaminating host proteins were substantially reduced by including polyethylene glycol in the purification protocol. Purified virus had high A260/280 nm values and contained virus particles of c 25—26 nm in diameter; most particles were isometric but a few were bacilliform. The M.Wts. of the four nucleic acids of ApMV-J, determined in non-denaturing and denaturing agarose gels, were estimated at 1.16, 0.98, 0.68 and 0.35 × 106 and 1.0, 0.91, 0.66 and 0.35 × 106, respectively. Abstract .The susceptibility of field-collected Culicoides bolitinos to infection by oral ingestion of bluetongue virus serotypes 1, 3 and 4 (BLU 1, 3 and 4) was compared with that of field-collected C. imicola and laboratory reared C. variipennis sonorensis . The concentration of the virus per millilitre of bloodmeal was 105.0 and 106.0TCID50 for BLU 4 and 107.2TCID50 for BLU 1 and 3. Of 4927 C. bolitinos and 9585 C. imicola fed, 386 and 287 individual midges survived 10 days extrinsic incubation, respectively. Midges were assayed for the presence of virus using a microtitration assay on BHK-21 cells and/or an antigen capture ELISA. Infection prevalences for the different serotypes as determined by virus isolation ranged from 22.7 to 82.0% in C. bolitinos and from 1.9 to 9.8% in C. imicola; infection prevalences were highest for BLU 1, and lowest for BLU 4 in both species. The mean log10 TCID50 titre of the three BLU viruses per single fly was higher in C. bolitinos than in C. imicola . The results suggested that C. bolitinos populations are capable vectors of the BLU viruses in South Africa. A high correlation was found between virus isolation and ELISA results for the detection of BLU 1, and less for BLU 4; the ELISA failed to detect the presence of BLU 3 in infected flies. The C. v. sonorensis colonies had a significantly lower susceptibility to infection with BLU 1, 3 and 4 than C. bolitinos and C. imicola . However, since infection prevalence of C. v. sonorensis was determined only by ELISA, this finding may merely reflect the insensitivity of this assay at low virus titres, compared to virus isolation.  相似文献

A polymerase chain reaction (PCR) assay was developed for detection of pathogenic, virulent strains of Yersinia enterocolitica . By using both virulence loci virF and ail as markers for pathogenicity, detection of species with a virulence factor present was possible. DNA preparation in the presence of hexadecyl trimethy ammonium bromide (CTAB) was followed by two 44 cycle amplification reactions, one for each of the markers. As few as 102 Y. enterocolitica cells were detected in ground pork in the presence of 105–106 bacteria of other species. The described PCR assay provides a sensitive robust assay for the detection of virulent Y. enterocolitica in food.  相似文献

The natural 15N abundance of amide-exporting nodules was compared to that of shoots in 12 plant species. Nodules were statistically less abundant in 15N than shoots in one of three cultivars of Pisum sativum L., in Vicia faba L. and in Medicago sativa L., but the 15N depletion of nodules was very samall. Nodules were statistically more abundant in 15N than shoots in Trifolium pratense L., depending on time during the growing season, Cyamopsis tetragonaloba L. Taub. and 7 Lupinus species, but the enrichment was small except for C. tetragonalova and 6 Lupinus species. Nodules of 3 Lupinus species infected with Rhizobium lupini isolated from Lupinus subcarnosa Hook, were only slightly enriched in 15N, but nodules of two of these species were substantially enriched in 15N when infected with a mix of other Rhizobium lupini strains. The third species, L. texensis Hook., was not infected by this mix of strains. Differences in 15N abundance between nodules and other tissues of amide-exporting and ureide-exporting nodules from several studies are tabulated. All ureide-exporting nodules in this tabulation are enriched in 15N. Amide-exporting nodules are considerably more variable in this regard. These results confirm that events associated with ureide synthesis or transport cannot be the sole cause of the substantial 15N enrichment seen in nodules.  相似文献

Abstract Cells of Rhodocyclus gelatinosus were radioactively labeled by addition of [32P]orthophosphate, [14C]inosine or [14C]orotic acid during anaerobic growth on citrate in the light. Protein analysis by two-dimensional gel electrophoresis and autoradiography of the gels revealed the presence of several radioactively labeled protein species in this organism. The molecular mass and the isoelectric point of all these proteins were determined. Treatment of the 32P-labeled protein fractions with acid and alkaline phosphatase clearly showed that at least 8 protein species were modified by phosphorylation. The experiments conducted with the 14C-labeled precursors of purines and pyrimidines indicated the presence of 4 protein species which were modified by a compound containing a purine and phosphate, and a single protein simultaneously being labeled with pyrimidine and phosphate.  相似文献

Abstract Oral immunization of suckling mice with Cryptosporidium parvum results in a humoral response to a limited set of antigens. Six-day-old BALB/c mice were each inoculated orally with 1 × 106 viable oocysts and subsequently administered oral inoculations of 2 × 106 viable oocysts at 30 and 60 days following the primary infection. After 45 days, mice were boosted with 1 × 106 oocysts orally, plus soluble extracts equivalent to 2 × 106 and 1 × 106 oocysts given intravenously and intraperitoneally, respectively. Four days later, splenic lymphocytes were fused to Ag8 myeloma cells. Using this method, we have been able to select for monoclonal antibodies that predominately recognize sporozoite surface and apical complex antigens.  相似文献

The planktonic and benthic bacterial populations of Lough Neagh, Northern Ireland, were studied over a one-year period. Direct counts of bacteria in the water column averaged 6 times 107/ml with limited spatial or temporal variation; viable counts, however, showed a pronounced late spring maximum of 1.7 times 106/ml and were consistently higher at a littoral sampling station. Direct counts of bacteria in the profundal sediments averaged 8 times 109/ml whilst viable benthic counts rose steeply during spring to reach a June maximum of 1 times 108/ml. Direct: viable count ratios were much greater in the more sandy littoral zone. The predominant benthic isolate was an Aeromonas sp. which was also common in samples from the water column. These results confirm the eutrophic status of Lough Neagh indicated by other biological and chemical surveys.  相似文献

Abstract Using a modified procedure large indigenous plasmids were detected in cells of three strains belonging to a group of phenotypically similar actinomycetes isolated from the rhizoplane and root nodules of Alnus spp. M r values of the plasmids were estimated to be about 80 · 106 and 120 · 106. The plasmid profiles of different strains of the group were found to be almost identical. This remarkable plasmid similarly is discussed in relation to the common source of isolation.  相似文献

The embryos, kept at 20°C for 3 hr–6 hr from the time of fertilization (at the morula stage), were cultured in sea water containing cycloheximide (10–16 mM) for successive 3 hr and then transferred to normal sea water. The embryos, thus treated, became vegetalized larvae. With the same treatment performed at a developmental stage prior to 3 hr of fertilization, most of embryos developed to small blastulae filled with mesenchyme-like cells. The treatment at a stage after 6 hr of fertilization yielded normal plutei. From the embryos exposed to both 14C-leucine and 3H-thymidine during the treatment, labelled chromatin was isolated. Only in the presumptive vegetalized embryos obtained by the cycloheximide treatment of morulae, ratio of 14C-radioactivity found in proteins of chromatin to 3H-radioactivity in DNA was markedly lower than that observed in chromatin from control embryos. The rate of 3H-radioactivity-decrease by DNase I treatment was higher in chromatin isolated from the presumptive regetalized embryos than that observed in chromatin isolated from control ones. Probable failure of chromatin structure formation, due to cycloheximide-inhibition of chromatin protein synthesis, seems to disturb the determination in the embryos at the morula stage, resulting in an induction of vegetalized embryos.  相似文献

Abstract: Over 60 Salsola species of Chenopodiaceae from South Africa were studied for their photosynthesis type, using δ13C analysis and light microscopy of leaf anatomy. These species cover about 70 % of the total list of Southern African Salsola species and grow naturally in South and Southwest African desert regions. All species are shrubby forms and belong to the single subsection Caroxylon. Only C4 photosynthesis was found in the Salsola species determined with 13C/12C carbon isotope discrimination values that ranged from - 11.04 to - 14.03 % (PDB), plus the presence of a Kranz type assimilation tissue anatomy. The apparent absence of C3 in Salsola in South and Southwest Africa and the known presence of C3 and C3 - C4 intermediate photosynthesis in Caroxylon, Salsola species in Asia strongly indicate that the genus Salsola originated in Asia and later migrated to South Africa.  相似文献

Abstract An examination of samples obtained from a commercial fish smoker, using seawater agar with incubation at 4°, 15° and 37°C for up to 28 days, revealed the presence of large bacterial populations in smoked fish. However, initially only low bacterial numbers, i.e., 2 × 103/g, were present in the muscle of fresh, whole haddock ( Melanogrammus aeglefinus ). With filleting, there was a sudden increase in numbers to 9.2 × 105/g. Yet immediately after smoking, the bacterial populations decreased (5 × 105/g), followed by a gradual increase with storage (e.g., 2 × 106/g after 24 h). Representative colonies were presumptively identified as Acinetobacter, Alcaligenes , coryneforms, Pseudomonas and Vibrio spp.  相似文献

Bacteria grew to viable populations of 108–109 cfu/g during the fermentation of soybeans into tempe with the fungus, Rhizopus oligosporus. Bacillus pumilus and B. brevis were the predominant bacterial species, reaching populations of approximately 108 cfu/g during the 48 h fermentation. Species of Streptococcus faecium, Lactobacillus casei, Klebsiella pneumoniae and Enterobacter cloacae also contributed to the fermentation and achieved populations of 106–107 cfu/g. and accepted 25 May 1989  相似文献

SUMMARY. 1. Published data are used to construct a tentative carbon budget for the ecosystem of the lower river Rhine and its sedimentation areas in The Netherlands.
2. It is estimated that 287 × 106kg Cy−1 of particulate organic material is transported by the river Rhine, and 100 × 106 kgCy−1 of this material is deposited in the delta.
3. Phytoplankton and nitrifying bacteria seem to contribute significantly to the carbon budget, by producing c. 78 × 106kgCy−l.
4. The mineralization in the water (estimated from routine determinations of biological oxygen demand and from in situ diurnal fluctuations of oxygen) and in the bottom (estimated from the denitrification rates in the delta, from the production rate of methane and from overall oxygen consumption) is shown to degrade c. 50% of the carbon input plus autochthonous production.
5. The carbon budget and oxygen regime in the lower Rhine suggest that after decades of severe organic pollution the river has more or less resumed the normal, slightly heterotrophic state of a large lowland river.  相似文献
