A series of novel 6-methoxy-2-(piperazin-1-yl)-4H-chromen-4-one and 5,7-dimethoxy-2-(piperazin-1-ylmethyl)-4H-chromen-4-one derivatives of biological interest were prepared and screened for their pro-inflammatory cytokines (TNF-α and IL-6) and antimicrobial activity (antibacterial and antifungal). Among all the compound screened (5aj and 10kt), the compounds 5c, 5g, 5h, 10l, 10m, 10n and 10r found to have promising anti-inflammatory activity (up to 65–87% TNF-α and 70–93% IL-6 inhibitory activity) at concentration of 10 μM with reference to standard dexamethasone (71% TNF-a and 84% IL-6 inhibitory activities at 1 μM) while the compounds 5b, 5i, 5j, 10s and 10t found to be potent antimicrobial agent showing even 2 to 2.5-fold more potency than that of standard ciprofloxacin and miconazole at the same MIC value of 10 μg/mL.  相似文献

A series of novel 2-imino-4-thiazolidinone derivatives 4a,b was synthesized through reaction of unsymmetrical thioureas 3a,b with chloroacetic acid. Condensation of 4a,b with aromatic aldehydes 5a-eyielded the corresponding 5-arylidene derivatives 6a-j. In addition, the reaction of 4a,b with 4-arylazo-3-hydroxybenzaldehydes 8a-c furnished the respective mono-arylazo-4-thiazolidinones10a-f. All the newly synthesized compounds were confirmed by their elemental analysis and spectral data. The antifungal activity of the newly synthesized compounds was assessed and the compounds 6d, 6e, 6i, 6j, 9a,b and 10a-frevealedhigher antifungal activity towards Alternaria solani than to the standard Ridomil gold plus. Moreover, the DNA toxicity of 4-thiazolidinone derivatives 6d, 9a, 10b, 10c and 10f on the nucleic acid of Alternaria solani (KT354939) was performed and the results showed qualitatively more than 70% cleavage. Also, compounds 6i, 6j, 9b, 10c and 10f were docked inside the active site of 1ZOYenzyme and suitable binding with the active site of amino acids, were displayed according to their bond lengths, angles and conformational energy.  相似文献

Mucor genevensis were used to bioconvert sinenxan A [2α,5α,10β,14β-tetraacetoxy-taxa-4(20),11-diene], a taxoid isolated from callus tissue cultures of Taxus spp., and 10 metabolites were obtained. On the basis of chemical and spectroscopic data analyses, their structures were determined as 10β-methoxy-2α,5α,14β-triacetoxy-taxa-4(20),11-diene (2), 10β-hydroxy-2α,5α,14β-triacetoxy-taxa-4(20),11-diene (3), 2α,5α,10β,14β-tetraacetoxy-4β,20-epoxy-taxa-11(12)-ene (4), 6α-hydroxy-2α,5α,10β,14β-tetraacetoxy-taxa-4(20),11-diene (5), 9α-hydroxy-2α,5α,10β,14β-tetraacetoxy-taxa-4(20),11-diene (6), 10β-hydroxy-2α,5α,14β-triacetoxy-4β,20-epoxy-taxa-11(12)-ene (7), 6α,10β-dihydroxy-2α,5α,14β-triacetoxy-taxa-4(20),11-diene (8), 6α-hydroxy-2α,5α,10β,14β-tetraacetoxy-4β,20-epoxy-taxa-11(12)-ene (9), and 9α,10β-dihydroxy-2α,5α,14β-triacetoxy-taxa-4(20),11-diene (10), and 9α,10β-O-(propane-2,2-diyl)-2α,5α,14β-triacetoxy-taxa-4(20),11-diene (11). Among them, metabolites 2, 4, and 9 were three new compounds. The three major metabolites 2, 3, and 4 along with 1 were pharmacologically evaluated for their multi-drug resistance (MDR) reversal activities towards taxol-resistant A549 tumor cells, and the results showed that 4 possessed about two-fold activity as verapamil, while 2, and 3 possessed lower activity than verapamil and 1.  相似文献

Syntheses of ten derivatives of mycophenolic acid (MPA) at C-6′ position, and structure–activity relationship study among these derivatives, MPA and mycophenolic hydroxamic acid (MPHA) led to discovery of N-(2,3,5-triazolyl)mycophenolic amide 4, (7′S) mycophenolic epoxyketone 9 and (7′R) mycophenolic epoxyketone 10 having potent inhibitory activity against human inosine-5′-monophosphate dehydrogenase (IMPDH) type I and II as well as antiproliferative activity on human leukemia K562 cells. Compounds 4, 9, and 10 showed induction activity of erythroid differentiation in K562 cells. Inhibitory effects of 4 and 10 against IMPDH were attenuated by supplemental guanosine in K562 cells. In contrast, attenuation effect by supplemental guanosine was not significant in the case of 9. Compound 9 weakly inhibited the enzyme activity of HDAC in the nuclear lysate of K562 cells at 10 μM. These observations suggest that the primary target of 4, 9, and 10 is IMPDH, whereas compound 9 partially inhibits a certain type of HDAC.  相似文献

Three 7-acylaminoimidazo[4,5-b]pyridines, namely 7-pentanoylaminoimidazo[4,5-b]pyridine (1), 7-benzoylaminoimidazo[4,5-b]pyridine(2), and 7-(2-furoylamino)imidazo[4,5-b]pyridine(3), six 4-acylaminoimidazo[4,5-c]pyridines, namely 4-propionylaminoimidazo[4,5-c]pyridine(4), 4-butyryl-aminoimidazo[4,5-c]pyridine(5), 4-pentanoylaminoimidazo[4,5-c]pyridine(6) 4-hexanoylaminoimidazo[4,5-c]pyridine(7),4-benzoylaminoimidazo[4,5-c]pyridine(8), and 4-(2-furoylamino)imidazo[4,5-c]-pyridine(9), and seven 7-acylaminoimidazo[4,5-c]pyridines, namely 7-propionylaminoimidazo[4,5-c]-pyridine(10), 7-butyrylaminoimidazo[4,5-c]pyridine(11), 7-pentanoylaminoimidazo[4,5-c]pyridine(12), 7-hexanoylaminoimidazo[4,5-c]pyridine(13), 7-benzoylaminoimidazo[4,5-c]pyridine(14), 7-phenylacetylaminoimidazo[4,5-c]pyridine(15), and 7-(2-furoylamino)imidazo[4,5-c]pyridine(16) were synthesized and tested for their cytokinin activity with the tobacco callus bioassay. 2 showed a cytokinin activity at 1 × 10−8 M and gave a callus yield about 72% of that produced by kinetin at 1 × 10−6 M. 1, 3 and 8 showed the optimum growth responses in the range of 10−7−10−6 M. 4, 5, 7, 9–16 were slightly active. These results support previous reports that a nitrogen atom at the 3-position in the purine ring plays an important role in conferring high cytokinin activity.  相似文献

A new cytotoxic Annonaceous acetogenin, annojahnin (1), was isolated from the twigs of Annona jahnii (Annonaceae) by bioactivity-directed fractionation using lethality to brine shrimp. Compound 1 represents an unusual type of C–37 Annonaceous acetogenin, lacking either tetrahydrofuran (THF) or epoxide rings, bearing a keto group at C–10, and possessing a double bond located two methylenes away from a vicinal diol. The structure and absolute configuration of 1 were elucidated by 1H and 13C NMR, COSY, and single-relayed COSY and from chemical derivatives. 4-Deoxy-18/21-trans-annomontacin 10-one (4) and 4-deoxy-18/21-cis-annomontacin-10-one (5), two semisynthetic mono-THF acetogenins, were prepared from 1 by reactions that mimic the biogenetic pathways. These acetogenins showed selective cytotoxicities, comparable or superior to adriamycin, among six human solid tumor cell lines. Reduction of the 10-keto of 1, to the racemic 10-OH derivative (3), retained the bioactivities as did the conversion of 1 to 4 and 5.  相似文献

The first phytochemical investigation on the vine stems of Millettia dorwardi Coll. et Hemsl led to the isolation of ten flavonoids (isoafrormosin 1, formononetin 2, afrormosin 3, padmakastein 4, liquiritigenin 5, 4H-1-Benzopyran-4-one,7-hydroxy-5,8-dimethoxy-3-(4-methoxyphenyl)-isoflavone 6, 4H-1-Benzopyran-4-one,7-hydroxy-3-(3-hydroxy-5-methoxyphenyl)-6-methoxy 8, 4H-1-Benzopyran-4-one,6-methoxy-3-(4-methoxyphenyl)-6,4′-dimethoxyisoflavone 9, irisolidone 10, prunetin 11), one heterocycle (5-5′-dibuthoxy-2-2′-bifuran 7) and one new isoflavone glycoside (4H-1-Benzopyran-4-one,5-hydroxymethyl-3-(4-methoxyphenyl)-6-β-d-glucopyranoside-isoflavone 12). Their structures were determined by extensive analysis of their spectroscopic data. Among them, compounds 4, 610, 12 were for the first time isolated from this genus. The chemotaxonomic importance of these compounds was also summarized.  相似文献

A series of long-chain derivatives of chrysin (compounds 322) were synthesized to evaluate for their antiproliferative activities against the human liver cancer cell line HT-29 and EGFR inhibitory activity. Among the compounds tested, compounds hexadecyl 2-(5-hydroxy-4-oxo-2-phenyl-4H-chromen-7-yloxy)acetate (10) and N-hexadecyl 2-(5-hydroxy-4-oxo-2-phenyl-4H-chromen-7-yloxy)acetamide (20) displayed potent EGFR inhibitory activity with IC50 values of 0.048 μM and 0.035 μM), comparable to the positive control erlotinib. Docking simulation of compounds 10 and 20 was carried out to illustrate the binding mode of the molecular into the EGFR active site, and the result suggested that compound 10 and 20 can bind the EGFR kinase well. Thus, compounds 10 and 20 with potent EGFR inhibitory activity would be potential anticancer agents.  相似文献

Lipoprotein lipase (LPL) is a key physiological regulator of triglycerides and atherosclerosis risk. Random screening identified a compound designated C10, showing greater LPL agonist activity than NO-1886, a known LPL agonist. Structure-activity relationship (SAR) exploration of C10 led to the identification of C10d exhibiting at least two fold greater LPL activation than NO-1886. Unlike NO-1886, novel LPL agonists C10 and C10d reversed the LPL inhibition by angiopoietin-like 4 (ANGPTL4), a physiological inhibitor of LPL.  相似文献

A combinatorial library of β-chlorovinyl chalcones (4) were synthesized by Claisen–Schmidt condensation reaction. Catalytic reaction of substituted 3-chloro-3-phenyl-propenal (2) and 1-(2,4-dimethoxy-phenyl)-ethanone or 1-(4-methoxy-phenyl)-ethanone (3) in alkaline conditions furnished the target compound 5-chloro-1-(2,4-dimethoxy-phenyl)-5-phenyl-penta-2,4-dien-1-one (4). The synthesized compounds were screened for their biological activity viz. anticancer, anti-inflammatory and antimicrobial activities. Synthesized compounds 4g and 4h revealed promising anti-inflammatory activity (66–67% TNF-α and 95–97% IL-6 inhibitory activity at 10 μM). Cytotoxicity of the compounds checked using CCK-8 cell lines and found to be nontoxic to slightly toxic. Furthermore, the anticancer activity (30–40%) was shown by compounds 4d, 4e, 4h and 4b at 10 μM concentrations against ACHN followed by Calu 1, Panc1, HCT116 and H460 cell lines. Some of the compounds 4d, 4e, 4a, 4i and 4b revealed promising antimicrobial activity at MIC 50–100 μg/mL against selected pathogenic bacteria and fungi.  相似文献

The wood and stem bark of Ervatamia heyneana (Apocynaceae) yielded 14 indole alkaloids and 3 triterpenoids. Six of these isolates, camptothecin (2), 9-methoxycamptothecin (3), coronaridine (1), pericalline (25), heyneatine (18) and 10-methoxyeglandine- N-oxide (4) displayed cytotoxic activity. Three of the indole alkaloids 18, 4 and 10-hydroxycoronaridine (8) are new members in the iboga series and their structures were determined by a combination of spectral interpretation and chemical correlation.  相似文献

The nonproteinogenic amino acid, 4-hydroxyisoleucine (1) has been isolated in large quantities from the fenugreek (T. foenum-graecum) seeds. Few novel derivatives (311 and 1318) were prepared from the naturally occurring 4-hydroxyisoleucine (1) and screened for their in vitro glucose uptake stimulatory effect in L-6 skeletal muscle cells. The derivatives 6, 7, 8, 10 and 11 exhibited better glucose uptake stimulatory activity than parent compound, 4-hydroxyisoleucine at 5 and 10 µM concentrations and compounds 7 and 11 enhanced translocation of insulin sensitive glucose transporters-4 in skeletal muscle cells.  相似文献

Valiolamine (1), a branched-chain aminocyclitol α-d-glucosidase inhibitor, has been synthesised as the racemic penta-N,O-acetyl derivative (10) from dl-(1,3/2,4)-1,2,3-triacetoxy-4-bromo-6-methylenecyclohexane (2). Epoxidation of 2 with m-chloroperbenzoic acid, followed by hydrolysis and acetylation, gave exclusively dl-(1,2,4/3,5)-2,3,4-triacetoxy-1-C-acetoxymethyl-5-bromocyclohexanol (4), from which 10 was obtained by azidolysis in N,N-dimethylformamide, and successive catalytic hydrogenation and acetylation. In contrast, azidolysis in aqueous 2-methoxyethanol gave dl-(1,2,4/3,5)-2,3,4-triacetoxy-1-C-acetoxymethyl-5-azidocyclohexanol, which was converted into the 5-epimer of 10. Hydroxylation of 2 with osmium tetraoxide and hydrogen peroxide, followed by acetylation, gave the 1-epimer (6) of 4. The 1,5-diepimer of 10 was prepared from 6 by the same sequence.  相似文献

Two new diterpenoid quinones, salvianolactone acid J and K (1 and 2), as well as eight known compounds (310) were isolated from the roots of Salvia miltiorrhiza. Their structures were elucidated by comprehensive analysis. All compounds (1-10) were evaluated for their protective effects against the BEAS-2B cells injury induced by LPS in vitro and compounds 1, 3, and 4 showed significantly protective activity at the concentration of 10 μM. Besides, compounds 1, 3, and 4 ameliorated the nuclear transport of p-P65 in LPS-induced BEAS-2B cells.  相似文献

A novel series of bis-indoles derived from naturally occurring marine alkaloid 4 were synthesized and evaluated as inhibitors of methicillin-resistant Staphylococcus aureus (MRSA) pyruvate kinase (PK). PK is not only critical for bacterial survival which would make it a target for development of novel antibiotics, but it is reported to be one of the most highly connected ‘hub proteins’ in MRSA, and thus should be very sensitive to mutations and making it difficult for the bacteria to develop resistance. From the co-crystal structure of cis-3-4-dihydrohamacanthin B (4) bound to S. aureus PK we were able to identify the pharmacophore needed for activity. Consequently, we prepared simple direct linked bis-indoles such as 10b that have similar anti-MRSA activity as compound 4. Structure–activity relationship (SAR) studies were carried out on 10b and led us to discover more potent compounds such as 10c, 10d, 10k and 10m with enzyme inhibiting activities in the low nanomolar range that effectively inhibited the bacteria growth in culture with minimum inhibitory concentrations (MIC) for MRSA as low as 0.5 μg/ml. Some potent PK inhibitors, such as 10b, exhibited attenuated antibacterial activity and were found to be substrates for an efflux mechanism in S. aureus. Studies comparing a wild type S. aureus with a construct (S. aureus LAC Δpyk::ErmR) that lacks PK activity confirmed that bactericidal activity of 10d was PK-dependant.  相似文献

Five sesquiterpenoids, 1α,8α-epidioxy-4α-hydroxy- 5αH-guai-7(11),9-dien- 12,8-olide. (1), 8,9-seco-4β-hydroxy-1α,5βH-7(11)-guaen-8,10-olide (2), 8α-hydroxy-1α, 4β,7βH-guai-10(15)-en- 5β,8β-endoxide(3), 7β,8α-dihydroxy-1α,4αH-guai-10(15)-en-5β,8β-endoxide(4) and 7-hydroxy-5(10),6,8-cadinatriene-4-one(5), together with seven known analogs were isolated from the rhizomes of Curcuma wenyujin. Their structures and relative configurations were determined on the basis of spectroscopic methods including 2D NMR techniques, and the structures of 1 and 2 were confirmed by single-crystal X-ray diffraction experiment. Compounds 1–10 and 12 showed significant in vitro antiviral activity against the influenza virus A with IC50 values ranged from 6.80 to 39.97 μM, and SI values ranged from 6.35 to 37.25.  相似文献

VirB8 is a critical component of the Brucella suis type IV secretion system (T4SS). We previously showed that the transmembrane (TM) domain plays an essential role in interactions of this protein with itself and the other proteins of the T4SS. We report that a point mutation in this TM domain stabilizes homodimers of VirB8 and heterodimers with VirB10. A similar variant of Agrobacterium tumefaciens VirB8 showed the same phenotype. The B. suis VirB8 variant was unable to complement a virB8 mutant and displayed a dominant negative phenotype when expressed in wild type B. suis. In a search for novel multifunctional anti-Alzheimer agents, a congeneric set of seventeen flavone-8-acrylamide derivatives (8a─q) were synthesized and evaluated for their cholinesterase inhibitory, antioxidant, neuroprotective and modulation of Aβ aggregation activities. The target compounds showed effective and selective inhibitory activity against the AChE over BuChE. In addition, the target compounds also showed moderate anti-oxidant activity and strong neuroprotective capacities, and accelerated dosage-dependently the Aβ aggregation. Also, we presented here a complete study on the interaction of 8a, 8d, 8e, 8h and 8i with AChE. Through fluorescence emission studies, the binding sites number found to be 1, binding constants were calculated as 2.04 × 104, 2.22 × 104, 1.18 × 104, 9.8 × 103 and 3.2 × 104 M−1 and free energy change as −5.83, −5.91, −5.51, −5.41 and −6.12 kcal M−1 at 25 °C which were well agreed with the computational calculations indicating a strong binding affinity of flavones and AChE. Furthermore, the CD studies revealed that the secondary structure of AChE became partly unfolded upon binding with 8a, 8d, 8e, 8h and 8i.  相似文献

In this work, the benzimidazole-pyrrole conjugates 6ah and benzimidazole-tetracycles conjugates 1214 were prepared. The cytotoxicity of the compounds 3, 4ah, 6ah, 8, 10 and 1214 was tested against lung cancer cell line A549. Compound 6b exhibited higher activity than the bis-benzoxazole natural product (UK-1), the standard. The tested 4g,h, 6ah, 10 and 1214 exhibited remarkable cytotoxicity activity against breast cancer cell line MCF-7 with higher activity than tamoxifen. Furthermore, compound 4h was found to be also more potent than doxurubicin. The antitumor promotion activity of synthesized compounds 4g,h, 6ah, 10 and 1214 has been estimated by studying their possible inhibitory effects on EBV-EA activation induced by 12-O-tetradecanoylphorbol-13-acetate (TPA). Among the studied compounds, the inhibitory activities of compounds 8, 13 and 14 demonstrated strong inhibitory effects on the Epstein–Barr virus early antigen (EBV-EA) activation without showing any cytotoxicity on the Raji cells and their effects being stronger than that of a representative control, oleanolic acid.Moreover, the molecular docking of the new compounds into plasminogen activator (uPA) receptor has been in correlation with the antitumor activity. All synthesized compounds 3, 4ah, 6ah, 8, 10 and 1214 were docked into same groove of the binding site of the native co-crystalized (4-iodobenzo[b]thiophene-2-carboxamidine) ligand (PDB code:1c5x) for activity explaination. Compounds 4h, 6b and 13, giving the best docking results, were further studied to estimate their effect on the level of uPA using AssayMax human urokinase (uPA) ELISA kit. In case of A549 cell line, compound 6 exhibited similar activity to MMC, and for MCF-7 cell line, compound 4h exhibited similar activity to doxorubicin, in inhibiting the expression of uPA.  相似文献

Two series of novel 4-chlorophenyl N-alkyl phosphoramidates of 3′-O-(t-butoxycarbonyl)-5-fluoro-2′-deoxyuridine (3′-BOC-FdU) (9a9j) and 5-fluoro-2′-deoxyuridine (FdU) (10a10j) were synthesized by means of phosphorylation of 3′-BOC-FdU (4) with 4-chlorophenyl phosphoroditriazolide (7), followed by a reaction with the appropriate amine. Phosphoramidates 9a9j were converted to the corresponding 10a10j by removal of the 3′-t-butoxycarbonyl protecting group (BOC) under acidic conditions. The synthesized phosphoramidates 9a9j and 10a10j were evaluated for their cytotoxic activity in five human cancer cell lines: cervical (HeLa), nasopharyngeal (KB), breast (MCF-7), liver (HepG2), osteosarcoma (143B) and normal human dermal fibroblast cell line (HDF) using the sulforhodamine B (SRB) assay. Two phosphoramidates 9b and 9j with the N-ethyl and N-(methoxy-(S)-alaninyl) substituents, respectively, displayed remarkable activity in all the investigated cancer cells, and the activity was considerably higher than that of the parent nucleoside 4 and FdU. Among phosphoramidates 10a10j compound 10c with the N-(2,2,2-trifluoroethyl) substituent showed the highest activity. Phosphoramidate 10c was more active than the FdU in all the cancer cell lines tested.  相似文献
