N,N′-Dilithiated 1,1′-bis(trimethylsilylamino)ferrocene (2) reacts with boron halide adducts (HBBr2-SMe2; BF3-OEt2 and BBr3-SMe2), boron halides (BCl3, BBr3, BCl2(OPh) and BCl2(Ph)) and 1,1-bis(dimethylamino)dichlorodiborane(4) to give the corresponding 1,3-bis(trimethylsilyl)-1,3,2-diazabora-[3]ferrocenophanes (3)-(8) and the 2,3-bis(dimethylamino)-1,4-bis(trimethylsilyl)-1,4,2,3-diazadibora-[4]ferrocenophane (9). All new complexes were characterised by multinuclear magnetic resonance spectroscopy in solution, and the solid-state molecular structures of the hydride (3), fluoride, chloride (4, 5), and of the phenoxy and phenyl derivatives (7, 8) were determined by X-ray analysis.  相似文献

The CNS of the cockroach Periplaneta americana contains saturable, specific binding sites for [3H]GABA, [3H]flunitrazepam and [35S]TBPS. The [3H]GABA binding site exhibits a pharmacological profile distinct from that reported for mammalian GABAA and GABAB receptors. The most potent inhibitors of [3H]GABA binding were GABA and muscimol, whereas isoguvacine, thiomuscimol and 3-aminopropane sulphonic acid were less effective. Bicuculline methiodide and baclofen were ineffective. Binding of [35S]TBPS was partially inhibited by 1.0 × 10−6 M GABA, whilst binding of [3H]flunitrazepam was enhanced by 1.0 × 10−7 M GABA. The pharmacological profile of the [3H]flunitrazepam binding site showed some similarities with the peripheral benzodiazepine binding sites of vertebrates, with Ro-5-4864 being a far more effective inhibitor of binding than clonazepam. Thus a class of GABA receptors with pharmacological properties distinct from mammalian GABA receptor subtypes is present in insect CNS.  相似文献

Extracts of Valeriana officinalis have been used in folkloric medicine for its sedative, hypnotic, tranquilizer and anticonvulsant effects, and may interact with -aminobutyric acid (GABA) and/or benzodiazepine sites. At low concentrations, valerian extracts enhance [3H]flunitrazepam binding (EC50 4.13 × 10–10 mg/ml). However, this increased [3H]flunitrazepam binding is replaced by an inhibition at higher concentrations (IC50 of 4.82 × 10–1 mg/ml). These results are consistent with the presence of at least two different biological activities interacting with [3H]flunitrazepam binding sites. Valerian extracts also potentiate K+ or veratridine-stimulated release of radioactivity from hippocampal slices preloaded with [3H]GABA. Finally, inhibition of synaptosomal [3H]GABA uptake by valerian extracts also displays a biphasic interaction with guvacine. The results confirm that valerian extracts have effects on GABAA receptors, but can also interact at other presynaptic components of GABAergic neurons.  相似文献

The compound [Cr(en)3][ZnCl4]Cl has been synthesized by reaction of CrCl3·6H2O, Zn and [Cr(en)3]2(SO4)3 in HCl. Its molecular and crystalline structure was determined by X-ray diffraction methods, being monoclinic, P21/c, a=21.215(3), b=12.532(2), c=13.707(2) Å, β=95.21°, V= 3629(2) Å3, Dx=1.738g cm−3, MW=474.9, Z= 8, F(000)=1928, λ(Mo Kα)=0.71069 Å, μ(Mo Kα)= 27.04 cm−1, 288 K. No significant exchange interactions between Cr(III) cations in the crystalline lattice were found. Curie-Weiss behavior was found in the three directions tested (g1=2.06±0.02,g2= 2.08±0.02,g3=2.09±0.01), T=1.2-1.4 K.  相似文献

Synaptosomal fractions were isolated from frog retina: a fraction enriched in photoreceptor terminals (P1) and a second one (P2) containing interneurons terminals. We compared the binding of [3H]glycine and [3H]strychine to membranes of these synaptosomes. The binding of both radioactive ligands was saturable and Na+-independent. [3H]Glycine bound to a single site in P1 and P2 synaptosomal fractions, with KD=12 and 82 nM and BMax=3.1 and 3.06 pmol/mg protein respectively. [3H]Strychnine bound to two sites in each one of the synaptosomal fractions. For P1 KD values were 3.9 and 18.7 nM, and BMax values were 1.1 and 7.1 pmol/mg protein, respecitively. Membranes from the P2 synaptosomal fraction showed KD's of 0.6 and 48 nM and BMax's of 0.4 and 4.5 pmol/mg. Specific [3H]glycine binding was displaced by -alanine, l-serine, d-serine and HA966, but not by strychnine 7-chlorokynurenic or 5,7-dichloro-kynurenic acids. Specific [3H]strychnine, binding was partially displaced by glycine and related aminoacids and totally displaced only by 2-NH2-strychnine. Our results indicate the presence of high affinity binding sites for glycine and strychnine in frog retinal synaptosomal membranes. The pharmacological binding pattern indicates the presence of the strychnine sensitive glycine receptor as well as other sites. These might not include the NMDA receptor-associated glycine site.  相似文献

This study aimed at comparing the binding characteristics of [3H]ketanserin, a high-affinity serotonin 2A (5-HT2A) receptor antagonist, in the prefrontal cortex, hippocampus and striatum of human brain post-mortem. The results indicated the presence of a single population of binding sites in all the regions investigated, with no statistical difference in maximum binding capacity (Bmax) or dissociation constant (Kd) values. The pharmacological profile of [3H]ketanserin binding was consistent with the labeling of the 5-HT2A receptor, since it revealed a competing drug potency ranking of ketanserin = spiperone > clozapine = haloperidol > methysergide > mesulergine > 5-HT. In conclusion, the 5-HT2A receptor, as labeled by [3H]ketanserin, would seem to consist of a homogenous population of binding sites and to be equally distributed in human prefronto-cortical, limbic and extrapyramidal structures.  相似文献

The fate of [14C] gibberellin A3 and [3H] gibberellin A1 was examined in senescing fruit of Shamouti orange (Citrus sinensis L. Osbeck) and tomato (Lycopersicon esculentum Mill.). Gibberellin A3 was highly persistent in Citrus peel (t 1/2=18 days) and to a lesser degree in tomato (t 1/2=5.5 days). Ethylene and ethephon caused a slight enhancement of gibberellin A3 metabolism in Citrus and tomato fruit, respectively. Gibberellin A1 was metabolized by Citrus peel at a relatively high rate (t 1/2 < 24 h) and ethylene slightly reduced this rate. Summary Movement of both [3H]GA1 and [14C]GA3 through root segments from P. coccineus seedlings was basipetally polarised. The basipetal/acropetal ratio of radioactivity from [3H]GA1 in agar receiver blocks was 9.2 for apical, elongating segments, and 4.0 for more basal, non-elongating segments. Polarity of gibberellin transport was restricted to the stele, and absent from cortical tissues. Transport of [14C]IAA through root segments to agar receivers was preferentially acropetal, particularly so in the stele. Despite the existence of basipetal polarity of gibberellin transport in the root, [3H]GA1 injected into cotyledons moved into and acropetally along the seedling root.  相似文献

Two models of perturbed cerebellar ontogenesis were obtained by a single administration of methylazoxymethanol (MAM), a potent antimitotic agent, to mouse pups either on the day of birth (MAM0 mice) or at postnatal day 5 (MAM5 mice). The alterations of the cerebellar GABAergic system were studied by measuring glutamic acid decarboxylase activity, [3H]muscimol binding sites, which are known to be concentrated in the GABAA receptors in the internal granular layer, and [3H]flunitrazepam binding sites, which are more abundant in the molecular layer. The primary target of the antimitotic agent are the precursors of the glutamatergic and GABAceptive granule cells. In both models GABAergic structures, as revealed by GAD activity measurements, appear to be relatively spared, and recovery of granule cell numbers occurs during development in MAM5 mice. In MAM treated mice the number of [3H]muscimol binding sites (on a per cerebellum basis) decrease as the number of granule cells decrease, although some recovery occurred in MAM5 mice, but not in MAM0 mice. In MAM5 mice, [3H]flunitrazepam binding sites (on a per cerebellum basis) were relatively unaffected, while they were decreased significantly, but to a lesser extent than [3H]muscimol binding sites, in MAM0 animals. The more significant reduction of granule cell numbers and the cytoarchitectural disruption resultant from the more precocious application of the antimitotic appear responsible for the significant alteration and lack of recovery in MAM0 mice.  相似文献

Structural changes between [OsIIL3]2+ and [OsIIIL3]3+ (L: 2,2′-bipyridine; 1,10-phenanthroline) and molecular and electronic structures of the OsIII complexes [OsIII(bpy)3]3+ and [OsIII(phen)3]3+ are discussed in this paper. Mid-infrared spectra in the ν(bpy) and ν(phen) ring stretching region for [OsII(bpy)3](PF6)2, [OsIII(bpy)3](PF6)3, [OsII(phen)3](PF6)2, and [OsIII(phen)3](PF6)3 are compared, as are X-ray crystal structures. Absorption spectra in the UV region for [OsIII(bpy)3](PF6)3 and [OsIII(phen)3](PF6)3 are dominated by very intense absorptions (ε = 40 000-50 000 M−1 cm−1) due to bpy and phen intra-ligand π → π transitions. In the visible region, relatively narrow bands with vibronic progressions of ∼1500 cm−1 appear, and have been assigned to bpy or phen-based, spin-orbit coupling enhanced, 1π → 3π electronic transitions. Also present in the visible region are ligand-to-metal charge transfer bands (LMCT) arising from π(bpy) → t2g(OsIII) or π(phen) → t2g(OsIII) transitions. In the near infrared, two broad absorption features appear for oxidized forms [OsIII(bpy)3](PF6)3 and [OsIII(phen)3](PF6)3 arising from dπ-dπ interconfigurational bands characteristic of dπ5OsIII. They are observed at 4580 and 5090 cm−1 for [OsIII(bpy)3](PF6)3 and at 4400 and 4990 cm−1 for [OsIII(phen)3](PF6)3. The bpy and phen infrared vibrational bands shift to higher energy upon oxidation of Os(II) to Os(III). In the cation structure in [OsIII(bpy)3](PF6)3, the OsIII atom resides at a distorted octahedral site, as judged by ∠N-Os-N, which varies from 78.78(22)° to 96.61(22)°. Os-N bond lengths are also in general longer for [OsIII(bpy)3](PF6)3 compared to [OsII(bpy)3](PF6)2 (0.010 Å), and for [OsIII(phen)3](PF6)3 compared to [OsII(phen)3](PF6)2 (0.014 Å). Structural changes in the ligands between oxidation states are discussed as originating from a combination of dπ(OsII) → π (bpy or phen) backbonding and charge redistribution on the ligands as calculated by natural population analysis.  相似文献

Looking for optimised analogues of compound 2 that might be useful in colon cancer therapy, we here explore the in vitro cytotoxicity against MDA-MB 231 human breast carcinoma, A-549 human lung carcinoma and HT-29 human colon carcinoma cell lines of several analogues and derivatives. The effect of the R2-substituent and/or the introduction of an arylmethyl side-chain at C-3, as well as the presence of a double bond in the skeleton or a methoxy group at C-1 have been investigated. New 6,15-iminoisoquino[3,2-b]3-benzazocine compounds, related to the saframycin family, in which the C(7)–N(8)–C(9)-substructure contains a lactam function, a fused oxazolidine or an aminonitrile function were also studied, and many of them showed low micromolar GI50 values.  相似文献

A synthesis of 25-hydroxy[23,24-3H]vitamin D3 leading to a radiochemically pure product with a specific acitivity of 78 Ci/mmol is described. The structure of the product was confirmed by comparison with unlabeled material and its biological activity was established by in vitro conversion to 1α,25-dihydroxy[23,24-3H]vitamin D3 using the chick kidney 1α-hydroxylase system.  相似文献

Serotonin (5HT) binding sites were studied in goldfish retinal membranes by radioligand experiments. The binding site of [3H]5HT was sensitive to pre-treatment of the membranes at 40° or 60° C. 5HT and 5-methoxy-N,N-dimethyltryptamine were the best inhibitors of [3H]5HT binding to retinal membranes. The 5HT2 agonist, 1-(-naphtyl)piperazine, was also a potent inhibitor, however, (+)-1-2,5-dimethoxy-4-iodopheny1-2-aminopropane was less efficient. The catecholaminergic agents haloperidol and clonidine did not display an important inhibition. Propranolol, also reported as 5HT1B antagonist, was a relatively potent blocker. Monoamine uptake blockers did not show potent inhibition. The GTP analog, GppNHp, inhibited the binding. The iterative analysis of saturation curves revealed two classes of binding sites, a high affinity component (Bmax 2.45 pmol/mg of protein, kd 6.86 nM), and a low affinity component (Bmax 53.46 pmol/mg of protein, Kd 232.07 nM). Analysis of the association and dissociation kinetics suggested a binding site (Kd 2 nM). The semilogarithmic plot of the dissociation kinetics gave curves concave to the upper side. The selectivity of the binding and the inhibition by GppNHp suggest the existance of 5HT1 receptors in goldfish retina. Ameliorating effects of ginseng were observed on neuronal cell death associated with ischemia or glutamate toxicity. Ginseng saponins are transformed by intestinal microflora and the transformants would be absorbed from intestine. In the present study, we have investigated the effects of transformed ginsenoside Rg3, Rh2 and compound K on the modulation of NMDA receptor and GABAA receptor binding in rat brain. The NMDA receptor binding was analyzed by quantitative autoradiography using [3H]MK-801 binding, and GABAA receptor bindings were analyzed by using [3H]muscimol and [3H]flunitrazepam binding in rat brain slices. Ginsenoside Rg3, Rh2 and compound K were infused (10 g/10 l/h) into rat brain lateral ventricle for 7 days, through pre-implanted cannula by osmotic minipumps (Alzet, model 2ML). The levels of [3H]MK-801 binding were highly decreased in almost all regions of frontal cortex and hippocampus by ginsenoside Rh2 and compound K. The levels of [3H]muscimol binding were elevated in part of frontal cortex and granule layer of cerebellum by the treatment of ginsenoside Rh2 and compound K. However, the [3H]flunitrazepam binding was not modulated by any tested ginsenosides. Ginsenoside Rh2 and compound K induced the downregulation of the [3H]MK-801 binding as well as upregulation of the and [3H]muscimol binding in a region-specific manner after prolonged infusion into lateral ventricle. However, ginsenoside Rg3 did not show the significant changes of ligand bindings. In addition, ginsenoside Rh2 decreased the expression of nNOS in the hippocampus although Rg3 decreased the expression in the cortex. These results suggest that biotransformed ginsenoside Rh2 and compound K could play an important role in the biological activities in the central nervous systems and neurodegenerative disease.  相似文献

The electronic structure and spectroscopic properties of [Au3(μ-C(OEt) = NC6H4CH3)3]n-(C6F6)m and [Au3(μ-C2,N3-bzim)3]n-(Ag+)m were studied at the B3LYP, PBE and TPSS levels. The interaction between the [Au3] cluster and L (C6F6, Ag+) was analyzed. Grimme’s dispersion correction is used for those functionals. Weak π-interactions (Au-C6F6) were found to be the main contribution short-range stability in the models; while in the models with Ag+, an ionic interaction is obtained. The absorption spectra of these models at the PBE level agree with the experimental spectra.  相似文献

We have shown that diazepam (ED50 2.4 M), flunitrazepam (ED50 10.2 M) and Ro5-4864 (ED50 5 M) are able to enhance both total and specific [3H]phenytoin binding. Picrotoxin (IC50 1.43 M) and chloride, either NaCl or KCl (IC50 42.4 M) inhibit both the increase in total and specific binding of [3H]phenytoin, Ro 15-1788 does not. The optimum time for this enhancement was 3–4 hours. While the ED50's for the benzodiazepines are high their order of potency suggests that an involvement of both the peripheral type benzodiazepine receptor and the GABA-chloride ionophore complex is likely. Clonazepam (IC50 23 M), oxazepam (IC50 12 M) chlordiazepoxide (IC50 35 M) and Ro8682-10, a convulsant benzodiazepine (IC50 16 M) all inhibit both total and specific [3H]phenytoin binding. These effects were not blocked by chloride ions, picrotoxin or Ro 15-1788, and reached equilibrium within 45 minutes. This order of potency also parallels that for the peripheral benzodiazepine receptor in rat brain. These data suggest the presence of a micromolar benzodiazepine receptor site which may play a role in the control of CNS excitability. Nitrazepam, medazepam, bromazepam and the tetralobenzodiazepines U38335, U42794, U43434, and U37834 had no effect on total or specific [3H]phenytoin binding nor on the actions of the other benzodiazepines described in concentrations up to 50 M.  相似文献

The effects of chronic pentobarbital (PB) treatment on the binding characteristics of [3H]flunitrazepam (FLU) in rat brain were examined. Saline or sodium PB (500 g/10l/hr) was infused into the lateral cerebral ventricles of rats for 6 days using osmotic pumps. Immediately before withdrawal, there were no significant differences in [3H]FLU binding constants (KD and Bmax) between saline and PB groups. However, 24 hr withdrawal caused an increase in Bmax with no changes in KD. The enhancement of [3H]FLU binding by in vitro addition of chloride ions and PB was not affected after the PB infusion. The PB enhancement of [3H]FLU binding was inhibited by the convulsant, picrotoxicin. PB withdrawal did not cause significant differences in the binding constants of [3H]Ro 15-1788, a benzodiazepine (BZ) antagonist, between the saline and PB groups. Pretreatment of membranes with 0.02 mM of 3-[(3-cholamidopropyl)-dimethylammonio]-1-propanesulfonate (CHAPS), a zwitterionic detergent, caused decreases in both KD and Bmax in FLU binding in PB-withdrawal membrane, but not in the saline-treated membrane. The enhancement of [3H]FLU binding by chloride ions and PB was not affected by the CHAPS treatment. These results suggest that the change in BZ receptors induced by PB withdrawal is functionally linked to the GABA-BZ-barbiturate receptor complex and that PB withdrawal induces some conformational changes in BZ receptors.  相似文献

The changes of [3H]yohimbine and [3H]clonidine binding sites in rat vas deferens on treatments with adenosine receptor agonists (2-chloroadenosine, adenosine) or reserpine were examined. Treatment with adenosine agonist in vitro increased [3H]clonidine binding sites but had no influence on affinity and number of binding sites of α2-antagonist, [3H]yohimbine. Amount of [3H]yohimbine binding sites was found to be higher than that of [3H]clonidine with or without the treatment. Inhibition curves of α2-agonists, clonidine and norepinephrine, on [3H]yohimbine binding were less than unity though α2-antagonist inhibited with about 1.0 of nH. The treatment with adenosine agonist reduced the IC50 value of agonists on the [3H]yohimbine binding but had no influence on the inhibitory effect of antagonist. These effect of adenosine agonists was completely blocked by theophylline. Accordingly it was considered that activation of adenosine receptor caused configurational change in α2-adrenergic receptor from low affinity state for agonist to the high affinity state, though both states had same affinity for antagonist.On the other hand, treatment with reserpine in vivo increased the affinity of clonidine for α2-adrenergic receptors and also increased the amount of the α2-receptors.  相似文献

The binding of (1)-[3H]vesamicol was characterized in several subcellular fractions and brain regions of the rat. Binding to a lysed P2 fraction from the rat cerebral cortex reached equilibrium within 4 min at 37°C and was reversible (dissociation half-time 4.9 min). At least two binding affinities were found in P2 fractions from the cerebral cortex (Kd:21 nM and 980 nM), striatum (Kd:28 nM and 690 nM), and cerebellum (Kd:22 nM and 833 nM). High affinity Bmax values were highest in striatum (1.17 pmol/mg protein), followed by cerebellum (0.67 pmol/mg protein), and cerebral cortex (0.38 pmol/mg protein). Low affinity Bmax values were highest in cerebellum (5.2 pmol/mg protein), with similar values for cerebral cortex (3.7 pmol/mg protein) and striatum (3.8 pmol/mg protein). High affinity but not low affinity binding in each brain region was stereospecific. Another inhibitor of vesicular ACh-transport also displaced 1-vesamicol binding potently (IC50:17 nM) and efficaciously (over 90%). Both high affinity and low affinity Bmax values for [3H]vesamicol-binding were highest in a partially purified synaptic vesicle fraction, followed by puriffied synaptosomes, crude membranes and P2 fractions. Specific binding was not observed in a mitochondria-enriched fraction. Crude membrane preparations of primary, neuron-enriched whole brain cultures also exhibited high (64 nM) and low affinity (1062 nM) [3H]vesamicol binding. Isoosmotic replaement of 0.18 M KCl in the binding-buffer with NaCl had no effect on binding. These results suggest that at least some high affinity [3H]vesamicol binding in rat brain preparations may be associated with synaptic vesicles, some of which may not be cholinergic in origin.  相似文献
