Heat shock proteins (HSPs) are ubiquitous protective proteins that play crucial roles in plant development and adaptation to stress, and the aim of this study is to characterize the HSP gene in alfalfa. Here we isolated a small heat shock protein gene (MsHSP17.7) from alfalfa by homology-based cloning. MsHSP17.7 contains a 477-bp open reading frame and encodes a protein of 17.70-kDa. The amino acid sequence shares high identity with MtHSP (93.98 %), PsHSP17.1 (83.13 %), GmHSP17.9 (74.10 %) and SlHSP17.6 (79.25 %). Phylogenetic analysis revealed that MsHSP17.7 belongs to the group of cytosolic class II small heat shock proteins (sHSP), and likely localizes to the cytoplasm. Quantitative RT-PCR indicated that MsHSP17.7 was induced by heat shock, high salinity, peroxide and drought stress. Prokaryotic expression indicated that the salt and peroxide tolerance of Escherichia coli was remarkably enhanced. Transgenic Arabidopsis plants overexpressing MsHSP17.7 exhibited increased root length of transgenic Arabidopsis lines under salt stress compared to the wild-type line. The malondialdehyde (MDA) levels in the transgenic lines were significantly lower than in wild-type, although proline levels were similar between transgenic and wild-type lines. MsHSP17.7 was induced by heat shock, high salinity, oxidative stress and drought stress. Overexpression analysis suggests that MsHSP17.7 might play a key role in response to high salinity stress.  相似文献

The photosynthetic rate, light saturation point, light compensation point, changes in the MDA and SOD activities, and protein expression of two different drought-resistance species, Spiraea fritschiana and Spiraea trichocarpa, were assessed in this study. Furthermore, the drought-resistant physiological mechanisms of both species were analyzed at the protein level. The photosynthetic capacities of two Spiraea species decreased under drought stress, and the light saturation point and light compensation point decreased. However, their capacities to use weak light increased. Spiraea fritschiana, which demonstrated a stronger drought resistance, showed a better ability to adapt to weak light than S. trichocarpa. The content of MDA in S. fritschiana was notably lower than that in S. trichocarpa, indicating that the concentration of the membrane peroxidation products of S. fritschiana was less than those of S. trichocarpa. Compared with S. trichocarpa, S. fritschiana’s SOD activity was higher, and its ability to remove ROS was also better. Sixty-six proteins were identified with significantly different expression behavior and included regulatory, redox homeostasis, metabolism and energy, and cytoskeleton proteins. The results showed that the photosynthesis of S. trichocarpa was significantly affected by the drought stress. Enzymes in photosynthesis changed significantly; the expression of the RuBisCo large subunit decreased; and RuBisCo carboxylase, the chlorophyll a–b binding protein, ATP synthase, OEC 33 kD photosystem II protein and 23 kD OEC protein greatly increased. In addition, four antioxidant enzymes greatly increased, GroES chaperonin decreased, and eIF5A significantly increased under light stress. When S. fritschiana Schneid encountered serious drought stress, in addition to those enzymes that changed significantly under light drought stress in S. trichocarpa Nakai, NAD(P)H-quinone oxidoreductase and eIF5A were up-regulated. Specifically, three heat-shock proteins were induced. The expression of the enzymes of the two Spiraea that were related to photosynthesis, oxidation–reduction and regulation were all affected, but their species and expression patterns were different. In S. trichocarpa Nakai and S. fritschiana Schneid, there were significant changes in the proteins related to energy metabolism and the proteins related to energy transport, respectively. Thus, we considered that, in the case of protein involvement, the differences in the metabolic pathways and adjustment levels might contribute to S. trichocarpa having a weaker drought tolerance than S. fritschiana.  相似文献

DNA-binding proteins (Dps) induced during starvation play an important role in gene regulation and maintaining homeostasis in bacteria. The nitrogen-fixing cyanobacterium, Anabaena PCC7120, has four genes annotated as coding for Dps; however, the information on their physiological roles is limiting. One of the genes coding for Dps, ‘all3940’ was found to be induced under different abiotic stresses in Anabaena and upon overexpression enhanced the tolerance of Anabaena to a multitude of stresses, which included salinity, heat, heavy metals, pesticide, and nutrient starvation. On the other hand, mutation in the gene resulted in decreased growth of Anabaena. The modulation in the levels of All3940 in Anabaena, achieved either by overexpression of the protein or mutation of the gene, resulted in changes in the proteome, which correlated well with the physiological changes observed. Proteins required for varied physiological activities, such as photosynthesis, carbon-metabolism, oxidative stress alleviation, exhibited change in protein profile upon modulation of All3940 levels in Anabaena. This suggested a direct or an indirect effect of All3940 on the expression of the above stress-responsive proteins, thereby enhancing tolerance in Anabaena PCC7120. Thus, All3940, though categorized as a Dps, is possibly a general stress protein having a global role in regulating tolerance to multitude of stresses in Anabaena.  相似文献

Members of the plant glycine-rich RNA-binding proteins (GR-RBPs) family have been reported in flowering, development, circadian rhythms, biotic and abiotic stresses. Particularly, GR-RBPs are reported to function as RNA chaperones, promoting growth and acclimation during cold shock. It is indispensable to further question the efficacy and mechanism of GR-RBPs under various environmental strains. Monitoring the expression of stress-regulated proteins under stress conditions has been a beneficial strategy to study their functional roles. In an effort to elucidate the NtGR-RBP1 function, stress markers such as salinity, drought, low temperature and heat stresses were studied. The NtGR-RBP1 gene was expressed in E. coli followed by the exposure to stress conditions. Recombinant E. coli expressing NtGR-RBP1 were more tolerant to stresses, e.g., salinity, drought, cold and heat shock. Recombinants exhibited higher growth rates compared to control in spot assays. The tolerance was further confirmed by monitoring the growth in liquid culture assays. Cells expressing NtGR-RBP1 under salt (500 mM NaCl), drought (20% PEG), cold (4 and 20 °C) and heat stresses (50 °C) had enhanced growing ability and better endurance. Our study supports the notion that the protective role of NtGR-RBP1 may contribute to growth and survival during diverse environmental stresses.  相似文献

The heat stress resistance of Drosophila melanogaster females carrying a hypomorphic mutation of the DILP6 insulin-like protein gene (dilp6 41 ) under a change in the level of stress-related hormones (juvenile hormone and octopamine) is studied. It is revealed that the dilp6 41 mutation decreases the heat stress resistance of mature D. melanogaster females. An experimental decrease in the level of juvenile hormone is shown to restore the stress resistance of mutant females to the level of stress resistance observed in wild type Canton S females. These data suggest that the effects of the dilp6 41 mutation on the stress resistance of females are mediated by an increased level of juvenile hormone. An experimental increase in the octopamine level that causes an increase in juvenile hormone level supports this hypothesis: the resistance to heat stress decreases in females of both lines and this decrease is more significant in mutant females than in the control line. Thus, it is established for the first time that the effect of the hypomorphic dilp6 gene mutation on the heat stress resistance of D. melanogaster females is mediated by juvenile hormone.  相似文献

Salt stress is a critical factor that affects the growth and development of plants. Salicylic acid (SA) is an important signal molecule that mitigates the negative effects of salt stress on plants. To elucidate salt tolerance in large pink Dianthus superbus L. (Caryophyllaceae) and the regulatory mechanism of exogenous SA on D. superbus under different salt stresses, we conducted a pot experiment to evaluate leaf biomass, leaf anatomy, soluble protein and sugar content, and the relative expression of salt-induced genes in D. superbus under 0.3, 0.6, and 0.9% NaCl conditions with and without 0.5 mM SA. The result showed that exposure of D. superbus to salt stress lead to a decrease in leaf growth, soluble protein and sugar content, and mesophyll thickness, together with an increase in the expression of MYB and P5CS genes. Foliar application of SA effectively increased leaf biomass, soluble protein and sugar content, and upregulated the expression of MYB and P5CS in the D. superbus, which facilitated in the acclimation of D. superbus to moderate salt stress. However, when the plants were grown under severe salt stress (0.9% NaCl), no significant difference in plant physiological responses and relevant gene expression between plants with and without SA was observed. The findings of this study suggest that exogenous SA can effectively counteract the adverse effects of moderate salt stress on D. superbus growth and development.  相似文献

It is important to reveal the mechanism of plants coping with heat stress, which results in a severe retardation in crop growth and development. Although Synaptotagmin A (SYTA) regulates the cell endocytosis and the plasma membrane reparation of Arabidopsis, its roles in heat resistance are not well understood. In this study, we obtained the new finding that SYTA is related to the heat resistance of Arabidopsis. In the SYTA overexpression strains, the germination rate of the seeds and the survival rate of the seedlings improved after heat shock treatment, and their degree of membrane peroxidation was reduced. However, syta mutant showed the opposite results. Meanwhile, the expressions of some heat stress signal pathway genes were higher in SYTA overexpression strains than that in wild-type strains, and were lower in syta mutant strains. These results suggested that SYTA responded positively to heat shock and was involved in the heat stress signal pathway.  相似文献

Stress-associated proteins (SAPs) are a novel class of zinc finger proteins that extensively participate in abiotic stress responses. To date, no overall analysis and expression profiling of SAP genes in woody plants have been reported. Populus euphratica is distributed in desert regions and is extraordinarily adaptable to abiotic stresses. Thus, it is regarded as a promising candidate for studying abiotic stress resistance mechanisms of woody plants. In this study, 18 non-redundant SAP genes were identified from the genome of P. euphratica using basic local alignment search tool algorithms and functional domain verification. Among these 18 PeuSAP genes, 15 were intronless. To investigate the evolutionary relationships of SAP genes in P. euphratica and other Salicaceae plants, phylogenetic analyses were performed. Subsequently, the expression profiles of the 18 PeuSAP genes were analyzed in different tissues and under various stresses (drought, salt, heat, cold, and abscisic acid (ABA) treatment) using quantitative real-time PCR. Tissue expression analysis indicated that PeuSAPs showed no tissue specificity. PeuSAPs were induced by multiple abiotic stresses, especially drought, salt, and heat stresses, perhaps because of abundant cis-acting heat shock elements and drought-inducible elements in the promoter regions of the PeuSAPs. Moreover, single nucleotide polymorphisms (SNPs) variant analysis revealed many synonymous and non-synonymous SNPs in PeuSAP genes, but the zinc finger structure was conserved during evolution. These results provide an overview of the SAP gene family in P. euphratica and a reference for further functional research on PeuSAP genes.  相似文献

Bacterial species of Bacillus, Lactobacillus, and Bifidobacterium in the intestinal tract have been used as probiotics. Selections for probiotic candidates by the culture-based approaches are time-consuming and labor-consuming. The aim of this study was to develop a new method based on sequencing strategies to select the probiotic Bacillus, Lactobacillus, and Bifidobacterium. The Illumina-based sequencing strategies with different specific primers for Bacillus, Clostridium, and Bifidobacterium were applied to analyze diversity of the genera in goat feces. The average number of different Bacillus, Clostridium, and Bifidobacterium OTUs (operational taxonomic units) at the 97% similarity level ranged from 1922 to 63172. The coverage index values of Bacillus, Clostridium, and Bifidobacterium calculated from the bacterial OTUs were 0.89, 0.99, and 1.00, respectively. The most genera of Bacillus (37.9%), Clostridium (53%), and Bifidobacterium (99%) were detected in goat feces by the Illumina-based sequencing with the specific primers of the genera, respectively. Higher phylogenetic resolutions of the genera in goat feces were successfully established. The results suggest that the selection for probiotic Bacillus, Clostridium, and Bifidobacterium based on the Illumina sequencing with their specific primers is reliable and feasible, and the core Bacillus, Clostridium, and Bifidobacterium species of healthy goats possess the potentials as probiotic microbial consortia.  相似文献

Heat stress is one of the main abiotic stresses that limit plant growth. The effects of high temperature on oxidative damage, PSII activity and D1 protein turnover were studied in three wheat varieties with different heat susceptibility (CS, YN949 and AK58). The results showed that heat stress induced lower lipid peroxidation in AK58 and YN949 than CS, which was related to different changes of SOD, CAT, POD and H2O2. Similarly, AK58 and YN949 performed better PSII photochemical efficiency (Fv/Fm, ΦPSII and ETR) under high temperature, which was attributed to rapid synthesis and degradation of D1 protein. Moreover, higher expression of D1 protein turnover-related genes (PsbA, STN8, PBCP, Deg1, Deg2, Deg5, Deg8, FtsH1/5 and FtsH2/8) and SOD activity in AK58 and YN949 under normal conditions also established a basis for acclimatizing high temperatures, thereby alleviating PSII photoinhibition and reducing oxidative damage when exposed to heat stress.  相似文献

In both prokaryotes and eukaryotes, the survival at temperatures considerably exceeding the optimum is supported by intense synthesis of the so-called heat shock proteins (HSPs), which act to overcome the adverse effects of heat stress. Among mycoplasmas (class Mollicutes), which have significantly reduced genomes, only some members of the Acholeplasmataceae family possess small HSPs of the α-crystallin type. Overproduction of a recombinant HSP IbpA (Hsp20) from the free-living mycoplasma Acholeplasma laidlawii was shown to increase the resistance of Escherichia coli to short-term heat shock. It has been long assumed that IbpA prevents protein aggregation and precipitation thereby increasing viability of E. coli cells. Several potential target proteins interacting with IbpA under heat stress were identified, including biosynthetic enzymes, enzymes of energy metabolism, and components of the protein synthesis machinery. Statistical analysis of physicochemical properties indicated that IbpA interaction partners significantly differ in molecular weight, charge, and isoelectric point from other members of the E. coli proteome. Upon shortterm exposure to increased temperature, IbpA was found to preferentially interact with high-molecularweight proteins having a pI of about 5.1, significantly lower than the typical values of E. coli proteins.  相似文献

Small heat shock proteins (sHSPs) have been shown to be involved in stress tolerance. However, their functions in Prunus mume under heat treatment are poorly characterized. To improve our understanding of sHSPs, we cloned a sHSP gene, PmHSP17.9, from P. mume. Sequence alignment and phylogenetic analysis indicated that PmHSP17.9 was a member of plant cytosolic class III sHSPs. Besides heat stress, PmHSP17.9 was also upregulated by salt, dehydration, oxidative stresses and ABA treatment. Leaves of transgenic Arabidopsis thaliana that ectopically express PmHSP17.9 accumulated less O2 ? and H2O2 compared with wild type (WT) after 42 °C treatment for 6 h. Over-expression of PmHSP17.9 in transgenic Arabidopsis enhanced seedling thermotolerance by decreased relative electrolyte leakage and MDA content under heat stress treatment when compared to WT plants. In addition, the induced expression of HSP101, HSFA2, and delta 1-pyrroline-5-carboxylate synthase (P5CS) under heat stress was more pronounced in transgenic plants than in WT plants. These results support the positive role of PmHSP17.9 in response to heat stress treatment.  相似文献

Phosphoenolpyruvate carboxylase (PEPC) is a tightly controlled enzyme located at the core of plant carbohydrate metabolism. Plant PEPCs belong to a small multigene family encoding several plant-type PEPC genes, along with at least one distantly related bacterial-type PEPC gene. The PEPC genes have been intensively studied in Arabidopsis, but not in peanut (Arachis hypogaea L.). Previously, we isolated five PEPC genes (AhPEPC1, AhPEPC2, AhPEPC3, AhPEPC4 and AhPEPC5) from peanut. Here, due to the sequencing of the peanut genome, we analyzed the complexity of its PEPC gene family, including phylogenetic relationships, gene structure and chromosome mapping. The results showed that AhPEPC1, AhPEPC2, AhPEPC3 and AhPEPC4 encoded typical plant-type enzymes, while AhPEPC5 was a bacterial-type PEPC. The recombinant proteins of these genes were expressed in Escherichia coli, and the calculated molecular weights of the recombinant proteins were 110.8 kD (AhPEPC1), 110.7 kD (AhPEPC2), 110.3 kD (AhPEPC3), 110.8 kD (AhPEPC4), and 116.4 kD (AhPEPC5). The expression patterns of AhPEPC1-5 were analyzed under cold, salt and drought conditions. Our results indicated that the expression of AhPEPC3 was rapidly and substantially enhanced under abiotic stress, whereas the expression of AhPEPC1 and AhPEPC2 was slightly enhanced under certain stress conditions. Some genes were down-regulated in leaves under stress: AhPEPC1, AhPEPC4 and AhPEPC5 under salt stress and AhPEPC4 and AhPEPC5 under drought stress. These results suggest that peanut PEPC proteins may differ in their functions during acclimation to abiotic stresses.  相似文献

Background and aims

Soil salinization with high pH condition is a major abiotic stress to plant growth and crop productivity. Helianthus tuberosus L. is an important stress tolerant plant and can survive in the saline-alkali soil and semiarid areas. The aim of this study is to identify the effect of alkali stress on H. tuberosus through global proteomics analysis and improve understanding of the alkalinity resistance of plants.

Methods

H. tuberosus seedlings were exposed to different level alkali stress for 7 days. Protein profiling was quantified by conducting MS-based comparative proteomics analysis. RT-PCR study was carried out to analyze the mRNA expression levels of candidate alkali stress response proteins.

Results

The response of H. tuberosus to alkali stress was detected at both physiological and molecular levels. 104 differentially expressed proteins from H. tuberosus leaves response to Na2CO3 treatment were successfully identified. Functional categorization of these identified proteins showed that the accumulation level of proteins involved in glycolysis, TCA cycle, PSI system, ROS scavenging and signal transduction increased under alkali stress.

Conclusions

Based on the observation of plant growth and the investigation of molecular regulation, H.tuberosus could resist certain alkali stress by modulating carbohydrate metabolism and redox homeostasis. These findings provide a new sight into the underlying molecular mechanisms of alkali resistance in plant.
