以10种我国市场上常见的黄瓜品种为材料,建立了一个简单高效的黄瓜子叶离体再生系统。从种子萌发到获得再生植株仅需6周。子叶外植体在MS=0.5mg/L BA培养基上不经愈伤组织诱导阶段而直接走器官发生途径。4~5天的子叶外 植体的不定芽发生能力最强。供试10种基因型中,“碧春”和“甜翠绿”的再生率高达100%。待不定芽长度超过1.0mc时,直接将不定芽转移到MS培养基生根成苗,幼苗移栽温室后正常结实  相似文献

本文以"京欣1号"母本和"伊选"西瓜4天苗龄子叶为外植体,研究离体培养植株高频率再生体系.结果表明"伊选"子叶远轴端外植体的再生频率仅为10%,子叶近轴端外植体在5mg/LBA+0.1mg/L IAA的激素组合下植株再生频率为100%,平均每个外植体的丛生芽数在所有组合中最多,为10.3个;"京欣1号"母本子叶近轴端外植体在2mg/L BA+0.5mg/L IAA激素组合下植株再生频率为100%,平均每个外植体的丛生芽数在所有组合中最多,达6.9个.本试验条件下,子叶近轴端外植体接种4天即分化出不定芽,至再生苗的移栽仅需40天,在MS+0.1mg/L NAA的生根培养基上的生根率为97.3%,移栽成活率达98.5%.  相似文献

以6个草莓品种为试材,研究了影响草莓不定芽再生的各种因素,建立离体叶片高效再生系统。结果表明,外植体基因型、激素种类及配比、叶龄等是影响草莓再生的主要因子,其中‘鬼露甘’叶片最佳芽诱导培养基为MS 2.0 mg/L 6-BA 0.1 mg/L IBA,‘嫜姬’叶片愈伤组织的诱导以MS 3 mg/L 6-BA 0.2 mg/L 2,4-D较好,而且1周左右的暗培养可以防止外植体的褐化。芽伸长的最适培养基为MS 0.5 mg/L 6-BA 0.5 mg/L IBA,生根的最适培养基为MS 0.2 mg/L IBA,试管苗移栽后成活率为87%。  相似文献

成熟种子的子叶离体后,在无外源激素和营养供应的滤纸保湿(26±1℃,2.92Wm-2,14h光照)条件下培养。0—4d内子叶的面积大小和鲜重迅速增加而干重基本保持不变,4d后子叶面积和鲜重继续增加但速度减慢,干重迅速下降;呼吸速率和叶绿素含量4d前稳定上升,第4天达到峰值后下降.离休后6d内,子叶中矿质元素含量发生了一定的变化;除Put含量在第2天有所下降外,子叶中Spm、SPd和Cad含量均不断上升,但上升幅度不相同,因此,离体并没有削弱黄瓜子叶多胺合成;GA3和ABA含量有不同程度的上升,但IAA和ZT含量剧增,表明离休子叶有很好的激素合成能力.离体后2d内,子叶中DNA含量、RNA含量、酸溶蛋白含量均成倍增加,此后DNA含量迅速下降,酸溶蛋白含量呈彼动状,RNA含量基本保持同样水平;0—4d碱溶蛋白含量不断减少,这表明子叶离体0—4d有活跃的核酸、蛋白质代谢活动.  相似文献

以‘08S555’和‘陕秋白’大白菜子叶为外植体,研究了激素配比和AgNO3对不定芽再生的影响。结果表明:‘陕秋白’的再生率高于‘08S555’,但出芽系数比‘08S555’低。在6-BA与NAA组合时,‘陕秋白’再生率最高,为68.33%,‘08S555’为28.33%;再生系数分别为1.09和2.81。在TDZ与NAA组合时,二者不定芽的再生频率分别为79.17%和45.83%;再生系数分别为1.24和2.55。这说明与6-BA相比,TDZ与NAA组合对两个大白菜子叶再生率的效果更好。AgNO3与细胞分裂素及生长素配合使用,能提高‘陕秋白’子叶不定芽的再生频率。当TDZ浓度为0.3mg·L^-1、NAA为0.5mg·L^-1和AgNO3为6mg·L^-1时,‘陕秋白’再生率最高,达到87.50%。  相似文献

以对生玉米的雌、雄幼穗为外植体,研究了不同质量浓度激素及其组合对愈伤组织诱导、再分化苗和试管苗生根的影响,建立了对生玉米离体培养再生体系。结果表明:长度为15-17 mm的雌、雄幼穗能够诱导出质量较好的愈伤组织,但只有来自于雄幼穗的愈伤组织才能再生成苗。适合愈伤组织诱导的培养基为Ms+1.5-2.0 mg·L-12,4-D+0.5 mg·L-16-BA+0.5 mg·L-1 NAA+500 mg·L-1脯氨酸+1000 mg·L-1水解酪蛋白+30 g·L-1蔗糖;适合愈伤组织再分化培养基为MS+1.5-2.0 mg·L-1 6-BA+0.1 mg·L-1 NAA+500 mg·L-1水解酪蛋白+30 g·L-1 蔗糖;适合试管苗生根培养基为1/2 MS+0.25-0.5 mg·L-1 IBA+20 g·L-1蔗糖。  相似文献

Summary A system has been developed for efficient regeneration of shoots from Brassica campestris in vitro. Using 4-day old cotyledons with petioles as expiants and a combination of BA and NAA in the regeneration media, up to 70% of expiants produced shoots after 2 weeks in culture. The optimal conditions for regeneration were found to include a BA concentration of 2mgL–1 and NAA concentration of 1mgL–1. Light intensity had a profound effect on regeneration potential. The use of silver ions as an inhibitor of ethylene action reduced regeneration rates in this system. Rooting occured simultaneously with shoot formation on these media and the resultant shoots could be rooted readily on minimal medium. The genotype dependency was investigated and indicated that this method would be widely applicable to B. campestris cultivars. Regeneration of one cultivar, a high erucic acid type (R-500), was inefficient in the system described here. Histological studies indicated the development of multiple shoot primordia from the petiolar cut ends of the expiants after the initiation of meristematic activity in the cells about 100m from the cut site within 2 days of culture initiation. The system described is compatible with previously reported Agrobacterium — mediated transformation protocols involving cotyledonary petioles.  相似文献

An effective protocol has been developed for plant regeneration from cotyledon explants of Swainsona salsula Taubert (Saline swainsona), a medicinal and agronomic shrub. Adventitious shoots were obtained from 83.2% of cotyledon explants from 3-day seedlings cultured on Murashige and Skoog (MS) medium containing 2.0 mg l−1 thidiazuron (TDZ), with an average of 9.3 shoots per explant. Individual elongated shoots were rooted on half strength MS medium supplemented with 2.0 mg l−1 indole-3-butyric acid (IBA), with 59.3% success. Regenerated plants with well developed shoots and roots were successfully transferred to soil, without detectable variants. Histological observation revealed that shoots developed from cotyledon explants via organogenesis, with little callus. Twenty-one lines of Brassica juncea were screened for frequency of in vitro shoot formation from cultured cotyledons of 8-day old seedlings. All brown-seeded Indian lines showed good shoot regeneration with 20–50% of cotyledons giving rise to shoots. Very poor shoot regeneration (0–12%) was observed for the predominantly yellow-seeded Chinese/European lines, including two erucic acid-free lines. Germination of seedlings in hydroponic nutrient solution markedly enhanced subsequent shoot regeneration frequency from cotyledons of three Indian lines but had no effect on shoot formation from the recalcitrant Chinese/European lines.  相似文献

In vitro plant regeneration has been obtained from Capsicum annuum cvs. Pico and Piquillo. Shootbuds were induced from hypocotyl and cotyledon segments after 15–20 days of culture on MS basal medium supplemented with IAA and BAP or Zeatin. Shoot-buds grew into rosettes that rooted in MS plus NAA (0.1 mg/l) and IBA (0.05 mg/l) after 15 days. The small plantlets were successfully transferred to pots with a mixture of peat and perlite and maintained under greenhouse conditions. Five different genotypes from in vitro as well as greenhouse grown melon plants were shown to be highly responsive for in vitro shoot formation from leaf explants when placed on basic MS medium supplemented with 1 mg/l 6-benzylaminopurine. In addition, a very suitable regeneration system was obtained when cotyledon pieces of mature seeds were incubated on the same culture medium. In this case, the first shoots already appeared after 10 days of incubation, and hundreds of shoots were formed on the cut surface 3 to 4 weeks later. Summary A system using cotyledon pieces as explants and a BAP/NAA containing medium was developed for in vitro mass propagation of Sesbania grandiflora, a tropical nitrogen-fixing leguminous tree. The age and the lighting conditions of seedlings providing the explants were shown to be critical factors for both bud induction and bud elongation. Optimal choice for an efficient and reproducible bud induction process consisted of dark-grown seedlings, 24/36 h-old-post-imbibition, that yielded up to 96% of explants producing more than 30 buds each, after one week in culture. Bud development occurred throughout a direct organogenesis pathway, from the proximal and adaxial cut surface of the explants as proved by histological studies. Additional sites of regeneration were also obtained after wounding on the epidermal surface of explants, suggesting a large distribution of regenerative cells all along the explants. Bud elongation, i.e. stem differentiation and leaf growth, was improved by bud isolation from cotyledon explants and their further subculture in liquid bud elongation media for one week. Ultrasonic treatment (0.5–2 min) stimulated multiple shoot regeneration to high levels in vitro from recalcitrant cotyledon explants of commercial squash (Cucurbita pepo L.) cultivars Ma’yan and Bareqet, on Murashige and Skoog [Physiol Plant 15:473–497, 1962] (regeneration) medium augmented with 4.4 μM benzyladenine. At this stage, unsonicated control explants regenerated only a few very small shoots or bud-like structures. Ultrasound also stimulated massive explant growth. Ultrasound treatment resulted in further multiple shoot production (five times greater than control) after explant transfer to elongation medium (Murashige and Skoog [Physiol Plant 15:473–497, 1962] medium with 0.44 μM benzyladenine and 2.9 μM gibberellic acid). Longer ultrasonic treatments (5 or 10 min) promoted multiple shoot regeneration and explant growth accompanied by hyperhydration. Scanning electron microscope observations showed that 2 min ultrasound changed the joint area between epidermal cells and removed some of the surface from the cotyledon epidermal cells, without gross surface injury to the explants. Longer periods of ultrasound (5–10 min) caused further surface erosion. Rubbing the explant contact surface with chloroform or sandpaper emulated the effect of sonication on shoot regeneration and explant growth, demonstrating that ultrasound exerts its morphogenic influence by surface removal. Sonication of explants from other batches of squash seeds (of cultivars Ma'yan and True French), that regenerated without such treatment, reduced regeneration and caused hyperhydration. This is the first report of stimulation of in vitro regeneration by ultrasound treatment. Summary. An efficient and fast regeneration system from cotyledon protoplasts was established for Arabidopsis thaliana accessions C24, Columbia, and Wassilewskija. Culture conditions and media compositions were optimised for the development of protoplasts embedded in thin alginate layers. Unexpectedly, the absence of cytokinins had a positive effect on cell development. Moreover, combined adjustment of -naphthylacetic acid and dicamba concentrations resulted in high plating efficiencies of up to 30%, followed by shoot regeneration within only 19 days after protoplast isolation. The protocol is reproducible, efficient, extremely fast, and regenerated plants are fertile. Summary A plant regeneration system applicable to 17 cowpea genotypes was developed. Cotyledons were initiated on 1/3 MS medium containing 15 to 35 mg N6-benzyladenine (BA) per 1 (66.6 to 155.3 μM) for 5 to 15 d. For shoot regeneration, the explants were transferred to a medium containing 1 mg BA per 1 (4.4 μM). Within 1 wk, shoot formation was visible at the proximal end of the cotyledons. Regeneration percentages (1% to 11%) and the numbers of shoots (4 to 12 per explant) were significantly influenced by genotype. Culture duration and BA concentration in the initiation stage significantly affected regeneration capacity. Explants initiated on media containing 15 mg BA per 1 for 5 d resulted in the highest percentage of explants capable of regeneration. Conversely, the highest number of shoots was obtained from explants initiated on media supplemented with 35 mg BA per 1. Whole plants were obtained on a plant growth regulator-free medium. To our knowledge, this is the first report of plant regeneration from U.S. commercial cowpea cultivars and breeding lines. This system is adaptable to diverse cowpea genotypes and will facilitate cowpea genetic transformation. In vitro plantlet regeneration systems for the seed geranium (Pelargonium x hortorum Bailey) using cotyledon, hypocotyl and root explants were optimized by studying the influence of seedling age, growth regulators and excision orientation on organogenesis. Indole-3-acetic acid combined with zeatin yielded the highest rate of shoot production on cotyledon explants (0.2–2 shoots per explant). More shoots were produced on explants cut from the most basal region of cotyledons from 2 to 4-day-old seedlings than from older seedlings or more distal cut sites. Hypocotyl explants produced the highest number of shoots, up to 40 shoots per explant, on indole-3-acetic acid (2.8–5.6 mM) + zeatin (4.6 mM) or thidiazuron (4.5 mM). Maximum shoot formation (0.3–1.4 shoots per explant) on root explants occurred when they were cultured on medium containing zeatin. Regenerated shoots rooted best on a basal medium containing no growth regulators.
