目的:研究海南汉族人群MICB等位基因的多态性与肺癌易感性之间的关联性。方法:采用PCR-SSP(PCR sequence-specific primers)和PCR-SBT(PCR sequence-based typing)方法对样本MICB等位基因的多态性进行检测。结果:肺癌患者中检出14种MICB等位基因;和对照组相比较,MICB*00502等位基因在肺癌患者组分布频率较少(43.5%vs 57.8%),MICB*016等位基因在肺癌患者组分布较多(5.9%vs 0.6%);MICB*016等位基因可能对肺癌易感(OR=11.19,95%CI:2.59-48.24,Pc0.05);MICB*00502等位基因可能对肺癌不易感(MICB*00502:OR=0.56,95%CI:0.42-0.76,Pc0.05)。结论:MICB等位基因的多态性与肺癌的易感性之间存在关联性。 相似文献

摘要目的:探讨新疆地区维吾尔族、汉族脑啡肽酶(Neprilysin,NEP)基因单核苷酸多态性与阿尔茨海默病(Alzheimer’SdiseaseAD)的关系。方法:对新疆地区维吾尔族、汉族≥50岁8284名人群进行AD流行病学调查,参照ADRDA.NINCDS的标准,选取散发性阿尔茨海默病(sporadicAlzheimer’s disease,SAD)患者209例(AD组)与正常对照220例(对照组),应用聚合酶链反应一限制性片段长度多态性技术(PCR)检测NEP基因多态性,采用病例一对照的关联分析方法对NEP基因rs3736187位点进行基因型和等位基因频率分析。结果:(1)新疆维吾尔族、汉族两民族AD组与对照组间NEP基因基因型频率和等位基因频率分布差异均有统计学意义(P〈0.05)。携带T等位基因个体出现AD的危险性高于携带c等位基因的个体(0R=1.981,P〈0.05)。(2)新疆维吾尔族、汉族不同民族之间比较NEP基因基因型频率和等位基因频率分布差异均无统计学意义(P〉0.05),而在同一民族中AD组和对照组之间比较NEP基因等位基因频率分布差异均具有统计学意义(P〈0.05)。(3)两个年龄分段(〈65岁及≥65岁)之间NEP基因基因型频率和等位基因频率分布差异均无统计学意义(P〉0.05),而在同一年龄段内部AD组与对照组间等位基因频率分布差异具有统计学意义(P〈0.05)。(4)男性、女性之间NEP基因基因型频率和等位基因频率分布差异均无统计学意义,而在女性AD组与对照组间等位基因频率分布差异有统计学意义(P〈0.05)。结论:NEP基因rs3736187位点基因型频率和等位基因频率在新疆维吾尔族、汉族两民族间的分布相似;NEP基因的T等位基因是SAD的危险因素,在新疆维吾尔族、汉族两民族及女性SAD的发病中起重要作用。 相似文献

探讨白介素17A基因多态性与胃癌预后的关系。129例研究对象纳入生存分析,分成死亡和存活两组,用基因测序的方法检测血液样本IL-17A基因SNP位点rs3748067、rs17880588基因型分布。rs3748067位点有3种基因型T/T、C/T、C/C,rs17880588位点有2种基因型A/G、G/G。比较存活组和死亡组之间的基因型分布频率和单点等位基因分布频率,发现rs3748067的基因型C/T在死亡组的分布频率较存活组高,基因型T/T和C/C在死亡组的分布频率低于存活组,两组之间分布频率差异有统计学意义(P0.05)。杂合型C/T基因型在存活组分布低于死亡组(OR=2.051,CI=0.016~1.420),该位点基因型杂合可能为胃癌预后的一种危险因素。rs17880588的两种基因型A/G、G/G在存活组和死亡组的分布频率差异无统计学意义(P0.05)。结论:IL-17A基因rs3748067位点SNP与胃癌预后可能有相关性。 相似文献

中国湖南人群GSTM1和GSTT1基因多态性与肺癌易感性关系的研究 总被引:3,自引:0,他引:3
应用病例-对照分析研究(对照组205例,肺癌病例组104例),抽提静脉血基因组DNA,采用PCR及多重PCR方法,检测谷胱甘肽转移酶GSTM1和GSTT1单独及联合缺失基因型的遗传多态性在中国湖南人群中肺癌患者和正常人群体中的分布,探讨这些多态性基因型与肺癌易感性的关系.结果显示GSTM1-/-基因型在湖南地区居民肺癌群体和正常对照人群中的频率分别为62.5%和46.3%(P<0.05);肺癌患者组GSTT1-/-基因型的频率(66.3%)显著高于正常对照组(42.4%)(P<0.05).GSTM1-/-和GSTT1-/-联合基因型在肺癌组和正常对照组中的频率分别为41.3%和22.4%(P<0.05).SPSS11.5软件统计学分析表明,这些基因型在肺癌患者组和正常对照组人群中的发生频率具有显著性差异.由此可知GSTM1基因缺失和GSTT1基因缺失分别与肺癌的易感性相关;GSTM1和GSTT1基因联合缺失与肺癌的发生和发展呈现显著正相关. 相似文献

目的:研究CYP4F3基因单核苷酸多态性(SNP)在中国汉族人群中的分布,为进一步研究该基因群体遗传学特征及与疾病易感性的相关性提供更为详实的数据。方法:对CYP4F3基因进行重测序,构建连锁不平衡模式,选择标签SNP在192例北京和424例广州汉族个体中进行基因分型。结果:CYP4F3基因重测序共检出30个SNP,连锁不平衡分析显示广州和北京地区人群的连锁不平衡模式不同,但选择的8个标签SNP的等位基因和基因型频率分布在2个人群中的差异无统计学意义。结论:中国北京地区汉族与广州地区汉族人群CYP4F3基因多态性无显著差异,但不同种族间存在差异。 相似文献

目的:探讨细胞色素P450 3A5基因(CYP3A5)单核苷酸多态位点rs3800959与氯吡格雷抵抗(Clopidogrel resistance,CR)发生的关系。方法:于2010年3月至2011年10月期间,连续入选在沈阳军区总医院心内科住院的接受标准双联抗血小板治疗(阿司匹林+氯吡格雷)的冠心病患者共800例。以光学比浊法测定20μmol/L浓度ADP诱导的残余血小板聚集率(Residual plateletagglutination,RPA),并定义RPA≥70%为CR,所有入选患者分为CR组和氯吡格雷非抵抗组(Non-clopidogrel resistance,NCR)。所有入选病例提取血液白细胞基因组DNA后,采用直接测序的方法测定CYP3A5基因rs3800959单核苷酸多态位点的基因型及等位基因。结果:所入选的800例病人中,CR组为150例,NCR组为650例,CR发生率为18.75%。rs3800959基因型频率在CR组为TT型110例(73.3%)、CT型39例(26.0%)及CC型1例(0.7%);NCR组rs3800959基因型频率分别为477例、159例及14例(73.4%、24.5%及2.1%)。两组间各基因型频率分布无统计学差异(P=0.460,x2=1.554);T、C等位基因分布频率在两组间亦无明显差异(P=0.784,OR=0.942,95%CI=0.655~1.356)。结论:CYP3A5基因单核苷酸多态位点rs3800959与冠心病人CR的发生无相关关系。 相似文献

目的:探讨阿片样物质受体(μ1 opioid receptor,OPRM1)(A118G)基因多态性与肺癌癌痛患者镇痛效果的相关性。方法:选取本院2017年3月至2019年10月收治的360例肺癌患者作为研究对象,判断患者阿片耐受与不良反应发生情况。收集患者血液指标,检测OPRM1(A118G)基因多态性情况并进行相关性分析。结果:在360例患者中,阿片耐受78例(耐受组),耐受率为21.7%;耐受组的性别、年龄、体重指数、肿瘤最大直径等与非耐受组对比差异无统计学意义(P0.05),两组临床分期与淋巴结转移等对比差异有统计学意义(P0.05)。OPRM1(A118G)基因共有AA、AG、GG三种基因型,两组人群的OPRM1(A118G)基因型分布均符合Hardy-Weinberg平衡定律;两组OPRM1(A118G)基因型分布差异具有统计学意义,耐受组的OPRM1(A118G)基因GG基因型比例显著高于非耐受组(P0.05),等位基因G频率显著高于非耐受组(P0.05);耐受组的呼吸抑制、恶心呕吐、头晕、皮肤瘙痒等不良反应发生率为35.9%,显著高于非耐受组的2.8%(P0.05)。直线相关性分析显示OPRM1(A118G)基因GG基因型与阿片耐受、淋巴结转移、临床分期都呈现相关性(P0.05);二分类变量Logistic回归分析显示OPRM1(A118G)基因GG基因型、临床分期、淋巴结转移为影响阿片耐受的主要因素(P0.05)。结论:肺癌癌痛患者在镇痛中存在阿片耐受情况,与患者的OPRM1(A118G)基因多态性与治疗不良反应显著相关,OPRM1(A118G)基因GG基因型、临床分期、淋巴结转移为影响阿片耐受的主要因素。 相似文献

人类rDNA顺序既偏偏保守性积蓄高的转录区,又有变异性较大的不转录间隔区。随着对rDNA结构的不断认识,发现对不转录间隔区特征的研究在分子群体遗传学上具有重要的价值。我们运用rDNA基因探针和Southern印迹法对甸汉族和彝族rRNA基因3'端不转录间隔区作多态性分析,RFLP特征揭示两个民族有广泛的共性,又有各自的特点,这一遗传标志对少数民族群体遗传学乃至我国民族的迁移和演化的研究有重要意义。 相似文献

CYP7A1(cholesterol 7α-hydroxylase )在胆固醇向胆汁酸代谢途径中起着至关重要的作用.为研究该基因启动子区-204位点A/C多态性是否影响基因表达, 利用荧光素酶作为报告基因,将含有A或C等位基因的启动子区片段分别正向和反向插入不含启动子的pGL3 basic质粒载体中,再以重组体转染4种细胞株,采用双荧光素酶报告基因检测系统测定酶活性并进行比较.实验结果表明,2种基因型的正向序列启动子活性均高于相应的反向序列,含有A等位基因的启动子片段活性比含有C等位基因的片段低约1/3.TRANSFAC数据库分析显示,当-204位点等位基因为C时,可能存在1个Zic3结合位点.研究结果提示,CYP7A1基因启动子区-204位点A/C变异可减少启动子活性从而影响基因表达,其原因可能为1个潜在的Zic3结合位点的丧失. 相似文献

Deficiency of drug glucuronidation in the cat is one of the major reasons why this animal is highly sensitive to the side effects of drugs. The characterization of cytochrome P450 isoforms belonging to the CYP1A subfamily, which exhibit important drug oxidation activities such as activation of pro-carcinogens, was investigated. Two cDNAs, designated CYP1A-a and CYP1A-b, corresponding to the CYP1A subfamily were obtained from feline liver. CYP1A-a and CYP1A-b cDNAs comprise coding regions of 1554 bp and 1539 bp, and encode predicted amino acid sequences of 517 and 512 residues, respectively. These amino acid sequences contain a heme-binding cysteine and a conserved threonine. The cDNA identities, as well as the predicted amino acid sequences containing six substrate recognition sites, suggest that CYP1A-a and CYP1A-b correspond to CYP1A1 and CYP1A2, respectively. This was confirmed by the kinetic parameters of the arylhydrocarbon hydroxylase and 7-ethoxyresorufin O-deethylase activities of expressed CYPs in yeast AH22 cells and by the tissue distribution of each mRNA. However, theophylline 3-demethylation is believed to be catalyzed by CYP1A1 in cats, based on the high V(max) and low K(m) seen, in contrast to other animals. Because feline CYP1A2 had a higher K(m) for phenacetin O-deethylase activity with acetaminophen, which cannot be conjugated with glucuronic acid due to UDP-glucuronosyltransferase deficiency, it is supposed that the side effects of phenacetin as a result of toxic intermediates are severe and prolonged in cats. 相似文献

Drugs and carcinogens are substrates of a group of metabolic enzymes including cytochrome p450 enzymes and gluthatione S-transferases. Many of the genes encoding these enzymes exhibit functional polymorphisms that contribute individual cancer susceptibility and drug response. Molecular studies based on these polymorphic enzymes also explain the aetiology of cancer and therapeutic management in clinics. We analysed the cytochrome p4501A1 (CYP1A1) and 2D6 (CYP2D6) variant genotype and allele frequencies by PCR-RFLP in Turkish individuals (n=140). The frequency of the CYP1A1*2A mutant allele was found to be 15.4%, and the CYP2D6*3 and *4 mutant allele (poor metabolizer) frequencies were 2.5% and 13.9%, respectively. This study presents the first results of CYP1A1 and CYP2D6 mutant allele distributions in the Turkish population and these data provide an understanding of epidemiological studies that correlate therapeutic approaches and aetiology of several types of malignancy in Turkish patients. 相似文献

CYP1A1 and CYP1A2 enzymes metabolize polycyclic aromatic hydrocarbons (PAHs) to the reactive oxyderivatives. PAHs can induce the activity of both enzymes, which increases its conversion and enhances risk of carcinogenesis. Thus, the inhibition of CYP enzymes is recognized as a cancer chemoprevention strategy. A well‐known group of chemopreventive agents is isothiocyanates, which occur naturally in Brassica vegetables. In this paper, a naturally occurring sulforaphane and its two synthetic analogues isothiocyanate‐2‐oxohexyl and alyssin were investigated. The aim of the study was to determine whether the differences in the isothiocyanate structure change its ability to inhibit CYP1A1 and CYP1A2 activity induced by benzo[a]pyrene in HepG2 and Mcf7 cells. Also a mechanistic study was performed including isothiocyanates' influence on CYP1A1 and CYP1A2 catalytic activity, enzymatic protein level, and AhR translocation. It was shown that both enzymes were significantly induced by benzo[a]pyrene, and isothiocyanates were capable of decreasing the induced activity. The inhibitory properties depend on the types of isothiocyanate and enzyme. In general, CYP1A2 was altered in the more meaningful way than CYP1A1 by isothiocyanates. Sulforaphane exhibited weak inhibitory properties, whereas both analogues were capable of inhibiting BaP‐induced activity with the similar efficacy. The mechanistic study revealed that analogues decreased the CYP1A2 activity via the protein‐level reduction and CYP1A1 directly. The results indicate that isothiocyanates can be considered as potent chemopreventive substances and the change in the sulforaphane structure increases its chemopreventive potency. © 2009 Wiley Periodicals, Inc. We report molecular cloning and single nucleotide polymorphism detection of the buffalo DGAT1 gene. Diacylglycerol acyltransferase (DGAT1) is considered the key enzyme in controlling the rate of synthesis of triglycerides.
The DGAT1 gene was recently identified as a strong functional candidate gene affecting milk yield and composition in cattle. A full-length
buffalo DGAT1 genomic DNA was amplified by iterative PCR based on homolog cloning. The buffalo DGAT1 gene comprises 17 exons and spans approximately 8.3 kb. The genomic structures of DGAT1 are highly conserved among mammal species. The deduced protein of buffalo DGAT1 contains 489 amino acids, showing high-sequence
similarity with mammal homologs. Through PCR-SSCP analysis and sequencing, seven polymorphic positions were detected in the
complete genomic region of buffalo DGAT1, and their frequencies were observed from a collection of 117 buffalo. The SNP (C/T) detected at position 11785 in exon 17
creates a substitution change for the amino acid sequence, resulting in an Ala residue (GCG) transition to a Val residue (GTG)
in position 484 of buffalo DGAT1 protein. Information provided in this study will be useful in further studies to determine
the role DGAT1 plays in the regulation of milk fat synthesis and quality improvement for milk in buffalo.

目的:氯吡格雷主要由CYP3A4催化使其激活,CYPlA2也参与氯吡格雷活化。关于氯吡格雷对肝微粒体酶的影响国内外文献报道不多,因此本实验通过检测肝细胞色素氧化酶CYP3A4和CYPlA2的表达,探讨氯吡格雷对大鼠肝药物酶的影响。方法:生理盐水为对照组,氯吡格雷设高、中、低三个剂量组(27,13.5,6.75mg/kg/d),雄性健康大鼠连续灌胃给药7天,脱臼处死,取肝组织,通过westernblot法检测大鼠肝脏CYP3A4和CYPlA2蛋白表达情况。结果:1)、氯吡格雷抑制大鼠CYP3A4蛋白表达,氯吡格雷高中低剂量组分别比生理盐水组大鼠CYP3A4蛋白表达量降低(P〈0.05);氯吡格雷低中高剂量组间进行比较,大鼠CYP3A4蛋白表达量呈梯度减少(P〈0.05);2)、氯吡格雷抑制大鼠CYPlA2蛋白表达,氯吡格雷高中低剂量组分别比生理盐水组大鼠CYPlA2蛋白表达量降低(P〈0.05),氯吡格雷低中高剂量组间进行比较,大鼠CYPlA2蛋白表达量呈梯度减少(P〈0.05)。结论:氯吡格雷使肝细胞色素氧化酶CYP3A4和CYPlA2的表达量减少,因此氯吡格雷高、中、低3个剂量组均不同程度的抑制大鼠肝脏CYP3A4和CYPlA2的表达,提示当氯吡格雷与某些主要经CYP3A4和CYPlA2代谢的药物合用时,发生代谢性相关作用的可能性大。 相似文献
