Conidia of Trichoderma reesei QM9414 were treated with colchicine. Nuclei in colchicine-treated conidia enlarged. When the concentration of colchicine or the treatment time with colchicine increased, the diameter of nuclei became larger. Colchicine-treated conidia generated sectors on a medium containing benomyl. Some sectors formed many conidia or could not produce clear zones on the plate assay medium for cellulase production. According to the DNA assay of conidia, colchicine-treated strains were autopolyploid.  相似文献

Aspergillus oryzae has been reported to form conidia with multinuclei. In order to analyze nuclei in living cells, we developed an expression system of the A. nidutans histone H2B protein tagged by EGFP (H2B::EGFP). In both A. oryzae niaD300 and A. nidulans FGSC89 transformants expressing H2B::EGFP, fluorescence was detected in nuclear regions of hyphae and conidia. While a conidium contained only one fluorescent spot in the A. nidulans transformant, approximately 66% of conidia had two, 24% had one, and 10% had three or more in the A. oryzae transformant. The conidia expressing H2B::EGFP were put through FACS (fluorescence-activated cell sorting) analysis and two sharp peaks, corresponding to one and two nuclei in each conidium, were noted in the A. oryzae transformant. In addition, the A. oryzae uninucleate conidia that were successfully isolated by FACS reproduced conidia with almost the same number distribution of nuclei as that of the original. Conidia of five A. oryzae strains used in sake brewing were scored for the number of nuclei, showing that a varied number of nuclei existed in each conidium and some strains had a small number of uninucleate conidia.  相似文献

Conidia ofTrichoderma reesei QM 9414 were treated with colchicine in order to obtain polyploids (diploids; tetraploids). Cellulase production by diploids (mononucleate conidia) was almost twice as great as that of the original strain, but that of tetraploids (binucleate conidia) was not increased. When these latter conidia were re-treated with 2.0% (w/v) colchicine, multiple nuclei were produced in each conidium, and their diameter was almost the same as that of the original nucleus. Cellulase production of the diploid was almost the same in either mononucleate or multinucleate nature. However, cellulase production by the tetraploid which produced multinucleate conidia was greater than that of the binucleate tetraploid and that of the diploid. The multinucleation technique can contribute to enhancing cellulase production.  相似文献

The cells of haploid Aspergillus niger strains contain, on the average, 7-9 nuclei, a fragment of a thin hypha 100 me long comprising 11-19 nuclei. The cells of a diploid strain are 1.5-2.6 times larger in volume. The diploid cells contain less nuclei and more cytoplasm per nucleus as compared to the haploid strains. The primary sterigmae of Aspergillus niger comprise 3-13 nuclei, the secondary sterigmae and conidia, one nucleus. The conidia of the diploid strains are 1.8-2.0 times larger in volume and contain twice as much DNA as compared to the haploid strains.  相似文献

Protoplasts of cycloheximide-resistant strains from Aspergillus oryzae IFO 5239 were fused with those of kabicidin-resistant strains from Aspergillus niger IFO 4407. By nuclear staining in conidia, it appeared that all of the fusant conidia had two kinds of nuclei. Small nuclei seemed to be derived from A. oryzae and large nuclei seemed to be derived from A. niger. However, three types of antibiotic resistance were shown among the conidia of fusants. Almost all were kabicidin resistant. Conidia of fusants were multinuclear and had various DNA contents and various sizes. By the comparison with the growth rates of parental strains, the growth rates of A. niger were superior to those of A. oryzae. The inclination in the distribution of antibiotic resistance of fusant conidia seemed to owe more to the differences of growth rates between parental strains than the influence of the multinucleate nature of a parental strain.  相似文献

Wild-type Aspergillus nidulans conidia are uninucleate. The mutation bncA1 (binucleated conidia) was first described as a single mutation located on chromosome IV that caused formation of approximately 25% binucleate and 1% trinucleate conidia. In this study, we show that bncA1 conidia exit G1 arrest earlier than the wild type. Germlings have hyphal elements with abnormal morphology, elevated numbers of randomly distributed nuclei and an irregular septation pattern. Older hyphal elements undergo mitotic catastrophe, suggesting the nuclear division cycle of internal (nonterminal) elements is not arrested. The bncA1 mutation also causes aberrant morphogenesis of the asexual reproductive structure, the conidiophore. Metulae and phialides are elongated and have incorrect numbers of nuclei. Phialides also have internal septation that appears to delineate hyphal-like elements. Heterokaryon analysis using strains with contrasting auxotrophic markers showed that the bncA1 mutation resulted in a higher frequency of diploid and multinucleated prototrophic conidia than control heterokaryons. These results suggest that in bncA1 strains multiple nuclei can move from the conidiophore vesicle to the metulae and/or from the phialide to the conidium. The bncA1 mutant also showed hypersensitivity to the anti-microtubule drugs thiabendazole and nocodazole, which is consistent with the defects in cell cycle regulation and nuclear movement. We propose that bncA has an important role in correctly regulating both the cell division cycle and nuclear movement.  相似文献

Mitotic activity in the cells of the germinative region of wool follicle bulbs was quantified by using small (0.1-0.5 ml) intradermal doses of colchicine and selective staining of the metaphase-blocked nuclei using either crystal violet, iodine and eosin or haematoxylin and eosin. The number of metaphase nuclei present 3 h after colchicine administration increased with colchicine dose from 0 to 1 microgram and thereafter remained relatively constant up to 200 micrograms colchicine. The accumulation of metaphase nuclei was linear for up to 6 h after intradermal colchicine. The metaphase-blocking effect of intradermal colchicine was confined to a radius of less than 5 cm from the injection site, allowing a number of estimates of mitotic rates to be made over a small area of skin. Such estimates revealed little variation in mitotic activity over the midside region of the sheep, although there were substantial differences in follicle activity at different sites over the body. The technique is simple, allows serial or concurrent estimates of mitotic activity to be made in the same animal, and eliminates problems associated with intravenous colchicine administration. It was used to derive the relationship between follicle activity and fibre production after nutritional changes, and to define the time course of mitotic events after administration of the antimitotic defleecing agent cyclophosphamide.  相似文献

Nuclear migration is indispensable for normal growth, differentiation, and development, and has been studied in several fungi including Aspergillus nidulans and Neurospora crassa. To better characterize nuclear movement and its consequences during conidiophore development, conidiation, and conidial germination, we performed confocal microscopy and time-lapse imaging on A. nidulans and Aspergillus oryzae strains expressing the histone H2B-EGFP fusion protein. Active trafficking of nuclei from a vesicle to a phialide and subsequently into a conidium provided the mechanistic basis for the formation of multinucleate conidia in A. oryzae. In particular, the first direct visual evidence on multinucleate conidium formation by the migration of nuclei from a phialide into the conidium, rather than by mitotic division in a newly formed conidium, was obtained. Interestingly, a statistical analysis on conidial germination revealed that conidia with more nuclei germinated earlier than those with fewer nuclei. Moreover, multinucleation of conidia conferred greater viability and resistance to UV-irradiation and freeze-thaw treatment.  相似文献

The V10 deteriorated variant of Aspergillus nidulans has hyphae, metulae, phialides and conidia with abnormal nuclear distributions. The alterations observed were: increase in the number of nuclei in hyphae, metulae and phialides, presence of anucleate, uninucleate and multinucleate conidia, abnormal vegetative growth and defective conidiation. When 0.5 M NaCl was added to the medium, an increase in the number of conidia was observed but their morphology and number of nuclei were not modified. The gene responsible for these alterations was named anuA1. The anuA1 gene is located on linkage group VII and is possibly involved in nuclear migration to hyphae, metulae, phialides and conidia.  相似文献

Helminthosporium euphorbiae is a pathogen of the weedEuphorbia heterophylla, which causes severe losses in soybean (Glycine max) crops. The fungus causes leaf loss and affects germination, making it a promising biocontrol agent for this weed. In order to start a breeding program for this species, four isolates were examined for number of nuclei in the conidia and hyphae and nuclear behavior at different cultivation stages. The conidia were multinucleated with about 20 nuclei per conidium, and 5 to 7 nuclei were observed in the hyphae compartments. The high number of nuclei makes the genetic manipulation of this species diffucult, so the protoplast formation is an alternative for obtaining cells with a reduced number of nuclei. Thus the experimental conditions for the production and regeneration of protoplasts inH. euphorbiae were determined by assessing three enzymatic complexes and seven osmotic stabilizers. The efficiency of formation and regeneration frequency of the protoplasts varied depending on isolates, stabilizers and enzyme mixture used. The number of nuclei estimated per protoplast was reduced to 1 to 6, depending on the stage of mycelial growth during the protoplast formation process.  相似文献

The effect of colchicine and boric acid on the substrate mycelium of Trichothecium roseum was studied in the course of trichothecin biosynthesis. Addition of boric aicd (0.01%) and colchicine (0.1%) to the medium for biosynthesis increased the antibiotic activity of the fungus, this being due to the specific effect of polyploidogenous factors on growth of the mycelium and the proportion of nuclei in it. The number of nuclei increased in cells of the substrate mycelium correlating with a higher antibiotic activity.  相似文献

The mycelial mat of Trichoderma reesei strain QM 6a was treated with 0.1% (w/v) colchicine solution for 14 days and designated M14. The cellulase productivity of strain M14 was not much higher than that of the original strain. When conidia of M14 were treated with ethylmethane sulphonate (EMS) solution, the cellulase hyperproducers, M14-1 and M14-2, were isolated using a selection medium containing Avicel. The DNA content of M14-1 and M14-2 was higher than that of the original strain. Cellulase productivity per mycelium of these strains increased and was higher than that of the original strain. The cellulase productivity did not change through ten generations when these strains were cultivated successively on a medium containing Avicel. It was concluded that cellulase hyperproducers, whose cellulase productivity per mycelium increased, could be obtained when the conidia of strain M14 were treated with EMS.  相似文献

During growth of conidia in 3.22 M ethylene glycol the increase in the number of the nuclei is proportional to the increase in volume only in the phase of maximum growth rate and is lower in the preceding and in the following periods of growth. DNA synthesis similarly initiates later and decreases faster than protein synthesis. The dilution of ethylene glycol is followed by the germination of giant conidia, which is characterized by the absence of a lag phase, a high degree of synchrony and the formation of more than one germ tube per conidium. The number of germ tubes is dependent on the volume reached by conidia at the end of the treatment and does not increase with time. The resuming of DNA synthesis after germination is preceded by a sharp increase in protein synthesis and the division of almost half of the nuclei and shows a synchronized pattern. Results are discussed in the light of models of growth proposed for eukaryotic cells.  相似文献

The intent of the present study was to find out whether dexamethasone pretreatment may affect the induction of Fos protein in cell nuclei of the cerebellar vestibular neuronal complex (CVNC) elicited by central administration of colchicine. Specifically, the rate of the dexamethasone-sensitive cell population was analyzed and compared at different levels of the CVNC using a light microscopic avidin-biotin peroxidase immunohistochemistry.2. Male Wistar rats were pretreated with dexamethasone 3 days prior (2.5 mg/kg/day, s.c.) and 24 h after an intracerebroventricular delivery of colchicine (60 g/10 L). Animals were sacrificed 48 h after colchicine treatment by a transcardial perfusion with fixative.3. Dexamethasone in itself had no effect on the activity of cells of the CVNC. However, in colchicine treated animals, which exhibited a large number of Fos-positive cells over the entire CVNC, the dexamethasone elicited a substantial reduction in the number of the Fos-immunoreactive cells over the CVNC. Distinct dexamethasone dependent reduction (50–90%) of Fos-immunoreactivity was observed in each of the deep cerebellar nuclei. On the other hand, less number of dexamethasone-sensitive cells were recognized in the vestibular structures. From these, maximal Fos-inhibition by dexamethasone was recognized in the medial vestibular nucleus, however, even in this case the number of suppressed cells did not exceed 50%.4. The results provide for the first time evidence about the dexamethasone dependent reduction of Fos-immunoreactivity in the cells of the CVNC in response to stimulation elicited by colchicine. The data also indicate that the glucocorticoids might be involved in the regulation of some functions of the CVNC under stress conditions.  相似文献

The effect of a single intracerebroventricular injection of colchicine on the distribution of organelles in neurons of the mesencephalic nucleus of the trigeminal nerve, the inferior colliculus and the deep cerebellar nuclei was studied. In the mesencephalic nucleus of the trigeminal nerve colchicine produced a dramatic accumulation of neurofilament bundles in the soma of these neurons and did not produce a reduction in the number of lysosomes. In other neuronal populations studied, colchicine produced neurofilament bundles in the dendrites and a reduction of lysosomes from the soma of neurons.  相似文献

The effectiveness of colchicine in destroying or preventing the development of the spindle is determined by the concentration of the colchicine and the degree of development of the spindle at the time of exposure; the greater the concentration of colchicine, the greater will be its effectiveness in destroying the spindle or interfering with its development; the more completely the spindle is developed at the time of exposure to colchicine, the greater is the concentration of colchicine required to destroy it or prevent its further development.The series of changes in chromosome orientation that take place during the course of colchicine action, namely, approximation of centromeres, formation of “stars,” the breaking up of one “star” into several, and complete chromosome disorientation, represent successive stages in the destruction of the spindle.Destruction of the completely formed spindle is typically accompanied by the accumulation of the spindle material outside the diminishing spindle in a hyaline globule. Similarly, interference of colchicine with spindle development leads to the accumulation of the presumptive spindle material, i.e., the karyolymph, in one or more hyaline globules. It is suggested that colchicine does not destroy the spindle material but merely alters its molecular orientation, so that it comes to comprise a spherical mass with no mitotic function.Strong concentrations of colchicine cause middle and late prophase nuclei to revert to early prophase. Somewhat lower concentrations applied to late prophase nuclei occasionally delay the breakdown of the nuclear membrane without altering the rate of chromosome contraction. In greatly retarded late prophase nuclei the chromosomes lose their intranuclear orientation, which lends support to the concept that the centromeres normally maintain a fixed position within the nucleus.  相似文献

S ummary . Heterocaryons and diploids from Aspergillus oryzae were investigated with respect to nuclear number/conidium and to conidial size. Heterocaryons usually had larger conidia and more nuclei/conidium than diploids and the haploid parent mutants. Diploids contained significantly fewer nuclei/conidium than haploids. However, they could not be distinguished from haploids by measurement of conidial size. The strains were examined for the production of α-amylase. All auxotrophic mutants produced less α-amylase than the prototrophic wild type. Heterocaryons gave yields which were intermediate between that of their parent mutants or the same as the best producing parent. Diploids which produced more α-amylase than the best producing parent strain were synthesized. The highest yield from a diploid was of the same order of magnitude as the yield from the wild type.  相似文献

西瓜枯萎病是一种世界范围的西瓜毁灭性病害,其病原菌为尖孢镰刀菌西瓜专化型(Fusarium oxysporum f.sp.niveum,FON)。研究病原菌生长发育和侵染的机制是解决病害的根本途径。利用荧光蛋白对细胞或细胞器进行标记,是病原菌研究中的重要方法。该研究利用绿色荧光蛋白和红色荧光蛋白对FON的细胞核和过氧化物酶体进行了荧光标记。通过农杆菌介导转化(Agrobacterium tumefaciens-mediated transformation,AtMT),该文将3种不同的荧光定位载体分别导入FON,获得了细胞核红色荧光标记的转化子(潮霉素抗性,含mCherry-H2B融合蛋白),以及过氧化物酶体绿色(潮霉素抗性,含GFP-PTS1融合蛋白)和红色(潮霉素抗性,含DsRED-PTS1融合蛋白)荧光标记的转化子各1种。在标记细胞核的菌株中,菌丝、孢子都可见明亮、圆形的红色荧光点,荧光点与DAPI染色标记的细胞核区域完全重合。在过氧化物酶体标记的菌株中,菌丝、孢子中可见明亮的红色或绿色荧光成小点状分布,符合过氧化物酶体的分布特征,而且在脂类物质诱导的条件下,荧光点的数量明显增加。此外,该文还利用细胞壁荧光染色剂卡氏白对3种荧光蛋白标记菌株进行染色。结果显示,卡氏白染色产生的蓝色荧光与红、绿荧光蛋白的荧光在FON中互不干扰。转化子继代培养和初步分析表明,其表型与野生型无差异,菌株继代后荧光表达稳定、定位明显。该结果为进一步研究FON细胞器动态、生长发育与致病分子机制提供了方法和工具。  相似文献

蛹虫草菌株在继代培养和低温长期保藏过程中极易退化导致子实体产量下降,对产业造成重大影响。本研究以正常菌株、PDA斜面长期4 ℃保藏导致退化的菌株和连续继代培养的菌株为材料,观察其子实体、菌落、菌丝形态和分生孢子数量,对菌丝细胞核、线粒体、活性氧积累和芽生孢子内脂滴进行染色观察;并对菌丝中虫草素、腺苷和麦角甾醇含量进行比较分析。结果表明PDA斜面长期4 ℃保藏和连续继代培养导致的蛹虫草菌株退化表型大多数一致,即退化菌株较正常菌株子实体产量降低、菌丝粘连打结、分生孢子数量显著降低、菌丝活性氧含量升高、细胞内线粒体数量减少、芽生孢子脂滴由弥散的小脂滴融合为大脂滴。然而,在菌落见光转色方面,长期低温保藏退化菌株基本不转色,而继代培养退化菌株转色不稳定;长期保藏退化菌株菌丝细胞核数目无明显变化,继代培养退化菌株细胞核数量明显减少;长期保藏退化菌株菌丝中虫草素、腺苷和麦角甾醇含量较正常菌株降低58%、41%和70%,继代培养退化菌株麦角甾醇含量无明显变化。因此,显微观察菌丝是否出现粘连打结及氮蓝四唑NBT检测菌丝活性氧含量,操作简单、用时较短,可用于大规模生产中蛹虫草菌株退化的检测手段;线粒体和脂滴染色也可作为退化菌株的鉴别方法;而生产中常用的通过菌落见光转色判断菌株优劣的方法需要慎重。继代培养菌株第4代开始出现明显的退化特征,因此在生产中使用的菌株最好控制在继代培养3代以内。  相似文献
