The aim of the study was to determine changes incurred by polyphenolic compounds from selected fruits in the lipid phase of the erythrocyte membrane, in liposomes formed of erythrocyte lipids and phosphatidylcholine liposomes. In particular, the effect of extracts from apple, chokeberry, and strawberry on the red blood cell morphology, on packing order in the lipid hydrophilic phase, on fluidity of the hydrophobic phase, as well as on the temperature of phase transition in DPPC liposomes was studied. In the erythrocyte population, the proportions of echinocytes increased due to incorporation of polyphenolic compounds. Fluorimetry with a laurdan probe indicated increased packing density in the hydrophilic phase of the membrane in presence of polyphenolic extracts, the highest effect being observed for the apple extract. Using the fluorescence probes DPH and TMA-DPH, no effect was noted inside the hydrophobic phase of the membrane, as the lipid bilayer fluidity was not modified. The polyphenolic extracts slightly lowered the phase transition temperature of phosphatidylcholine liposomes. The studies have shown that the phenolic compounds contained in the extracts incorporate into the outer region of the erythrocyte membrane, affecting its shape and lipid packing order, which is reflected in the increasing number of echinocytes. The compounds also penetrate the outer part of the external lipid layer of liposomes formed of natural and DPPC lipids, changing its packing order.  相似文献

Cell membranes show complex behavior, in part because of the large number of different components that interact with each other in different ways. One aspect of this complex behavior is lateral organization of components on a range of spatial scales. We found that lipid-only mixtures can model the range of size scales, from approximately 2 nm up to microns. Furthermore, the size of compositional heterogeneities can be controlled entirely by lipid composition for mixtures such as 1,2-distearoyl-sn-glycero-3-phosphocholine (DSPC)/1,2-dioleoyl-sn-glycero-3-phosphocholine (DOPC)/1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine (POPC)/cholesterol or sphingomyelin (SM)/DOPC/POPC/cholesterol. In one region of special interest, because of its connection to cell membrane rafts, nanometer-scale domains of liquid-disordered phase and liquid-ordered phase coexist over a wide range of compositions.  相似文献

Cytoplasmic membrane deterioration during petal senescence ofcut rose flowers entails a sequence of interrelated physicalchanges in the lipid bilayer. Wide angle X-ray diffraction revealedthat microsomal membranes from flowers at the tight bud stage(day 1) contained liquid-crystalline lipid and only traces ofgel phase lipid. However, with advancing senescence, the proportionof gel phase lipid increased, particularly between days 6 and9. The lipid phase transition temperature also increased duringsenescence from 38 ?C at day 1 to 48 ?C at day 9. An order parameter(S), reflecting molecular order close to the membrane surfaceand determined by electron spin resonance (ESR) of microsomalmembranes labelled with 5-doxyl stearate, increased during theearly stages of senescence between days 1 and 3. The rotationalcorrelation time (c), a motion parameter obtained from microsomalmembranes labelled with 16-doxyl stearate, increased throughoutsenescence, reflecting a rise in lipid microviscosity deep inthe hydrophobic core of the membrane bilayer. Activation energies,calculated from continuously linear Arrhenius plots of rc andreflecting the degree of molecular order in the hydrophobiccore of the membrane, increased during the later stages of senescencebetween days 6 and 9. The physiological implications of thesephysical changes in the lipid bilayer are discussed.  相似文献

Free polyamine levels (spermine (SPM), spermidine (SPD), and putrescine (PUT)) were determined using thin-layer chromatography and fluorometric method in loquat (Eriobotrya japonica Lindl. cv. Dahongpao) fruits stored at 1℃ and 12℃ and in postharvest SPM treated fruits stored at 1℃ respectively to investigate the relationship between changes in polyamines and chilling injury. In the loquat fruits stored at 1℃, SPM level decreased gradually in the first two weeks, then increased sharply and reached a peak value after three weeks, thereafter it decreased rapidly. SPD level decreased steadily during the first three weeks and increased significantly afterwards. PUT level evolved in a similar way as the SPM level did except that it increased slowly in the first two weeks. The fruit showed symptom of chilling injury manifested as flesh leatheriness after three weeks. However, no significant increase and decrease of these three polyamines was detected during storage at the nonchilling temperature (12℃). The SPM-treated fruits maintained high levels of SPM and SPD and remained low level of PUT during storage at 1℃, and no symptom of chilling injury was observed. These results suggested that the increase in SPM level in response to chilling exposure might serve as a defense mechanism against chilling injury while the accumulation of PUT could be a cause of the stress-induced injury and the increase in SPD level could be a consequence of this kind of stress.  相似文献

The stability and shapes of domains with different bending rigidities in lipid membranes are investigated. These domains can be formed from the inclusion of an impurity in a lipid membrane or from the phase separation within the membrane. We show that, for weak line tensions, surface tensions and finite spontaneous curvatures, an equilibrium phase of protruding circular domains or striped domains may be obtained. We also predict a possible phase transition between the investigated morphologies.  相似文献

Quarantine treatment enables export of avocado fruit (Persea americana) to parts of the world that enforce quarantine against fruit fly. The recommended cold-based quarantine treatment (storage at 1.1°C for 14 days) was studied with two commercial avocado cultivars ‘Hass’ and ‘Ettinger’ for 2 years. Chilling injuries (CIs) are prevalent in the avocado fruit after cold-quarantine treatment. Hence, we examined the effect of integrating several treatments: modified atmosphere (MA; fruit covered with perforated polyethylene bags), methyl jasmonate (MJ; fruit dipped in 2.5 μM MJ for Hass or 10 μM MJ for Ettinger for 30 s), 1-methylcyclopropene (1-MCP; fruit treated with 300 ppb 1-MCP for 18 h) and low-temperature conditioning (LTC; a gradual decrease in temperature over 3 days) on CI reduction during cold quarantine. Avocado fruit stored at 1°C suffered from severe CI, lipid peroxidation, and increased expression of chilling-responsive genes of fruit peel. The combined therapeutic treatments alleviated CI in cold-quarantined fruit to the level in fruit stored at commercial temperature (5°C). A successful therapeutic treatment was developed to protect ‘Hass’ and ‘Ettinger’ avocado fruit during cold quarantine against fruit fly, while maintaining fruit quality. Subsequently, treated fruit stored at 1°C had a longer shelf life and less decay than the fruit stored at 5°C. This therapeutic treatment could potentially enable the export of avocado fruit to all quarantine-enforcing countries. Similar methods might be applicable to other types of fruit that require cold quarantine.  相似文献

Total proteins from pericarp tissue of different chronological ages from normally ripening tomato (Lycopersicon esculentum Mill. cv Rutgers) fruits and from fruits of the isogenic ripening-impaired mutants rin, nor, and Nr were extracted and separated by sodium dodecylsulfate-polyacrylamide gel electrophoresis. Analysis of the stained bands revealed increases in 5 polypeptides (94, 44, 34, 20, and 12 kilodaltons), decreases in 12 polypeptides (106, 98, 88, 76, 64, 52, 48, 45, 36, 28, 25, and 15 kilodaltons), and fluctuations in 5 polypeptides (85, 60, 26, 21, and 16 kilodaltons) as normal ripening proceeded. Several polypeptides present in ripening normal pericarp exhibited very low or undetectable levels in developing mutant pericarp. Total RNAs extracted from various stages of Rutgers pericarp and from 60 to 65 days old rin, nor, and Nr pericarp were fractionated into poly(A)+ and poly(A) RNAs. Peak levels of total RNA, poly(A)+ RNA, and poly(A)+ RNA as percent of total RNA occurred between the mature green to breaker stages of normal pericarp. In vitro translation of poly(A)+ RNAs from normal pericarp in rabbit reticulocyte lysates revealed increases in mRNAs for 9 polypeptides (116, 89, 70, 42, 38, 33, 31, 29, and 26 kilodaltons), decreases in mRNAs for 2 polypeptides (41 and 35 kilodaltons), and fluctuations in mRNAs for 5 polypeptides (156, 53, 39, 30, and 14 kilodaltons) during normal ripening. Analysis of two-dimensional separation of in vitro translated polypeptides from poly(A)+ RNAs isolated from different developmental stages revealed even more extensive changes in mRNA populations during ripening. In addition, a polygalacturonase precursor (54 kilodaltons) was immunoprecipitated from breaker, turning, red ripe, and 65 days old Nr in vitro translation products.  相似文献

以陕西杨凌地区主栽的番茄品种'金棚1号'为试验材料,通过固相微萃取和GC/MS联用技术,对番茄果实不同成熟阶段的香气成分及其组成变化进行了研究.结果表明,'金棚1号'番茄果实共检测到54种香气成分,主要成分为醛类、酸类、醇类、酮类、酯类、酚类等.在果实的不同发育阶段,香味组分及其含量差异较大.醛类物质在绿熟期相对含量较高,为45.87%,在半熟期、硬熟期、完熟期的相对含量分别为12.65%、16.62%、17.15%,其中C6醛在绿熟期占43.7%,完熟期占15.27%,为醛类物质的主要成分;酸类物质含量在4个发育时期中先上升后下降,在半熟期含量达到最高,为15.2%,在完熟期酸类物质含量下降,为6.93%;酮类物质在完熟期含量达到最大,为18.27%;在绿熟期检测到4种重要的番茄特征香气物质,半熟期检测到5种番茄特征香气物质,硬熟期和完熟期各检测到6种番茄特征香气物质.说明随着果实的成熟,特征香气物质种类增多.  相似文献

The transition temperature of the leaf polar lipids and the critical temperature for chill-induced inhibition of photosynthesis was determined for three altitudinal ecotypes of the wild tomato Lycopersicon hirsutum. Photosynthesis was measured as CO2-dependent O2 evolution at 25°C after leaf slices were exposed to chilling temperatures for 2 hours at a moderate photon flux density of 450 micromoles per square meter per second. The transition temperature of the leaf polar lipids was detected from the change in the temperature coefficient of the fluorescence intensity of trans-parinaric acid. Chill-induced photoinhibition was evident in the three tomato ecotypes when they were chilled below a critical temperature of 10°, 11°, and 13°C, respectively, for the high (LA1777), mid (LA1625), and low (LA1361) altitudinal ecotypes. The temperature differential, below the critical temperature, required to produce a 50% inhibition was also similar for the three ecotypes. A transition was detected in the leaf polar lipids of these plants at a temperature similar to that of the critical temperature for photoinhibition. The results show that the three tomato ecotypes are similar with respect to their critical temperature for chilling-induced photoinhibition and the rate of their response to the chilling stress. They are, thus, similarly sensitive to chilling.  相似文献

The effects of lipid degradation on proteins of smooth microsomalmembranes isolated from young bean cotyledons have been examinedby three techniques, viz. fluorescence energy transfer fromtryptophan to cis-parinaric acid; protein spin-labelling with3-maleimido PROXYL; and SDS-PAGE. Lipid degradation was inducedin isolated membranes by activating phospholipase D and phosphatidicacid phosphatase through the addition of Ca2+, by treatmentwith exogenous phospholipase C to simulate the concerted actionsof phospholipase D and phosphatidic acid phosphatase or by treatmentwith exogenous phospholipase A2 to generate endogenous substratefor lipoxygenase. All of the treatments induced time-dependentchanges in lipid-protein interaction and in protein conformation,and the treatment with phospholipase A2 also engendered proteolysis.The effects of the Ca2+ and phospholipase C treatments on lipid-proteininteraction and protein conformation can presumably be partlyattributed to an accumulation of diacylglycerol in the membrane,whereas the induction of proteolysis by phospholipase A2 appearsto be due to activated oxygen derived from the lipoxygenasereaction and ensuing lipid peroxidation. Lipid degradation inducedby these treatments simulates that which occurs during naturalsenescence of the cotyledons and hence these observations suggestthat loss of protein function and proteolysis in senescing membranesis facilitated by lipolytic and peroxidative activity withinthe lipid bilayer. When cucumber plants are chilled at 5 ?C and 85 per cent, r.h.the leaves wilt rapidly and lose water. Chlorophyll is lostin the light, but not in the dark. If chilled leaves are placedin water, electrolytes leak out rapidly, the amount dependingon the duration of chilling. There are marked reductions inthe rates of respiration and photosynthesis; Q10 for respirationis 5.0 between 15 and 5 ?C. Phospholipid levels decline, butmore slowly than the rate of water loss. Leaves recover theirinitial fresh weight, rate of electrolyte leakage, and respirationand photosynthesis rates if the plants are returned to warmconditions within 1 to 2 d. When leaves are chilled at 100 per cent r.h. they do not losewater or phospholipids; electrolyte leakage is little more thanin controls, but respiration and photosynthesis are reducedas at 85 per cent r.h. It is envisaged that the leaf cell membranes suffer a physicalchange at 5 ?C which reduces the rates of respiration and photosynthesis,but allows the passage of water and electrolytes out of thecells; the water evaporates away but electrolytes become concentratedin the walls and can be leached out of the leaf. As water islost from the leaf symptoms of dehydration appear. Phospholipidloss may be associated with enzyme action following the lossof compart-mentation.  相似文献

The phase behaviour of smooth microsomal membranes from senescing cultures of Scenedesmus quadricauda has been examined by wide-angle x-ray diffraction. The algae were grown in Bristol's medium at 22°C under continuous illumination. The transition temperature, taken to be the highest temperature at which crystalline (gel) phase lipid can be detected, increased with culture age from a low of 0°C for young cultures to a high of about 70°C for 140-day-old cultures. This indicates that for young cultures the membrane lipid is entirely liquid-crystalline (fluid) at physiological temperatures, but as the cultures age portions of the lipid become crystalline. The increase in transition temperature showed a close temporal correlation with loss of chlorophyll and loss of protein per g dry weight, and can thus be construed as an index of senescence. The unsaturated to saturated fatty acid ratio of the membrane lipid, while fluctuating with culture age, did not show any consistent trend that could be related to the change in transition temperature. Thus the formation of gel phase lipid does not appear to be due to a change in fatty acid saturation.  相似文献

Dopamine-β-hydroxylase (DBH), an enzyme that catalyzes the conversion of dopamine (DA) to norepinephrine (NE) in adrenal medullary chromaffin granules, increases the electrical conductance of bimolecular lipid membranes. The conductance increase requires both DA and Ca2+ and occurs in discrete steps. The conductance, which increases as the square of the DBH concentration, is nonselective for cations over anions and requires the native conformation of DBH. NE cannot replace DA.  相似文献

Phenolic acids are components of the plant defence system; however few studies had been done with fruits. The objective of this experiment was to elucidate the changes in phenolic acid content in tomato fruits in response to pathogenic attack. Tomato fruit ‘Pinto’ was inoculated with spores of Alternaria alternata and stored for 10 days at 25°C and 90–92% of relative humidity. Sampling of epicarp and mesocarp tissues of control and infected tomato fruits were done every 2 days. Phenolic acids were extracted from each tissue and identified by HPLC coupled to a mass spectrometer. Quantification was done based in standard curves. In vitro evaluation of the phenolic acid effect either one‐by‐one and in a mixture of phenolic acids (MPA) over A. alternata spore germination was done with 25, 50, 100, 200 and 500 mm . It was found caffeic, chlorogenic and vanillic acids in both epicarp and mesocarp tomato tissues before the beginning of the experiment. Infected tomato fruits showed significantly higher concentrations of vanillic acid in the epicarp, only. Chlorogenic acid and the MPA inhibit spore germination by 30%, whereas caffeic acid and vanillic acid inhibit 16% at 500 mm . It was concluded that chlorogenic, vanillic and caffeic acids are phytoanticipins in tomato fruit. The concentration of vanillic acid also increases in tomato fruit epicarp as part of the defence system during a pathogenic attack.  相似文献

Spermidine is a natural polyamine involved in many important cellular functions, whose supplementation in food or water increases life span and stress resistance in several model organisms. In this work, we expand spermidine’s range of age-related beneficial effects by demonstrating that it is also able to improve locomotor performance in aged flies. Spermidine’s mechanism of action on aging has been primarily related to general protein hypoacetylation that subsequently induces autophagy. Here, we suggest that the molecular targets of spermidine also include lipid metabolism: Spermidine-fed flies contain more triglycerides and show altered fatty acid and phospholipid profiles. We further determine that most of these metabolic changes are regulated through autophagy. Collectively, our data suggests an additional and novel lipid-mediated mechanism of action for spermidine-induced autophagy.  相似文献

Mitochondria were isolated from `Fuerte' avocado fruit (Persea americana Mill.) at four different stages of the respiratory climacteric. Preclimacteric fruit had the highest rate of succinate oxidation and the postclimacteric mitochondria the lowest. X-ray scattering features induced by aggregates of alamethicin (Alm) were obtained in oriented stacks of model membranes of DOPC(diC18:1PC) and diC22:1PC. The first feature obtained near full hydration was Bragg rod in-plane scattering near 0.11 ?−1 in DOPC and near 0.08 ?−1 in diC22:1PC at a 1:10 Alm:lipid ratio. This feature is interpreted as bundles consisting of n Alm monomers in a barrel-stave configuration surrounding a water pore. Fitting the scattering data to previously published molecular dynamics simulations indicates that the number of peptides per bundle is n = 6 in DOPC and n ≥ 9 in diC22:1PC. The larger bundle size in diC22:1PC is explained by hydrophobic mismatch of Alm with the thicker bilayer. A second diffuse scattering peak located at q r ≈ 0.7 ?−1 is obtained for both DOPC and diC22:1PC at several peptide concentrations. Theoretical calculations indicate that this peak cannot be caused by the Alm bundle structure. Instead, we interpret it as being due to two-dimensional hexagonally packed clusters in equilibrium with Alm bundles. As the relative humidity was reduced, interactions between Alm in neighboring bilayers produced more peaks with three-dimensional crystallographic character that do not index with the conventional hexagonal space groups.  相似文献
