A cell-free preparation of hydrogenase was obtained from acetone-driedcells of Anabaena cylindrica. This preparation was capable ofcatalyzing the reduction of various redox dyes by molecularhydrogen and evolution of hydrogen from reduced methyl viologen.PMS, methylene blue, DPIP and toluidine blue were effective,in this order, as hydrogen acceptors. Ferricyanide, p-quinone,phenosafranine and neutral red were not reduced by the presentpreparation. PPNR obtained from this alga was effective on neitherreduction of NAD and NADP by hydrogen nor stimulation of hydrogenuptake with methylene blue and PMS. A growth inhibitor, amarinin was isolated from the seeds ofLuffa amara. Cell lines producing monoclonal antibodies specific for non-derivatizedgibberellins (GAs) were prepared from spleen cells of a mouseimmunized with an immunogen that carried 16-substituted GA4as a hapten. The effects of some fluorinated pyrimidines on the growth ofexcised pea embryos {Pisum sativum var. Alaska) in sterile culturewere studied. Even the lowest concentrations of the compoundstested inhibited growth in length of the embryos. In order ofdecreasing activity, the compounds tested were : 5-fluorodeoxyuridine,5-fluorodeoxycytidine, 5-fluorouridine, 5-fluoroorotic acidand 5-fluorouracil. Inhibition of growth in length of the rootprimordia was found to be mainly due to inhibition of cell divisionwith no effect on cell elongation. In a search for callus-inducing compounds for indica rice thatare superior in this regard to 2,4-dichlorophenoxyacetic acid(2,4-D), we tested 13 derivatives of phenoxyacetic acids, namely,mono- and di- chloro-, mononitro- and 4-halogeno-substitutedphenoxyacetic acids for their ability to stimulate the inductionof calluses from mature seeds of rice (Oryza sativa L. cv. Chiemchanh). Five analogues of benzoic acid were also tested. Thecallus-forming ability and the optimal concentration for inductionof calluses were differed from compound to compound. The optimal concentration for callus induction of 4-fluorophenoxyaceticacid (4FPA) was relatively high (250 µM), but its callus-formingactivity was stronger than that of 2,4-D, even when comparedwith 2,4-D at its optimal concentration (50 µM). 4FPA-inducedcalluses retained their regenerative ability for at least 15months with 9 subcultures, while calluses induced by 2,4-D losttheir regenerative ability after 3 subcultures. 4FPA-induced calluses showed higher regenerative ability thandid calluses induced by 2,4-D, in almost all of 20 varietiesof indica tested, including three indica-japonica hybrids. 3Present address: Kibun Co.Ltd., Ginza, Tokyo, 104 Japan 4Present address: Hyogo Agricultural Research Institute, Kasai,Hyogo, 679-01 Japan (Received October 25, 1989; Accepted May 17, 1990)  相似文献

Etiolated oat protoplasts were treated with dibutyryl cAMP tostudy possible function of cAMP in the development by measuringthe protoplast swelling. The mean diameter of protoplasts inthe absence of any chemical treatment was 33.58±1.26(SE) µm, which increased to 36.96±0.86 µmin the presence of 100 µM dibutyryl cAMP. Prostacyclin,a potent activator of adenyl cyclase, also showed a significantswelling effect (diameter 38.01±0.98 µm). TA [4-ethoxy-1-(p-tolyl)-s-triazine-2,6 (1H, 3H)-dione] wasprepared chemically by intramolecular cyclization of IU [2-ethyl-3-methoxycarbonyl-1-(p-tolylcarbamoyl)-isourea],which has been reported to be an effective GA-synergist. TAalone slightly stimulated the shoot growth of rice seedlings,and in combination with GA showed a distinctly synergistic effecton the growth of rice shoots. The results suggest that TA andIU are nearly equal in their physiological activity. From theresults of the rice seedling test of these two compounds andtheir analogs, structure-activity relationships of isoureaswere correlated with those of triazinones. The results of applicationof IU, at 100 mg/liter to rice seedlings followed by extractionand silica gel thin-layer chromatography, suggested that IUwas biologically converted into its closed-ring form, TA, whichis a stable and potent GA-synergist.   1. Comparative studies were performed on growth, photosyntheticand respiratory activities, and pigment content in Rhodopseudomonaspalustris.
  2. The growth of the organism, as influenced by variousculturalconditions such as light, aerobiosis, anaerobiosisand nutritionalfactors was investigated.
  3. The respiratoryactivity of the bacterium was found to be higherin dark-growncells than in cells grown in the light. The photosyntheticactivitydid not significantly depend on the growth conditionsof theculture. Cells of younger cultures were found to be moreactivethan those of older cultures, with respect both to respirationand photosynthesis.
  4. The pigment content was found to be higherin the light-growncells than in the dark-grown ones. The ratiophotosyntheticactivity/bacteriochlorophyll was significantlyhigher in thelatter than in the former.
  5. Light, as well asvarious nutritional factors, was found toexert a marked accelerationon pigment formation, although ithas not yet been possibleto culture cells completely lackingin photosynthetic pigmentsand accordingly in photosyntheticactivity.

Pyruvate, orthophosphate dikinase (EC 2.7.9.1 [EC] ) was activatedin the light and inactivated following a dark treatment in intactmaize mesophyll chloroplasts. Addition of catalase (100–250units/ml) to the assay medium was necessary to obtain good activationand to keep the enzyme in an active state during illumination.Arsenate and carbonyl cyanide m-chlorophenyl-hydrazone, uncouplersof photophosphorylation, inhibited the activation. Pyruvate,which has been proposed to have a critical role in supportingthe light activation of pyruvate, orthophosphate dikinase, actuallyinhibited the activation. The pyruvate level in the chloroplastsuspension decreased when the enzyme was light-activated. Glutamine synthesis in germinating seeds of Cucurbita moschata   总被引:1,自引:0,他引:1  
During germination, an increase in glutamine and glutamine synthetase[L-glutamate: ammonia ligase (ADP), EC 6.3.1.2 [EC] ] occurred inthe cotyledons reaching a maximum at 4 to 6 days. The enzymehad a Km of 4.5 nun for L-glutamate, and 0.67 mu for hydroxylamine.Hydroxylamine exhibited substrate inhibition kinetics. The enzymewas inhibited by calcium ion, fluoride ion and p-hydroxymercuribenzoatebut not by EDTA, sodium ion or chloride ion. The sulf hydrylinhibition was reversed by dithiothreitol. The biphasic reaction course, fallover, of carboxyla-tion catalysedby ribulose 1,5-bisphosphate carboxylase/ox-ygenase (RuBisCO)has been known as a characteristic of the enzyme from higherland plants. Fallover consists of hysteresis in the reactionseen during the initial several minutes and a very slow suicideinhibition by inhibitors formed from the substrate ribulose-l,5-bisphosphate(RuBP). This study examined the relationship between occurrenceof fallover and non-catalytic RuBP-binding sites, and the putativehysteresis-inducible sites (Lys-21 and Lys-30S of the largesubunit in spinach RuBisCO) amongst RuBisCOs of a wide varietyof photosynthetic organisms. Fallover could be detected by followingthe course of the carboxylase reaction at 1 mM RuBP and thenon-catalytic binding sites by alleviation of fallover at 5mM RuBP. RuBisCO from Euglena gracilis showed the same linearreaction course at both RuBP concentrations, indicating an associationbetween an absence of fallover and an absence of the non-catalyticbinding sites. This was supported by the results of an equilibriumbinding assay for this enzyme with a transition state analogue.Green macroalgae and non-green algae contained the plant-type,fallover enzyme. RuBisCOs from Conjugatae, Closterium ehrenbergii,Gona-tozygon monotaenium and Netrium digitus, showed a muchsmaller decrease in activity at 1 mM RuBP than the spinach enzymeand the reaction courses of these enzymes at 5 mM RuBP werealmost linear. Methenyltetrahydrofolate cyclohydrolase (E.C. 3.5.4.9 [EC] ), whichis responsible for the enzymatic conversion of 5,10-methenyl-H4FAto 10-formyl-H4FA, has been found in various plant tissues.The enzyme was partially purified from pea seedlings and someof its properties were investigated. It was unstable, but wasstabilized by the addition of 25% glycerol. The enzyme was purifiedabout 60-fold by fractionation with ammonium sulfate and columnchromatography on DEAE-cellulose in the presence of 25% glycerol.Optimum pH for the reaction was 7.7. Michaelis constants for5,10-methenyl-H4FA in the forward reaction, and for 10-formyl-H4FAin the reverse reaction were 4x10–5M and 2x10–4M,respectively. The apparent equilibrium constant for the reactionwas calculated as 50. Enzyme activity was greatly inhibitedby the reduced forms of folate derivatives. The probable participationof this enzyme in the regulation of folate coenzyme levels inplant tissues has been suggested. 1 Studies on the enzymatic synthesis and metabolism of folatecoenzymes in plants, VI. A novel photorespiratory mutant of Arabidopsis thaliana, designatedgld2, was isolated based on a growth requirement for abnormallyhigh levels of atmospheric CO2. Photosynthetic CO2 fixationwas inhibited in the mutant following illumination in air butnot in atmosphere containing 2% O2. Photosynthetic assimilationof 14CO2 in an atmosphere containing 50% O2 resulted in accumulationof 48% of the soluble label in glycine in the mutant comparedto 9% in the wild type. The rate of glycine decarboxylationby isolated mitochondria from the mutant was reduced to 6% ofthe wild type rate. In genetic crosses, the mutant complementedtwo previously described photorespiratory mutants of A. thalianathat accumulate glycine during photosynthesis in air due todefects in glycine decarboxylase (glyD, now designated gld1)and serine transhydroxymethylase (stm). Because glycine decarboxylaseis a complex of four enzymes, these results are consistent witha mutation in a glycine decarboxylase subunit other than thataffected in the gld1 mutant. The planktonic calanoid copepod, Sinocalanus doerrii, a nativeof the rivers of mainland China was found in 1978 in California'sSacramento-San Joaquin Estuary during routine plankton sampling.Previous plankton surveys in 1963 and from 1972 to the presentindicate that the introduction occurred a relativery short timebefore specimens were first caught. The most probable mode ofintroduction is ballast water from Japanese freighters previouslydocking in China. Membranes containing protease were isolated from the cotyledonsof germinating pea seeds by differential and two successivesucrose density gradient centrifugations. Membranes were recoveredin the microsomal fraction, but could be clearly separated frommembranes carrying antimycin A-insensitive NADH-cytochrome creductase EC 1.6.2.1 [EC] ), which seem to come from the endoplasmicreticulum. The density of protease-containing membranes was1.135. Membranes contained a lower amount of phospholipid ascompared with the endoplasmic reticulum. Photosynthetic functions in leaves of cucumber (Cucumis sativusL.) and rice (Oryza sativa L.) were examined before and aftervarious chilling treatments. Cucumber leaves lost the capacityfor the photosynthetic oxygen evolution after chilling at 0°Cin the dark for 48 h. Thyla-koids isolated from such leaveswere not able to reduce dichloroindophenol (DCIP), but the additionof diphenylcarbazide (DPC), an electron donor to PS II, restoredthe ability to reduce DCIP, indicating that the site of damageis in the water-splitting machinery of PS II. In moderate light (500 jumol quanta m–2s–1), chillingof cucumber leaves at 5°C for 5 h was sufficient to inducethe complete loss of the capacity for photosynthetic oxygenevolution. Electron transport rates measured in thylakoids wereunaltered, but thylakoids were totally permeable to protons.Since the addition of dicyclohexylcarbodiimide (DCCD) restoredcoupling and the capacity for proton uptake, the primary siteof damage was deduced to be in the ATPase.
