目的利用激光共聚焦显微镜(Confocal Laser Scanning Microscopy, CLSM)观察大蒜素对铜绿假单胞菌PA01菌株生物膜(biofilms,BF)形成过程的影响,并用ISA(Image Structure Analyzer)软件对BF结构进行定量化分析。方法体外建立1d.3d和7d组PA01菌株BF模型,每组分3小组,分别用生理盐水、大蒜素128μg/mL、大蒜素10μg/mL作用6h。通过荧光探针SYT09/PI标记,利用CLSM进行动态观察,并用ISA软件对其BF结构进行定量分析。结果(1)生理盐水对照组铜绿假单胞菌生物膜厚度随时间的增加而增加,但在后3d增加的速度减慢;同时区域孔率(Areal Porosity,AP)呈下降趋势,平均扩散距离(Average Diffusion Distance,ADD)有逐渐增加趋势,结构熵(Textural Entropy,TE)有增加的趋势。(2)7d组模型中,大蒜素128μg/mL作用后,BF厚度由(28.83±0.67)μm减低到(16.50±0.69)μm;AP由0.76±0.10增加到0.92±0.02;TE由6.78±0.93减低到5.61±0.55。10μg/mL大蒜素干预后,有同样趋势,但效应不如高浓度明显。(3)1d、3d模型组,大蒜素作用后同对照组相比,厚度、AP、TE的差异均有统计学意义,趋势同7d组。结论通过ISA软件对干预前后的BF进行结构定量分析,大蒜素对各个时间段BF的形成均有影响,高浓度效果更显著。  相似文献

细菌生物被膜是细菌持续性致病的重要机制。研究细菌生物被膜的形成和发展可为顽固性细菌感染防治提供新的思路与策略。环二腺苷酸c-di-AMP(Cyclic diadenosine monophosphate)是继c-di-GMP之后在细菌中新发现的一种核苷酸第二信使分子。研究发现,c-di-AMP参与调节细菌多种生理功能,包括细菌生长代谢、生物被膜形成、细胞壁的合成以及细菌毒力因子等。本文综述了c-di-AMP参与调控细菌生物被膜形成的不同方式及其分子机制。鉴于c-di-AMP在调控细菌生物被膜中的重要性,其可作为抗细菌生物被膜感染新药研发的潜在靶点。  相似文献

The Asian citrus psyllid (Diaphorina citri) is a major pest of the citrus industry and is also the vector for Candidatus Liberibacter asiaticus (CLas), a destructive Huanglongbing (HLB) disease of citrus trees. Insect endosymbionts and gut bacteria play important roles in vector-pathogen interactions and host immunity. Thus, our aim was to evaluate the correlation between CLas infection and the microbiome in D. citri by conducting 16S rRNA amplification sequencing on insects successfully and unsuccessfully infected with CLas (CLas-infected and non-infected). Genera Candidatus Profftella, Wolbachia, and Candidatus Carsonella were highly abundant genera in all tested samples. Compared with the non-infected and control groups, CLas-infected samples harboured more observed OTUs and showed higher alpha diversity metrics. Principal coordinate analysis based on beta-diversity metrics indicated two distinct clusters between the CLas-infected samples and non-infected/control samples. Subsequent LEfSe analysis revealed that Candidatus Profftella was more abundant in the non-infected group than in the control and CLas-infected groups. The interaction network also indicated a co-exclusion relationship between Candidatus Profftella and CLas, while CLas co-existed with Wolbachia, several Enterobacteriaceae spp., and multiple other bacteria. Our study provides insight into the interaction between the microbiome community in D. citri and CLas, which can facilitate the management of this pest and its associated pathogen.  相似文献

目的构建惰性材料塑料输液管内壁细菌生物膜体外模型,观察细菌生物膜的结构,探讨输液管内壁细菌生物膜形成影响因素。方法建立铜绿假单胞菌生物膜和铜绿假单胞菌、肺炎克雷伯菌混合生物膜,分别于培养1、3和7d用扫描电镜动态观察生物膜形成过程。结果混合菌生物膜的生长速度高于铜绿假单胞菌单独成膜。结论输液管是形成细菌生物膜的良好支持材料,混合细菌培养可以加速细菌形成生物膜。  相似文献

目的通过生物膜定量分析仪来观察铜绿假单胞菌(Pseudomonas aeruginosa PAO1),变形链球菌(Streptococcus mutans UA159)以及大肠埃希菌(Escherichia coli MG1655)生物膜形成能力的不同,并以各菌株的吸光度值A600为参考,对3种菌株早期生物膜形成能力进行比较。方法通过向生物膜培养悬液中加入与细菌直径相近的磁性小珠,利用这些小珠在磁场中受到生物膜的位移约束力的原理,采用生物膜定量分析仪,定量比较3种菌株在生物膜形成上的差别。结果实验发现铜绿假单胞菌PAO1和大肠埃希菌MG1655的细菌增长速度基本相同,但铜绿假单胞菌PAO1的生物膜形成明显快于大肠埃希菌MG1655。大肠埃希菌MG1655和变形链球菌UA159的生物膜形成速度基本相同,但大肠埃希菌MG1655的细菌增长速度明显高于变形链球菌UA159。结论不同细菌有各自的生物膜形成模式。生物膜定量分析仪作为一种高效简便的检测手段,可用于生物膜早期形成的动态分析。  相似文献

Although biofilm formation is widely documented on Earth, it has not been demonstrated in the absence of gravity. To explore this possibility, Pseudomonas aeruginosa, suspended in sterile buffer, was flown in a commercial payload on space shuttle flight STS-95. During earth orbit, biofilm formation was induced by exposing the bacteria to sterile media through a 0.2-microm (pore size) polycarbonate membrane. Examination of these membranes by confocal microscopy revealed biofilms to be present and that these biofilms could persist in spite of vigorous agitation. These results represent the first report of biofilm formation under microgravity conditions.  相似文献

The development and use of small-molecule inhibitors of the adherence of Porphyromonas gingivalis to oral streptococci represents a potential therapy for the treatment of periodontal disease as these organisms work in tandem to colonize the oral cavity. Earlier work from these laboratories demonstrated that a small synthetic peptide was an effective inhibitor of the interaction between P. gingivalis and Streptococcus gordonii and that a small-molecule peptidomimetic would provide a more stable, less expensive and more effective inhibitor. An array of 2-(azidomethyl)- and 2-(azidophenyl)-4,5-diaryloxazoles having a full range of hydrophobic groups were prepared and reacted with substituted arylacetylenes to afford the corresponding ‘click’ products. The title compounds were evaluated for their ability to inhibit P. gingivalis’ adherence to oral streptococci and several were found to be inhibitory in the range of (IC50) 5.3–67 μM.  相似文献

细菌通过自身合成的水合多聚物粘附在固体表面,以固着的方式生长从而形成生物膜,细菌生物膜的形成涉及到几个明显的阶段,包括起始的附着、细胞与细胞之间的吸附与增殖、生物膜的成熟、及最后细菌的脱离等四个阶段,生物膜的形成增加了细菌对抗生素的抗性以及帮助细菌逃逸寄主的免疫攻击等,从而引起临床上持续性的慢性感染等各种问题;生物膜结构非常复杂,除了细菌分泌的各种胞外多糖,胞外蛋白质外,最新的研究表明,DNA也是生物膜的一个重要成分.针对近年来的最新文献报道分别对生物膜的形成、结构以及调控机制等进行综述.  相似文献

Bacterial communities such as biofilms are widely recognized as being important for survival and persistence of bacteria in harsh environments. Mechanistic models of biofilm growth indicate that the way in which the surface is seeded can effect the morphology of simulated biofilms. Experimental studies indicate that genes which are important for chemotaxis also influence biofilm formation, perhaps by influencing aggregation on a surface. Understanding aggregation and microcolony formation could therefore help clarify factors influencing biofilm formation and illuminate how groups influence the fitness of bacteria. In this paper I develop an individual based model to examine how different behaviors involved in microcolony formation on a surface determine patterns of group sizes and link patterns to bacterial fitness.  相似文献

白念珠菌是人体重要的条件性致病真菌。形态的多样性和可塑性是白念珠菌典型的生物学特征,这与它的致病性、宿主适应能力以及有性生殖过程密切相关。白念珠菌生物被膜(Biofilm)是由不同形态细胞(包括酵母型、菌丝和假菌丝)以及胞外基质组成的致密结构,也是毒性和耐药性形成的重要因子。生物被膜对抗真菌药物、宿主免疫系统和环境胁迫因子等都表现出较强的抵抗力和耐受性,是临床上病原真菌感染防治的重大挑战。随着基因表达谱和遗传操作技术的发展,白念珠菌生物被膜的形成及其耐药性的获得所依赖的遗传调控通路和分子调控机制越来越清楚。主要包括MAPK和cAMP介导的信号途径以及Bcr1和Tec1等因子介导的转录调控。此外,白念珠菌生物被膜的形成与形态转换和有性生殖之间存在密切的联系。文中综述了白念珠菌生物被膜形成的遗传调控机制,重点介绍了细胞壁相关蛋白、转录因子和交配型对该过程的调控以及生物被膜的耐药机制。  相似文献

Many bacteria utilize sophisticated regulatory systems to ensure that some functions are only expressed when a particular population density has been reached. The term 'quorum-sensing' has been coined to describe this form of density-dependent gene regulation which relies on the production and perception of small signal molecules by bacterial cells. As in many pathogenic bacteria the production of virulence factors is quorum-sensing regulated, it has been suggested that this form of gene regulation allows the bacteria to remain invisible to the defence systems of the host until the population is sufficiently large to successfully establish the infection. Here we present first evidence that polyphenolic compounds can interfere with bacterial quorum-sensing. Since polyphenols are widely distributed in the plant kingdom, they may be important for promoting plant fitness.  相似文献

Many bacterial pathogens have defined in vitro virulence inducing conditions in liquid media which lead to production of virulence factors important during an infection. Identifying mutants that no longer respond to virulence inducing conditions will increase our understanding of bacterial pathogenesis. However, traditional genetic screens require growth on solid media. Bacteria in a single colony are in every phase of the growth curve, which complicates the analysis and makes screens for growth phase-specific mutants problematic. Here, we utilize fluorescence-activated cell sorting in conjunction with random transposon mutagenesis to isolate bacteria grown in liquid media that are defective in virulence activation. This method permits analysis of an entire bacterial population in real time and selection of individual bacterial mutants with the desired gene expression profile at any time point after induction. We have used this method to identify Vibrio cholerae mutants defective in virulence induction.  相似文献

Monitoring bacterial communities is critical for assessing biodeterioration among other processes. This study presents a strategy and an example of comparative analysis of bacterial communities developing in a cave environment, Altamira Cave which contains unique paleolithic paintings. The analyzed question was whether white colonizations discovered throughout the cave corresponded to similar or different bacterial communities. Molecular fingerprints were obtained by PCR–DGGE from DNA and RNA and statistically compared. Results based on DNA analysis showed that a similar bacterial community was present in white colonizations throughout the cave. Fingerprints based on RNA confirmed the similarity of the major metabolically active components of these communities. The proposed procedure confirmed that white colonizations in Altamira Cave were a consequence of the development of a single complex bacterial community, and the method proves to be highly useful for comparative analysis of microbial communities, including biodeteriorating processes and any other comparative analysis of bacterial communities.  相似文献

The bacterial and eukaryotic communities forming biofilms on six different antifouling coatings, three biocidal and three fouling-release, on boards statically submerged in a marine environment were studied using next-generation sequencing. Sequenced amplicons of bacterial 16S ribosomal DNA and eukaryotic ribosomal DNA internal transcribed spacer were assigned taxonomy by comparison to reference databases and relative abundances were calculated. Differences in species composition, bacterial and eukaryotic, and relative abundance were observed between the biofilms on the various coatings; the main difference was between coating type, biocidal compared to fouling-release. Species composition and relative abundance also changed through time. Thus, it was possible to group replicate samples by coating and time point, indicating that there are fundamental and reproducible differences in biofilms assemblages. The routine use of next-generation sequencing to assess biofilm formation will allow evaluation of the efficacy of various commercial coatings and the identification of targets for novel formulations.  相似文献

Total heavy metal concentrations in marine sediments are not sufficient to reliably predict detrimental biological effects. Here we provide evidence that only bioavailable heavy metals have a significant impact on benthic microbial loop functioning. Sediment samples collected along 250 km of the Apulian coast (Mediterranean Sea) were analysed for total and bioavailable heavy metals (Cr, Cd, Pb and Cu), organic matter content, bacterial abundance, biomass and carbon production and -glucosidase activity. Sampling strategy was specifically designed to cover a wide range of environmental conditions and types of anthropogenic influences. Total heavy metal concentrations in the sediments were tightly coupled with organic matter content, whereas bioavailable heavy metal concentrations displayed an opposite pattern. Bioavailable Cr concentrations were up to 10-fold higher than values observed for the other bioavailable metals and significantly inhibited benthic bacterial metabolism and turnover. Results from this study suggest that functional microbial variables are highly sensitive to heavy metal contamination and could be used as bioindicators of stress conditions in coastal sediments.  相似文献

Abstract A screening of twenty-two marine isolates was made to examine their effects on corrosion of carbon steel ASTM A619. In batch cultures, sixteen of the isolates gave a lower corrosion than the control. Aerobic and anaerobic biofilm populations were formed by immersing iron coupons in natural seawater under aerobic and anaerobic conditions. The effects of the biofilms depended on a balance between the presence of oxygen and the type of population. An anaerobic population attached to the surface increased the corrosion rate if immersed in a suspension of Vibrio sp. DW1. The vibrio population probably 'protected' the anaerobic population from oxygen and may have provided nutrients, thereby creating conditions that allowed production of corrosive metabolites close to the metal. In contrast, coupons without a biofilm showed a decrease in the corrosion when immersed in the same vibrio suspension. The protective effect of a dense suspension of bacteria found earlier [5,6] was tested in situ in seawater. Iron coupons were immersed in dialysis bags with a suspension of Vibrio sp. DW1. Coupons immersed in dialysis bags with DW1 showed a lower degree of corrosion than coupons immersed in bags with seawater.  相似文献

AIM: To study the effect of antiseptics on bacterial biofilm formation. METHODS AND RESULTS: Biofilm formation and planktonic growth were tested in microtiter plates in the presence of antiseptics. For Escherichia coli G1473 in the presence of chlorhexidine or benzalkonium chloride, for Klebsiella pneumoniae CF504 in the presence of chlorhexidine and for Pseudomonas aeruginosa PAO1 in the presence of benzalkonium chloride, biofilm development and planktonic growth were affected at the same concentrations of antiseptics. For PAO1 in the presence of chlorhexidine and CF504 in the presence of benzalkonium chloride, planktonic growth was significantly inhibited by a fourfold lower antiseptic concentration than biofilm development. For Staphylococcus epidermidis CIP53124 in the presence of antiseptics at the minimal inhibitory concentration (MIC), a total inhibition of biofilm formation was observed. For Staph. epidermidis exposed to chlorhexidine at 1/2, 1/4 and 1/8 MIC, or to benzalkonium chloride at 1/8, 1/16 or 1/32 MIC, biofilm formation was increased from 11.4% to 22.5% without any significant effect onto planktonic growth. CONCLUSIONS: Chlorhexidine and benzalkonium chloride inhibited biofilm formation of different bacterial species but were able to induce biofilm development for the Staph. epidermidis CIP53124 strain at sub-MICs. SIGNIFICANCE AND IMPACT OF THE STUDY: Sublethal exposure to cationic antiseptics may contribute to the persistence of staphylococci through biofilm induction.  相似文献

Bacteria can switch between planktonic forms (single cells) and biofilms, i.e., bacterial communities growing on solid surfaces and embedded in a matrix of extracellular polymeric substance. Biofilm formation by pathogenic bacteria often results in lower susceptibility to antibiotic treatments and in the development of chronic infections; thus, biofilm formation can be considered an important virulence factor. In recent years, much attention has been directed towards understanding the biology of biofilms and towards searching for inhibitors of biofilm development and of biofilm-related cellular processes. In this report, we review selected examples of target-based screening for anti-biofilm agents: We focus on inhibitors of quorum sensing, possibly the most characterized target for molecules with anti-biofilm activity, and on compounds interfering with the metabolism of the signal molecule cyclic di-GMP metabolism and on inhibitors of DNA and nucleotide biosynthesis, which represent a novel and promising class of biofilm inhibitors. Finally, we discuss the activation of biofilm dispersal as a novel mode of action for anti-biofilm compounds.  相似文献
