In the leopard frog (Rana pipiens), thymic lymphocytes do notoriginate from blood-borne stem cells that migrate into thethymus anlage; rather they arise in situ from elements in thethymic rudiment itself. After thymic differentiation, the lymphocytes(or their descendants) leave the thymus and extensively seedthe peripheral lymphoid organs. Indeed, virtually all the lymphocytesin the spleen, kidney, and bone marrow are ontogenically derivedfrom thymic cells. In postmetamorphic life, the thymus representsan organ in which lymphopoiesis is genuinely self-sustaining.Throughout the juvenile life of the frog, there is no indicationof an inward afferent stream of cells entering the intact thymus.  相似文献

A histological study was undertaken of the differentiation of the lymphomyeloid complex of larvae of the common leopard frog, Rana pipiens, reared at 18–21 °C. Immature lymphocytes are first recognised in the thymus and pronephros at stage I of Taylor and Kollros ('46). By the end of stage II, small lymphocytes are regularly found among the predominant larger lymphoid cells in the thymus, in which corticomedullary differentiation has begun. At this time, a few small lymphocytes are also apparent in the ventral cavity bodies, lymph gland, pronephros, mesonephros and intestine, but rarely occur in the spleen. During stages III and IV extensive development of these components of the lymphomyeloid complex occurs. The organs now contain large numbers of mature lymphocytes and have attained states of differentiation that remain essentially similar in subsequent larval stages. By stage V, small epithelium-associated lymphoid accumulations are abundant throughout the length of the gastrointestinal tract. The anlagen of the procoracoid body and bone marrow appear just prior to metamorphosis. The kidneys are the main sites of blood formation in the larva. Masses of granulocytes are also usually found in the abundant ventral cavity bodies. A consideration of the roles of each of the organs provides insight into the ontogeny of the immune system of the Anura in general.  相似文献

Summary A study of the ultrastructure of the developing thymus of the leopard frog (Rana pipiens) revealed that the thymus had undergone all of the major changes which would persist through larval life and metamorphosis by the time that the animals had reached larval stage IV of Taylor and Kollros (1946). These changes included development of an outer, lymphoid cortical region and an inner, essentially nonlymphoid medulla; mitotic activity among lymphoid cell precursors and the formation of the first small lymphocytes; development of complex cysts containing PAS-positive material and the appearance of other signs of secretory activity among epithelial cells of the medulla; and differentiation of large myoid cells containing bundles of striated muscle fibrils. Extirpation of the thymus, during its differentiation as a lymphoid organ, in the early larval life of the leopard frog was attended by a marked impairment of allograft immunity. In cases in which thymectomy was most successful—i.e., in which no thymic tissue was found in the larvae on later autopsy—the survival of allogeneic grafts was greatly prolonged. When thymectomy was less successful—i.e., when fragments ranging widely in size were found in the larvae at autopsy—the results were variable: the larvae either rejected allografts more slowly than control larvae or responded to them at the same rate as the control larvae. The “less successful” cases of thymectomy most likely represented instances of the regeneration of thymic tissue.  相似文献

This paper examines the thymic dependence of alloimmunity inamphibians. In Xenopus, the presence of a thymus during thefirst 2 weeks of life is essential for the development of normalfirst-set skin allograft immunity. Thymectomy during this earlyperiod always impairs the alloimmune response of young adulttoads. However, most of these thymectomized animals are ableto completely destroy skin allografts, albeit with prolongedrejection times. Chronic graft rejection, rather than tolerance,still occurs following thymectomy as early as 5 days, when thethymus contains no small lymphocytes. In contrast to the considerabledifferences in first-set allograft survival times in controland early-thymectomized Xenopus, second-set grafts, appliedsubsequent to first-set destruction, are rejected in acute fashion(<3 weeks) in both groups. That the defect in first-set alloimmunityis specifically related to absence of thymus has been confirmedby implanting allogeneic thymus 2 weeks post-thymectomy. Thedonor thymus remains healthy and restores the allograft responseto normal. In contrast, allogeneic spleen does not reconstituteand itself often undergoes destruction. Preliminary autoradiographicexperiments on lymphoid tissue involvement in first-set allograftrejection are also described.  相似文献

Wetland hydroperiod is a key factor for the reproductive success of pond-breeding amphibians. Ground-water withdrawals may cause intermittent ponds to dry prematurely, potentially affecting amphibian development. In three intermittent ponds, we monitored hydrology and tracked oviposition, larval development, and metamorphosis for three frog species that represented a range of breeding phenologies. The three species were the southern leopard frog (Lithobates sphenocephalus), spring peeper (Pseudacris crucifer), and Pine Barrens treefrog (Hyla andersonii). We simulated ground-water withdrawals by subtracting from 5 to 50 cm (in 5-cm increments) from the measured water-depth values at the ponds over a short-term (2-year) period and a long-term (10-year) period to estimate the potential impact of hydroperiod alterations on frog development. Short-term simulations indicated that 5 and 10 cm water-depth reductions would have resulted in little or no impact to hydroperiod or larval development and metamorphosis of any of the species. Noticeable impacts were estimated to occur for reductions ≥15 cm. Long-term simulations showed that impacts to the appearance of the first pre-metamorphs and metamorphs would have occurred at reductions ≥10 cm and impacts to initial egg deposition would have occurred at reductions ≥20 cm. For all simulations, successively greater reductions would have caused increasing impacts that varied by species and pond, with the 50-cm reductions shortening hydroperiods enough to practically eliminate the possibility of larval development and metamorphosis for all three species. Compared to the spring peeper and southern leopard frog, the estimated impacts of the simulations on the various life stages were the greatest for the Pine Barrens treefrog.  相似文献

Cirripede larvae can occur year-round in temperate and tropicalwaters, often in significant numbers, yet the species compositionof the ‘Balanus sp.’ component is rardy studied.Weekly plankton samples were analyzed qualitatively for larvalcirripede species and stage over a year (1977–1978) attwo Rhode Island stations. Six species of larvae were foundin Lower Narragansett Bay (30° salinity). Semibalanus balanoidesand Balanus balanus have a single winter brood. S. balanoidesis the predominant winter breeder with a minor release of naupliiin early December and major release in March followed by cypridsin mid-April.B. balanus populations release all larvae in Marchwith cyprids in mid-April. Balanus crenatus is mainly a winterbreeder, but has multiple broods; it does not breed in July-Septemberwhen the water temperature is above 18°C. Balanus venustusis the predominant summer breeder, and larvae were observedfrom May through December (water >8°C). Larvae of Chthamalusfragilis and Balanus eburneus occur in low numbers from May-October.At the Pettaquamscutt River site (12 salinity), Balanus improvisuslarvae predominate and early stage nauplii (I-II) occur in samplesyear round (0–27°C). Two peaks of later stage naupliiand cyprids occur in late spring (May) and early winter (Nov.-Jan.).Continued temperatures bdow 5°C or above 20°C appearto inhibit larval development. Comparison of results with existing literature reveals severalsignificant findings. The bimodal rdease of S. balanoides larvaeis unusual and may be in response to the phytoplankton dynamicsof the year; however, the existence of distinct races of S.balanoides may also be a factor. Larvae of B. venustus predominatein the lower bay during the summer, yet this species is unreportedin past studies. B. improvisus nauplii are more cold tolerantthan previously reported. Respiratory activity and seasonal changes in carbohydrate contentof the storage organs of Rubus chamaemorus L. have been investigated.Leaf dark respiration rate increases in a non-linear mannerfrom 0·7 mg CO2 evolved dm–2 h–1 at 0 °Cto 4·6 rng CO2 evolved dm–2 hh–1 at 30 °C.Root and rhizome respiration rates increase from 1 µ1O2 uptake g–1 fresh weight h–1 at 0.7 ° C to10 µ10, uptake g–1 f. wt h–1 at 20 °C.Rhizome carbohydrate reserves decline from a September peakof 33 per cent alcohol insoluble d. wt to 16 per cent in May. Experimental studies have demonstrated that for many marineinvertebrate species, variability in larval condition or qualityat settlement may have important effects on post-settlement,early juvenile performance. Relatively few studies, however,explicitly examine natural variability in larval condition atsettlement. This study examines natural variability in larvalattributes (size and lipid index) at settlement for terminal-stagelarvae of intertidal mussels (Mytilus sp.) and barnacles (Pollicipespolymerus and Chthamalus dalli) from southern California. Despitesignificant differences among cohorts in larval attributes,for all 3 species a greater percentage of the variance in larvallength (80–100%) and lipids (58–83%) occurred amongindividuals within a cohort, rather than among cohorts. Forall 3 species, coefficients of variation within a cohort forlength were much smaller (3–8%) than those for lipid index(30–93%), suggesting that lipid storage is a much moreplastic attribute than size for larvae. For mussels, settlementintensity and larval attributes were decoupled, such that averagelarval condition of a cohort was not related to the number oflarvae that settled. At the cohort level, Mytilus and Pollicipessettling together across 3 dates showed similar trends of decreasinglipid index over time, suggesting that environmental conditionsmay influence co-occurring planktonic larvae similarly acrossspecies. This work highlights the need for further experimentsin the field on the effects of larval history on recruitmentsuccess in natural populations, and further studies to determinewhat factors influence larval attributes for planktonic larvaein the field.  相似文献

Larvae of Mithrax caribbaeus were reared in the laboratoryin a factorial experiment employing three temperatures (22,25 and 28°C) and three salinities (32, 35 and 38). Survivaland duration of larval stages were recorded. Ovigerous femalesof M.caribbaeus were collected from the south-eastern coastof Margarita Island, Venezuela, and maintained in individualaquaria until hatching. Eggs from three of the females hatchedin the laboratory. Larvae from each hatching were subdividedinto groups of 10 and reared in plastic bowls containing 200ml filtered and UV-irradiated sea water at different temperature–salinitycombinations. Larvae were transferred daily to clean bowls withnewly hatched Artemia nauplii, and the number of molts and mortalitywithin each bowl was recorded. Complete larval development ofM.caribbaeus occurred under all experimental conditions. Salinityhad the greatest effect on percentage survival of each larvalstage and complete development up to the first crab stage. Thefirst zoeal stage exhibited the highest survival rate. Maximumsurvival for this stage occurred at 25°C, 32–35. Survivalin the second zoeal stage and the megalopa was affected onlyby salinity. Effects of temperature and salinity on survivaldecreased with advance in development. The duration of the twozoeal stages, the megalopa, and development to the first crabstage showed a gradual reduction with increasing temperature.Salinity showed an effect on the duration of zoeal stages butnot on the megalopal stage. Development from hatching to thefirst crab stage required 8–18 days, depending on thetemperature–salinity combination, and was inversely relatedto temperature, averaging 14.3 days at 22°C, 11.8 days at25°C and 9.2 days at 28°C.  相似文献

Development of T lymphocytes in Xenopus laevis was studied using a mouse monoclonal antibody (mAb), XT-1, that was produced against surface determinants on thymocytes of J strain frog. Ontogenic studies, employing immunofluorescence, showed that cells positive for the determinant recognized by XT-1 mAb (XT-1+ cells) were first detected in the thymus of J strain Xenopus by Nieuwkoop and Faber stage 48 (7 days postfertilization) and then in the spleen, liver and kidney by stage 52 (20 days postfertilization). Percentages of XT-1+ cells in the thymus increased rapidly by stage 49 (10 days postfertilization) and reached adult levels by stage 52, and those in the spleen, liver, and kidney reached adult levels by stage 56 (40 days postfertilization). Electron microscopic immunohistochemistry revealed that most XT-1+ cells in thymuses from stage 56 larvae were typical small lymphocytes (4–7 μm in diameter). In contrast, many XT-1+ cells in larval thymuses at stage 49 are large (8–10 μm in diameter) lymphoblastoid cells. Thymectomy at stage 46 (5 days postfertilization) depleted XT-1+ cells in larval and adult lymphoid organs to background levels. These results suggest that the XT-1+ cells are differentiated from the lymphoid precursor cells in the thymus before the appearance of small lymphocytes and migrate into peripheral lymphoid organs. The cell surface determinant recognized by the XT-1 mAb may provide an important marker for the differentiation of T lymphocytes in Xenopus.  相似文献

The turions of Myriophyllum verticillatum, an aquatic vascularplant, develop in the fall and function in propagation and dispersalas well as in over-wintering. Experiments with controlled evnironmentsindicate that both temperature and photoperiod regulate turionformation. Turions can be induced at 15°C or lower, butnot at 20°C. At 15°C, turions form in both 8- and 12-hrdays, but not in 16-hr days. Plants collected in early springdo not form turions readily in response to short days unlesspreviously exposed to long days; thus, turion formation is along-day-short-day response. This combination of photoperiodand temperature requirements probably prevents turion developmentin early spring when the temperature and photoperiod are similarto those in the fall. Treatment of plants with ABA (10–5M) enhances turion development under marginally inductive conditions(12-hr days at 15°C) but cannot induce it under long days.On the other hand, the cytokinin benzyladenine (10–5 M)blocks turion formation. GA3 (10–5 M) and AMO-1618 (10–5M) exert only small qualitative effects on turion development,while IAA (10–5 M) retards it. During turion development,the level of ABAlike activity and of one or two unidentifiedinhibitors increases. Cytokinin activity decreases at the startof turion formation, increases during development, then decreasesat abscission. The relationship between the induction of tracheary elementdifferentiation and exogenous L-methionine was examined in agar-growncultures of soya bean callus initiated from Glycine max L. ‘Wayne’and ‘Clark 63’. Although Wayne is a normal cultivarsoya bean, seedlings of Clark 63 exhibit abnormal growth at25 °C due to exessive ethylene biosynthesis at this temperature.Wayne callus showed increased xylogenesis in the presence ofexogenous L-methionine (3.7 µg 1–1) in comparisonto IAA–KN controls at both 20 and 25 °C. Clark 63callus produced greater numbers of tracheary elements in responseto exogenous L-methionine only at 25 °C. The induction ofxylem differentiation was independent of the maintenance temperatureof the stock cultures of both cultivars. Xylogenesis initiatedbyan IAA–KN medium was inhibited by the addition of AgNO3(20 mg 1–1) to the extent of 76.5 per cent in cv. Wayneand 6 per cent in cv. Clark 63. The inhibitory effect was partiallyreversed by the addition of L-methionine (3.7 µg 1–1)to the IAA–KN–AgNO2 medium. The environmental control of flowering in some arctic-alpineCarexspecies has been studied in controlled environments.Carex nigra,C. brunnescens, C. atrata, C. norwegica andC. serotina all hada dual induction requirement for flowering. In all exceptC.nigra either low temperature (12 °C or lower) or short days(SD) over a wider range of temperatures were needed for primaryfloral induction and inflorescence formation. InC. nigra primaryfloral induction took place in SD only (9–21 °C),8–10 weeks of exposure being required for a full response.In all these species long days (LD) were required for, or stronglypromoted, culm elongation and inflorescence development (secondaryinduction). Quantitative ecotype differences in both primaryand secondary induction were demonstrated. Unlike the otherspecies,C. bicolor proved to be a regular LD plant which requiredLD only for inflorescence initiation and development. In allspecies leaf growth was strongly promoted by LD, especiallyin the higher temperature range (15–21 °C). In SDand temperatures below 15 °C the leaves became senescentand the plants entered a semi-dormant condition which was immediatelyreversed by LD. Two limpet species occur intertidally on subantarctic SouthGeorgia, the patellid Nacella concinna and the siphonarlid Kerguelenellalateralis. N. concinna is confined to the lower shore closeto LWS; K. lateralis occurs in middle shore pools, so theirdistributions do not overlap. N. concinna has a much narrowerthermal niche (–12.9°C to +15.6°C) than K. lateralis(–17.8°C to +31.8°C). Environmental data are presentedto show that the upper lethal temperature of N. concinna islow enough to prevent the limpet living higher on the shore.Both limpet species are slow-moving, but K. lateralis showsincreasing speed with rising temperature, peaking at 15–20°C.In contrast, N. concinna moves actively down to –1.9°C(when sea water freezes), but there is a steady decrease inspeed of locomotion above +2°C. Locomotion ceases at 14°Cin N. concinna (c.f. 30°C in K. lateralis). Both speciesexhibit very low tenacities, but in N. concinna tenacity decreaseswith increasing shell length. In K. lateralis there is no effectof temperature on tenacity. Both species show a positive allometricrelationship between foot area and shell length. N. concinnafeeds upon microbial films and microepiflora, but K. lateraliseats colonial diatoms and Enteromorpha bulbosa. Observationson shell middens of the kelp gull Larus dominicanus showed thatthe gulls did not eat K. lateralis, though they ate great quantitiesof the less accessible N. concinna. Gulls ate N. concinna assmall as 11 mm shell length (within the size range of K. lateralis).Experiments on gulls demonstrated an unwillingness to eat K.lateralis, probably because the siphonariid extrudes a viscidwhite mucus when the foot is touched. ABA Levels and Effects in Chilled and Hardened Phaseolus vulgaris   总被引:3,自引:0,他引:3  
Leaf abscisic acid (ABA) levels of chilled P. vulgaris weremeasured after 18 h chilling at 5°C, at a saturation deficitof 1.24 g m–3 (SD), and after chilling in a water-saturatedatmosphere. Changes were also followed during a chill hardeningperiod of 4 d at 12°C, 2.1 g m–3 SD. It was foundthat hardening resulted in an almost 5. fold increase in ABAlevels after 3 d at 12°C, and this decreased to approximatelycontrol levels on the fourth day. Subsequent chilling of hardenedplants produced no change in ABA levels from that of controlplants (22° C). In contrast, non-hardened plants chilledat 1.24 g m–3 SD had ABA levels almost 3 times the levelof control plants. However, chilling in a water-saturated atmosphereresulted in a decrease in ABA levels. In addition, the response of leaf diffusion resistance (LDR)to exogenous ABA fed via the transpiration stream was measuredat 5 ° C and 22° C in hardened and non-hardened plants.Use of tritium-labelled ABA was made to calculate the stomatalsensitivity to ABA. It was found that exogenous ABA caused anincreased in LDR at 22°C in both hardened and non-hardenedplants. However, the sensitivity of the hardened plants to ABAwas greater in terms of rate of closure and amount of ABA requiredto close the stomata. At 5°C, however, ABA caused stomatalopening and the maintainance of open stomata in non-hardenedplants. In hardened plants, ABA caused stomatal closure at 5°C.These results are discussed in relation to the locking-openresponse of chilled P. vulgaris stomata. Centropages abdominalis is a neritic, omnivorous, temporallyabundant copepod present throughout the subarctic Pacific andits marginal seas. The two main objectives of this study wereto determine how temperature influences the development of C.abdominalis and whether growth rates of in situ populationsmay be limited by available food. At 6.9°C, median developmenttime from eggs laid to 50% adults was 42 days and the averageweight-specific somatic growth rate was 0.17 day–1. At4.6°C, median development time to adult was 59 days (projected)and growth rate averaged 0.08 day–1, suggesting that 4.6°Cmay be approaching the lower temperature for development andgrowth in this species. The functional relationship betweendevelopment time and temperature was established over the temperaturerange in which this species occurs. The in situ adult growthrates between 10 and 13°C averaged 0.14 day–1 andwere generally lower than the laboratory-reared juvenile growthrates, which may indicate that adult C. abdominalis are foodlimited in the field during summer and autumn.  相似文献

The variation in the extent to which autolysis of Aspergillusniger van Tieghem occurs in response to alteration of temperatureand of the length of the period of autolysis has been studied.The degree of autolysis at 10, 15, 20, 25, 30 and 35 °Cwas 15–8, 23–9, 35–2, 40–8, 49–1and 56–2 per cent, respectively. Autolysis increased sharplywith the length of the period of autolysis reaching a plateautowards the second day at the lowest temperatures (0–20°C) and towards the fourth day at the higher temperatures.Theextent of the increase of autolysis plotted against temperatureindicates a linear relationship between these two parameters.The release of protein from autolysing mycelium of A. nigeris greatly affected by temperature, amounting to 1–9 percent at 10 °C, whereas at 30 °C it reached 5–1per cent. On the other hand, gradual increase of temperatureduring autolysis seems not to alter the content of mycelialcarbohydrates of A. niger.  相似文献

Maximum daily consumption was estimated for Mysis relicta fedad libitum rations of Daphnia pulex at 4,10,15 and 18°C.Gut-residence time was also evaluated for M.relicta fed clado-ceranprey at 4, 10 and 157deg;C. Mean daily consumption (g dry weightof Daphnia g–1 dry weight of Mysis day–1) rangedfrom 6% at 4%C to 12% at 10°. At 18°C, Mysis feedingrate declined to 9% day1. Mean, weight-adjusted consumptionrates exhibited a ‘dome-shaped’ response in relationto water temperature. Consumption rate was highest at 10°Cand lowest at 4°C. Estimated Q10 was more sensitive from4 to 10°C (Q10= 3) than from 10 to 15°C (Q10=1.2). Gut-residencetime for Mysis was inversely related to water temperature, implyingthat evacuation rate increases linearly with water temperature.Feeding and gut-evacuation rates become disassociated at watertemperatures >10°C. As water temperature increased above1°C, relative evacuation rate increased, whereas feedingrate declined. Simultaneous quantitative analyses have been made of the endogenouslevels of auxin- and gibberellin like substances, growth inhibitors,and auxin-oxidizing enzyme activity in the cold-requiring Chrysanthemummorifolium cv. Sunbeam subjected to different daylength, lightintensity and temperature regimes known to affect flowering.While little hormone or enzyme activity was found in extractsfrom unvernalized plants, a striking rise in auxin-oxidizingenzyme activity occurred rapidly after the end of cold treatment.Increased auxin activity was also recorded shortly after vernalization.At 28 °C both enzyme and auxin activity declined over aperiod of 3–4 weeks; at 20 °C this response was delayed.Gibberellin activity at 28 °C rose steeply about 2 weeksfrom vernalization and declined several weeks later; at 20 °Ca similar response was less marked. Low light intensity treatment,which may have increased endogenous auxin levels, or exogenousauxin application reduced gibberellin-like substance levelsand cause d devernalization.Phosphon D treatment also loweredgibberellin levels and prevented flowering. An extract fromvernalized plants containing gibberellin-like substances intensifiedthe flowering of partially vernalized test plants. Persistenceof high auxin activity in vernalized plants on long days wasassociated with failure to form normal flower buds. Stem elongationrates correlated in general with levels of endogenous auxin-and gibberellin-like substances. Significant amounts of an abscisin-likeinhibitor were found in extracts of flower buds. The mechanismof natural devernalization is discussed in relation to theseobservations.  相似文献
