建立稳定表达人RANTES基因的夏枯草细胞克隆   总被引:1,自引:0,他引:1  
为了在中药细胞中表达人源有用基因 ,以增强其特异药理活性 ,将克隆自人外周血淋巴细胞 (PBL)mRNA的RANTES基因经Ti质粒衍生的中间表达载体pROKII导入携带pAL44 0 4质粒的根癌农杆菌LBA44 0 4菌株中 ,并采用叶盘共培养法转化离体培养夏枯草细胞 ,经Southern杂交确认RANTES基因在转化细胞基因组中的整合 ,用RT -PCR扩增、Western印迹杂交和酶联免疫吸附测定 (ELISA)分析转化细胞中RANTES基因的表达 ,以PBL的过氧化物酶活性作为重组RANTES对细胞趋化性诱导的检测指标。结果表明 ,RANTES基因已在转基因夏枯草细胞中整合 ,并已形成稳定表达RANTES基因的细胞克隆 ,为进一步培育具有特异药理活性的转基因夏枯草植株打下了基础。  相似文献

Mitogenic potential of the partially purified lectin from P. vulgaris isolated by ammonium sulphate precipitation was assessed by lymphocyte transformation test (LTT) and was compared with three commercially available phytohemagglutinins (PHA). The blast inducing capacity and mitotic index analysis revealed that this preparation has potential to be used in the indigenous commercial production of PHA which is routinely used for human chromosomal studies from the peripheral blood culture at, at present, is imported.  相似文献

Human Tamm-Horsfall glycoprotein inhibits lymphocyte transformation induced by leucoagglutinin and haemagglutinin from Phaseolus vulgaris (red kidney bean). The glycoprotein interacts with the two lectins, giving insoluble precipitates. The interaction with leucoagglutinin is highly specific, and the shape of the precipitin curve is that of an antigen-antibody reaction; precipitation is specifically inhibited by N-acetyl-D-galactosamine. Results are discussed, and it is suggested that inhibition of lymphocyte transformation is due to competition between human Tamm-Horsfall glycoprotein and carbohydrate receptors on lymphocytes for the two lectins. The interaction between human Tamm-Horsfall glycoprotein and Phaseolus vulgaris lectins has been used to develop a one-step procedure for the separation of the two lectins by affinity chromatography on (human Tamm-Horsfall-glycoprotein)-Sepharose.  相似文献

The estuarine shrimp Palaemonetes pugio and P. vulgaris co-occur across a broad range in salinity. Field surveys and laboratory lethality tests suggest that low salinity conditions favor P. pugio over P. vulgaris. This study compared energetic expenditures across a range in salinity (0.5-35 ppt) to test the hypothesis that P. pugio experiences lower maintenance-energy requirements than P. vulgaris in low-salinity habitats. Standard metabolic rate (SMR) was measured as the minimum oxygen consumption rate at 25 degrees C to estimate daily maintenance-energy expenditure. SMR was greatest at 0.5 and least at 10 ppt (irrespective of species), and greater for P. vulgaris than for P. pugio. At low salinity (1.5-3 ppt), SMR was lower for P. pugio than for P. vulgaris. Energy expenditure did not coincide with the pattern predicted based upon osmoregulatory costs alone; no reduction was observed near the reported hemolymph concentration for P. vulgaris, although a minimum was observed near the hemolymph isoionic concentration for P. pugio. A local minimum in SMR by P. pugio between 1.5 and 3 ppt contrasted with relatively high SMR of P. vulgaris across this range. It appears that low-salinity habitats are energetically more favorable for P. pugio than for P. vulgaris. The results suggest an adaptive mechanism allowing P. pugio to occupy environments that present its competitor, P. vulgaris, with greater energetic challenges. While long-term exposure to very low salinity may be detrimental to both species, energetic traits of P. pugio may confer it with greater success than P. vulgaris in habitats in which salinity periodically fluctuates to low values.  相似文献

目的:探讨夏枯草胶囊对自身免疫性甲状腺炎(AITD)大鼠甲状腺组织病理学及Fas、FasL表达的影响。方法:将40只雌性SD大鼠按照随机数表法随机分为空白对照组、模型组、夏枯草胶囊组、硒酵母组,每组10只,空白对照组予以正常饲养,模型组和药物组予以PTg皮下注射建立AITD模型,夏枯草胶囊组、硒酵母组分别给予夏枯草胶囊与硒酵母片的生理水溶液灌胃,药物干预6周后处死大鼠,取甲状腺组织,HE染色电镜下观察各组大鼠甲状腺组织形态,免疫组化法检测各组大鼠甲状腺组织Fas、FasL蛋白的表达。结果:1)模型组大鼠甲状腺组织滤泡大量破坏,形态不规则,胶质分布不均匀,滤泡间隙增大,可见大量淋巴细胞及浆细胞浸润。夏枯草胶囊组滤泡形态尚规则,胶质含量较空白组少,滤泡间有少量淋巴细胞浸润。与模型组对比,夏枯草胶囊组与硒酵母组滤泡破坏及淋巴细胞浸润有显著改善。2)对照组大鼠甲状腺组织中Fas、FasL蛋白仅呈少量表达,模型组大鼠甲状腺组织中Fas、FasL蛋白表达较对照组显著增加(P0.01),夏枯草胶囊组大鼠甲状腺组织中Fas、FasL蛋白表达较模型组及硒酵母组均显著减少(P0.01)。结论:夏枯草胶囊可能通过减少Fas、FasL的表达,抑制甲状腺滤泡细胞凋亡,从而减轻甲状腺滤泡上皮的破坏。  相似文献

The present study was designed to evaluate the effects of Prunella Vulgaris (P. vulgaris) on the immune function in mice. The mice were randomly divided into one control group and three treatment groups of 10 mice each. The control group received pure water and the treatment groups received P. vulgaris extract at concentrations of 0.15, 0.30 and 0.90 g/kg BW orally for 30 days, respectively. Changes in cell immune function, non-specific immunity and humoral immunity function were evaluated. Active lymphocytes and T lymphocyte subsets were determined by fluorescence-activated cell sorting (FACS). Certain Serum concentrations of cytokines were measured by enzyme-linked immunosorbent assay (ELISA). The results showed that, for cell immune function, compared with the control group, foot pad thickness in high dose group increased significantly (p<0.01), whereas no significant difference in the proliferative ability of splenic lymphocytes was observed among all groups (p>0.05). For non-specific immunity, NK cell activity increased significantly in a dose-dependent manner in P. vulgaris treated mice (p<0.01), mononuclear-macrophage function in medium and high dose P. vulgaris treated mice were significantly higher than that of the control group (p<0.05). For humoral immunity, no significant differences were observed in terms of the half value of hemolysis (HC50), number of hemolytic plaques and serum IgG level (p>0.05). The percentage of active T and Th lymphocytes of mice peripheral blood in high dose group were significantly higher than that of the control group (p<0.01). There was no significant difference in serum levels of IL-1β, IL-4, IL-10 and IFN-γ among all of the four groups (p>0.05). The data indicated that 0.90 g/kg BW P. vulgaris extract (equivalent to 7.5 g/kg BW crude drug) had some effect on cellular immune function and non-specific immune function in mice.  相似文献

Eighty bacterial isolates from root nodules of the leguminous plants Phaseolus vulgaris, Campylotropis spp. and Cassia spp. grown in China were classified into five groups by phenotypic analyses, SDS-PAGE of whole-cell proteins, PCR-based 16S rRNA gene restriction-fragment-length-polymorphism and sequencing. Thirty-three isolates from the three plant genera were identified as Agrobacterium tumefaciens because they are closely related to the type strain of A. tumefaciens. Fourteen isolates from P. vulgaris grown in Yunnan and Inner Mongolia were classified as R. leguminosarum bv. phaseoli based on their close relationship with the type strain in numerical taxonomy and in 16S rDNA phylogeny. Twenty-seven isolates from Campylotropis delavayi, P. vulgaris and four species of Cassia grown in the central zones of China were classified into three groups within the genus Bradyrhizobium. One of these three groups could be defined as Bradyrhizobium japonicum. Our results demonstrated that P. vulgaris and the species of Campylotropis and Cassia could form nodules with diverse rhizobia in Chinese soils, including novel lineages associated with P. vulgaris. These results also offered information about the convergent evolution between rhizobia and legumes since the rhizobial populations associated with P. vulgaris in Chinese soils were completely different from those in Mexico, the original cite of this plant. Some rhizobial species could be found in all of the three leguminous genera.  相似文献

The gamma subunit of the human T lymphocyte T3 antigen is rapidly phosphorylated on serine residues in vivo during the initiation of T cell activation by a polyclonal mitogen (Phaseolus vulgaris phytohemagglutinin), an activator of protein kinase C (phorbol 12,13-dibutyrate), and an elevator of intracellular calcium (ionomycin). The sites of phosphorylation were identified by comparing tryptic peptide analyses of T3 gamma chains labeled in vivo with various synthetic peptides, corresponding to portions of the cytoplasmic domain of the gamma chain that had been labeled in vitro using purified protein kinase C. Two sites, serines 123 and 126, were phosphorylated in response to ionomycin, whereas a single site, serine 126, was phosphorylated when T lymphocytes were stimulated by P. vulgaris phytohemagglutinin or when protein kinase C was directly activated by phorbol 12,13-dibutyrate. Immune activation of T cells via the protein kinase C pathway thus induces phosphorylation of a single site on the T3 gamma chain, namely serine 126.  相似文献

Sets of primers for the species-specific detection of P. mirabilis and P. vulgaris by the polymerase chain reaction (PCR) were developed. As targets for these primers beta-lactamase and 16S rRNA gene fragments were chosen on the basis of the multiple leveling of the sequences of the DNA of all known P. mirabilis and P. vulgaris isolates. For differential detection oligonucleotides were selected in such a way that primers, specific for P. vulgaris, contained the non-paired nucleotide for P. mirabilis isolate at the 3'-end, and all other nucleotides were complementary to the beta-lactamase gene fragment. Primers, specific for gene 16S rRNA of P. mirabilis, contained the non-paired nucleotide for P. vulgaris isolates at the 3'-end. Standard PCR was carried out for 6 P. mirabilis and P. vulgaris strains. The use of PCR species-specific primers to P. vulgaris DNA made it possible to amplify the DNA fragment of the expected length only for P. vulgaris isolates, while the result of PCR for P. mirabilis was negative. PCR with primers specific to P. mirabilis permitted the detection of amplicon sized 101 nucleotides pairs only for P. mirabilis strains. These primers were optimized so as to use them in the specific differentiation of closely related P. mirabilis and P. vulgaris species by multiplex PCR. Genus-specific primers permitted the detection of bacterial gyrB gene of the genus Proteus were developed also.  相似文献

Lecithinase production is described as a new biochemical property of P. vulgaris strains grown in a selective agar medium containing brilliant green, crystal violet and lecithin (BCL agar), the authors' own modification of egg-yolk culture medium. By using this BCL agar as a medium inhibiting the swarming growth of P. vulgaris cultures the authors succeeded in identifying 12 lecithinase-positive strains among the P. vulgaris isolates obtained from patients with Crohn's disease. Of 50 P. mirabilis strains tested in parallel none gave the positive test for lecithinase production in this medium.  相似文献

The term 'dead-end trap cropping' has recently been proposed to identify a plant that is highly attractive for oviposition by an insect pest, but on which offspring of the pest cannot survive. The potential of the wild crucifer Barbarea vulgaris R. Br. to allure and serve as a dead-end trap crop for the diamondback moth Plutella xylostella (L.), an important pest of cruciferous crops worldwide, was examined in laboratory experiments. When P. xylostella adults were provided with a dual-choice of plants of B. vulgaris, and Chinese cabbage Brassica campestris (L.), in one arena, adult moths laid 2.5-6.8 times more eggs on the former than on the latter. When P. xylostella adults were provided with a dual-choice of plants of B. vulgaris and common cabbage Brassica oleracea L., adult moths laid virtually all their eggs on the former and ignored the latter. Nearly all P. xylostella eggs laid on the three species of plants hatched successfully, but nearly all individuals on plants of B. vulgaris died as neonates or early instar larvae, while 87-100% of the larvae on Chinese cabbage and common cabbage survived to pupation. Dual choice tests with a Y-tube olfactometer showed that volatiles from B. vulgaris were much more attractive to P. xylostella adults than those from common cabbage. The results demonstrate that B. vulgaris has a great potential as a dead-end trap crop for improving management of P. xylostella. Factors that may influence the feasibility of using B. vulgaris as a trap crop in the field are discussed, and ways to utilize this plant are proposed.  相似文献

The specificity and affinity of eight lectins (concanavalin A, L. culinaris, P. sativum, phytohemagglutinin P, D. biflorus, soybean agglutinin, T. purpureus, and T. vulgaris) to B, T, T gamma, and T mu lymphocytes from the blood of normal subjects were determined. Lectins attached to latex particles were used to evaluate the binding of each lectin to individual cells. The rosette percentage found in each lymphocyte population expresses the specificity index and the specific sugar concentrations needed to decrease the rosette percentage by 50% is taken as the affinity index. B Lymphocytes showed a major subclass, with respect to T lymphocytes, with receptors for WGA, SBA, D. biflorus, L. culinaris, and P. sativum lectins. In contrast, T lymphocytes exhibit a greater number of cells with specific receptors for Con A, T. purpureus, and PHA lectins than B lymphocytes, the T gamma subpopulation being responsible for the specificity of the first two lectins and the T mu subpopulation for the PHA lectin.  相似文献

A microbiological assay is described for determining gas produced by Clostridium perfringens (Veilon and Zuber) Holland from materials believed to be flatulent. A rationale for its use is given and analyzed. The fractionation and the results of the assay for several components of lima beans, Phaseolus lunatus L., and navy beans, P. vulgaris L., are given. In particular, a polysaccharide was isolated from dried P. vulgaris seeds. Only about one-seventh as much could be isolated from green P. lunatus seeds as from P. vulgaris seeds. The polysaccharide stimulates voluminous gas production by C. perfringens and meets all criteria of the rationale for flatulent sus pects. It has not been tested on humans; however, data from the microbiological assay correlate well with human results for green and dry lima beans and navy beans.  相似文献

This study focuses on lead accumulation in roots, stems and leaves of three plant species of the Fabacea family: Vicia faba, Pisum sativum and Phaseolus vulgaris grown hydroponically in a medium supplemented with 1 mM concentration of lead. The largest amount of lead, up to 75 mg Pb/g dry weight, was accumulated in roots of P. vulgaris. The highest rate of Pb ions uptake from the medium took place during the first 10 h of incubation with lead and after 96 h of incubation lead content in the medium decreased by half. Thus, it was suggested that P. vulgaris could be used in rhizofiltration--the use of plant roots to absorb pollutants from water contaminated with lead. At the same time we studied the influence of lead on acid soluble thiol, glutathione, homoglutathione contents and the synthesis of phyto- and homophytochelatins in roots of V. faba, P. sativum and P. vulgaris grown hydroponically. Activation of the detoxicative-phytochelatin system was observed in the cytosol of root cells of the tested plants. This system was composed of phytochelatins (PCs) in roots of V. faba, homophytochelatins (hPCs) in P. vulgaris roots and both PCs and hPCs in P. sativum roots. The total content of PCs and hPCs in roots of P. sativum was very high and reached around 4800 (expressed in nmol SH x g(-1)FW) and induction of their synthesis occurred after only 2 h of treatment with 1 mM Pb.  相似文献

从四川鲶科鱼类养殖场濒死的大口鲇(Silurus meridionalis Chen)体内分离到3株优势菌,人工感染试验表明菌株TWN3致病性最强。通过形态学观察、生理生化测定,结合DNA的G Cmol%、16S rDNA序列同源性和系统发育分析,确定TWN3为普通变形杆菌(Proteus vulgaris)。再根据生理生化特性,可进一步确定TWN3属于普通变形杆菌(Proteus vulgaris)中的P.vulgarisBG3。该菌株不溶血;对庆大霉素等4种药物敏感;对小鼠有较强的致病性。生长特性研究表明,该菌最适生长温度37℃、最适pH6.0、37℃时盐度1.5%生长最适。普通变形杆菌致病性广泛,但作为养殖大口鲇的致病菌国内外未见报道。另外,对该菌的分类学研究、生长特性、感染途径及防治进行了讨论。  相似文献

西花蓟马Frankliniella occidentalis(Pergande)是传入我国的重要入侵害虫,在贵阳地区对豆科蔬菜为害十分严重。在25℃条件下,组建了西花蓟马取食大豆叶片、豇豆叶片、四季豆叶片和四季豆豆荚时的实验种群生命表。结果表明:西花蓟马未成熟期取食四季豆豆荚时存活率最高,取食其它3种豆科蔬菜时存活率相差不大。西花蓟马取食四季豆豆荚时内禀增长力最高为0.1626,取食豇豆叶片时最低为0.0834,净增值率也是取食四季豆豆荚时最高,取食大豆叶片时最低,表明四季豆豆荚最有利于西花蓟马的生长发育和繁殖。在不同豆科蔬菜上的稳定年龄组配中未成熟期所占的比例很大,而成虫期所占的比例相对较小。  相似文献

The data on the study of the antibiotic response to 42 Proteus strains isolated from different sources in the hospitals of Kharkov are presented. The isolates belonged to P. mirabilis and P. vulgaris. Many strains were resistant to gentamicin, ampicillin and carbenicillin irrespective of the isolation source. 58.0 and 90.3 per cent of the strains isolated from patients with intestinal infections, 66.6 and 100 per cent of the strains isolated from patients with otitis, 33.3 and 66.6 per cent of the strains isolated from patients with bronchopulmonary affections and 100 and 100 per cent of the strains isolated from patients with urological diseases were resistant to gentamicin and carbenicillin, respectively. As for ampicillin, the respective figures were 74.2, 66.6, 66.6 and 100 per cent. All the strains of P. vulgaris isolated from patients with otitis, urological diseases and bronchopulmonary affections were resistant to ampicillin. The MIC of carbenicillin for all the strains except 4 indole-positive strains of P. vulgaris isolated from the faeces and bronchial excreta was much higher than the borderline values.  相似文献

The haemolytic activities of Proteus mirabilis and P. vulgaris strains were studied under different conditions. No filterable alpha haemolysin could be detected in P. mirabilis uropathogens provided from patients with urinary tract infections. Together with the results presented in the accompanying paper, in which three clinical isolates with temporary ability to produce a soluble haemolysin were described, the occurrence of alpha haemolytic P. mirabilis isolates did not exceed 3%. Cell bound beta haemolysin is present in nearly 35% of P. mirabilis urinary strains. Another kind of haemolytic activity was observed when P. mirabilis and P. vulgaris strains were grown in liquid media supplemented with erythrocytes. During the logarithmic growth phase nearly 100% of P. mirabilis and P. vulgaris strains of various origin haemolyzed 100-50% of erythrocytes. Except for Serratia, the other representatives of Enterobacteriaceae did not demonstrate such activity in the same conditions. The preliminary characteristics of this phenomenon is given.  相似文献

Dichogamy in five taxa of Pulsatilla (P. patens, P. pratensis, P. vernalis, P. vulgaris ssp. gotlandica and P. vulgaris ssp. vulgaris ) is incomplete with overlap between the female and male phases of a flower. The species are protogynous with the duration of the obligate female phase lasting 0 to 10 d and the duration of stigma receptivity of unpollinated flowers lasting 3.5 to 32 d in the greenhouse. The pollen presentation time varied between 1.5 and 10 d. There were significant differences among species in duration of both female and male phases. Hand pollination decreased the duration of stigmatic receptivity and pollen presentation significantly. The spatial separation of pollen presentation and receptive surfaces in a flower changed with time due to the continuing growth of the androecium and the gynoecium, and due to the temporal pattern of pollen presentation within the androecium. Hand pollination experiments indicate that seed set may be partly pollen-limited in a natural population of P. vulgaris ssp. vulgaris. The level of cross-pollination was high in this population as shown by high levels of pollen deposition on the stigmas during the obligate female phase.  相似文献
