Aedes aegypti, the main vector of dengue viruses in Asia, displays variation in population density over time. The larval habitats of this species being unevenly distributed and transient (depending on cycles of drought and flood), the forces generating temporal variation in gene frequencies in populations are studied. We sampled seven mosquito populations from Ho Chi Minh City (Vietnam) and its suburbs on five occasions between April 1999 and August 2000. We investigated genetic variation by studying isoenzyme and microsatellite polymorphism and susceptibility to a dengue 2 virus strain. Ae. aegypti populations collected during the dry season (January-April) showed genetic differentiation (F(ST) = 0.016, P < 10(-6) for isoenzymes) and showed more differentiated infection rates of the dengue 2 virus. The genetic structure of the population is less marked during the rainy season (F(ST) = 0.081, P < 10(-6)). Thus, environmental factors, such as rainfall and factors related to human activity, such as breeding site density and insecticide treatment, control the genetic structure of Ae. aegypti populations in the short term. The implications of studies of this kind for the design of future control programmes are discussed.  相似文献

Background

Aedes aegypti is a cosmopolite mosquito, vector of arboviruses. The worldwide studies of its insecticide resistance have demonstrated a strong loss of susceptibility to pyrethroids, the major class of insecticide used for vector control. French overseas territories such as French Guiana (South America), Guadeloupe islands (Lesser Antilles) as well as New Caledonia (Pacific Ocean), have encountered such resistance.

Methodology/Principal Findings

We initiated a research program on the pyrethroid resistance in French Guiana, Guadeloupe and New Caledonia. Aedes aegypti populations were tested for their deltamethrin resistance level then screened by an improved microarray developed to specifically study metabolic resistance mechanisms. Cytochrome P450 genes were implicated in conferring resistance. CYP6BB2, CYP6M11, CYP6N12, CYP9J9, CYP9J10 and CCE3 genes were upregulated in the resistant populations and were common to other populations at a regional scale. The implication of these genes in resistance phenomenon is therefore strongly suggested. Other genes from detoxification pathways were also differentially regulated. Screening for target site mutations on the voltage-gated sodium channel gene demonstrated the presence of I1016 and C1534.

Conclusion /significance

This study highlighted the presence of a common set of differentially up-regulated detoxifying genes, mainly cytochrome P450 genes in all three populations. GUA and GUY populations shared a higher number of those genes compared to CAL. Two kdr mutations well known to be associated to pyrethroid resistance were also detected in those two populations but not in CAL. Different selective pressures and genetic backgrounds can explain such differences. These results are also compared with those obtained from other parts of the world and are discussed in the context of integrative research on vector competence.  相似文献

Field survival of Aedes aegypti females is a key parameter for estimating the dengue transmission potential of a mosquito population. The objectives of this study were to explore the dynamics of these survival rates at different times of the year in French Guiana and to analyze the results from the perspective of dengue patterns. The mosquitoes were captured, marked, released, and recaptured during four consecutive days in six houses every month, for three to 24 months, from January 1997 to December 1998. Laboratory experiments showed no effects on female survival but some effect on the survival of males. Females' daily survival in the field varied from 0.525 to 1 but was mostly between 0.8 and 0.95 during the entire year, with a mean value of 0.913. The field survival of Ae. aegypti females in French Guiana was thus in agreement with the likely transmission of dengue and the dengue endemic patterns throughout the year. On the other hand, heavy rainfalls during this time were less favorable to Ae. aegypti survival, which may explain part of the El Ni?o effect on dengue epidemics in French Guiana. The methods and results on Ae. aegypti survival will be implemented in a global dengue surveillance network in French Guiana.  相似文献

Geographically disjunct populations are unusual in marine species, but the Houtman Abrolhos Islands, Western Australia, provide opportunities to study highly disjunct peripheral isolates of several species. The intertidal snail Batillariella estuarina occurs in isolated tidal ponds in the Abrolhos Islands, where it is at its northern limit, disjunct from mainland populations by 600-900 km. The species is thus disjunct both geographically and among the peripherally isolated populations in the Abrolhos Islands. Comparisons of allozymes at 11 polymorphic loci were made among populations from 10 ponds in the Abrolhos Islands and six sites from relatively continuous tidal flats at Albany, 900 km away, the nearest major set of populations. Among all 16 populations, subdivision was high (FST = 0.455). Although there were subtle differences between the geographical regions, the large majority of divergence occurred among the isolated ponds in the Abrolhos (FST = 0.441), and divergence on the tidal flats at Albany was only moderate (FST = 0.085). Characteristic of peripheral isolates, the pond populations have less polymorphism and fewer alleles than the more connected populations at Albany. Combined with evidence of genetic divergence in the gastropods Bembicium vittatum and Austrocochlea constricta, which have very similar geographical distributions to that of B. estuarina, these results indicate the potential evolutionary significance of peripherally isolated marine populations in the unusual habitats of the Abrolhos Islands.  相似文献

In French Guiana, pyrethroids and organophosphates have been used for many years against Aedes aegypti. We aimed to establish both the resistance level of Ae. aegypti and the ultra low volume spray efficacy to provide mosquito control services with practical information to implement vector control and resistance management. Resistance to deltamethrin and fenitrothion was observed. In addition, the profound loss of efficacy of AquaK'othrine? and the moderate loss of efficacy of Paluthion? 500 were recorded. Fenitrothion remained the most effective candidate for spatial application in French Guiana until its removal in December 2010. Further investigation of the mechanism of resistance to deltamethrin demonstrated the involvement of mixed-function oxidases and, to a lesser extent, of carboxylesterases. However, these observations alone cannot explain the level of insecticide resistance we observed during tube and cage tests.  相似文献

FST was estimated for 67 communities and 6 small towns of Kostroma province. The mean FST value for rural and urban populations was (0.83 +/- 0.08).10(-3) and (0.29 +/- 0.07).10(-3), respectively. The connection between FST values and the load of autosomal-recessive disorders was established; the coefficient of correlation (by Chuprov) was 0.34 (chi 2 = 8.45; P less than 0.05). The matrices of genetic distances for two groups of districts of Kostroma province, based on surnames frequencies, were calculated. Dendrogramms were constructed using genetic distances, which reflect the degree of genetical similarity of the populations. The conclusion drawn from the analysis of these dendrogramms is that there is distinct isolation by distance in populations of Kostroma province. It is shown that genetical subdivision of populations is dependent on geographical and some other factors and on the load of autosomal-recessive pathology in the population.  相似文献

Recent studies of Anopheles gambiae, the principal mosquito vector of malaria in Africa, suggested that the eastern Rift Valley and its surrounding areas act as a barrier to gene flow. To quantify the unique effect of these areas on gene flow, we measured genetic variation within and between populations from each side of the Rift. Low differentiation was measured between populations on each side of the Rift (mean FST < 0.008, mean RST < 0.002). However, high differentiation was measured across the Rift (mean FST = 0.104; mean RST = 0.032). Genetic diversity within populations was lower in eastern populations, suggesting that the effective population sizes (Ne) of these populations were lower than those of western populations. We partitioned the overall differentiation across the Rift into three factors: variation in Ne between populations contributed 7-20%; distance contributed 10-30%, and the remainder, corresponding to the unique effect of the Rift was 50-80%. The Rift's effect was highly significant based on FST. The greater sensitivity of FST in measuring differentiation indicated that drift and not mutation generated the differences between populations. Restricted gene exchange across several hundred kilometers on the face of intense human transportation implies that active mosquito dispersal is the major form of migration, and that migration is a multistep process, where step length is relatively short.  相似文献

We assessed the effects of long-term habitat fragmentation on genetic (random amplified polymorphic DNA) diversity in 11 Peromyscus maniculatus populations in the Lake Superior watershed. We analysed genetic structure at two spatial scales and the effect of island size and isolation on genetic diversity. At the regional scale, island populations differed from mainland populations (FST = 0.36), but mainland populations did not differ from each other (FST = 0.01). At the local scale, populations of the main island of Isle Royale differed from adjacent islet populations (P < 0.001; Monte Carlo approximation of Fisher's exact test), but not from each other (combined P = 0.63). Although geographical distance and genetic distance were positively correlated (P < 0.01; Mantel test), cluster analysis revealed some inconsistencies. Finally, genetic diversity was inversely related to isolation (P = 0.01), but had an unexpectedly negative relationship with island area (P = 0.03). The genetic structure of P. maniculatus populations in portions of the Lake Superior watershed appears to have been affected by long-term habitat fragmentation.  相似文献

Using our results and literature data on multilocus DNA fingerprinting, we propose a method of obtaining unbiased estimates of the between--population genetic similarity index and a measure of population subdivision based on modified Wright's FST-statistics. On the basis of multiple comparison T2 Hotelling's test and Holmes' procedure, the FST-statistics was applied to assess differentiation of four (Pacific and Atlantic) subpopulations of humpback whale Megaptera novaeangliae, six populations of California island gray fox Urocyon littoralis, and geographically isolated Ob' and Yakutia populations of Siberian white crane Crus leucogeranus. It was shown that the regional humpback whale subpopulations do not constitute a single panmictic unit (P < 10(-4)). The subdivision index of the Pacific and Atlantic populations expressed in terms of FST-statistics varied from 0.101 to 0.157. The differentiation estimates for the island fox populations, which ranged from 0.2109 to 0.4027, indicate that subdivision of these populations is a function of the distance between the islands, island size, and population size. In particular, the smallest and the greatest differences were found respectively between the populations of the geographically closest northern islands (FST = 0.2157, FST = 0.2109) and between those of the most distant northern and southern islands (FST = 0.4027, FST = 0.3869). Subdivision of the island populations with minimum areas and low population number was intermediate (FST = 0.3789). Mean values of heterozygosity, within-population genetic similarity index, and the number of coinciding fragments for two random individuals of Siberian white crane from the Ob' and Yakutia population were not statistically significantly different (P > or = 0.852, (P > or = 0.491, (P > or = 0.325). However, pairwise comparisons of mean FST values indicated that the differentiation estimates for samples from these populations fall within the limits of population subdivision (P = 0.01). The subdivision estimate (0.108-0.133) of various groups of Siberian white cranes is comparable to interregional subdivision of humpback whale. Based on the results of this study, we recommend the approach based on modified Wright's FST-statistics for studying genetic population structure aimed at detecting population subdivision.  相似文献

In recent years, the development of new tools to gather field information about vector ecological parameters has increased. This report evaluated the BG-Sentinel Trap (BGS-Trap), a promising new attempt to improve collection of the dengue vector, Aedes aegypti. The efficacy of the BGS-Trap was compared with the CDC backpack aspirator, one of the commonest used methods for capturing adult mosquitoes. BGS-Traps captured significantly more Ae. aegypti males (chi2 = 21.774, df = 1, P < 0.05) and females (chi2 = 56.007, df = 1, P < 0.05) than CDC aspirator during all days of field collection. However, CDC aspirator was significantly more efficient to capture Culex quinquefasciatus males (chi2 = 5.681, df = 1, P < 0.05) and females (chi2 = 6.553, df = 1, P < 0.05). BGS-Traps captured host-seeking females (varying between 68.75 to 89.8%) in detriment of females in other behavioral and physiological stages. BGS-Traps proved to be efficient and can be used for monitoring adult mosquito populations.  相似文献

We assayed mtDNA haplotype [300 base pairs (bp) control region] geography and genealogy in the Indo-Pacific tasselfish, Polynemus sheridani from its contiguous estuarine distribution across northern Australia (n = 169). Eight estuaries were sampled from three oceanographic regions (Timor Sea, Gulf of Carpentaria and the Coral Sea) to assess the impact of Pleistocene sea level changes on the historical connectivity among P. sheridani populations. Specifically, we investigated the genetic consequences of disruption to Indian-Pacific Ocean connectivity brought about by the closure of the Torres Strait. Overall there was significant population subdivision among estuaries (FST = 0.161, PhiST = 0.187). Despite a linear distribution, P. sheridani did not show isolation by distance over the entire sampled range because of genetic similarity of estuaries greater than 3000 km apart. However, significant isolation by distance was detected between estuaries separated by less than 3000 km of coastline. Unlike many genetic studies of Indo-Pacific marine species, there was no evidence for an historical division between eastern and western populations. Instead, phylogeographical patterns were dominated by a starlike intraspecific phylogeny coupled with evidence for population expansion in both the Gulf of Carpentaria and the Coral Sea but not the Timor Sea. This was interpreted as evidence for recent west to east recolonization across of northern Australia following the last postglacial marine advance. We argue that although sufficient time has elapsed postcolonization for populations to approach gene flow-drift equilibrium over smaller spatial scales (< 3000 km), the signal of historical colonization persists to obscure the expected equilibrium pattern of isolation by distance over large spatial scales (> 3000 km).  相似文献

The amphibian chytrid fungus Batrachochytrium dendrobatidis (Bd) is a purported agent of decline and extinction of many amphibian populations worldwide. Its occurrence remains poorly documented in many tropical regions, including the Guiana Shield, despite the area’s high amphibian diversity. We conducted a comprehensive assessment of Bd in French Guiana in order to (1) determine its geographical distribution, (2) test variation of Bd prevalence among species in French Guiana and compare it to earlier reported values in other South American anuran species (http://www.bd-maps.net; 123 species from 15 genera) to define sentinel species for future work, (3) track changes in prevalence through time and (4) determine if Bd presence had a negative effect on one selected species. We tested the presence of Bd in 14 species at 11 sites for a total of 1053 samples (306 in 2009 and 747 in 2012). At least one Bd-positive individual was found at eight out of 11 sites, suggesting a wide distribution of Bd in French Guiana. The pathogen was not uniformly distributed among the studied amphibian hosts, with Dendrobatidae species displaying the highest prevalence (12.4%) as compared to Bufonidae (2.6 %) and Hylidae (1.5%). In contrast to earlier reported values, we found highest prevalence for three Dendrobatidae species and two of them displayed an increase in Bd prevalence from 2009 to 2012. Those three species might be the sentinel species of choice for French Guiana. For Dendrobates tinctorius, of key conservation value in the Guiana Shield, smaller female individuals were more likely to be infected, suggesting either that frogs can outgrow their chytrid infections or that the disease induces developmental stress limiting growth. Generally, our study supports the idea that Bd is more widespread than previously thought and occurs at remote places in the lowland forest of the Guiana shield.  相似文献

The genetic population structure of Polyommatus coridon (Poda 1761) over large regions of France, Italy and Germany was studied by allozyme electrophoresis. The genetic diversity within populations was high for all parameters analysed (number of alleles 2.72; observed and expected heterozygosity 19.6% and 20.3%, respectively; percentage of polymorphic loci: total: 76.4% and, with polymorphism if the frequency of the commonest allele is below 95%: 53.1%), whereas genetic differentiation between populations was comparatively low (FST = 0.021 +/- 0.002). The mean number of alleles declined significantly from southern to northern populations (r = -0.53, P = 0.0005). Similar effects were found also for other parameters of genetic diversity. This is interpreted as a loss of genetic diversity during postglacial expansion. However, samples from France and Italy had similar patterns of genetic diversity indicating no significant loss in this region. Populations from southern Germany were genetically uniform, well differentiated from French populations and showed a significant loss of genetic diversity. Probably, this is due to a bottleneck during passing through the Burgundian Gap, which is a migration corridor from north-eastern France to southern Germany. In contrast to southern German populations, western German populations were not well differentiated from French populations. Nevertheless, they were genetically impoverished, probably as a result from local bottlenecks and post-expansion phenomena.  相似文献

Methods of estimating within- and between-population gene diversity in parthenogenetic species using mini- and microsatellite DNA markers and modified Wright's FST statistic are presented with special reference to model populations of lizards of the genus Darevskia (D. dahli, D. armeniaca, D. unisexualis). We used DNA fingerprinting data for several populations of these species examined earlier. The effects of variation in M13, minisatellite, (GACA)n and (TCC)n microsatellite loci on the formation of within-population gene diversity in parthenogenetic species D. dahli and D. armeniaca were shown to be different. The equality of the realized gene diversity H and its maximum possible value Hmax in two populations of D. dahli (Hmax = 0.032, H = 0.031, P < < 0.0431; Hmax = 0.024, H = 0.027, P = 0.09) and D. armeniaca (Hmax = 0.05, H = 0.053, P = 0.03; Hmax = 0.054, H = 0.055, P = 0.02) suggests that variation in (GACA)n loci substantially contributes to the maintenance of within-population genetic diversity. Analysis of between-population genetic diversity using loci M13, (GACA)n, and (TCC)n showed differentiation of D. dahli populations from northeastern and northwestern Armenia (FST = 0.0272, P = 3 x 10(-13)) and genetic homogeneity of the Armenian and Introduced to the Ukraine populations of D. armeniaca characteristic of one clone (FST = 0, P = 1).  相似文献

In a climate of growing concern that Plasmodium falciparum may be developing a drug resistance to artemisinin derivatives in the Guiana Shield, this review details our current knowledge of malaria and control strategy in one part of the Shield, French Guiana. Local epidemiology, test-treat-track strategy, the state of parasite drug resistance and vector control measures are summarised. Current issues in terms of mobile populations and legislative limitations are also discussed.  相似文献

Many tropical regions lack models predicting the biological and environmental conditions expected in any given area, thus precluding the implementation of reference condition-based water policies. We focused on streams of French Guiana, and tested two predictions: geomorphology determines ecological sub-regions that have typical invertebrate communities, and diversity declines as anthropogenic pressure increases. Sixty-five stream sites were sampled for benthic invertebrates and physical–chemical variables across various watersheds. We used the Self-Organizing Map algorithm (neural network) to model relationships between invertebrate communities and environmental variables. Sites characterized by invertebrate communities clustered into two major subsets matching French Guiana’s hydro-ecoregions: the coastal alluvial plain characterized by recent sediment and low elevations, and the Guiana Shield characterized by an eroded rocky substrate and dense rainforests. Changes in community composition, and to a lesser extent taxonomic richness within each sub-region revealed ecological impacts of gold mining and logging, further clustering hydro-ecoregions into subsets of reference and impaired sites. Further analyses would, however, be needed to identify tipping points between natural and disturbed states, especially in remote headwater streams where gold mining had the harsher impact upon freshwater diversity, making upstream communities resembling the most downstream impacted ones.  相似文献

A genetic study was carried out on a neotropical fish species to illustrate the refuge theory, whose main principles are summarized. The geographical structure of the anostomid species Leporinus friderici was analysed in French Guiana and Brazil by horizontal starch gel electrophoresis. Fifteen enzymatic systems corresponding to 21 loci were studied, revealing the following particularities: (1) specimens of fish from six independent coastal rivers of French Guiana form two groups geographically situated on either side of the Kourou River; (2) two alleles can serve as geographic markers, Ldh2 ( l30 ) showing an affinity between the populations in eastern French Guiana and Brazil, and Me 1 ( 300 ) being specific to the west.
These observations are probably related to the refuges that were formed in the Quaternary in South America, especially on the Guiana Shield. We consider that the populations of French Guiana are separated into two groups at the boundary between the two migratory flows, one arising in the west from the refuge of the Guiana Shield and the other in the east from a nearby undetermined Amazonian refuge.  相似文献
