The effect of different temperatures on the shoot and root formation in isolated petiole segments of Begonia × cheimantha was determined after 10 weeks on a modified White medium containing 0.1 mg/1 NAA and 0.5 mg/1 BA. Temperature proved to be important for the induction of shoot and root formation. At a constant temperature the best plants were obtained at 18 to21°C. If the temperature was higher, fewer cultures survived and the number of roots and shoots were lower. Lower temperatures inhibited the development of plants. A pretreatment at 15 or 18°C for two to four weeks improved the number and size of the shoots developed during a following 24°C treatment. High temperatures throughout the growing period reduced the number of shoots severely. A pretreatment of three days at 24°C or one day at 28°C reduced the shoot number by 50 %. After seven days at 28°C there was not a single shoot in any of the cultures. However, after two weeks at 15 or 18°C it was no longer possible to inhibit the shoot formation by a 24°C treatment. It is concluded that the formation of shoots in petiole segments takes place during the first two weeks after excision, and that high temperature is detrimental to the shoot initiation process.  相似文献

Root segments taken from aseptically-grown seedlings of Brassicaoleracea var. italica cv. Green Comet were used in an investigationof factors affecting in vitro regeneration. Shoot regenerationwas found to increase with seedling age and to be highest inroot segments adjacent to the hypocotyl and lowest in segmentsadjacent to the root tip. In a comparison of a range of mediaand agar concentrations shoot formation was favoured by complexmedia containing reduced nitrogen and was higher on gelled mediathan in liquid medium. The effects of various cytokinins andauxins were investigated; KN was the best cytokinin and IAAand Picloram the best auxins for shoot induction. Root segmentsfrom six other Brassica cultivars were grown on the medium devisedfor Green Comet; shoots were regenerated from two B. oleraceacultivars and two B. napus cultivars, but not from the B. campestriscultivars tested. Lateral shoot tips from young Asparagus setaceus (Kunth) Jessop (syn. A. plumosus Baker) shoots were grown on a modified Murashige and Skoog medium. Tips from 5 to 20 mm lateral shoots had significantly better growth and development than tips from lateral shoots (2 mm) still covered by leaf-scale. The optimum temperature for growth and development of the explants was 17 to 24°C. The initial growth was fast at 24°C but stopped after about 4 weeks. At 17°C the growth was slow but in return the cultures continued to grow. Kinetin was necessary for growth. Without any kinetin all cultures died. Optimum growth was found with 2 mg/l kinetin. There was no growth at all with IAA alone. A low IAA concentration had no effect, but at high concentrations IAA inhibited the kinetin induced growth.  相似文献

Lateral shoot tips from young shoots of Asparagus setaceus (Kunth) Jessop (syn. A. plumosus Baker) were grown on a modified Murashige and Skoog medium. PBA was the most effective cytokinin for growth and development of Asparagus plumosus shoot tips. The optimal range of PBA was between 0.02 and 2 mg/1. Zeatin was less effective, and kinetin, BA and 2iP were poor as cytokinins for A. plumosus. The optimal temperature for growth and development was 21°C. There was interaction between the temperature and the PBA concentration. With increasing temperature from 17 to 24°C the need for PBA increased from 0.2 to 2 mg/1.  相似文献

Hypocotyl sections and isolated meristem-tips from 7- to 10-day-old seedlings of flax were cultured on a Murashige and Skoog (1962) medium. The optimum concentrations of naphthaleneacetic acid (NAA) and benzyladenine (BA) were determined for shoot and root initiation from hypocotyls. Meristem-tip cultures differentiated shoots and roots in response to these concentrations. The frequency and yield of shoots, promoted by BA, decreased if NAA or gibberellic acid (GA3) was included in the medium. At high NAA concentration GA3 and BA inhibited root initiation. Morphological development of cultures differed depending on the growth regulators in the medium.  相似文献

White clover (Trifolium repens L.) was grown in controlled environmentsto determine the distinct effects of root and shoot temperatureon the accumulation of total and fixed (15 N dilution) nitrogenat two levels of nitrate (10 and 75 mM). Nitrogen fixation(BNF) showed a positive response to higher shoot temperature(23 vs. 13 C day temperature), irrespective of whether or notroot temperature was increased in parallel. Low root temperature(5 C) caused a marked reduction in the accumulation of totalnitrogen at both nitrate levels, and led to a lower proportionof N derived from BNF. The temperature response of BNF was attributedfor the major part to an adaptation to the demand for fixedN. It is therefore concluded that BNF is not primarily responsiblefor the reduced clover growth at low temperatures. Abstract: Winter wheat plants ( Triticum aestivum L. cv. Urban), grown in nutrient solution, were exposed to differential shoot/root temperatures (i.e., 4/4, 4/20, 20/4 and 20/20 C) for six weeks. Leaves grown at 4C showed an increase in frost tolerance from - 4C down to - 11 C, irrespective of root temperature. In 4/20 C plants, high root temperature decreased the rate of hardening of the leaves, but did not influence the final level of frost tolerance. In roots grown at 4C frost tolerance increased from - 3 C down to - 4 C, independently of shoot temperature. An accumulation of soluble sugars in the leaves was only observed when both shoot and root were grown at 4C and was not correlated with final frost tolerance achieved. However, the rate of hardening was correlated with the soluble sugar concentration. An increase in root soluble sugar concentration was exclusively observed in roots exposed to a temperature of 4C, irrespective of shoot temperature. Proline concentration only increased in plant parts exposed to a temperature of 4C. The present results indicate that the importance of root temperature in low-temperature hardening of winter wheat is limited, even though exposure to differential root and shoot temperatures brought about pronounced changes in growth, soluble sugar concentration, insoluble sugar concentration and proline concentration in roots and leaves.  相似文献

The growth of three populations of greater lotus (Lotus uliginosusSchkuhrsyn.L. pedunculatusCav.) was compared at photoperiods of 10,12 and 14 h at a maximum day/minimum night temperature of 21/16°C and at maximum day/minimum night temperatures of 27/22,21/16, 18/13 and 15/10 °C at a photoperiod of 12 h. Shortdays (10 h) favoured root and rhizome development compared tolong days (14 h). A temperature regime of 15/10 °C restrictedrhizome development compared to the 18/13 and 21/16 °C regimes.Shoot growth was restricted at the highest temperature regime(27/22 °C). The cultivar Sharnae had fewer, but heavier,rhizomes than Grasslands Maku; this may indicate adaptationto the dry summers at its site of origin (Algarve, Portugal).The response of rhizome growth to temperature and photoperiodexplains part of the performance of greater lotus in the fieldat a wide range of latitudes. Grazing management to encouragethe persistence ofL. uliginosusin pasture in temperate environmentsmay include the exclusion of grazing livestock in autumn. The effect of temperature on respiration of mitochondria and tissue segments from three wheat (Triticum aestivum L.) and one rye (Secale cereale L.) cultivar grown at 2 and 24 C has been examined. Discontinuities in Arrhenius plots of respiratory activity against temperature were observed for mitochondria and tissue segments from seedlings grown at both temperatures. The rates of respiration decreased abruptly below the transition temperatures, resulting in increased energy of activation values for respiration. Transition temperatures were observed from 6 to 10 C during tissue segment respiration, and from 10 to 14 C during respiration by isolated mitochondria. Respiratory control and efficiency of phosphorylation were not affected markedly by either reaction temperature or growth temperature of the seedlings. No correlation was observed between the cold hardiness of the cultivars and the temperature at which structural transitions occurred in the mitochondria. Dry matter content of the seedlings increased markedly during growth at 2 C, but no appreciable changes in the levels of mitochondrial protein were observed. The results support the view that changes other than fatty acid unsaturation are involved in the abrupt change in mitochondrial membrane properties at low temperature.  相似文献

Established red maple (Acer rubrum L.) callus was cultured on media varying in auxin (NAA or 2,4-D) and cytokinin (BA) concentrations. Callus growth was positively affected by the presence of both an auxin and cytokinin in the medium. Two grape cultivars, susceptible French Colombard and tolerant Rubired, and four nematodes, Meloidogyne incognita, Pratylenchus vulnus, Tylenchulus semipenetrans, and Xiphinema index, were used to quantify the equilibrium between root (R) and shoot (S) growth. Root and shoot growth of French Colombard was retarded by M. incognita, P. vulnus, and X. index but not by T. semipenetrans. Although the root growth of Rubired was limited by all the nematodes, the shoot growth was limited only by X. index. The R:S ratios of Rubired were higher than those of French Colombard. The reduced R:S ratios of Rubired were primarily an expression of reduction in root systems without an equal reduction in shoot growth, whereas in French Colombard the reduced R:S ratios were due to a reduction in both shoot growth and root growth and to a greater reduction in root growth than shoot growth. All nematodes reproduced equally well on both cultivars. Both foliage and root growth of French Colombard were significantly reduced by M. incognita and P. vulnus. Nematodes reduced the shoot length by reducing the internode length. Accumulative R:S ratios in inoculated plants were significantly smaller than those in controls in all nematode treatments but not at individual harvest dates. Bud break was delayed by X. index and was initiated earlier by P. vulnus and M. incognita. All buds in nematode treatments were less vigorous than in controls.  相似文献

The elongation of the internodes and petioles of Vicia fabahas been studied in the plumules of normal seedlings, in seedlingsfrom which part or all of the cotyledonary reserve has beenremoved, and in axillary shoots developed on seedlings of variousages. The curves obtained by plotting the lengths of the internodesagainst their plastochrone ages reveal constant differencesin behaviour between plumules and axillary branches. As thegrowth of any shoot progresses a greater contribution to thetotal elongation rate cornea to be made by the internodes fartherremoved from the apex. The length of each internode depends throughout its developmenton its position in the stem as well as on the length of theinternode below. The extent to which the elongation of an internodelags behind that of the one below increases upwards, a relationshipsimilar to that previously shown to exist in respect of thevascularization of the internodes. In plumules the length of petiole corresponding to any givenlength of internode undergoes a decrease followed by an increaseas the plumule is traversed from base to apex, a relationshipexactly oppsite to that previously demonstrated between thevascularization of a leaf-trace bundle and that of the internodein which it runs. The effects of age and of removal of cotyledonary reserves aredescribed, and it is concluded that it is unlikely that thecharacteristic difference in internode length within the limitsof a single shoot are wholly due to changes in the availabiltyof reserve substances.  相似文献

Maize (Zea mays L.) and spring wheat (Triticum aestivum L.)were grown in nutrient solution at uniformly high air temperature(20 °C), but different root zone temperatures (RZT 20, 16,12 °C). To manipulate the ratio of shoot activity to rootactivity, the plants were grown with their shoot base includingthe apical meristem either above (i.e. at 20 °C) or withinthe nutrient solution (i.e. at 20, 16 or 12 °C). In wheat, the ratio of shoot:root dry matter partitioning decreasedat low RZT, whereas the opposite was true for maize. In bothspecies, dry matter partitioning to the shoot was one-sidedlyincreased when the shoot base temperature, and thus shoot activity,were increased at low RZT. The concentrations of non-structuralcarbohydrates (NSC) in the shoots and roots were higher at lowin comparison to high RZT in both species, irrespective of theshoot base temperature. The concentrations of nitrogen (N) inthe shoot and root fresh matter also increased at low RZT withthe exception of maize grown at 12 °C RZT and 20 °Cshoot base temperature. The ratio of NSC:N was increased inboth species at low RZT. However this ratio was negatively correlatedwith the ratio of shoot:root dry matter partitioning in wheat,but positively correlated in maize. It is suggested that dry matter partitioning between shoot androots at low RZT is not causally related to the internal nitrogenor carbohydrate status of the plants. Twelve pasture species were grown in the same aerial environment,but with five constant soil temperatures ranging from 5 to 35°C, to determine the influence of root temperature on theweight of roots per unit weight of foliage (R/S ratio). Thisratio varied by a factor of 2 to 8 within species. Using maximum yield of foliage to indicate the optimum soiltemperature for each species, it was found that the R/S ratiowas lowest at the optimum soil temperature, and was progressivelyhigher at soil temperatures above and below the optimum withonly slight exceptions. Soil application of CCC reduced stem and leaf growth in Begonia plants. This effect was evident with all concentrations tested at 18°C, whereas at 21 and 24°C no growth–retarding effect was observed with 2 × 10?2 M CCC, and with 5 × 10?3 M growth was even stimulated. Flowering was promoted by CCC in long day and neur–critical temperature, particularly under low light intensity in the winter. The formation of adventitious buds in leaves of plants grown at 21 and 24°C was stimulated when the plants received 5 × 10?2 and 2 × 10?2 M CCC, while 8 7times; 10?2 M was inhibitory. In plants grown at 18°C bud formation was inhibited by all CCC concentrations. Root formation in the the leaves was usually stimulated by high CCC concentrations, while root elongation was reduced. The level of ether–extractable. acidic auxin (presumably IAA) in the leaves was lowered by CCC treatment of the plants, hut this required higher CCC concentrations at higt than at low temperature. When applied to detached leaves CCC stimulated bud formation at concentrations ranging from 10?4 to 10?2 M in leaves planted at 18 and 21°C. At 24°C budding was inhibited by 10?2 M CCC, the lower concentrations being stimulatory also at this temperature. Root formation and growth were not much affected by CCC treatment of the leaves, but increased with the temperature. Soil application of Phosfon (4 × 10?4 M) had no effect on growth and flowering, nov did it affect the subsequent regeneration of buds and roots in the leaves. In detached leaves Phosfon stimulated bud formation with au optimum at 10?6 M. Root formation was stimulated by Phosfon at all temperatures, the optimal concentration being 10?5 M, whereas root length was conversely affected. Foliar application of B-995 to intact plants and treatment of detached leaves greatly inhibited the formation of buds and had little effect on root formation. B-99D reduced the growth and delayed flowering in the plants.  相似文献

In roots two distinct polar movements of auxin have been reported that may control different developmental and growth events. To test the hypothesis that auxin derived from the shoot and transported toward the root controls lateral root development, the two polarities of auxin transport were uncoupled in Arabidopsis. Local application of the auxin-transport inhibitor naphthylphthalamic acid (NPA) at the root-shoot junction decreased the number and density of lateral roots and reduced the free indoleacetic acid (IAA) levels in the root and [3H]IAA transport into the root. Application of NPA to the basal half of or at several positions along the root only reduced lateral root density in regions that were in contact with NPA or in regions apical to the site of application. Lateral root development was restored by application of IAA apical to NPA application. Lateral root development in Arabidopsis roots was also inhibited by excision of the shoot or dark growth and this inhibition was reversible by IAA. Together, these results are consistent with auxin transport from the shoot into the root controlling lateral root development.  相似文献

A micro-growth measuring technique was used to determine the growth response of stem segments of Avena sativa cv. Avon to a variety of gibberellic acid (GA3) concentrations over a range of incubation temperatures. Growth rate varied with GA3 concentration, the temperature at which the rate was measured, and the growth temperature of the plants prior to excision of the segments. The curves relating segment extension rates to temperature were affected by GA3 such that the linear portion of the curve was shifted to higher rates as GA3 concentration was increased. The results seem to be analogous to the GA3-induced shifts of thermally induced phase transitions in glucose leakage from liposomes, observed earlier (Wood, Paleg 1974 Aust J Plant Physiol 1: 31-40).  相似文献
