Cd accumulation in roots and shoots of durum wheat: the roles of transpiration rate and apoplastic bypass 总被引:3,自引:0,他引:3
Cadmium is readily taken up from soils by plants, depending on soil chemistry, and variably among species and cultivars; altered transpiration and xylem transport and/or translocation in the phloem could cause this variation in Cd accumulation, some degree of which is heritable. Using Triticum turgidum var. durum cvs Kyle and Arcola (high and low grain Cd accumulating, respectively), the objectives of this study were to determine if low-concentration Cd exposure alters transpiration, to relate transpiration to accumulation of Cd in roots and shoots at several life stages, and to evaluate the role of apoplastic bypass in the accumulation of Cd in shoots. The low abundance isotope (106)Cd was used to probe Cd translocation in plants which had been exposed to elemental Cd or were Cd-na?ve; apoplastic bypass was monitored using the fluorescent dye PTS (8-hydroxy-1,3,6-pyrenetrisulphonate). Differential accumulation of Cd by 'Kyle' and 'Arcola' could be partially attributed to the effect of Cd on transpiration, as exposure to low concentrations of Cd increased mass flow and concomitant Cd accumulation in 'Kyle'. Distinct from this, exposure of 'Arcola' to low concentrations of Cd reduced translocation of Cd from roots to shoots relative to root accumulation of Cd. It is possible, but not tested here, that sequestration mechanisms (such as phytochelatin production, as demonstrated by others) are the genetically controlled difference between these two cultivars that results in differential Cd accumulation. These results also suggest that apoplastic bypass was not a major pathway of Cd transport from the root to the shoot in these plants, and that most of the shoot Cd resulted from uptake into the stele of the root via the symplastic pathway. 相似文献

Cadmium (Cd) accumulation has been found to vary between cultivars of durum wheat (Triticum turgidum var. durum), and it is hypothesized that low-molecular-weight organic acids (LMWOAs) produced at the soil-root interface (rhizosphere) may play an important role in the availability and uptake of Cd by these plants. The objective of this study, therefore, was to (1) investigate the nature and quantity of LMWOAs present in the rhizosphere of durum wheat cultivars Arcola (low Cd accumulator) and Kyle (high Cd accumulator) grown in three different soils: Yorkton, Sutherland and Waitville, and (2) determine the relationship between Cd accumulation in these plants and LMWOAs present in the rhizosphere. Plants were grown for two weeks in pot-cultures under growth chamber conditions. Oxalic, fumaric, succinic, L-malic, tartaric, citric, acetic, propionic and butyric acids were found and quantified in the water extracts of rhizosphere soil, with acetic and succinic acids being predominant. No water extractable LMWOAs were identified in the bulk soil. Total amount of LMWOAs in the rhizosphere soil of the high Cd accumulator (Kyle) was significantly higher than that for the low Cd accumulator (Arcola) in all three soils. Furthermore, large differences in amounts of LMWOAs were found in the rhizosphere soil for the same cultivars grown in different soils and followed the pattern: Sutherland > Waitville > Yorkton. Extractable soil Cd (M NH4Cl) and Cd accumulation in the plants also followed the same soil sequence as LMWOA production. Cadmium accumulation by the high and low Cd accumulating cultivars was proportional to the levels of LMWOAs found in the rhizosphere soil of each cultivar. These results suggest that the differing levels of LMWOAs present in the rhizosphere soil played an important role in the solubilization of particulate-bound Cd into soil solution and its subsequent phytoaccumulation by the high and low Cd accumulating cultivars. 相似文献

Background and Aims
Cultivars of water spinach (Ipomoea aquatica Forsk.) differ widely in their shoot cadmium (Cd) concentration. Previously, we suggested that low-Cd cultivars are better able to retain Cd in their roots and thus prevent root-to-shoot Cd translocation. In this study, we explored the roles of roots and shoots in Cd accumulation in a high-Cd (T308) and low-Cd cultivar (QLQ).Methods
We used reciprocal grafting to determine the importance of roots and shoots in Cd accumulation, and a dithizone histochemical method to investigate Cd distribution in the roots.Results
The T308 scion with QLQ rootstock accumulated less Cd than the shoot of non-grafted T308. The QLQ scion with T308 rootstock showed a significantly higher Cd concentration than that in the shoot of non-grafted QLQ. Cadmium induced thicker phellem formation in the main roots of QLQ than in those of T308 and only QLQ showed thickening of the outer cortex cell walls in lateral roots.Conclusions
Shoot Cd accumulation was primarily determined by root-to-shoot Cd translocation, not root Cd uptake. The thicker phellem and outer cortex cell walls in QLQ than in T308 may be one reason why QLQ roots were able to retain more Cd, and thus reducing Cd translocation to shoots. 相似文献4.

Cadmium (Cd) in rice is a major source of Cd intake for people on a staple rice diet. The mechanisms underlying Cd accumulation in rice plant are still poorly understood. Here, we characterized the physiology and genetics of Cd transport in a high-Cd-accumulating cultivar (Jarjan) of rice (Oryza sativa). Jarjan showed 5- to 34-fold higher Cd accumulation in the shoots and grains than the cultivar Nipponbare, when it was grown in either a non-Cd-contaminated or a Cd-contaminated soil. A short-term uptake experiment showed no significant difference in Cd uptake by the roots between the two cultivars. However, Jarjan translocated 49% of the total Cd taken up to the shoots, whereas Nipponbare retained most of the Cd in the roots. In both concentration- and time-dependent experiments, Jarjan showed a superior capacity for root-to-shoot translocation of Cd. These results indicate that the high-Cd-accumulation phenotype in Jarjan results from efficient translocation of Cd from roots to shoots. Genetic analysis using an F(2) population derived from Jarjan and Nipponbare revealed that plants showing high- and low-Cd-accumulation phenotypes segregated in a 1:3 ratio, indicating that high accumulation in Jarjan is controlled by a single recessive gene. Furthermore, we isolated OsHMA3, a gene encoding a tonoplast-localized Cd transporter from Jarjan. The OsHMA3 protein was localized in all roots cells, but the sequence has a mutation leading to loss of function. Therefore, failure to sequester Cd into the root vacuoles by OsHMA3 is probably responsible for high Cd accumulation in Jarjan. 相似文献

High Cd content in durum wheat (Triticum turgidum L. var durum) grain grown in the United States and Canada presents potential health and economic problems for consumers and growers. In an effort to understand the biological processes that result in excess Cd accumulation, root Cd uptake and xylem translocation to shoots in seedlings of bread wheat (Triticum aestivum L.) and durum wheat cultivars were studied. Whole-plant Cd accumulation was somewhat greater in the bread wheat cultivar, but this was probably because of increased apoplastic Cd binding. Concentration-dependent 109Cd2+-influx kinetics in both cultivars were characterized by smooth, nonsaturating curves that could be dissected into linear and saturable components. The saturable component likely represented carrier-mediated Cd influx across root-cell plasma membranes (Michaelis constant, 20–40 nm; maximum initial velocity, 26–29 nmol g−1 fresh weight h−1), whereas linear Cd uptake represented cell wall binding of 109Cd. Cd translocation to shoots was greater in the bread wheat cultivar than in the durum cultivar because a larger proportion of root-absorbed Cd moved to shoots. Our results indicate that excess Cd accumulation in durum wheat grain is not correlated with seedling-root influx rates or root-to-shoot translocation, but may be related to phloem-mediated Cd transport to the grain. 相似文献

Zinc effects on cadmium accumulation and partitioning in near-isogenic lines of durum wheat that differ in grain cadmium concentration 总被引:9,自引:0,他引:9
Here, we examined the effectiveness of two approaches for reducing cadmium (Cd) accumulation in durum wheat (Triticum turgidum L. var durum) grain: the application of supplemental zinc (Zn), and the use of cultivars exhibiting reduced grain Cd concentrations. Two durum wheat near-isogenic lines (NIL) that differ in grain Cd accumulation were grown to maturity in solution culture containing a chelating agent to buffer the free activities of Zn and Cd at levels approximating those of field conditions. The low Cd accumulating (L-Cd) isoline had Cd concentrations, in grains and shoot parts, which were 60-70% lower than those of the high Cd accumulating (H-Cd) isoline. Increasing the Zn activities in the nutrient solution from deficient to sufficient levels reduced the concentration of Cd in grains and vegetative shoot parts of both isolines. The results suggest that supplemental Zn reduces Cd tissue concentrations by inhibiting Cd uptake into roots. Cd partitioning patterns between roots and shoots and between spike components suggest that the physiological basis for the low Cd trait is related to the compartmentation or symplasmic translocation of Cd. 相似文献

roots increased with increasing external Cd concentrations. Relationships between corrected parameters of growth, photosynthesis
and fluorescence and corrected Cd concentrations in shoots and roots could be explained by the regression model Y = K/(1 + exp(a + bX)). Jing 411 was found to be Cd tolerant considering parameters of chlorophyll content, photosynthesis and chlorophyll fluorescence
in which less Cd translocation was from roots into shoots. The high Cd concentrations were in shoots and roots in Yangmai

The main objective of this study was to investigate the accumulation and distribution of strontium (Sr) in 26 cultivars of wheat (Triticum aestivum L.), husk oat (Avena sativa L) and naked oat (Avena nuda), and barley (Hordeum vulgare L.) for their potential use in phytoremediation.Sr levels had no effect on the accumulation of shoot biomass at tillering or at maturity. Mean shoot Sr concentration of naked oat and barley at tillering was significantly (P < 0.05) higher than that of wheat; Neimengkeyimai-1, a naked oat cultivar, had the highest Sr concentrations. At maturity, of four naked oat cultivars, Neimengkeyimai-1 had the highest Sr content at all measured Sr levels. Leaves had the highest Sr concentrations, followed by roots and straw, and then grain with the lowest. Mean enrichment coefficients from soil to shoots ranged from 0.521 to 1.343; the percentage of stable Sr removed from the soil to the shoots at harvest time was more than 1.4% after 120 days. Neimengkeyimai-1 could be used as a model for further research to find more effective cultivars; and naked oat plants could be selected for phytoremediation to clean up contaminated soil. 相似文献

Effect of zinc (Zn) nutritional status on uptake of inorganic 65Zn was studied in rye (Secale cereale, cv. Aslim), three bread wheat (Triticum aestivum, cvs. Dagdas, Bezostaja, BDME-10)
and durum wheat (Triticum durum, cv. Kunduru-1149) cultivars grown for 13 days in nutrient solution under controlled environmental
conditions. The cultivars were selected based on their response to Zn deficiency and to Zn fertilization in calcareous soils
under field conditions. When grown in Zn-deficient calcareous soil in the field, the rye cultivar had the highest, and the
durum wheat the lowest Zn efficiency. Among the bread wheats, BDME-10 showed higher susceptibility to Zn deficiency and Bezostaja
and Dagdas were less affected by Zn deficiency. Similarly to field conditions, in nutrient solution visual Zn deficiency symptoms
(i.e. necrotic lesions on leaf blade) appeared to be more severe in Kunduru-1149 and BDME-10 and less severe in rye cultivar
Aslim. Under Zn deficiency, shoot concentrations of Zn were similar between all cultivars. Cultivars with adequate Zn supply
did not differ in uptake and root-to-shoot translocation rate of 65Zn, but under Zn deficiency there were distinct differences; rye showed the highest rate of Zn uptake and the durum wheat
the lowest. In the case of bread wheat cultivars, 65Zn uptake rate was about the same and not related to their differential Zn efficiency. Under Zn deficiency, rye had the highest
rate of root-to-shoot translocation of 65Zn, while all bread and durum wheat cultivars were similar in their capacity to translocate 65Zn from roots to shoots. When Zn2+ activity in uptake solution ranged between 117 p M and 34550 pM, Zn-efficient and Zn-inefficient bread wheat genotypes were
again similar in uptake and root-to-shoot translocation rate of 65Zn.
The results indicate that high Zn efficiency of rye can be attributed to its greater Zn uptake capacity from soils. The inability
of the durum wheat cultivar Kunduru-1149 to have a high Zn uptake capacity seems to be an important reason for its Zn inefficiency.
Differential Zn efficiency between the bread wheat cultivars used in this study is not related to their capacity to take up
inorganic Zn.

Phytochelatins (PCs) are peptides that function in heavy-metal chelation and detoxification in plants and fungi. A recent study showed that PCs have the ability to undergo long-distance transport in a root-to-shoot direction in transgenic Arabidopsis (Arabidopsis thaliana). To determine whether long-distance transport of PCs can occur in the opposite direction, from shoots to roots, the wheat (Triticum aestivum) PC synthase (TaPCS1) gene was expressed under the control of a shoot-specific promoter (CAB2) in an Arabidopsis PC-deficient mutant, cad1-3 (CAB2TaPCS1/cad1-3). Analyses demonstrated that TaPCS1 is expressed only in shoots and that CAB2TaPCS1/cad1-3 lines complement the cadmium (Cd) and arsenic metal sensitivity of cad1-3 shoots. CAB2TaPCS1/cad1-3 plants exhibited higher Cd accumulation in roots and lower Cd accumulation in shoots compared to wild type. Fluorescence HPLC coupled to mass spectrometry analyses directly detected PC2 in the roots of CAB2:TaPCS1/cad1-3 but not in cad1-3 controls, suggesting that PC2 is transported over long distances in the shoot-to-root direction. In addition, wild-type shoot tissues were grafted onto PC synthase cad1-3 atpcs2-1 double loss-of-function mutant root tissues. An Arabidopsis grafting technique for mature plants was modified to obtain an 84% success rate, significantly greater than a previous rate of approximately 11%. Fluorescence HPLC-mass spectrometry showed the presence of PC2, PC3, and PC4 in the root tissue of grafts between wild-type shoots and cad1-3 atpcs2-1 double-mutant roots, demonstrating that PCs are transported over long distances from shoots to roots in Arabidopsis. 相似文献

Accumulation of cadmium (Cd) in crop plants is of great concern due to the potential for food chain contamination through the soil-root interface. Although Cd uptake varies considerably with plant species, the processes which determine the accumulation of Cd in plant tissues are affected by soil factors. The influence of soil type on Cd uptake by durum wheat (Triticum turgidum var. durum L.) and flax (Linum usitatissimum L.) was studied in a pot experiment under environmentally controlled growth chamber conditions. Four cultivars/lines of durum wheat (Kyle, Sceptre, DT 627, and DT 637) and three cultivars/lines of flax (Flanders, AC Emerson, and YSED 2) were grown in two Saskatchewan soils: an Orthic Gray Luvisol (low background Cd concentration; total/ABDTPA extractable Cd: 0.12/0.03 mg kg-1, respectively) and a Dark Brown Chernozem (relatively high background Cd concentration; total/ABDTPA Cd: 0.34/0.17 mg kg-1 respectively). Plant roots, stems, newly developed heads, and grain/seeds were analyzed for Cd concentration at three stages of plant growth: two and seven weeks after germination, and at plant maturity. The results showed that Cd bioaccumulation and distribution within the plants were strongly affected by both soil type and plant cultivar/line. The Cd concentration in roots leaves and stems varied at different stages of plant growth. However, all cultivars of both plant species grown in the Chernozemic soil accumulated more Cd in grain/seeds than plants grown in the Orthic Gray Luvisol soil. The different Cd accumulation pattern also corresponded to the levels of ABDTPA extractable and metal-organic complex bound soil Cd found in both soils. Large differences were found in grain Cd among the durum wheat cultivars grown in the same soil type, suggesting the importance of rhizosphere processes in Cd bioaccumulation and/or Cd transport processes within the plant. Distribution of Cd in parts of mature plants showed that durum grain contained up to 21 and 36% of the total amount of Cd taken up by the plants for the Orthic Gray Luvisol and Chernozemic soils, respectively. These results indicate the importance of studying Cd speciation, bioaccumulation and cycling in the environment for the management of agricultural soils and crops. 相似文献

Uptake and retranslocation of leaf-applied radiolabeled cadmium (109Cd) was studied in three diploid (Triticum monococcum, AA), four tetraploid (Triticum turgidum, BBAA) and two hexaploid (Triticum aestivum, BBAADD) wheat genotypes grown for 9 d under controlled environmental conditions in nutrient solution. Among the tetraploid wheats, two genotypes were primitive (ssp. dicoccum) and two genotypes modern wheats (ssp. durum). Radiolabelled Cd was applied by immersing the tips (3 cm) of mature leaf into a 109Cd radiolabelled solution. There was a substantial variation in the uptake and export of 109Cd among and within wheat species. On average, diploid wheats (AA) absorbed and translocated more 109Cd than other wheats. The largest variation in 109Cd uptake was found within tetraploid wheats (BBAA). Primitive tetraploid wheats (ssp. dicoccum) had a greater uptake capacity for 109Cd than modern tetraploid wheats (ssp. durum). In all wheats studied, the amount of the 109Cd exported from the treated leaf into the roots and the remainder of the shoots was poorly related to the total absorption. For example, bread wheat cultivars were more or less similar in total absorption, but differed greatly in the amount of 109Cd retranslocated. The diploid wheat genotype 'FAL-43' absorbed the lowest amount of 109Cd, but retranslocated the greatest amount of 109Cd in roots and remainder of shoots. The results indicate the existence of substantial genotypic variation in the uptake and retranslocation of leaf-applied 109Cd. This variation is discussed in terms of potential genotypic differences in binding of Cd to cell walls and the composition of phloem sap ligands possibly affecting Cd transport into sink organs. 相似文献

以2个耐镉(Cd)性不同的甘蓝品种为材料,研究了不同Cd浓度(0、20、50、100μmol.L-1)对甘蓝植株生长、叶片光合特性和养分吸收的影响.结果表明:Cd敏感品种在低浓度Cd(20μmol.L-1)处理下生长受到明显抑制,叶片净光合速率(Pn)、气孔导度(Gs)、PSⅡ光化学效率(Fv/Fm)、PSⅡ光合电子传递量子效率(ΦPSⅡ)及地上部、根系干质量显著降低;Cd耐性品种在高浓度Cd(50和100μmol.L-1)处理下生长和光合特性受到显著影响;Cd胁迫降低了甘蓝叶片叶绿素a和b含量,尤其对叶绿素a的影响较大,进而抑制了叶片光合能力.Cd胁迫显著降低了植株对Mn的吸收,抑制了Mg和Fe从根部向地上部的转运,且Cd敏感品种受抑制幅度更大;Cd胁迫促进了Cd耐性品种对P和S的吸收,而Cd敏感品种相反.因此,Cd胁迫下甘蓝敏感品种叶片Mn、Fe、Mg、S和P含量的降低是影响其叶片光合作用,进而抑制植株生长的重要生理原因. 相似文献

Improved nitrogen nutrition enhances root uptake, root-to-shoot translocation and remobilization of zinc ((65) Zn) in wheat 总被引:4,自引:0,他引:4
This study focussed on the effect of increasing nitrogen (N) supply on root uptake and root-to-shoot translocation of zinc (Zn) as well as retranslocation of foliar-applied Zn in durum wheat (Triticum durum). Nutrient solution experiments were conducted to examine the root uptake and root-to-shoot translocation of (65) Zn in seedlings precultured with different N supplies. In additional experiments, the effect of varied N nutrition on retranslocation of foliar-applied (65) Zn was tested at both the vegetative and generative stages. When N supply was increased, the (65) Zn uptake by roots was enhanced by up to threefold and the (65) Zn translocation from roots to shoots increased by up to eightfold, while plant growth was affected to a much smaller degree. Retranslocation of (65) Zn from old into young leaves and from flag leaves to grains also showed marked positive responses to increasing N supply. The results demonstrate that the N-nutritional status of wheat affects major steps in the route of Zn from the growth medium to the grain, including its uptake, xylem transport and remobilization via phloem. Thus, N is a critical player in the uptake and accumulation of Zn in plants, which deserves special attention in biofortification of food crops with Zn. 相似文献

Previous studies on the fate of fertiliser nitrogen applied to winter wheat in temperate climates have shown that nitrogen
(N) applied early, at tillering for wheat, was less efficiently taken up than N applied later in the growth cycle. We examined
the extent to which the soil microbial N immobilisation varied during the wheat spring growth cycle and how microbial immobilisation
and plant uptake competed for nitrogen. We set up a pulse-15N labelled field experiment in which N was applied at eight development
stages from tillering (beginning of March) to anthesis (mid-June). Each application was 50 kg N ha-1 as 15N labelled urea
except for the first application which was 25 kg N ha-1. The distribution of fertiliser 15N in shoots, roots, mineral and
organic soil N was examined by destructive sampling 7 and 14 days after each 15N pulse. The inorganic 15N pool was almost
depleted by day 14. The N uptake efficiency increased with later applications from 45% at tillering to 65% at flowering. N
immobilisation was rather constant at 13–16% of N applied, whatever the date of application. The increase in plant 15N uptake
resulted in an increase in the total 15N recovery in the plant-soil system (15N in soil +15N in plant), suggesting that gaseous

以抗旱性不同的小麦品种为材料,在小麦的水分临界期开花期进行缓慢脱水处理,分别在脱水的不同阶段取样测定叶片及根系的渗透调节能力及渗透调节物质。结果表明:随着土壤含水量的降低,叶片与根系的饱和渗透势同步下降,表现出叶片与根系对水分胁迫反应的一到场生,但根系的渗透调节能力低于叶片。根系与叶片的渗透调节物质,一方面在物质总含量方面,表现出与渗透调节能力的一致性,另一方面各种物质的相对含量又有一定差异,叶片中可溶性糖与K+含量及增加量都高于根系,而根系中的游离氨基酸与Ca^2 的相对增加量则大于叶片。 相似文献

Identification of RAPD markers linked to a gene governing cadmium uptake in durum wheat. 总被引:17,自引:0,他引:17
Temperature sweep gel electrophoresis in combination with random amplified polymorphic DNA analysis was employed to detect two markers for a single gene governing low cadmium uptake in western Canadian durum wheat (Triticum turgidum L. var. durum). Analysis of progeny derived from a cross of the high cadmium accumulating cultivar Kyle by the low cadmium accumulating cultivar Nile resulted in linkage estimates of 4.6 (OPC-20) and 21.2 (UBC-180) cM. The closest marker (OPC-20) was shown to be useful for making low cadmium uptake selections from two other sources of low cadmium, 'Biodur' and 'Hercules'. The marker was further used to determine the genetic basis of resistance in 20 introduced durum wheat lines. Within this diverse range of germplasm the marker was correlated with cadmium contents as expected in all but two cases. Plant breeding selection for low cadmium genotypes is hindered by the high cost of chemical determination of cadmium content. Marker assisted selection for a low cadmium uptake gene offers an effective alternative. 相似文献

Cadmium (Cd) tolerance and accumulation in wheat varieties were investigated at seedling stage under a controlled environmental
condition. The 46 leading wheat varieties cultivated from the 1950s to 2000s in China were treated at the three-leaf stage
with a 50 μM CdCl2 solution for 24 days. Growth and photosynthesis parameters were measured and the Cd-tolerance index (ratio of a given parameter
under Cd treatment to that of the control) was determined. Cd accumulation in shoots and roots and Cd translocation were also
determined. It was found that Cd tolerance and accumulation of these wheat varieties varied over the different decades. Principal
components analysis (PCA) showed that wheat varieties in 1950s and 1980s were tolerant while varieties from the last decade
were sensitive to Cd stress. Wheat varieties in 1960s and 1970s were particularly tolerant to Cd stress for the parameters
of shoot height, secondary root numbers, net photosynthesis and transpiration rate while the varieties in the 1990s were sensitive
to Cd stress for shoot dry weight and root dry weight. Comparing each decade to the average Cd translocation ratio from the
roots to the shoots for the whole period, the varieties from the 1950s and 1960s had a higher translocation ratio, while varieties
in the 1970s were below that average. Varieties from the 1980s to 2000s showed an average translocation ratio. Using cluster
analysis (CA), Shannongfu 63, Yangmai 1 and Yangmai 158 were the most Cd-tolerant varieties in which Cd translocation ratio
were low, and Yumai 18 and Huaimai 20 were the most Cd-sensitive varieties in which Cd translocation ratio were high. The
results indicating that wheat varieties from different decades were different in Cd tolerance and accumulation, and could

Abstract An isolate of Pseudomonas aureofaciens from the phylloplane of sugar beet which was chromosomally modified for monitors purposes by the insertion of two gene cassettes (kmr - xyl E and lac ZY) was introduced to the phytosphere of spring wheat in a number of experiments and the resulting microbial perturbations quantified. Such studies involving innocuous bacterial isolates can serve as a guide in the assessment of risk associated with the release of functionally modified microorganisms. Introductions of P. aureofaciens on seeds caused large microbial perturbations (up to 2 log units) at the seedling stage on seeds and roots. As the inoculated plants matured (tillering, flowering and ripening), perturbations of total microbial populations were found to be non-significant. Microbial perturbation on maturing wheat roots as a result of seed inoculations with P. aureofaciens could only be detected using more sensitive monitoring procedures describing the Pseudomonas community in terms of colony appearance rate on a selective Pseudomonas medium. Spray applications of the marked P. aureofaciens isolate onto the leaf surface of wheat caused no significant perturbations of the indigenous microbial present on the phylloplane. 相似文献

Sedum alfredii (Crasulaceae) is the only known Cd-hyperaccumulating species that are not in the Brassica family; the mechanism of Cd hyperaccumulation in this plant is, however, little understood. Here, a combination of radioactive techniques, metabolic inhibitors, and fluorescence imaging was used to contrast Cd uptake and translocation between a hyperaccumulating ecotype (HE) and a non-hyperaccumulating ecotype (NHE) of S. alfredii. The K(m) of (109)Cd influx into roots was similar in both ecotypes, while the V(max) was 2-fold higher in the HE. Significant inhibition of Cd uptake by low temperature or metabolic inhibitors was observed in the HE, whereas the effect was less pronounced in the NHE. (109)Cd influx into roots was also significantly decreased by high Ca in both ecotypes. The rate of root-to-shoot translocation of (109)Cd in the HE was >10 times higher when compared with the NHE, and shoots of the HE accumulated dramatically higher (109)Cd concentrations those of the NHE. The addition of the metabolic inhibitor carbonyl cyanide m-chlorophenylhydrazone (CCCP) resulted in a significant reduction in Cd contents in the shoots of the HE, and in the roots of the NHE. Cd was distributed preferentially to the root cylinder of the HE but not the NHE, and there was a 3-5 times higher Cd concentration in xylem sap of the HE in contrast to the NHE. These results illustrate that a greatly enhanced rate of root-to-shoot translocation, possibly as a result of enhanced xylem loading, rather than differences in the rate of root uptake, was the pivotal process expressed in the Cd hyperaccumulator HE S. alfredii. 相似文献
