The DNA probes, P1887, P2405, P2056 (being specific tags for Aedes aegypti genes coding for ribosomal RNA) and a centric heterochromatin probe, K20-1A5, were chosen to hybridize the metaphase chromosomes from the testes of four mosquito species, Culex pipiens, Aedes aegypti, Aedes albopictus and Aedes triseriatus. In addition, a single plasmid, P2392, which contained the three probes, P1887, P2405 and P2056, was also used as chromosome landmark in aedine species. Only the Aedes aegypti metaphase chromosome 1-specific tag, P1887, was conserved in Aedes albopictus, Aedes triseriatus, and Culex pipiens metaphase chromosomes. Aedes triseriatus exhibited two gene loci, on chromosomes 1 and 3, coding for ribosomal RNA per haploid genome. When the specific probes for chromosomes 2 and 3, 2405 and 2056, were used in the fluorescence in situ hybridization technique against the metaphase chromosomes the fluorescent signals were not seen in Aedes albopictus, Aedes triseriatus or Culex pipiens. Also the centric heterochromatin probe, K20-1A5, exhibited strong fluorescent signals on chromosomes 1, 2 and 3 of Aedes aegypti. These fluorescent signals were not observed in metaphase chromosomes derived from the other aedine species, indicating that the centromere sequence can vary within the species.This paper was presented at the Second Arab Conference on Biotechnology and Genetic Engineering, held in the Kingdom of Bahrain, 15–17 April, 2002 and is published here with the endorsement of the Co-ordinator of the Scientific Committee, Professor Essam H. Summary Replicated divergent selection based upon the time taken to recover from the larval alarm reaction in the mosquito Aedes aegypti resulted in lines which recovered faster and slower than the control lines. Estimates of the realized heritability were consistent, ranging from 0.21 to 0.24 in the fast replicates and 0.19 to 0.20 in the slow replicates. After 11 generations of selection an apparent change in the fitness was examined using an application of the path analysis. The relevance of the findings to natural selection is also discussed.  相似文献

Aedes aegypti and Aedes albopictus are potential arboviral vectors leading to high human fatality worldwide. Efforts in the present study were made to differentiate the eggs of A. aegypti and A. albopictus morphologically and morphometrically using scanning electron microscopy (SEM). Morphometrically, these species’ eggs were 48.48% significantly different of the 33 attributes including egg dimensions, micropylar apparatus, dimensions and density of outer chorionic cells (OCCs), tubercles and width of exochorionic network. In comparison to A. aegypti eggs, A. albopictus eggs were significantly smaller and more tapered at the posterior end; however, the micropylar disc of A. aegypti was wider and had incomplete circular sectors whereas it was a narrower polygon without sectors in A. albopictus. These species were also significantly different with regards to OCC which enclose both large central and small peripheral tubercles. Specifically, the exochorionic networks in A. aegypti were interwoven, reticulated and extensively wide whereas they were narrow, prominent and solid-wall-like in A. albopictus. This feature may strengthen A. albopictus eggs against desiccation, when they are laid in containers. The morphometrical and morphological analysis of the egg’s attributes of A. aegypti and A. albopictus may be helpful in understanding egg biology as well as in species confirmation.  相似文献

Very simply constructed aquatic sticky light traps employing a chemical solution emitting a bright light attracted large numbers of all larval instars, but few pupae of Aedes cantans in trials in England. During April numbers of larvae per trap ranged from 73±14.2 to 163±19.9, whereas dipping with a ladle yielded only 6.9±5.5 to 11.8±7.7 larvae/dip. The traps were also effective in sampling larvae of Culiseta morsitans and in Sierra Leone larvae of Aedes aegypti. It was concluded that sticky light traps could be valuable in sampling relative numbers of mosquito larvae and merited further evaluation.
Résumé Des pièges adhésifs lumineux faciles à réaliser sont construits pour les captures des larves de moustiques. Ils sont formés de paires de boîtes de Pétri en plastique contenant une solution émettrice de lumière provenant de bâtons lumineux chimiques Cyalume®. Chaque piège est recouvert d'une feuille de plastique de 16×16 cm enduite d'une substance qui reste adhésive sous l'eau. Les larves de moustiques attirées vers la source lumineuse viennent se coller à la feuille adhésive.Ces pièges ont été utilisés avec succès en Angleterre pour l'échantillonnage de tous les stades larvaires d'Aedes cantans, mais pas avec le même efficacité pour les nymphes. Des tests plus restreints ont montré que la technique était également utilisable pour les larves de Culiseta morsitans. Les nombres moyens d'A. cantans capturés pendant une piode de 12 h (73±14,2–163±19,9) étaient supérieux à ceux obtenus en plongeant une louche (6,9±5,5–11,8±7,7). En utilisant les pièges adhésifs lumineux, les captures moyennes de C. morsitans étaient également supérieures (12,6±4,7–35,8±8,9) contre (5,7±6,1–8,9±7,8) avec la méthode de la louche.En Sierra Leone, des stades larvaires immatures d'A. aegypti se développant dans des containers de stockage d'eau ont également pu être capturés à l'aide de ces pièges.Bien qu'une évaluation plus poussée soit encore nécessaire, il ne fait aucun doute que ces pièges adhésifs lumineux aient un intérêt considérable pour l'échantillonnage larvaire dans les sites inaccessibles, tels que les trous d'arbres ou les trous de crabes, où les méthodes d'échantillonage classiques sont souvent d'un emploi difficile.

The lack of eye pigment in the Aedes aegypti WE (white eye) colony was confirmed to be due to a mutation in the kynurenine hydroxylase gene, which catalyzes one of the steps in the metabolic synthesis of ommochrome eye pigments. Partial restoration of eye color (orange to red phenotype) in pupae and adults occurred in both sexes when first or second instar larvae were reared in water containing 3-hydroxykynurenine, the metabolic product of the enzyme kynurenine hydroxylase. No eye color restoration was observed when larvae were reared in water containing kynurenine sulfate, the precursor of 3-hydroxykynurenine in the ommochrome synthesis pathway. In addition, a plasmid clone containing the wild type Drosophila melanogaster gene encoding kynurenine hydroxylase, cinnabar (cn), was also able to complement the kynurenine hydroxylase mutation when it was injected into embryos of the A. aegypti WE strain. The ability to complement this A. aegypti mutant with the transiently expressed D. melanogaster cinnabar gene supports the value of this gene as a transformation reporter for use with A. aegypti WE and possibly other Diptera with null mutations in the kynurenine hydroxylase gene.  相似文献

Summary Two pure translocation homozygote stocks, T1/T1 and T3/T3, were used to produce a double translocation heterozygote system designated T1/T3, employing T1/T1 as the male and T3/T3 as the female parent. The double heterozygote showed 73 % sterility when mated to wild females. Tests on mating competitiveness, recombination frequency in the differential segment, insemination rate and inheritance of sterility after release, for four generations in laboratory cages, have been carried out to evaluate the efficiency of this strain as an agent for a population control programme.  相似文献

The left (5) inverted terminal repeat (ITR) of the Mos1 mariner transposable element was altered by site-directed mutagenesis so that it exactly matched the nucleotide sequence of the right (3) ITR. The effects on the transposition frequency resulting from the use of two 3 ITRs, as well as those caused by the deletion of internal portions of the Mos1 element, were evaluated using plasmid-based transposition assays in Escherichia coli and Aedes aegypti. Donor constructs that utilized two 3 ITRs transposed with greater frequency in E. coli than did donor constructs with the wild-type ITR configuration. The lack of all but 10 bp of the internal sequence of Mos1 did not significantly affect the transposition frequency of a wild-type ITR donor. However, the lack of these internal sequences in a donor construct that utilized two 3 ITRs resulted in a further increase in transposition frequency. Conversely, the use of a donor construct with two 3 ITRs did not result in a significant increase in transposition in Ae. aegypti. Furthermore, deletion of a large portion of the internal Mos1 sequence resulted in the loss of transposition activity in the mosquito. The results of this study indicate the possible presence of a negative regulator of transposition located within the internal sequence, and suggest that the putative negative regulatory element may act to inhibit binding of the transposase to the left ITR. The results also indicate that host factors which are absent in E. coli, influence Mos1 transposition in Ae. aegypti.Communicated by G. P. Georgiev  相似文献

A new cell line, UM-AVE1, was established from embryos of the mosquito Aedes vexans. Banding patterns for the isozymes lactate dehydrogenase (LDH), malate dehydrogenase (MDH), isocitrate dehydrogenase (IDH), xanthine dehydrogenase (XDH), and esterases were compared with those of larval Aedes vexans tissues as well as those of four other mosquito cell lines and one moth cell line. Karyotype analyses confirmed that the dipteran cell lines were not contaminated with lepidopteran cells, because in all mosquito lines the modal number of chromosomes was 6 (=2n) or 7. Isozyme electrophoresis established a specific profile for each cell line. Two isozymes present in UM-AVE1 (LDH, IDH) were not detected in larvae; this could be a reflection of the different stages used for cell line isolation and enzyme analysis, or lability of sample preparations. It is significant that extracts from UM-AVE1 cells and Aedes vexans larvae had an identical double band for XDH, while all other cell lines examined exhibited only a single band.  相似文献

Essential oils obtained from the flowers of Dendropanax morbifera were extracted and the chemical composition and larvicidal effects were studied. The analyses were conducted by gas chromatography and mass spectroscopy (GC–MS) revealed that the essential oil of D. morbifera contained 27 compounds. The major chemical components identified were γ-elemene (18.59%), tetramethyltricyclohydrocarbon (10.82%), β-selinene (10.41%), α-zingibirene (10.52%), 2-isopropyl-5-methylbicylodecen (4.2%), β-cubebene (4.19), and 2,6-bis(1,1-Dimethylethyl)-4-phenol (4.01%). The essential oil had a significant toxic effect against early fourth-stage larvae of Aedes aegypti L. with an LC50 value of 62.32 ppm and an LC90 value of 131.21 ppm. The results could be useful in search for newer, safer, and more effective natural larvicidal agents against A. aegypti.  相似文献

We studied interspecific competition between the larvae of the two mosquitoesAedes albopictus andTripteroides bambusa, which are predominantly found in water-filled bamboo stumps in northern Kyushu, south-western Japan, using microcosms with dead bamboo leaves in the laboratory. We compared short-term competition between single cohorts of the two species and long-term competition involving four cohorts of each species, which were introduced at 6-day intervals. In the single cohort experiment,A. albopictus grew faster thanT. bambusa. However, in the multiple cohort experiment, although the first cohort ofA. albopictus grew faster and began to pupate earlier than that ofT. bambusa, molting rates of later cohorts ofA. albopictus, that were introduced on the 12th and the 18th day, were lower than those ofT. bambusa. The survival rate ofA. albopictus became lower than that ofT. bambusa after the 18th day. The cumulative number of the pupatedT. bambusa individuals exceeded that ofA. albopictus on the 96th day. The final pupation success was higher inT. bambusa than inA. albopictus, especially when additional leaves were supplied on the 48th and the 96th days. The reversed outcomes between short- and long-term interspecific competition and the variation in the lifespans of small aquatic sites may contribute to the coexistence of the two mosquito species in bamboo groves.  相似文献

Dengue is a human disease transmitted by the mosquito Aedes aegypti. For this reason geographical regions infested by this mosquito species are under the risk of dengue outbreaks. In this work, we propose a mathematical model to study the spatial dissemination of dengue using a system of partial differential reaction-diffusion equations. With respect to the human and mosquito populations, we take into account their respective subclasses of infected and uninfected individuals. The dynamics of the mosquito population considers only two subpopulations: the winged form (mature female mosquitoes), and an aquatic population (comprising eggs, larvae and pupae). We disregard the long-distance movement by transportation facilities, for which reason the diffusion is considered restricted only to the winged form. The human population is considered homogeneously distributed in space, in order to describe localized dengue dissemination during a short period of epidemics. The cross-infection is modeled by the law of mass action. A threshold value as a function of the model's parameters is obtained, which determines the rate of dengue dissemination and the risk of dengue outbreaks. Assuming that an area was previously colonized by the mosquitoes, the rate of disease dissemination is determined as a function of the model's parameters. This rate of dissemination of dengue disease is determined by applying the traveling wave solutions to the corresponding system of partial differential equations.  相似文献

Aedes aegypti from the Brazilian cities of São José do Rio Preto (SJ) and Goiânia (GO) were analyzed as to their esterase patterns and the results were compared with data obtained about 5 years before for SJ population. Esterase bands not detected in the previous study were now observed in mosquitoes from both SJ and GO populations, being the last considered a population resistant to insecticides. Other similarities between SJ and GO populations in this study, and some differences in comparison with the previous data on SJ were observed, involving, in addition to changes in band type, changes in frequency of mosquitoes expressing them and differential gene activation during development. As it is generally true for genetic features, changes in the esterase patterns are expected to be the result of factors such as selection by environmental conditions and genetic drift. In the present case, continuous use of insecticides aiming mosquito population size control in SJ by sanitary authorities could be involved in the observed changes. Changed esterases were classified as carboxylesterases and cholinesterases, which are enzymes already shown to take part in the development of resistance in several organisms. In addition, data obtained in the elapsed time by authorities responsible for the mosquito control has shown increasing insecticide resistance of SJ population mosquitoes parallel to increase in the total amount of esterases, reinforcing the mentioned possibility.  相似文献

Invasive arthropods that vector pathogens have the potential to influence pathogen transmission both directly, by becoming a novel pathogen vector, or indirectly, by interacting with native vectors. Adult mosquito size is influenced by food availability in the larval stage, and smaller, nutrient-deprived mosquitoes are, in some studies, more efficient viral vectors in the laboratory. This is the first study to examine the indirect impacts that larval competition between Aedes albopictus, an introduced mosquito species, and Ochlerotatus triseriatus, a native mosquito species and the primary vector for La Crosse virus (LACV) in the US, has on native mosquito larval survival, adult size, and vector competence. A. albopictus presence decreased Oc. triseriatus larval survival, but surviving Oc. triseriatus females were larger, potentially owing to a release from intraspecific competition. These larger, native females were more likely to develop both midgut and disseminated LACV infections than females emerging from monospecific treatments. Collectively, these results suggest a need to better understand the ecology of both native and invasive vector species, their interactions, and the potential for those interactions to alter vector-borne disease transmission.  相似文献

Predator-mediated coexistence of competitors occurs when a species that is superior in competition is also more vulnerable to a shared predator compared to a poorer competitor. The invasive mosquito Aedes albopictus is usually competitively superior to Ochlerotatus triseriatus. Among second instar larvae, A. albopictus show a lesser degree of behavioral modification in response to water-borne cues from predation by the larval midge Corethrella appendiculata than do O. triseriatus, rendering A. albopictus more vulnerable to predation by C. appendiculata than O. triseriatus. The hypothesis that C. appendiculata predation favors coexistence of these competitors predicts that C. appendiculata abundances will be negatively and positively correlated with A. albopictus and O. triseriatus abundances, respectively, and that coexistence will occur where C. appendiculata are common. Actual abundances of O. triseriatus, A. albopictus, and C. appendiculata in three habitats fit this prediction. In natural container habitats like tree holes, C. appendiculata were abundant and competitors co-existed at similar densities. In cemeteries and tires, which occur primarily in non-forested, human-dominated habitats, A. albopictus dominated, with abundances twice those found in tree holes, but C. appendiculata and O. triseriatus were rare or absent. We also tested for whether antipredatory behavioral responses of A. albopictus differed among habitats or populations, or were correlated with local C. appendiculata abundances. We could detect no differences in A. albopictus antipredatory behavioral responses to water-borne cues from predation. Tree hole habitats appear to promote co-existence of O. triseriatus and A. albopictus through interactions with predatory C. appendiculata, and this predator effect appears to limit invasion success of A. albopictus in tree holes. There are many studies on predator-mediated coexistence in natural habitats but to our knowledge this is the first study to suggest differential predator-mediated coexistence between natural and man-made habitats. The larvicidal effects of polyphenols of natural crude decomposed alder leaf litter and commercially available tannic acid were experimentally compared with those of two common conventional insecticides (Bacillus thuringiensis ssp. israelensis: microbial insecticide; temephos: organophosphate insecticide). Comparative standard bioassays using third instar larval Aedes aegypti, A. albopictus, Culex pipiens and Coquillettidia richiardii as references indicated that Aedes and Culex taxa are far more sensitive to alder leaf litter than to tannic acid and conventional insecticides. C. richiardii is far more resistant to conventional insecticides than Aedes and Culex taxa, but its sensitivity to tannic acid is close to that of those taxa. Dietary vegetable polyphenols are thus proposed as new, practical, alternative chemicals for mosquito control when conventional insecticides are difficult and costly to be used (e.g., in the management of Aedes and Culex populations in man-made breeding sites and Coquillettidia control strategy).  相似文献

Four regions of hexokinase activity are detected by starch gel electrophoresis of adult Aedes aegypti. Three of the regions, Hk-2, Hk-3, and Hk-4, are produced by three tightly linked loci, located on the third chromosome 7.25 map units from the locus fuzzy. Analysis of larval Aedes aegypti midgut using scanning electron microscopy, nuclear and mitochondrial dyes, response to Bacillus thuringiensis israelensis CryIVB toxin, and electrophysiology is described. The anterior ventriculus ("stomach") region is found to have much lower mitochondrial densities than other midgut regions. The transitional region is distinguished by apical surface architecture, and by region-specific effects of CryIVB endotoxin. In this region CryIVB causes holes ranging from 1.0 to 7.0 microm in diameter (mean 3.3+/-0.53 microm, N=12), blisters 16.9+/-1.54 microm in diameter (N=10), and separation of adjacent cells. The holes are not consistent with damage due to the colloid osmotic lysis model of delta-endotoxin activity. The posterior ventriculus possesses a distinctive cellular architecture consisting of hemispherical, domed apical membranes surrounded by deep clefts. Functional and morphological heterogeneity is revealed within the posterior ventriculus, with the anterior end dominating the electrical profile of isolated, perfused preparations and showing the greatest response to serotonin. Hyperpolarization of the transepithelial potential by serotonin occurred in conjunction with a decrease in the space constant lambda, ruling out closure of ion channels as the mechanism of action of serotonin.  相似文献

An electrophoretic survey of esterases in 7 wild-type and 10 mutant strains of the mosquito Aedes (Finlaya) togoi was undertaken using thin-layer agar gels. Three esterases (designated the Est-1, Est-2, and Est-3 loci in decreasing order of electrophoretic mobility) could be detected from fourth-instar larvae, pupae, and 2- to 5-day-old adults. Homogenates of the larvae gave the most intensely stained bands in the gels, especially for Est-3. The three esterases were designated carboxylesterases based on their response to the two esterase inhibitors, eserine and paraoxon (diethyl-p-nitrophenyl phosphate). The Est-3 locus was found to have five alleles including at least one null. The linkage results of six backcrosses suggest that Est-3 is located only 5–8 map units from the sex allele (m) and the gene arrangement is Est-3-m-s (straw-colored larva) in linkage group I.This work was supported by National Institutes of Health Grant AI 16983-01.  相似文献

A series of experiments were conducted to determine the infection sites of the entomopathogenic fungus Beauveria bassiana after ingestion by the larvae of Aedes aegypti. The timing of the host death in relation to larval molting, the principle sites of fungal infection and development in host tissues were studied. Fungal conidiospores (CS) and blastospores (BS) were used separately for treatment of mosquito larvae. Although in most instances CS germinated and developed within the host, in others there was a premature abortion of the fungal development cycle. On the other hand, BS ingested by the larvae showed differences in the fungal development stages in the larval tissues. While the two primary infection sites were the head and the anal region, the most preferred site for fungal development was the larval gut. No more than two cycles of fungal development can occur in the host. Although both CS and BS are effective as larvicides, BS is far more pathogenic.
Résumé Une série d'expériences a été menée afin de déterminer les emplacements d'infection du fongus entomopathogénique Beauveria bassiana (souche GK 2016) lequel avait été ingesté des larves d'Aedes aegypti. On a étudié le moment de la mort des hôtes en relation des mues des larves, les emplacements principaux où se conduit l'infection fongale et le développement du fongus dans des tissus divers d'hôte. On a utilisé les conidiospores (CS) et les blastospores (BS) séparément pour traitement des larves des moustiques. Bien que dans la plupart des cas CS ait germiné et se soit développé chez l'hôte, en d'autres cas, CS a germiné mais n'avait pas produit du BS évidemment grâce à l'avortement prématuré du cycle de développement des fongus chez l'hôte. Le BS qui avait été ingesté dans les larves présentait des stages différents de développement du fongus dans les tissus des larves. Quoique les deux emplacements placements principaux étaient la tête et la région anale, l'emplacement préféré pour le développement du fongus était l'intestin des larves. Nous montrons qu'il n'y a plus de deux cycles de développement du fongus chez l'hôte. Bien que CS et BS soient efficaces comme larvicides, BS est de loin plus pathogène.
