Polycyclic aromatic hydrocarbons (PAHs) are one of the toxic persistent organic pollutants, have global environmental concern. Seed germination and early seedling development are the initial handicaps for plant establishment in phytoremediation program. Assisted phytoremediation by plant growth regulators may be a potential solution for these problems. Hence, we investigated the effects of seed treatment with various concentrations (0.01, 1.0, 100?nM) of 24-epibrassinolide (EBR) in tomato under graded levels (30, 100, 300???M) of a three-ring PAH namely phenanthrene (PHE). Delayed and decreased seed germination, reduced length and fresh weight (FW) of shoot and root were observed following 10?days of PHE exposure in a dose dependent manner. Chlorophyll fluorescence study suggested a possible photoinhibition and damage to photosynthetic apparatus under PHE exposure. However, seed treatment with EBR improved seed germination and increased length and FW of shoot and root. In addition, EBR remarkably restored the studied chlorophyll fluorescence parameters towards control levels. Different responses in antioxidant enzymes were observed following PHE exposure, while malondialdehyde (MDA) content was increased in a concentration dependent manner. EBR treatment prior to PHE exposure remarkably increased the activities of antioxidant enzymes over PHE alone, but decreased the MDA contents both in shoot and root of young tomato seedlings. Considering all the studied parameters, seed treatment with 1.0?nM EBR was most effective followed by 100 and 0.01?nM for the improvement of germination and seedling growth under PHE stress in tomato.  相似文献

Hydrogen peroxide (H2O2) promotes seed germination of cereal plants and ascorbic acid which acts as antioxidant suppresses the germination of wheat seeds, but the role of H2O2 scavenging on germination during seed maturation has not been demonstrated. We investigated relationship of germination, ascorbate, H2O2 scavenging enzymes and sensitivity to ascorbic acid (AsA) maturing seeds of two typical wheat (Triticum aestivum L.) cultivars, cvs. Shirogane-Komugi and Norin61. Shirogane-Komugi had marked high germination ability than Norin61 during seed maturation. Although the H2O2 content had no difference in the two culti-vars, sensitivity to AsA of Norin61 seeds was higher than that of Shirogane-Komugi seeds during seed maturation. The sensitivity to AsA closely correlated with germination characteristic in the two cultivars. Especially, at 28 days after pollination (DAP), sensitivity to AsA in Norin61 seeds was remarkably high. At that stage, no significant differences were observed in endogenous AsA level, ascorbate peroxidase (APX, EC 1.11.1.11) and dehydroascorbate reductase (DHAR, EC 1.8.5.1) activities in the two cultivars. However, catalase (CAT, EC 1.11.1.6) activity and CAT mRNA in Norin61 were remarkably higher than in Shirogane-Komugi. Sensitivity to AsA at 35 and 42 DAPs kept high levels in Norin61, and endogenous AsA and CAT activity in the seeds were significantly higher than in Shirogane-Komugi. These results revealed a direct correlation between germination and antioxidant sensitivity during the developmental stages of wheat seeds.Key words: ascorbic acid, germination, hydorogen peroxide, maturation, wheat seed  相似文献

Tea seed is believed to be recalcitrant based on its sensitivity to chilling or drying stress. Reactive oxygen species (ROS) and alterations in cytosolic redox status have been implicated in intolerance to desiccation by recalcitrant seed, but there is little information available regarding how ROS are regulated in seeds susceptible to drying stress. We investigated changes in protein expression and activity in tea embryo in response to desiccation using physiological and proteomic methods. Results showed that desiccation treatment dramatically induced the accumulation of H2O2 in tea embryos, accompanied by increased activities of antioxidant enzymes like ascorbate peroxidase (APX) and superoxide dismutase (SOD). Proteomic analyses also demonstrated that 23 proteins associated with defense response, metabolism and redox status were up-regulated following desiccation. Increase in antioxidants, ascorbic acid (AsA) and catalase (CAT) (H2O2 scavengers) partially assuaged desiccation damage to tea seed, resulting in improved germination rates. Higher accumulation of H2O2 aggravated desiccation damage to seeds leading to lower germination activity. We propose that desiccation causes an over-accumulation of ROS that are not efficiently scavenged by increased levels of antioxidant enzymes. High levels of ROS alter the redox status and are detrimental to seed viability. Reducing ROS to appropriate concentrations is an efficient way to reduce desiccation damage and improve germination rates of recalcitrant seeds.  相似文献

Hydrogen peroxide (H2O2), an active oxygen species, is widely generated in many biological systems and mediates various physiological and biochemical processes in plants. In this study, we demonstrated that exogenous H2O2 was able to improve the tolerance of wheat seedlings to salt stress. Treatments with exogenous H2O2 for 2 days significantly enhanced salt stress tolerance in wheat seedlings by decreasing the concentration of malondialdehyde (MDA), the production rate of superoxide radical (O2 ), and increasing the activities of superoxide dismutase (SOD), peroxidase (POD), catalase (CAT) and ascorbate peroxidase (APX), and the concentration of glutathione (GSH) and carotenoids (CAR). To further clarify the role of H2O2 in preventing salt stress damage, CAT and ascorbate (AsA), the specific H2O2 scavengers, were used. The promoting effect of exogenous H2O2 on salt stress could be reversed by the addition of CAT and AsA. It was suggested that exogenous H2O2 induced changes in MDA, O2 , antioxidant enzymes and antioxidant compounds were responsible for the increase in salt stress tolerance observed in the experiments. Therefore, H2O2 may participate in antioxidant enzymes and antioxidant compounds induced tolerance of wheat seedlings to salt stress. The results also showed that exogenous H2O2 had a positive physiological effect on the growth and development of salt-stressed seedlings.  相似文献

We induced an oxidative stress by means of exogenous hydrogen peroxide in two wheat genotypes, C 306 (tolerant to water stress) and Hira (susceptible to water stress), and investigated oxidative injury and changes in antioxidant enzymes activity. H2O2 treatment caused chlorophyll degradation, lipid peroxidation, decreased membrane stability and activity of nitrate reductase. Hydrogen peroxide increased the activity of antioxidant enzymes, glutathione reductase and catalase. These effects increased with increasing H2O2 concentrations. However, no change was observed in the activity of superoxide dismutase and proline accumulation.  相似文献

Reactive carbonyl species methylglyoxal (MG) has always been regarded as a cytotoxic metabolite, but now is emerging to function as signal molecule in plants. However, whether MG can induce salt tolerance is elusive. In this study, treatment of wheat seeds with NaCl reduced seed germination, plant height, root length, fresh weight, and dry weight, indicating the inhibitive effects of NaCl on seed germination and seedling growth. The inhibitive effects of NaCl were alleviated by applying exogenous MG, but aggravated by the MG scavenger N-acetyl-L-cysteine (NAC), suggesting that MG could induce the salt tolerance of wheat. In addition, MG increased glyoxalase I and glyoxalase II activities and decreased endogenous MG content in wheat seedlings under NaCl stress, whereas coapplication of NAC weakened glyoxalase activity and enhanced the endogenous MG level. Also, MG activated superoxide dismutase, catalase, ascorbate peroxidase, and glutathione reductase activities; increased glutathione and ascorbic acid levels; and decreased superoxide radical production and H2O2 and malondialdehyde contents under NaCl stress, while NAC reversed these physiological parameters. Furthermore, MG also induced the accumulation of proline, glycine betaine, and soluble sugar under NaCl stress, whereas this accumulation was weakened by NAC. This work reported for the first time that MG could induce the salt tolerance of wheat, and the acquisition of this salt tolerance was involved in the activation of the glyoxalase system and antioxidant system, as well as the accumulation of osmolytes.  相似文献

Proanthocyanidins (PAs) are the end products of the flavonoid biosynthetic pathway in many seeds, but their biological function is rarely unknown during seed germination. In the present study, we observed that PAs pretreatment accelerated cucumber seeds germination with maximum efficiency at 0.15% by measuring germination percentage and radical length. Using inhibitors of abscisic acid (ABA), gibberellins (GA) and alternative oxidase (AOX) and H2O2 scavenger pretreatment and gene expression analysis, we found that the accelerated effect of 0.15% PAs on seed germination was due to the decreased ABA biogenesis and enhanced GA production. ROS are induced by PAs pretreatment. Then, the enhanced ROS contributed to GA and ethylene accumulation and ABA decrease in seeds. Moreover, the improvement of GA was involved in the further induction of antioxidant enzymes activities. Therefore, our findings uncover a novel role of PAs in seed germination and clarify the relationships between ROS, ABA, GA and ethylene during seed germination.  相似文献

Magnesium (Mg) deficiency in plants is a widespread problem, affecting productivity and quality in agriculture. The mechanism of Mg deficiency inducing antioxidant enzyme activities has not been elucidated in rice. We examined the relationship among abscisic acid (ABA), H2O2, and antioxidant enzymes in the leaves of rice seedlings grown under conditions of Mg deficiency. The expression of OsRab16A, an ABA responsive gene, was used to determine the content of ABA. Mg deficiency resulted in increased ABA content in leaves of rice seedlings. The production of H2O2 was examined by 3,3-diaminobenzidine staining and a colorimetric method. Mg deficiency also induced H2O2 production in leaves, which was blocked by dipehnyleneiodonium chloride (DPI), an NADPH oxidase inhibitor. Tungstate (Tu), an ABA biosynthesis inhibitor, was effective in reducing Mg deficiency-increased ABA content, as well as Mg deficiency-induced H2O2 production. Both Tu and DPI were effective in reducing Mg deficiency-induced activities of superoxide dismutase, ascorbate peroxidase, glutathione reductase, and catalase in the leaves. Mg deficiency-induced ABA accumulation may trigger increased production of H2O2, which may involve plasma-membrane NADPH oxidase, and, in turn, up-regulates the activities of antioxidant enzymes in leaves of rice seedlings.  相似文献

The effect of exogenously applied hydrogen peroxide on salt stress tolerance was investigated in Panax ginseng. Pretreatment of ginseng seedlings with 100 μM H2O2 increased the physiological salt tolerance of the ginseng plant and was used as the optimum concentration to induce salt tolerance capacity. Treatment with exogenous H2O2 for 2 days significantly enhanced salt stress tolerance in ginseng seedlings by increasing the activities of ascorbate peroxidase, catalase and guaiacol peroxidase and by decreasing the concentrations of malondialdehyde (MDA) and endogenous H2O2 as well as the production rate of superoxide radical (O2 ?). There was a positive physiological effect on the growth and development of salt-stressed seedlings by exogenous H2O2 as measured by ginseng dry weight and both chlorophyll and carotenoid contents. Exogenous H2O2 induced changes in MDA, O2 ?, antioxidant enzymes and antioxidant compounds, which are responsible for increases in salt stress tolerance. Salt treatment caused drastic declines in ginseng growth and antioxidants levels; whereas, acclimation treatment with H2O2 allowed the ginseng seedlings to recover from salt stress by up-regulation of defense-related proteins such as antioxidant enzymes and antioxidant compounds.  相似文献

A hydroponics experiment was conducted to test the effects of sodium nitroprusside (SNP, a donor of NO) supplied with different concentrations on copper (Cu) toxicity in ryegrass seedlings (Lolium perenne L.). Excess Cu (200 µM) reduced chlorophyll content, resulting a decrease in photosynthesis. Cu stress induced the production of hydrogen peroxide (H2O2) and superoxide anion (O2? ?), leading to malondialdehyde (MDA) accumulation. Furthermore, activities of antioxidant enzymes in Cu-treated seedlings such as superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT) were decreased. In addition, Cu stress inhibited the uptake of K, Mg, Fe, and Zn and increased Ca content in roots. Moreover, in leaves of Cu-stressed seedlings, K, Fe, and Zn contents were decreased and the contents of Ca and Mg were not affected significantly. In Cu-treated seedlings, Cu concentration in roots was higher than in leaves. Addition of 50, 100, 200 µM SNP in Cu-mediated solutions increased chlorophyll content and photosynthesis, improved antioxidant enzyme activities, reduced Cu-induced oxidative damages, kept intracellular ion equilibrium under Cu stress, increased Cu concentration in roots and inhibited Cu accumulation in leaves. In particular, addition of 100 µM SNP had the best effect on promoting growth of ryegrass seedlings under Cu stress. However, the application of 400 µM SNP had no obvious alleviating effect on Cu toxicity in ryegrass seedlings.  相似文献

Wheat (Triticum aestivum L. cv. ‘Zyta’) seedlings were treated with 10, 100 and 200 μM Ni. Tissue Ni accumulation, length, relative water content (RWC), proline and H2O2 concentrations as well as the activities of superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APX), guaiacol peroxidase (POD) and glutathione S-transferase (GST) were studied in the shoots and roots after 6 days of Ni exposure. Treatment with Ni, except for its lowest concentration, resulted in a significant reduction in wheat growth. In comparison to the shoots, the roots showed greater inhibition of elongation, which corresponded with higher accumulation of Ni in these organs. Both shoots and roots responded to Ni application with a decrease in RWC and enhancement in proline concentration. Greater dehydration of the shoot tissue was accompanied by more intense accumulation of proline. Treatment of the wheat seedlings with the highest concentration of Ni led to about 60% increase in H2O2 concentration in both studied organs. Apart from CAT, constitutive activities of antioxidative enzymes were much higher in the roots than in the shoots. Exposure of the seedlings to Ni resulted in SOD activity decline, which was more marked in the roots. While the shoots showed a substantial decrease (up to 30%) in CAT activity, in the roots the activity of this enzyme remained unchanged. After Ni application APX, POD and GST activities increased several-fold in the shoots, whereas in the roots they were not significantly altered. The results suggest that differential antioxidative responses of the shoots and roots of wheat seedlings to Ni stress might be related to diverse constitutive levels of antioxidant enzyme activities in both organs.  相似文献

Antioxidant response of wheat roots to drought acclimation   总被引:1,自引:0,他引:1
Wheat (Triticum aestivum L.) seedlings of a drought-resistant cv. C306 were subjected to severe water deficit directly or through stress cycles of increasing intensity with intermittent recovery periods. The antioxidant defense in terms of redox metabolites and enzymes in root cells and mitochondria was examined in relation to membrane damage. Acclimated seedlings exhibited higher relative water content and were able to limit the accumulation of H2O2 and membrane damage during subsequent severe water stress conditions. This was due to systematic up-regulation of superoxide dismutase, ascorbate peroxidase (APX), catalase, peroxidases, and ascorbate–glutathione cycle components at both the whole cell level as well as in mitochondria. In contrast, direct exposure of severe water stress to non-acclimated seedlings caused greater water loss, excessive accumulation of H2O2 followed by elevated lipid peroxidation due to the poor antioxidant enzyme response particularly of APX, monodehydroascorbate reductase, dehydroascorbate reductase, glutathione reductase, and ascorbate–glutathione redox balance. Mitochondrial antioxidant defense was found to be better than the cellular defense in non-acclimated roots. Termination of stress followed by rewatering leads to a rapid enhancement in all the antioxidant defense components in non-acclimated roots, which suggested that the excess levels of H2O2 during severe water stress conditions might have inhibited or down-regulated the antioxidant enzymes. Hence, drought acclimation conferred enhanced tolerance toward oxidative stress in the root tissue of wheat seedlings due to both reactive oxygen species restriction and well-coordinated induction of antioxidant defense.  相似文献

Changes in the antioxidant defense system and proline accumulation were examined at different growth stages (vegetative, boot and reproductive) in plants of two hexaploid spring wheat cultivars (S-24, salt tolerant; MH-97, salt sensitive), grown in hydroponics and salinity-affected with 0, 50, 100 and 150 mM of NaCl. Salt stress provoked a marked decline in plant dry mass and ascorbic acid contents, and increased proline, total soluble proteins and H2O2 contents in both wheat cultivars at all growth stages. However, higher accumulation of proline and H2O2 contents was recorded at the vegetative and boot stages, respectively, in both wheat cultivars. Salt stress caused a consistent rise in the activities of some key antioxidant enzymes (CAT, SOD, POD, and APX) at all growth stages only in the salt tolerant cultivar S-24, whereas such pattern of enhanced activities of enzymatic antioxidants in cv. MH-97 was found only at the vegetative stage under saline regimes. Maximum activities of various enzymatic antioxidants were observed at the vegetative stage in both wheat cultivars under varying external salt treatments. The results showed that high salinity tolerance of cv. S-24, as manifested by lower decrease in its dry matter under salt stress, was associated with higher activities of antioxidant enzymes, increased accumulation of proline, and lower levels of H2O2, as compared with cv. MH-97 at all growth stages under saline regimes.  相似文献

This study aimed to investigate seed germination, seedling growth, and antioxidative responses in two wheat cultivars, Ningchun and Xihan, exposed to different H2O2 concentrations. Ningchun exhibited higher germination rate but lower root and shoot growth than Xihan when exposed to H2O2 treatment. Assays using fluorescent dye H2DCFDA and propidium iodide showed a significantly enhanced H2O2 content and a cell elongation inhibition in H2O2-treated roots. The malondialdehyde content was elevated with increasing exogenous H2O2 concentration. Moreover, treatments of seedlings with H2O2 scavenger, catalase (CAT), and antioxidant, butylated hydroxytoluene, partly abolished H2O2-induced negative effect on root growth. In both untreated and H2O2-treated leaves, SOD activity in Ningchun was higher than that in Xihan, but POD and APX activities in Ningchun were lower than those in Xihan, leading to elevated H2O2 level in Ningchun leaves but decreased H2O2 content in Xihan ones under H2O2 treatment.  相似文献

The aim of this study is to investigate the impacts of exogenous salicylic acid (SA) pretreatments on hydrogen peroxide (H2O2) accumulation, protein oxidation, and H2O2-scavenging enzymes in leaves of Cd-treated flax seedlings. Cd-enhanced H2O2 levels were related to increased activities of guaiacol peroxidase (POX, EC 1.11.1.7) and ascorbate peroxidase (APX, EC 1.11.1.11), and were independent of changes in catalase (CAT, EC 1.11.1.6) and superoxide dismutase (SOD, EC 1.15.1.1) activities. In control flax seedlings, exogenous SA pretreatments inhibited the activity of CAT, resulted in an enhanced production of H2O2 suggesting that SA requires H2O2 to initiate an oxidative stress. However, although leaves of Cd-free flax seedlings pretreated with SA accumulated in vivo H2O2 by 1.2-fold compared with leaves of Cd-only exposed ones; the damage to growth and proteins after the exposure to Cd was significantly less, indicating that SA can regulate the Cd-induced oxidative stress. Moreover, the Cd-treated seedlings primed with SA exhibited a higher level of total antioxidant capacities and increased activities of H2O2-detoxifying enzymes.  相似文献

To investigate the effects of ozone on winter wheat and flixweed under competition, two species were exposed to ambient, elevated and high [O3] for 30 days, planted singly or in mixculture. Eco-physiological responses were examined at different [O3] and fumigating time. Ozone reduced the contents of chlorophyll, increased the accumulation of H2O2 and malondialdehyde in both wheat and flixweed. The effects of competition on chlorophyll content of wheat emerged at elevated and high [O3], while that of flixweed emerged only at high [O3]. The increase of H2O2 and malondialdehyde of flixweed was less than that of wheat under the same condition. Antioxidant enzyme activities of wheat and flixweed were seriously depressed by perennial and serious treatment using O3. However, short-term and moderate fumigation increased the activities of SOD and POD of wheat, and CAT of flixweed. The expression levels of antioxidant enzymes related genes provided explanation for these results. Furthermore, the increase of CAT expression of flixweed was much higher than that of SOD and POD expression of wheat. Ozone and competition resulted in significant reductions in biomass and grain yield in both winter wheat and flixweed. However, the negative effects on flixweed were less than wheat. Our results demonstrated that winter wheat is more sensitive to O3 and competition than flixweed, providing valuable data for further investigation on responses of winter wheat to ozone pollution, in particular combined with species competition.  相似文献

Physiological and biochemical responses of wheat seedlings to drought, UV-B radiation, and combined stress were investigated. Drought, UV-B, and combined stresses retarded seedling growth by 26.5, 29.1, and 55.9%, respectively. One reason for growth retardation may be the oxidative damage indicated by an increase in the H2O2 content and lipid peroxidation degree. Furthermore, there was negative correlation between shoot fresh weight and H2O2 content, fresh weight and the content of thiobarbituric acid-reacting substances (TBARS), and the positive correlation between H2O2 content and TBARS (R 2 = 0.9251, 0.9005, and 0.9007, respectively). The activities of superoxide dismutase, guaiacol peroxidase, and ascorbate peroxidase increased under drought, UV-B, and the combination of stresses, while catalase activity decreased under the combined stress as compared to the control. The combination of drought and UV-B caused more severe damage to wheat seedlings than stress factors applied separately. Thus, the combined application of drought and UV-B had more strong adverse effects on wheat seedlings. The addition of 0.2 mM sodium nitroprusside (SNP) enhanced wheat seedling growth under drought, UV-B, and combined stress, likely, due to decreasing the accumulation of H2O2 and lipid peroxidation as well as activating the antioxidant enzymes. However, SNP treatment decreased the proline content. Published in Russian in Fiziologiya Rastenii, 2007, Vol. 54, No. 5, pp. 763–769. The text was submitted by the authors in English.  相似文献

The histochemical and cytochemical localization of abscisic acid (ABA)-induced H2O2 production in leaves of maize (Zea mays L.) plants were examined, using 3,3-diaminobenzidine (DAB) and CeCl3 staining, respectively, and the relationship between ABA-induced H2O2 production and ABA-induced subcellular activities of antioxidant enzymes was studied. H2O2 generated in response to ABA treatment was detected within 0.5 h in major veins of the leaves and maximized at about 2–4 h. In mesophyll and bundle sheath cells, ABA-induced H2O2 accumulation was observed only in apoplast, and the greatest accumulation occurred in the walls of mesophyll cells facing large intercellular spaces. Meanwhile, ABA treatment led to a significant increase in the activities of the leaf chloroplastic and cytosolic antioxidant enzymes superoxide dismutase (SOD), ascorbate peroxidase (APX) and glutathione reductase (GR), and pretreatment with the NADPH oxidase inhibitor diphenyleneiodonium (DPI), the O 2 scavenger Tiron and the H2O2 scavenger dimethylthiourea (DMTU) almost completely arrested the increase in the activities of these antioxidant enzymes. Our results indicate that the accumulation of apoplastic H2O2 is involved in the induction of the chloroplastic and cytosolic antioxidant enzymes. Moreover, an oxidative stress induced by paraquat (PQ), which generates O 2 and then H2O2 in chloroplasts, also up-regulated the activities of the chloroplastic and cytosolic antioxidant enzymes, and the up-regulation was blocked by the pretreatment with Tiron and DMTU. These data suggest that H2O2 produced at a specific cellular site could coordinate the activities of antioxidant enzymes in different subcellular compartments.  相似文献

The influence of cold hardening of rye (Secale cereale L.) and wheat (Triticum aestivum L.) seedlings on their resistance to the oxidative stress (OS) agents, namely, 50 mM hydrogen peroxide or 5 mM iron (II) sulfate was studied. Unhardened rye seedlings were more resistant to hydrogen peroxide than those of wheat, since their growth was less inhibited, and they accumulated lesser amounts of lipid peroxidation products after a treatment with H2O2. The interspecific differences in responses to FeSO4 were less significant. The unhardened seedlings of rye, in comparison with those of wheat, possessed more active guaiacol peroxidase (GPO) and more levels of anthocyanins and proline. In response to the OS agents, the unhardened rye seedlings enhanced activities of superoxide dismutase and catalase, whereas the wheat seedlings enhanced GPO activity and proline content. The cold hardening (6 days at 2°C) increased activities of antioxidant (AO) enzymes, contents of proline, sugars, and anthocyanins in seedlings of both species, and made the seedlings more resistant to the OS agents. After the cold hardening, rye seedlings were more resistant to OS than wheat seedlings. The hardened seedlings of both species activated the AO enzymes in response to H2O2 or FeSO4 greater than the unhardened ones. However, the hardened wheat seedlings, in contrast to the unhardened ones, did not augment the proline content in contact with the OS agents. The conclusion was drawn on different contributions of AO enzymes and low-molecular weight compounds to the basal and induced by the cold—hardening resistances of rye and wheat seedlings to OS.  相似文献
