The effect of exposure to copper in seawater (0.005, 0.01, and 0.02 mg/liter) on the development of Mytilus trossulus was examined in the stages of fertilization, blastula, early veliger, veliger, and veliconch. Copper in a concentration of 0.01 and 0.02 mg/liter inhibited the development and growth of the embryos and larvae. At 0.005 mg Cu/liter, the embryos and larvae were capable of adaptation. If M. trossulus embryos and larvae were maintained for 1–2 days in seawater containing 0.01 mg Cu/liter and then transferred to clean water, the consequences were largely dependent on the developmental stage at which the exposure to copper took place. We examined the effects of various protease inhibitors on Xenopus laevis embryogenesis. Aprotinin, a serine protease inhibitor, was found to inhibit embryogenesis markedly, but other protease inhibitors had virtually no effect. The inhibitory effect of aprotinin was specific for embryos at the blastula or gastrula stage. These results suggest that an aprotinin-sensitive protease involved in embryonic development is secreted from the embryos or appears on the surface of embryonic cells at these stages. We found that various serine proteases are in fact secreted from the embryos with their development and that some of them are sensitive to aprotinin.  相似文献

Using Drosophila melanogaster, the relative sensitivities of various larval stages to the toxic effects of growth on media supplemented with either 0.44 or 0.88 ppm aflatoxin B1 (AFB1) were determined. Two strains of fruit flies were tested: strain A-11 which is relatively resistant to AFB1 induced toxicity, and strain A-9 which is quite sensitive. Eggs, mid-first, mid-second and early-, mid- and late-third instar larvae were transferred onto AFB1 media and allowed to complete larval and pupal development and eclose as adults. At the 0.44 ppm concentration, strain A-11 showed no effect, while only first instar larvae of strain A-9 showed significant mortality rates for first instar larvae, but the A-9 larvae die at higher rates than the A-11 larvae. In addition, second and third instar larvae of strain A-9 show significant mortality rates when grown at 0.88 ppm AFB1, while these stages are not affected in strain A-11.  相似文献

以小麦为供试材料,分别采用发芽试验、溶液培养、土柱栽培等毒理试验方法,研究了小麦萌芽期、幼苗期及成株期受重金属污染毒害的指标体系。结果表明,①小麦根伸长抑制率可作为萌芽期重金属污染评价的一项生物指标;Cd、Cr、Pb对小麦根生长的7d半效应浓度(EC50)值分别为1.39、0.20mmo.lL-1和2.75mmol.L-1,据此得到3种重金属对小麦同一性状的毒性次序为CrCdPb;此外各性状抑制率与胁迫浓度的关系可用双曲线模型y=x/(a+x/100)或指数曲线模型y=a(1-exp(-bx))较好模拟。②Cd、Cr、Pb单一污染胁迫对小麦幼苗性状的毒性次序均以影响叶面积和冠部干重为主,表明叶面积和冠部干重为幼苗期污染监测的敏感指标;重金属对除根干重外所有性状的毒性次序:CrCdPb,该毒性次序是根据引起50%抑制的临界浓度即EC50值确定的;各性状抑制率与胁迫浓度的关系可用直线模型、或双曲线模型、或指数曲线模型较好模拟。③重金属单一污染胁迫造成小麦籽粒产量下降幅度是CdCrPb;二元互作对产量影响的重要性次序为CdCrCdPbCrPb;在Cd、Cr、Pb复合污染胁迫条件下,穗数可以作为成株期重金属污染监测的首选指标。以上指标可为农田小麦重金属污染的监测和综合治理提供一些理论依据。  相似文献

Somatic embryogenesis is an important biotechnological tool that has significant potential to be used in studies related to environmental stress. In this study, embryogenic cell masses of Abies alba and Picea abies were grown on media enriched with 50–500 µM cadmium (Cd2+) or lead (Pb2+). The effects of cadmium and lead were evaluated during the subsequent stages of somatic embryogenesis: proliferation, maturation, and germination. The following characteristics were evaluated: proliferation potential, cell viability, average number of somatic embryos obtained per 1 g of fresh weight, and morphology of the developed somatic embryos. The tested heavy metals significantly reduced the proliferation rate of A. alba and P. abies embryogenic cell masses. The highest tested cadmium concentration markedly slowed or stopped the growth of embryogenic cell masses in both species. Unexpectedly, the proliferation ratio remained fairly high for the P. abies cell lines treated with lead at all concentrations tested. During the maturation stage, the total number of somatic embryos declined under cadmium exposure. The formation of early precotyledonary and cotyledonary somatic embryos in both species was similarly reduced, although cadmium caused a higher death rate and was more toxic than lead. To the best of our knowledge, this is the first report to study the effects of heavy metals on A. alba embryogenic cell masses during the proliferation stage as well as on the maturation and germination processes of both species. This in vitro system can be used for testing the response of large sets of genotypes, and the best performing lines can be used in the future for in vivo performance tests of heavy metal-polluted soils.  相似文献

A study was made of acute toxic effects of Cd+, Cu+, Al+, Pb+, Ni+, Mn+ and Zn+ ions on embryos, juveniles, and adults of Asellus aquaticus L. at 19 and 29 degrees C, when examined in different seasons of the same year. The highest sensitivity to heavy metals was displayed by embryos. The metal sensitivity commonly decreases with age, however, during hormonal rearrangements associated with reproduction this was seen to increase again. For all age groups the heavy metal toxicity increases with increased temperature rises. The importance of a more strict control of water quality in the system of lakes and rivers is suggested, especially in spring and summer time when the mass reproduction of hydrobionts occurs.  相似文献

本文研究了温度、盐度和重金属等几种因子对缢蛏和长牡蛎幼体趋地性和趋光性行为的影响。结果表明:1)降低温度能增强缢蛏幼体的正趋地性,而长牡蛎幼体却由正趋地性转变为负趋地性,同时两种幼体的正趋光性受到抑制作用;升高温度对缢蛏幼体趋光性的影响不明显,但能加强牡蛎幼体的正趋光性。2)盐度提高时缢蛏幼体的正趋光性趋于增强,正趋地性趋于减弱。3)重金属Cu和Zn在各自的亚致死浓度下具有提高缢蛏幼体正趋地性和降低正趋光性的作用,而牡蛎幼体的正趋地性却趋于减弱,正趋光性趋于增强。本文据上述幼体的行为变化,还讨论了这几种因子对幼体垂直分布的调节作用。  相似文献

The effects of water temperatures of 7, 15 and 20°C and pollution of sea water by copper ions in concentrations of 0.005 and 0.01 mg/l on the early development of the Pacific mussel Mytilus trossulus were studied. It was shown that copper additives were most toxic at the initial stages of development, from fertilization up to the veliger stage. It was determined that larvae of the Pacific mussel were able to adapt to copper at a concentration of 0.005 mg/l. The copper toxicity increased to a greater extent with an increase in water temperature from its average value of 15°C, than with a temperature decrease.  相似文献

Cocaine is used by over 20% of women of reproductive age. Although there have been numerous studies focusing on its effects on reproductive processes, none has evaluated its direct effect on preimplantation development. We have investigated the effect of cocaine and its major metabolite, benzoylecgonine, on in vitro preimplantation mouse embryogenesis. One-cell embryos were exposed at the one-, two-, four-, or eight-cell stage for 24 hr to medium containing 0-400 micrograms/ml cocaine or benzoylecgonine and then cultured to the blastocyst stage. Cocaine had its strongest inhibitory effect at the earliest stages of development. At the one- and two-cell stages, there was a significant inhibition of blastocyst formation following exposure to cocaine concentrations of 25-400 micrograms/ml, and at the four-cell stage there was an inhibitory effect at 100 and 400 micrograms/ml cocaine. Benzoylecgonine inhibited the development of embryos to blastocyst only at the one- and two-cell stages, at concentrations of 100-400 micrograms/ml. These findings suggest that cocaine is capable of blocking preimplantation embryogenesis, particularly following exposure at the earliest stages, and that this toxicity may abate as cocaine is biotransformed to benzoylecgonine.  相似文献

The sensitivity of Spodoptera littoralis second and fourth instar larvae to the heavy metals cadmium (Cd) and lead (Pb) was investigated under laboratory conditions. Both Cd and Pb, even, at very low concentrations, had relatively high toxic effects on S. littoralis larvae. Cd was more toxic to S. littoralis larvae than Pb. In addition, the mean percentage feeding deterrence (FDI%) of Cd and Pb was concentration-dependent. The nutritional indices (consumption index, growth rate, efficiency of converted ingested and digested food, and approximate digestibility) of S. littoralis second and fourth instar larvae were more affected in Cd-treatments compared with those of Pb-treatments. Therefore, the presence of such heavy metals in the environment would exert an adverse impact on S. littoralis larvae and their development.  相似文献

The unsolved problem of cryopreservation of the yolk-rich teleost embryos may be related, in part, to their sensitivity to chilling and cryoprotective agents. The aim of this study was to gain data on the sensitivity of carp embryos to low temperatures at different developmental stages and on the possible protective and toxic effects of cryoprotectants. A total of 86,400 morulae, half-epiboly and heartbeat-stage embryos was selected and then placed in water or in 1 M methanol, dimethyl sulfoxide (Me2SO), glycerol or 0.1 M sucrose solution at 0, 4 or 24 degrees C for 5 min or 1 h. Following these treatments, the embryos were held in a 24 degrees C water bath until the evaluation of hatching rates. In every developmental stage a significant decrease of hatching rates following exposure to 4 or 0 degree C was detected. Sensitivity to chilling changed significantly with development (heartbeat < morula < half-epiboly). Half-epiboly stage embryos were less sensitive to a short period of exposure to cryoprotectants than morula and heartbeat stages. A 1-h exposure to cryoprotectants revealed a stage dependent sensitivity. Toxicity increased in the order of methanol < Me2SO < glycerol in morula and half-epiboly stages, and methanol < glycerol < Me2SO in the heartbeat stage. The results show morulae are partially protected against chilling in Me2SO and sucrose, half-epiboly in Me2SO, sucrose and methanol, and heartbeat-stage in methanol and glycerol. The results further suggest that carp embryos are sensitive to chilling and that toxicity and protective effects against chilling of cryoprotectants are stage-dependent. The finding on the low chilling sensitivity of heartbeat-stage embryos and the protective effect of certain cryoprotectants may be useful in designing cryopreservation protocols.  相似文献

The toxic effects of dissolved versus bioconcentrated tributyl tin (TBT) on oyster larvae were compared. Water column TBT levels, which had no effect in solution, inhibited natural attachment and metamorphosis of oyster larvae on bottom surfaces due to bioconcentration by biofilms. This study investigated the toxicity of zinc pyrithione (Zpt) on the early stages of development of the ascidian Ciona intestinalis. Larval morphological abnormalities were studied after the exposure of C. intestinalis embryos at different stages of development. The median effective concentrations (EC50) ranged from 226-590 nM. The larval settlement stage was the most sensitive to Zpt. Toxic effects of Zpt on larval settlement were detected at 9 nM (EC10). The inhibition of C. intestinalis embryonic development was also used to study the loss of toxicity in Zpt solutions exposed to direct sunlight and laboratory UV light. The results showed that the toxicity of Zpt solutions decreased but did not disappear after 4 h exposure to direct sunlight (EC50 = 484 nM) or UV light (EC50 = 453 nM), compared to control Zpt solutions prepared in dark conditions. On the basis of the present data, predicted no effect concentrations of Zpt to C. intestinalis larvae are lower than predicted environmental concentrations of Zpt in certain polluted areas and therefore, may pose a risk to C. intestinalis populations.  相似文献

含镉量子点是典型的量子点,近年来受到广泛研究。含镉量子点的潜在毒性是其在生物成像及生物医药方面应用和发展的关键制约因素,因此,对其毒性作用的研究具有重要意义。目前对含镉量子点的体外毒性研究主要集中在人肝癌细胞(HepG2)、神经分泌细胞(PC12)等细胞实验及斑马鱼胚胎体外培养实验。体内毒性研究包括小鼠等动物实验。这些研究证实,量子点对HepG2等细胞系和小鼠、贻贝等动物均具细胞毒性。研究者们普遍认为,量子点是通过释放其组成中的重金属,诱导生物体产生活性氧自由基,进而引发细胞凋亡或自噬,但对量子点的具体毒性作用机制并不完全清楚。该文对含镉量子点的体内和体外毒性研究工作进展进行了综述,包括含镉量子点对肝肾细胞、神经细胞、血液细胞及免疫细胞等体外毒性研究工作,对陆生及水生动物等的体内毒性研究工作,旨在更好、更全面地评估含镉量子点的毒性,为今后对量子点的毒性作用机制研究提供方向,促进含镉量子点在生物医学方面的发展和应用。  相似文献

The effects of toxic boron (B) concentrations on the growth of wheat genotypes at organ level and cellular level were investigated using excised root culture techniques. At the organ level, wheat genotypes differed for root elongation and lateral root development in response to toxic B concentrations. Genotypes classified as resistant from field studies had longer root axes and more lateral roots than sensitive or moderately sensitive genotypes, but there was no difference in axis elongation between sensitive and moderately sensitive genotypes. At the cellular level, callus production of root explants among genotypes still different at toxic B concentrations: resistant genotypes could produce more callus than sensitive or moderately sensitive genotypes. These results suggest that differences among genotypes in resistance to toxic B concentrations may be related to cell membrane permeability to B since they were also expressed in undifferentiated callus cells. The distinct and consistent differences among genotypes in response to B toxicity both at the organ level and at the cellular level could serve as a basis for selection in a breeding program. Vitellogenins (Vtgs) are the major yolk proteins in all oviparous animals. Systematic and regulated processing of these during embryogenesis is crucial for embryonic development. In the present study, toxicant-induced disturbance of Vtg degradation processes during Danio rerio (DR) embryogenesis was analysed to establish a sensitive tool for monitoring toxic stress at the molecular level. A 2-DE-based proteomic approach for whole DR embryos was established to study Vtg cleavage products (lipovitellin (Lv) derivatives). Ethanol was chosen as a positive control for a toxicity related change in the proteome of whole zebra fish embryos. Protein extracts from embryos treated with two ethanol concentrations, 0.5 and 2% v/v, showing either no or very strong visible effects, like absent heartbeat and blood circulation, were examined. Significant changes in the Lv pattern were detected for both conditions. The results are interpreted as scope for the use of the high abundant Lv derivatives as sensitive stress indicators in zebra fish embryos reflecting the overall fitness of the intact organisms.  相似文献

Although estrogens have been detected in some echinoderm species, their role is not clearly understood; so we examined the effects of estrogens administered to sea urchin embryos and larvae. A typical malformation was exogastrulation, induced by the exposure to ethynylestradiol (EER) in a defined period of 12 h from 12 h after fertilization (HAF). Morphogenesis for gastrulation was delayed in the treated embryos: protrusion of the archenteron started at 30 HAF when gastrulation had already finished in normal embryos. Exogastrulation induced by EER was cancelled by the antiestrogen chemical, ICI182,780. Feeding larvae were less sensitive to estrogens than those in early embryogenesis and, at certain concentrations, developed without abnormal morphology. The effect of estrogens was examined at the level of gene expression of the major yolk protein (MYP). MYP expression started during the larval stage and was suppressed by estrone at the six-armed stage, but not by β-estradiol, and in later stage larvae, the expression was not affected by treatment with either estrogen. Estrogens affect sea urchins in the early stage of embryogenesis, leading to abnormal morphogenesis and interference with gene expression.  相似文献
