We report here on a series of studies aimed at characterization of the structural and dynamical properties of the synthetic lipid diphytanoyl phosphatidylcholine, in multilamellar dispersions and vesicle suspensions.This lipid exhibits no detectable gel to liquid crystalline phase transition over a large temperature range (?120°C to +120°C).Examination of proton nuclear magnetic resonance (NMR) free induction decays obtained from multilayer dispersions of diphytanoyl phosphatidylcholine provided an estimate of the methylene proton order parameter. The estimated magnitude of 0.21 is comparable to those determined for other phospholipids.Sonication of aqueous dispersions of diphytanoyl phosphatidylcholine led to formation of bilayer vesicles as determined by the measurement of the outer/inner choline methyl proton resonances, vesicle sizes in electron micrographs, and comparison of proton NMR linewidths between multilayer and sonicated dispersions. Ultracentrifugation studies of diphytanoyl phosphatidylcholine vesicles in H2O and 2H2O media yielded a value of 1.013 ± 0.026 ml/g for the partial specific volume of this lipid.We have measured spin lattice relaxation rates for the methyl and methylenemethyne protons of the hydrocarbon chains of diphytanoyl phosphatidylcholine in bilayer vesicles over a range of temperatures and at two NMR frequencies (100 and 220 MHz). The observed relaxation rates for the methylene protons in this system were approximately twice those previously reported for dipalmitoyl phosphatidylcholine at comparable temperatures and resonance frequencies, whereas the relaxation rates measured for the methyl protons were greater than those of the straight chain lipid by an order of magnitude.Measurement of the spin lattice relaxation rates of the hydrocarbon protons of the diphytanoyl phosphatidylcholine in a 10 mol% mixture of the branched-chain lipid in a deuterated host lipid, diperdeuteropalmitoyl phosphatidylcholine, showed a discontinuity in the temperature dependence of the proton NMR longitudinal relaxation rates of the branched-chain lipid in the region of the gel to liquid crystalline phase transition temperature of the deuterated dipalmitoyl phosphatidylcholine host lipid. This result may be taken as evidence of lateral phase separation of a liquid cyrstalline phase enriched in diphytanoyl phosphatidylcholine from a gel phase enriched in diperdeuteropalmitoyl phosphatidylcholine at temperatures below the phase transition temperature of deuterated host lipid. This conclusion is supported by the observation of an abrupt change in the hydrocarbon methylene linewidth (at 100 MHz) of 10 mol% diphytanoyl phosphatidylcholine in diperdeuteropalmitoyl phosphatidylcholine over the temperature range where lateral phase separation is taking place according to differential thermograms.  相似文献

The association of water (D2O) with phospholipid membranes was studied by using pulsed-electron spin resonance techniques. We measured the deuterium electron spin echo modulation of spin-labeled phospholipids by D2O in membranes of dipalmitoyl phosphatidylcholine with and without 50 mol% of cholesterol. The Fourier transform of the relaxation-corrected two-pulse echo decay curve reveals peaks, at one and two times the deuterium NMR frequency, that arise from the dipolar hyperfine interaction of the deuterium nucleus with the unpaired electron spin of the nitroxide-labeled lipid. For phosphatidylcholine spin-labeled at different positions down the sn-2 chain, the amplitude of the deuterium signal decreases toward the center of the membrane, and is reduced to zero from the C-12 atom position onward. At chain positions C-5 and C-7 closer to the phospholipid headgroups, the amplitude of the deuterium signal is greater in the presence of cholesterol than in its absence. These results are in good agreement with more indirect measurements of the transmembrane polarity profile that are based on the 14N-hyperfine splittings in the conventional continuous-wave electron spin resonance spectrum.  相似文献

Activation of the first component of human complement (C1) by bilayer-embedded nitroxide spin label lipid haptens and specific rabbit antinitroxide antibody has been measured. The nitroxide spin label hapten was contained in host bilayers of either dimyristoyl phosphatidylcholine or dipalmitoyl phosphatidylcholine in the form of both liposomes and vesicles. At a temperature of 32 degrees C, which is intermediate between the hydrocarbon chain-melting temperatures of the two phospholipids, activation of C1 in such vesicles and liposomes is more efficient in the fluid membrane. Studies of C1 activation in binary mixtures of cholesterol and dipalmitoyl phosphatidylcholine indicate that the activation of C1 is not limited by the lateral diffusion of the lipid haptens in these membranes.  相似文献

The adsorption of human serum albumin (HSA) to dipalmitoyl phosphatidylcholine (DPPC) bilayer membranes containing poly(ethylene glycol)-grafted dipalmitoyl phosphatidylethanolamine (PEG-DPPE) was studied as a function of content and headgroup size of the polymer lipid. In the absence of protein, conversion from the low-density mushroom regime to the high-density brush regime of polymer-lipid content is detected by the change in ESR outer hyperfine splitting, 2Amax, of chain spin-labelled phosphatidylcholine in gel-phase membranes. The values of 2Amax remain constant in the mushroom regime, but decrease on entering the brush regime. Conversion between the two regimes occurs at mole fractions XPEG(mb)≈0.04, 0.01-0.02 and 0.005-0.01 for PEG-DPPE with mean PEG molecular masses of 350, 2000 and 5000 Da, respectively, as expected theoretically. Adsorption of HSA to DPPC membranes is detected as a decrease of the spin label 2Amax hyperfine splitting in the gel phase. Saturation is obtained at a protein/lipid ratio of ca. 1:1 w/w. In the presence of polymer-grafted lipids, HSA adsorbs to DPPC membranes only in the mushroom regime, irrespective of polymer length. In the brush regime, the spin-label values of 2Amax are unchanged in the presence of protein. Even in the mushroom regime, protein adsorption progressively becomes strongly attenuated as a result of the steric stabilization exerted by the polymer lipid. These results are in agreement with theoretical estimates of the lateral pressure exerted by the grafted polymer in the brush and mushroom regimes, respectively.  相似文献

The temperature and hydration dependences of lipid lateral diffusion in model membrane/D2O multilayers of dipalmitoyl (DPL), dilauryl (DLL) and egg yolk (EYPC) lecithins were measured using pulsed gradient proton nuclear magnetic resonance (NMR) spin echo techniques. Oriented samples were used to minimize anisotropic dipolar interactions and permit formation of a spin echo. Significantly lipid lateral diffusion is hydration dependent over the range studied (15–40% D2O w/w), varying in DPL over this range for example by a factor of 2. For the saturated lipids at the same hydration and temperature, diffusion decreases monotonically as the chain length increases. The results tend to be larger, by factors of 2–5, than the earlier electron spin resonance (ESR) spin label results, the differences being attributable in part to the differences in hydration and to the absence of probe effects in this work. The addition of cholesterol (28.6 mol%) decreases diffusion of the lipids. Comparisons with other methods of lateral diffusion measurements are made.  相似文献

In order to clarify, in dipalmitoyl phosphatidylcholine multilayers, the effect of the 34 degrees C thermal pretransition on the acyl chain intramolecular disordering process, Raman spectra of dipalmitoyl phosphatidylcholine gels at 20 and 34 degrees C were compared in the 1000--1200 cm-1 skeletal C-C stretching region. In addition to an overall intensity decrease associated with a change in chain packing characteristics, the growth of intensity in the 1080--1090 and 1122 cm-1 regions in the 34--20 degrees C) difference spectrum clearly indicates that the thermal pretransition is accompanied by an increase in the population of hydrocarbon chain gauche rotamers toward the center of the bilayer.  相似文献

The now well-established use of Raman spectroscopy to examine the structure of biomembranes is extended through an examination of the origins of the structure-sensitive features of phospholipid spectra and the development of quantitative order-parameters. One parameter gives a quantitative measure of the fraction of all-trans bonds in the hydrocarbon chains while the other provides a semiquantitative estimate of the lateral crystal-like order between the chains. The parameters are used to study the differences between vesicles and dispersions of dipalmitoyl phosphotidylcholine, dimyristoylcholine and egg lecithin. We find that the vesicles of dipalmitoyl phosphotidylcholine are substantially less ordered than the dispersions in terms of both longitudinal and lateral order which are greatly decreased. A very careful measurement of the order as a function of temperature shows that there is a pre-melting transition in the dispersions of dipalmitoyl phosphotidylcholine which does not exist in the vesicles. Remarkable agreement is obtained between the Raman technique and that previously reported by calorimetric measurements and theoretical calculations.  相似文献

The proportionality constant between the equipotency concentrations of a series of solutes and the fraction of a solute in the membrane phase is directly related to the solute to lipid mol ratio. Experimental measurements of partition coefficient and of several alkanol-induced effects show that the solute/lipid mol ratlos for a series of alkanols are not constant at their equipotency concentrations. The deviations in the solute/lipid ratios are similar in the various systems, and these deviations seem to depend primarily upon the chain length and branching in alkanols. It is suggested that such intrinsic differences in the perturbing ability of alcohols arise from a specificity of interaction between alkanols and lipid bilayer. We have correlated partition coefficients (in n-octanol, in egg phosphatidylcholine liposomes, and in dipalmitoyl phosphatidylcholine liposomes) for thirteen alkanols to the equipotency concentrations for their ability to modify the order-disorder thermotropic transition in dipalmitoyl phosphatidylcholine, ability to stimulate the hydrolysis of phosphatidylcholine in a bilayer by bee venom phospholipase A2, and for the activation of the galactoside transport system in Escherichia coli. Significant correlation is found between equipotency concentrations for perturbing the order-disorder transition, the activation of phospholipase A2-catalyzed hydrolysis and the activation of galactoside transport system.  相似文献

The statistical mechanical cluster theory of Fisher as applied by Kanehisa and Tsong to phospholipid bilayers is modified to describe the effects of hydrostatic pressure on the state of an aqueous dispersion of the phospholipid dipalmitoyl phosphatidylcholine. A high pressure Raman scattering cell has been built to obtain the Raman spectra of aqueous dispersions of phospholipids as a function of the applied hydrostatic pressure from 0 to 100 atmospheres. Predicted thermal and pressure-induced phase transitions are compared with an experimentally obtained Raman order parameter derived from the ratio of two bands in the C-H stretching region of the Raman spectrum of the sample. The parameters of the theory are adjusted to obtain a satisfactory fit of the Raman order parameter versus temperature. The theory is then found to give an excellent prediction of the observed pressure dependence of the Raman order parameter with no changes in the adjustable parameters. The implications of the success of the theoretical fit is discussed. Particularly of interest is the rather high value of the critical temperature, Tc, for lipid bilayers which is predicted by the model.  相似文献

Raman spectroscopic frequency differences between selected carbon-carbon stretching modes of lipid hydrocarbon chains were determined as a function of temperature for use in monitoring lipid phase transition behavior and acyl chain disorder in both multilamellar and single-wall vesicles. Transition temperatues detected by this procedure for pure dipalmitoyl phosphatidylcholine and dimyristoyl phosphatidylcholine multilayers were observed at 39±1 °C and 23±1 °C, respectively. Although the phase transition for unilamellar vesicles of dipalmitoyl phosphatidylcholine occurred at nearly the same temperature as the multilayers, the crystal-liquid crystalline transition for the single-shell vesicles appeared to span a slightly broader temperature range, a characteristic consistent with irregularities in the packing arrangement of the hydrocarbon chains. Within the precision of the Raman spectroscopic method, however, the temperature behavior of both the multilamellar and the unilamellar dimyristoyl phosphatidylcholine assemblies appeared nearly identical. The temperature profile for the Raman frequency differences of an excess water sonicate of 25 mol percent cholesterol in dipalmitoyl phosphatidylcholine served as an example of the effect upon lipid phase transition characteristics of a bilayer component intercalated between the acyl chains. For this particular cholesterol-lipid system the phase transition was broadened over a 30 °C temperature range, in contrast to the narrow 5?4 °C range observed for pure multilayer and single-shell vesicle particles.  相似文献

Computer simulation analysis of the structure sensitive features in the Raman spectrum of aqueous dipalmitoyl phosphatidylcholine (DPPC) multilayers was made for the 1000–1200 cm−1 region. The composite triplet was resolved into 4 Lorentzian bands with the possibility to follow their parameters (amplitude, width and position) in the temperature interval 25–60°C. The analysis reveals the apparent inward shift of the 1130 and 1065 cm−1 bands towards both sides of the broad intense 1087 cm−1 feature which is due to the relative changes in amplitudes of these bands in the course of the lipid phase transition. The more accurate method for the evaluation of the trans order parameter (Strans) confirms the underestimation of the trans segment content above the phase transition temperature when using both the relative amplitude and area of the 1130 cm−1 band for the quantitative characterization of chain conformation. Calculated changes in the bilayer thickness following the frequency shift of the 1100 cm−1 band are in good agreement with the previously reported deuterium NMR data and X-ray diffraction studies.  相似文献

Multilamellar spin labelled liposomes were prepared from dipalmitoyl or dimyristoyl phosphatidylcholine, dicetyl phosphate, and the spin probe 12-doxyl stearate methyl ester. The effects of a series of benzene and adamantane derivatives, on fatty acyl chain motion was measured through changes in the electron spin resonance spectra of these liposomes. All the compounds tested, increased lipid chain motion to a variable degree. In general, molecules possessing a polar group were more potent than those lacking such a group and lipophilicity per se correlated poorly with the relative order of these compounds. Within the adamantane series separating the polar group from the cage structure by the insertion of methylene groups further enhanced the capacity of the molecule to increase hydrocarbon chain mobility. These observations are consistent with the hypothesis that the location of the additive within the bilayer is the main determinant of its effectiveness in increasing fatty acyl chain motion.  相似文献

The temperature dependence of the Raman spectral transitions assigned to the acyl chain C-C stretching modes of dipalmitoyl phosphatidylcholine was determined for the gel, phase transition and liquid crystalline states of the lipid multilayers. The van't Hoff enthalpy differences ΔHVH between trans and gauche rotational isomers were obtained from the Raman spectral data for the temperature region characteristics of each bilayer state. An average size for the cooperative unit undergoing the chain melting process during the phase transition was estimated from the ratio of the appropriate van't Hoff enthalpy to an adjusted calorimetric enthalpy.  相似文献

The interaction of antidepressant drug trazodone hydrochloride (TRZ) with dipalmitoyl phosphatidylcholine (DPPC) multilamellar liposomes (MLVs) in the presence and absence of cholesterol (CHO) was investigated as a function of temperature by using Electron Paramagnetic Resonance (EPR) spin labeling, Fourier Transform Infrared (FTIR) Spectroscopy and Differential Scanning Calorimetry (DSC) techniques. These interactions were also examined for dimyristoyl phosphatidylcholine (DMPC) multilamellar liposomes by using Electron Paramagnetic Resonance (EPR) spin labeling technique. In the EPR spin labeling studies, 5- and 16-doxyl stearic acid (5-DS and 16-DS) spin labels were used to monitor the head group and alkyl chain region of phospholipids respectively. The results indicated that TRZ incorporation causes changes in the physical properties of PC liposomes by decreasing the main phase transition temperature, abolishing the pre-transition, broadening the phase transition profile, and disordering the system around the head group region. The interaction of TRZ with unilamellar (LUV) DPPC liposomes was also examined. The most pronounced effect of TRZ on DPPC LUVs was observed as the further decrease of main phase transition temperature in comparison with DPPC MLVs. The mentioned changes in lipid structure and dynamics caused by TRZ may modulate the biophysical activity of membrane associated receptors and in turn the pharmacological action of TRZ.  相似文献

Spin lattice relaxation times in both the lab and rotating frame, have been measured for deuterons (2H) in a number of unsonicated dispersions of 1,2 dipalmitoyl phosphatidylcholine in D2O over a range of resonant frequencies from 13 MHz to 1 MHz for temperatures from ?20°C to 65°C.The proton (1H) spin lattice relaxation time for the lecithin was measured for resonant frequencies of 8.5 MHz, and 40 MHz over a similar range of temperatures.The results agree with broadline measurements by Salsbury et al. [1], and for the liquid crystal phase are consistent with an anisotropic tumbling model of the water molecules bound to the lecithin headgroup. This tumbling occurs with correlation times of ≤10?10 sec and ≈ 10?6 sec about axes parallel to and perpendicular to the bisector of the D-O-D angle within a D2O molecule, hydrogen bonded to the negatively charged phosphate headgroup.  相似文献

Approximate phase diagrams describing lateral phase separations are given for binary mixtures of dimyristoyl phosphatidylcholine with dipalmitoyl phosphatidylcholine, distearoyl phosphatidylcholine, and dipalmitoyl phosphatidylethanolamine. These diagrams are based in part on freeze-fracture electron microscopic data presented here, as well as other earlier spin-label, calorimetric, and X-ray data. These phase diagrams represent an improvement over previous studies in that both solid phases (Pβ' and Lβ') of the phosphatidylcholines are included. Further consideration is given to the problem of binary mixtures in which there are two Pβ' phases that do not form a continuous range of solid solutions.  相似文献

We used wide angle x-ray scattering (WAXS) from stacks of oriented lipid bilayers to measure chain orientational order parameters and lipid areas in model membranes consisting of mixtures of 1,2-dioleoyl-sn-glycero-3-phosphocholine (DOPC)/cholesterol and 1,2-dipalmitoyl-sn-glycero-3-phosphocholine (DPPC)/cholesterol in fluid phases. The addition of 40% cholesterol to either DOPC or DPPC changes the WAXS pattern due to an increase in acyl chain orientational order, which is one of the main properties distinguishing the cholesterol-rich liquid-ordered (Lo) phase from the liquid-disordered (Ld) phase. In contrast, powder x-ray data from multilamellar vesicles does not yield information about orientational order, and the scattering from the Lo and Ld phases looks similar. An analytical model to describe the relationship between the chain orientational distribution and WAXS data was used to obtain an average orientational order parameter, Sx-ray. When 40% cholesterol is added to either DOPC or DPPC, Sx-ray more than doubles, consistent with previous NMR order parameter measurements. By combining information about the average chain orientation with the chain-chain correlation spacing, we extended a commonly used method for calculating areas for gel-phase lipids to fluid-phase lipids and obtained agreement to within 5% of literature values.  相似文献
